当今社会中微生物的生长可在必须控制微生物水平的应用中呈现严重的问题。在健康产业以及水输送、处理和过滤等应用中,微生物生长至不健康的水平会导致普遍的疾病。另外,水过滤和输送装置中微生物的生长也会导致生物污染,从而缩短设备的有效寿命。在加热、通风和空气调节(HVAC)系统中,如果不进行处理,在潮湿空气管道中繁殖的微生物会导致难闻的气味和健康问题。此外,对于水中的容器,容器湿润区域上微生物的不受控制的生长会通过破坏壳体形状和产生阻力而降低壳体的流体动力学效率,从而降低燃料效率。

抗微生物膜

在一些实施方式中,石墨烯材料与交联剂材料的质量比可以为约1:1000至约10:1。在一些实施方式中,石墨烯材料与聚合物材料的质量比可以为约1:100至约5:1,或约3:100。
在一些实施方式中,支撑物可以是膜的一部分。这种支撑物的非限制性实例包括反渗透膜、带材或可用作基材的柔性或非柔性的任何物质。在一些实施方式中,支撑物材料可以是聚合物。在一些实施方式中,支撑物材料可包含中空纤维。在其他实施方式中,支撑物可以是要保护免于微生物生长的物品。在一些实施方式中,要保护的物品可以是不期望生物生长的任何物品。实例包括但不限于船体、处理池、管道、脱盐过滤器、空气过滤器、HVAC系统部件、医院设备和装饰、台面、盥洗室装饰等。

在一些实施方式中,膜的选择性渗透性可以反映在水蒸气的渗透率与至少一种选定气体(例如氧气和/或氮气)的渗透率的比率上。在一些实施方式中,膜可以表现出水蒸气渗透率与气体渗透率之比,例如WVTR/OTR,大于50,大于100,大于200,和/或大于400。在一些实施方式中,选择性渗透性可以是在上述水平下水蒸气:气体渗透率/转移率之比的量度。上文已经公开了确定水蒸气渗透率和/或气体渗透率的合适方法。
在一些实施方式中,膜可具有抗微生物特性,或阻止工作流体中微生物的生长。在一些实施方式中,生长被阻止的微生物可包含escherichia coli(8739,美国模式培养物保藏所(ATCC),Manassas,VA USA)。在一些实施方式中,膜可具有2.0或更高的抗菌效力。抗菌效力可通过标准JIS Z 2801(2010)测定。在一些实施方式中,工作流体可以是液体、气体或其组合(例如,饱和空气)。液体工作流体的非限制性实例可以是脱盐设备中的卤水/盐水或淡水、废物处理设备中的水、船舶的海水、HVAC系统中的空气或封闭空间中的空气。
在一些实施方式中,抗微生物膜可以设置在要保护的物体和流体贮存器之间。在一些实施方式中,流体贮存器可含有微生物。在一些实施方式中,膜可以阻止膜上微生物的生长。
在抗微生物元件的一些实施方式中,复合材料涂层还包含足够的酸以实现水解缩合。在一些实施方式中,可以将约0.05ml至约5ml的1N HCl加入到约1gm至约20gm的0.01重量%氧化石墨烯水性分散体,约1gm至约20g的10重量%PVA水溶液,相对于PVA约5重量%的TEOS。例如,可以将11.5g 0.01重量%氧化石墨烯水性分散体添加到11.5g 10重量%PVA水溶液(Aldrich,St.Louis,MO,USA);0.065克或相对于PVA 5重量%的TEOS(Aldrich);和0.2mL 1N HCl水溶液(Aldrich)的混合物中。
在一些实施方式中,抗微生物元件还可包含分散剂。在一些实施方式中,分散剂可以是铵盐,例如NH4Cl;Flowlen;鱼油;长链聚合物;硬脂酸;氧化鲱鱼油(MFO);二羧酸,例如但不限于琥珀酸,乙二酸,丙二酸,戊二酸,己二酸,庚二酸,辛二酸,壬二酸,癸二酸,邻苯二甲酸和对苯二甲酸;脱水山梨糖醇单油酸酯;及其混合物。一些实施方式使用氧化的MFO作为分散剂。
