背景技术

本发明部分地基于这样的发现:即,生物提取物中的分子组合对IL-1α具有抑制活性,因此可用于治疗具有炎性基础的疾病或病症。提取物可用于治疗全身性疾病或病症(包括NASH)或者皮肤疾病或病症,包括痤疮(包括不是由痤疮丙酸杆菌引起的痤疮)、瘢痕、瑕疵、湿疹和衰老的影响。而且,生物提取物中分子的组合刺激细胞生长,引起组织再生,因此可用于同时治疗炎性疾病或病症以及刺激组织再生。这种同时效应对于治疗诸如痤疮(包括不是由痤疮丙酸杆菌引起的痤疮)、湿疹、秃头或脱发和NASH之类的病症来说是特别有利的,这些病症具有炎性成分,因而能从抗炎活性中受益,而且,这些病症还具有可从组织再生中受益的成分。
因此,在第一方面,本发明提供一种具有抗炎活性的生物提取物。
术语“生物提取物”在本文用于指从受试者获得的样品,其中该受试者是动物、植物、真菌或细菌。生物提取物可以是粗提取物、部分纯化的提取物、基本纯化的提取物或纯化的提取物。当生物提取物是粗提取物时,它可以含有多种活性成分,其中活性成分是具有抗炎活性和/或刺激新细胞生长的能力的成分。
由于从粗提取物到纯化提取物的每一级纯化的缘故,提取物中活性成分的量会减少。活性成分可以选自包括以下各项的组中的一种或多种:蛋白质、肽、有机小分子和脂肪酸。优选地,纯化的生物提取物具有至少两种活性成分。甚至更优选地,纯化的生物提取物包含具有抗炎活性的活性成分以及具有刺激新细胞生长的能力的活性成分,因而提供了具有组合效应的提取物。
优选地,生物提取物从植物、真菌或细菌获得。在特别优选的实施方案中,生物提取物从节螺藻属中的一种或多种蓝藻细菌中获得。也就是说,生物提取物可获自节螺藻属中的任何合适的物种,所述物种包括但不限于极大节旋藻(Arthrospira maxima)。可选地,生物提取物可以来源于节螺藻属物种的混合物。
在优选的实施方案中,生物提取物是由生理应激的极大节旋藻制备的。
优选地,生理应激的极大节旋藻的粗提取物经过至少一个纯化阶段,以提供部分纯化的提取物、基本纯化的提取物或纯化的提取物。当与粗提取物的氨基酸含量相比时,该至少一个纯化阶段优选地使至少丙氨酸的相对氨基酸含量增加。优选地,由于该至少一个纯化阶段的缘故,丙氨酸含量增加了3-4摩尔%。甚至更优选地,由于纯化的缘故,选自由以下各项组成的组的一种或多种氨基酸的相对氨基酸含量增加了:丙氨酸、半胱氨酸、谷氨酸/谷氨酰胺、甘氨酸、天冬氨酸/天冬酰胺、脯氨酸、亮氨酸、缬氨酸、异亮氨酸、丝氨酸以及苯丙氨酸。由于该至少一个纯化阶段的缘故,这些氨基酸中每一种的含量优选独立地增加0.2-4摩尔%范围内的任一值。

在另一具体实施方案中,纯化阶段另外地使提取物中γ-亚麻酸的量减少12-16%,亚麻酸的量减少8-12%并且油酸的量减少1-2%。

在第四方面,本发明提供了具有抗炎活性和刺激新细胞生长的能力的生物提取物。
优选地,抗炎活性与刺激新细胞生长的能力联合发挥作用。例如,皮肤疾病或病症将受益于抗炎活性和刺激新细胞生长的联合,所述皮肤疾病或病症例如具有炎性成分并且也会引起细胞损伤(如瘢痕和纤维化)的痤疮(包括不是由痤疮丙酸杆菌引起的痤疮)和湿疹。

