Abstract
本发明公开一种兼顾浅色黄姑鱼鱼鳔品质与抗氧化能力的人工饲料,包括如下重量百分比的原料制备而成:蛋白源原料、脂肪源原料、糖源,辅料和重量百分比为0.5~2.5%的L‑脯氨酸;其中,所述饲料的营养成分按重量百分比组成如下:粗蛋白47.099~47.454%,粗脂肪10.154~10.681%,余下为碳水化合物、粗纤维、灰分和水分。本发明的人工配合饲料对浅色黄姑鱼生长和饲料利用没有显著的影响,但能显著提高鱼体蛋白含量,并显著增加了鱼鳔中胶原蛋白和Hyp含量,从而提高肌肉和鱼鳔的品质;另外,该人工配合饲料对SOD、CAT、GSH、GSH‑PX、MDA等抗氧化/氧化指标均有显著影响,从而提高了浅色黄姑的抗氧化性能。

浅色黄姑鱼(Nibea coibor),俗名白奈、金丝,其生长迅速,在人工网箱养殖状态下,一年体重可达1kg,三周年可达2.5kg以上。浅色黄姑鱼肉质细嫩,口感独特,味道鲜美,无肌间刺,是一种被广泛接受的优质食用鱼。它还有一个显著的特点就是鳔大,其鱼鳔可加工制成高级食品鱼胶,是名贵的中药补品,有非常好的滋补养颜效果,具有很高的市场价值。
鱼鳔,又名鱼泡、鱼肚、鱼白、压胞,以富有胶质著称,是四大海珍品之一(鲍鱼、海参、鱼翅和鱼肚),又有″海洋人参″之誉。鱼鳔是理想的高蛋白低脂肪食品,其蛋白质含量一般在80%(干重)以上,而脂肪含量较低。鱼鳔营养成分主要为胶原蛋白、粘多糖、氨基酸和多种维生素等成分,并且还含有一定量的Ca、Fe、Mg、Zn等矿物质元素。鱼鳔中含有的生物大分子胶原蛋白质,是人体补充、合成蛋白质的重要原料,且易于吸收和利用,它以水结合的形式贮存于人体组织中,从而改善组织的营养状况和新陈代谢。鱼鳔富含胶原蛋白,是一种传统的功能性食品,可以改善大脑功能,维持正常的内分泌状态,调节免疫功能。此外,鱼鳔具有很高的药用价值。现代中医学认为,鱼鳔味甘性平,有滋阴养血、止血、补肾固精的功能。在我国,食用鱼鳔有着悠久的历史,北魏贾思勰《齐民要术》中就有把鱼鳔进行加工食用的记载。浅色黄姑鱼的鱼鳔产品——“白奈鱼胶”(“白花胶”)相比于其他鱼胶更有营养,其药用及营养价值仅次于金钱鮸,导致白花胶在市场上供不应求。因此,浅色黄姑鱼的海水网箱人工养殖,深受闽、粤等地广大养殖户的青睐,具有良好的发展前景。
然而,目前浅色黄姑鱼养殖以投喂冰鲜野杂鱼为主,存在成本高、养殖环境污染严重等缺点。同时,冰鲜鱼营养结构不均衡,致使鱼鳔的品质不稳定。因此,对浅色黄姑鱼的营养需求进行系统的研究,开发出一种能够促进浅色黄姑鱼健康养殖,又能提高其鱼鳔品质的饲料尤为必要。然而目前,缺乏浅色黄姑鱼人工配合饲料的开发的研究,关于L-脯氨酸对浅色黄姑鱼鱼鳔品质、抗氧化性能的影响的研究更是未见报道。
发明内容
本发明的目的在于,提供一种兼顾浅色黄姑鱼鱼鳔品质与抗氧化能力的人工饲料,为浅色黄姑鱼的健康养殖、优质鱼鳔的生产提供理论指导。
为了解决上述技术问题,本发明采用如下技术方案:
一种兼顾浅色黄姑鱼鱼鳔品质与抗氧化能力的人工饲料,包括如下重量百分比的原料制备而成:蛋白源原料、脂肪源原料、糖源,辅料和重量百分比为0.5~2.5%的L-脯氨酸;其中,所述饲料的营养成分按重量百分比组成如下:粗蛋白47.099~47.454%,粗脂肪10.154~10.681%,余下为碳水化合物、粗纤维、灰分和水分。
