Abstract
The invention discloses a traditional Chinese medicine composition for inhibiting formation of dental plaque, a preparation method and application thereof. The action principle of the invention is that the phytochemical extract is utilized to adjust the dynamic balance of flora in the oral cavity, and the growth and adhesion of pathogenic bacteria such as streptococcus mutans and the like are inhibited to avoid forming dental plaque, thereby relieving periodontitis and halitosis. The trial tests of people show that the composition can effectively inhibit the formation of dental plaque, and the five components have synergistic effect. The composition can be developed into products such as mouth wash, oral spray and solid beverage.
Description
Technical Field
The invention relates to the technical field of medicines, and particularly relates to a traditional Chinese medicine composition for inhibiting dental plaque formation, a preparation method and application thereof.
Background
Dental plaque is a soft, unmineralised bacterial biofilm formed by a large number of bacteria that colonise the tooth surface, between teeth or on the surface of dentures (dentures, restorations), together with exfoliated oral epithelial cells and food debris, has strong adhesion and is difficult to remove by mouth rinsing and tooth brushing. The growth, reproduction and acid production of bacteria in the bacterial plaque can remove the enamel, which is a common cause of tooth diseases such as periodontal disease, decayed tooth and the like. In addition, plaque can calcify to form calculus, resulting in bad breath. Since bacterial growth is continuous, the key to plaque control is the frequent inhibition of harmful oral bacteria, particularly anaerobic bacteria such as streptococci and filamentous bacteria.
In order to inhibit the formation of dental plaque, some collutory and toothpaste products added with chemical pharmaceutical ingredients such as alcohol, oxidant, lysozyme, antibiotics and the like appear in the market, and can effectively reduce the quantity of pathogenic microorganisms in the oral cavity. However, the perceived comfort and acceptance of the patient is reduced due to the corrosive, irritating odor and staining of the teeth of some formulations. Therefore, there is a need to develop a more safe and effective dental plaque inhibitor.
In view of this, the invention is particularly proposed.
Disclosure of Invention
The invention aims to overcome the defects in the prior art and provide a traditional Chinese medicine composition for inhibiting dental plaque formation, a preparation method and application thereof.
The invention is realized by the following steps:
the embodiment of the invention provides a traditional Chinese medicine composition for inhibiting dental plaque formation, which comprises the following components in parts by weight: 5-10 parts of houttuynia cordata, 10-15 parts of dandelion, 15-20 parts of mint, 20-30 parts of honeysuckle and 10-15 parts of liquorice.
The embodiment of the invention also provides a preparation method of the traditional Chinese medicine composition for inhibiting dental plaque formation, which comprises the following steps: mixing herba Houttuyniae, herba Taraxaci, herba Menthae, flos Lonicerae and Glycyrrhrizae radix at a certain proportion, extracting with solvent, filtering, and concentrating the obtained filtrate into soft extract.
The embodiment of the invention also provides application of the traditional Chinese medicine composition for inhibiting formation of dental plaque in preparation of a medicine for treating or inhibiting chemical stains, dental plaque, acid erosion or tartar on the surface of teeth.
The embodiment of the invention also provides application of the traditional Chinese medicine composition for inhibiting dental plaque formation in preparing a medicine for treating or preventing oral diseases.
The embodiment of the invention also provides application of the traditional Chinese medicine composition for inhibiting formation of dental plaque in preparation of food, medicines or oral care products for preventing dental caries, relieving periodontal diseases or treating halitosis.
The invention has the following beneficial effects:

Detailed Description
In order to make the objects, technical solutions and advantages of the embodiments of the present invention clearer, the technical solutions in the embodiments of the present invention will be clearly and completely described below. The examples, in which specific conditions are not specified, were conducted under conventional conditions or conditions recommended by the manufacturer. The reagents or instruments used are not indicated by the manufacturer, and are all conventional products available commercially.
