Abstract
本发明公开了一种工程化外泌体在糖尿病足溃疡的皮肤再生修复中的应用及其制备方法,所述的工程化外泌体是应用外泌体作为基因载体、装载促进血管再生的基因,同时,应用干细胞来源的外泌体自身含有多种活性因子,促进创面抗感染和血管网络、神经的修复。本发明利用成体干细胞来源的外泌体替代因免疫原性、致瘤性原因导致临床使用严格的干细胞,且协同装载的活性分子的基因货物,具有多功能,治疗效果好的特性,可广泛应用于临床上皮肤创面缺损的修复,尤其是糖尿病足溃烂皮肤的再生修复。
Description
技术领域
本发明属于生物医药和组织工程领域,具体涉及一种新型工程化外泌体在糖尿病足溃疡的皮肤再生修复中的应用及其制备方法。
背景技术
外泌体是由多种类型细胞分泌的直径约为40-200nm的胞外囊泡,含有不同种类的microRNAs、蛋白质和脂质等具有生物学活性的物质,在不同细胞间进行信息传递,调节细胞间的信号传导,在创伤修复、炎症反应和肿瘤发生发展等许多生理和病理过程中发挥着重要的功能。干细胞是具有高度自我更新能力与多向分化潜能的干细胞,目前已应用于临床治疗多种疾病。但是直接采用干细胞进行临床治疗具有较大的免疫排斥的风险。因此,利用干细胞分泌的外泌体治疗疾病可以避免干细胞的免疫原性与致瘤性的风险。
随着对外泌体的进一步深入研究,人们将目光从外泌体本身的功能转移至其优越的结构特性,具有磷脂双分子层结构的外泌体是一种天然的生物活性载体,与传统的药物基因载体相比,外泌体来源于细胞具有完美的生物相容性,并且能够逃避机体的免疫识别,从而将装载的货物高效地转运至功能细胞中。因此,将具有一些特定功能的基因装载外泌体中,形成一种工程化的外泌体,协同外泌体自身发挥更多的功能。
糖尿病足是由糖尿病引发的下肢缺血性疾病,糖尿病患者不仅体内血糖高,而且下肢血管病变造成血管闭塞和供血障碍导致足部组织得不到足够的营养物质,微小的创伤即可引起微生物的侵袭与感染,并且感染易于扩散,导致下肢持续反复感染,出现难愈性溃疡。这种因血管病变引发的皮肤溃疡与普通的皮肤创伤不同,目前临床上针对糖尿病足溃疡的治疗方法有限,一般为抗感染的药物,甚至面临截肢的风险,然而长时间施用抗生素会产生一定的耐药性,截肢更是会严重影响患者的生活水平和质量,使患者长时间饱受身体和心理创伤的困扰。
因此,研究如何能使糖尿病足溃疡皮肤尽快修复愈合,避免反复感染,已成为临床医生迫切需要解决的难题。
发明内容
针对现有技术以上缺陷或改进需求中的至少一种,本发明提供了一种能够针对糖尿病足发病机理而设计的工程化外泌体,本发明的目的是提供了工程化外泌体在糖尿病足溃疡的皮肤再生修复中的应用。工程化外泌体是应用外泌体作为基因载体装载促进血管再生的基因,同时,应用干细胞来源的外泌体自身含有多种活性因子,促进创面抗感染和血管网络、神经的修复。
为实现上述目的,按照本发明的一个方面,提供了一种工程化外泌体在糖尿病足溃疡的皮肤再生修复中的应用,所述工程化外泌体是由用于促进血管再生的基因导入到外泌体中所获得。
优选地,所述外泌体来源于成体干细胞,所述成体干细胞包括骨髓间充质干细胞、造血干细胞、脂肪干细胞。
优选地,所述促进血管再生的基因包括VEGF、FGF、angs、TGF-β、TNF。
本发明针对糖尿病足的发病机制,设计用于糖尿病足溃疡皮肤修复的新型工程化外泌体。糖尿病足因下肢血管病变造成血管闭塞和供血障碍导致足部组织得不到足够的营养物质,微小的创伤即可引起微生物的侵袭与感染,并且感染易于扩散,导致下肢持续反复感染,出现难愈性溃疡。因此,本发明通过引入干细胞来源的外泌体及促进血管再生的基因制备出工程化外泌体来协同刺激皮肤创面血管的再生,进而促进创面愈合。