在一些实施方式中,抗微生物元件还可包含增塑剂。在一些实施方式中,增塑剂可以是1型增塑剂,其通常可以降低玻璃化转变温度(Tg),例如使其更加柔韧,邻苯二甲酸酯(邻苯二甲酸正丁酯,邻苯二甲酸二丁酯,邻苯二甲酸二辛酯,邻苯二甲酸丁基苄基酯,缺失的酯(missed esters)和邻苯二甲酸二甲酯);和/或2型增塑剂,其可以使成为更柔韧、更易变形的层,并且可能减少由层压产生的空隙的量,例如二醇类(聚乙烯;聚亚烷基;聚丙烯;三亚乙基;二丙基二醇苯甲酸酯)。
1型增塑剂可包括但不限于邻苯二甲酸丁基苄酯,基于二羧酸/三羧酸酯的增塑剂,例如但不限于基于邻苯二甲酸酯的增塑剂,例如但不限于邻苯二甲酸二(2-乙基己基)酯,邻苯二甲酸二异壬酯,邻苯二甲酸二(正丁基)酯,邻苯二甲酸丁基苄酯,邻苯二甲酸二异癸酯,邻苯二甲酸二正辛酯,邻苯二甲酸二异辛酯,邻苯二甲酸二乙酯,邻苯二甲酸二异丁酯,邻苯二甲酸二正己酯及其混合物;基于己二酸酯的增塑剂,例如但不限于己二酸双(2-乙基己基)酯,己二酸二甲酯,己二酸单甲酯,己二酸二辛酯及其混合物;基于癸二酸酯的增塑剂,例如但不限于癸二酸二丁酯和马来酸酯。
2型增塑剂可包括但不限于马来酸二丁酯,马来酸二异丁酯及其混合物,聚亚烷基二醇,例如但不限于聚乙二醇,聚丙二醇及其混合物。可以使用的其他增塑剂包括但不限于苯甲酸酯,环氧化植物油;磺酰胺,例如但不限于N-乙基甲苯磺酰胺,N-(2-羟丙基)苯磺酰胺,N-(正丁基)苯磺酰胺;有机磷酸酯,例如但不限于磷酸三甲苯酯,磷酸三丁酯;二醇类/聚醚类,例如但不限于三乙二醇二己酸酯,四乙二醇二庚酸酯及其混合物;柠檬酸烷基酯,例如但不限于柠檬酸三乙酯,柠檬酸乙酰基三乙酯,柠檬酸三丁酯,柠檬酸乙酰基三丁酯,柠檬酸三辛酯,柠檬酸乙酰基三辛酯,柠檬酸三己酯,柠檬酸乙酰基三己酯,柠檬酸丁酰基三己酯,柠檬酸三甲酯,烷基磺酸苯酯和及其混合物。
在一些实施方式中,溶剂也可以存在于抗微生物元件中。用于制造材料层的溶剂包括但不限于水,低级烷醇,例如但不限于乙醇,甲醇,异丙醇,二甲苯,环己酮,丙酮,甲苯和甲基乙基酮,以及它们的混合物。一些实施方式使用二甲苯和乙醇的混合物作为溶剂。
在一个实施方式中,如图1所示,抗微生物膜100可至少包含基材元件120和前述复合材料涂层110。该涂层暴露于工作流体130。在一些实施方式中,如图2和3所示,基材120可包含要保护的物品。在图2中,要保护的物品是反渗透膜,膜位于膜的表面上。在图3中,要保护的物品是船体,膜是船体上的涂层。由于膜的存在,可以阻止所述膜和受保护物品上微生物的生长。
在一些实施方式中,可以在抗微生物膜100中包含增强或改善膜100与工作流体130的相互作用的材料。在一些实施方式中,添加的材料或间隔材料可以改善工作流体在膜100上方或穿过膜100的通量或移动。在一些实施方式中,添加的材料在抗微生物膜100内产生空间或体积。在一些实施方式中,添加的材料产生或增强抗微生物膜100表面的粗糙度或不规则性。在一些实施方式中,添加的材料是二氧化硅,例如二氧化硅纳米颗粒,或产生期望的流体通量或表面构造的其它合适材料。在利用二氧化硅颗粒或纳米颗粒的一些实施方式中,颗粒的尺寸可以为1nm至500nm、40nm至300nm或70nm至250nm。在一些实施方式中,粒度为5nm、7nm、10nm、20nm、60nm、80nm、100nm、120nm、140nm、160nm、180nm、200nm或220nm。另外,具有相似尺寸和性能特征的其他纳米颗粒包括Fe3O4、TiO2、ZrO2或Al2O3的纳米颗粒。在一些实施方式中,相对于复合材料涂层110的总重量,间隔材料具有约1%至约10%的重量百分比。在一些实施方式中,相对于复合材料涂层110的总重量,间隔材料具有约6%或约6.6%的重量百分比。
在一些实施方式中,复合材料涂层110的厚度为约10nm至约10μm。复合材料涂层110的厚度可以为50nm、100nm、110nm、150nm、180nm、200nm、220nm、300nm、400nm、500nm、600nm、1μm、1.4μm、5μm或接近这些值或介于这些值之间的任何值。在一些实施方式中,厚度小于约20μm、小于约15μm、小于10μm或小于约5μm。在一些实施方式中,复合材料涂层110的厚度不足以自支撑。换句话说,在一些实施方式中,复合材料涂层110必须施加或粘附到支撑结构或表面,例如基材元件120。
