技术领域
本发明涉及化妆品领域,具体涉及一种促进皮肤成纤维细胞分泌结构蛋白的多效乳霜。
背景技术
皮肤弹性问题是所有人都会面临的问题,随着年龄增大,弹性蛋白的流失是不可避免的,因皮肤缺少弹性而生出褶皱是人们所关注的重要皮肤问题之一,社会上多数比较有效的方式是手术和预防,而手术是被少部分人认同的方法,对于已经缺失皮肤弹性的人用处不大。
现有技术中,通过在乳霜中引入弹性蛋白肽,弹性蛋白肽脂质体能够通过促进成纤维细胞在皮肤真皮部分产生弹性蛋白和胶原蛋白,直接进入面部血液循环,从而使皮肤恢复饱满有弹性的状态。但是,由于蛋白质多肽类药物需要保持蛋白质分子的化学结构来维持其活性,其存在稳定性差、半衰期短、膜通透性差、体内清除率高,易受酶、细菌甚至体液破坏,非注射给药生物利用度极低的问题。
发明内容
针对人体面部皮肤弹性蛋白流失和现有含有弹性蛋白肽乳霜存在的上述问题,本发明的目的提供了一种促进皮肤成纤维细胞分泌结构蛋白的多效乳霜,将弹性蛋白肽制备成脂质体加入到乳霜中,使弹性蛋白肽脂质体能够被皮肤有效吸收,从而恢复面部皮肤弹性。
本发明解决其技术问题采用的技术方案如下:
本发明的多效乳霜由以下重量份的原料组成:水170~190份、卡波姆0.3~0.7份、甘油10~15份、尿囊素1~2份、矿油13~17份、羟苯丙酯0.2~0.3份、羟苯甲酯0.2~0.3份、聚二甲基硅氧烷0.3~0.6份、十六十八醇11~15份、香精0.07~0.1份、弹性蛋白肽脂质体20~24份、生育酚糖苷0.8~1.5份、冰片0.15~0.25份、硬脂酸甘油酯2.5~3份、吐温201.0~1.5份、丙二醇6~9份、乳木果油9~11份、乙二胺四乙酸二钠0.4~0.6份和氨甲基丙醇适量。
在某一实施方案中,所述乳霜由以下重量份的原料组成:水181份、卡波姆0.5份、甘油12.5份、尿囊素1.5份、矿油15份、羟苯丙酯0.25份、羟苯甲酯0.25份、聚二甲基硅氧烷0.45份、十六十八醇12.5份、香精0.1份、弹性蛋白肽脂质体21.5份、生育酚糖苷1.5份、冰片0.25份、硬脂酸甘油酯2.8份、吐温20 1.3份、丙二醇7.5份、乳木果油10份、乙二胺四乙酸二钠0.5份和氨甲基丙醇适量。
进一步,所述弹性蛋白肽脂质体由以下重量份的原料组成:泊洛沙姆20份、氢化大豆磷脂20份、胆固醇6份、吐温-80 2份、丙二醇4份、甘露醇150份和弹性蛋白肽100份。
更进一步,所述弹性蛋白肽脂质体通过以下制备方法得到:先将聚维酮溶解于水中,加入弹性蛋白肽完全溶解后装于西林瓶中,于-30℃冷冻5小时,得到弹性蛋白肽固态冻干品;再取氢化大豆磷脂、胆固醇、吐温80和十六醇共同溶于叔丁醇,在65℃水浴中溶解,加入上述弹性蛋白肽固态冻干品,完全溶解,加入甘露醇,分散均匀后装于西林瓶中,于-30℃冷冻5小时,得到弹性蛋白肽脂质体固态冻干品。
本发明的多效乳霜中各种原料在乳霜体系中的作用为:
弹性蛋白肽脂质体能够通过透皮吸收,刺激皮下纤维母细胞产生弹性蛋白原形成弹性纤维,达到增加皮肤弹性的作用。
乳木果油作为纯天然成分,拥有抗皮肤衰老因子,和弹性蛋白肽脂质体配合可使皮肤减少皱纹产生,与甘油配合使用可以保持皮肤湿润状态。
生育酚糖苷和冰片均作为透皮吸收促进剂,使平时难以透过皮肤吸收的弹性蛋白肽脂质体能够被人体皮肤所吸收,同时生育酚糖苷还可以抗氧化,去除皮肤表面自由基的作用,丙二醇用于溶解冰片,使其在水相中稳定存在。