为实现上述目的,按照本发明的另一个方面,还提供了一种如前所述的应用中的工程化外泌体的制备方法,包括以下步骤:
S1、干细胞外泌体的提取:采用超高速离心法提取外泌体;
S2、干细胞外泌体的鉴定:将步骤S1中得到的干细胞外泌体采用透射电镜观察外泌体形貌、蛋白质免疫印迹对外泌体标志蛋白进行鉴定、纳米颗粒跟踪分析评估外泌体的粒径分布;
S3、促进血管再生的基因的导入:将促进血管再生的基因导入到步骤S2通过鉴定后的干细胞外泌体中。
优选地,步骤S1包括:
S1.1:将3-5代干细胞生长至汇合度80%时,弃干细胞完全培养基,换为不含血清的培养基饥饿48h,收集饥饿后的培养基;
S1.2:将步骤S1.1所得的培养基于4℃,2000×g离心10min,弃沉淀,收集上清;
S1.3:将步骤S1.2所得的上清于4℃,10000×g离心1h,弃沉淀,收集上清;
S1.4:将步骤S1.3所得的上清用0.22μm真空过滤器过滤,收集滤液;
S1.5:将步骤S1.4所得的滤液于4℃,100000×g离心1h,弃上清,收集沉淀;
S1.6:将步骤S1.5所得的沉淀用与培养基等体积的PBS将沉淀重悬,于4℃,100000×g离心1h,弃上清,收集沉淀;
S1.7:将步骤S1.6所得的沉淀用100-200μL PBS重悬,得到外泌体。
优选地,在步骤S2中,若同时满足以下三个条件:
透射电镜观察外泌体形貌:茶杯托样;
蛋白质免疫印迹对外泌体标志蛋白进行鉴定:标记物抗体呈阳性;
纳米颗粒跟踪分析评估外泌体的粒径分布:40-200nm;
则鉴定出步骤S1的提取物为外泌体。
优选地,步骤S3具体为:采用电穿孔的方法将促进血管再生的基因导入到步骤S2通过鉴定后的干细胞外泌体中。
优选地,步骤S3具体为:采用电穿孔的方法将促进血管再生的基因导入到步骤S2通过鉴定后的干细胞外泌体中。
优选地,步骤S3包括:
S3.1:将促进血管再生的基因和干细胞外泌体按1:3wt加入0.4cm厚度的电转杯中,用电穿孔缓冲液稀释至400μL;
S3.2:将步骤S3.1的电转杯放入电转仪中,设置电转的参数,对基因和外泌体进行电转;
S3.3:将步骤S3.2的电转产物转移至1.5mL EP管中,于4℃,25000×g离心1h,去上清,得到纯化后导入了基因的外泌体,即工程化外泌体。
上述优选技术特征只要彼此之间未构成冲突就可以相互组合。
总体而言,通过本发明所构思的以上技术方案与现有技术相比,具有以下有益效果:
1、本发明提供了一种能够针对糖尿病足发病机理而设计的工程化外泌体,并提供了工程化外泌体在糖尿病足溃疡的皮肤再生修复中的应用。工程化外泌体是应用外泌体作为基因载体装载促进血管再生的基因,同时,应用干细胞来源的外泌体自身含有多种活性因子,促进创面抗感染和血管网络、神经的修复。

为了使本发明的目的、技术方案及优点更加清楚明白,以下结合附图及实施例,对本发明进行进一步详细说明。应当理解,此处所描述的具体实施例仅仅用以解释本发明,并不用于限定本发明。此外,下面所描述的本发明各个实施方式中所涉及到的技术特征只要彼此之间未构成冲突就可以相互组合。下面结合具体实施方式对本发明进一步详细说明。
实施例1
本发明提供了一种工程化外泌体在糖尿病足溃疡的皮肤再生修复中的应用及其制备方法。
