Abstract
The invention discloses donkey-hide gelatin vitality securing paste and a preparation method of nano-scale paste thereof. The donkey-hide gelatin vitality securing cream comprises the following preparation raw materials: colla Corii Asini, semen Sesami Niger, semen Juglandis, fructus Jujubae, fructus Lycii, flos Rosae Rugosae, brown sugar, collagen peptide, yellow wine, maltodextrin, maltose syrup and water. The preparation is carried out by adopting the processes of crushing, colloid milling and the like, and the nutritional ingredients and the functional ingredients of the raw materials are completely reserved; the low-temperature freezing and crushing technology is adopted, so that the problems of local overheating of equipment, material adhesion, equipment blockage and the like are solved, and the nutritional ingredients and the functional ingredients of the raw materials are not damaged, so that the fineness and the activity of the material particles are ensured.

Technical Field
The invention relates to the technical field of food processing, in particular to donkey-hide gelatin vitality-strengthening paste and a preparation method of nano-scale paste thereof.
Background
Nano-food refers to food that employs nanotechnology means or tools in the production, processing, or packaging process. The nanometer food has molecular and atomic re-programming and some structure changed by means of nanometer technology, and this can raise the absorption and utilization of some components in human body, speed the transportation of nutrients in body, strengthen original function and obtain new function. The nanometer food has effects in improving nutrition, improving body constitution, preventing diseases, recovering health, regulating body rhythm, and delaying aging.
The vitality securing paste is a traditional medicated diet in China and is a medicated diet which is considered to be very popular in the New year of Cixi. The formula is also created by royal of the Tang dynasty poplar, and the formula can nourish blood and moisten skin and blacken hair after being eaten frequently. At present, most of vitality securing pastes on the market are prepared from donkey-hide gelatin, red dates, black sesame, walnut kernels, longan, crystal sugar and yellow wine. Some formulas are required by different people, and medlar, spina date seed, honey, lotus seed and the like can be added.
Although the vitality securing cream has a long history, the formula and the process are too solidified, the vitality securing cream is rarely improved, only a plurality of raw materials are simply mixed, the shape is mostly blocky, the taste is more greasy, the absorption effect of the nutrient components by a human body is not good, and the efficacy of the vitality securing cream is not brought into full play at all.

Preferably, the donkey-hide gelatin vitality securing paste comprises the following raw materials in parts by weight: 1-10 parts of donkey-hide gelatin, 4-15 parts of black sesame, 2-10 parts of walnut kernel, 0.5-5 parts of red date, 2-10 parts of medlar, 0.1-2 parts of rose, 1-5 parts of brown sugar, 0.1-3 parts of collagen peptide, 0.1-2 parts of yellow wine, 3-15 parts of maltodextrin, 30-60 parts of maltose syrup and 10-40 parts of water.
Preferably, the donkey-hide gelatin vitality securing paste comprises the following raw materials in parts by weight: 1-5 parts of donkey-hide gelatin, 8-12 parts of black sesame, 3-5 parts of walnut kernel, 2-4 parts of red date, 1-4 parts of medlar, 0.1-0.5 part of rose, 1-3 parts of brown sugar, 0.1-0.6 part of collagen peptide, 0.2-1 part of yellow wine, 2-5 parts of maltodextrin, 38-45 parts of maltose syrup and 25-30 parts of water.
The donkey-hide gelatin vitality securing cream body is in a nanometer level, is prepared by the following method, and comprises the following steps:
(1) crushing: mixing semen Sesami Niger, semen Juglandis, fructus Jujubae, fructus Lycii, and flos Rosae Rugosae, coarse pulverizing, sieving with 10-40 mesh sieve, freezing at low temperature, pulverizing, sieving with 100-300 mesh sieve; pulverizing colla Corii Asini independently and sieving with 10-40 mesh sieve;
(2) boiling: adding the crushed materials into a boiling pot, and adding a proper amount of water; melting colla Corii Asini powder with yellow wine and appropriate amount of hot water, adding into a decocting pot, stirring, sealing, heating with steam to 95-120 deg.C, keeping the temperature for 0.5-2 hr, stirring for 1-2 times to soften the raw materials;
(3) rubber grinding: carrying out colloid milling on the boiled materials by using a colloid mill;
(4) nano-crystallization: homogenizing and nanocrystallizing the abrasive material by using a high-pressure homogenizer at the homogenizing pressure of 100-300MPa to obtain a nano liquid material;
(5) preparing materials: putting the nano liquid material into an emulsifying pot, dissolving the brown sugar, the collagen peptide and the maltodextrin with a proper amount of hot water, adding the dissolved materials and the maltose syrup into the emulsifying pot, and stirring and shearing to uniformly mix the materials to obtain the nano donkey-hide gelatin vitality securing cream.