卡波姆为丙烯酸键合烯丙基蔗糖或季戊四醇烯丙醚的高分子聚合物,作为凝胶基质有效促进油相及其水相的稳定性。
尿囊素是尿基海因,具有增加和促进接合基质的水合能力,还能作用于角蛋白,使其结合水的能力增加,保持皮肤光滑、滋润和柔韧,且尿囊素的尿素还有抗霉菌和抗细菌作用。
羟苯丙酯和羟苯甲酯复配具有抑制真菌以及细菌的效果,用作抑菌防腐剂,提高乳霜防腐性能,延长其货架期。
氨甲基丙醇作为pH调节剂,有效调节霜体pH值使其符合人体皮肤所需。
乙二胺四乙酸二钠可与细菌生长所必需的微量金属离子螯合,增强抗氧化效果,同时也能提高乳霜防腐性能,延长乳霜货架期。另外,其还具有减慢反应、保持化学平衡、降低表面张力以及防止光、热分解或氧化分解等作用。
乳化剂,有一定的自增稠性,对无论是霜体还是乳液都拥有良好稳定性,且乳化后的霜体外观细腻光亮、铺展性较好,易涂抹、赋予产品很舒适的涂抹特性,有效发挥功效成分吸收。和吐温20、聚二甲基硅氧烷配合使用可作为乳霜基质。
十六十八醇和聚二甲基硅氧烷可以稳定水相、油相以及主要功能组分之间的稳定性,使其不易分层,使功能成分分散均匀。

(ⅲ)制备组分C:按比例将所述弹性蛋白肽脂质体、生育酚糖苷和部分水混合后备用;
(ⅳ)制备组分D:按比例将尿囊素和香精混合共熔后备用;
(ⅴ)制备乳霜:将上述水相A和油相B转入反应罐中,在真空度为0.01~0.05Mpa的条件下于85~90℃搅拌混合25~35r/min,2300~2800r/min均质6~8min,再于45~55℃和25~32r/min的条件下搅拌3~4min与组分C混合,2300~2800r/min第二次均质4~6min,25~32r/min搅拌降温到33~37℃,出料。
与现有方案相比,本发明具有以下有益效果:
1、本发明中,主要功能成分弹性蛋白肽脂质体能够在冰片和生育酚糖苷的辅助作用下,刺激皮肤生成新的弹性蛋白,乳木果油所含有的抗衰老成分和弹性蛋白肽脂质体配合使用,可有效减少皱纹产生和促进皮肤弹性。
2、羟苯甲酯和羟苯丙酯复配使用,且在乙二胺四乙酸二钠的配合下,可以有效抑菌、防腐和延长乳霜的货架期,乙二胺四乙酸二钠和生育酚糖苷配合还可以增强抗氧化能力。另外,十六十八醇、聚二甲基硅氧烷和卡波姆的添加使霜体变稠、各物质混合后均匀分散,便于涂抹,同时有效促进油相以及水相的稳定性。
3、本发明的弹性蛋白肽脂质体乳霜制备方法简单易控制,适用于工业应用,同时可有效提高稳定性以及渗透力。

为使本发明的技术方案和优点更加清楚,下面将对本发明实施例中的技术方案进行详细说明。实施例中未注明具体条件者,按照常规条件或制造商建议的条件进行,所用试剂或仪器未注明生产厂商者,均为可以通过市售购买获得的常规产品。
以下实施例中多效乳霜的制备方法步骤为:
(1)制备水相A
按比例将水相原料冰片、丙二醇、乳木果油、甘油、卡波姆、乙二胺四乙酸二钠、羟苯甲酯、羟苯丙酯、氨甲基丙醇和部分水投入水相锅中,于80~90℃搅拌混合25~35r/min,80~85℃保温,得水相A。
(2)制备油相B
按比例将油相原料矿油、聚二甲基硅氧烷、十六十八醇、吐温20、硬脂酸甘油酯加入油相锅中,于80~90℃搅拌混合25~35r/min,80~85℃保温,得油相B。
(3)制备组分C
按比例将所述弹性蛋白肽脂质体、生育酚糖苷和部分水混合后备用。
(4)制备组分D
