Abstract
本发明涉及一种原位构筑人造细胞外基质的自组装材料及其制备方法和应用,由靶向肽、自组装多肽和双芘荧光信号分子组成,其化学结构如式(Ⅰ)所示;其中,R1来自于分子内具有多氢键的自组装多肽;R2和R3均来自于肿瘤靶向肽。其制备方法为:以氨基酸和双芘荧光信号分子为原料,通过固相合成法,合成所述自组装材料。该自组装材料可主动靶向肿瘤部位,同时在肿瘤部位可被诱导发生形变,形成纳米纤维网状结构,即可原位构筑人工细胞外基质,其与天然细胞外基质相连接共同构成长期的屏障,抑制肿瘤的转移和入侵,还可以和天然细胞外基质竞争整合素的结合位点,降低金属基质蛋白酶的表达,更加抑制肿瘤的转移。

技术领域
本发明涉及生物医药领域,具体涉及一种自组装材料及其制备方法和应用,尤其涉及一种原位构筑人造细胞外基质的自组装材料及其制备方法和应用。
背景技术
肿瘤是指机体在各种致瘤因子作用下,局部组织细胞增生所形成的新生物。肿瘤分为良性肿瘤和恶性肿瘤,恶性肿瘤严重危害人们的健康,癌症死亡率居高不下的原因主要是肿瘤转移快,因此抑制肿瘤转移是肿瘤治疗的关键因素。肿瘤转移是一个极其复杂的过程,其中,通过抑制金属基质蛋白酶的活性来实现肿瘤转移的抑制是一种常用的策略,因为金属基质蛋白酶降解细胞外基质是导致肿瘤转移的主要因素,但是该方法的收效甚微。另外,现有技术中还有一些关于肿瘤转移抑制策略的报道。
CN103550192A公开了一种天然化合物在制备治疗乳腺癌迁移、侵袭药物的应用。本发明所述的天然化合物能够抑制转录因子NF-κB的转录活性,抑制NF-κB对其下游基因基质金属蛋白酶MMP-9的转录表达,进而抑制MMP-9的酶活性对肿瘤细胞迁移和侵袭的促进作用。该天然化合物同时能够克服化学致癌剂佛波酯对肿瘤侵袭的促进作用,在天然化合物的肿瘤迁移、侵袭靶向治疗方面达到一个新水平。
CN108553479A公开了一种阳离子修饰的琼脂糖和核酸药物组合物及其制备方法和靶向淋巴结免疫治疗用于阻断肿瘤迁移的应用,阳离子修饰的琼脂糖中氨基修饰的方式是伯氨基或仲氨基或叔氨基,氨基化形式为单氨基化或多氨基化,连接方式和数量不受限制;核酸药物为质粒、小片段的DNA、小片段的RNA或小片段的DNA以及RNA的杂合。所述靶向淋巴结的载体-核酸药物给药体系可以有效破坏淋巴结中的免疫抑制性环境,恢复淋巴结固有的免疫应答体制,从而达到阻断肿瘤经淋巴结转移的目的。
CN101445535A公开了一种小分子干扰RNA在制备抑制肿瘤转移药物中的用途,这种小分子干扰RNA可以沉没survivin基因表达,可以改变乳腺癌、胃癌、肝癌细胞对IV型胶原蛋白的黏附力,抑制乳腺癌、胃癌、肝癌细胞二维、三维水平上的迁移能力。这种小分子干扰RNA可以制备抑制肿瘤转移的药物。
综上,现有技术中关于抑制肿瘤转移的治疗策略还比较少,因此,开发一种的新型的疗效显著的能够抑制肿瘤转移的治疗策略是非常重要的。
发明内容
针对现有技术的不足,本发明的目的在于提供一种自组装材料及其制备方法和应用,尤其提供一种原位构筑人造细胞外基质的自组装材料及其制备方法和应用。
为达到此发明目的,本发明采用以下技术方案:
一方面,本发明提供一种原位构筑人造细胞外基质的自组装材料,所述自组装材料由靶向肽、自组装多肽和双芘荧光信号分子组成,其化学结构如式(Ⅰ)所示:
其中,R1来自于分子内具有多氢键的自组装多肽;
R2和R3均来自于肿瘤靶向肽。
所述R1来自于分子内具有多氢键的自组装多肽,该自组装多肽以酰胺键的连接方式分别与双芘荧光信号分子、R2或R3连接,所述R2和R3均来自于肿瘤靶向肽,该肿瘤靶向肽以酰胺键的连接方式与R1连接。