The invention also discloses a donkey-hide gelatin Yuan-Gu tablet, which comprises the following preparation raw materials: colla Corii Asini, semen Sesami Niger, semen Juglandis, fructus Jujubae, fructus Lycii, flos Rosae Rugosae, brown sugar powder, collagen peptide, yellow wine, white sugar powder, mannitol, silicon dioxide and magnesium stearate.
Preferably, the donkey-hide gelatin Yuan-Gu tablet formula can also be added with ingredients meeting food regulations, including but not limited to ossein peptide, fish collagen peptide, sea cucumber peptide, bird's nest peptide, oyster peptide, maca, Chinese yam, rhizoma polygonati, dendrobium officinale leaf (flower), pseudo-ginseng stem leaf, ginseng, cistanche, codonopsis pilosula, astragalus mongholicus, polygonum multiflorum, longan pulp, lotus seed, gordon euryale seed, bird's nest, roxburgh rose powder, acerola cherry powder, sorbitol, lactose, starch, maltodextrin, microcrystalline cellulose, calcium carbonate, sucralose, stevioside, aspartame, acesulfame potassium, sodium cyclamate and the like.

(4) nano-crystallization: homogenizing and nanocrystallizing the abrasive material by using a high-pressure homogenizer at the homogenizing pressure of 100-300MPa to obtain a nano liquid material;
(5) and (3) drying: drying the nano liquid material, carrying out superfine grinding, and sieving with a sieve of more than 200 meshes;
(6) tabletting: mixing the above powders with brown sugar powder, collagen peptide, white sugar powder, mannitol, silicon dioxide, and magnesium stearate, and tabletting.
Compared with the prior art, the invention has the following beneficial effects:
1. adopts the processes of crushing, colloid milling and the like (rather than the extraction and concentration processes), completely reserves the nutrient components and the functional components of the raw materials.
2. By adopting the low-temperature freezing and crushing technology, the problems of local overheating of equipment, material adhesion, equipment blockage and the like are solved, and the nutritional ingredients and the functional ingredients of the raw materials are not damaged, so that the particle fineness and the activity of the materials are ensured.
3. The particle size of the particles of the vitality securing cream prepared by the invention reaches the nanometer level (the particle size range is 200-400 nm), the dissolubility of raw materials (especially some functional components with poor water solubility or extremely difficult dissolution) can be obviously improved, the vitality securing cream is easier to be absorbed by a human body, the efficacy is obviously improved, and the efficacy of the vitality securing cream is best exerted.
4. The nanometer donkey-hide gelatin vitality securing cream provided by the invention can replenish qi and blood, tonify the kidney and nourish qi, secure the body resistance and reinforce the vitality, has the function of enhancing immunity, and is very suitable for daily invigoration; the nano donkey-hide gelatin vitality securing tablet provided by the invention also has the characteristics of high content of functional components, convenience in eating and portability.

The following examples are further illustrative of the present invention and are not intended to be limiting thereof. The components of the formulations in the following examples are, unless otherwise specified, conventional commercial products.
The flow chart of the preparation process of the nanometer paste of the donkey-hide gelatin solid paste of the following examples is shown in figure 1.
Example 1
The donkey-hide gelatin vitality securing paste comprises the following raw materials in parts by weight:
1 part of donkey-hide gelatin, 10 parts of black sesame, 5 parts of walnut kernel, 2 parts of red date, 3 parts of medlar, 0.1 part of rose with multiple petals, 1 part of brown sugar, 0.1 part of collagen peptide, 0.2 part of yellow wine, 5 parts of maltodextrin, 45 parts of maltose syrup and 26 parts of water.