在一些实施方式中,通过下述方法来制备膜100:将复合材料涂层110施加到基材元件120上,然后将所得膜暴露于升高的温度一段时间。在一些实施方式中,该方法使膜100固化。在一些实施方式中,在施加到基材元件120之后,允许复合材料涂层110风干一段时间,然后暴露于升高的温度。在一些实施方式中,升高的温度的范围为约30℃至约300℃,约60℃至约200℃,或约70℃至约150℃。在一些实施方式中,升高的温度为约70℃,约85℃,约90℃,约130℃,约140℃,或接近这些值或在这些值之间的任何值。在一些实施方式中,暴露时间为约1分钟至约180分钟、约2分钟至约150分钟、约3分钟至约120分钟。在一些实施方式中,暴露时间为约3分钟、约6分钟、约8分钟、约20分钟、约30分钟、约60分钟、约90分钟、约120分钟或接近这些值或在这些值之间的任何值。
防止微生物生长的方法
在一些实施方式中,可描述防止表面上微生物生长的方法,如图4中所示。在一些实施方式中,该方法可包括提供任何前述抗微生物膜。在一些实施方式中,提供任何前述膜可包括用任何所述膜涂布的要保护的表面。在一些实施方式中,膜可包含复合材料涂层。在一些实施方式中,膜可包含支撑物和在支撑物材料上的复合材料涂层。在一些实施方式中,复合材料涂层可包含氧化石墨烯和交联剂。在一些实施方式中,交联剂可以是聚乙烯醇。在一些实施方式中,氧化石墨烯与交联剂的质量比可以为1:1000至10:1。在一些实施方式中,石墨烯材料与交联剂的质量比可以为约1:100至约5:1,或约3:100。在一些实施方式中,支撑物可以是多孔的。在其他实施方式中,支撑物可以是无孔的。在一些实施方式中,支撑物可以是涂层的一部分。在其他实施方式中,支撑物可以与涂层分离。在一些实施方式中,当支撑物与涂层分离时,支撑物可包含要保护免于微生物生长的物品。实例包括但不限于船体、处理池、管道、脱盐过滤器、空气过滤器、HVAC系统部件、医院设备和装饰、台面、盥洗室装饰等。
在一些实施方式中,所述方法还包括将膜暴露于工作流体。在一些实施方式中,工作流体可含有微生物,其中由于暴露于工作流体,膜阻止微生物生长。在一些实施方式中,受控微生物可包含escherichia coli(8739,ATCC)。在一些实施方式中,膜可具有2.0或更高的抗菌效力。抗菌效力可通过标准JIS Z 2801(2012)测定。在一些实施方式中,工作流体可包括空气。在一些实施方式中,工作流体可包括水。在一些实施方式中,工作流体可包括空气和水蒸气的混合物。在一些实施方式中,空气和水蒸气的混合物可以在约100%相对湿度至约0%相对湿度的范围内。
实施例
已经发现,本文所述的抗微生物膜元件的实施方式具有改善的对微生物的抗性。通过以下实施例进一步显示这些益处,这些实施例旨在说明本公开的实施方式,但不旨在以任何方式限制范围或基本原理。
实施例1.1:制备氧化石墨烯
GO制备:使用改进的Hummers方法由石墨制备GO。将石墨片(2.0g,Aldrich,100目)在NaNO3(2.0g)、KMnO4(10g)和浓缩的98%H2SO4(96mL)的混合物中在50℃下氧化15小时。然后将得到的糊状混合物倒入冰(400g)中,随后加入30%过氧化氢(20mL)。将所得溶液搅拌2小时以还原二氧化锰,通过滤纸过滤,并用去离子(DI)水洗涤。收集固体并通过搅拌分散在DI水中,然后以6300rpm离心40分钟,并倾析出水层。将剩余的固体分散在去离子水中,并重复洗涤过程4次。然后通过超声处理(20W)2.5小时将纯化的GO分散在DI水中,得到了GO分散体(0.4重量%)。
实施例1.2:制备抗微生物元件