该自组装材料中的自组装多肽具有良好的生物相容性和自组装能力,可通过与双芘荧光信号分子段的亲疏水平衡使式(Ⅰ)所示的自组装材料自组装形成纳米球形颗粒,经EPR效应被动富集于肿瘤部位,同时在肿瘤部位可被诱导发生形变,自组装形成β-折叠纳米纤维网状结构,即人造细胞外基质,该人造细胞外基质可以和天然细胞外基质相连接,共同构成长期的屏障抑制肿瘤的转移和入侵;另外,该人造细胞外基质可以和天然细胞外基质竞争整合素的结合位点,从而降低金属基质蛋白酶(MMP-9)的表达,进而更加抑制肿瘤的转移。
该自组装材料中的肿瘤靶向肽可以实现对肿瘤部位的主动靶向,提高材料的生物安全性和生物利用度;而该自组装材料中的双芘荧光信号分子可在聚集状态中自发绿色荧光,因此可以实现自组装材料的实时生物成像。
优选地,所述R1来自于多肽序列Lys-Leu-Val-Phe-Phe或Leu-Pro-Phe-Phe-Asp。

其中,Lys-Leu-Val-Phe-Phe这一多肽序列来自于β-淀粉样蛋白(Aβ42肽),这段多肽的强氢键作用会诱导纳米颗粒形成纳米纤维。Asp-Gly-Arg序列能够靶向肿瘤细胞表面的整合素,Cys-Arg-Glu-Lys-Ala序列能够靶向纤粘连蛋白的纤维蛋白结合位点。
另一方面,本发明提供一种如上所述的原位构筑人造细胞外基质的自组装材料的制备方法,所述制备方法为:
以末端氨基和侧链氨基均得到保护的氨基酸以及末端氨基得到保护的双芘荧光信号分子为原料,通过固相合成法,合成所述原位构筑人造细胞外基质的自组装材料。
所述双芘荧光信号分子的结构式如式(Ⅷ)所示:
优选地,所述制备方法具体包括如下步骤:
(1)将载体树脂进行溶胀;
(2)采用末端氨基得到Fmoc保护、侧链氨基得到Boc保护的氨基酸以及末端氨基得到Fmoc保护的双芘荧光信号分子为原料,首先,按照R3的氨基酸顺序,将R3第一个氨基酸加入至载体树脂,与载体树脂进行偶联反应并连接;脱去R3第一个氨基酸上的Fmoc保护基,将R3第二个氨基酸与R3第一个氨基酸进行偶联反应并连接;直至完成R3中所有氨基酸的缩合;
(3)脱去R3最后一个氨基酸的Fmoc保护基,按照R1的氨基酸顺序,将R1第一个氨基酸与R3最后一个氨基酸进行偶联反应并连接;脱去R1第一个氨基酸上的Fmoc保护基,将R1第二个氨基酸与R1第一个氨基酸进行偶联反应并连接;直至完成R1中所有氨基酸的缩合;
(4)脱去R1最后一个氨基酸的Fmoc保护基,将双芘荧光信号分子的羧基端与R1最后一个氨基酸进行偶联反应并连接;
(5)脱去双芘荧光信号分子的Fmoc保护基,按照R1的氨基酸顺序,将R1第一个氨基酸与双芘荧光信号分子的氨基端进行偶联反应并连接;脱去R1第一个氨基酸上的Fmoc保护基,将R1第二个氨基酸与R1第一个氨基酸进行偶联反应并连接;直至完成R1中所有氨基酸的缩合;
(6)脱去R1最后一个氨基酸的Fmoc保护基,按照R2的氨基酸顺序,将R2第一个氨基酸与R1最后一个氨基酸进行偶联反应并连接;脱去R2第一个氨基酸上的Fmoc保护基,将R2第二个氨基酸与R2第一个氨基酸进行偶联反应并连接;直至完成R2中所有氨基酸的缩合并脱去R2最后一个氨基酸上的Fmoc保护基;
(7)将步骤(6)得到的产物从载体树脂上脱除,并脱除侧链氨基保护,得到所述原位构筑人造细胞外基质的自组装材料。
优选地,步骤(1)所述载体树脂为Wang树脂。
优选地,步骤(1)所述溶胀所使用的试剂为N,N-二甲基甲酰胺。
优选地,所述脱去Fmoc保护基所使用的脱保护剂为N,N-二甲基甲酰胺与六氢吡啶的混合溶剂。
优选地,所述N,N-二甲基甲酰胺与六氢吡啶的体积比为4:1。
