技术领域
本发明涉及生物技术领域,尤其是涉及一种猪脂肪酸组成相关的分子标记和基因及应用。
背景技术
生猪产业是我国畜牧业中的支柱产业。猪肉作为我国最为主要的肉类食品之一,其占全部肉类食品产量和消费量的比值超过60%。猪肉肌纤维粗细适中,结缔组织较少,含有较多的肌间脂肪,风味和口感鲜美;瘦肉中不仅富含优质动物蛋白,还含有各种微量元素尤其是铁,深受人们的喜爱。同时,猪是一种特殊的高脂肪物种,其体内丰富的脂肪,不仅能为人体提供足够的能量,促进脂溶性维生素的吸收,也可以提高猪肉的风味;并且这些脂肪也是重要动物油脂和工业原料。
猪脂肪主要由饱和高级脂肪酸甘油酯与不饱和高级脂肪酸甘油脂组成,其中饱和高级脂肪酸甘油酯含量更高,具有一定的营养,并且能提供极高的热量。也正因如此,尽管猪油与一般植物油相比,有着不可替代的特殊香味,可以获得用其他调料难以达到的美味,但我们不宜多食。饱和脂肪酸(SFA)含量高则不易被身体吸收,研究发现,SFA长期摄入量过多会导致高血脂症,形成动脉粥样硬化,增加心脑血管疾病的发生风险。相反,临床研究证明,多不饱和脂肪酸(PUFA)有利于人体健康,因其能调节人体脂质代谢,减少血液中胆固醇和甘油三酯含量,从而降低血液黏稠度并改善血液微循环,对心脑血管疾病的治疗和预防有重要价值;同时还被证明,其能增强人体免疫防御功能以及排除过量SFA形成的多余脂肪,具有减肥的功效。
其中,n-3系列PUFAs是发挥这些功能的主要成分,其对健康的积极影响主要通过调节和抑制类二十烷酸的酸合成途径,进而改变炎症反应和相关分子蛋白表达活性、调节与正常及病理细胞功能有关的各种信号通路中分子及酶活性,且n-3脂肪酸可并入膜磷脂直接影响基因的表达,所有这些通路均高度相关,n-3脂肪酸对健康和疾病的生物有益作用是靠多种协调机制完成的。然而,对于PUFA,人体自身大多不能合成,需要从外界摄取。由此,多不饱和脂肪酸在食品、健康辅料、化妆品和制药中的潜在市场是十分巨大的。目前,不饱和脂肪酸仅能从高等植物种子和深海鱼油中获取。但是受限于其易氧化的性质和工艺复杂的提炼过程,无法满足市场及广大人民需求。
因此,长期以来,人们在不断探求可替代的生物资源,因猪肉具有普遍性,生猪产业量庞大,所以增加猪肉和猪脂肪中不饱和脂肪酸比例,对提高其食用价值和推动相关产业发展具有重要意义。
有鉴于此,特提出本发明。
发明内容
本发明的第一目的在于提供一种猪脂肪酸组成相关的分子标记,所述分子标记为位于猪MSTN基因内含有如SEQ ID NO.1所示片段。
本发明的第二目的在于提供一种猪脂肪酸组成相关的基因。
本发明的第三目的在于提供一种上述猪脂肪酸组成相关的分子标记或基因的应用。

根据本发明的一个方面,本发明提供了一种猪脂肪酸组成相关的分子标记,所述分子标记为位于猪MSTN基因内含有如SEQ ID NO.1所示序列的片段;
所述猪脂肪酸组成包括猪饱和脂肪酸含量、单不饱和脂肪酸含量、n-3系列多不饱和脂肪酸在脂肪酸中的占比和n-6系列多不饱和脂肪酸在脂肪酸中的占比。
优选地,所述猪脂肪酸组成包括猪背最长肌中脂肪酸组成、猪板油中脂肪酸组成和猪皮下脂肪中脂肪酸组成。
