Abstract
本发明提供了一种嵌合性抗原受体及表达其的巨噬细胞、调节巨噬细胞极化的方法和应用,涉及生物技术领域。该嵌合性抗原受体的胞内结构域中含有IFN‑γ受体,表达该嵌合性抗原受体的免疫细胞对IFN‑γ的响应能力增强。表达该嵌合性抗原受体的巨噬细胞可以相对较长时间保持M1型状态,增强肿瘤细胞抗原与嵌合性抗原受体结合后巨噬细胞M1型活性。

技术领域
本发明涉及生物技术领域,尤其是涉及一种嵌合性抗原受体及表达其的巨噬细胞、调节巨噬细胞极化的方法和应用。
背景技术
巨噬细胞(英语:Macrophages,缩写为)是一种位于组织内的白细胞,源自单核细胞,而单核细胞又来源于骨髓中的前体细胞。巨噬细胞和单核细胞皆为吞噬细胞,在脊椎动物体内参与非特异性防卫(先天性免疫)和特异性防卫(细胞免疫)。它们的主要功能是以固定细胞或游离细胞的形式对细胞残片及病原体进行噬菌作用(即吞噬以及消化),并激活淋巴球或其他免疫细胞,令其对病原体做出反应。
巨噬细胞是先天固有免疫的重要组成部分,在炎症和宿主防御中发挥核心作用。在响应各种环境因素(例如,微生物产物,受损细胞,活化的淋巴细胞)或在不同的病理生理条件下,巨噬细胞转化为不同的功能表型,即经典活化性巨噬细胞(classicallyactivated macrophages,M1)和选择活化性巨噬细胞(alternatively activatedmacrophages,M2)。成熟的巨噬细胞在各种因素下出现表型及形态分化,即巨噬细胞的极化现象。根据对环境刺激反应的不同,巨噬细胞主要被激活为M1和M2两种表型。
在实体肿瘤组织周围,巨噬细胞约占肿瘤细胞数目的一半以上,充分说明在肿瘤的发生,增殖及转移中巨噬细胞发挥重要作用。在长期肿瘤微环境下,经IFN-γ和LPS等信号活化得到M1型,主要具有抗肿瘤和增强免疫的作用,能分泌炎症因子、趋化因子、效应分子和TNF-α等,其中以膜分子CD80、表面标记CD64等为代表。经IL-4和IL-13等因子活化得到M2型,主要具有抑制免疫反应、促进血管发生、组织修复和促进肿瘤生长的潜能,更多地分泌IL-10、TGF-β、VEGF等因子,以CD163和CD206 相对高表达。肿瘤组织周围的巨噬细胞通过初期抗肿瘤作用(M1型巨噬细胞),逐渐转变为促进肿瘤发展的M2型巨噬细胞。因此,如何利用巨噬细胞调节肿瘤微环境,使其保持M1型状态,并且发挥其天然免疫细胞功能及抗原提呈性,这对肿瘤免疫治疗至关重要。
有鉴于此,特提出本发明。
发明内容
本发明的第一目的在于提供一种嵌合性抗原受体,表达该嵌合性抗原受体的免疫细胞对IFN-γ的响应能力增强。
本发明的第二目的在于提供一种调节巨噬细胞极化的方法。
本发明的第三目的在于提供一种表达所述嵌合性抗原受体的巨噬细胞。
本发明的第三四的在于提供上述嵌合性抗原受体、调节巨噬细胞极化的方或表达所述嵌合性抗原受体的巨噬细胞的应用。
为解决上述技术问题,本发明特采用如下技术方案:
根据本发明的一个方面,本发明提供了一种,所述嵌合性抗原受体含有IFN-γ受体,所述IFN-γ受体位于所述嵌合性抗原受体的胞内结构域。
可选地,所述嵌合性抗原受体的胞内结构域含有共刺激结构域和胞内活化区域,所述IFN-γ受体位于胞内活化区域;
可选地,所述IFN-γ受体的氨基酸序列如SEQ ID NO.2所示;
可选地,所述共刺激结构域含有4-1BB、CD80、CD86和CD28中的至少一种,优选含有4-1BB,4-1BB的氨基酸序列如SEQ ID NO.3所示;
可选地,所述胞内活化区域还含有FcγRI或CD3ζ,优选含有CD3ζ, CD3ζ的氨基酸序列优选如SEQ ID NO.4所示。

