Abstract
本发明的目的在于提供脂质运载蛋白型前列腺素D2合成酶(L‑PGDS)产生促进剂、更具体而言提供周细胞或从周细胞脱分化得到的缺血诱导性多能性干细胞(iSC)中的L‑PGDS产生促进剂。本发明中,确认了痘苗病毒接种炎症组织提取物中含有具有L‑PGDS产生促进作用的物质。L‑PGDS产生促进剂作为可以期待由L‑PGDS表达促进所带来的效果的疾病、即、脑梗塞等的脑血管障碍、阿尔茨海默氏病等的认知症或睡眠障碍的预防、治疗或复发预防药物等,有用性高。
Description
技术领域
本发明涉及脂质运载蛋白(lipocalin)型前列腺素D2合成酶(以下,有时记作“L-PGDS”。另外,被称为“脂质运载蛋白型前列腺素D合成酶”的酶也可以包括在本发明中。)的产生促进剂等。
背景技术
前列腺素D2合成酶(PGDS)已知有分布于中枢神经系统、雄性生殖器官、心脏等处的脂质运载蛋白型(L-)PGDS、和分布于肥大细胞和Th2细胞的造血型(H-)PGDS的2种。L-PGDS具有催化从作为前列腺素生物合成的共通中间体的前列腺素H2(PGH2)向前列腺素D2(PGD2)的异构化反应的活性。另一方面,L-PGDS在结构上属于发挥作为脂溶性物质的载体的功能的脂质运载蛋白家族。因此,L-PGDS为具有PGD2生物合成酶和脂溶性载体的两面性的多功能蛋白。
分布于中枢神经系统的L-PGDS在脑脊髓液中也有检出,与其他的脂质运载蛋白相比较,具有巨大的亲油性袋,因此被认为发挥作为脑内的各种脂溶性分子的输送蛋白质和清除剂的作用。作为那样的L-PGDS的脂质运载蛋白的功能的例子,可以列举在蛛网膜下腔出血后,脑内L-PGDS上升,结合成为神经毒性的原因的胆红素,由此避免神经毒性(InuiT.et al.,J.Cereb.Blood Flow Metab.34,1558-1567,2014);L-PGDS在阿尔茨海默氏病的患者或动物模型中,与淀粉样蛋白β蛋白质(Aβ、老人斑)的低聚物形成所必须的区域牢固地结合,抑制脑脊髓液中的Aβ凝集形成和细胞毒性(Kanekiyo T.et al.,Proc.Natl.Acad.Sci.USA 104,6412-6417,2007)等。
另外,存在如下报道:小鼠的缺血脑中PGD2的产生显著增加,通过L-PGDS和H-PGDS、或PGD2受体DP1的敲除,而缺血后的脑水肿发病重症化(Tanigichi H.et al.,J.Neurosci.27,4303-4312,2007);通过L-PGDS的敲除,而脑梗塞灶和脑水肿增大(SaleemS.et al.,Neuroscience 160,248-254,2009)。由此可以认为在脑缺血时由L-PGDS或H-PGDS产生的PGD2通过由其受体介导的作用而发挥脑保护的作用。
此外,报道了,半乳糖基神经酰胺酶缺损引起的脱髓鞘疾病克拉伯病的模型小鼠(Twitcher小鼠)中,在对脱髓鞘显示抗性的轴突区被观察到L-PGDS基因表达的上升,如果进一步导入L-PGDS的缺损,则会发生少突胶质细胞的消失引起的脱髓鞘的重症化(TaniikeM.et al.,J.Neurosci.22,4885-4896,2002)。另外报道了,在L-PGDS缺损小鼠中,诱导末梢神经的脱髓鞘,发现作为雪旺氏细胞的PGD2受体的Gpr44对神经髓鞘化是必须的(TrimarcoA.et al.,Nature Neurosci.17,1682-1992,2014)。由这些发现可以认为,少突胶质细胞(中枢神经)或雪旺氏细胞(末梢神经)中的神经轴索髓鞘化及其维持中,L-PGDS及其产物的PGD2是必须的。
此外,还报道了,关于通过L-PGDS的保护作用,不限于神经细胞,作为PGD2的代谢产物的15-脱氧-前列腺素J2也可以避免过氧化应激引起的胃肠道神经胶质细胞的细胞死亡(Abdo H.et al.,J.Physiol.590,2739-2750,2012)。
另外,自古以来已知由L-PGDS合成的脑内PGD2具有睡眠调节作用(Ueno R.etal.,Biochem.Biophys.Res.Commun.109,576-582,1982)。关于其机理,由日本的研究者进行了详细研究,其结果被认为是由于PGD2经由眼底部蛛网膜DP1受体而分泌腺苷,从而刺激睡眠中枢的缘故。L-PGDS对作为酶产物的PGD2具有高的亲和性,因此被推测确保脑脊髓液中的PGD2的稳定性并且将PGD2向附近的受体输送(Urade Y.and Hayaishi O.,Biochim.Biophys.Acta 1482,259-271,2000),被认为极大参与经由PGD2的睡眠调节。
如上所述,L-PGDS被认为是作为合成PGD2的酶蛋白质发挥作用,并且发挥脑内的各种疏水性低分子的结合、输送和作为清除剂的作用,具有脑内环境调整、脑保护功能和睡眠调节功能等各种功能的蛋白质。因此可以认为,促进L-PGDS的产生使得这些功能有效地发挥,对于L-PGDS相关的疾病的预防、治疗等有用。然而,目前为止,还没有在生物体内促进L-PGDS的产生的物质的报道。
虽然在本发明中,确认了痘苗病毒接种炎症组织提取物(本提取物)具有优异的L-PGDS产生促进作用,但关于本提取物或含有本提取物的制剂已知有非常多的作用、效果。例如作为本提取物对脑的作用、效果,已知有对脑梗塞等缺血性疾病的治疗效果(日本特开2000-16942号公报)和BDNF等的神经营养因子的产生促进作用(国际公开WO2011/162317号公报),但本提取物具有L-PGDS的产生促进作用目前为止是未知的。
发明内容