The preparation method of the nanometer paste of the donkey-hide gelatin vitality securing paste comprises the following steps:
(1) crushing: mixing semen Sesami Niger, semen Juglandis, fructus Jujubae, fructus Lycii, and flos Rosae Rugosae, coarse pulverizing, sieving with 10 mesh sieve, freezing at low temperature, pulverizing, and sieving with 100 mesh sieve; pulverizing colla Corii Asini independently and sieving with 20 mesh sieve;
(2) boiling: adding the crushed materials into a boiling pot, and adding a proper amount of water; melting colla Corii Asini powder with yellow wine and appropriate amount of hot water, adding into a decocting pot, stirring, sealing, heating with steam to 95 deg.C, keeping the temperature for 2 hr, and stirring for 1 time to soften the raw materials;
(3) rubber grinding: carrying out colloid milling on the boiled materials by using a colloid mill;
(4) nano-crystallization: homogenizing and nanocrystallizing the abrasive material by using a high-pressure homogenizer at the homogenizing pressure of 100MPa to obtain a nano liquid material;
(5) preparing materials: putting the nano liquid material into an emulsifying pot, dissolving the brown sugar, the collagen peptide and the maltodextrin with a proper amount of hot water, adding the dissolved materials and the maltose syrup into the emulsifying pot, and stirring and shearing to uniformly mix the materials to obtain the nano donkey-hide gelatin vitality securing cream.
(6) Packaging nanometer colla Corii Asini paste, and sterilizing to obtain final product.
Example 2
The donkey-hide gelatin vitality securing paste comprises the following raw materials in parts by weight:
3 parts of donkey-hide gelatin, 8 parts of black sesame, 3 parts of walnut kernel, 3 parts of red date, 4 parts of medlar, 0.5 part of rose with double petals, 2 parts of brown sugar, 0.4 part of collagen peptide, 0.5 part of yellow wine, 4 parts of maltodextrin, 39 parts of maltose syrup and 30 parts of water.
The preparation method of the nanometer paste of the donkey-hide gelatin vitality securing paste comprises the following steps:
(1) crushing: mixing semen Sesami Niger, semen Juglandis, fructus Jujubae, fructus Lycii, and flos Rosae Rugosae, coarse pulverizing, sieving with 20 mesh sieve, freezing at low temperature, pulverizing, and sieving with 150 mesh sieve; pulverizing colla Corii Asini independently and sieving with 30 mesh sieve;
(2) boiling: adding the crushed materials into a boiling pot, and adding a proper amount of water; melting colla Corii Asini powder with yellow wine and appropriate amount of hot water, adding into a decocting pot, stirring, sealing, heating with steam to 100 deg.C, keeping the temperature for 1 hr, and stirring for 1 time to soften the raw materials;
(3) rubber grinding: carrying out colloid milling on the boiled materials by using a colloid mill;
(4) nano-crystallization: homogenizing and nanocrystallizing the abrasive material with a high-pressure homogenizer at the homogenizing pressure of 150MPa to obtain a nano liquid material;
(5) preparing materials: putting the nano liquid material into an emulsifying pot, dissolving the brown sugar, the collagen peptide and the maltodextrin with a proper amount of hot water, adding the dissolved materials and the maltose syrup into the emulsifying pot, and stirring and shearing to uniformly mix the materials to obtain the nano donkey-hide gelatin vitality securing cream.
(6) Packaging nanometer colla Corii Asini paste, and sterilizing to obtain final product.
Example 3
The donkey-hide gelatin vitality securing paste comprises the following raw materials in parts by weight:
the donkey-hide gelatin porridge comprises, by weight, 5 parts of donkey-hide gelatin, 11 parts of black sesame, 4 parts of walnut kernels, 3 parts of red dates, 1 part of wolfberry fruits, 0.2 part of rose with double petals, 3 parts of brown sugar powder, 0.6 part of collagen peptide, 1 part of yellow wine, 2 parts of maltodextrin, 38 parts of maltose syrup and 25 parts of water.
