技术领域
本发明属于功能食品领域,具体涉及一种具有减肥功能的植物益生菌发酵物及其制备方法和用途。
背景技术
随着摄入热量的增加和运动量的减少,人群中的肥胖率越来越高。相应地,与减肥、瘦身相关的食品数量激增。
然受制于国内法规与加工工艺的限制,能达到预期效果的减肥瘦身产品并不多见。更有不良商家为了追求效果不惜在食品、保健食品中非法添加药品,对消费者造成极大的身体与精神伤害。
传统中医认为,肥胖与痰、湿、气虚等有关。如《内经》曰:“素嗜肥甘,好酒色,体肥痰盛”。另有一些医者提到“肥人多痰湿”,“肥白人多湿”,“肥人沉困怠情是气虚”。中药减肥强调益气、健脾、化痰等着手,利用中药的优势,通过益消泻的方法,调整人体各器官恢复原有功能,将人体多余的脂肪转化成能量,促进在人体内的能量代谢,从而达到健康减肥的目的。
目前,以药食同源的中药材发酵制取减肥食品的报道还比较鲜见。
发明内容
本发明的首要目的在于提供一种具有减肥功能的植物益生菌发酵物及其制备方法,该方法将优选的药食同源药材粉碎,经不同益生菌、不同发酵条件发酵,发酵完成后榨汁,浓缩或喷雾干燥而成。发酵过程中益生菌发酵产生的有机酸、各种酶协同作用,使药材的组织更有效地分解、细胞壁更容易破碎,进而提高药材有效成分的浸出比例,增强原药材的功能;从而达到减肥的效果。此外益生菌的代谢产物还能促进亚油酸、油酸转化为共轭亚油酸,进一步提成产品效果。
本发明的另一目的在于提供上述植物益生菌发酵物的用途。

一种植物益生菌发酵物,原料包括按重量份计的以下组分:

本发明植物益生菌发酵物的原料配方中,决明子含蒽醌类化合物,主要为大黄素,具有降压、调脂的功效,泻下作用轻微;在多种益生菌发酵的体系中大黄素可减少氧化,并在各类酶的作用下与糖结合成苷类(大黄素-1-O-β-D-葡萄糖苷和大黄素-8-O-β-D-葡萄糖苷),则可发挥较强的泻下作用。
荷叶含有荷叶碱,荷叶碱是一种应用于降脂,降压,消脂保健成分,它有效分解体细胞脂肪,提高其对胰岛素的感知度,防止高脂疾病发生。在发酵过程中,枯草芽孢杆菌分泌的各类酶可加速荷叶粉中的细胞壁、纤维分解,进而使荷叶碱的浸出比例增加。
郁李仁所含郁李仁甙对实验动物有强烈泻下作用。山楂有清脂和加速排解体内废物的功能,所含的解脂酶能促进脂肪类食物的消化,促进胃液分泌和增加胃内酶素等功能。中医认为,山楂具有消积化滞、收敛止痢、活血化淤等瘦身功效。
共轭亚油酸可以有效地降低体内脂肪组织比例,促进体内新陈代谢能力,形成良性循环,使减肥更易实现。共轭亚油酸中约有10-20%油酸、3%的亚油酸,在植物乳酸杆菌发酵过程中产生的代谢产物异构化酶及乳酸可以促使油酸、亚油酸转化为共轭亚油酸,起到协同、增效作用。
本发明采用的益生菌中,枯草芽孢杆菌(菌种编号:CMCC(B)63501,来源于广东省微生物研究所)能够迅速消耗发酵体系内的游离氧,从而为植物乳酸杆菌、乳双歧杆菌等厌氧菌优化生产环境。枯草芽孢杆菌还能够分泌淀粉酶、脂肪酶、蛋白酶、和纤维素酶等多种内源酶。内源酶与植物乳酸杆菌发酵产生的乳酸协同作用,加速植物纤维、淀粉的分解,进而析出有效成分。此外能合成维生素B1、B2、B6、烟酸等多种B族维生素为后续发酵提供养分。
植物乳酸杆菌(菌种编号:R1012采购自加拿大拉曼集团)兼性厌氧,分解糖的能力强,分解蛋白质类的能力极低,分解糖的主要终产物是乳酸。发酵过程产出特有的乳酸杆菌素,乳酸杆菌素是一种生物型的防腐剂;产出的DL-乳酸、1,6~二磷酸果糖醛缩酶、异构化酶、单磷酸己糖可促进油酸、亚油酸转化为共轭亚油酸。
乳双歧杆菌(菌种编号:Bb-12采购自科汉森公司)专性厌氧,可分泌多种酶类、有机酸,促进发酵体系内的细胞降解;并在发酵过程中合成丙氨酸、天冬氨酸和苏氨酸等人体必需的营养物质,在减肥的同时维持人体的营养需求。另外乳双歧杆菌发酵产物具有良好抗氧化效果,可保护决明子中大黄素的氧化。乳双歧杆菌利用枯草芽孢杆菌发酵产生的维生素、烟酸等养分;在经枯草芽孢杆菌、植物乳酸杆菌发酵后的低氧环境下迅速繁殖;植物中存在的无机盐、维生素、氨基酸为乳双歧杆菌发酵的必备养分。
上述植物益生菌发酵物的制备方法,包括以下步骤:
(1)将决明子、荷叶、郁李仁、山楂粉碎,连同共轭亚油酸、低聚果糖、蛋白胨投入发酵罐中,加水混匀,调节pH值为5.5-5.8,然后加入枯草芽孢杆菌,30-40℃下、低速搅拌、敞口发酵24小时以上;
枯草芽孢杆菌为需氧菌,在特定的pH值和温度下可快速增殖;设置搅拌可增加发酵液中的含氧量,有助于充分发酵。
(2)调节步骤(1)所得混合液的pH值为5.7-6.2,加入植物乳酸杆菌,设置温度为35-42℃,密闭发酵24小时以上;其中,每间隔不长于8小时开启发酵罐搅拌一次,每次搅拌不短于5分钟,低速搅拌;
步骤(1)和(2)所述低速搅拌的转速为1-3rpm;
植物乳酸杆菌为兼性厌氧菌,间隔搅拌可保证植物乳酸杆菌发酵所需供氧量,也可进一步消耗发酵发酵罐内的氧气,为下一步密闭发酵创造条件。
(3)通过加料口向步骤(2)发酵得到的混合液加入乳双歧杆菌,30-35℃下、密闭发酵24小时以上;发酵完成后,将混合液中的固体分离,将发酵液加热浓缩灭活益生菌,得到植物益生菌发酵物;
步骤(3)所述的加热浓缩,加热温度100℃,加热时间30分钟以上;
步骤(3)所述将混合液中的固体分离,可以采用榨汁、离心或过滤等方法;
步骤(3)所得的植物益生菌发酵物,可以直接用于生产饮料、口服液等食品;也可进一步喷雾干燥,制得合适使用的粉料。
本发明的植物益生菌发酵物可以用制备具有减肥功效的保健品和食品。
本发明相对于现有技术具有如下的优点及效果:
1、本发明提供的具有减肥功能的植物益生菌发酵物,依照中医理论选用药食同源的药材,通过微生物发酵的方法使药材含有的减肥效用的成分充分浸出,使组方达到理想的减肥效果。
2、本发明提供的植物益生菌发酵物的制备方法中,三个发酵步骤依次进行,上一次发酵为下一次发酵提供条件;下一步发酵利用上一步发酵的发酵产物。三个发酵步骤的不同发酵产物协同作用于食同源药材,增加有效成分的得率。
3、本发明提供的植物益生菌发酵物的制备方法中,三个发酵步骤产生不同的益生菌代谢产物,与组方中的成分协同作用,达到增强减肥效果的目的;各个发酵阶段产生的有机酸、酶增强了植物纤维、细胞壁的分解,增强有效成分的浸出比例。
4、本发明提供的植物益生菌发酵物的制备方法中,采用高温浓缩,以杀灭发酵过程中的益生菌,便于满足后期制剂需求。

实施例1
一种植物益生菌发酵物,原料包括按重量份计的以下组分:决明子:20份、荷叶:20份、郁李仁:20份、山楂:15份、共轭亚油酸:4份、低聚果糖:7份、蛋白胨:4份、植物乳酸杆菌:1份、乳双歧杆菌:1.5份、枯草芽孢杆菌:2份;水:375份。
该植物益生菌发酵物的制备方法包括以下步骤:
1)决明子、荷叶、郁李仁、山楂,分别粉碎过20目筛;
2)将1)产生的药材粉、共轭亚油酸、低聚果糖、蛋白胨投入带搅拌、控温功能的不锈钢发酵灌装中,加入5倍的水,搅拌均匀,的混合料液。