发明要解决的技术问题
本发明提供促进具有脑保护作用、睡眠作用等的L-PGDS的产生的物质、以及以L-PGDS产生促进作用为指标的具有脑保护作用、睡眠作用等的药剂的筛选体系等。另外,提供含有该物质作为有效成分的、在脑梗塞等的脑血管障碍、阿尔茨海默氏病等的认知症、失眠症等的L-PGDS参与的疾病的预防、治疗或复发预防中,有效且安全性高的医药品。此外,本发明涉及的L-PGDS产生促进剂具有抑制、缓和脑血管障碍时等的脑的缺血障碍、神经细胞损伤等的作用。此外,本发明中使用的“治疗”包括“减轻”、“改善”、“进展抑制”等的意思。另外,本发明中有时将具有睡眠作用的药剂记作“安眠药”,但被称为“睡眠剂”、“睡眠改善药”、“睡眠导入剂”、“催眠药”的物质也可以包括在本发明中。
用于解决技术问题的技术方案
本发明的发明人由利用小鼠脑梗塞模型(大脑中动脉永久性闭塞模型)的研究,在通过血流的阻断而成熟神经细胞不断死亡的梗塞区域内,发现了能够分化成包括神经细胞、星形胶质细胞、少突胶质细胞等的神经系统细胞在内的各种细胞的干细胞,将其命名为缺血诱导性多能性干细胞(ischemia-induced multipotent stem cells,iSCs)。通过在iSC中强表达的神经干细胞标志物巢蛋白(nestin)在组织化学上分布于脑软膜至大脑皮质的血管周围;另外,iSC表达PDGFRβ(血小板源性生长因子受体β)、NG2(神经胶质抗原2,neuron-glial antigen 2)这样的血管周细胞(周细胞,pericyte)标志物等,因此可以认为iSC的来源是分布于血管周围的周细胞。周细胞与神经细胞、星形胶质细胞、血管内皮细胞一起,构成作为脑的功能性的器质性基本单元的神经血管单元(neurovascular unit,NVU),被认为发挥参与血脑屏障的形成、维持和神经功能的调节、脑内排出系统(glymphatic system)等的重要作用。表示iSC在缺血等的脑损伤时周细胞被重编程而产生,由于血管内皮细胞等所形成的微小环境而分化成神经细胞。由此可以认为iSC是中风后的血流再建后的神经修复中发挥主要作用的干细胞。
本发明的发明人作为使用iSC的研究的一环,进行了痘苗病毒接种家兔炎症皮肤提取物(本提取物)对iSC的作用的研究。而且,在iSC中添加本提取物培养后,进行了利用约2万8千个基因的DNA芯片的全面的基因表达解析,结果发现本提取物具有选择性表达促进编码L-PGDS的基因PTGDS的作用。另外,在蛋白质水平上,也确认了本提取物促进L-PGDS的产生。此外,通过免疫组织化学方法研究的结果,缺血脑中表达的L-PGDS显示与周细胞标志物共分布。由此可以认为L-PGDS来自于周细胞或从周细胞脱分化的iSC。
如上所述,L-PGDS被认为是发挥作为合成PGD2的酶蛋白质的作用、并且主要在脑内表达、且发挥各种疏水性低分子的结合、输送和作为清除剂的作用、并具有脑内环境调整、脑保护功能和睡眠调节功能等各种功能的蛋白质。由此可以认为L-PGDS产生促进剂作为脑梗塞等的脑血管障碍、阿尔茨海默氏病等的认知症、失眠症等的L-PGDS所参与的疾病的预防、治疗或复发预防药物有用。本提取物被确认具有优异的L-PGDS产生促进作用,使阿尔茨海默型认知症模型小鼠的脑内的L-PGDS量增加,并且使Aβ量减少,进而改善认知功能。因此,本提取物或本提取物中所含有的具有L-PGDS产生促进作用的物质、或含有本提取物的制剂作为L-PGDS产生促进剂非常有用。
另外,本发明的发明人发现以iSC的L-PGDS产生促进作用为指标,作为对L-PGDS所参与的疾病的治疗有用的药剂的筛选方法有用。该筛选方法被认为特别有助于具有脑保护作用或睡眠作用的药剂的开发。另外,本发明提供以iSC的L-PGDS产生促进作用为指标、对本提取物或含有本提取物的制剂进行试验,由此判定或评价本提取物或含有本提取物的制剂的作用、效果,进而确保本提取物或含有本提取物的制剂的药效的方法。本发明的发明人基于以上的发现,完成了本发明。
即,本发明包括例如下述的方案。

发明的效果
本发明L-PGDS产生促进剂通过增强周细胞或iSC表达的L-PGDS的作为脂质运载蛋白的功能,发挥作为排出脑内的各种疏水性分子的转运蛋白的作用,强烈期待发挥保护脑不受被认为是脑缺血时的损伤或认知症的原因的物质的损害的作用。另外,本发明L-PGDS产生促进剂在脑脊髓液中分泌在细胞内合成的PGD2,通过向脑内的PGD2受体输送,被期待发挥睡眠调节作用等。特别是得知了本提取物在小鼠的缺血脑中促进L-PGDS的产生,此外,确认了通过将本提取物向阿尔茨海默型认知症模型小鼠投予,脑内的L-PGDS量上升,并且Aβ量减少,认知功能改善。这样,本提取物促进L-PGDS的产生并具有优异的药理作用在动物实验中也得到了确认。另外,含有本提取物的制剂作为副作用等问题少的安全性高的药剂被长年使用。因此,本发明的有用性极高。
另外,本提取物是包含数量非常庞大的成分的多成分体系的物质,所以通过单一或多个含有成分的含量等判定或评价其作用、确保药效等极为困难。该点,根据以本发明的L-PGDS的表达促进作用作为指标的本提取物或含有本提取物的制剂的判定或评价方法,能够简便地判定或评价本提取物或含有本提取物的制剂的作用,进而能够用于确保本提取物或含有本提取物的制剂的药效。从这一方面而言,本发明的有用性也高。此外,本发明中的“判定或评价”包括通过试验、检查等调查对象物来确定效果、作用、是否适用等的全部概念。

具体实施方式