The preparation method of the nanometer paste of the donkey-hide gelatin vitality securing paste comprises the following steps:
(1) crushing: mixing semen Sesami Niger, semen Juglandis, fructus Jujubae, fructus Lycii, and flos Rosae Rugosae, coarse pulverizing, sieving with 40 mesh sieve, freezing at low temperature, pulverizing, and sieving with 300 mesh sieve; pulverizing colla Corii Asini independently and sieving with 40 mesh sieve;
(2) boiling: adding the crushed materials into a boiling pot, and adding a proper amount of water; melting colla Corii Asini powder with yellow wine and appropriate amount of hot water, adding into a decocting pot, stirring, sealing, heating with steam to 120 deg.C, keeping the temperature for 0.5 hr, and stirring for 2 times to soften the raw materials;
(3) rubber grinding: carrying out colloid milling on the boiled materials by using a colloid mill;
(4) nano-crystallization: homogenizing and nanocrystallizing the abrasive material with a high-pressure homogenizer at the homogenizing pressure of 300MPa to obtain a nano liquid material;
(5) preparing materials: putting the nano liquid material into an emulsifying pot, dissolving the brown sugar, the collagen peptide and the maltodextrin with a proper amount of hot water, adding the dissolved materials and the maltose syrup into the emulsifying pot, and stirring and shearing to uniformly mix the materials to obtain the nano donkey-hide gelatin vitality securing cream.
(6) Packaging nanometer colla Corii Asini paste, and sterilizing to obtain final product.
Example 4
The donkey-hide gelatin vitality securing paste comprises the following raw materials in parts by weight:
10 parts of donkey-hide gelatin, 4 parts of black sesame, 2 parts of walnut kernel, 0.5 part of red date, 2 parts of medlar, 0.1 part of rose with multiple petals, 1 part of brown sugar, 0.1 part of collagen peptide, 2 parts of yellow wine, 3 parts of maltodextrin, 60 parts of maltose syrup and 10 parts of water.
The preparation procedure is as in example 1.
Example 5
The donkey-hide gelatin vitality securing paste comprises the following raw materials in parts by weight:
1 part of donkey-hide gelatin, 15 parts of black sesame, 10 parts of walnut kernel, 5 parts of red date, 10 parts of medlar, 2 parts of rose with double petals, 5 parts of brown sugar, 3 parts of collagen peptide, 0.1 part of yellow wine, 15 parts of maltodextrin, 30 parts of maltose syrup and 40 parts of water.
The preparation procedure is as in example 1.
Example 6
The donkey-hide gelatin Yuan Gu pian of the embodiment comprises the following raw materials in parts by weight:
1 part of donkey-hide gelatin, 6 parts of black sesame, 4 parts of walnut kernel, 1 part of red date, 3 parts of medlar, 0.1 part of rose with multiple petals, 1 part of black sugar powder, 0.1 part of collagen peptide, 0.2 part of yellow wine, 10 parts of white granulated sugar powder, 6 parts of mannitol, 0.4 part of silicon dioxide and 1 part of magnesium stearate.

Comparative example 1 donkey-hide gelatin vitality securing paste comprises the following raw materials in parts by weight:
3 parts of donkey-hide gelatin, 8 parts of black sesame, 3 parts of walnut kernel, 3 parts of red date, 4 parts of medlar, 0.5 part of rose with multiple petals, 2 parts of brown sugar, 0.4 part of collagen peptide, 0.5 part of yellow wine, 4 parts of maltodextrin and 39 parts of maltose syrup.
The preparation method adopts an extraction and concentration preparation process, and specifically comprises the following steps:

The results show that the donkey-hide gelatin solid paste prepared in examples 1 to 3 of the present invention was superior to the donkey-hide gelatin solid paste prepared in comparative example 1, commercially available donkey-hide gelatin paste a and commercially available donkey-hide gelatin paste B in all of the three evaluations of color, flavor and taste.
Test example II evaluation of Immunity-enhancing effect of donkey-hide gelatin Guyuan paste
1. Test samples: the donkey-hide gelatin original-strengthening paste obtained in examples 1-3 and comparative examples 1-2, and a commercially available donkey-hide gelatin paste A and a commercially available donkey-hide gelatin paste B.