药物组合物中生物提取物的浓度可以根据应用而变化。例如,药物组合物中生物提取物的浓度可以是约0.01%至约100%之间的任意值。优选地,药物组合物中生物提取物的浓度为约5%至约20%。因此,药物组合物中生物提取物的浓度可以是约5、6、7、8、9、10、11、12、13、14、15、16、17、18、19或约20%。
药物组合物可以以任何合适的形式施用。例如,药物组合物可以是适合口服的形式。适合于口服的形式包括固体,诸如胶囊剂、片剂、丸剂、粉剂和颗粒剂。适合于口服的形式还包括液体,诸如乳剂、微乳剂、溶液剂、混悬剂、糖浆和酏剂。
在替代实施方案中,药物组合物可以是适合局部或经皮施用的形式。适合于局部或经皮施用的形式包括软膏、糊剂、乳膏、洗剂、凝胶剂、粉剂、溶液剂、气雾剂、吸入剂和贴剂。
在第九方面,本发明提供一种治疗受试者中炎性病变的方法,包括将治疗有效量的第一、第二或第三方面所提供的生物提取物或者第五、第六或第七方面所提供的药物组合物施用于该受试者的步骤。
治疗的受试者可以是人类、哺乳动物或动物。优选地,受试者是人类或其它类型的哺乳动物。
“治疗有效量”是指对治疗炎性病变有效的量。炎性病变可以是具有炎性基础的任何疾病或病症。因此,炎性病变可以是皮肤的疾病或病症,诸如痤疮(包括不是由痤疮丙酸杆菌引起的痤疮)、湿疹。可选地,炎性病变可以是涉及肉芽肿的疾病或病症,诸如嵌甲。在另外的替代方案中,炎性疾病或病症可以是NASH。
施用的步骤可以通过任何适当的手段进行。优选地,治疗有效量的生物提取物被口服施用或局部施用。
在第十方面,本发明提供一种治疗受试者中的皮肤病症(其中没有微生物感染或侵染)的方法,包括将治疗有效量的第一、第二或第三方面所提供的生物提取物或者第五、第六或第七方面所提供的药物组合物施用于该受试者的步骤。
皮肤病症可以是没有微生物感染或侵染的任何皮肤疾病或病症。优选地,皮肤病症具有炎性基础。例如,皮肤病症可以是痤疮(诸如不是由痤疮丙酸杆菌引起的痤疮)、湿疹、色素沉着、瑕疵、瘢痕、肉芽肿或老化。
应当理解,提供以下实施例,用于说明本发明的优选实施方案的目的。因此,应当理解的是,本发明不应被认为仅局限于实施例中所描述的特征。
实施例
实施例1——包含生理应激的节螺藻属的产品的抗炎活性的体外评价
将包含正常人表皮角质细胞并以综合三维细胞培养模型(Skinethic公司,法国尼斯)生长的重建人工人类皮肤模型用作人类皮肤的体外模拟。由于分化角质层的存在,该模型表现出正常的屏障功能。将正常人表皮角质细胞接种在胶原蛋白基质上,并使其在无血清培养基中生长,以形成表面有分化角质层的多层结构。在培养的第16天,直接从Skinethic公司购买0.5cm2直径的表皮单位。将表皮层置于多孔膜上的通透性隔离腔中。在使用和不使用0.5%SLS进行预处理(30分钟)的情况下,将待测产品的未稀释重复样品(10至15mg)施加在皮肤的上角质化层上。在37℃,5%CO2下16小时后,取出样品,用PBS清洗皮肤两次。然后,执行MTT测定以评价细胞存活率,由此评估样品的刺激潜力。
所测试的两个样品是18%生理应激的节螺藻属的皮肤医药(dermaceutical)乳膏(‘Amycot 18%’)和8%生理应激的节螺藻属的皮肤医药洗剂(‘Amycot 8%’)。
MTT测定的关键成分是在溶液中呈黄色的3-[4,5-二甲基噻唑-2-基]-2,5-二苯基四唑溴化物(MTT)。活细胞的线粒体脱氢酶裂解四唑环,导致形成不溶于水性溶液的紫色晶体。将这些晶体重新溶解在酸化异丙醇中,并用分光光度法测量所得的紫色溶液。细胞数量的增加或减少导致所形成的甲臜的量的伴随变化,从而指示出由测试材料引起的细胞毒性程度。
在本测定中,将细胞暴露于测试材料后,用Dulbecco的PBS洗涤细胞。在去除PBS后,将MTT培养基加入各培养孔中,并在37℃孵育细胞4小时。孵育之后,去除MTT培养基,在每个孔中加入MTT溶解溶液(酸化异丙醇)。使培养板在旋转板振荡器上振荡20至30分钟,确保所有晶体均溶解并形成均匀溶液。利用背景去除,在酶标仪上测量每种溶液的吸光度。结果示于表1中并根据等式1以存活力表示:

数据表明,在暴露于SLS(0.5%)16小时之后,细胞存活率降低95%。不含SLS的测试化合物在16小时时没有表现出细胞死亡。因此,结果表明,样品与皮肤之间具有良好的生物相容性,此外,样品没有对皮肤产生细胞毒性和/或刺激作用。
用市售ELISA测试试剂盒测量对IL-1α释放的抑制作用,作为皮肤刺激的指标。在利用基于抗体的比色反应进行特异性鉴定后,利用细胞因子IL-1α设计的标准曲线图允许通过读取定义波长处的吸光度来在细胞培养基中进行滴定。针对本测定,在6小时和16小时在孔的下隔室中收集200μL培养基。将SLS溶液(0.5%)用作阳性对照。
结果示于表2。

还测试了测试产品对SLS诱导的IL-1α释放的抑制作用,结果示于表3中。

结果表明,用测试产品处理6小时之后对SLS诱导的IL-1α释放的抑制作用,该结果还在图2中示出。
结果表明,在处理6小时之后,Amycot 18%和Amycot 8%都能抑制SLS诱导的IL-1α释放,然而,在处理16小时之后,这种作用消失。这两种产品确实导致了未处理细胞中IL-1α释放的可见(sight)增加,但这种增加远远低于由0.5%SLS的刺激作用所导致的增加。
使用培养的角质细胞进行细胞存活率测定,以确定暴露于测试样品后的细胞增殖。在细胞存活率测定中使用MTT测定。此外,使用Bradford测试,在暴露于测试样品的系列稀释液之后的不同端点处进行角质细胞中总细胞蛋白含量的测定。所测试的两个样品是18%生理应激的节螺藻属的皮肤医药乳膏(Amycot 18%)和8%生理应激的节螺藻属的皮肤医药洗剂(Amycot 8%)。
使用了由成人皮肤建立的细胞系(HaCaT)。该细胞系由转化人角质细胞组成,并具有完全表皮分化能力。
将HaCaT细胞接种于96孔板(2,500个细胞/孔)中的杜氏改良伊格尔培养基(DMEM)和10%胎牛血清(FCS)中24小时。然后加入新鲜培养基,仅补充5%的FCS以及溶解于培养基中的两种样品(即Amycot 18%和Amycot8%)的标量稀释液。将未处理细胞用作阴性对照。针对每种稀释液,均进行两次重复试验,并重复两次。在24小时、48小时和72小时后,使用MTT测定来测试细胞存活力,并使用Bradford测定来测试总蛋白含量。
对于MTT测定,如果细胞是活的,则细胞内的线粒体脱氢酶可以裂解MTT的四唑环,导致形成不溶于水性溶液的紫色晶体。将这些晶体溶解在酸化异丙醇中,并用分光光度法测量所得的紫色溶液。使HaCaT细胞暴露于样品后,用Dulbecco的PBS洗涤细胞。将MTT培养基添加到每个培养孔中,在37℃孵育细胞。孵育之后,去除MTT培养基,用MTT溶解溶液处理细胞。然后,使板在旋转板振荡器上振荡,确保所有晶体均从细胞中溶解并形成均匀溶液。利用背景清除,在酶标仪上测量吸光度。Amycot 18%和Amycot 8%的结果分别在表4和表5中示出,并根据等式1以存活力表示:

此结果表明,在0.001mg/mL下暴露24小时之后,相比未处理细胞,细胞增殖增加了。还在图3中示出了暴露于Amycot 18%后的细胞增殖结果。

此结果表明,在不同浓度下暴露24小时和72小时之后,相比未处理细胞,细胞增殖增加了。在0.1mg/mL浓度下暴露24小时后观察到最高活性。还在图4中示出了暴露于Amycot8%后的细胞增殖结果。
用Bio-Rad蛋白测定试剂盒进行总蛋白含量的Bradford测定。细胞用0.1N NaOH裂解,接着用Hank溶液进行稀释,然后再用蛋白质专用染料染色。在595nm处读取吸光度测量结果,根据与通过白蛋白的滴定曲线相比测得的光密度(OD),计算蛋白质滴定。相同数量细胞的总蛋白相对于Amycot 18%的浓度作图,并与阴性对照(未处理细胞)进行比较,如图5所示。图6中示出了与未处理细胞相比的百分比增长。
图7和图8中示出了用Amycot 8%处理的细胞的相应数据。
数据显示,对于在0.1mg/mL下暴露48小时后和在0.001mg/mL下暴露24小时后的Amycot 18%,与未处理细胞相比,蛋白质合成增加了。相比之下,Amycot 8%的使用使得蛋白质合成在暴露24小时、48小时和72小时后增加了,特别是在0.001mg/mL的浓度。
综上所述,结果表明,Amycot 8%在暴露24、48和72小时后能有效刺激皮肤来源的细胞(如角质细胞)的生长,Amycot 18%在暴露24小时后增加细胞增殖。此外,尽管Amycot8%和Amycot 18%使得蛋白质含量增加,但Amycot 8%在刺激角质细胞的蛋白质新合成(neosynthesis)方面比Amycot 18%更加有效,其中在0.001mg/mL浓度下暴露48小时后作用最大。
在由嵌甲引起的肉芽肿上测试包含8%生理应激的节螺藻属的产品的抗炎活性。受试者是16岁的女性,肉芽肿带来了很大的疼痛。在图9A中示出了处理之前的肉芽肿的图像。用包含8%生理应激的节螺藻属的局部用产品对肉芽肿进行处理。在最初处理的12小时内,受试者报告疼痛有所减轻。图9B中示出了处理后24小时的肉芽肿。
在患有痤疮的受试者上测试了包含12%生理应激的节螺藻属的产品的作用。在图10A和图10B中,示出了患有痤疮的受试者。粉刺非常明显并呈红色,这表明存在炎症。将包含12%生理应激的节螺藻属的局部用产品施用到每位受试者的患病区域。在图10C和图10D中示出了局部处理四周后的受试者。粉刺变得不那么明显,红肿有所减少,这说明该局部用产品具备抗炎作用。
实施例5——生理应激的节螺藻属对湿疹(汗疱疹)的作用
在患有湿疹(汗疱疹)的受试者上测试了包含12%生理应激的节螺藻属的产品的作用。受试者是42岁的女性,湿疹呈红色、瘙痒且已存在达8年。市面上可用的湿疹疗法并没有成功地清除湿疹。用包含12%生理应激的节螺藻属的局部用产品对湿疹进行了处理。在图11中,示出了在处理之前以及在处理期间的第1、3、5、7和10天的受试者右手的系列图。在最初处理的24小时(第1天)内,红肿减轻,然后到第10天湿疹已经消失,表明该局部用产品具有抗炎作用。
针对受试者的皮肤瑕疵,测试了包含12%生理应激的节螺藻属的产品的作用。受试者为55岁的女性。在图12A中示出了处理之前的皮肤瑕疵的图像。将包含12%生理应激的节螺藻属的局部用产品每天施用于皮肤瑕疵,持续两周。图12B中示出了两周的每日处理之后的皮肤瑕疵区域。皮肤瑕疵不再可见,这表明该局部用产品对皮肤瑕疵有作用,即使是在没有微生物感染或侵染的情形下。
对经加工的极大节旋藻进行定量氨基酸分析,其中分别进行半胱氨酸分析和色氨酸分析。对未加工的原始(source)极大节旋藻也进行定量氨基酸分析、半胱氨酸分析和色氨酸分析,以便与经加工的极大节旋藻进行比较。
为了进行氨基酸定量分析,将经加工的极大节旋藻和原始极大节旋藻的样品在6M盐酸中于110℃进行液体水解,持续24小时。采用过甲酸氧化且随后在110℃酸水解24小时来进行半胱氨酸分析。利用氢氧化物在110℃水解24小时来进行色氨酸分析。
水解之后,用AccQ-Tag化学(Waters Corporation)分析氨基酸含量。样品一式两份被分析,结果以平均值表示,列于表6至表9中。