本提取物是含有从接种痘苗病毒并发痘的动物的发炎组织提取分离的非蛋白性的活性物质的提取物。本提取物在提取状态下是液体,但也能够通过干燥制成固体。本制剂作为医药品是非常有用的。作为本制剂,在申请人在日本制造并销售的具体的商品中,有“含有接种痘苗病毒的家兔的炎症皮肤的提取液的制剂”(商品名:神经妥乐平/NEUROTROPIN〔注册商标〕)(以下称为“神经妥乐平”。)。神经妥乐平中有注射剂和片剂,都为医疗用医药品(ethical drug)。
神经妥乐平的注射剂的适应症为“腰痛症、颈肩腕综合症、症状性神经痛、伴随皮肤疾病(湿疹、皮炎、荨麻疹)的瘙痒、过敏性鼻炎、亚急性脊髓视神经病(SMON)后遗症状的寒冷感、感觉异常、疼痛”。神经妥乐平的片剂的适应症为“带状疱疹后遗神经痛、腰痛症、颈肩腕综合症、肩周炎、变形性关节症”。本制剂是申请人创制并作为医药品开发出来的,其有效性和安全性上的优异特长受到赞赏,已经销售多年,并已经在日本医药品市场中确立了稳固的地位。
本发明中的痘苗病毒接种炎症组织提取物能够通过如下操作得到:接种痘苗病毒,将发痘的炎症组织破碎,加入提取溶剂,除去组织碎片后,进行除蛋白处理,使其吸附于吸附剂,然后将有效成分洗脱。即,例如为如下的工序。
(A)收集接种痘苗病毒并使其发痘的兔、小鼠等的皮肤组织等,将发痘组织破碎,加入水、苯酚水、生理盐水或苯酚加甘油水等提取溶剂后,通过过滤或离心分离,得到提取液(滤液或上清液)。
(B)将上述提取液调整为酸性的pH并加热,进行除蛋白处理。接着,将除蛋白后的溶液调整为碱性并加热后,进行过滤或离心分离。
(C)使所得到的滤液或上清液成为酸性,使其吸附于活性炭、高岭土等吸附剂。
(D)在上述吸附剂中加入水等提取溶剂,调整为碱性的pH,将吸附成分洗脱,由此能够得到痘苗病毒接种炎症组织提取物。然后,根据希望,适当将洗脱液在减压下蒸发干燥固化或冻结干燥,由此也能够得到干燥固化物。
作为用于接种痘苗病毒得到炎症组织的动物,能够使用兔、牛、马、绵羊、山羊、猴、大鼠、小鼠等感染痘苗病毒的各种动物,作为炎症组织,优选兔的炎症皮肤组织。兔只要是属于兔目即可,可以使用任何兔子。作为例子,有:穴兔(Oryctolagus cuniculus)、家兔(驯养的穴兔)、野兔(日本野兔)、鼠兔、雪兔等。其中,适合使用家兔。在日本,有从过去被饲养的作为家畜或实验用动物而频繁使用的称为家养兔(イエウサギ)的兔,这也是家兔的别称。在家兔中存在多个品种(breed),能够优选使用称为日本白色种和新西兰白色种(新西兰白)的品种。
痘苗病毒(vaccinia virus)可以是任意株的痘苗病毒。作为例子,可以列举李斯特(Lister)株、大连(Dairen)株、池田(Ikeda)株、EM-63株、纽约市公众卫生局(New YorkCity Board of Health)株等。
关于上述的本提取物的基本的提取工序(A)~(D),更详细而言,例如,能够作为如下的工序实施。
关于工序(A)
在兔的皮肤中将痘苗病毒进行皮内接种使其发痘,收集炎症皮肤组织。收集的皮肤组织以苯酚溶液等进行清洗、消毒。将该炎症皮肤组织破碎,加入其1至5倍量的提取溶剂。其中,所谓破碎是指使用绞碎机等细细地破碎为肉末状。另外,作为提取溶剂,能够使用蒸馏水、生理盐水、弱酸性至弱碱性的缓冲液等,也可以适当添加苯酚等的杀菌-防腐剂、甘油等的稳定剂、氯化钠、氯化钾、氯化镁等的盐类等。此时,也能够通过冷冻融化、超声波、细胞膜溶解酶或表面活性剂等的处理来破坏细胞组织,使提取变得容易。将得到的悬浮液放置5日至12日。在该期间,可以边适当搅拌或不搅拌边加温到30至45℃。通过对得到的液体进行固液分离(过滤或离心分离等)除去组织碎片,得到粗提取液(滤液或上清液)。
关于工序(B)
对在工序(A)中得到的粗提取液进行除蛋白处理。除蛋白能够由通常进行的公知方法实施,能够应用加热处理、利用蛋白质变性剂(例如酸、碱、脲、胍、丙酮等有机溶剂等)的处理、等电点沉淀、盐析等的方法。接着,利用除去不溶物的通常方法,例如使用滤纸(纤维素、硝酸纤维素等)、玻璃过滤器、硅藻土、塞茨过滤板(Seitz filter)等的过滤、超滤、离心分离等,得到除去了析出的不溶蛋白质的滤液或上清液。
关于工序(C)
将在工序(B)中得到的滤液或上清液调整为酸性、优选调整为pH3.5至5.5,进行向吸附剂的吸附操作。作为能够使用的吸附剂,能够列举活性炭、高岭土等,在提取液中添加吸附剂进行搅拌,或使提取液通过吸附剂填充柱,能够使有效成分吸附于该吸附剂。在提取液中添加吸附剂时,通过过滤或离心分离等除去溶液,能够得到吸附有活性成分的吸附剂。
关于工序(D)
为了使活性成分从工序(C)中得到的吸附剂中洗脱(脱离),在该吸附剂中加入洗脱溶剂,调整为碱性、优选调整为pH9至12,在室温或适当加热、或者搅拌进行洗脱,以过滤或离心分离等通常的方法除去吸附剂。作为所使用的洗脱溶剂,能够使用碱性溶剂,例如,能够使用调整为碱性pH的水、甲醇、乙醇、异丙醇等或它们的适当的混合溶液,能够优选使用调整为pH9至12的水。洗脱溶剂的量能够适当设定。为了将这样操作得到的洗脱液作为原药使用,能够将pH适当地调整到中性附近等,最终得到接种痘苗病毒的兔炎症皮肤提取物(本提取物)。
本提取物在制成时为液体,因此也能够通过适当浓缩、稀释制成所希望浓度的提取物。从本提取物制造制剂时,优选实施加热灭菌处理。为了制成注射剂,例如,能够加入氯化钠等配制与生理盐水等渗的溶液。另外,也能够通过以液体或凝胶等的状态口服给药,也能够通过对本提取物进行适当的浓缩干燥固化等的操作,制造片剂等的口服用固体制剂。从本提取物制造这样的口服用固体制剂的具体方法记载于日本专利第3818657号和日本专利第4883798号的说明书中。这样操作得到的注射剂或口服用制剂等是本制剂的例子。
