Abstract
The invention relates to a composition for improving memory and preventing senile dementia and a preparation method and application thereof, belonging to the technical field of medicines. The composition comprises the following raw materials for preparation: deer blood, deer heart, haematococcus pluvialis, phytosterol ester, spirulina and dihydromyricetin. The composition disclosed by the invention is definite in components, safe and effective, has no toxic or side effect, and has the effects of obviously improving memory and preventing senile dementia.
Description
Technical Field
The invention relates to the technical field of medicines, in particular to a composition for improving memory and preventing senile dementia and a preparation method and application thereof.
Background
With the increasing speed of the aging and aging of the population and the increasing life span of the population, the number of patients suffering from senile dementia is gradually increased, and the senile dementia becomes a serious social problem. In the prior art, chemical medicines are mostly adopted to treat senile dementia, but the chemical medicines have side effects and dependence.

Preferably, the deer blood is deer blood powder, and the preparation method of the deer blood powder comprises the following steps: dehydrating sanguis Cervi, lyophilizing, and pulverizing to obtain sanguis Cervi powder.
Preferably, the deer heart is deer heart powder, and the preparation method of the deer heart powder comprises the following steps: cleaning Cor Cervi, cutting into pieces, lyophilizing, and pulverizing to obtain Cor Cervi powder.
Preferably, the dosage form of the composition comprises an oral preparation, and the oral preparation comprises powder, granules, tablets or pills.
The invention also provides a preparation method of the composition in the technical scheme, which comprises the following steps:
1) cleaning Cor Cervi, cutting into pieces, lyophilizing, and pulverizing to obtain Cor Cervi powder;
2) dehydrating sanguis Cervi, lyophilizing, and pulverizing to obtain sanguis Cervi powder;
3) sterilizing and drying deer heart powder and deer blood powder to obtain deer heart micro powder and deer blood micro powder;
4) mixing Cor Cervi micropowder, sanguis Cervi micropowder, Haematococcus pluvialis, phytosterol ester, Spirulina and dihydromyricetin to obtain composition;
the step 1) and the step 2) are not limited in chronological order.
The invention also provides application of the composition in the technical scheme or the composition prepared by the preparation method in the technical scheme in preparation of health-care food or medicine for improving memory and preventing senile dementia.
The invention provides a composition for improving memory and preventing senile dementia. The composition consisting of the raw materials of deer blood, deer heart, haematococcus pluvialis, phytosterol ester, spirulina and dihydromyricetin which can be eaten for a long time is used for improving memory and preventing senile dementia, is safe and effective, can be used for preparing health-care food and health food with the functions of improving memory and preventing senile dementia, can also be prepared into medicinal preparations with the functions of improving memory and preventing senile dementia, is easy to accept by patients, and accords with the general trend of social development. The composition disclosed by the invention is definite in components, safe and effective, has no toxic or side effect, and has the effects of obviously improving memory and preventing senile dementia. Detailed Description
The invention provides a composition for improving memory and preventing senile dementia, which comprises the following raw materials for preparation: deer blood, deer heart, haematococcus pluvialis, phytosterol ester, spirulina and dihydromyricetin.
The deer blood powder is prepared by dehydrating deer blood by a conventional operation method, freeze-drying and crushing to obtain the deer blood powder, the deer blood powder is not specially limited in source, and conventional deer blood commercial products well known to technicians in the field can be adopted.
The deer heart powder is prepared by the following steps of cleaning and cutting deer hearts, freeze-drying by a conventional method and crushing to obtain the deer heart powder, and particularly, the deer heart powder is preferably prepared by cleaning fresh deer hearts with deionized water and cutting the cleaned deer hearts into 2-3 cm squares.