优选的,所述L-脯氨酸的重量百分比为1.5%
脯氨酸Pro是胶原蛋白中的主要氨基酸,约占胶原蛋白氨基酸总量的25%,对胶原蛋白的生物合成、结构和强度至关重要。且Pro在蛋白质合成、结构、代谢(特别是合成精氨酸、多胺和谷氨酸代谢等)、营养、伤口愈合、抗氧化反应、免疫应答等多方面发挥重要作用。也有研究表明,当饮食中严重缺乏Pro时,会导致小鼠伤口愈合缺陷等疾病,这主要是因为Pro参与胶原蛋白合成,而伤口的愈合也与胶原形成密切相关。一般认为,在胶原合成较旺盛的组织(如鱼鳔)、幼年等生长旺盛的个体、动物受伤或产后对Pro的需要特别高。此外,近年来研究表明,Pro还是一种非常有效的抗氧化剂,可以清除超氧阴离子自由基、羟自由基等活性氧,与机体的抗氧化系统(超氧化物氧化物歧化酶SOD,过氧化氢酶CAT、抗坏血酸、还原型谷胱甘肽GSH等)协同调控细胞的活性氧平衡。许多鱼类对Pro的要求特别高,为了满足最大的生长,必须从饮食中获得足够的Pro,因而常将其归为鱼类的条件性必需氨基酸或功能性氨基酸。但是,对于鱼类来说,这样的摄入并非越多越好,过多地摄入Pro会导致鱼类生长减缓、饲料利用率降低,反而造成了蛋白质积累的减少,这可能与Pro增多导致膳食氨基酸平衡被破坏,影响了鱼类对营养物质的消化利用有关。鱼体应该摄入多少Pro最为合适,能在不影响正常生长的情况下,兼顾蛋白质积累与提高抗氧化能力,一直是亟待解决的问题。
优选的,所述蛋白源原料为鱼粉、发酵豆粕和大豆浓缩蛋白中的一种或多种;所述脂肪源原料为鱼油和卵磷脂中的一种或多种;所述糖源原料为木薯淀粉;所述辅料为自由氯化胆碱、微晶纤维素、复合维生素、复合矿物质、磷酸二氢钙、丙氨酸、混合氨基酸中的一种或几种。
优选的,所述饲料包括如下重量百分比的原料制备而成:鱼粉22~26%,发酵豆粕8~12%,大豆浓缩蛋白14~16%,鱼油6~8%,卵磷脂0-4%,木薯淀粉15~20%,混合氨基酸10~16%,微晶纤维素1~5%,氯化胆碱1%,α-淀粉2~6%,磷酸二氢钙1%,谷氨酸0.5~2.5%,复合维生素1%,混合矿物质1%,L-脯氨酸0.5~2.5%。
优选的,所述饲料包括如下重量百分比的原料制备而成:鱼粉24.48%,发酵豆粕9.8%,大豆浓缩蛋白14.2%,鱼油7.6%,卵磷脂2%,木薯淀粉16.5%,混合氨基酸12.8%,微晶纤维素1.72%,氯化胆碱1%,α-淀粉4.4%,磷酸二氢钙1%,丙氨酸1%,复合维生素1%,混合矿物质1%,L-脯氨酸1.5%
优选的,所述饲料通过如下方法制备:
将除所述鱼油、所述卵磷脂外的其他原料及辅料预先粉碎并过80~120目筛,混合后加入水分,混匀后膨化制粒,膨化温度为100~120℃,最后将所述卵磷脂溶于所述鱼油并混匀后,喷涂在膨化粒表面,完全吸收后,即得所述饲料成品。卵磷脂,又称为蛋黄素,被誉为与蛋白质、维生素并列的"第三营养素"。卵磷脂属于一种混合物,其构成成分包括磷酸、胆碱、脂肪酸、甘油、糖脂、甘油三酸酯以及磷脂(如磷脂酰胆碱、磷脂酰乙醇胺和磷脂酰肌醇)。卵磷脂在动物体中起着重要的作用,在卵磷脂充足时,动物的新陈代谢能力才会更强,而且也更富有生命力,适量的卵磷脂可以起到很好的保护肝脏和防止脂肪肝、同时还能抗氧化和延缓衰老等。但过多的摄食卵磷脂也会导致一些副作用,如消化不良、食欲减退、血脂降低影响生长等。在本发明的饲料中饲料添加卵磷脂,可以与Pro协同增强浅色黄姑鱼的抗氧化能力。