按比例将尿囊素和香精混合共熔后备用。
(5)制备乳霜
将上述水相A和油相B转入反应罐中,在真空度为0.01~0.05Mpa的条件下于85~90℃搅拌混合25~35r/min,2300~2800r/min均质6~8min,再于45~55℃和25~32r/min的条件下搅拌3~4min与组分C混合,2300~2800r/min第二次均质4~6min,25~32r/min搅拌降温到33~37℃,出料。
上述所用的弹性蛋白肽脂质体由以下重量份的原料组成:泊洛沙姆20份、氢化大豆磷脂20份、胆固醇6份、吐温-80 2份、丙二醇4份、甘露醇150份和弹性蛋白肽100份,还包括叔丁醇15500份和水7000份;通过以下制备方法得到:先将聚维酮溶解于水中,加入弹性蛋白肽完全溶解后装于西林瓶中,于-30℃冷冻5小时,得到弹性蛋白肽固态冻干品;再取氢化大豆磷脂、胆固醇、吐温80和十六醇共同溶于叔丁醇,在65℃水浴中溶解,加入上述弹性蛋白肽固态冻干品,完全溶解,加入甘露醇,分散均匀后装于西林瓶中,于-30℃冷冻5小时,得到弹性蛋白肽脂质体固态冻干品。
弹性蛋白肽制作过程为:选用鲣鱼韧带洗净绞碎后与磷酸缓冲液混合均匀,加入弹性蛋白酶;将加入弹性蛋白酶后的溶液每1L溶液中加入8.5gNaCl,超声波处理,并在25℃震荡培养2h,通过多层纱布过滤并弃去滤渣,将滤液经过活性炭脱色后置于超过滤装置中通过半透膜;将半透膜滤过后的弹性蛋白收集后溶于纯水中,调节pH值至9.5,静置30min,在转速为15000r/min的条件下离心分离15min,取上清液,使用透析袋提取所需分子量的弹性蛋白肽。
实施例1
采用上述制备方法制备多效乳霜,其由以下重量份数的原料制备而成:水181份、卡波姆0.5份、甘油12.5份、尿囊素1.5份、矿油15份、羟苯丙酯0.25份、羟苯甲酯0.25份、聚二甲基硅氧烷0.45份、十六十八醇12.5份、香精0.1份、弹性蛋白肽脂质体固态冻干品21.5份、生育酚糖苷1.5份、冰片0.25份、硬脂酸甘油酯2.8份、吐温20 1.3份、丙二醇7.5份、乳木果油10份、乙二胺四乙酸二钠0.5份和氨甲基丙醇适量。
实施例2
本实施例中多效乳霜的制备方法同实施例1,不同之处在于其由以下重量份数的原料制备得到:水173份、卡波姆0.3份、甘油13份、尿囊素1份、矿油14份、羟苯丙酯0.2份、羟苯甲酯0.2份、聚二甲基硅氧烷0.5份、十六十八醇14份、香精0.1份、弹性蛋白肽脂质体固态冻干品22份、生育酚糖苷1.2份、冰片0.15份、硬脂酸甘油酯2.7份、吐温20 1份、丙二醇8份、乳木果油10份、乙二胺四乙酸二钠0.4份和氨甲基丙醇适量。
实施例3
本实施例中多效乳霜的制备方法同实施例1,不同之处在于其由以下重量份数的原料制备得到:水185份、卡波姆0.6份、甘油14份、尿囊素1.5份、矿油17份、羟苯丙酯0.2份、羟苯甲酯0.3份、聚二甲基硅氧烷0.5份、十六十八醇14份、香精0.08份、弹性蛋白肽脂质体固态冻干品22份、生育酚糖苷1.1份、冰片0.20份、硬脂酸甘油酯2.9份、吐温20 1.4份、丙二醇8份、乳木果油9份、乙二胺四乙酸二钠0.4份和氨甲基丙醇适量。
实施例4