4)二次发酵:设置发酵罐内温度为42℃;使用柠檬酸、碳酸氢钠为调节剂,调节pH值为5.7;向发酵罐内加入植物乳酸杆菌。关闭发酵罐密封口,发酵24小时备用。其中,每间隔不长于8小时开启发酵罐搅拌一次,每次搅拌不短于5分钟,低速搅拌(搅拌转速为1-3转/分钟);
5)三次发酵:设置发酵罐内温度为35℃;向发酵罐内加入乳双歧杆菌,关闭搅拌桨。关闭发酵罐密封口,发酵24小时备用。
6)经三次发酵的混合料液通过压榨机榨汁,备用。
7)使用常压浓缩设施,设置浓缩温度为100℃,将发酵液浓缩至相对密度为1.18,浓缩时间不小于30分钟,制成植物益生菌发酵浓缩液。
所述植物益生菌发酵浓缩液可直接用于口服液、饮料、颗粒剂制剂使用。
实施例2
一种植物益生菌发酵物,包括按重量份计的以下组分:决明子:15份、荷叶:15份、郁李仁:15份、山楂:10份、共轭亚油酸:2份、低聚果糖:4份、蛋白胨:2份、植物乳酸杆菌:0.5份、乳双歧杆菌:1份、枯草芽孢杆菌:1份、水:275份。
该植物益生菌发酵物的制备方法包括以下步骤:
1)决明子、荷叶、郁李仁、山楂,分别粉碎过20目筛;
2)将1)产生的药材粉、共轭亚油酸、低聚果糖、蛋白胨投入带搅拌、控温功能的不锈钢发酵灌装中,加入5倍的水,搅拌均匀,的混合料液。
3)一次发酵:设置发酵罐内温度为40℃;使用柠檬酸、碳酸氢钠为调节剂,调节pH值为5.5;向发酵罐内加入枯草芽孢杆菌,设置搅拌转速为1转/分钟。敞口发酵24小时备用。

8)使用喷雾干燥设备,选用适当的辅料,对浓缩液进行喷雾干燥处理,得干燥粉料。
所述具有减肥功能植物益生菌发酵物喷雾干燥粉使用的辅料为麦芽糊精、乳糖等。
所述具有减肥功能植物益生菌发酵物喷雾干燥粉因在喷雾过程中辅料对料液有包埋作用,保存时间长,功效成分不易流失。可直接用于固体饮料、压片糖果等产品制剂使用。
实施例3
一种植物益生菌发酵物,原料包括按重量份计的以下组分:决明子:25份、荷叶:25份、郁李仁:25份、山楂:20份、共轭亚油酸:6份、低聚果糖:10份、蛋白胨:6份、植物乳酸杆菌:1.5份、乳双歧杆菌:2份、枯草芽孢杆菌:3份;水:475份。
该植物益生菌发酵物的制备方法与实施例1一致。
试验例1:
1.1根据《中华人民共和国药典》2015年版一部中荷叶碱的测试方法,测试组方经益生菌发酵(本发明方法)及未经益生菌发酵的组方荷叶碱的含量。
1.2色谱条件与系统适用性试验:以十八烷基硅烷键合硅胶为填充剂;以乙腈:水:三乙胺:冰醋酸(27:70.6:1.6:0.78)为流动相;检测波长为270nm。理论板数按荷叶碱峰计算应不低于2000。
1.3对照品溶液的制备:取荷叶碱对照品适量,精密称定,加甲醇制成每1ml含16μg的溶液,即得。
1.4样品制备:发酵液样品制备:取实施例1最后制得的发酵液,按1:10比例用甲醇稀释,过0.45μm微孔滤膜备用;
未发酵样品制备:使用实施例1相同的发酵原料(不含三种益生菌),相同的步骤;制得的中药复方提取物,浓缩至与发酵液组相同浓度(相对密度为1.18);按1:10比例用甲醇稀释,过0.45μm微孔滤膜备用。
1.5测定法:分别精密吸取标准品溶液、发酵液样品、未发酵液样品各20μL,注入液相色谱仪,测定,即得。
所得荷叶碱液相图谱如图1所示。由图可见发酵液与未发酵液所含成分相似,在15.50min左右出现荷叶碱峰型,发酵液峰面积明显大于未发酵液峰面积。试验得出发酵可使荷叶碱的浸出比例大幅增加。
试验例2:
2.1动物实验参考《保健食品检验与评价技术规范(2003版)》制定。
2.2试验动物:选用雄性大鼠,适应期结束时,体重200±20g。
2.3试验材料:
发酵液组样品:取实施例1制备的具有减肥功能益生菌发酵物作为样品;
未发酵组样品:与实施例1相同的发酵原料(不含三种益生菌),相同的步骤;制得的中药复方提取物,浓缩至与发酵液组相同浓度(相对密度为1.18),作为未发酵组供试样品;
空白对照组样品:纯化水。
2.4实验步骤:适应期结束后按体重随机分成2组,10只大鼠给予维持饲料作为空白对照组,80只大鼠给予高热量模型饲料。每周记录给食量、撒食量、剩食量,称量体重1次。
维持饲料配方为:东北玉米、小麦、西班牙苜蓿草、秘鲁鱼粉、美国鸡肉粉、豆粕、豆油、葵花籽油、复合氨基酸、复合维生素矿物质。饲料采购自江苏省协同医药生物工程有限责任公司,产品符合符合GB 14924.1《实验动物配合饲料通用质量标准》、GB 13078《饲料卫生标准》、GB 14924.2《实验动物配合饲料卫生标准》和GB 14924.3《实验动物配合饲料营养成分》要求。
高热量模型饲料配方为:78.8%维持饲料、1%胆固醇、10%蛋黄粉、10%猪油、0.2%胆盐。
喂养2周后,给予高热量饲料的80只大鼠按体重增重排序,淘汰体重增重较低的40只肥胖抵抗大鼠。将筛选出的40只肥胖敏感大鼠再给予高热量饲料6周,空白对照组同时给予维持饲料。
造模期结束后,40只肥胖敏感大鼠按体重随机分成4组,分别为空白对照组、未发酵组、发酵液两个剂量组。每周记录给食量、称量体重1次。空白对照组、未发酵组和发酵液两个剂量组给予高热量模型饲料。发酵液两个剂量组:低剂量组5ml/天;高剂量组10ml/天;未发酵组、空白对照组给予高剂量组等量的纯化水;投喂6周。
试验结束后,称体重,1%戊巴比妥钠(0.5ml/100g BW)麻醉,解剖取肾周围脂肪、睾丸周围脂肪,并称重,计算脂/体比。
2.5试验结果

