Abstract
本发明公开了一种鸭蛋减产综合征病毒,其编码的VP1蛋白的氨基酸序列如SEQ ID NO:7所示,或者与SEQ ID NO:7所示氨基酸序列的同源性在97.1%以上的氨基酸序列。本发明还公开了一种预防鸭蛋减产综合征的疫苗。本发明的鸭蛋减产综合征病毒,是分离、鉴定获得的一株全新的引发鸭蛋减产综合征的鸭小RNA病毒,利用该病毒研制出的疫苗可有效预防鸭蛋减产综合征。

技术领域
本发明涉及生物工程技术领域,尤其涉及一种鸭蛋减产综合征病毒及其疫苗。
背景技术
中国是一个养禽大国,养禽业迅猛发展,家禽的年饲养量已达120亿羽,居世界首位,我国是世界第一水禽养殖大国和消费大国。虽然近几年水禽饲养量在逐年增长,规模化、产业化经营水平也得到快速提升,但是中国水禽养殖技术相对落后,尤其是水禽传染病的发生,导致水禽养殖的淘汰率高,致使我国水禽产业每年损失高达数百亿元人民币,成为困扰着我国水禽养殖业健康发展的制约因素。
中国的养鸭业面临新发传染性疾病的严重威胁。2016年以来,首先在种鸭场陆续发生了不明原因的产蛋下降,鸭群产蛋率下降20~30%,数周后有一定恢复,但很难恢复到发病前的产蛋率,从而导致种鸭场遭受巨大经济损失。该病的主要症状是蛋鸭产蛋下降,因此将该病命名为鸭产蛋下降综合征。经剖检病鸭,发现此病发病前期肝脏有淤血样变,卵巢和输卵管有不同程度损伤。该病在中国不断蔓延,目前全国主要的蛋鸭生产地区都有该病的暴发。
发明内容
本发明要解决目前缺少有效的鸭蛋减产综合征疫苗、对鸭蛋减产综合征的预防手段有限的技术问题,提供一种新型的鸭蛋减产综合征病毒及其疫苗,该病毒是本发明分离、鉴定的一株全新的鸭蛋减产综合征病毒,利用该病毒研发的疫苗可有效预防鸭蛋减产综合征。
为了解决上述技术问题,本发明通过如下技术方案实现:
本发明提供了一种鸭蛋减产综合征病毒,其5’非编码区的核苷酸序列如SEQ IDNO:1所示或者与SEQ ID NO:1所示的核苷酸序列的同源性在98%以上。
本发明提供了一种鸭蛋减产综合征病毒,其3’非编码区的核苷酸序列如SEQ IDNO:2所示或者与SEQ ID NO:2所示的核苷酸序列的同源性在98%以上。
本发明提供了一种鸭蛋减产综合征病毒,其编码的VP0蛋白的氨基酸序列如SEQID NO:3所示或者与SEQ ID NO:3所示的氨基酸序列的同源性在98%以上。