作为向患者的给药方法,没有特别限定,可以根据治疗目的适当选择。例如除了口服给药以外,还可以列举皮下、肌肉内、静脉内给药、经皮给药等,给药量可以根据痘苗病毒接种炎症组织提取物的种类适当设定。关于市售的制剂中确认的给药量,基本上以内服时为1天16NU,注射剂时1天投予3.6至7.2NU的方式作为医疗用医药品进行了表示,但可以根据疾病的种类、重症度、患者的个人差异、给药方法、给药期间等适当增减(NU:神经妥乐平单位。神经妥乐平单位是使用疼痛阈值比正常动物低的慢性应激动物的SART应激小鼠,根据炎症足加压(Randall-Selitto)改良法进行试验,以镇痛效力的ED50值进行规定的。1NU为表示ED50值为100mg/kg时的神经妥乐平制剂的含有镇痛活性的成分1mg的活性。)。
以下,表示本提取物的制造方法的例子、以及本提取物的关于新型的药理作用、L-PGDS产生促进作用的药理试验结果,但本发明不受这些实施例的记载的任何限制。
实施例
实施例1本提取物的制造
在健康的成熟家兔的皮肤中皮内接种痘苗病毒,切下并采集发痘的皮肤。将采集的皮肤以苯酚溶液进行清洗、消毒后,除去多余的苯酚溶液,进行破碎,加入苯酚溶液混合,放置3~7日后,进而边搅拌3~4天,边加温至35~40℃。然后,利用盐酸将固液分离得到的提取液调整为pH4.5~5.2,以90~100℃加热处理30分钟后,过滤除去蛋白。再利用氢氧化钠将滤液调整为pH9.0~9.5,以90~100℃加热处理15分钟后,进行固液分离。
利用盐酸将所得到的除蛋白液调整为pH4.0~4.3,加入除蛋白液质量的2%量的活性炭,搅拌2小时后,进行固液分离。在采集的活性炭中加水,利用氢氧化钠调整为pH9.5~10,以60℃搅拌90~100分钟后,进行离心分离得到上清液。向通过离心分离而沉淀的活性炭再加入水,之后利用氢氧化钠调整为pH10.5~11,以60℃搅拌90~100分钟,之后进行离心分离,得到上清液。合并两上清液,利用盐酸中和,得到本提取物。
实施例2药理试验
接着,表示使用上述实施例1中得到的本提取物作为受试物质的关于L-PGDS产生促进作用的药理试验的方法和结果。此外,在下述的药理试验中,关于脑梗塞向C.B-17小鼠的导入、从该脑梗塞灶的iSC的分离培养,根据Nakagomi,T.et al.Eur.J.Neurosci.,29,1842_1852,2009中记载的方法进行。
试验例1:受试物质处理iSC中的综合基因表达解析
对C.B-17小鼠(3只)实施大脑中动脉闭塞造成的缺血负荷,在第3天由梗塞巢分离3株iSC。对于添加了各个培养iSC(2%胎牛血清(FBS)、20ng/mL成纤维细胞增殖因子(FBS)、20ng/mL上皮生长因子(EGF)和1%N2添加剂(supplement)的Dulbecco改良的Eagle培养基F12(2%FBS DMEM/F12 F/E/N)、)添加受试物质(50、1000mNU/mL)或生理盐水(对照),在培养第4天通过RNeasy〔注册商标、以下同样〕Mini Kit(QIAGEN公司)回收总RNA(全部9个培养)。在综合基因表达解析中,使用小鼠用的基因芯片SurePrint G3Mouse GE微阵列8×60K(24,321种RNA和4,576种非编码RNA、GE公司),通过受试物质的添加,筛选3株均在两个浓度下显示相同方向1/2以下的降低或2倍以上的表达变化的基因。结果作为所筛选的各基因相对于对照的表达量比来表示(3株的平均值±标准误差)。将结果的一例表示于表1。

上述解析的结果,如表1所示,筛选到显示表达减少的COCH基因以及显示表达上升的GBP6基因和PTGDS基因的合计3个基因。它们之中,编码L-PGDS的PTGDS基因的表达量对于受试物质显示强的用量依赖性。
试验例2-1:对L-PGDS基因表达的评价(实时RT-PCR法)
与试验例1同样,对培养iSC(不含FBS的DMEM/F12 F/-/-、 )添加受试物质(50、500mNU/mL)或生理盐水(对照)(n=1),培养7天后,通过Isogen II〔注册商标〕按照制造商(Nippon Gene公司)的手册回收RNA。以260nm和280nm的吸光度为指标检验RNA纯度后,在随机引物存在下通过SuperScript〔注册商标、以下同样〕IV RTase(Invitrogen公司)进行逆转录反应,得到对总RNA的单链cDNA。对所得到的cDNA,通过使用PTGDS特异性引物的定量PCR法(Prism〔注册商标〕7900HT、Applied Biosystems公司),以持家基因β-肌动蛋白为标准进行转录产物的定量(阈值再循环比较法)。其中,使用的PTGDS和β-肌动蛋白用引物的序列如下〔PTGDS:5'-gactctgaaggacgagctgaag-3'(序列编号1)、5'-tcttgaatgcacttatccggttgg-3'(序列编号2)、·-肌动蛋白:5'-tacagcttcaccaccacagc-3'(序列编号3)、5'-aaggaaggctggaaaagagc-3'(序列编号4)〕。结果以对照为1时的PTGDS相对于·-肌动蛋白的表达量比来表示。将结果的一例表示于表2和图1。

从表2和图1可知,与试验例1所示的结果同样,确认到受试物质在iSC中用量依赖性地促进L-PGDS基因(PTGDS)的表达。
试验例2-2:对L-PGDS基因表达的评价(经典的RT-PCR法)