数据表明,当与原始极大节旋藻的相对氨基酸含量相比时,对粗的原始极大节旋藻进行加工的步骤使得加工后的极大节旋藻中丙氨酸、缬氨酸、异亮氨酸、亮氨酸、甘氨酸、脯氨酸、半胱氨酸、天冬氨酸/天冬酰胺、谷氨酸/谷氨酰胺、丝氨酸和苯丙氨酸的相对氨基酸含量增加。同时,可以看出,加工后的极大节旋藻中赖氨酸、精氨酸、组氨酸、酪氨酸、色氨酸和甲硫氨酸的相对氨基酸含量与原始极大节旋藻的相对氨基酸含量相比有所降低。
对加工后的极大节旋藻进行脂肪酸分析。对未加工的原始极大节旋藻也进行脂肪酸分析,以便与加工后的极大节旋藻进行比较。

Claims (14)
1.一种源自生理应激的节螺藻属(Arthrospira)的提取物,其中所述提取物具有抗炎活性。
2.根据权利要求1所述的提取物,其中所述抗炎活性是对IL-1α的抑制活性。
3.根据权利要求1或2所述的提取物,其还具有刺激新细胞生长的能力。
4.一种药物组合物,其包含根据权利要求1至3中任一项所述的提取物以及药学上可接受的载体、溶剂、碱或赋形剂。
5.一种治疗受试者中炎性病变的方法,其包括将治疗有效量的根据权利要求1至3中任一项所述的提取物或者根据权利要求4所述的药物组合物施用于所述受试者的步骤。
6.一种治疗受试者中皮肤病症的方法,其中所述皮肤病症不是由微生物感染或侵染引起的,所述方法包括将治疗有效量的根据权利要求1至3中任一项所述的提取物或者根据权利要求4所述的药物组合物施用于所述受试者的步骤。
7.一种治疗受试者中炎性病变和皮肤病症的方法,其中所述皮肤病症不是由微生物感染或侵染引起的,所述方法包括将治疗有效量的根据权利要求1至3中任一项所述的提取物或者根据权利要求4所述的药物组合物施用于所述受试者的步骤。
8.治疗有效量的根据权利要求1至3中任一项所述的提取物或者根据权利要求4所述的药物组合物,其用于治疗炎性病变。
9.治疗有效量的根据权利要求1至3中任一项所述的提取物或者根据权利要求4所述的药物组合物,其用于治疗皮肤病症,其中所述皮肤病症不是由微生物感染或侵染引起的。
10.治疗有效量的根据权利要求1至3中任一项所述的提取物或者根据权利要求4所述的药物组合物,其用于治疗炎性病变和皮肤病症,其中所述皮肤病症不是由微生物感染或侵染引起的。
11.治疗有效量的根据权利要求1至3中任一项所述的提取物在制造用于治疗炎性病变的药物中的用途。
12.治疗有效量的根据权利要求1至3中任一项所述的提取物在制造用于治疗皮肤病症的药物中的用途,其中所述皮肤病症不是由微生物感染或侵染引起的。
13.治疗有效量的根据权利要求1至3中任一项所述的生物提取物在制造用于治疗炎性病变和皮肤病症的药物中的用途,其中所述皮肤病症不是由微生物感染或侵染引起的。
14.一种针对抗炎活性筛选生理应激的节螺藻属的提取物的方法,所述方法包括测定所述提取物的IL-1α抑制活性,其中IL-1α抑制活性指示抗炎活性。