优选地,所述脱去Fmoc保护基的持续时间为10-15min,例如10min、10.5min、11min、11.5min、12min、13min、14min或15min等。
优选地,所述脱去Fmoc保护基在20-30℃下进行,例如20℃、22℃、23℃、24℃、25℃、26℃、27℃、28℃或30℃等。
优选地,所述脱去Fmoc保护基后对反应产物进行清洗。

优选地,步骤(7)所述将步骤(6)得到的产物从载体树脂上脱除,并脱除侧链氨基保护的具体方法为:
收缩树脂,并进行真空干燥,用裂解液对干燥后的树脂进行裂解,裂解完成后,抽滤并旋蒸滤液,将旋干产物中的多肽析出,然后将多肽离心后干燥。
优选地,所述收缩树脂所使用的溶剂为甲醇。
优选地,所述收缩树脂的持续时间为20-40min,例如20min、23min、25min、28min、30min、32min、35min、37min或40min等。
优选地,所述裂解液按总质量为100%计包括去离子水2.5%、三异丙基硅烷2.5%、1,2-乙二硫醇2.5%和三氟乙酸92.5%。
优选地,所述将多肽析出所使用的试剂为无水乙醚。
再一方面,本发明提供一种如上所述的原位构筑人造细胞外基质的自组装材料在制备抑制肿瘤转移和入侵的药物中的应用。
相对于现有技术,本发明具有以下有益效果:
本发明所涉及的自组装材料结构中的肿瘤靶向肽可以实现对肿瘤部位的主动靶向,提高材料的生物安全性和生物利用度;结构中的双芘荧光信号分子可在聚集状态中自发绿色荧光,实现自组装材料的实时生物成像;结构中的自组装多肽具有良好的生物相容性和力学性能,可通过β折叠使自组装材料自组装形成纳米球形颗粒,经EPR效应被动富集于肿瘤部位,同时在肿瘤部位可被诱导发生形变,自组装形成纳米纤维网状结构,即可原位构筑人工细胞外基质,其与天然细胞外基质相连接共同构成长期的屏障,抑制肿瘤的转移和入侵,该人工细胞外基质还可以和天然细胞外基质竞争整合素的结合位点,从而降低金属基质蛋白酶(MMP-9)的表达,进而更加抑制肿瘤的转移。

具体实施方式
下面通过具体实施方式来进一步说明本发明的技术方案。本领域技术人员应该明了,所述实施例仅仅是帮助理解本发明,不应视为对本发明的具体限制。
实施例1
本实施例构建一种原位构筑人造细胞外基质的自组装材料,所述自组装材料由靶向肽、自组装多肽和双芘荧光信号分子组成,其化学结构如下所示;
其制备方法包括如下步骤:
(1)将Wang树脂用N,N-二甲基甲酰胺进行溶胀;
(2)采用末端氨基得到Fmoc保护、侧链氨基得到Boc保护的氨基酸以及末端氨基得到Fmoc保护的双芘荧光信号分子为原料,首先,按照Ala-Lys-Glu-Arg-Cys的氨基酸顺序,将丙氨酸加入至Wang树脂,与Wang树脂进行偶联反应并连接;脱去丙氨酸上的Fmoc保护基,将赖氨酸与丙氨酸进行偶联反应并连接;直至完成Ala-Lys-Glu-Arg-Cys中所有氨基酸的缩合;
(3)脱去甲硫氨酸的Fmoc保护基,按照Lys-Leu-Val-Phe-Phe的氨基酸顺序,将赖氨酸与甲硫氨酸进行偶联反应并连接;脱去赖氨酸上的Fmoc保护基,将异亮氨酸与赖氨酸进行偶联反应并连接;直至完成Lys-Leu-Val-Phe-Phe中所有氨基酸的缩合;
(4)脱去苯丙氨酸的Fmoc保护基,将双芘荧光信号分子的羧基端与苯丙氨酸进行偶联反应并连接;
(5)脱去双芘荧光信号分子的Fmoc保护基,按照Phe-Phe-Val-Leu-Lys的氨基酸顺序,将苯丙氨酸与双芘荧光信号分子的氨基端进行偶联反应并连接;脱去苯丙氨酸上的Fmoc保护基,将另一个苯丙氨酸与此苯丙氨酸进行偶联反应并连接;直至完成Phe-Phe-Val-Leu-Lys中所有氨基酸的缩合;
