Description

External traditional Chinese medicine care solution for promoting wound healing
Technical Field
The invention belongs to the technical field of traditional Chinese medicines, and particularly relates to an external traditional Chinese medicine care solution for promoting wound healing.
Background
The skin is the first barrier of the body to the outside world, and can prevent the invasion of bacteria, viruses and the like, but when the skin is damaged to form wounds, the human body or other animals can be invaded by the bacteria and the viruses. The wound is treated correctly and can heal rapidly, otherwise, the wound can be suppurative and infected for a long time and even complicated with systemic infection, gas gangrene, tetanus and the like which endanger life. Clinically, skin injuries caused by various reasons are common diseases and high morbidity, and scars formed after the wounds bring huge damage to the physiology and the psychology of patients. At present, common medicines for disinfecting and debriding skin comprise medical alcohol, diluted iodine tincture, iodophor, furacilin solution, benzalkonium bromide solution and the like, but clinical application finds that the medicines have strong irritation, cannot be applied in a large area, have unsatisfactory debridement effect and even have anaphylactic reaction. In addition, the medicines have no functions of promoting granulation growth, easing pain and the like, basically stay in the stage of waiting for the natural healing of the wound surface, and have very limited effect of promoting the healing of the wound surface. Some medicines contain some growth factors, although the growth factors have stronger cell proliferation promoting effect and can promote wound healing, the safety of the medicines is always the focus of attention of people because the wound healing needs a plurality of growth factors, the compatibility is complex, and the excessive proliferation of the cells can form proliferative scars and even cause canceration. Some medicines mainly take antibiotic treatment medicines, and the excessive use of the antibiotic medicines can easily cause pathogenic bacteria to generate drug resistance and reduce human immunity.
With the importance of Chinese medicine and the continuous acceleration of social rhythm, more people like or choose the method of external treatment of Chinese medicine to treat diseases or to keep health, and the Chinese medicine has the advantages of simplicity, convenience, low price, safety, reliability, quick curative effect, easy popularization and the like. Natural medicines are an important source for finding new medicines at present, and have attracted wide attention of all countries in the world; the Chinese medicine is an important component of the Chinese medicine and is also an advantageous field of new medicine development in China. The traditional Chinese medicine is externally used for promoting wound healing, has unique curative effect, takes 'pus accretion per nestle', 'removing putrefaction and promoting tissue regeneration', 'promoting blood circulation and removing blood stasis' as main treatment principles, plays a multi-target, multi-link and multi-level comprehensive regulation and control role aiming at main influencing factors of wound healing, improves the curative effect of the medicine, reduces adverse reactions and reduces the medication cost. At present, a plurality of traditional Chinese medicine preparations for promoting the healing of skin wounds have been developed successively at home and abroad, but the traditional Chinese medicine preparations for promoting the healing of the wounds have advantages and have defects of different degrees, so that the field needs to further research and develop a novel external traditional Chinese medicine nursing liquid which has good effect on promoting the healing of the skin wounds, simple preparation process, low cost, and outstanding antibacterial, anti-inflammatory, astringent, hemostatic, putrefaction-removing, granulation-promoting and granulation-promoting effects.

First, antibacterial experiment
Taking slant cultures of staphylococcus aureus, escherichia coli and pseudomonas aeruginosa, respectively inoculating the slant cultures into a meat soup culture medium, culturing the slant cultures at 36.5 ℃ for 24 hours, respectively taking 0.5 ml of the slant cultures, concentrating the slant cultures into 13 plate culture media, respectively taking the same amount of the medicines prepared in the examples 1-7 and the comparative examples 1-6, and respectively dripping the medicines into the 13 plate culture media in equal amount; acting for 5 minutes to detect the sterilization rate; the results are shown in the following table:
Figure BDA0002412995640000061
as can be seen from the table above, the wound care solution prepared by the invention has good antibacterial effect. Compared with the prior art, the wound care solution prepared by the raw materials and the proportion of the raw materials has better antibacterial effect on most pathogenic bacteria than other care solutions.