在0(对照;生理盐水)、1、5、50、100mNU/mL的各用量的受试物质存在下,用含有FGF的培养基(DMEM/F12 F/-/-)或不含FGF的培养基(DMEM/F12-/-/-)对iSC培养4天。对通过RNeasy Mini Kit(QIAGEN公司)回收得到的总RNA,通过使用SuperScript III One-StepRT-PCR System with Platinum(Invitrogen公司)的经典的RT-PCR法对L-PGDS基因(PTGDS)的表达量进行半定量(35个循环)。将PCR产物用2%琼脂糖凝胶电泳分离,将PTGDS和GAPDH的条带用溴乙锭染色并可视地进行检测。其中,所使用的PTGDS和GAPDH用引物的序列如下〔PTGDS:5'-cctccaactcaagctggttc-3'(序列编号5)、5'-atagttggcctccaccactg-3'(序列编号6)、GAPDH:5'-atcactgccacccagaagac-3'(序列编号7)、5'-cacattgggggtaggaacac-3'(序列编号8)〕。将结果的一例表示于图2。
关于iSC中的L-PGDS基因(PTGDS)表达促进作用,用更细致的受试物质的用量进行调查,结果如图2所示,受试物质与培养基是否含有FGF无关,用量依赖性地使L-PGDS基因的表达量增加。
试验例3-1:对L-PGDS蛋白的表达的评价(蛋白质印迹法)
将iSC在血清非存在下添加受试物质(0、1、5、50、100mNU/mL)培养4天(DMEM/F12F/-/-、)后回收,用磷酸缓冲液清洗后,通过RIPA缓冲液(4℃、50mMTris盐酸缓冲液(pH 7.6)、150mM氯化钠、1%Nonidet〔注册商标〕P-40(NP-40)、0.5%脱氧胆酸钠和0.1%十二烷基硫酸钠)溶解,进行匀浆。调整使得总蛋白量均一后,将匀浆液用SDS-PAGE(BIO-RAD Any kD(商标))分离,转印到聚偏二氟乙烯(PVDF)膜(Immun-Blot PVDF膜、BIO-RAD公司),用Bloking One(nacalai tesque公司)封闭后,通过使用特异性抗体的蛋白质印迹法,检测L-PGDS(抗前列腺素D合酶(脂质运载蛋白)抗体(anti-ProstaglandinD Synthase(Lipocalin)antibody)[EP12357]、Abcam公司、1﹕2000)和β-肌动蛋白(β-肌动蛋白单克隆抗体(Monoclonal anti-β-Actin)[A1978]、Sigma公司、1﹕100000)。检测使用高灵敏度化学发光法(Chemi-Lumi One L,nacalai tesque公司)。将结果的一例表示于图3。
从图3可知,确认到受试物质促进iSC细胞内的L-PGDS蛋白的表达。
试验例3-2:对L-PGDS蛋白的表达的评价(斑点印迹法)
将由受试物质(0、1、10、50、100mNU/mL)培养了4天(DMEM/F12F/-/-、)的iSC的培养上清500μL点样到PVDF膜(Immun-Blot PVDF膜、BIO-RAD公司)上,用Bloking One(nacalai tesque公司)封闭后,通过使用抗L-PGDS抗体(抗前列腺素D合酶(脂质运载蛋白)抗体[EP12357]、Abcam公司、1﹕2000)的斑点印迹法,检测L-PGDS。将结果的一例表示于图4。
L-PGDS在其N末端具有典型的分泌信号序列和信号肽酶识别序列,因此被认为分泌到细胞外。因此,测定添加了受试物质的iSC的培养上清中的L-PGDS蛋白,结果如图4可知,确认了培养上清中(细胞外)的L-PGDS蛋白量也依赖于受试物质的添加用量而增加。
试验例3-3:对L-PGDS蛋白质的表达的评价(ELISA法)
将由受试物质(0〔对照〕、1、10、50、100、1000mNU/mL)处理了4天的iSC的培养(DMEM/F12 F/-/-、)的上清回收。离心分离(1500rpm、10分钟、4℃)后,使用特异性ELISA法(人L-PGDS用ELISA试剂盒(Prostaglandin D Synthase 21kDa(Brain)、型号:SEA724Hu、Cloud-Clone Corp.)按照制造者的手册测定培养上清中的L-PGDS量。将结果的一例表示于表3和图5。

由表3和图5可知,与试验例3-2同样,在由受试物质处理了的iSC的培养上清(细胞外)中,确认到了L-PGDS蛋白量增加。由试验例3的结果可以确认到受试物质在iSC中不仅在基因水平,而且在蛋白水平上都促进L-PGDS的产生,进而确认到使所产生的L-PGDS分泌。
试验例4:对L-PGDS的酶活性的评价(液相色谱(HPLC)质谱法)
L-PGDS是以PGH2作为底物而生物合成PGD2的酶(EC5.3.99.2)。为了确认在iSC中由受试物质促进产生的L-PGDS通过其酶活性进而产生PGD2,对iSC的A:细胞内和B:细胞外,PGD2及其代谢产物、以及与PGD2同样以PGH2作为底物所产生的PGE2等的前列腺素类进行了全面的分析。
A:iSC细胞内酶活性的评价
从添加受试物质(0、10、50mNU/mL)培养3天(不含FBS的DMEM/F12 F/-/-、)的iSC中,通过RIPA缓冲液处理(4℃、20min)制备细胞提取液。通过HPLC质谱法,分别测定细胞内提取液中的前列腺素类(PGD2、PGJ2、15-脱氧-Δ12,14-PGJ2、13,14-二氢-15-酮-PGD2和PGE2)的浓度。HPLC分离使用AQUITY UPLC HSS T3色谱柱(Waters公司),作为检测器和质谱仪分别使用API4000 LC/MS/MS系统和TripleQuadrupole(均为AB Sciex公司)。将结果的一例表示于表4和图6。

