Abstract
The invention belongs to the field of cosmetics, and discloses a composition with an antibacterial effect, which comprises the following components in parts by weight: 0.001-8 parts of nisin, 0.001-5 parts of polylysine, 0.001-5 parts of cordycepin, 0.001-2 parts of lysozyme, 0.1-20 parts of 1, 2-hexanediol, 0.01-1 part of p-hydroxyacetophenone, 60-90 parts of water and 0.1-5 parts of pH regulator. Compared with the prior art, the composition prepared by the invention has good antibacterial and bacteriostatic effects through the proper selection of nisin, polylysine and cordycepin and the proper selection of the dosage, and particularly has good antibacterial and bacteriostatic effects on heat-resistant coliform, pseudomonas aeruginosa, white streptococcus, aspergillus niger and saccharomycetes, so that the composition can replace the traditional preservative when being applied to skin care products, so that the potential harm to the skin caused by the conventional preservative is avoided, and the safety of the product is improved.
Description
Technical Field
The invention belongs to the field of cosmetics, and particularly relates to a composition with an antibacterial effect and application thereof in a skin care product without additives.
Background
In the field of cosmetics, formulators add preservatives to inhibit the growth and proliferation of microorganisms in cosmetics, and to extend the shelf life of cosmetics. Traditional preservatives are classified into formaldehyde and formaldehyde donors, parabens, isothiazolinones, phenols and acids, and the like. The traditional preservative is used for a long time, so that the skin of a human body is adversely affected, for example, formaldehyde and formaldehyde donors have the risk of carcinogenesis, parabens can react with ultraviolet rays to cause skin aging, isothiazolinone has the risk of sensitization and the like. How to reduce or even eliminate the use of these preservatives, avoid potential hazards and concerns, and ensure the shelf life of cosmetics is one of the product trends in the cosmetics industry.
Disclosure of Invention
Aiming at the defects of the prior art, the invention provides a composition with an antibacterial effect and application thereof in a non-additive skin care product. The composition has obvious bacteriostatic effect on gram-positive bacteria, gram-negative bacteria and heat-resistant coliform bacteria. The composition is safe when added into cosmetics, and can ensure long shelf life of the cosmetics.
In addition, the invention also provides a preparation method of the composition with the antibacterial effect.

Preferably, the water is deionized water subjected to sterilization treatment.
Preferably, the cordycepin is derived from an extract of Cordyceps sinensis.
The molecular weight of the polylysine is between 3600-4300, the antibacterial activity of the polylysine is the best, and when the molecular weight is lower than 1300, the polylysine loses the antibacterial activity.
Preferably, the pH adjusting agent is citric acid.
Preferably, the pH of the composition is 4 to 6.5; further preferably, the pH of the composition is 5.2 to 5.8.
A method for preparing a composition with antibacterial effect comprises the following steps:
(1) weighing the components according to the formula ratio, adding a pH regulator into water, and regulating the pH to 4.0-6.0 to prepare a mixture A for later use;
(2) mixing p-hydroxyacetophenone and 1, 2-hexanediol, heating to 40-50 ℃, stirring, and then cooling to 15-35 ℃ to obtain a mixture B for later use;
(3) adding the mixture B prepared in the step (2) into the mixture A prepared in the step (1), uniformly stirring, adding nisin, polylysine, cordycepin and lysozyme, and stirring for dissolving until the prepared mixture is transparent, thus preparing the composition with antibacterial effect.
Preferably, the pH is adjusted to 5.2-5.8 in step (1).
Preferably, the temperature in step (2) is raised to 40-45 ℃.
Preferably, the temperature in step (2) is as hereinbefore described at 25-35 ℃.
The skin care product comprises the composition, and the composition accounts for 0.5-5% of the skin care product in percentage by mass.
When the composition disclosed by the invention is added into a skin care product, the skin care product does not need to be added with a traditional preservative, so that the skin care product is called an additive-free skin care product.
Compared with the prior art, the invention has the following beneficial effects:
(1) the composition disclosed by the invention has good antibacterial and bacteriostatic effects by optimizing nisin, polylysine, cordycepin and lysozyme and optimizing the dosage, is added into cosmetics, does not cause adverse effects on skin after being used for a long time, is very safe, and can also play roles in conditioning skin and benefiting skin health.
(2) The polylysine also has the effects of supplementing skin nutrition and conditioning skin, and the 1, 2-hexanediol also has good moisturizing effect and solvent effect.
(3) The composition has remarkable antibacterial and bacteriostatic effects on heat-resistant coliform bacteria, pseudomonas aeruginosa, white streptococcus, aspergillus niger and saccharomycetes, so that the composition can be applied to skin care products to replace the traditional preservative, so that the potential harm to skin caused by the conventional preservative is avoided, and the safety of the product is improved. The composition is added into cosmetics, and the shelf life of the cosmetics can be as long as 3 years.