本实施例中多效乳霜的制备方法同实施例1,不同之处在于其由以下重量份数的原料制备得到:水175份、卡波姆0.6份、甘油12份、尿囊素1份、矿油15份、羟苯丙酯0.2份、羟苯甲酯0.25份、聚二甲基硅氧烷0.35份、十六十八醇12份、香精0.1份、弹性蛋白肽脂质体固态冻干品23份、生育酚糖苷1份、冰片0.25份、硬脂酸甘油酯2.6份、吐温20 1.2份、丙二醇9份、乳木果油11份和氨甲基丙醇适量,且其不含乙二胺四乙酸二钠。
效果例1
弹性蛋白肽对于皮肤成纤维细胞效果(激活人体真皮成纤维细胞)的体外评价试验,其中:
细胞:人真皮成纤维细胞;
试验期:5天;
试验材料:弹性蛋白肽50、500、5000ng/mL。
评价方法:利用相对值方法,通过CTG(CELL TITER-GLO)发光法测定鲣鱼弹性蛋白肽对于人体成纤维细胞的增殖的影响,设定对照组细胞数为100%。表1是鲣鱼弹性蛋白肽添加量对于人体真皮成纤维细胞的增殖的影响(*试验前后比较P≤0.05;**试验前后比较P≤0.01。)

图1是体外测试结果,表明人体真皮成纤维细胞数随着鲣鱼弹性蛋白肽的添加而增加(表1),且该促进作用呈剂量依赖关系。
效果例2
弹性蛋白肽对于促皮肤成纤维细胞的胶原蛋白合成影响(促进胶原蛋白的生成)的体外评价试验,其中:
细胞:人真皮成纤维细胞;
试验期:5天;
试验材料:弹性蛋白肽0、0.1、1、10mg/mL。
评价方法:利用相对值方法,运用羟脯氨酸法胶原蛋白定量检测技术分析鲣鱼弹性蛋白肽对人真皮成纤维细胞生成胶原蛋白的含量影响,设定对照组细胶原蛋白的产生量为100%;表2鲣鱼弹性蛋白肽添加量对于人体真皮成纤维细胞合成胶原蛋白的影响(*试验前后比较P≤0.05;**试验前后比较P≤0.01)。
图2是体外测试结果,与对照组相比,鲣鱼弹性蛋白肽能明显促进人真皮成纤维细胞生成胶原蛋白(表2),且该促进作用也呈剂量依赖关系。

效果例3:多效乳霜临床试验验证
实验对象:男性25例,女性25例,年龄30~45岁。
随机分组:对照组、试用组1(实施例1产品)、试用组2(实施例2产品)、试用组3(实施例3产品)、试用组4(实施例4产品),其中,试用组1~4分别使用实施例1~4制备的多效乳霜,每日2次,每次1~3g,连续使用30天后经行皮肤纹理和皱纹测试。
测试方法:用干净棉球蘸蒸馏水将被测皮肤擦洗干净,擦干15分钟后用皮肤测定仪测定皮肤纹理和皱纹,测试结果如表3所示。

试用30天后,四个试用组受试者皮肤纹理及皱纹指标均有不同程度的改善,相比而言,试用组1中改善程度最为显著,说明本发明促进皮肤成纤维细胞分泌结构蛋白的多效乳霜具有理想抗皱、改善皮肤弹性效果,且其效果明显优于改变配方或制备工艺获得的样品(试用组2~4)。
以上对本发明优选的具体实施方式和实施例作了详细说明,但是本发明并不限于上述实施方式和实施例,在本领域技术人员所具备的知识范围内,还可以在不脱离本发明构思的前提下作出各种变化。

Claims (7)

1.弹性蛋白肽脂质体,其特征在于,其具有脂质体包覆弹性蛋白肽结构,且所述脂质体为亚微米级脂质体,所述弹性蛋白肽的分子量小于1000,所述弹性蛋白肽脂质体的平均包封率不低于80%;其中:
所述弹性蛋白肽脂质体由以下重量份的原料组成:泊洛沙姆20份、氢化大豆磷脂20份、胆固醇6份、吐温-80 2份、丙二醇4份、甘露醇150份和弹性蛋白肽100份。
2.根据权利要求1所述的弹性蛋白肽脂质体,其特征在于,所述弹性蛋白肽脂质体通过以下制备方法得到:
先将泊洛沙姆溶解于水中,加入弹性蛋白肽完全溶解后装于西林瓶中,于-30℃冷冻5小时,得到弹性蛋白肽固态冻干品;
再取氢化大豆磷脂、胆固醇、吐温80和丙二醇共同溶于叔丁醇,在65℃水浴中溶解,加入上述弹性蛋白肽固态冻干品,完全溶解,加入甘露醇,分散均匀后装于西林瓶中,于-30℃冷冻5小时,得到弹性蛋白肽脂质体固态冻干品。
3.根据权利要求1或2所述的弹性蛋白肽脂质体,其特征在于,所述弹性蛋白肽通过以下制备方法得到:选用鲣鱼韧带洗净绞碎后与适量磷酸缓冲液混合均匀,加入弹性蛋白酶,将加入弹性蛋白酶后的每1L溶液中加入8.5gNaCl,超声波处理,并在25℃震荡培养2h,通过多层纱布过滤并弃去滤渣,将滤液经过活性炭脱色后置于超过滤装置通过其半透膜;将半透膜滤过后的弹性蛋白收集后溶于纯水中,调节pH值至9.5,静置30min,15000r/min离心分离15min,取上清液,并将上清液通过1000分子量的透析袋。
4.一种促进皮肤成纤维细胞分泌结构蛋白的多效乳霜,其特征在于,所述乳霜内含有权利要求1~3任一项所述弹性蛋白肽脂质体。
5.根据权利要求4所述的多效乳霜,其特征在于,所述多效乳霜由以下重量份的原料组成:所述弹性蛋白肽脂质体20~24份、水170~190份、卡波姆0.3~0.7份、甘油10~15份、尿囊素1~2份、矿油13~17份、羟苯丙酯0.2~0.3份、羟苯甲酯0.2~0.3份、聚二甲基硅氧烷0.3~0.6份、十六十八醇11~15份、香精0.07~0.1份、生育酚糖苷0.8~1.5份、冰片0.15~0.25份、硬脂酸甘油酯2.5~3份、吐温20 1.0~1.5份、丙二醇6~9份、乳木果油9~11份、乙二胺四乙酸二钠0.4~0.6份和氨甲基丙醇适量。
6.根据权利要求4或5所述的多效乳霜,其特征在于,所述乳霜由以下重量份的原料组成:所述弹性蛋白肽脂质体21.5份、水181份、卡波姆0.5份、甘油12.5份、尿囊素1.5份、矿油15份、羟苯丙酯0.25份、羟苯甲酯0.25份、聚二甲基硅氧烷0.45份、十六十八醇12.5份、香精0.1份、生育酚糖苷1.5份、冰片0.25份、硬脂酸甘油酯2.8份、吐温20 1.3份、丙二醇7.5份、乳木果油10份、乙二胺四乙酸二钠0.5份和氨甲基丙醇适量。
7.权利要求4~6任一项所述多效乳霜的制备方法,其特征在于,包括以下步骤:
(ⅰ)制备水相A:按比例将水相原料冰片、丙二醇、乳木果油、甘油、卡波姆、乙二胺四乙酸二钠、羟苯甲酯、羟苯丙酯、氨甲基丙醇和部分水投入水相锅中,于80~90℃搅拌混合25~35r/min,80~85℃保温,得水相A;
(ⅱ)制备油相B:按比例将油相原料矿油、聚二甲基硅氧烷、十六十八醇、吐温20、硬脂酸甘油酯加入油相锅中,于80~90℃搅拌混合25~35r/min,80~85℃保温,得油相B;
(ⅲ)制备组分C:按比例将所述弹性蛋白肽脂质体、生育酚糖苷和部分水混合后备用;
(ⅳ)制备组分D:按比例将尿囊素和香精混合共熔后备用;
(ⅴ)制备乳霜:将上述水相A和油相B转入反应罐中,在真空度为0.01~0.05Mpa的条件下于85~90℃搅拌混合25~35r/min,2300~2800r/min均质6~8min,再于45~55℃和25~32r/min的条件下搅拌3~4min与组分C混合,2300~2800r/min第二次均质4~6min,25~32r/min搅拌降温到33~37℃,出料。