注:与空白组相比:**P<0.01;摄食量不显著低于空白组。
由表1所列结果可见,试验末发酵液高剂量组动物体重增重低于空白对照组,且差异具有高度显著性(P<0.01);同时两组动物的摄食量不显著低于空白组。

由表2所列结果可见,试验末发酵液高剂量组动物“体脂/终体重”低于空白组,且差异具有显著性(P<0.05);发酵液高剂量组动物“体脂/终体重”低于未发酵高剂量组,且差异具有显著性(P<0.05);同时两组动物的摄食量不显著低于空白组。
本次动物减肥降血脂实验结果表明:本发明的植物益生菌发酵物每日摄入量为10ml时对大鼠具有减肥降脂作用。实验末发酵液高剂量组动物,在体重增加,“体脂/终体重”低于未发酵高剂量组,且差异具有显著性(P<0.05)。
上述实施例为本发明较佳的实施方式,但本发明的实施方式并不受上述实施例的限制,其他的任何未背离本发明的精神实质与原理下所作的改变、修饰、替代、组合、简化,均应为等效的置换方式,都包含在本发明的保护范围之内。
Claims (10)
2.根据权利要求1所述的植物益生菌发酵物,其特征在于:所述的益生菌由植物乳酸杆菌、乳双歧杆菌和枯草芽孢杆菌组成。
3.根据权利要求1所述的植物益生菌发酵物,其特征在于:所述的益生菌由0.5-1.5份植物乳酸杆菌、1-2份乳双歧杆菌和1-3份枯草芽孢杆菌组成。
5.权利要求1-4任一项所述的植物益生菌发酵物的制备方法,其特征在于包括以下步骤:
(1)将决明子、荷叶、郁李仁、山楂粉碎,连同共轭亚油酸、低聚果糖、蛋白胨投入发酵罐中,加水混匀,调节pH值为5.5-5.8,然后加入枯草芽孢杆菌,30-40℃下、低速搅拌、敞口发酵24小时以上;
(2)调节步骤(1)所得混合液的pH值为5.7-6.2,加入植物乳酸杆菌,设置温度为35-42℃,密闭发酵24小时以上;其中,每间隔不长于8小时开启发酵罐搅拌一次;
(3)通过加料口向步骤(2)发酵得到的混合液加入乳双歧杆菌,30-35℃下、密闭发酵24小时以上;发酵完成后,将混合液中的固体分离,将发酵液加热浓缩灭活益生菌,得到植物益生菌发酵物。
6.根据权利要求5所述的制备方法,其特征在于:步骤(2)所述开启发酵罐搅拌一次,是每次搅拌不短于5分钟,低速搅拌。
7.根据权利要求5或6所述的制备方法,其特征在于:步骤(1)和(2)所述低速搅拌的转速为1-3rpm。
8.根据权利要求5所述的制备方法,其特征在于:步骤(3)所述的加热浓缩,加热温度100℃,加热时间30分钟以上。
9.根据权利要求5所述的制备方法,其特征在于:步骤(3)所述将混合液中的固体分离,是采用榨汁、离心或过滤方法。
10.权利要求1-4任一项所述的植物益生菌发酵物在保健品和食品中的应用。