The haematococcus pluvialis composition also comprises 2-100 parts by weight of haematococcus pluvialis, more preferably 10 parts by weight of haematococcus pluvialis, the source of the haematococcus pluvialis is not particularly limited, the haematococcus pluvialis is obtained by adopting a conventional commercial product of the haematococcus pluvialis well known to a person skilled in the art, the haematococcus pluvialis has a remarkable anti-aging effect, the aging of a human body is mainly caused by free radical oxidation, and natural astaxanthin in the haematococcus pluvialis can penetrate through the outer wall of cells of the human body, directly eliminate oxygen radicals in the cells, enhance the regeneration capacity of the cells, protect the health of the cells and DNA (deoxyribonucleic acid), reduce the accumulation of aged cells and play a role in delaying aging.
The composition also comprises 2-100 parts of phytosterol ester, and more preferably 10 parts. The source of the phytosterol ester is not particularly limited in the present invention, and conventional commercial products of phytosterol ester well known to those skilled in the art can be used. The phytosterol ester added in the invention has the effects of protecting the heart, reducing cholesterol and high-density lipoprotein so as to improve coronary atherosclerosis, and in addition, the phytosterol ester also has obvious anti-inflammatory and anti-oxidation effects, and improves the inflammation and oxidative damage of hippocampal nerve cells of senile dementia patients.
The composition also comprises 2-100 parts by weight of spirulina, and more preferably 10 parts by weight of spirulina. The source of the spirulina is not particularly limited in the present invention, and conventional commercially available spirulina products well known to those skilled in the art may be used. The spirulina contains rich spirulina polysaccharide, iron, vitamin B12, chlorophyll, gamma-linolenic acid, linoleic acid and other unsaturated fatty acid substances, can improve the immunologic function of an organism, promote the synthesis and release of excitatory transmitter of nerve cells, regulate the functions of the nerve cells, improve the excitability and the stress of a nervous system, has stronger effects of improving memory and preventing senile dementia, can also increase the oxygen carrying capacity of hemoglobin, improve the blood oxygen saturation, enhance the blood fat metabolism function, obviously reduce the total cholesterol concentration and the arteriosclerosis index of serum, has obvious functions of resisting anoxia and regulating blood fat, and has the auxiliary treatment effect on memory loss and senile dementia.
The composition also comprises 3-130 parts by weight of dihydromyricetin, and more preferably 15 parts by weight of dihydromyricetin. The source of the dihydromyricetin is not particularly limited in the invention, and conventional dihydromyricetin conventional commercial products well known to those skilled in the art can be adopted. Dihydromyricetin is a flavanonol compound and widely exists in Ampelopsis plants in Ampelopsis of Ampelopsis, and researches in recent years find that the dihydromyricetin has wide and remarkable pharmacological effects. Can obviously inhibit the generation of Malondialdehyde (MDA) in the homogenate of the cardiac muscle, the liver and the brain tissue of the rat, and the MDA generation inhibiting effect is increased along with the increase of the concentration of the dihydromyricetin. The dihydromyricetin can obviously inhibit the generation of MDA in the grease, and the antioxidant effect is enhanced along with the increase of the purity (60-90%) of the dihydromyricetin; dihydromyricetin can obviously inhibit the rise of blood sugar of mice induced by alloxan, epinephrine, streptozotocin and the like, and simultaneously, the serum insulin level is raised, the lymphocyte infiltration in pancreatic tissues is obviously reduced, the inflammatory reaction is obviously relieved, the number of pancreatic islets is obviously increased, the serum Triacylglycerol (TG) content of hyperglycemic rats induced by streptozotocin is also obviously reduced, and the dihydromyricetin has obvious functions of reducing blood sugar, resisting inflammation and reducing blood fat, and has auxiliary effects of improving memory and preventing senile dementia.
In the present invention, the dosage form of the composition includes an oral preparation, which includes powder, granules, tablets or pills. The preparation method of the above dosage forms of the present invention is not particularly limited, and conventional methods for preparing powders, granules, tablets and pills, which are well known to those skilled in the art, may be used.