In recent years, researchers have begun to look for natural phytochemicals having antibacterial effects as an alternative ingredient to chemically synthesized antibacterial agents. The preparation method comprises the steps of preparing aqueous phase and ethanol extracts of medicinal and edible plant materials such as houttuynia cordata, dandelion, mint, chrysanthemum, honeysuckle, boat-fruited sterculia seed, lophatherum gracile, clove and liquorice, comparing the capacity of the extracts for inhibiting the proliferation of streptococcus mutans (Streptococcus salivarius), preferably selecting the ethanol extracts of the houttuynia cordata, the dandelion, the honeysuckle, the mint and the liquorice to have strong antibacterial capacity, obtaining mixed extracts with good taste and antibacterial effect by optimizing the proportion of the five plant materials, and gargling 3 times after meals by using gargle prepared by the extracts through the self contrast test of 80 volunteers, so that the formation of dental plaque can be effectively inhibited, and the extract mixture can be added into chewing gum, toothpaste and beverage to form a special product for inhibiting the formation of the dental plaque.
In order to achieve the above purpose, the following technical scheme is adopted:
in a first aspect, the embodiment of the present invention provides a traditional Chinese medicine composition for inhibiting dental plaque formation, which includes the following components in parts by weight: 5-10 parts of houttuynia cordata, 10-15 parts of dandelion, 15-20 parts of mint, 20-30 parts of honeysuckle and 10-15 parts of liquorice.
The embodiment of the invention provides a traditional Chinese medicine composition for inhibiting dental plaque formation, which comprises the following components in part by weight: the traditional Chinese medicine composition comprises the components of houttuynia cordata, dandelion, mint, honeysuckle and liquorice, and can be a medicine and food dual-purpose resource combination, and can be formed by directly mixing raw medicinal materials or combining extracting solutions, wherein the traditional Chinese medicine composition in the embodiment of the invention is prepared according to the monarch and minister compatibility principle of the traditional Chinese medicine, and specifically comprises the following components in parts by weight:
the herba Houttuyniae is fresh whole plant or dried aerial part of houttuynia cordata Thunb of Saururaceae, has effects of clearing heat and detoxicating, resolving carbuncle, discharging pus, inducing diuresis and treating stranguria, and can be used for treating lung carbuncle, empyema, phlegm heat, asthma, dysentery, heat stranguria, carbuncle, and suppurative sore. The active ingredients of the houttuynia cordata, such as decanoyl acetaldehyde, methyl n-nonanone, myrcene, sunflower aldehyde and the like, have broad-spectrum antibacterial activity and have obvious inhibiting effect on beta coccus, staphylococcus aureus, influenza bacillus and pneumococcus.
The dandelion is also named as herba violae, is wild all over the land, is dry whole herb of perennial herb dandelion of the compositae, is cold in nature, bitter and sweet in taste, enters liver and stomach channels, has the effects of clearing heat and removing toxicity, and eliminating carbuncles and dissipating stagnation, and belongs to heat and toxic materials in Chinese herbal medicines.
Herba Menthae is dry aerial part of Mentha haplocalyx of Labiatae, belongs to pungent and cool exterior syndrome relieving Chinese medicinal materials, and is used for dispelling pathogenic wind and heat, refreshing mind, relieving sore throat, and promoting eruption. The herba Menthae contains phytochemicals such as pulegone, carvone, L-menthol and L-menthone, and has antibacterial and antiinflammatory effects.
The flos Lonicerae is dried bud or flower with initial bloom of Lonicera Japonica of Caprifoliaceae, belongs to heat-clearing and detoxicating traditional Chinese medicine, and can be used for treating carbuncle, furuncle, wind heat, epidemic febrile disease, and bloody dysentery. The phenolic acid substances in the honeysuckle have obvious bacteriostatic effects on bacillus subtilis, staphylococcus aureus, escherichia coli and pseudomonas aeruginosa.
The licorice root is a perennial herb of Leguminosae, and is used as a medicine by root and rhizome, is a tonifying Chinese herbal medicine, is used for spleen deficiency and weak qi, cough and asthma, carbuncle-abscess sore and toxin, urethral pain, harmonizing medicine property and the like, is used for clearing fire, is used for raw use, and is used for reinforcing middle-jiao and moxibustion.