与现有技术相比,本发明具有如下有益效果:
仅表达嵌合性抗原受体的巨噬细胞,在肿瘤微环境状态下很难长时间保持M1型状态,巨噬细胞在非M1型状态下,无法刺激杀伤性T细胞活性,从而使肿瘤微环境持续性处于抑制状态。本发明提供的嵌合性抗原受体,将IFN-γ受体插入到嵌合性抗原受体胞内段,可以大大增强IFN-γ对巨噬细胞的激活效应,增强巨噬细胞对IFN-γ的响应,使巨噬细胞在体内可以相对较长时间保持M1型状态,增强肿瘤细胞抗原与嵌合性抗原受体结合后巨噬细胞M1型活性。此时,M1型巨噬细胞可以刺激杀伤性T细胞分泌更多的IFN-γ,继而活化的T细胞可以再刺激巨噬细胞维持M1型状态。使其在最大程度上发挥吞噬肿瘤及抗原提呈能力,激活T细胞,从而充分调节肿瘤微环境,最终改变肿瘤微环境的免疫抑制状态。

具体实施方式
下面将结合实施例对本发明的技术方案进行清楚、完整地描述,显然,所描述的实施例是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
根据本发明的一个方面,本发明提供了一种嵌合性抗原受体。嵌合性抗原受体(Chimeric Antigen Receptor,CAR)主要由胞外结构域、跨膜区和胞内结构域三部分构成。胞外结构域是嵌合性抗原受体在细胞外结合抗原的区域,具有特异性识别并结合肿瘤特异性抗原/相关性抗原的功能,赋予表达嵌合性抗原受体的细胞抗原依赖性吞噬作用;跨膜区通常是由免疫球蛋白超家族组成;胞内结构域是胞内实现信号传导的区域。装载嵌合性抗原受体的免疫细胞与肿瘤细胞表面抗原结合后,膜外抗原结合区将信号传递到胞内信号活化区,启动免疫细胞活化反应。
本发明提供的嵌合性抗原受体中含有IFN-γ受体,IFN-γ受体位于所述嵌合性抗原受体的胞内结构域,能够增强表达该嵌合性抗原受体的免疫细胞。本发明将IFN-γR序列构建在嵌合性抗原受体胞内序列中,增强表达该嵌合性抗原受体的免疫细胞对IFN-γ的响应。当使用巨噬细胞表达本发明提供的嵌合性抗原受体,能够增强巨噬细胞对IFN-γ的响应,增强肿瘤细胞抗原与嵌合性抗原受体结合后巨噬细胞M1型活性。
本发明提供的嵌合性抗原受体的构思是在嵌合性抗原受体的胞内结构域设置IFN-γ受体,以增强细胞对IFN-γ的响应能力,因此可以理解的是,本发明对嵌合性抗原受体中的其他结构部分没有特别的限制,可以根据本领域对嵌合性抗原受体的一般设计原则设置嵌合性抗原受体的其他结构域。

在一些可选的实施例中,所述嵌合性抗原受体的胞外结构域识别间皮质素。间皮质素是位于胞膜上的糖蛋白,在生理情况下,间皮质素仅表达于机体的间皮细胞,在很多肿瘤中都有表达,有研究表明,间皮质素在卵巢癌、胸膜间皮瘤、胰腺癌、胆管癌等均有表达,间皮质素在促进肿瘤的发生、发展的过程中发挥着重要作用。在一些可选的实施例中,所述嵌合性抗原受体的胞外结构域含有靶向间皮质素的单链抗体(Mesothelin-scFv),Mesothelin-scFv的氨基酸序列优选如SEQ ID NO.5所示。
用于实现信号传导的胞内结构域主要含有共刺激结构域和胞内活化区域。在一些优选的实施方式中,IFN-γ受体位于胞内活化区域,IFN-γ受体的氨基酸序列优选如SEQID NO.2所示。共刺激结构域可以增强受体信号,共刺激结构域含有但不限于4-1BB(CD137)、CD80、CD86和CD28中的至少一种,优选含有4-1BB,4-1BB的氨基酸序列如SEQ IDNO.3所示。胞内活化区域含有例如可以但不限于FcγRI或CD3ζ,优选含有CD3ζ, CD3ζ的氨基酸序列优选如SEQ ID NO.4所示。