根据本发明的另一个方面,本发明还提供了一种猪脂肪酸组成相关的基因,所述基因为猪MSTN基因缺失上述含有如SEQ ID NO.1所示序列的片段后的基因;

根据本发明的另一个方面,本发明还提供了上述猪脂肪酸组成相关的分子标记、上述猪脂肪酸组成相关的基因、上述生物材料或上述调控猪脂肪酸组成的方法在(x1)鉴定猪脂肪酸组成;(x2)选育种猪;(x3)制备不饱和脂肪酸;或(x4)制备包括猪肉原料的食品中的应用。
与现有技术相比,本发明具有如下有益效果:
本发明提供的猪脂肪酸组成相关的分子标记,该分子标记为位于猪MSTN基因内含有如SEQ ID NO.1所示序列的片段。不存在上述分子标记的猪,饱和脂肪酸含量更低,单不饱和脂肪酸含量更高,n-3和n-6系列多不饱和脂肪酸在脂肪酸中的占比分别更高。
本发明提供的猪脂肪酸组成相关的基因为猪MSTN基因缺失上述含有如SEQ IDNO.1所示序列的片段后的基因。该基因和降低猪饱和脂肪酸含量,提高单不饱和脂肪酸含量,提高n-3系列多不饱和脂肪酸在脂肪酸中的占比相关以及和提高n-6系列多不饱和脂肪酸在脂肪酸中的占比相关。使猪表达上述猪脂肪酸组成相关的基因能够调控猪脂肪酸组成。
上述分子标记和上述基因,可以用于选育不饱和脂肪酸含量较高的种猪,为生产实践中快速鉴定高不饱和脂肪酸含量的猪提供了新途径,不仅有利于打开从猪脂肪中提炼多不饱和脂肪酸的市场,也有利于提高猪肉营养价值和食用品质,极大的提高了猪肉的营养和食用价值,对人类心脑血管健康,改善人类膳食健康具有重要意义。
具体实施方式
下面将结合实施例对本发明的技术方案进行清楚、完整地描述,显然,所描述的实施例是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
猪肌生成抑制素(MSTN)可以抑制成肌细胞的增殖、调控成肌细胞分化以及控制其对糖的摄取和代谢过程。MSTN基因的失活会促进骨骼肌纤维的增生和肥大,现有研究揭示,通过敲除或沉默MSTN基因可以增加瘦肉产量。本发明通过采集活体猪耳组织,提取若干猪DNA信息,并设计一对跨MSTN基因插入或缺失区域的引物,进行PCR扩增反应,对PCR产物进行测序并分析发现,猪MSTN基因中一段11bp的片段(ttgaagctttt,如SEQ ID NO.1所示)和猪脂肪酸组成相关。
本发明的实验结果揭示了猪MSTN基因中SEQ ID NO.1所示序列的片段缺失后,猪脂肪酸组成至少会发生如下改变:降低饱和脂肪酸含量、提高单不饱和脂肪酸含量、提高n-3系列多不饱和脂肪酸在脂肪酸中的占比和提高n-6系列多不饱和脂肪酸在脂肪酸中的占比。
基于上述实验结果,根据本发明的一个方面,本发明提供了一种猪脂肪酸组成相关的分子标记,该分子标记为位于猪MSTN基因内含有如SEQ ID NO.1所示序列的片段。通过检测上述分子标记,可以预估待检测猪的脂肪酸组。
所述猪脂肪酸组成包括猪饱和脂肪酸含量、单不饱和脂肪酸含量、n-3系列多不饱和脂肪酸在脂肪酸中的占比和n-6系列多不饱和脂肪酸在脂肪酸中的占比。不存在上述分子标记的猪,饱和脂肪酸含量更低,单不饱和脂肪酸含量更高,n-3和n-6系列多不饱和脂肪酸在脂肪酸中的占比分别更高。