The embodiment of the invention provides a traditional Chinese medicine composition for inhibiting formation of dental plaque, which takes representative bacteria streptococcus mutans (streptococcus mutans) in dental plaque as a test strain, and 5 extracts with the lowest effective inhibitory concentration (MIC) are screened from 10 medicine and food dual-purpose resources with reported antibacterial activity, wherein the extracts comprise houttuynia cordata, dandelion, mint, honeysuckle and liquorice. Wherein the dandelion, the honeysuckle and the houttuynia cordata are all heat-clearing and detoxifying traditional Chinese medicines which are monarch medicines, and the dandelion, the honeysuckle and the houttuynia cordata are in compatibility with each other; the mint auxiliary monarch drug strengthens the drugs for treating the major diseases and the major symptoms, and is a drug for diminishing inflammation, sterilizing and playing a role in treating concurrent diseases or concurrent symptoms; the liquorice is a channel-inducing medicine in the recipe, and has compatibility with the first two important types, so that the medicines in the recipe directly reach the affected part, and can coordinate the medicines, enhance the effects of clearing away heat and toxic materials and relieving exterior syndrome with pungent and cool natured drugs, and make the medicines cooperate to eliminate pathogenic factors. The raw materials are compatible to play a role in preventing and adjusting oral flora, and the tests of people prove that the oral bacterial plaque can effectively inhibit the formation of dental plaque, and is beneficial to preventing oral diseases such as oral ulcer and periodontal disease caused by certain pathogenic bacteria, so that the oral health condition is improved.
In a second aspect, the embodiment of the present invention provides a preparation method of the above traditional Chinese medicine composition for inhibiting dental plaque formation, including: mixing herba Houttuyniae, herba Taraxaci, herba Menthae, flos Lonicerae and Glycyrrhrizae radix at a certain proportion, extracting with solvent, filtering, and concentrating the obtained filtrate into soft extract.
Further, the preparation method comprises the following steps: drying herba Houttuyniae, herba Taraxaci, herba Menthae, flos Lonicerae and Glycyrrhrizae radix respectively, mechanically pulverizing, micronizing, adding into water or ethanol, extracting, concentrating, and vacuum filtering.
Further, the leaching temperature is 40-70 deg.C, and the leaching time is 30-60 min.
Further, the concentration conditions were: centrifuging and filtering the leaching solution, collecting supernatant, and concentrating under reduced pressure at 50-70 deg.C to 35-45% of the original volume;
furthermore, the aperture of the filter membrane subjected to suction filtration is less than or equal to 100 nm.
In a third aspect, the embodiments of the present invention further provide an application of a traditional Chinese medicine composition for inhibiting formation of dental plaque in preparation of a medicament for treating or inhibiting chemical stains, dental plaque, acid erosion, or tartar on a tooth surface.
In a fourth aspect, the embodiment of the present invention further provides an application of a traditional Chinese medicine composition for inhibiting dental plaque formation in the preparation of a medicament for treating or preventing oral diseases.
In a fifth aspect, the embodiment of the present invention further provides an application of a traditional Chinese medicine composition for inhibiting dental plaque formation in preparing a food, a drug or an oral care product for preventing dental caries, alleviating periodontal disease or treating halitosis.
Further, the traditional Chinese medicine composition for inhibiting the formation of dental plaque is prepared into mouthwash, spray, toothpaste, buccal tablets, effervescent tablets, chewable tablets, granules or powder solid beverages. Wherein the herbal composition further comprises one or more agents selected from the group consisting of diluents, bicarbonate salts, pH modifying agents, surfactants, foam modulators, viscosity modifiers, humectants, sweeteners, flavorants, pigments, anticaries agents, anticalculus or tartar control agents, abrasives, and mixtures thereof.
The features and properties of the present invention are described in further detail below with reference to examples.
Example 1
Screening the bacteriostatic action of common traditional Chinese medicines on streptococcus mutans:

as can be seen from table 1 above, compared with the negative control, the difference of the bacteriostatic ratio of the extract of five plant materials is extremely significant, and the bacteriostatic ratio of the alcohol extract is usually higher than that of the corresponding water extract, and the extract is sequentially arranged by houttuynia cordata, dandelion, mint, honeysuckle and liquorice.