跨膜区通常是由免疫球蛋白超家族组成,跨膜区含有但不限于CD8或 CD28,优选含有CD8,例如含有CD8α,CD8α的氨基酸序列如SEQ ID NO.6 所示。
本发明提供的嵌合性抗原受体除了胞外结构域、跨膜区和胞内结构域外,还可以含有其他功能的结构域,包括但不限于用于铰链区,或用于标记所述嵌合性抗原受体的结构域,例如报告基团等,本发明对此不作限制。
在一些优选的实施方式中,所述嵌合性抗原受体用于表达于巨噬细胞,所述嵌合性抗原受体的胞外结构域含有靶向间皮质素的scFv (Mesothelin-scFv),以赋予巨噬细胞抗原依赖性吞噬作用;跨膜区域含有 CD8α;胞内结构域含有共刺激分子4-1BB,以用于活化巨噬细胞;胞内活化区域含有用于增强巨噬细胞抗肿瘤效果的CD3ζ和用于促使巨噬细胞M1型(抗肿瘤)极化IFN-γ受体。所述嵌合性抗原受体的氨基酸序列优选如 SEQ ID NO.1所示。所述嵌合性抗原受体的氨基酸序列也可以为与SEQ ID NO.1所示序列至少90%以上同源性,同时使所述嵌合性抗原受体和与SEQ ID NO.1所示序列编码的嵌合性抗原受体具有相同功能的氨基酸序列。
根据本发明的另一个方面,本发明还提供了一种调节巨噬细胞极化的方法,所述方法包括使巨噬细胞表达所述嵌合性抗原受体。由于本发明提供的嵌合性抗原受体胞内结构域中含有IFN-γR,能够增强表达该嵌合性抗原受体的巨噬细胞对IFN-γ的响应,从而增强巨噬细胞M1型活性,促进巨噬细胞向M1型极化。
根据本发明的另一个方面,本发明还提供了表达上述嵌合性抗原受体的巨噬细胞。表达上述嵌合性抗原受体的巨噬细胞对IFN-γ诱导其转变为 M1型的效率更高,继而这种维持在M1型的巨噬细胞与肿瘤抗原结合后,将刺激T细胞活性,活化的T细胞可分泌更多IFN-γ,诱导巨噬细胞转变为 M1型。其次,当含有IFN-γR的嵌合性抗原受体巨噬细胞与肿瘤抗原结合后,同样会诱导巨噬细胞发挥M1型巨噬细胞功能。使其在最大程度上发挥吞噬肿瘤及抗原提呈功能,从而充分调节肿瘤微环境,最终改变肿瘤微环境的免疫抑制状态。
在一些优选的实施方式中,所述巨噬细胞表达的嵌合性抗原受体的结构如下:胞外结构域含有靶向间皮质素的scFv;跨膜区域含有CD8α;胞内结构域含有共刺激分子4-1BB;胞内活化区域含有CD3ζ和IFN-γ受体。其氨基酸序列优选为如SEQ ID NO.1所示序列,或与SEQ ID NO.1所示序列至少具有90%以上同源性,并且具有相同功能的氨基酸序列。
在一些优选的实施方式中,巨噬细胞包括人诱导多能干细胞(hiPSC) 诱导分化来源的巨噬细胞,或人外周血单核细胞(PBMC)分化得到的巨噬细胞。
根据本发明的另一个方面,本发明还提供了编码所述嵌合性抗原受体的核酸。本发明所述的“核酸”指的是任何长度的核苷酸的聚合物形式,核苷酸包括核糖核苷酸和/或脱氧核糖核苷酸。核酸的实例包括但不限于单链、双链或多链DNA或RNA、基因组DNA、cDNA、DNA-RNA杂合体或者包含嘌呤和嘧啶碱基或其他天然、化学或生化修饰、非天然或衍生的核苷酸碱基的聚合物。所述核酸中除去含有用于编码所述嵌合性抗原受体的区域外,还可以含有其他功能单元,包括但不限于启动子、终止子、增强子、酶切位点、编码标记区域的核酸序列,例如编码荧光蛋白或抗性基因的单元;以及,编码载体的区域等。
根据本发明的另一个方面,本发明还提供了所述嵌合性抗原受体,调节巨噬细胞极化的方法,表达所述嵌合性抗原受体的巨噬细胞或所述核酸在制备用于治疗肿瘤产品中的应用。所述应用可以用于制备对肿瘤具有治疗作用的产品,例如用于制备含有表达所述嵌合性抗原受体的试剂或试剂盒;也可以用于制备用于治疗肿瘤的中间产品,例如用于制备含有编码所述嵌合性抗原受体核酸的载体,例如质粒或慢病毒,或用于增殖上述载体的重组微生物或细胞系等。