本发明的一株优选的鸭蛋减产综合征病毒,已于2018年11月22日保藏于中国典型培养物保藏中心(简称CCTCC,地址:湖北省武汉市武昌珞珈山武汉大学保藏中心),保藏号为CCTCC NO:V201866,其分类命名为鸭蛋减产综合征病毒AH204株。
具体实施方式
实施例1病原分离
取鸭蛋减产综合征发病鸭的肝脏、脾脏和肺脏等组织,按1:1(m:v)的量加入灭菌PBS(含100单位青霉素和50ug的链霉素)进行研磨。离心后,取上清,经0.2um滤器除菌后,接种鹅胚。弃掉24h内死亡的鸡胚;收集24h后死亡的鸡胚,收集尿囊液。
实施例2病毒的纯化
将收集到的尿囊液,采用蔗糖密度梯度离心法纯化病毒,操作步骤如下:1)以7500rpm高速离心30min,获取上清,再以10000rpm高速离心30min,获取上清。2)将所得上清以40000rpm超速离心2h,将沉淀用10ml STE溶解。3)先分别在两个38ml的超速离心管中加入5ml所获得的上述经STE溶解的病毒液,然后用长针头从离心管底部依次加入15﹪、30﹪、45﹪和60%的蔗糖溶液,以40000rpm超速离心2.5h,结果可以在30﹪与45﹪之间发现一条清晰的带,吸取此条带的液体至38ml的超速离心管中。4)去蔗糖:用STE缓冲液稀释纯化的病毒,然后以40000rpm超速离心3h,所得沉淀用2mlSTE缓冲液溶解,即为所纯化的病毒液。
实施例3病毒鉴定
3.1病原核酸类型的鉴定
将纯化浓缩的病毒用QIAGEN RNeasy plus mini Kit提取病毒RNA,5ulRNA提取物加1ulRNase(20mg/ul)37℃温育30min;DNase消化:10×buffer 1ul,RNase Inhibitor0.5ul,DNaseⅠ1ul,H20(RNase-free)2.5ul,RNA提取物5ul,37℃温育30min。电泳结果发现,经DNase消化后产物和病毒基因组都可见电泳条带,而经RNase消化的产物无电泳条带。分析可知该病毒为RNA病毒。
3.2病原的PCR鉴定
利用M-MLV反转录酶(Takara公司)和随机引物将病毒RNA反转录,以反转录产物为模板分别利用禽流感病毒、新城疫病毒、网状内皮增生病病毒、I型鸭肝炎病毒、呼肠孤病毒、细小病毒、双RNA病毒的特异性引物进行了PCR扩增,PCR结果均为阴性。
3.3病毒粒子的电镜观察
一定浓度的病毒液经磷酸钨负染,用JEM2100透射电镜进行观察。在此实验结果中可见镜下病毒粒子大小约为30-40nm,椭圆形,无囊膜(图1)。
3.4病毒囊膜的鉴定
取1ml含病毒尿囊液,加入50ul分析纯的氯仿,置于4℃振荡混匀10min,随后500rpm,离心5min,吸取上清,十倍比稀释为10-1,10-2,10-3,10-4,每个滴度取100ul经尿囊腔接种3枚10日龄鸭胚,37℃孵化箱内孵育,观察记录鸭胚死亡情况;对照组的病毒尿囊液中加入相同体积的生理盐水代替氯仿,同样处理和滴定。实验结果显示,氯仿处理病毒后,病毒仍然具有感染能力,因此,此病毒无囊膜。
3.5鸭蛋减产综合征病毒的基因组序列测定
将纯化病毒,用QIAGEN RNeasy plus mini Kit提取病毒的RNA,利用Takara公司的cDNA Larbrary Construction Kit试剂盒构建cDNA文库,从文库中取出部分菌液,涂布于LB/Amp琼脂平板培养基上培养,随机挑选细菌克隆,提取质粒后进行测序,利用Seqman软件(DNASTAR)进行拼接,去除宿主(鸡胚)基因序列后,获得该病毒的基因序列。本发明鸭蛋减产综合征病毒分离株基因组结构示意图如图2所示。其中,鸭蛋减产综合征病毒5’非编码区的核苷酸序列如SEQ ID NO:1所示;鸭蛋减产综合征病毒3’非编码区的核苷酸序列如SEQID NO:2所示;鸭蛋减产综合征病毒VP0蛋白基因的核苷酸序列如SEQ ID NO:4所示,鸭蛋减产综合征病毒VP0蛋白的氨基酸序列如SEQ ID NO:3所示;鸭蛋减产综合征病毒VP3蛋白基因的核苷酸序列如SEQ ID NO:6所示,鸭蛋减产综合征病毒VP3蛋白的氨基酸序列如SEQ IDNO:5所示;鸭蛋减产综合征病毒VP1蛋白基因的核苷酸序列如SEQ ID NO:8所示,鸭蛋减产综合征病毒VP1蛋白的氨基酸序列如SEQ ID NO:7所示;鸭蛋减产综合征病毒2A蛋白基因的核苷酸序列如SEQ ID NO:10所示,鸭蛋减产综合征病毒2A蛋白的氨基酸序列如SEQ ID NO:9所示;鸭蛋减产综合征病毒2B蛋白基因的核苷酸序列如SEQ ID NO:12所示,鸭蛋减产综合征病毒2B蛋白的氨基酸序列如SEQ ID NO:11所示;鸭蛋减产综合征病毒2C蛋白基因的核苷酸序列如SEQ ID NO:14所示,鸭蛋减产综合征病毒2C蛋白的氨基酸序列如SEQ ID NO:13所示;鸭蛋减产综合征病毒3A蛋白基因的核苷酸序列如SEQ ID NO:16所示,鸭蛋减产综合征病毒3A蛋白的氨基酸序列如SEQ ID NO:15所示;鸭蛋减产综合征病毒3B蛋白基因的核苷酸序列如SEQ ID NO:18所示,鸭蛋减产综合征病毒3B蛋白的氨基酸序列如SEQ ID NO:17所示;鸭蛋减产综合征病毒3C蛋白基因的核苷酸序列如SEQ ID NO:20所示,鸭蛋减产综合征病毒3C蛋白的氨基酸序列如SEQ ID NO:19所示;鸭蛋减产综合征病毒3D蛋白基因的核苷酸序列如SEQ ID NO:22所示,鸭蛋减产综合征病毒3D蛋白的氨基酸序列如SEQ ID NO:21所示。
3.6鸭蛋减产综合征病毒分类学
通过研究P1、P2和P3区域氨基酸序列,发现鸭蛋减产综合征病毒与小RNA病毒科各个属中的病毒的同源性都很低(表1),根据国际病毒分类委员会制定的规则,鸭蛋减产综合征病毒属于小RNA病毒科未指定病毒属的病毒。从不同蛋白进化树上分析,鸭蛋减产综合征病毒主要蛋白P1蛋白(图3)在2C蛋白(图4)和3CD蛋白(图5)均独立于小RNA病毒科其他病毒属。该病毒属的病毒是本发明首次报道。