卵磷脂和鱼油一样不耐高温,易氧化,若在膨化阶段前加入,则容易在膨化过程因高温氧化而丧失营养价值。卵磷脂不溶于水,但与鱼油的相容性甚佳,因此一般在添加时先将卵磷脂溶于液态的鱼油,然后在饲料加工过程中通过油脂喷涂到饲料表面,自然吸收风干。
一种上述兼顾浅色黄姑鱼鱼鳔品质与抗氧化能力的人工饲料的应用,用于提高浅色黄姑鱼鱼鳔品质与抗氧化能力。
实施本发明,具有如下有益效果:
本发明将适当比例的L-脯氨酸添加入浅色黄姑鱼配合饲料中,对浅色黄姑鱼的生长没有显著影响,但可以促进蛋白质沉积、显著提高鱼体蛋白含量,并显著增加了鱼鳔中胶原蛋白和Hyp含量,从而提高肌肉和鱼鳔的品质;另外,该人工配合饲料对SOD、CAT、GSH、GSH-PX、MDA等氧化指标均有显著影响,从而对浅色黄姑鱼表现出一定的抗氧化活性,为浅色黄姑鱼人工配合饲料的开发及优质鱼鳔的生产提供理论指导。

实施例1
一种兼顾浅色黄姑鱼鱼鳔品质与抗氧化能力的人工饲料,包括如下重量百分比的原料制备而成:鱼粉24.6%,发酵豆粕9%,大豆浓缩蛋白15%,鱼油6%,卵磷脂4%,木薯淀粉16%,混合氨基酸12%,微晶纤维素3%,氯化胆碱1%,磷酸二氢钙1%,α-淀粉4.4%,丙氨酸1.5%,复合维生素1%,混合矿物质1%,L-脯氨酸0.5%。
实施例2
一种兼顾浅色黄姑鱼鱼鳔品质与抗氧化能力的人工饲料,包括如下重量百分比的原料制备而成:鱼粉25%,发酵豆粕12%,大豆浓缩蛋白14%,鱼油8%,卵磷脂2%,木薯淀粉15%,混合氨基酸10%,微晶纤维素3%,氯化胆碱1%,磷酸二氢钙1%,α-淀粉4.5%,丙氨酸1.5%,复合维生素1%,混合矿物质1%,L-脯氨酸1%。
实施例3
一种兼顾浅色黄姑鱼鱼鳔品质与抗氧化能力的人工饲料,包括如下重量百分比的原料制备而成:鱼粉25%,发酵豆粕10%,大豆浓缩蛋白15%,鱼油6%,卵磷脂2.6%,木薯淀粉18%,混合氨基酸10%,微晶纤维素2%,氯化胆碱1%,磷酸二氢钙1%,α-淀粉4.4%,丙氨酸1.5%,复合维生素1%,混合矿物质1%,L-脯氨酸1.5%。
实施例4
一种兼顾浅色黄姑鱼鱼鳔品质与抗氧化能力的人工饲料,包括如下重量百分比的原料制备而成:鱼粉24.48%,发酵豆粕9.8%,大豆浓缩蛋白14.2%,鱼油7.6%,卵磷脂2%,木薯淀粉16.5%,混合氨基酸12.8%,微晶纤维素1.72%,氯化胆碱1%,α-淀粉4.4%,磷酸二氢钙1%,丙氨酸0.5~2.5%,复合维生素1%,混合矿物质1%,L-脯氨酸0.5~2.5%。
实施例5
本发明饲料的效果检测:
养殖实验在广东省汕头市南澳县汕头大学海洋生物临海实验站中进行。浅色黄姑鱼购自当地的一个苗种孵化场(中国广东饶平)。450条试验鱼(8.64±0.138g)被随机分入18个网箱(1m×1m×1.5m,长×宽×深),有盖网,以防逃逸和水鸟。按实施例4的配方(表1)添加对应量的L-脯氨酸,三个网箱一组被随机分配到六种饮食处理组。设计L-脯氨酸含量分别为0、0.5、1、1.5、2、2.5%六个梯度的饲料,实测饲料中脯氨酸含量分别为0.828%,1.291%,1.636%,2.098%,2.460%和2.828%,编号为P1~P6,饲料配方如表1所示(数值为百分比),其氨基酸组成如表2所述。在8周的饲养期内,每天人工投饵两次,同时记录好饲料投喂量,称重记录生长情况、观察记录鱼的健康状况及环境变化等,在实验过程中,温度23℃~30℃,pH值7.8~8.1,氨氮低于0.05mg L-1,盐度31~33gL-1,溶解氧5.2~6mg L-1。

养殖试验结束后,对每个试验组所有鱼进行称重,计算鱼的生长和饲料利用率见表3,每个网箱随机选取4尾鱼储存在-20℃冰箱中用于体成分分析。另外随机从每个重复中选取6尾鱼,测量个体体重、体长,并用于血液样本的采集,血液利用尾静脉采血法抽取,静置离心分离后,取上层血清存于-20℃,用于血清生化及抗氧化测定。采血后的试验鱼进行屠宰,称量肝脏重量和内脏重量,计算肥满度、肝体指数和脏体指数,同时采集组织样本(肝脏和鱼鳔),刮净表面脂肪后装于密封袋中,-20℃冰箱保存,以备测定鱼鳔营养成分及鱼鳔胶原蛋白含量。

由表3的生长指标可以看出,各组浅色黄姑鱼增重率、特定生长率、饲料利用率、蛋白质效率、肥满度及鳔体比基本不受膳食Pro添加量的影响,说明饲料中添加Pro不会影响影响浅色黄姑鱼生长性能。

由表4的全鱼体成分分析得出,Pro对全鱼水分、粗脂肪和粗灰分无显著影响,但L-脯氨酸可显著提高全鱼粗蛋白含量,且呈现出先升高后降低的趋势,最大值为P4组。以饲料中所添加的L-脯氨酸含量(百分比)为自变量,浅色黄姑鱼全鱼蛋白含量为因变量,通过分段回归分析得到,在基础饲料(P1组)中添加1.473%(干物质)Pro时,全鱼中蛋白积累量最高(如附图1所示)。说明饲料中添加Pro可以提高浅色黄姑鱼蛋白的沉积,在基础日粮基础上以添加1.473%(干物质)Pro时效果最佳。

由表5和表6,浅色黄姑鱼血液及肝脏生化指标可以看出,摄食不同水平Pro饲料后,浅色黄姑鱼的许多生化指标均发生了显著的变化。其中抗氧化指标受影响较大,血清GSH、GSH-PX、SOD、MDA、CAT和肝脏GSH、GSH-PX、CAT均受膳食Pro水平的显著影响,其中血清GSH、GSH-PX、SOD、CAT(表5)和肝脏GSH、GSH-PX、CAT(表6)随膳食Pro水平的增加有增加的趋势(P<0.05);相反血清、肝脏MDA水平随膳食Pro水平的增加有降低的趋势(表6)(P<0.05),说明饲料中添加Pro可以显著提高浅色黄姑鱼的抗氧化性能。而在这几个与抗氧化性能直接相关的指标中,血清里的SOD值、肝脏里的GSH值分别最早出现显著提高(P<0.05)(如附图2所示),此时Pro的添加量为1.5%(干物质)。