根据本发明的另一个方面,本发明还提供了用于治疗肿瘤的产品,所述用于治疗肿瘤的产品例如可以为但不限于为:含有表达所述嵌合性抗原受体的巨噬细胞的试剂或试剂盒;表达所述嵌合性抗原受体的细胞,或用于复制编码所述嵌合性抗原受体核酸的重组微生物或重组细胞系;含有编码所述嵌合性抗原受体的载体,例如质粒载体或慢病毒载体等。
本发明所述的在制备用于治疗肿瘤产品中的应用,或用于治疗肿瘤的产品中所述的“肿瘤”,包括但不限于急性淋巴母细胞性白血病、急性骨髓源性白血病、胆管癌、乳腺癌、子宫颈癌、慢性淋巴细胞性白血病、慢性骨髓源性白血病、结肠直肠癌、子宫内膜癌、食道癌、胃癌、头颈部癌、霍奇金氏淋巴瘤、肺癌、甲状腺髓样癌、非霍奇金氏淋巴瘤、多发性骨髓瘤、肾癌、卵巢癌、胰腺癌、神经胶质瘤、黑素瘤、肝癌、前列腺癌和尿路膀胱癌等。在不同肿瘤中的应用,可以通过调整所述嵌合性抗原受体中胞外结构域中的用于结合抗原区域靶向的抗原实现,预期任何类型的肿瘤和任何类型的肿瘤抗原都可被靶向。需要说明的是,本领域技术人员应该认识到实际上任何类型的肿瘤相关抗原都是已知的。
下面结合优选实施例和对比例进一步说明本发明的技术方案和有益效果。
下述中使用的巨噬细胞的制备方法如下:
(1)人诱导多能干细胞分化而来的巨噬细胞:hiPSC分化为巨噬细胞过程依次经过拟胚体(EB)的形成、造血干祖细胞、髓系祖细胞、单核细胞最终形成单核巨噬细胞。在EB向造血干细胞分化过程中加入:BMP4、 bFGF、VEGF三种生长因子刺激诱导分化;造血干细胞向髓系分化过程加入:IL-3、M-CSF、GM-CSF三种细胞因子;髓系分化为单核巨噬细胞加入 GM-CSF细胞因子,最终获得成熟的巨噬细胞,如图1所示。
(2)人外周单核细胞(PBMC)诱导分化得到的巨噬细胞:利用密度梯度离心法获得外周血单核细胞,每2×106个PBMC用含终浓度100ng/mL 的GM-CSF的10%胎牛血清的RPMI1640重悬后进行培养,隔天换液一次,培养5-7天后,获得贴壁的巨噬细胞,如图2所示。
实施例1
(一)构建靶向间皮素抗原的嵌合性抗原受体:
将包含靶向间皮素单链抗体片段Mesothelin-scFv,CD8跨膜区,4-1BB 共刺激分子,CD3ζ以及IFN-γ受体的全部序列,整合至慢病毒载体质粒。重组慢病毒载体质粒包括EF-1α启动子序列、Mesothelin单链抗体(scFv)、 CD8跨膜区、4-1BB共刺激分子区域、CD3活化区域、IFN-γ受体序列。
第三代慢病毒质粒包括氨苄青霉素抗性基因AmpR序列、原核复制子 pUC Ori序列、PGK启动子、慢病毒5'LTR、慢病毒3'LTR、RRE顺式元件、cPPT顺式元件、eWPRE增强型土拨鼠乙肝病毒转录后调控元件。
(二)构建表达
利用慢病毒包装质粒,将外源基因即嵌合性抗原受体序列整合至慢病毒载体。利用293T细胞过表达整合后的慢病毒,收集病毒,进一步感染巨噬细胞,后通过嘌呤霉素筛选阳性细胞,建立稳定转染的细胞株,得到表达嵌合性抗原受体的巨噬细胞。利用Mesothelin流式抗体标记巨噬细胞,通过流式细胞术检测嵌合性抗原受体的巨噬细胞表达情况。
对比例1
本对比例与实施例1的区别在于,嵌合性抗原受体中不含有IFN-γR,氨基酸序列如SEQ ID NO.7所示。
效果例
利用IFN-γ刺激上述实施例和对比例两种嵌合性抗原受体巨噬细胞,比较二者经IFN-γ刺激后,巨噬细胞向M1型极化变化状态。