In order to illustrate the technical solution of the present invention, the following examples are given for illustration. It should be noted that the following examples are not intended to limit the scope of the claimed invention.
Example 1
The composition with the antibacterial effect comprises the following components in parts by weight:
the molecular weight of the polylysine is 3900-4000.
The cordycepin is derived from Cordyceps sinensis extract.
The pH regulator is citric acid.
A method for preparing a composition with antibacterial effect comprises the following steps:
(1) weighing the components according to the formula ratio, adding a pH regulator into water, and regulating the pH to 5.2 to prepare a mixture A for later use;
(2) mixing p-hydroxyacetophenone and 1, 2-hexanediol, heating to 40 ℃, stirring, and then cooling to 30 ℃ to prepare a mixture B for later use;
(3) adding the mixture B prepared in the step (2) into the mixture A prepared in the step (1), uniformly stirring, adding nisin, polylysine, cordycepin and lysozyme, and stirring for dissolving until the prepared mixture is transparent, thus preparing the composition with antibacterial effect.

(3) adding the mixture B prepared in the step (2) into the mixture A prepared in the step (1), uniformly stirring, adding nisin, polylysine, cordycepin and lysozyme, and stirring for dissolving until the prepared mixture is transparent, thus preparing the composition with antibacterial effect.
Example 3
The composition with the antibacterial effect comprises the following components in parts by weight:

(3) adding the mixture B prepared in the step (2) into the mixture A prepared in the step (1), uniformly stirring, adding nisin, polylysine, cordycepin and lysozyme, and stirring for dissolving until the prepared mixture is transparent, thus preparing the composition with antibacterial effect.
Comparative example 1
In comparison with example 2, comparative example 1 produced a composition in which poly-arginine was substituted for poly-lysine. The remaining components and preparation were the same as in example 2.
Comparative example 2
Compared with example 2, the composition prepared in comparative example 2 does not contain cordycepin. The remaining components and preparation were the same as in example 2.
Comparative example 3
In contrast to example 2, the composition prepared in comparative example 3 did not contain nisin. The remaining components and preparation were the same as in example 2.
Comparative example 4
Compared with the example 2, the composition prepared in the comparative example 4 comprises 0.001 part of nisin, 10 parts of polylysine and 0.001 part of cordycepin, and the rest of the components and the preparation process are the same as those in the example 2.
Comparative example 5
In contrast to example 2, comparative example 5 produced a composition in which 1, 2-octanediol was used instead of 1, 2-hexanediol. The remaining components and preparation were the same as in example 2.
Product effectiveness testing
The heat-resistant coliform group, the pseudomonas aeruginosa, the streptococcus albus, the aspergillus niger and the yeast are respectively put into corresponding culture media (sold in the market) to be cultured at a proper temperature (as shown in the table 1), and a colony which can be seen by naked eyes (namely, the area corresponding to the initial colony) is formed.

then, the compositions prepared in examples 1 to 3 and comparative examples 1 to 5 and distilled water were sprayed on colonies of heat-resistant coliform bacteria, pseudomonas aeruginosa, streptococcus albus, aspergillus niger and yeast in equal amounts, and the area of each colony was counted after 1 day, and the antibacterial ratio (antibacterial ratio [ initial colony area-area of colony counted after 1 day ]/initial colony area 100%) was calculated and the results are shown in table 2.