The traditional Chinese medicine composition for inhibiting dental plaque formation comprises the following components in parts by weight: 1 part of houttuynia cordata extract, 1.5 parts of dandelion extract, 2 parts of mint extract, 3 parts of honeysuckle extract and 1.5 parts of liquorice extract.
The bacteriostatic effect of the traditional Chinese medicine composition for inhibiting dental plaque formation provided in this example was evaluated:
evaluation of bacteriostatic effect referring to fig. 1, the lowest effective inhibitory concentration of each plant extract and composition in fig. 1 for inhibiting the proliferation of streptococcus mutans is shown as statistical difference in letters. As can be seen from FIG. 1, the lowest effective inhibitory concentration of the composition obtained by combining the 5 Chinese medicinal extracts is obviously lower than that of the single component, which indicates that the effective inhibitory effect can be achieved by combining the 5 different Chinese medicaments.
The effect of the traditional Chinese medicine composition for inhibiting dental plaque formation provided in this example was evaluated:
80 healthy adult volunteers were recruited, smokers and patients with severe dental disease were excluded, and plaque formation evaluation test was performed for 2 weeks. During the experiment, the consumption of sweets such as candies and the like and the drinking of colored beverages such as strong tea, coffee and the like are avoided, other medicines are forbidden, the subjects brush their teeth once respectively before getting up in the morning and sleeping at night, and the toothpaste is common toothpaste and is provided by a subject group. Five teeth were thoroughly cleaned supragingivally before the experiment for each subject, at which the Plaque Index (PI) was calculated to be 0.

Sixthly, gargling twice with honeysuckle extract after each meal
Seventh group, rinsing twice with Glycyrrhrizae radix extractive solution after meals
Eighth group, rinsing twice with the extract composition after each meal
After 14 days each examined tooth surface was stained by chewing a plaque indicator and 5 tooth Plaque Indices (PI) were calculated.
Evaluation of the effect of inhibiting plaque formation referring to fig. 2, the effect of gargling each plant extracts and compositions on plaque formation in fig. 2 is shown with statistical differences in letters. As can be seen from fig. 2: the plant extract provided by the embodiment of the invention has no difference with the dental plaque index of the chemical preparation mouthwash, and compared with a clear water control, the plant extract can effectively reduce the dental plaque index, and the dental plaque index of the compound is the lowest, which indicates that the plant extract has a synergistic effect.
Example 2
A traditional Chinese medicine composition for inhibiting dental plaque formation comprises the following components in parts by weight: 5 parts of cordate houttuynia, 15 parts of dandelion, 20 parts of mint, 25 parts of honeysuckle and 15 parts of liquorice.
Example 3
A traditional Chinese medicine composition for inhibiting dental plaque formation comprises the following components in parts by weight: 10 parts of houttuynia cordata, 10 parts of dandelion, 15 parts of mint, 25 parts of honeysuckle and 10 parts of liquorice.
Example 4
A traditional Chinese medicine composition for inhibiting dental plaque formation comprises the following components in parts by weight: 10 parts of heartleaf houttuynia herb, 10 parts of dandelion, 20 parts of mint, 20 parts of honeysuckle and 10 parts of liquorice.
Example 5
The preparation method of the traditional Chinese medicine composition for inhibiting dental plaque formation comprises the following steps:
fully drying, mechanically crushing and ultramicro-crushing 10 parts of houttuynia cordata, 10 parts of dandelion, 15 parts of mint, 20 parts of honeysuckle and 15 parts of liquorice according to the weight ratio of 1: feeding 12 of the raw materials into water or 60% ethanol (prepared by edible alcohol), stirring and leaching for 30 minutes at 60 ℃, centrifuging by a sedimentation centrifuge, removing filter residues, concentrating the centrifugate at 60 ℃ under reduced pressure to 40% of the original volume, ensuring that the ethanol is completely distilled, and performing suction filtration by a 100nm filter membrane to obtain clear filtrate as an extracting solution raw material.