将IFN-γ刺激后的两种巨噬细胞与肿瘤细胞(卵巢癌细胞系HO8910、 OVCAR3)共培养,效靶比1:1、3:1、5:1,使用CD68/CD11b或iNOS标记巨噬细胞,间皮素(Mesothelin)标记肿瘤细胞,利用qPCR和ELISA检测巨噬细胞相关促炎因子和趋化因子分泌及表达情况。
①将两种嵌合性抗原受体,即T-CAR(不包含IFN-γR)和IFN-γ R-T-CAR病毒(病毒滴度为6×108TU/mL)转染至两种来源的巨噬细胞 (hiPSC诱导分化的巨噬细胞和PBMC来源的原代巨噬细胞),转染巨噬细胞(细胞数量为2×106个细胞/mL),由于病毒质粒含有GFP绿色荧光蛋白,转染后在48-96小时镜下观察绿色荧光在巨噬细胞表面的表达情况,再通过嘌呤霉素药物筛选获得成功表达CAR分子的巨噬细胞。利用IFN-γ(浓度为100ng/ml)刺激巨噬细胞(1×106个细胞/mL)24小时后,qPCR结果显示,与无IFN-γR的巨噬细胞相比,含有IFN-γR的巨噬细胞对IFN-γ刺激后M1型marker,如趋化因子CCL2,CCR7,CCL8和CXCL9表达显著升高。
实验结果如图3和图4所示。图3所示:以不表达CAR的巨噬细胞作为空白对照组(control组),T-CAR(不包含IFN-γR)巨噬细胞与IFN-γ R-T-CAR巨噬细胞对IFN-γ的刺激响应均有不同程度的提高,其中IFN-γ R-T-CAR巨噬细胞在接收IFN-γ的刺激后M1型marker显著升高(p< 0.05),图4所示:仅比较T-CAR(不包含IFN-γR)巨噬细胞与IFN-γR-T-CAR巨噬细胞对IFN-γ的刺激响应程度,结果显示,与T-CAR(不包含IFN-γR)巨噬细胞相比,IFN-γR-T-CAR巨噬细胞在接收IFN-γ刺激后,M1型巨噬细胞标志抗原表达升高。
②两种嵌合性抗原受体的巨噬细胞经IFN-γ诱导后,体外与肿瘤细胞 (卵巢癌细胞系HO8910)共培养48小时,利用qPCR进行检测,结果显示,与无IFN-γR的巨噬细胞相比,含有IFN-γR的巨噬细胞经IFN-γ刺激再与肿瘤细胞共培养后,检测M1型促炎因子IL-6,IL-12,IL-23和IL-1β表达及分泌情况。实验结果如图5和图6所示:图5所示:以不表达CAR的巨噬细胞作为空白对照组(control组),T-CAR(不包含IFN-γR)巨噬细胞与IFN-γR-T-CAR巨噬细胞经IFN-γ刺激后,再与肿瘤细胞接触48 小数后,IFN-γR-T-CAR巨噬细胞M1型促炎因子表达显著升高(p<0.05),图6所示:仅比较T-CAR(不包含IFN-γR)巨噬细胞与IFN-γR-T-CAR 巨噬细胞经IFN-γ刺激后,再与肿瘤细胞接触48小数后,结果显示,与 T-CAR(不包含IFN-γR)巨噬细胞相比,IFN-γR-T-CAR巨噬细胞M1型促炎因子表达显著升高。此结果说明,两种嵌合性抗原受体在接收IFN-γ刺激后,由于所表现出M1型巨噬细胞表型有所不同,继而与肿瘤细胞共培养后具有更显著的促炎功能(体现在M1型促炎因子表达升高)。
③将两种巨噬细胞与肿瘤细胞(卵巢癌细胞系HO8910)共培养,观察肿瘤抗原激活嵌合性抗原受体后,巨噬细胞极化状态的改变。结果显示,与无IFN-γR的巨噬细胞相比,含有IFN-γR的巨噬细胞与肿瘤细胞共培养后,M1型促炎因子表达显著升高。实验结果如图7和图8所示。图7所示:以不表达CAR的巨噬细胞作为空白对照组(control组),T-CAR(不包含IFN-γR)巨噬细胞与IFN-γR-T-CAR巨噬细胞对肿瘤抗原刺激所发生的极化均有不同程度的响应,其中IFN-γR-T-CAR巨噬细胞在接收肿瘤抗原的刺激后M1型marker IL-6显著升高(p<0.05),图8所示:仅比较T-CAR (不包含IFN-γR)巨噬细胞与IFN-γR-T-CAR巨噬细胞对肿瘤抗原的刺激极化变化程度,结果显示,与T-CAR(不包含IFN-γR)巨噬细胞相比,IFN-γR-T-CAR巨噬细胞在接收肿瘤抗原刺激后,M1型促炎因子IL-6、 IL-12表达升高。