as can be seen from table 2, the antibacterial rate is negative when the antibacterial test is directly performed with distilled water, which indicates that the distilled water cannot exert the antibacterial effect. As can be seen from the results of the antibacterial ratio of examples 1-3 and comparative examples 1-5, the composition prepared according to the technical scheme of the invention has the best antibacterial effect. The combined use of cordycepin, nisin and polylysine and the appropriate choice of the amounts have an important influence on the antibacterial effect of the composition. In addition, as can be seen from the results of comparative example 5, the present invention also has a certain selectivity to 1, 2-hexanediol which acts as a solvent.
The composition prepared in the example 2 is added into a skin care product (such as moisturizing and repairing mask essence), the mass percentage of the composition prepared in the example 2 in the cosmetic is 1%, and the skin care product does not have the phenomena of skin redness and swelling, allergy and the like after being continuously used for 1 year.
The skin care product containing the composition prepared in example 2 is placed at 48 ℃ for 10 hours, then at 0 ℃ for 10 hours, and then at 48 ℃ for 10 hours, and the circulation is repeated for 3 times, so that the skin care product (such as the moisturizing repair mask essence) still has good beautifying and moisturizing effects, and the skin care product still has good antibacterial effects on heat-resistant coliform bacteria, pseudomonas aeruginosa, white streptococcus, aspergillus niger and saccharomycetes.
The skin care product containing the composition prepared in example 2 was stored at normal temperature and pressure, and the shelf life of the skin care product was 3 years, but the shelf life of the cosmetic product not containing the composition prepared in example 2 was only 2 years.
Application example 1
The composition prepared in example 2 is added to a formula of the moisturizing and repairing mask essence instead of a preservative, and the specific formula is shown in table 3.

As can be seen from table 4, after 7 days of each inoculation, the viable count of each strain in the mask essence is detected, the viable count of all the strains is lower than 20CFU/ml, the bacteriostasis rate reaches over 99.999%, and after 28 days of the third inoculation, the bacteriostasis rate of each strain in the mask essence reaches 100%. The composition prepared in example 2 has excellent preservative effect on moisturizing repair mask essence by repeated inoculation challenge according to CTFA (american cosmetic association) and domestic microbial challenge evaluation criteria.
Application example 2
The composition prepared in example 2 was added to a moisturizing repair cream, and the formulation thereof is shown in table 5.

As can be seen from Table 6, challenge experiments were performed using the single inoculation microbiological method. After inoculation, the results of viable counts in the product are shown in the table above, with samples of heat-resistant coliform and Pseudomonas aeruginosa of the order of 106CFU, Candida albicans in the order of 105CFU, Aspergillus niger order of 104CFU, meeting the microbial challenge test bacteria number order regulation of cosmetics。
After 7 days of inoculation, the number of viable bacteria of each strain is less than 10CFU/ml, and the bacteriostasis rate of each strain reaches more than 99.998 percent; the bacteriostasis rate of each strain on the 14 th day and the 28 th day reaches 100 percent. According to CTFA and domestic microbial challenge evaluation standards, the added antibacterial composition has an excellent preservative effect on the moisturizing repair cream.
Application example 3
The composition prepared in example 2 was added to whitening anti-wrinkle milk instead of a preservative, and the formula thereof is shown in table 7.

After 7 days of inoculation, the bacteriostasis rate of each strain reaches 100 percent. According to CTFA and domestic microbial challenge evaluation standards, the added antibacterial composition has excellent preservative effect on the whitening anti-wrinkle milk.

4. The composition of claim 1, wherein the pH adjusting agent is citric acid.
5. The composition of claim 1, wherein the composition has a pH of 4 to 6.5.
6. A method for preparing the composition of any one of claims 1 to 5, comprising the steps of:
(1) weighing the components according to the formula ratio, adding a pH regulator into water, and regulating the pH to obtain a mixture A for later use;
(2) mixing p-hydroxyacetophenone and 1, 2-hexanediol, heating, stirring, and then cooling to obtain a mixture B for later use;

8. The method according to claim 6, wherein the temperature in the step (2) is raised to 40 to 50 ℃, stirred, and then lowered to 25 to 35 ℃.
9. A skin care product comprising the composition according to any one of claims 1 to 5.
10. The skin care product of claim 9, wherein the composition is present in the skin care product in an amount of 0.5 to 5% by weight.