如表4和图6所示,PGD2及其非酶代谢产物PGJ2的细胞内水平随着添加受试物质而用量依赖性地增加。进而,在添加50mNU/mL的受试物质时,还确认到了PGJ2的非酶代谢产物的15-脱氧-Δ12,14-PGJ2的产生。此外,15-脱氧-Δ12,14-PGJ2的非酶代谢产物13,14-二氢-15-酮-PGD2在检测极限以下。另一方面,关于由与PGD2共通的底物PGH2产生的PGE2,没能观察到依赖于受试物质添加的用量的产生响应。B:iSC细胞外酶活性的评价
使L-PGDS的底物PGH2和谷胱甘肽(GSH)作用于将iSC添加受试物质(0、50、100、1000mNU/mL)培养3天(不含FBS的DMEM/F12 F/-/-、)后的培养上清(反应条件:100mM Tris-HCl(pH8.0)、1mM GSH、10mM PGH2、37℃、5min)。与上述A.同样,通过HPLC质谱法,分别测定反应液中的前列腺素类(PGD2、PGJ2、15-脱氧-Δ12,14-PGJ2、13,14-二氢-15-酮-PGD2和PGE2)的浓度。将结果的一例表示于表5和图7。

如表5和图7所示,受试物质在100mNU/mL以上的浓度时使反应液中的PGD2及其非酶代谢产物PGJ2、15-脱氧-Δ12,14-PGJ2的浓度上升,因此确认到在iSC培养上清中存在L-PGDS活性。另外,关于15-脱氧-Δ12,14-PGJ2的非酶代谢产物13,14-二氢-15-酮-PGD2,通过添加到100mNU/mL为止的浓度范围的受试物质,观察到了用量依赖性地产生,但在1000mNU/mL时低于检测极限。
已知L-PGDS是分泌型酶,但由上述A.和B.的结果确认到iSC向细胞外释放具有酶活性的L-PGDS,通过添加受试物质,促进其产生和释放。
试验例5-1:小鼠脑内的L-PGDS产生细胞的研究(免疫组织化学染色法)
制作实施了由大脑中动脉闭塞带来的缺血负荷的C.B-17小鼠(缺血3天后)的脑切片,使用对L-PGDS(图片(panel)B、绿;抗前列腺素D合酶(脂质运载蛋白)抗体[EP12357]、Abcam公司、1﹕1000)、周细胞标志物α-SMA(图片C、红;抗肌动蛋白、平滑肌、克隆ASM-1、Millipore公司、1﹕1000)和血管内皮细胞标志物CD31(图片D、红;抗小鼠CD31(PECAM-1)单克隆抗体(anti-mouse CD31(PECAM-1)monoclonal antibody)、550274、BD Pharmingen公司、1﹕1000)的特异性抗体,进行利用共聚焦激光显微镜的免疫组织化学研究。通过多重染色法,分别用标记了不同的荧光色素的特异性抗体检测L-PGDS(图片B)和周细胞标志物(图片C)或L-PGDS(图片B)和血管内皮细胞标志物(图片D),将图像重叠,对L-PGDS和周细胞标志物或L-PGDS和血管内皮细胞标志物的分布进行比较(图片A1、图片A2)。将结果的一例表示于图8。
在图8上段,图片B的绿色荧光表示L-PGDS的分布,图片C的红色荧光表示周细胞标志物的分布,但在将两者重叠的图片A1中,L-PGDS(绿色荧光)与周细胞标志物(红色荧光)的分布局部一致(200倍)。另外,在图8下段,图片B的绿色荧光表示L-PGDS的分布,图片D的红色荧光表示血管内皮细胞标志物的分布,在将两者重叠的图片A2中,L-PGDS(绿色荧光)尽管分布于血管内皮细胞标志物(红色荧光)的附近,但是没有得到显示共分布的观察(200倍)。
L-PGDS是主要分布于中枢神经系统的蛋白,存在于蛛网膜、软膜和侧脑室的脉络丛等,被认为分泌到脑脊髓液中(Urade Y.et al.,J Lipid Mediat.Cell Signal.14,71-82,1996),但关于脑内产生细胞并未充分解明。本发明的发明人通过免疫组织化学的方法进行了有关L-PGDS的脑内分布的研究,结果C.B-17小鼠的正常脑中的表达微弱,但在上述试验例4-1中,在该小鼠中导入大脑中动脉的永久性结扎,则在脑梗塞灶确认到明显的L-PGDS表达(图8的图片B)。另外,在血管内皮细胞的标志物CD31的周围确认到L-PGDS的分布,但两者的分布不重合(图8的图片A2)。另一方面,得知L-PGDS显示与周细胞的标志物α-SMA的一部分共分布(图8的图片A1)。由以上的结果可以认为在脑梗塞灶中被表达诱导的L-PGDS来自于周细胞(或者iSC)。这可以通过如下事实得到验证:通过后述的试验例4-2的免疫电子显微镜法也确认到L-PGDS阳性产物在与血管内皮细胞和基底膜接触存在的周细胞的细胞质内作为电子密度高的结构物(图9的图片A和B)。由这些结果可以想象作为缺血响应中的周细胞或iSC的生理学的作用,包括L-PGDS的表达诱导。
试验例5-2:小鼠脑内的L-PGDS产生细胞的研究(免疫电子显微镜法)
制作实施了通过大脑中动脉闭塞带来的缺血负荷的C.B-17小鼠(缺血3天后)的脑切片,通过使用对L-PGDS(抗前列腺素D合酶(脂质运载蛋白)抗体[EP12357]、Abcam公司、1﹕1000)的特异性抗体的免疫电子显微镜法,观察L-PGDS的表达部位。具体而言,用振动切片机(Vibratome)制作2μm厚的脑切片,使用亲和素-生物素辣根过氧化物酶(HRP)复合物试剂盒(Vector Laboratories)和3,3'-二氨基联苯胺四盐酸盐(3,3'-diaminobenzidinetetrahydrochloride)(DAB)的反应后,进行锇处理,环氧树脂(EPON)包埋后,制作超薄切片进行电子显微镜观察。将结果的一例表示于图9。