一、骨髓间充质干细胞外泌体的提取及鉴定
1、骨髓间充质干细胞外泌体的提取
采用超高速离心法提取外泌体,步骤如下:
步骤(1):骨髓间充质干细胞(购自Cyagen)用骨髓间充质干细胞完全培养基(购自Cyagen)传代培养至3-5代,进一步扩大培养待骨髓间充质干细胞生长至汇合度80%左右时,弃培养基,换为不含血清的DMEM-L培养基饥饿48h,收集饥饿后的培养基;
步骤(2):将步骤(1)所得的培养基于4℃,2000×g离心10min,弃沉淀,收集上清;
步骤(3):将步骤(2)所得的上清于4℃,10000×g离心1h,弃沉淀,收集上清;
步骤(4):将步骤(3)所得的上清用0.22μm真空过滤器(购自Millipore)过滤,收集滤液;
步骤(5):将步骤(4)所得的滤液于4℃,100000×g离心1h,弃上清,收集沉淀;
步骤(6):将步骤(5)所得的沉淀用与培养基等体积的PBS(购自Hyclone)将沉淀重悬,于4℃,100000×g离心1h,弃上清,收集沉淀;
步骤(7):将步骤(6)所得的沉淀用100-200μL PBS重悬,得到外泌体。
2、骨髓间充质干细胞外泌体的鉴定
将上述得到的骨髓间充质干细胞外泌体采用透射电镜(TEM)、蛋白质免疫印迹(Western Blot)、纳米颗粒跟踪分析(NTA)进行鉴定。若同时满足以下三个条件:透射电镜观察外泌体形貌:茶杯托样;蛋白质免疫印迹对外泌体标志蛋白进行鉴定:标记物抗体呈阳性;纳米颗粒跟踪分析评估外泌体的粒径分布:40-200nm;则鉴定出步骤S1的提取物为外泌体。
TEM观察外泌体形貌:将孔径为2nm的载样铜网放置于液体样品制样槽中,将提取步骤(7)得到的外泌体悬液用PBS稀释至100ng/μL,吸取20μL外泌体悬液,滴加到铜载网上,形成液滴,静置10min;用滤纸吸去载网上的多余液体,滴加20μL 2%的磷钨酸染液,形成液滴,负染5min左右至载网变色;用干燥滤纸吸去多余染液,用烤灯烘干,将载样铜网置于TEM样品小室内,TEM观察并拍照。
TEM结果如图1所示,骨髓间充质干细胞外泌体在TEM下呈茶杯托样。
Western Blot检测外泌体表面标记物:将超高速离心提取步骤(6)得到的外泌体沉淀用含PMSF的RIPA裂解液裂解,用BCA法检测蛋白浓度,加入5×蛋白上样缓冲液,95℃变性5min,得到外泌体样品。经过电泳、转膜、封闭、孵育抗体、显色、观察等步骤,分别得到外泌体表面标记物CD63、TSG101是否呈阳性的表达。
Western Blot结果如图2所示,骨髓间充质干细胞外泌体的标志性抗体CD63、TSG101均呈阳性。
NTA检测外泌体的粒径分布:将-80℃保存的100μL蛋白浓度为1200ng/μL的外泌体样品用干冰运输寄送上海逍鹏生物检测,所用仪器为ZetaView PMX 110(ParticleMetrix,Meerbusch,Germany),软件版本ZetaView 8.04.02SP2。
NTA结果如图3所示,骨髓间充质干细胞外泌体的粒径集中分布在118nm左右,呈单峰,密度均一,纯度较高。
二、工程化外泌体的制备
采用电穿孔的方法将VEGF质粒DNA导入到骨髓间充质干细胞外泌体中(其他促进血管再生的基因如FGF、angs、TGF-β、TNF的导入同理,不再赘述),步骤如下:
步骤(1):将10μg VEGF质粒DNA和30μg骨髓间充质干细胞外泌体加入0.4cm厚度的电转杯(Bio-Rad)中,用电穿孔缓冲液(0.4M sucrose(13.7%),2.4g/L HEPES,6g/L KCl,600mg/L CaCl2·2H2O,pH 7.2(with KOH))稀释至400μL;
步骤(2):将步骤(1)的电转杯放入电转仪中,设置电转的参数(电压、时间、脉冲次数)对VEGF质粒DNA和外泌体进行电转;
步骤(3):将步骤(2)的电转产物转移至1.5mL EP管中,于4℃,25000×g离心1h,去上清,得到纯化后导入了VEGF质粒DNA的外泌体,即工程化外泌体。
三、工程化外泌体在糖尿病足溃疡的皮肤再生修复中的应用
以所得的工程化外泌体为活性成分配合敷料用于糖尿病足溃疡的皮肤创面的再生修复。
实施例2
本发明提供了一种工程化外泌体在糖尿病足溃疡的皮肤再生修复中的应用及其制备方法。
一、造血干细胞外泌体的提取及鉴定
1、造血干细胞外泌体的提取
采用超高速离心法提取外泌体,步骤如下:
步骤(1):造血干细胞(购自ATCC)用造血干细胞培养基(购自Stemcell)传代培养至3-5代,进一步扩大培养待造血干细胞生长至汇合度80%左右时,弃培养基,换为不含血清的DMEM-L培养基饥饿48h,收集饥饿后的培养基;
步骤(2):将步骤(1)所得的培养基于4℃,2000×g离心10min,弃沉淀,收集上清;
步骤(3):将步骤(2)所得的上清于4℃,10000×g离心1h,弃沉淀,收集上清;
步骤(4):将步骤(3)所得的上清用0.22μm真空过滤器(购自Millipore)过滤,收集滤液;
步骤(5):将步骤(4)所得的滤液于4℃,100000×g离心1h,弃上清,收集沉淀;
步骤(6):将步骤(5)所得的沉淀用与培养基等体积的PBS(购自Hyclone)将沉淀重悬,于4℃,100000×g离心1h,弃上清,收集沉淀;
步骤(7):将步骤(6)所得的沉淀用100-200μL PBS重悬,得到外泌体。
2、造血干细胞外泌体的鉴定
将上述得到的造血干细胞外泌体采用透射电镜(TEM)、蛋白质免疫印迹(WesternBlot)、纳米颗粒跟踪分析(NTA)进行鉴定。

步骤(3):将步骤(2)的电转产物转移至1.5mL EP管中,于4℃,25000×g离心1h,去上清,得到纯化后导入了VEGF质粒DNA的外泌体,即工程化外泌体。
三、工程化外泌体在糖尿病足溃疡的皮肤再生修复中的应用
以所得的工程化外泌体为活性成分配合敷料用于糖尿病足溃疡的皮肤创面的再生修复。
实施例3
本发明提供了一种工程化外泌体在糖尿病足溃疡的皮肤再生修复中的应用及其制备方法。
一、脂肪干细胞外泌体的提取及鉴定
1、脂肪干细胞外泌体的提取
采用超高速离心法提取外泌体,步骤如下:
步骤(1):脂肪干细胞(购自Cyagen)用脂肪干细胞培养基(购自Cyagen)传代培养至3-5代,进一步扩大培养待脂肪干细胞生长至汇合度80%左右时,弃培养基,换为不含血清的DMEM-L培养基饥饿48h,收集饥饿后的培养基;
步骤(2):将步骤(1)所得的培养基于4℃,2000×g离心10min,弃沉淀,收集上清;

将上述得到的脂肪干细胞外泌体采用透射电镜(TEM)、蛋白质免疫印迹(WesternBlot)、纳米颗粒跟踪分析(NTA)进行鉴定。