在一些优选的实施方式中,所述猪脂肪酸组成包括猪背最长肌中脂肪酸组成、猪板油中脂肪酸组成和猪皮下脂肪中脂肪酸组成。猪脂肪主要包括板油和皮下脂肪。其中,板油一般加工后作为食品用油,以及制成药物;皮下脂肪除炼猪油外,也可以被收集起来作为化工原料。
根据本发明的一个方面,本发明提供了一种猪脂肪酸组成相关的基因,所述基因为猪MSTN基因缺失上述含有如SEQ ID NO.1所示序列的片段后的基因。其中猪脂肪酸组成相关的实例包括但不限于:和降低猪饱和脂肪酸含量相关;和提高单不饱和脂肪酸含量相关;和提高n-3系列多不饱和脂肪酸在脂肪酸中的占比相关;以及和提高n-6系列多不饱和脂肪酸在脂肪酸中的占比相关。含有上述基因的猪,饱和脂肪酸含量更低,单不饱和脂肪酸含量更高,n-3和n-6系列多不饱和脂肪酸在脂肪酸中的占比分别更高。其中猪脂肪酸组成优选包括猪背最长肌中脂肪酸组成、猪板油中脂肪酸组成和猪皮下脂肪中脂肪酸组成。
本发明不限制未缺失含有如SEQ ID NO.1所示序列的片段的野生型的猪MSTN基因序列,在一些优选的实施方式中,未缺失含有如SEQ ID NO.1所示序列的片段的野生型的猪MSTN基因表达的蛋白的氨基酸序列如SEQ ID NO.4所示;在一些优选的实施方式中,未缺失含有如SEQ ID NO.1所示序列的片段的野生型的猪MSTN基因的cDNA序列如SEQ ID NO.5所示。
在一些优选的实施方式中,所述猪脂肪酸组成相关的基因表达的蛋白的氨基酸序列如SEQ ID NO.2所示。在一些优选的实施方式中,所述猪脂肪酸组成相关的基因的cDNA序列如SEQ ID NO.3所示。
根据本发明的另一个方面,本发明还提供了和上述基因相关的生物材料,所述生物材料包括表达盒、载体、重组微生物或重组细胞系。
具体的,所述表达盒含有所述猪脂肪酸组成相关的基因和功能元件,功能元件的实例包括但不限于:启动子、核糖体结合位点、转录因子结合位点和增强子等。所述载体含有上述基因,或含有上述表达盒,载体的实例包括但不限于为质粒、噬菌粒、噬菌体和病毒基因组。所述重组微生物含有上述基因、表达盒或载体;所述细胞系含有上述基因、表达盒或载体。
根据本发明的另一个方面,本发明还提供了一种调控猪脂肪酸组成的方法,该方法包括使猪表达上述猪脂肪酸组成相关的基因。所述调控猪脂肪酸组成包括:(a)降低猪饱和脂肪酸含量;(b)提高单不饱和脂肪酸含量;(c)提高n-3系列多不饱和脂肪酸在脂肪酸中的占比;和(d)提高n-6系列多不饱和脂肪酸在脂肪酸中的占比。可以理解的是,所述调控猪脂肪酸组成的方法也不排除通过结合其他分子生物学方法,例如缺失其他生理功能的片段,或过表达其他生理功能的基因,以达到实现上述(a)、(b)、(c)和(d)中一种或几种的目的:例如但不限于只提高单不饱和脂肪酸含量,只提高n-3系列多不饱和脂肪酸在脂肪酸中的占比,或降低猪饱和脂肪酸含量的同时提高单不饱和脂肪酸含量。