在图9的图片A和B的任一图中,在对L-PGDS的免疫电子显微镜法中,在与血管内皮细胞和基底膜接触存在的血管周细胞(周细胞)的细胞质内,都确认到电子密度高的区域。
试验例6:人脑内的L-PGDS产生细胞的研究(免疫组织化学染色法)
根据Tatebayashi K.et al.,Stem Cells and Develop.26,787-797,2017中记载的方法,在从人脑梗塞灶(坏死组织)提取的培养iSC中,使用对L-PGDS(图片B、绿;抗前列腺素D合酶(脂质运载蛋白)抗体[EP12357]、Abcam公司、1﹕1000)和神经干细胞标志物巢蛋白(图片C、红;anti-巢蛋白、clone 10C2、Millipore公司、1﹕1000)的特异性抗体进行免疫组织化学的研究。细胞与Alexa Fluor〔注册商标、以下同样〕488-偶联抗体或Alexa Fluor555-偶联抗体(1﹕500;Molecular Probes、Eugene)反应后,进行利用4',6-二脒基-2-苯基吲哚(4',6-diamidino-2-phenylindole)(DAPI;1﹕1000;Kirkegaard&Perry Laboratories公司)的核染色。用荧光显微镜(Fluorescent microscope)(BX60;Olympus,Japan)进行荧光观察,将L-PGDS(图片B)和巢蛋白(图片C)的图像重叠,对L-PGDS和巢蛋白的分布进行比较(图片A)。将结果的一例表示于图10。
在图10中,图片B的绿色荧光表示L-PGDS的分布,图片C的红色荧光表示nesitin的分布,但在将两者重叠的图片A中,L-PGDS(绿色荧光)与巢蛋白(红色荧光)的分布一致(200倍)。即,确认到L-PGDS在表达巢蛋白的几乎所有iSC中都表达。由上可以认为L-PGDS不仅在小鼠脑中,而且在人脑中都在iSC或周细胞中产生。
试验例7:对阿尔茨海默型认知症模型动物的脑内淀粉样蛋白β沉积和L-PGDS表达
的评价
(1)关于认知能力的行动学解析和样品制备
将APPswe/PS1dE9(APP/PS1)小鼠(雌性3月龄)以各组体重均等的方式分为受试物质投予组和对照组(各组10~11只),在约3个月里分别将受试物质(100NU/kg体重)或生理盐水每周2次尾静脉注射。最终投予后,进行关于认知能力的行动学解析(Y字迷宫试验、新奇物体识别试验和莫里斯水迷宫试验)。在各试验中,A:Y字迷宫试验中测定交替行动率(%),B:新奇物体识别试验中测定对新奇物体的接触频度(%),以及在C:莫里斯水迷宫试验中测定到学习期间的结束时的4天(第6~9天)发现平台所需的平均时间(秒),对每个个体计算综合得分(A×B÷C)。将各组每2个个体的测定结果表示于表6。

由表6可知,观察到了由受试物质投予组得到的认知能力的改善作用,该作用在组间在统计学上有显著差异。
关于认知能力的行动学解析后(最终投予24小时后),在受试物质投予组和对照组中,在麻醉下断头,采集脑,将左半球用液氮浴急速冻结。另外,剩下的右半球通过(A)包含戊二醛的固定液(0.05%戊二醛、4%多聚甲醛、0.1M磷酸缓冲液)或(B)PLP固定液(0.01M偏高碘酸钠、0.075M赖氨酸、2%多聚甲醛)固定(4℃)。从固定组织标本通过低温恒温器(cryostats)(Leica CM1850)制作包含大脑皮质的冠状面切片(厚度20μm)。
(2)对大脑皮质中的淀粉样蛋白β沉积的评价(免疫组织化学染色法)
对上述(1)中制作的受试物质投予组和对照组的冠状面切片,进行利用抗淀粉样蛋白β抗体(抗β淀粉样蛋白(anti-β-amyloid)、1-16、[SIG-39300]、BioLegend公司、1﹕1000)的免疫组织化学染色。淀粉样蛋白β与Alexa Fluor 488偶联抗体(1﹕500;MolecularProbes、Eugene)反应后,进行利用4',6-二脒基-2-苯基吲哚(DAPI;1﹕1000;Kirkegaard&Perry Laboratories)的核染色,用荧光显微镜(BX60;Olympus,Japan)进行荧光观察。将结果的一例表示于图11。
如图11所示,在由受试物质投予确认了认知能力的改善的个体中,与对照组相比,脑淀粉样蛋白斑的数量和表面积降低。
(3)对大脑皮质中的L-PGDS表达的评价(免疫组织化学染色法)
对上述(1)中制作的受试物质投予组和对照组的冠状面切片,进行利用抗L-PGDS抗体(抗前列腺素D合酶(脂质运载蛋白)抗体[EP12357]、Abcam公司、1﹕1000)的免疫组织化学染色。L-PGDS使用亲和素-生物素辣根过氧化物酶(HRP)复合物试剂盒(VectorLaboratories公司)和3,3'-二氨基联苯胺四盐酸盐(DAB)的反应进行检测。将结果的一例表示于图12。
如图12所示,在受试物质投予组的脑中,在血窦(血管膣)附近确认到L-PGDS的特异染色(图片B的箭头部分),相对于此,在对照组的脑中,没有确认到L-PGDS的特异染色(图片A)。