(6)脱去赖氨酸的Fmoc保护基,按照Asp-Gly-Arg的氨基酸顺序,将天冬氨酸与赖氨酸进行偶联反应并连接;脱去天冬氨酸上的Fmoc保护基,将甘氨酸与天冬氨酸进行偶联反应并连接;直至完成Asp-Gly-Arg中所有氨基酸的缩合并脱去最后一个氨基酸精氨酸上的Fmoc保护基;
(7)用甲醇收缩树脂30min,并进行真空干燥,用裂解液对干燥后的树脂进行裂解,裂解完成后,抽滤并旋蒸滤液,用无水乙醚将旋干产物中的多肽析出,然后将多肽离心后干燥,得到所述原位构筑人造细胞外基质的自组装材料。
对制得的自组装材料用质谱进行表征,质谱表征结果如图1所示,由图1可知:通过质谱图的主峰可以看出和合成的多肽材料分子量一致,由此推出合成了目标分子,表明如上式结构的自组装材料被成功合成。
实施例2
本实施例制备自组装纳米颗粒溶液和纳米纤维分散液,具体方法如下:
将实施例1制得的自组装材料溶于DMSO溶剂中(自组装材料的浓度为3×10-3M),取上述溶液置于离心管中,再将去离子水缓慢加入离心管中,制备出不同含水量(0%,98%)的混合溶液,将自组装材料单体溶液(含水量0%)和自组装纳米颗粒溶液(含水量98%),分别用荧光分光光度计进行荧光检测,荧光图谱如图2所示,由图2可以看出:自组装纳米颗粒溶液(含水量98%)在525nm处的荧光强度出现峰值,表明双芘荧光分子的J型聚集体的形成。
用透射电镜对得到的自组装纳米颗粒进行表征,结果如图3所示,由图3可以看出:实施例1制得的自组装材料在水溶液中形成了自组装颗粒。
在上述得到的自组装纳米颗粒溶液中加入无水CaCl2溶液,使Ca2+浓度为3×10-5M,静置48h后即可得到纳米纤维分散液。用透射电镜对得到的纳米纤维进行表征,结果如图4所示,由图4可以看出:自组装多肽材料变为短纤维状,直径大小为8.4±2.4nm。
因为多肽材料中的Asp-Gly-Arg序列在靶向肿瘤细胞表面整合素时,是与整合素上的Ca2+进行螯合,从而达到特异性靶向的功能。所以,此处用Ca2+溶液来体外模拟肿瘤环境。
实施例3
激光共聚焦试验:
本实施例分别采用人源乳腺癌细胞MDA-MB-231和小鼠乳腺癌细胞4T1为细胞模型,在含10%的胎牛血清和100U/mL青霉素以及100μg/mL链霉素的DMEM培养基中培养,培养温度为37.0℃,CO2的浓度为5.0%。培养细胞至对数生长期且细胞状态良好时,用胰蛋白酶消化分散细胞3min,1000r/min离心3min,弃去上清液。将每毫升含有104个细胞的细胞悬浮液加至激光共聚焦小皿中,培养24h,然后在每个共聚焦皿中各加入1mL的30μM的实施例1制得的自组装材料,放入细胞培养箱中培养2h,将上清液倒掉用PBS清洗三遍,加入适量的PBS,进行单光子激光共聚焦成像试验。试验结果如图5和图6所示,由图5和图6可以看出:两种癌细胞的表面均有很强的绿色荧光出现,说明自组装材料能够主动靶向于肿瘤细胞。
实施例4
扫描电镜试验:
本实施例分别采用人源乳腺癌细胞MDA-MB-231和小鼠乳腺癌细胞4T1为细胞模型,并分为对照组和实验组。在培养皿底部放入硅片,使对照组细胞在含10%的胎牛血清和100U/mL青霉素以及100μg/mL链霉素的DMEM培养基中培养24h,实验组细胞在含10%的胎牛血清、100U/mL青霉素、100μg/mL链霉素的DMEM培养基中培养24h,然后加入30μM实施例1制得的自组装材料培养2h,培养温度为37.0℃,CO2的浓度为5.0%,使细胞在硅片上生长。