根据本发明的另一个方面,本发明还提供了上述分子标记、上述猪脂肪酸组成相关的基因、上述生物材料或上述调控猪脂肪酸组成的方法的应用。通过鉴定MSTN基因上含有如SEQ ID NO.1所示序列的插入或缺失来确定猪个体的基因型,可以选育或辅助选育不饱和脂肪酸含量较高的种猪,为生产实践中快速鉴定高不饱和脂肪酸含量的猪提供了新途径,不仅有利于打开从猪脂肪中提炼不饱和脂肪酸的市场,例如从猪板油和皮下脂肪中提炼工业用不饱和脂肪酸,也有利于提高猪肉营养价值和食用品质,极大的提高了猪肉的营养和食用价值,对人类心脑血管健康,改善人类膳食健康具有重要意义。因此本发明提供的上述分子标记、上述猪脂肪酸组成相关的基因、上述生物材料或上述调控猪脂肪酸组成的方法可应用于鉴定猪脂肪酸组成、选育种猪、制备不饱和脂肪酸和制备包括猪肉原料的食品等领域中。
下面结合优选实施例进一步说明本发明的技术方案和有益效果。
实施例
采集活体猪耳组织,使用试剂盒提取基因组DNA信息,设计一对跨MSTN基因插入或缺失区域的引物,进行PCR扩增反应,使用1.5%琼脂糖凝胶电泳检测PCR所得产物,将电泳结果显示符合片段预期大小的特异性明亮条带送公司测序,得到片段ttgaagctttt(SEQ IDNO.1)为大白猪群体中MSTN基因插入或缺失的片段。未缺失该片段的MSTN基因的cDNA序列如SEQ ID NO.5所示;缺失该片段的MSTN基因的cDNA序列如SEQ ID NO.3所示。
根据测序结果鉴定大白猪群体中MSTN基因片段缺失个体(InDel-11)和MSTN基因未缺失个体(InDel+11),选取MSTN基因片段插入或缺失个体猪各10头,进行集中屠宰和脂肪酸测定,然后统计两组猪的猪肉、板油和皮下脂肪中各脂肪酸含量之间的差异。
所有测量数据均以Mean±SEM表示。在未获得正态性时,采用非配对t检验比较片段插入和片段缺失两组猪的各脂肪酸数据。显著性水平设为0.05。数据分析使用统计软件包GraphPad Prism version 6.02 for windows(GraphPad Software,La Jolla,California)。实验结果如表1-表3所示:

从上述实验结果可以看出,缺失11bp片段的MSTN基因对大白猪产生如下影响:
(1)对于大白猪背最长肌(如表1所示),显著降低饱和脂肪酸含量,极显著提高单不饱和脂肪酸(MUFA)含量,n-3和n-6的所占脂肪酸比例分别得到显著和极显著提高。
(2)对于大白猪板油(如表2所示),极显著降低饱和脂肪酸含量,极显著提高单不饱和脂肪酸含量,n-3和n-6的所占脂肪酸比例分别极显著提高。
(3)对于大白猪皮下脂肪(如表3所示),显著降低饱和脂肪酸含量,极显著提高单不饱和脂肪酸含量,n-3和n-6的所占脂肪酸比例分别极显著提高。
本实施例中,猪基因组DNA的提取和检测的方法如下:
(1)剪取适量的猪耳组织,剪碎将其放进1.5ml的Axgen管中。
(2)取50ml BD离心管,将终浓度为0.4mg/ml的蛋白酶K和裂解缓冲液进行混合均匀,向装有猪耳朵组织的1.5ml的离心管中加入0.5ml裂解液进行裂解。
(3)离心管平行均匀的放置于恒温杂交炉的摇板上(严封管盖,以免液体外渗)。55度放置6个小时以上(消化过程中样品的充分混合是非常重要的。判断依据是无明显肉眼可见的猪耳组织,消化后的混合液成乳状);
(4)待样品充分裂解后,取出离心管,在每管中加入0.3ml饱和氯化钠溶液,进行颠倒充分混合6-8次,然后放于冰上,冰浴15分钟;
