Test example 4
Mouse anti-inflammatory assay (mouse ear swelling assay):
(1) subjects and groups:
60 KM male mice, weighing 18-22g, were randomly divided into 6 groups. The anti-inflammatory efficacy was compared using the clear water group as a blank group, the effective ingredients in comparative examples 1 and 2 as control groups 1-2, and the effective ingredients in examples 1-5 as experimental groups 1-5.
(2) Experimental procedure
Dipping 20 mul of liquid medicine of the experimental group and the control group of the drug on the front and back surfaces of the right ear of each mouse by a cotton swab, coating distilled water (20 mul/mouse) on the front and back surfaces of the right ear of each mouse in the blank group, dipping 20 mul of xylene with the cotton swab with the volume of about 100 percent on the front and back surfaces of the right ear of each mouse, uniformly and rapidly coating the xylene with the cotton swab on the front and back surfaces of the right ear of each mouse, recording the drug coating and sensitization time of each mouse, and repeatedly using the drug 0.5 h, 1.5 h and 2.5h after inflammation. Killing the animals 4h after inflammation, cutting two ears along the base line of auricle, punching the ears of the two ears by using a trephine with diameter of 6mm, measuring the thickness of the two ears, and taking the difference of the thicknesses of the two ears as swelling degree (mm); the swelling inhibition (%) was calculated as the difference in thickness (mm) between the two ear pieces, and the calculation formula was as follows:
swelling degree (mm) ═ right ear thickness (mm) -left ear thickness (mm)
The swelling inhibition ratio (%) - (blank group ear thickness difference (mg) — sample group ear thickness difference (mg))/blank group ear thickness difference (mg) × 100%.
The results of the mouse anti-inflammatory efficacy test are shown in the following table:
group of samples | Swelling degree (mm) | Swelling inhibition ratio (%) |
Example 1 | 0.207±0.050 | 81.38 |
Example 2 | 0.189±0.043 | 83.00 |
Example 3 | 0.139±0.071 | 87.50 |
Example 4 | 0.133±0.069 | 88.04 |
Example 5 | 0.110±0.055 | 90.10 |
Comparative example 1 | 0.890±0.036 | 19.96 |
Comparative example 2 | 0.834±0.088 | 25.00 |
Blank group | 1.112±0.061 | / |
As can be seen from the experimental results, the difference in ear thickness and swelling degree of the mice in examples 1 to 5 were significantly smaller than those of the control group and the blank group, indicating that the effective ingredient can improve the anti-inflammatory effect, with the anti-inflammatory effect of example 5 being the best example.