3.7鸭蛋减产综合征病毒进化分析
利用已经获得的鸭蛋减产综合征病毒的基因组序列设计引物,扩增国内不同省份分离到的鸭蛋减产综合征病毒的开放阅读框序列,测序后发现,这些病毒具有相同的基因组结构,各基因核苷酸同源性在97.9-100%之间(表2),对应的氨基酸序列同源性也在98-100%之间,根据病毒分类委员会规定各株病毒分离株的序列虽有差异,但这些病毒均属于同一个种的病毒,这些病毒在核苷酸序列进化树(图6)和氨基酸序列进化树(图7)上显示出高度同源性,因此其生物学特性是相似的。不同病毒分离株的ORF的核苷酸序列测定后,包含在本发明中,分离株GD512序列如SEQ ID NO:23所示,分离株GD519序列如SEQ ID NO:24所示,分离株JS361序列如SEQ ID NO:25所示,分离株SD609序列如SEQ ID NO:26所示。

3.8鸭蛋减产综合征病毒VP1蛋白同源性分析
本发明的VP1蛋白是鸭蛋减产综合征病毒的主要免疫抗原,因此疫苗研制主要与VP1蛋白相关。不同鸭蛋减产综合征病毒分离株的氨基酸序列的同源性在97.1-99.4%之间(表4)。因此,不仅可以利用含有与SEQ ID NO:7一致的鸭蛋减产综合征病毒分离株,也可利用含有与SEQ ID NO:7同源性在98%以上的鸭蛋减产综合征病毒毒株,来研制灭活疫苗、通过传代致弱或人工构建感染性克隆致弱的弱毒疫苗。利用表达载体,表达与SEQ ID NO:7一致的VP1蛋白;或表达与SEQ ID NO:7同源性在98%以上的VP1蛋白;以及表达部分连续的10个以上氨基酸与本发明病毒序列SEQ ID NO:7一致的氨基酸序列来研制DNA疫苗、病毒载体疫苗(包括副粘病毒载体、痘病毒载体、腺病毒载体、疱疹病毒载体、杆状病毒等)、真核表达多肽疫苗、原核表达多肽疫苗和合成肽疫苗。

