Abstract
The invention discloses a whitening composition containing ginseng ferment, which comprises, by mass, α -arbutin 5% -15%, nicotinamide 50% -90% and ginseng ferment 5% -15%.
Description
Technical Field
The invention relates to the technical field of cosmetics, and particularly relates to a whitening composition containing ginseng ferment and application thereof.
Background
Since ancient times, Asian women, especially oriental women, have a beautifying concept of 'white covering ugly', and until now, whitening is still one of the hottest requirements of the makeup industry, and whitening-related skin care products have become one of the mainstream of efficacy cosmetics.
The skin color of a human body is determined by intricate and complex combinations of various factors such as the physiological surface state of the skin, blood flow, moisture, and pigment, and the most important factor among these factors is melanin. Melanin is a high molecular biological pigment mainly distributed in epidermal cells and keratinocytes of the human body, and when melanin is excessively accumulated, it causes problems such as spots and freckles on the skin.
Melanin is synthesized by melanocytes and is metabolized as follows: tyrosine forms dopa under the action of tyrosinase, then the dopa is converted into dopaquinone, and the dopaquinone is subjected to a series of oxidative isomerization and polymerization reactions to finally form melanin. After melanin is formed, it is loaded by melanosomes and migrates toward surrounding keratinocytes along the dendritic projection ends extended from melanocytes, thereby being taken up by the keratinocytes. As the skin is cornified, melanin is pushed to the surface of the skin and is discharged from the skin with the exfoliation of the stratum corneum, and melanin completes the entire process from production to metabolism. The influence of the target object on the skin color of a human body, namely the whitening effect of the target object can be judged by blocking the formation of melanin or any stage of the delivery process and measuring the change of the melanin content (namely the melanin inhibition rate) caused by the blocking.
Phenylthiourea (PTU) is one of the best known tyrosinase inhibitors. The sulfur atom of PTU is combined with two copper ions of the active site of tyrosinase, and the activity of the enzyme is blocked, so that the synthetic process of melanin is cut off, and the melanin content is effectively reduced. Therefore, PTU is commonly used as a reference substance for skin whitening raw materials to measure the whitening efficacy of an object.
In addition to indirectly judging the whitening efficacy of a target substance by the melanin inhibition rate, it is a more direct judgment method by measuring the change in skin brightness. The Conicamidlata CR-400 chromatism meter is a certified successor of CR-100, CR-200 and CR-300 series, can be used for detecting the chromaticity of skin, expresses the whiteness of the skin by an L value, and measures the whitening effect of a target by a dL value (the difference of the L values after and before the treatment of the target). In addition to the data evidence of dL values, CR-400 also provides photographs of the skin to visualize changes in skin color.

The whitening efficacy was tested according to the composition and proportions of example 1:
the first, experimental principle:
to verify the whitening efficacy of α -arbutin/niacinamide/ginseng ferment composition, we employed the following experimental method:
1. culturing recombinant Asian tanning skin epidermal layer cells, namely performing different treatment blank groups (without adding any test substance), solvent groups (adding a solvent for dissolving α -arbutin/nicotinamide/ginseng ferment composition), PTU groups (adding PTU) and α -arbutin/nicotinamide/ginseng ferment compositions on the recombinant skin cells, measuring the melanin content in the melanin cells of each group after culturing the cells for several days, calculating the melanin inhibition rate between each treatment group and the blank groups to show the whitening effect of each treatment substance, if α -arbutin/nicotinamide/ginseng ferment compositions are compared with the blank groups, the melanin inhibition rate data show a significant difference, that is, α -arbutin/nicotinamide/ginseng ferment compositions can reduce the generation of the total melanin amount and can be considered to have a significant whitening effect, if α -arbutin/nicotinamide/ginseng ferment compositions are compared with the blank groups, α -arbutin/nicotinamide/ginseng ferment compositions can further verify the whitening effect of the arbutin/ginseng ferment compositions by adopting Nicotiana CR-400 to measure the color difference of the skin color difference, and further verify the skin whitening effect of the skin by comparing the data of the α -arbutin/nicotinamide/ginseng ferment compositions with the blank groups 35dL, and calculating the skin whitening effect data show a significant difference, if the skin whitening effect of the skin is calculated by adopting a comparison result of the nikojicamb-arbutin/nicotinamide/ginseng enzyme compositions which is poor, and the skin is further, and the skin whitening effect of the skin is calculated by adopting a comparison of α, and the nikojic score of the;
3) the blank group, the solvent group, the PTU group and the α -arbutin/nicotinamide/ginseng ferment composition are photographed and compared in a mode of directly photographing by a Conicamideak CR-400 color difference meter, and the whitening effect of the α -arbutin/nicotinamide/ginseng ferment composition is judged through the visual perception of the photograph.
Second, Experimental methods
1. Cell culture and processing
The 3-D human epidermal layer recombinant skin containing human keratinocytes and melanocytes was placed on a sterilized agarose plate, seeded in a 6-well cell plate filled with 1.5ml of a medium, cultured overnight at 37 ℃ under 5% CO2, and the state of the cells was maintained as required (a blank well was set, only recombinant skin of the corresponding concentration was added to the blank well) for 24 hours, then 25ul of the test substance (including a separate solvent control, phenylthiourea of 3mM concentration and α -arbutin/nicotinamide/ginseng ferment composition of the corresponding concentration) diluted with PBS/20% ethanol solution was added to each of the other cell plates except the blank well, and the incubation was continued (37 ℃, 5% CO2), and the above operations were repeated on days 3 and 6, and the medium was changed.
2. Melanin content detection
After the recombinant skin is incubated for 8 days, the skin culture is collected respectively and placed in a 1.5ml centrifuge tube, 360 mul of melanin extracting solution is added, water bath is carried out for 45 minutes at 100 ℃, then 150 mul of solution is absorbed from each centrifuge tube twice and transferred into a 96-well plate, and the absorbance value at 490nm is measured by an enzyme-linked immunosorbent assay.
3. Calculation of melanin inhibition ratio
Melanin inhibition (%) was 100% (relative value of melanin content in sample/relative value of melanin content in blank control group)
4. Measurement of L value of recombined skin by Konika minolta CR-400 chromatism meter
The CR-400 colorimeter needs to be calibrated before testing. For the blank and cultured reconstituted skin after treatment with each treatment agent, the L values were tested on day 1 and day 8, respectively, and dL values and dL control rates were calculated.
dL ═ L8 (day 8L value) -L1 (day 1L value)
dL control (sample dL value/blank dL value) 100%
5. Photographing contrast of Konika minolta CR-400 color chromatometer
For the change of the recombinant skin before and after the treatment of each treatment agent, except for the comparison and the test of the L value, the change is photographed at the same time for visual comparison.
The experimental result of the melanin inhibition effect is shown in fig. 1, and compared with the blank group, the melanin inhibition rate of the 0.1% α -arbutin/1% nicotinamide/0.1% ginseng ferment composition is significantly different, and the melanin inhibition effect of the composition is close to that of a famous tyrosinase inhibitor PTU.
The dL control rate is shown in fig. 2, and the dL control rate of the recombinant skin cultured after the treatment of 0.1% α -arbutin/1% nicotinamide/0.1% ginseng ferment composition is very different from that of the blank group, which indicates that the recombinant skin has very significant skin whitening effect and is superior to the famous tyrosinase inhibitor PTU.
The recombined skin cultured after the blank group and each treatment agent are compared by photographing through a Conicamidta CR-400 chromatism meter, and the recombined skin cultured after the α -arbutin/nicotinamide/ginseng ferment composition is treated intuitively shows the obvious skin whitening effect.