超氧化物歧化酶(Superoxide Dismutase,SOD)是生物体内存在的一种抗氧化金属酶,它能够催化超氧阴离子自由基歧化生成氧和过氧化氢,在机体氧化与抗氧化平衡中起到至关重要的作用,与很多疾病的发生、发展密不可分。SOD的催化作用是通过金属离子Mn+1(氧化态)和Mn(还原态)的交替电子得失实现的。一般认为超氧阴离子自由基首先与金属离子形成内界配合物,Mn+1被体内的超氧阴离子自由基还原为Mn,同时生成O2,Mn又被HO2·氧化为Mn+1,同时生成H2O2。而SOD又被氧化为初始氧化态的SOD。最后,H2O2在过氧化氢酶的作用下,被催化分解为水(H2O)和O2。
谷胱甘肽(glutathione,r-glutamyl cysteingl+glycine,GSH)是一种含γ-酰胺键和巯基的三肽,由谷氨酸、半胱氨酸及甘氨酸组成。GSH作为一种细胞内重要的调节代谢物质,其既是甘油醛磷酸脱氢酶的辅基,又是乙二醛酶及丙糖脱氢酶的辅酶,参与体内三羧酸循环及糖代谢,并能激活多种酶,如巯基(SH)酶-辅酶等,从而促进糖类、脂肪和蛋白质代谢。GSH分子特点是具有活性巯基(-SH),是最重要的功能集团,可参与机体多种重要的生化反应,保护体内重要酶蛋白巯基不被氧化、灭活,保证能量代谢、细胞利用。同时,其通过巯基与体内的自由基结合,可直接使自由基还原成酸性物质,从而加速自由基的排泄,并对抗自由基对重要脏器的损害。Haddad等研究发现,GSH参与了脂多糖诱导的细胞因子转录的调节及I-KB/NF-KB信号通路的调节。Armstrong等发现,GSH含量的降低是一种潜在的凋亡早期激活信号,随后产生的氧自由基促使细胞发生凋亡。
在血清里的SOD值、肝脏里的GSH值分别出现显著提高的情况下,可判断浅色黄姑鱼整体抗氧化性能也得到显著提高。说明在基础饲料中(P1)添加Pro达到1.5%附近,可在一定程度上明显提高浅色黄姑鱼的抗氧化性能。

由表7鱼鳔胶原蛋白含量显示,浅色黄姑鱼血浆游离Hyp随膳食Pro水平的增加而轻微增加。肝脏组织Hyp、鱼鳔组织Hyp和鱼鳔胶原蛋白含量均随膳食Pro水平增加先升高后降低,且在P4组达到最高值。以鱼鳔胶原蛋白含量评价膳食Pro的最适添加量,通过分段回归分析发现(如附图3所示),浅色黄姑鱼饲料中膳食Pro最适添加量为1.488%(干物质),表明在本实验的养殖条件下,在基础饲料(P1组)中添加1.488%(干物质)Pro时,鱼鳔中胶原蛋白积累量最高。
综合以上各指标,可知本饲料配方可以提高浅色黄姑鱼抗氧化性能,促进全鱼蛋白沉积,也增加了鱼鳔中胶原蛋白的沉积,进而提高了浅色黄姑鱼鱼鳔品质。尤其当饲料中的Pro添加量达到1.5%附近时,浅色黄姑鱼的鱼鳔品质与抗氧化能力同时得到兼顾:即抗氧化性能显著提高,且鱼鳔中的粗蛋白和胶原蛋白积累量达到最高。
以上所揭露的仅为本发明的较佳实施例而已,当然不能以此来限定本发明之权利范围,因此依本发明权利要求所作的等同变化,仍属本发明所涵盖的范围。

1.一种兼顾浅色黄姑鱼鱼鳔品质与抗氧化能力的人工饲料,其特征在于,包括如下重量百分比的原料制备而成:蛋白源原料、脂肪源原料、糖源,辅料和重量百分比为0.5~2.5%的L-脯氨酸;其中,所述饲料的营养成分按重量百分比组成如下:粗蛋白47.099~47.454%,粗脂肪10.154~10.681%,余下为碳水化合物、粗纤维、灰分和水分。
2.根据权利要求1所述兼顾浅色黄姑鱼鱼鳔品质与抗氧化能力的人工饲料,其特征在于,所述L-脯氨酸的重量百分比为1.5%。