然后弃去培养基,用PBS清洗三遍,加入4%的多聚甲醛固定1h,用PBS清洗两遍,然后用30%、50%、70%、90%、100%的乙醇/PBS溶液对细胞梯度脱水,每个浓度脱两遍,每次10min,最后用叔丁醇漂洗细胞30min。处理完后将细胞放到真空干燥箱干燥,干燥完成后用扫描电子显微镜进行观察扫描。试验结果如图7-10所示(图8和图10中的箭头所指为肿瘤细胞表面形成的纤维),从图中可以看出:在两种肿瘤细胞的表面均有很细的纤维生成,这证明了本发明所述的自组装材料在肿瘤细胞的表面形成了纤维。自组装材料利用氢键的作用以及受体和配体的相互作用,使其在肿瘤细胞的表面形成了纤维。
实施例5
细胞伤口愈合试验:
本实施例分别采用人源乳腺癌细胞MDA-MB-231和小鼠乳腺癌细胞4T1为细胞模型,并分为对照组和实验组。将细胞接种到24孔细胞培养板中,接种浓度为每毫升104个细胞,MDA-MB-231细胞在DMEM培养基,4T1细胞在1640培养基中培养24h,不更换培养基,用新的200μL枪头轻轻在单层细胞间划痕,划痕横穿过孔,枪头垂直于孔板底部。划痕后用培养基轻轻的清洗孔板2次,以除去脱落的细胞,然后在每孔中加入新鲜的培养基,与对照组相比,实验组的培养基中还添加30μM实施例1制得的自组装材料,细胞培养24h,PBS清洗2次,然后用无水甲醇固定30min,用0.1%的结晶紫(2%乙醇溶解)染色30min,PBS清洗3遍,显微镜拍照观察。试验结果如图11-14所示(图11-14中的虚线表示划痕的宽度,实线表示划痕经愈合后的宽度),从图中可以看出:MDA-MB-231模型和4T1模型的实验组中间空白的区域均比对照组宽,表示实验组抑制肿瘤横向迁移的能力更强,经过定量分析发现,经过自组装材料处理后,MDA-MB-231、4T1的创伤修复率分别下降到8.89%、56.97%,证明了本发明所述自组装材料能高效地抑制肿瘤的横向迁移。
实施例6
细胞侵袭试验:
本实施例分别采用人源乳腺癌细胞MDA-MB-231和小鼠乳腺癌细胞4T1为细胞模型,并分为对照组和实验组。将铺有人工基质胶的Transwell小室放入24孔板中,在上室加入300μL预温的无血清培养基室温下静置30min,使基质胶再水化,再吸去剩余培养液。
将培养中的MDA-MB-231细胞和4T1细胞撤血清饥饿处理12h,进一步除去血清的影响,胰酶消化MDA-MB-231细胞和4T1细胞制备细胞悬浮液,用培养基将细胞悬浮液调至每毫升含有5×105个细胞,取200μL加入Transwell小室内,并在上室加入30μM实施例1制得的自组装材料,在下室加入500μL含有10%的胎牛血清的培养基,培养细胞24h,将小室取出,用棉签轻轻刮去小室上表面的细胞,将小室下表面的细胞用无水甲醇固定30min,用0.1%的结晶紫(2%乙醇溶解)染色30min,PBS清洗3遍,显微镜拍照观察。试验结果如图15、16、17、18所示,从图中可以看出:MDA-MB-231模型和4T1模型的实验组的细胞数量相比于对照组的细胞数量显著减少,通过定量分析,自组装材料对MDA-MB-231和4T1肿瘤侵袭的抑制率分别为21.19%和29.33%,证明了本发明所述自组装材料可以有效的抑制肿瘤细胞的侵袭。
实施例7
细胞迁移试验:
该试验方法与实施例6的试验方法的区别仅在于将transwell小室替换成没有基质胶的transwell小室,其他均一致。试验结果如图19、20、21、22所示,从图中可以看出:MDA-MB-231模型和4T1模型的实验组细胞相对于对照组到达小室底部的数量显著减少,通过定量分析可以发现,自组装材料对MDA-MB-231和4T1细胞迁移的抑制率分别为16.12%和20.11%,证明了本发明所述自组装材料可以高效的抑制肿瘤细胞的纵向迁移。