Spraying conditions are as follows: the feeding temperature is 60 ℃, the air inlet temperature is 150 ℃, and the air outlet temperature is 80 ℃. In summary, the embodiment of the invention provides a traditional Chinese medicine composition for inhibiting dental plaque formation, a preparation method and an application thereof. According to the in vitro antibacterial activity, the alcohol extracts of five medicinal and edible resources such as houttuynia, dandelion, mint, honeysuckle, liquorice and the like are preferably selected, and are optimally combined to form a compound with better taste. The action principle of the invention is that the phytochemical extract is utilized to adjust the dynamic balance of flora in the oral cavity, and the growth and adhesion of pathogenic bacteria such as streptococcus mutans and the like are inhibited to avoid forming dental plaque, thereby relieving periodontitis and halitosis. The trial tests of people show that the composition can effectively inhibit the formation of dental plaque, and the five components have synergistic effect. The composition can be developed into products such as mouth wash, oral spray and solid beverage.
Compared with the prior art, the embodiment of the invention has the following beneficial effects:
(1) the cost is low, the operation is simple, and the plant extract can be obtained only by simple operation processes of drying, crushing, leaching, filtering and the like;
(2) the safety is good, the used materials are both medicinal and edible resources, and the ethanol extract is prepared from edible alcohol and has no organic solvent residue;
(3) the product has good stability, and polysaccharide and protein precipitate are removed by centrifugation after alcohol extraction;
(4) the product has multiple effects, can resist bacteria and viruses, has the aromatic components of the Chinese herbal medicines, and can be developed into tooth care products such as mouthwash toothpaste, chewing gum, beverage and the like and flavor beverage.

Claims (10)
1. A traditional Chinese medicine composition for inhibiting formation of dental plaque is used for treating or inhibiting chemical stains, dental plaque, acid erosion or tartar on tooth surfaces, and comprises the following components in parts by weight: 5-10 parts of houttuynia cordata, 10-15 parts of dandelion, 15-20 parts of mint, 20-30 parts of honeysuckle and 10-15 parts of liquorice.
2. A method of preparing a traditional Chinese medicine composition for inhibiting dental plaque formation according to claim 1, comprising: mixing herba Houttuyniae, herba Taraxaci, herba Menthae, flos Lonicerae and Glycyrrhrizae radix at a certain proportion, extracting with solvent, filtering, and concentrating the obtained filtrate into soft extract.
3. The method of claim 2, comprising the steps of: drying herba Houttuyniae, herba Taraxaci, herba Menthae, flos Lonicerae and Glycyrrhrizae radix respectively, mechanically pulverizing, micronizing, adding into water or ethanol, extracting, concentrating, and vacuum filtering.
4. A method according to claim 3, wherein the leaching is carried out at a temperature of 40-70 ℃ for a period of 30-60 min.
5. The method according to claim 3, wherein the concentration conditions are: centrifuging the leaching solution, collecting supernatant, and concentrating under reduced pressure at 50-70 deg.C to 35-45% of the original volume.
6. The method according to claim 3, wherein the pore size of the filtration membrane is 100nm or less.
7. Use of the traditional Chinese medicine composition for inhibiting formation of dental plaque according to claim 1 or the traditional Chinese medicine composition for inhibiting formation of dental plaque prepared by the preparation method according to any one of claims 2 to 6 in the preparation of a medicament for treating or inhibiting chemical stains, dental plaque, acid erosion or tartar on tooth surfaces.
8. Use of the traditional Chinese medicine composition for inhibiting formation of dental plaque according to claim 1 or the traditional Chinese medicine composition for inhibiting formation of dental plaque prepared by the preparation method according to any one of claims 2 to 6 in the preparation of a medicament for treating or preventing oral diseases.
9. Use of the traditional Chinese medicine composition for inhibiting formation of dental plaque according to claim 1 or the traditional Chinese medicine composition for inhibiting formation of dental plaque prepared by the preparation method according to any one of claims 2 to 6 in the preparation of a medicament for preventing dental caries, alleviating periodontal disease or treating halitosis.
10. The use of claim 9, wherein the composition is formulated as a spray, troche, effervescent tablet, chewable tablet or granule.