2. The test method comprises the following steps: selecting 240 SPF-grade healthy Kunming male mice, weighing 18-22 g, and dividing into 3 test scheme groups, 80 each test scheme, sequentially comprising 1) ConA-induced splenic lymphocyte transformation experiment (MTT method) of mice, 2) antibody-producing cell detection experiment (Jerne modified slide method) and 3) mouse carbon clearance experiment, wherein each test scheme is divided into 8 groups, each group comprises 10 mice, sequentially comprising an example 1-3 group, a comparative example 1-2 group, a commercial product A group, a commercial product B group and a negative control group, sequentially intragastrically administering the donkey-hide gelatin solid extract prepared in the example 1-3 and the comparative example 1-2, the commercial donkey-hide gelatin extract A and the commercial donkey-hide gelatin extract B, the intragastrically administering amount is 60 mg/kg. as can be seen from Table 3, when the donkey-hide gelatin solid cream 30d prepared in examples 1-3 of the present invention was orally administered to mice, the lymphocyte transformation capacity of the mice was higher than that of the negative control group (P > 0.05), indicating that the sample had the effect of promoting the lymphocyte proliferation and transformation capacity of the mice, and the effect was better than that of comparative examples 1-2, and commercial product A and commercial product B.
As is apparent from Table 4, when the donkey-hide gelatin solid cream 30d prepared in examples 1 to 3 of the present invention was orally administered to mice, the number of antibody-producing cells in the mice was higher than that in the negative control group (P > 0.05), indicating that the sample had the effect of promoting the proliferation of the antibody-producing cells in the mice, and the effect was superior to that in comparative examples 1 to 2, and that in commercial product A and commercial product B.
As can be seen from Table 5, when the donkey-hide gelatin solid cream 30d prepared in examples 1-3 of the present invention was orally administered to mice, the phagocytic index of the mice was higher than that of the negative control group (P > 0.05), indicating that the sample had the effect of promoting the carbon clearance of mononuclear-macrophages in the mice, and the effect was superior to that of comparative examples 1-2, and commercial product A and commercial product B.
In conclusion, the donkey-hide gelatin vitality securing cream prepared in the embodiments 1 to 3 of the present invention has the function of enhancing immunity. As can be seen from comparative example 1, the preparation of the donkey-hide gelatin vitality securing cream by adopting the conventional extraction and concentration preparation process can significantly reduce the function of the donkey-hide gelatin vitality securing cream in enhancing the immunity. As can be seen from the comparative example 2, the proper adjustment of the formulation of the donkey-hide gelatin primordial qi strengthening paste can affect the function of the donkey-hide gelatin primordial qi strengthening paste in enhancing the immunity.

(1) crushing: mixing semen Sesami Niger, semen Juglandis, fructus Jujubae, fructus Lycii, and flos Rosae Rugosae, coarse pulverizing, sieving with 10-40 mesh sieve, freezing at low temperature, pulverizing, sieving with 100-300 mesh sieve; pulverizing colla Corii Asini independently and sieving with 10-40 mesh sieve;
(2) boiling: adding the crushed materials into a boiling pot, and adding a proper amount of water; melting colla Corii Asini powder with yellow wine and appropriate amount of hot water, adding into a decocting pot, stirring, sealing, heating with steam to 95-120 deg.C, keeping the temperature for 0.5-2 hr, stirring for 1-2 times to soften the raw materials;
(3) rubber grinding: carrying out colloid milling on the boiled materials by using a colloid mill;
(4) nano-crystallization: homogenizing and nanocrystallizing the abrasive material by using a high-pressure homogenizer at the homogenizing pressure of 100-300MPa to obtain a nano liquid material;
(5) preparing materials: putting the nano liquid material into an emulsifying pot, dissolving the brown sugar, the collagen peptide and the maltodextrin with a proper amount of hot water, adding the dissolved materials and the maltose syrup into the emulsifying pot, and stirring and shearing to uniformly mix the materials to obtain the nano donkey-hide gelatin vitality securing cream.