TEM观察外泌体形貌:将孔径为2nm的载样铜网放置于液体样品制样槽中,将提取步骤(7)得到的外泌体悬液用PBS稀释至100ng/μL,吸取20μL外泌体悬液,滴加到铜载网上,形成液滴,静置10min;用滤纸吸去载网上的多余液体,滴加20μL 2%的磷钨酸染液,形成液滴,负染5min左右至载网变色;用干燥滤纸吸去多余染液,用烤灯烘干,将载样铜网置于TEM样品小室内,TEM观察并拍照。

Claims (9)
1.一种工程化外泌体在糖尿病足溃疡的皮肤再生修复中的应用,其特征在于:所述工程化外泌体是由用于促进血管再生的基因导入到外泌体中所获得。
2.如权利要求1所述的工程化外泌体在糖尿病足溃疡的皮肤再生修复中的应用,其特征在于:
所述外泌体来源于成体干细胞,所述成体干细胞包括骨髓间充质干细胞、造血干细胞、脂肪干细胞。
3.如权利要求1或2所述的工程化外泌体在糖尿病足溃疡的皮肤再生修复中的应用,其特征在于:
所述促进血管再生的基因包括VEGF、FGF、angs、TGF-β、TNF。
4.一种如权利要求1-3任一项所述的应用中的工程化外泌体的制备方法,其特征在于,包括以下步骤:
S1、干细胞外泌体的提取:采用超高速离心法提取外泌体;
S2、干细胞外泌体的鉴定:将步骤S1中得到的干细胞外泌体采用透射电镜观察外泌体形貌、蛋白质免疫印迹对外泌体标志蛋白进行鉴定、纳米颗粒跟踪分析评估外泌体的粒径分布;
S3、促进血管再生的基因的导入:将促进血管再生的基因导入到步骤S2通过鉴定后的干细胞外泌体中。
5.如权利要求4所述的工程化外泌体的制备方法,其特征在于:
步骤S1包括:
S1.1:将3-5代干细胞生长至汇合度80%时,弃干细胞完全培养基,换为不含血清的培养基饥饿48h,收集饥饿后的培养基;
S1.2:将步骤S1.1所得的培养基于4℃,2000×g离心10min,弃沉淀,收集上清;
S1.3:将步骤S1.2所得的上清于4℃,10000×g离心1h,弃沉淀,收集上清;
S1.4:将步骤S1.3所得的上清用0.22μm真空过滤器过滤,收集滤液;
S1.5:将步骤S1.4所得的滤液于4℃,100000×g离心1h,弃上清,收集沉淀;
S1.6:将步骤S1.5所得的沉淀用与培养基等体积的PBS将沉淀重悬,于4℃,100000×g离心1h,弃上清,收集沉淀;
S1.7:将步骤S1.6所得的沉淀用100-200μL PBS重悬,得到外泌体。
6.如权利要求4或5所述的工程化外泌体的制备方法,其特征在于:
在步骤S2中,若同时满足以下三个条件:
透射电镜观察外泌体形貌:茶杯托样;
蛋白质免疫印迹对外泌体标志蛋白进行鉴定:标记物抗体呈阳性;
纳米颗粒跟踪分析评估外泌体的粒径分布:40-200nm;
则鉴定出步骤S1的提取物为外泌体。
7.如权利要求4所述的工程化外泌体的制备方法,其特征在于:
步骤S3具体为:采用电穿孔的方法将促进血管再生的基因导入到步骤S2通过鉴定后的干细胞外泌体中。
8.如权利要求6所述的工程化外泌体的制备方法,其特征在于:
步骤S3具体为:采用电穿孔的方法将促进血管再生的基因导入到步骤S2通过鉴定后的干细胞外泌体中。
9.如权利要求7或8所述的工程化外泌体的制备方法,其特征在于:
步骤S3包括:
S3.1:将促进血管再生的基因和干细胞外泌体按1:3wt加入0.4cm厚度的电转杯中,用电穿孔缓冲液稀释至400μL;
S3.2:将步骤S3.1的电转杯放入电转仪中,设置电转的参数,对基因和外泌体进行电转;
S3.3:将步骤S3.2的电转产物转移至1.5mL EP管中,于4℃,25000×g离心1h,去上清,得到纯化后导入了基因的外泌体,即工程化外泌体。