实施例8
动物试验:
试验动物选用Balb/c雌性裸鼠20只,随机分为对照组和实验组,每组10只,预饲7天,在20只小鼠的右胸处皮下注射约5×106个MDA-MA-231细胞,建立小鼠肿瘤模型。当小鼠肿瘤体积约100mm3左右时,开始给药治疗。实验组小鼠每隔72小时静脉给药200μL,药物溶液为生理盐水溶解的实施例1制得的自组装材料,浓度100μM,共给药7次,对照组小鼠每隔72小时静脉给生理盐水200μL,给药7次。
结束后将小鼠进行解剖,取出小鼠肺组织进行观察,如图23和24所示(图23和图24中的箭头所指为小鼠的肺结节处),由图中可以看出:实验组小鼠的肺结节数相比于对照组的肺结节数显著减少。
对小鼠肺组织进行石蜡切片和HE染色观察,结果如图25(图25中的标示所指为小鼠的肺结节处)和26所示,由图中可以看出:实验组相比于对照组的转移灶显著减少,这和上述肺结节数的试验结果相对应。
申请人声明,本发明通过上述实施例来说明本发明的原位构筑人工细胞外基质的自组装材料及其制备方法和应用,但本发明并不局限于上述实施例,即不意味着本发明必须依赖上述实施例才能实施。所属技术领域的技术人员应该明了,对本发明的任何改进,对本发明产品各原料的等效替换及辅助成分的添加、具体方式的选择等,均落在本发明的保护范围和公开范围之内。
以上详细描述了本发明的优选实施方式,但是,本发明并不限于上述实施方式中的具体细节,在本发明的技术构思范围内,可以对本发明的技术方案进行多种简单变型,这些简单变型均属于本发明的保护范围。
另外需要说明的是,在上述具体实施方式中所描述的各个具体技术特征,在不矛盾的情况下,可以通过任何合适的方式进行组合,为了避免不必要的重复,本发明对各种可能的组合方式不再另行说明。

Claims (23)
2.如权利要求1所述的原位构筑人造细胞外基质的自组装材料的制备方法,其特征在于,所述制备方法为:
以末端氨基和侧链氨基均得到保护的氨基酸以及末端氨基得到保护的双芘荧光信号分子为原料,通过固相合成法,合成所述原位构筑人造细胞外基质的自组装材料。
3.如权利要求2所述的原位构筑人造细胞外基质的自组装材料的制备方法,其特征在于,所述制备方法具体包括如下步骤:
(1)将载体树脂进行溶胀;
(2)采用末端氨基得到Fmoc保护、侧链氨基得到Boc保护的氨基酸以及末端氨基得到Fmoc保护的双芘荧光信号分子为原料,首先,按照R3的氨基酸顺序,将R3第一个氨基酸加入至载体树脂,与载体树脂进行偶联反应并连接;脱去R3第一个氨基酸上的Fmoc保护基,将R3第二个氨基酸与R3第一个氨基酸进行偶联反应并连接;直至完成R3中所有氨基酸的缩合;
(3)脱去R3最后一个氨基酸的Fmoc保护基,按照R1的氨基酸顺序,将R1第一个氨基酸与R3最后一个氨基酸进行偶联反应并连接;脱去R1第一个氨基酸上的Fmoc保护基,将R1第二个氨基酸与R1第一个氨基酸进行偶联反应并连接;直至完成R1中所有氨基酸的缩合;
(4)脱去R1最后一个氨基酸的Fmoc保护基,将双芘荧光信号分子的羧基端与R1最后一个氨基酸进行偶联反应并连接;
(5)脱去双芘荧光信号分子的Fmoc保护基,按照R1的氨基酸顺序,将R1第一个氨基酸与双芘荧光信号分子的氨基端进行偶联反应并连接;脱去R1第一个氨基酸上的Fmoc保护基,将R1第二个氨基酸与R1第一个氨基酸进行偶联反应并连接;直至完成R1中所有氨基酸的缩合;
(6)脱去R1最后一个氨基酸的Fmoc保护基,按照R2的氨基酸顺序,将R2第一个氨基酸与R1最后一个氨基酸进行偶联反应并连接;脱去R2第一个氨基酸上的Fmoc保护基,将R2第二个氨基酸与R2第一个氨基酸进行偶联反应并连接;直至完成R2中所有氨基酸的缩合并脱去R2最后一个氨基酸上的Fmoc保护基;