另外,对上述(1)中制作的受试物质投予组的冠状面切片,使用对L-PGDS(抗前列腺素D合酶(脂质运载蛋白)抗体[EP12357]、Abcam公司、1﹕1000)和血管内皮细胞标志物CD31(抗小鼠CD31(PECAM-1)单克隆抗体、550274、BD Pharmingen公司、1﹕1000)的特异性抗体进行免疫组织化学的研究。在结合了上述的一次抗体的切片中,在与Alexa Fluor 488-偶联抗体或Alexa Fluor 555-偶联抗体(1﹕500;Molecular Probes、Eugene)反应后,进行利用4',6-二脒基-2-苯基吲哚(DAPI;1﹕000;Kirkegaard&Perry Laboratories公司)的核染色,用共聚焦激光显微镜(LSM780;Carl Zeiss、Jena,Germany)进行荧光观察。通过多重染色法,将L-PGDS(图片B)和CD31(图片C)的图像重叠,对L-PGDS和CD31的分布进行比较(图片A)。将结果的一例表示于图13。
如图13所示,在受试物质投予组的脑中,确认到L-PGDS(绿色荧光)分布于血管内皮细胞标志物CD31(红色荧光)的周围。由此,可以认为在响应受试物质的投予、以覆盖血管内皮细胞的方式存在的周细胞(或iSC)中,L-PGDS的表达上升。
(4)对脑内淀粉样蛋白β沉积和L-PGDS表达的评价(蛋白质印迹法)
将上述(1)采集后急速冻结的左半球脑用Potter型匀浆器匀浆而提取的蛋白用SDS-PAGE(BIO-RAD Any kD(商标))分离,转印于PVDF膜(Immun-Blot PVDF膜、BIO-RAD公司)。用Bloking One(nacalai tesque公司)封闭后,通过利用特异性抗体的蛋白质印迹法,检测淀粉样蛋白β(anti-β-amyloid、1-16、[SIG-39300]、BioLegend公司、1﹕1000)和L-PGDS(抗前列腺素D合酶(脂质运载蛋白)抗体[EP12357]、Abcam公司、1﹕2000)。检测使用高灵敏度化学发光法(Chemi-Lumi One L,nacalai tesque公司)。将结果的一例表示于图14。
如图14的图片A※标记部分所示,关于脑内淀粉样蛋白β量,可以认为在通过受试物质投予确认到认知能力的改善的受试物质投予组的小鼠(个体编号4、29)中,与对照组的小鼠(个体编号14、30)相比减少。该结果与在试验例7(2)的免疫组织化学染色法(图11)中,相对于对照组,在投予受试物质时,脑内淀粉样蛋白β抗体阳性产物的数量和表面积尺寸被抑制的结果相关。另外,如图14的图片B*标记部分所示,在受试物质投予组中脑内L-PGDS量与对照组相比增加,因此,可以认为受试物质增强了脑内L-PGDS产生。从上述试验例7的一系列的结果,通过受试物质使L-PGDS的脑内表达量增加,可以认为具有抑制脑内淀粉样蛋白β的沉积、进而改善认知功能的可能性。
试验例8:对来自人的周细胞中的L-PGDS表达促进原因的研究(经典的RT-PCR法)
如上所述,确认到对受试物质的响应性的iSC被认为来自于脑周细胞,因此在市售的来自人的周细胞株Human brain vascular pericytes(ScienCell公司)中,在不同的氧和/或葡萄糖浓度的条件下,调查受试物质的L-PGDS表达能力。具体而言,在0、50、500mNU/mL的各用量的受试物质存在下,分别在(1)iSC显示反应性的条件(4.5g/L葡萄糖和20%O2)、(2)低葡萄糖条件(90mg/L葡萄糖和20%O2)以及(3)低氧-低葡萄糖条件(90mg/L葡萄糖和1%O2)下,将iSC培养4天(不含FBS的DMEM培养基F/-/-)。对用RNeasy Mini Kit(QIAGEN公司)回收的总RNA,通过使用SuperScript III One-Step RT-PCR System with Platinum(Invitrogen公司)的经典的RT-PCR法,对L-PGDS基因(PTGDS)的表达量进行半定量(35个循环)。将PCR产物用2%琼脂糖凝胶电泳分离,用溴乙锭对PTGDS和β-肌动蛋白的条带进行染色而可视化检出。此外,PTGDS和β-肌动蛋白用引物使用与上述试验例2-1中使用的相同的引物。将结果的一例表示于图15。
如图15所示,虽然能够检测到条件(1)下培养的细胞的L-PGDS基因的转录,但没有观察到对受试物质的响应性。而在低氧(1%)且低葡萄糖浓度(90mg/L)的条件(3)下培养的细胞中,观察到了对受试物质明显的响应性。低氧-低葡萄糖条件被认为是模拟脑内的缺血状态,因此在这样的病理状态时可以认为周细胞(或者iSC)获得了对外界刺激的响应性,将L-PGDS这样的细胞保护性蛋白游离。
产业上的可利用性
L-PGDS主要在脑内表达,发挥着作为各种疏水性低分子的结合、输送和清除剂的作用,被认为是具有脑内环境调整、脑保护功能和睡眠调节功能等各种功能的蛋白质,因此本发明L-PGDS产生促进剂作为脑梗塞等的脑血管障碍、阿尔茨海默氏病等的认知症、失眠症等的L-PGDS参与的疾病的预防、治疗或复发预防药物有用。特别是本提取物和含有本提取物的制剂为优异的L-PGDS产生促进剂,并且作为副作用等问题少的安全性高的药剂,有用性高。另外,以周细胞或从周细胞脱分化得到的iSC中的L-PGDS产生促进作用作为指标,对L-PGDS参与的疾病的预防、治疗或复发预防有用的物质、特别是具有脑保护作用或睡眠作用的物质的本发明筛选方法是有助于开发新的治疗药物的非常有用的方法。

具有脑保护作用的物质为脑梗塞的预防、治疗或复发预防药物或者认知症的预防、治疗药物。
18.一种具有脑保护作用或睡眠作用的物质,其特征在于:
其是通过权利要求15~17中任一项所述的筛选方法得到的。

脑保护作用是通过淀粉样蛋白β的排出作用带来的。
