背景技术
胆碱类药物主要是针对胆碱受体、乙酰胆碱酯酶而开发的药品,分为拟胆碱药物、抗胆碱药物两大类。拟胆碱药物可用于青光眼、肠麻痹、血管痉挛性疾病等治疗;抗胆碱药物可用于抢救感染中毒性休克,解除有机磷农药中毒,阿斯综合症和内脏绞痛,也可用于麻醉前给药、散瞳或治疗角膜炎、虹膜炎等。
磷酰胆碱吗啉是合成胆碱类药物的中间体,不仅在生产前需要对其进行质量控制,而且在生产后也要监控其是否残留。然而目前并没有磷酰胆碱吗啉检测方法的相关研究。因此亟需开发磷酰胆碱吗啉的检测方法。
本方法简单便捷,可快速测定磷酰胆碱吗啉含量,具有较高的准确性、稳定性和分离度。
发明内容
本发明的目的是提供一种十八烷基键合相的色谱柱分离、蒸发光检测的高准确性、稳定性和分离度的磷酰胆碱吗啉的高效液相色谱检测方法。
为了实现本发明的目的,本发明的技术方案如下:试样经流动相溶解并定容,过膜后注入高效液相色谱仪,在设置仪器条件下采集色谱图,外标法定量;十八烷基键合相的色谱柱分离,柱温30~40℃,体积比为4/96~10/90的甲醇和水为流动相进行等度洗脱,柱流量0.7~1.0mL/min,进样体积10~20μL,蒸发光检测器检测。
有益效果:本发明采用十八烷基键合相的色谱柱分离,蒸发光检测,可快速测定磷酰胆碱吗啉含量,具有较高的准确性、稳定性和分离度,且方法简单便捷。

实施例一
采用的高效液相色谱仪,配蒸发光检测器。
精密称取磷酰胆碱吗啉待测样品0.1g(精确0.001g),用流动相溶解并定容至50mL。采用的色谱条件为:ODS-3色谱柱;40℃柱温;甲醇和水为流动相等度洗脱且体积比为4/96:1.0mL/min流速:10μL进样体积:蒸发光检测器检测。将样品溶液过膜后注入高效液相色谱仪,记录色谱图。4.19min为磷酰胆碱吗啉谱峰,保留时间适宜且分离度高,峰形对称性良好。

1.一种磷酰胆碱吗啉的高效液相色谱检测方法,其特征在于,试样经流动相溶解并定容,过膜后注入高效液相色谱仪,在设置仪器条件下采集色谱图,外标法定量;十八烷基键合相的色谱柱分离,柱温30~40℃,体积比为4/96~10/90的甲醇和水为流动相进行等度洗脱,柱流量0.7~1.0mL/min,进样体积10~20μL,蒸发光检测器检测。
