Description
Technical Field
The invention belongs to the field of medicine, health-care food and food processing, and particularly provides a preparation method of high-activity rhizoma polygonati paste.
Background
Polygonatum sibiricum (Polygonatum sibiricum) is a perennial herb of Liliaceae, and the root and stem of Polygonatum sibiricum are used as medicines, and has been recorded in Ming Dynasty's famous medical records for more than two thousand years. The sealwort has neutral nature and sweet taste, enters spleen, kidney and lung channels, has the effects of tonifying kidney and replenishing essence, relieving epigastric distention and benefiting vital energy, nourishing yin and moistening dryness, promoting the production of body fluid and tonifying spleen and the like, and is long-term applied to the treatment of symptoms such as weakness of spleen and stomach, deficiency of the kidney and deficiency, lung deficiency and dry cough, tiredness and hypodynamia and the like. Modern chemical component analysis shows that the main effective components of rhizoma polygonati include steroid saponin, polysaccharide, oligosaccharide, triterpenoid saponin, flavone and the like. Rhizoma Polygonati is listed in list of articles issued by Ministry of health, which are food and medicine, and has high safety.
At present, the method for processing polygonatum sibiricum at home and abroad mainly comprises the following methods: firstly, rhizoma polygonati is dried and then crushed into particles to be drunk as tea bags; secondly, processing rhizoma polygonati into prepared rhizoma polygonati by adopting a steaming and drying combined method, and using the rhizoma polygonati as an instant food; thirdly, polysaccharide or flavonoid ingredients in the sealwort are singly separated by using an organic solvent and a resin adsorption method, and the sealwort is used for producing medicines. The processing method has the problems of low extraction or conversion degree of active ingredients and nutrient substances of the polygonatum, backward technical method, low automation degree, inconvenient eating mode of products and the like, the due medicinal value and health care effect of the products are difficult to ensure, and the large-scale production and standardized production of the polygonatum are urgently solved to promote the upgrading of the polygonatum industry.

(2) carrying out enzyme hydrolysis conversion reaction on rhizoma polygonati: keeping the temperature of the rhizoma polygonati extract in the temporary storage tank to 25-40 ℃, adding fresh rhizoma polygonati blocks accounting for 1-5% of the weight of the extract, and performing enzyme hydrolysis conversion reaction on the rhizoma polygonati by using natural biological enzyme existing in rhizoma polygonati tubers and keeping the temperature for 1-4 hours at 25-40 ℃;
(3) filtering with a microporous membrane: conveying the extracting solution at high pressure and passing through a microporous membrane filter, wherein the pressure of a conveying pump is 0.3-0.5 MPa, and conveying the filtrate to a vacuum reduced pressure concentrator;
(4) and (3) concentrating under reduced pressure: controlling the parameters of a vacuum decompression concentrator, and carrying out decompression concentration on the sealwort conversion extract until the content of solid matters is 30-35% Brix (w/w), wherein the concentration temperature is 60-75 ℃, and the vacuum degree is-0.06-0.08 Mpa;
(5) and (3) secondary conversion of active ingredients of rhizoma polygonati: placing sealwort concentrated solution with the solid content of 30-35% Brix (w/w) in a scraper type concentration tank, adjusting the temperature to 85-90 ℃, stirring at normal pressure and keeping the temperature for 8-12 h, starting a vacuum pump, adjusting the vacuum degree to-0.05-0.07 Mpa, and carrying out reduced pressure concentration; repeating the steps for a plurality of times until the solid content is 60-65% Brix (w/w), and finishing concentration to obtain the high-activity rhizoma polygonati paste.
As a preferred technical scheme:
in the step (1), the slice thickness of the rhizoma polygonati is 0.5-1 cm.
In the step (2), the size of the fresh rhizoma polygonati blocks is a cube or a cuboid with the side length of 0.5-1 cm.
In the step (3), the filtering pore diameter of the microporous membrane filter is 1-5 μm.
In the step (5), the stirring and heat preservation under normal pressure and the concentration under reduced pressure are carried out for three times, the first concentration under reduced pressure is carried out until the solid content is 40-45% Brix (w/w), the second concentration under reduced pressure is carried out until the solid content is 45-50% Brix (w/w), and the third concentration under reduced pressure is carried out until the solid content is 60-65% Brix (w/w).
The Polygonatum kingianum of the invention comprises dried rhizome of Polygonatum kingianum Coll. et Hemsl, Polygonatum-tum sibiricum Red. or Polygonatum cyrtonema Hua of Liliaceae and processed product thereof.
The invention creatively uses the active enzyme hydrolysis reaction technology, the dynamic circulation extraction technology and the secondary conversion technology of the active ingredients of the sealwort, the processing technology is more advanced, the automation control degree of the production of sealwort products is greatly improved, the physical and chemical index stability of the products is increased, and the product quality is obviously improved. The invention optimizes the preparation process parameters of the high-activity rhizoma polygonati paste, defines the reaction temperature and time of natural rhizoma polygonati enzyme on rhizoma polygonati polysaccharide, steroid saponin, protein, starch and other substances, converts high-activity components such as rhizoma polygonati polysaccharide, steroid saponin and the like to the maximum extent, effectively improves the content of the high-activity components rhizoma polygonati polysaccharide and rhizoma polygonati steroid saponin in the rhizoma polygonati paste, wherein the content of the rhizoma polygonati polysaccharide is more than or equal to 6 percent, the content of the rhizoma polygonati steroid saponin is more than or equal to 1.0 percent, and the content of the active components in the rhizoma polygonati paste is improved by more than 30 percent compared. The invention overcomes the defects of extensive steaming and drying method, fussy manual manufacturing procedure, low quality standardization degree and the like in the traditional processing method of the rhizoma polygonati, and the implementation of the invention is beneficial to improving the additional value of the rhizoma polygonati industry and promoting the development of modernization of traditional Chinese medicine processing.