The invention starts from the aspects of improving blood fat, nourishing qi and calming the nerves, regulating cardiac muscle, enhancing blood circulation, resisting oxygen deficiency, oxidation and inflammation, and the like, and has the effects of improving memory and preventing senile dementia, in particular to a compound which takes deer blood, deer heart, haematococcus pluvialis, phytosterol ester, spirulina and dihydromyricetin as raw materials and is formulated according to different proportions, and a large number of screening tests show that the compound of the deer blood, the deer heart, the haematococcus pluvialis, the phytosterol ester, the spirulina and the dihydromyricetin contains sterols, amino acids, vitamins, fatty acids, trace elements and flavonoid compounds, has complementarity in the aspects of improving memory and preventing senile dementia, and has more obvious pharmacological activity compared with single deer blood, the deer heart, the haematococcus pluvialis, the phytosterol ester, the spirulina or the dihydromyricetin after the combination of the two compounds, can improve memory and prevent senile dementia from different angles. The composition disclosed by the invention can achieve the effects of improving memory and preventing senile dementia by regulating nerves, maintaining mind, improving blood fat, enhancing blood circulation, resisting oxidation and inflammation. The composition of the invention embodies the synergistic effect of the chemical components of the traditional Chinese medicine. The composition has the advantages of definite curative effect, no toxic or side effect, convenient taking and scientific formulation.
The invention also provides a preparation method of the composition in the technical scheme, which comprises the following steps:
1) cleaning Cor Cervi, cutting into pieces, lyophilizing, and pulverizing to obtain Cor Cervi powder;
2) dehydrating sanguis Cervi, lyophilizing, and pulverizing to obtain sanguis Cervi powder;
3) sterilizing and drying deer heart powder and deer blood powder to obtain deer heart micro powder and deer blood micro powder;
4) mixing Cor Cervi micropowder, sanguis Cervi micropowder, Haematococcus pluvialis, phytosterol ester, Spirulina and dihydromyricetin to obtain composition;
the step 1) and the step 2) are not limited in chronological order.
The deer heart powder is prepared by cleaning deer heart, cutting into blocks, freeze-drying and crushing. The preparation conditions of the deer heart powder are the same as those described above.
The invention dehydrates, freezes, dries and crushes the deer blood to obtain the deer blood powder. The preparation conditions of the deer blood powder are the same as those described above.
After obtaining the deer heart powder and the deer blood powder, the invention preferentially sterilizes and dries the deer heart powder and the deer blood powder to obtain the deer heart micro powder and the deer blood micro powder.
After obtaining the deer blood micro powder and the deer heart micro powder, the invention mixes the deer heart micro powder, the deer blood micro powder, haematococcus pluvialis, phytosterol ester, spirulina and dihydromyricetin to obtain the composition. In the present invention, the haematococcus pluvialis, phytosterol ester, spirulina and dihydromyricetin are preferably pulverized before being mixed, and are preferably sieved with a 300-mesh sieve after being pulverized. The preparation method is simple and low in cost.
The invention also provides application of the composition in the technical scheme or the composition prepared by the preparation method in the technical scheme in preparation of health-care food or medicine for improving memory and preventing senile dementia.
The composition for improving memory and preventing senile dementia, the preparation method and the application thereof according to the present invention will be described in further detail with reference to the following embodiments, which include but are not limited to the following examples.

The corresponding drugs are administrated to each group by intragastric administration, equal volume of distilled water is administrated to the normal group and the model group, the corresponding drugs are administrated to the administration group for 1 time every day and 28 days continuously, the mice of each group are administrated for 27 days and trained by a diving platform instrument (the mice can find and jump on a safe platform on an electrified flat plate), and the time (latency) required for finding and jumping on the safe platform is measured after administration for 1 hour on the 28 th day; performing water maze training 2 times a day on 23-27 days after drug administration; after the platform-skipping latency is measured on day 28, the Morris water maze is adopted for behavioural detection, the time for finding the platform (escape latency) and the swimming path (total distance) are recorded by the method, and comparison between groups is carried out.
