The invention has the following advantages and positive effects:
1. the corn protein is used as a raw material by utilizing a biological enzyme engineering technology to produce a digestion-free corn protein product with high branched chain amino acid enzyme activity and non-activity, and a branched chain amino acid mouse experiment has prolonged the service life of a mouse by 12 percent and provides a new biological food capable of prolonging the service life of the society. 6. separating the enzymolysis feed liquid by a membrane to remove indigestible protein larger than 500Da, purifying, and performing enzyme deactivation treatment or enzyme deactivation treatment on the feed liquid for 30 seconds at 90 ℃ to obtain high-purity and non-high-purity, enzyme activity and non-active high-branched-chain amino acid indigestible corn protein products;
7. concentrating the four feed liquids, blending according to the standard of liquid products, and aseptically filling to obtain four liquid products;
8. the material liquid of the treatment process 6 is concentrated by a double-effect evaporator to a solid content of more than 40 percent, and then is spray-dried to obtain four powdery products.
Example 2:
1. weighing 200kg of corn protein powder;
2. mixing soybean protein isolate raw material with 8-10 times of water, adding 2000kg of water in the experiment;
3. sterilizing the mixed material liquid at ultrahigh temperature, passivating physiological harmful substances and performing protein denaturation treatment, and keeping the temperature at least 135 ℃ for more than 50 seconds;
4. cooling the material liquid to 54-56 deg.c;
5. adding 0.3-0.5% of compound protease by weight of the raw materials for enzymolysis, wherein 0.6 kg of compound protease is added in the example, the enzymolysis time is 68 hours, and the pH value is not required to be adjusted in the enzymolysis process, but the slow stirring treatment is required;
6. separating the enzymolysis feed liquid by a membrane to remove indigestible protein larger than 500Da, purifying, and performing enzyme deactivation treatment or enzyme deactivation treatment on the feed liquid for 30 seconds at 90 ℃ to obtain high-purity and non-high-purity, enzyme activity and non-active high-branched-chain amino acid indigestible corn protein products;
7. concentrating the four feed liquids, blending according to the standard of liquid products, and aseptically filling to obtain four liquid products;
8. the material liquid of the treatment process 6 is concentrated by a double-effect evaporator to a solid content of more than 40 percent, and then is spray-dried to obtain four powdery products.
Claims (5)
1. The invention relates to a preparation method of a high branched chain amino acid digestion-free corn protein product, which takes corn protein powder as a raw material and is a digestion-free corn protein product containing high branched chain amino acid and finished by enzyme engineering, and the digestion-free corn protein product comprises the following components: the corn protein is a product mainly comprising composite amino acid with the average molecular weight of 120 daltons, small molecular dipeptide and tripeptide with the average molecular weight of 180 to 500 daltons, and the corn protein raw material contains about 22.24 percent of branched chain amino acid and has the highest content in vegetable protein, so the corn protein is used as the raw material to produce a digestion-free protein product with high branched chain amino acid.
2. The process adopts a membrane separation and purification method to separate out protein larger than 500Da to produce a high-purity and high-branched-chain amino acid digestion-free corn protein product, and can also produce the high-branched-chain amino acid digestion-free corn protein product without purification after enzymolysis.
3. The process adopts the treatment of more than 90 ℃ for more than 30 seconds, and needs to inactivate the protease so as to produce the enzyme inactive digestion-free protein product, and can also produce the product containing the protease activity without enzyme deactivation.
4. The feed liquid of the digestion-free protein of the high branched chain amino acid is concentrated and blended according to the standard of liquid products to produce liquid products, and the liquid products are concentrated, sprayed and dried to produce powdery products;
the preparation process comprises the following steps:
①, weighing the corn protein powder raw material;
②, mixing water (①) with the weight of 1: 8-10 times of the weight of the raw material into slurry;
③, carrying out ultra-high temperature sterilization and appropriate protein denaturation treatment on ② mixed slurry, wherein the temperature is at least 135 ℃ and needs to be kept for more than 50 seconds;
④, cooling ③ to reduce the temperature of the feed liquid to 54-56 ℃;
⑤, adding ④ into compound protease accounting for 0.3-0.5 percent of the weight of the raw materials for enzymolysis for 48-72 hours, wherein the pH value does not need to be adjusted in the enzymolysis process, but the raw materials need to be stirred slowly;
⑥, purifying the feed liquid after ⑤ enzymolysis by membrane separation, and carrying out enzyme deactivation treatment or enzyme deactivation treatment on the purified feed liquid at 90 ℃ for 30 seconds to produce a digestion-free corn protein product of high-purity, enzyme activity and inactive high-branched-chain amino acid;
⑦, carrying out enzymolysis on ⑤, and then carrying out enzyme deactivation treatment or enzyme deactivation treatment on the feed liquid at 90 ℃ for 30 seconds without membrane separation and purification to produce a digestion-free corn protein product with enzyme activity and inactive high-branched-chain amino acid;
⑧, concentrating the liquid of ⑥ and ⑦, blending according to the standard of the liquid product, and aseptically filling to obtain the liquid product;
⑨, concentrating ⑥, ⑦ and the feed liquid, and spray drying the solid matter to over 40% to obtain the powdery product.
5. The method of claim 1, 2, 3, or 4 for producing a highly branched chain amino acid digestion-free corn protein product, wherein the method comprises the steps of: taking zein as a raw material, and obtaining the digestion-free zein liquid and powder products of high purity or non-high purity, enzymatic activity and non-activity and high branched chain amino acid by pulp mixing, enzymolysis, membrane separation or non-membrane separation, and enzyme deactivation or non-enzyme deactivation.