实施例4病毒的致弱
将含鸭蛋减产综合征病毒尿囊液,10000rpm离心45min,取其上清用于病毒致弱。
4.1细胞致弱
1)取100ul病毒液加到生长良好的Vero细胞上,放于37℃细胞培养箱内吸附1~2h,随后用PBS洗一次,洗掉未吸附的病毒,重新加入新鲜培养液置于37℃含5﹪CO2的细胞培养箱内进行培养。2)48h后吸取细胞上清同样方法使病毒吸附于另一生长良好的Vero细胞,放于37℃含5﹪CO2的细胞培养箱内进行培养。3)连续传代至第40代后收获含病毒的细胞上清液,稀释至10-1、10-2、10-3、10-4,每个滴度接种三枚9日龄鸭胚,放于37℃孵化箱内孵育,弃掉24h内死亡的鸭胚;除去非特异性死亡的鸭胚,收集死亡鸭胚,计算ELD50,结果表明经过在Vero细胞上传代40次后病毒被明显的致弱。
4.2鸭胚致弱法
1)将病毒液上清稀释100倍,经尿囊腔接种10枚10日龄鸭胚,放于37℃孵化箱内孵育,弃掉24h内死亡的鸭胚,除去非特异性死亡的鸭胚,收集死亡鸭胚。2)取死亡鸭胚尿囊液,按上述同样的方法再次接种10枚9日龄鸭胚,放于37℃孵化箱内孵育,弃掉24h内死亡的鸭胚,除去非特异性死亡的鸭胚,收集死亡鸭胚,连续传代60代以上。结果表明经过在鸭胚上传代60次后病毒明显适应鸭胚。
4.3病毒致病力实验
收集细胞上清或尿囊液,感染26周龄产蛋鸭,检验病毒致弱情况。分为A、B和C三组,每组10只。A组:104TCID50细胞致弱病毒100ul经肌肉注射感染,正常饲养;B组:104ELD50鸭胚致弱病毒100ul经肌肉注射感染,正常饲养;C组接种100ul灭菌PBS作为对照。两天后所有鸭子采食量正常,未见精神沉郁,A组、B组和C组产蛋量没有明显差异。攻毒后第四天每组分别剖检三只,A组和B组鸭子肝脏未见明显的病变;脾脏不肿大。病毒滴定未在接种鸭体内发现病毒,表明病毒明显致弱了。
4.4弱毒疫苗研制
将上述致弱的病毒在相应的鸭胚或细胞上增殖后,离心去除杂质,与保护剂混合后分装,在冻干机上冻干,用于研制弱毒疫苗。按《兽用生物制品标准》进行弱毒疫苗研制。
实施例5灭活疫苗的制备
5.1病毒增殖
将含鸭蛋减产综合征病毒尿囊液,用灭菌PBS稀释十倍经尿囊腔接种40枚10日龄鸭胚,放于37℃孵化箱内孵育。弃掉24h内死亡的鸭胚;接种后144h,鸭胚全部死亡;收集尿囊液。
5.2病毒含量测定
将含鸭蛋减产综合征病毒尿囊液,用灭菌PBS作10倍系列稀释,取10-2、10-3、10-4、10-5、10-6五个稀释度分别经尿囊腔内接种10日龄鸭胚,每个滴度接种3枚胚,每胚0.lmL,置37℃继续孵育,24小时前死亡的胚弃去不计,将48~120小时死亡的鸭胚及时取出,逐个检查胚体,去除非特异性死亡胚,计算ELD50,结果见表5。

5.3病毒灭活
5.3.1灭活试验将收集到的含鸭蛋减产综合征病毒尿囊液,加入终浓度为2‰的甲醛,然后放于37℃灭活24h(以瓶内温度达到37℃开始计时),在灭活期间振摇3~4次,到时取出,置于4℃保存备用。
5.3.2灭活效果检验:取10日龄鸭胚10枚,每枚经尿囊腔接种灭活病毒液0.2ml,每天照蛋2次,连续观察6天,鸡胚无特异性死亡,144小时后对所有鸭胚进行检查,均无明显病变。
5.4疫苗制造方法
取油佐剂相放于胶体磨或乳化缸内,开动电机慢速转动搅拌,同时徐徐加入水相,加完后再以10000rpm搅拌2分钟,在终止乳化前加入1%硫柳汞溶液,使其最终浓度为0.01%,即成乳白色的油乳剂灭活疫苗。按相关内容进行质量检验。
5.5疫苗安全性试验
针对每批次疫苗试验2个不同鸭种组,28周龄蛋鸭、4周龄雏鸭各10只,随机分组。对于26周龄蛋鸭、4周龄雏鸭,采用颈部皮下注射该油乳剂灭活苗2mL/羽;另外,设置对照组,颈部皮下注射生理盐水2mL/羽,观察30天,有无体温升高,精神不振,食欲减退及其他异常反应,结果见表6。