Second, testing the hemostatic and healing promoting effects
Taking 156 healthy SD rats, randomly and evenly dividing into 13 groups, each group comprises 12 rats, performing pentobarbital anesthesia (30mg/kg), opening 1cm beside one side of back vertebral column, and performing full-layer skin excision with a sharp knife to form an area of 3cm2The wound surface is cut off from the circular full-thickness skin, and the skin at the symmetrical part on the opposite side is used as the normal self-contrast. The same amount of the external traditional Chinese medicine nursing liquid of examples 1 to 7 and comparative examples 1 to 6 was used for wound treatment three times a day. The dacron is thrown on the 3 rd and 7 th daysAnd (5) calculating the size of the wound surface by film drawing and calculating the healing rate of the wound surface. The results are as follows:
Figure BDA0002412995640000071
as can be seen from the above table, compared with the comparative examples, the embodiment of the invention has the advantages that the hemostasis speed is high, the wound healing rate in 3 rd and 7 th days is better than that in comparative examples 1-6, and particularly, the hemostasis and healing promotion effects of the embodiment 1 are optimal, so that the wound healing promotion effect of the invention is obvious.
In addition, 6 rats in example 1 and comparative examples 1-6 were selected at random on days 3 and 7 respectively and sacrificed by excessive anesthesia, granulation tissue at the wound margin boundary was cut off, expression of lysozyme was measured by S-P method, and in situ apoptosis was detected by Tunel-AP method.
Variation of the content of lysozyme (x. + -. s) for each group
Figure BDA0002412995640000081
As can be seen from the table, the lysozyme content of the rat wound surface of the example is larger on the 3 rd day, the lysozyme content of the wound surface of the comparative example is larger on the 7 th day, and the lysozyme content of the example 1 of the invention is higher than that of the comparative example on the 3 rd and 7 th days. The embodiment 1 has the advantages that the time for the lysozyme content is earlier than that of the comparative example group, and the embodiment shows that the macrophage activity and function are enhanced, the cell function is enhanced, the ruins cleaning effect is enhanced, and the debridement function is strong. The apoptosis index result shows that the apoptosis index of the rat wound apoptosis in the example 1 is the largest on the 3 rd day, the apoptosis index of the rat wound apoptosis in the example 1 group is obviously reduced on the 7 th day compared with the 7 th day, and the apoptosis index of the rat wound apoptosis in the comparative example is increased on the 7 th day. Therefore, the care solution provided by the embodiment of the invention has the effect of rapidly promoting local tissue cell apoptosis, particularly induces apoptosis of dying cells in a transition region between wound necrotic tissues and normal tissues, is beneficial to promoting shedding of the necrotic tissues, is beneficial to wound debridement and granulation tissue growth, and can promote skin wound healing.
The healing condition of the rest of the rats which are not killed is continuously observed, and by visual observation, no obvious bacterial infection condition is seen in 3 days after injury in the examples 1-7 of the invention, no fat liquefaction phenomenon exists, more than 90 percent of wound surfaces heal, and the wound surfaces are flat and have no dark color; comparative examples 2 and 6 had no obvious infection on the wound surface on day 3, but the wound surface healed for more than 7 days, and obvious scars remained after the wound surface healed; comparative examples 3, 4 and 5 no obvious infection of the wound surface on day 3, and wound surface healing was observed on day 10, and fine scars remained after wound surface healing; comparative example 1, small area of fat liquefaction occurred in the case of a small amount of bacterial infection on the wound surface on day 3, the wound surface healed on day 14, and fine scars remained after the wound surface healed.
It can be seen from this that: compared with the wound care solutions of comparative examples 1 to 6, the wound care solutions of examples 1 to 7 of the invention have higher hemostasis speed and wound recovery rate, better wound recovery condition and smoother healed wound surface. The wound sprays of examples 1 to 7 were also subjected to cytotoxicity, acute test, skin irritation, and sensitization test, and the results were: the spray has no potential cytotoxicity in cytotoxicity test, and has no adverse reaction in acute test, skin irritation test and skin sensitization test.
Although embodiments of the present invention have been shown and described, it will be appreciated by those skilled in the art that changes, modifications, substitutions and alterations can be made in these embodiments without departing from the principles and spirit of the invention, the scope of which is defined in the appended claims and their equivalents.