(7)将步骤(6)得到的产物从载体树脂上脱除,并脱除侧链氨基保护,得到所述原位构筑人造细胞外基质的自组装材料。
4.如权利要求3所述的原位构筑人造细胞外基质的自组装材料的制备方法,其特征在于,步骤(1)所述载体树脂为Wang树脂。
5.如权利要求3所述的原位构筑人造细胞外基质的自组装材料的制备方法,其特征在于,步骤(1)所述溶胀所使用的试剂为N,N-二甲基甲酰胺。
6.如权利要求3所述的原位构筑人造细胞外基质的自组装材料的制备方法,其特征在于,所述脱去Fmoc保护基所使用的脱保护剂为N,N-二甲基甲酰胺与六氢吡啶的混合溶剂。
7.如权利要求6所述的原位构筑人造细胞外基质的自组装材料的制备方法,其特征在于,所述N,N-二甲基甲酰胺与六氢吡啶的体积比为4:1。
8.如权利要求3所述的原位构筑人造细胞外基质的自组装材料的制备方法,其特征在于,所述脱去Fmoc保护基的持续时间为10-15min。
9.如权利要求3所述的原位构筑人造细胞外基质的自组装材料的制备方法,其特征在于,所述脱去Fmoc保护基在20-30℃下进行。
10.如权利要求3所述的原位构筑人造细胞外基质的自组装材料的制备方法,其特征在于,所述脱去Fmoc保护基后对反应产物进行清洗。
11.如权利要求10所述的原位构筑人造细胞外基质的自组装材料的制备方法,其特征在于,所述清洗所使用的试剂为N,N-二甲基甲酰胺和二氯甲烷。
12.如权利要求3所述的原位构筑人造细胞外基质的自组装材料的制备方法,其特征在于,所述偶联反应所使用的偶联剂为N,N-二甲基甲酰胺与N-甲基吗啉的混合溶剂。
13.如权利要求12所述的原位构筑人造细胞外基质的自组装材料的制备方法,其特征在于,所述N,N-二甲基甲酰胺与N-甲基吗啉的体积比为95:5。
14.如权利要求3所述的原位构筑人造细胞外基质的自组装材料的制备方法,其特征在于,所述偶联反应的反应时间为60-90min。
15.如权利要求3所述的原位构筑人造细胞外基质的自组装材料的制备方法,其特征在于,所述偶联反应的反应温度为20-30℃。
16.如权利要求3所述的原位构筑人造细胞外基质的自组装材料的制备方法,其特征在于,所述偶联反应完成后对反应产物进行清洗。
17.如权利要求16所述的原位构筑人造细胞外基质的自组装材料的制备方法,其特征在于,所述清洗所使用的试剂为N,N-二甲基甲酰胺和二氯甲烷。
18.如权利要求3所述的原位构筑人造细胞外基质的自组装材料的制备方法,其特征在于,步骤(7)所述将步骤(6)得到的产物从载体树脂上脱除,并脱除侧链氨基保护的具体方法为:
收缩树脂,并进行真空干燥,用裂解液对干燥后的树脂进行裂解,裂解完成后,抽滤并旋蒸滤液,将旋干产物中的多肽析出,然后将多肽离心后干燥。
19.如权利要求18所述的原位构筑人造细胞外基质的自组装材料的制备方法,其特征在于,所述收缩树脂所使用的溶剂为甲醇。
20.如权利要求18所述的原位构筑人造细胞外基质的自组装材料的制备方法,其特征在于,所述收缩树脂的持续时间为20-40min。
21.如权利要求18所述的原位构筑人造细胞外基质的自组装材料的制备方法,其特征在于,所述裂解液按总质量为100%计包括去离子水2.5%、三异丙基硅烷2.5%、1,2-乙二硫醇2.5%和三氟乙酸92.5%。
22.如权利要求18所述的原位构筑人造细胞外基质的自组装材料的制备方法,其特征在于,所述将多肽析出所使用的试剂为无水乙醚。
23.如权利要求1所述的原位构筑人造细胞外基质的自组装材料在制备抑制肿瘤转移和入侵的药物中的应用。