5.6免疫效力试验
1)26周龄蛋鸭的免疫效力试验,每羽颈部皮下注射0.3mL。试验情况见表7。

5.7最小免疫剂量测定
用26周龄蛋鸭和4周龄雏鸭以0.lmL,0.2mL,0.3mL,0.5mL,1mL不同的剂量颈部皮下接种,SPF鸭每组10只,4周龄雏鸭每组40只。对照组每组均为10只。21天采血后攻毒,攻毒后5天分离病毒。结果是,最小免疫剂量为0.2mL。
实施例6DNA疫苗研制
6.1VP1蛋白真核表达质粒pCAGGs-VP1的构建与鉴定
根据与SEQ ID NO:7所示的鸭蛋减产综合征病毒VP1蛋白氨基酸序列,通过密码子优化,使其更适合鸭体表达,在密码子优化时,在核苷酸序列前加上Kozak序列(GCCACC)和ATG、同时在首尾分别加上Hind III和Xho I酶切位点,将优化合成的序列通过Hind III和Xho I酶切位点插入到pCAGGs载体。连接体系如下:10×buffer 1ul,T4DNA连接酶1ul,酶切处理载体pCAGGs和PCR产物各1和7ul,室温连接2h。将这VP1蛋白基因的连接产物分别转化DH5α感受态细胞,并在含有氨苄青霉素的LB平板上37℃培养。16-20h后,挑取单个菌落,抽提质粒DNA进行酶切鉴定(Hind III和Xho I)。酶切产物在1.0%琼脂糖电泳观察。结果可见,重组质粒双酶切后能切出大小为530bp左右的条带。然后将pCAGGs-VP1重组质粒送测序。测序结果证实,所插入pCAGGs载体的VP1优化序列无误。这些结果证明pCAGGs-VP1重组质粒已构建成功。
6.2VP1基因DNA疫苗的研制
将含有pCAGGs-VP1重组质粒的DH5α大肠杆菌,接种于盛有3ml LB液体培养基的试管中,37℃过夜培养。菌液离心弃上清,菌体沉淀用OMEGA公司的去内毒素试剂盒进行超纯质粒抽提。电泳检测,并用Nanodrop 2000c紫外可见光分光光度计测量OD260、OD280,估算DNA含量与纯度。按上述操作获得的pCAGGs-VP1质粒用0.85%生理盐水稀释到1mg/ml,即为鸭蛋减产综合征DNA疫苗。
6.3鸭免疫保护实验
6.3.1免疫方法30只鸭随机分为3组,每组10只:1)重组质粒pCAGGs-VP1组;2)载体质粒pCAGGs组;3)空白对照组。用基因导入仪在鸭腿部肌注0.2ml(1mg/ml)质粒,1次/2周,连续6周。分别于末次加强免疫后两、三周、四周后取鸭血清。
6.3.2抗病毒保护率的测定鸭接种3次DNA疫苗后28天,各个实验组静脉注射接种104ELD50鸭蛋减产综合征病毒。每天观察并记录鸭的死亡、发病情况。攻毒后4天,剖杀所有鸭,检测各脏器中病毒。结果见表8,重组质粒pCAGGS-VP1免疫组攻毒后病毒分离率为0,表明鸭蛋减产综合征病毒DNA疫苗对鸭有一定的免疫保护,可作为DNA疫苗使用。

结论,表达VP1蛋白的DNA疫苗可以预防鸭蛋减产综合征病毒,表明VP1蛋白是鸭蛋减产综合征病毒的主要保护性抗原,可通过病毒载体表达VP1研制载体疫苗;真核表达或原核表达的VP1蛋白可用于研制亚单位。
以上所述实施例仅表达了本发明的实施方式,其描述较为具体和详细,但并不能因此而理解为对本发明专利范围的限制。应当指出的是,对于本领域的普通技术人员来说,在不脱离本发明构思的前提下,还可以做出若干变形和改进,这些都属于本发明的保护范围。因此,本发明专利的保护范围应以所附权利要求为准。