(5)冰浴过后,12000rpm室温离心15分钟,小心缓慢的将上清转移至新的1.5mlAxgen离心管中(小心避免沉淀随上清一起倒出,保持倒的手法要一致,保持每管倒得上清在量上保持相同);
(6)加入0.7ml的异丙醇(加入异丙醇的量随倒出的上清的量的变化而变化,二者等体积)于各管,颠倒直至溶液中有絮状沉淀出现(若无絮状沉淀,可将溶液-20℃冰箱放置2小时或4℃冰箱放置过夜);
(7)12000rpm室温离心15分钟,去掉上清液体(此过程中留意观察离心管底部的白色DNA沉淀,切勿将其与上清一起倒掉);
(8)在各离心管中加入0.5ml 70%乙醇,轻轻颠倒以充分冲洗上述沉淀出来的DNA;
(9)10000rmp离心30s,用200μl微量移液枪将离心管中的乙醇吸掉,将沉淀的DNA留在管中(当心勿将管中的DNA沉淀吸走,此步骤所用的枪头在各离心管间操作时可不用更换);
(10)自然风干DNA 10分钟;
(11)移液枪取0.1ml的TE缓冲液于各管中,将上述DNA沉淀重新溶解,置其于55℃放置2小时,定时摇晃数次,以使DNA充分溶解;
(12)待DNA充分溶解后,用紫外分光光度计测定所提浓度,并琼脂糖凝胶电泳检测所提质量。
(13)将DNA溶液放置4℃过夜,次日取1μl进行PCR。(若数星期内使用所提DNA,可在4℃保存;如若长时间不使用,需放置于-20℃环境下)
本实施例中,猪肉、板油和皮下脂肪中各脂肪酸含量测定方法如下:
取每头猪的背最长肌、板油和皮下脂肪各100g肉样冻干除水,在18℃、68%湿度条件下,利用剩余冻干粉测定脂肪酸含量,操作和方法参照《食品中脂肪酸的测定,GB5009.168-2016》中的水解-提取法。
水解-提取法:加入内标物的试样经水解-乙醚溶液提取其中的脂肪后,在碱性条件下皂化和甲酯化,生成脂肪酸甲酯,经毛细管柱气相色谱分析,内标法定量测定脂肪酸甲酯含量。依据各种脂肪酸甲酯含量和转换系数计算出总脂肪、饱和脂肪(酸)、单不饱和脂肪(酸)、多不饱和脂肪(酸)含量、n-3含量、n-6含量等。

1.猪脂肪酸组成相关的分子标记,所述分子标记为位于猪MSTN基因内含有如SEQ IDNO.1所示序列的片段;
所述猪脂肪酸组成包括猪饱和脂肪酸含量、单不饱和脂肪酸含量、n-3系列多不饱和脂肪酸在脂肪酸中的占比和n-6系列多不饱和脂肪酸在脂肪酸中的占比。
2.根据权利要求1所述的分子标记,其特征在于,所述猪脂肪酸组成包括猪背最长肌中脂肪酸组成、猪板油中脂肪酸组成和猪皮下脂肪中脂肪酸组成。

9.根据权利要求8所述的调控猪脂肪酸组成的方法,其特征在于,所述调控猪脂肪酸组成包括如下中的至少一种:
(a)降低猪饱和脂肪酸含量;
(b)提高单不饱和脂肪酸含量;
(c)提高n-3系列多不饱和脂肪酸在脂肪酸中的占比;
(d)提高n-6系列多不饱和脂肪酸在脂肪酸中的占比。
10.权利要求1或2所述的猪脂肪酸组成相关的分子标记、权利要求3-6任一项所述的猪脂肪酸组成相关的基因、权利要求7所述的生物材料或权利要求8或9所述的调控猪脂肪酸组成的方法在(x1)鉴定猪脂肪酸组成;(x2)选育种猪;(x3)制备不饱和脂肪酸;或(x4)制备包括猪肉原料的食品中的应用。