最后应说明的是:以上各实施例仅用以说明本发明的技术方案,而非对其限制;尽管参照前述各实施例对本发明进行了详细的说明,本领域的普通技术人员应当理解:其依然可以对前述各实施例所记载的技术方案进行修改,或者对其中部分或者全部技术特征进行等同替换;而这些修改或者替换,并不使相应技术方案的本质脱离本发明各实施例技术方案的范围。

Claims (10)
1.一种嵌合性抗原受体,其特征在于,所述嵌合性抗原受体含有IFN-γ受体,所述IFN-γ受体位于所述嵌合性抗原受体的胞内结构域。
2.根据权利要求1所述的嵌合性抗原受体,其特征在于,所述嵌合性抗原受体的胞内结构域含有共刺激结构域和胞内活化区域,所述IFN-γ受体位于胞内活化区域;
可选地,所述IFN-γ受体的氨基酸序列如SEQ ID NO.2所示;
可选地,所述共刺激结构域含有4-1BB、CD80、CD86和CD28中的至少一种,优选含有4-1BB,4-1BB的氨基酸序列如SEQ ID NO.3所示;
可选地,所述胞内活化区域还含有FcγRI或CD3ζ,优选含有CD3ζ,CD3ζ的氨基酸序列优选如SEQ ID NO.4所示。
3.根据权利要求1所述的嵌合性抗原受体,其特征在于,所述嵌合性抗原受体的胞外结构域含有scFv、Fab、scFab、或scIgG抗体片段;
可选地,所述胞外结构域含有scFv;
可选地,所述scFv靶向间皮质素;
可选地,靶向间皮质素的scFv的氨基酸序列如SEQ ID NO.5所示。
4.根据权利要求1所述的嵌合性抗原受体,其特征在于,所述嵌合性抗原受体的跨膜区域含有CD8或CD28,优选含有CD8,例如含有CD8α;
可选地,CD8α的氨基酸序列如SEQ ID NO.6所示。
5.根据权利要求1-4任一项所述的嵌合性抗原受体,其特征在于,所述嵌合性抗原受体的胞外结构域含有靶向间皮质素的scFv;跨膜区域含有CD8α;胞内结构域含有共刺激分子4-1BB,CD3ζ和IFN-γ受体;
可选地,所述嵌合性抗原受体的氨基酸序列如SEQ ID NO.1所示,或,如与SEQ ID NO.1所示序列至少90%以上同源性的序列所示。
6.一种调节巨噬细胞极化的方法,其特征在于,包括使巨噬细胞表达权利要求1-5任一项所述嵌合性抗原受体。
7.表达权利要求1-5任一项所述嵌合性抗原受体的巨噬细胞;
可选地,所述嵌合性抗原受体的胞外结构域含有靶向间皮质素的scFv,跨膜区域含有CD8α,胞内结构域含有共刺激分子4-1BB,CD3和IFN-γ受体;
可选地,所述巨噬细胞表达的嵌合性抗原受体如SEQ ID NO.1所示,或,如与SEQ IDNO.1所示序列至少90%以上同源性的序列所示;
可选地,所述巨噬细胞包括人诱导多能干细胞诱导分化来源的巨噬细胞,或人外周血单核细胞分化得到的巨噬细胞。
8.编码权利要求1-5任一项所述嵌合性抗原受体的核酸。
9.权利要求1-5任一项所述嵌合性抗原受体,权利要求6所述的调节巨噬细胞极化的方法,权利要求7所述的表达所述嵌合性抗原受体的巨噬细胞,或权利要求8所述的核酸在制备用于治疗肿瘤产品中的应用。
10.用于治疗肿瘤的产品,包括权利要求1-5任一项所述嵌合性抗原受体,权利要求7所述的巨噬细胞或权利要求8所述的核酸。