GO-PVA膜(AM-1)制备:利用去离子水将4mg/mL如实施例1.1中所述制备的氧化石墨烯(GO)水性分散体稀释至0.1重量%。然后,将32.0g所得的0.1重量%氧化石墨烯水性分散体添加到由40.0g 2.5重量%PVA水溶液(Aldrich)组成的混合物中。随后将所得混合物在室温下搅拌10分钟。通过以0.6g/90cm2将溶液滴在膜表面上,将所得溶液浇铸到反渗透(RO)膜(ESPA膜,Hydranautics)上。在空气中干燥后,将膜置于85℃的烘箱中30分钟以除去水并使膜交联,得到了厚度为1.4μm、GO/PVA的质量比分别为3.1重量%和96.9重量%(或1:31.26)的膜,或AM-1。
对比例1.1:制备原料基材
Hydranautics膜(CM-1)制备:在另一个实施例(CM-1)中,从来自Hydranautics的原料反渗透(RO)膜(ESPA膜,Hydranautics)上切下膜。
实施例2.1:测量抗微生物特性
为了检测膜的抗微生物特性,使用符合用于检测抗微生物产品效力的日本工业标准(JIS)Z 2801:2012(英文版,2012年9月出版)的程序测量实施例AM-1,所述文件整体并入本文中。用于验证抗微生物能力的生物是escherichia coli.(8739,ATCC)。
对于测试,通过下述方法来制备培养液:将8g营养粉(DifcoTM Nutrient Broth,Becton,Dickinson and Company,Franklin Lakes,NJ USA)悬浮于1L过滤的无菌水中,充分混合,然后加热并频繁搅动。为了溶解粉末,将混合物煮沸1分钟,然后在121℃下高压灭菌15分钟。测试前一晚,将escherichia coli.加入2-3mL制备的培养液中并培养过夜。
在测试当天,将所得培养物在新鲜培养基中稀释,然后使其生长至密度为108CFU/mL(或大致将1mL培养物稀释在9mL新鲜营养培养液中)。随后使所得溶液再生长2小时。然后将再生长物在无菌盐水(在1L蒸馏水中的NaCl 8.5g(Aldrich))稀释50倍,以达到约2x106CFU/mL的预期密度。50μL稀释物提供接种数。
然后将样品切成1英寸×2英寸的方块,并置于培养皿中,GO涂层面朝上。随后取50μL稀释物并接种试样。然后使用透明覆盖膜(0.75英寸×1.5英寸,3M,St.Paul,MN USA)来帮助扩散细菌接种物,确定扩散尺寸并减少蒸发。然后,用透明盖盖住培养皿,并静置以使细菌能够生长。
当达到2小时和24小时的期望测量点时,用无菌镊子将试样和覆盖膜转移到含有20mL盐水的50mL锥形管中,并通过将它们在涡旋混合器(120V,VWR Arlington Heights,ILUSA)中混合至少30秒来洗掉每个样品的细菌。然后使用泵(MXPPUMP01,EMD Millipore,Billerica,MA USA)和过滤器(Millflex-100,100mL,0.45μm,白色网格,MXHAWG124,EMDMillipore)将每种溶液中的细菌细胞分别转移到预装有胰蛋白酶大豆琼脂(MXSMCTS48,EMD Millipore)的各个盒中。
然后,倒转盒,将其置于37℃的培养箱中18小时。18小时后,对盒上的菌落数进行计数。如果没有菌落,则记录零。对于未处理的块,24小时后菌落数不少于1x 103个菌落。
测试细菌的结果显示在表1中。测试进行三次,并且在所有三次计数中,实验样品AM-1中的生物体计数为零,而对于两个对照样品(经洗涤的CM-1和未经洗涤的CM-1),有机体计数多得不能计数(TNTC)。该数据支持2.0或更高的抗菌活性。结果,确定GO-PVA涂层AM-1是有效的杀生物剂,其能够帮助防止表面上的微生物累积。

实施例3.1:另外的抗微生物元件的制备和测试
以与AM-1类似的方式制备一些抗微生物元件,其中使用不同比例的不同交联剂并以不同的厚度沉积。在一些样品中,使用另外的交联剂或其他材料来测试这些材料对性能的影响。每种抗微生物元件的细节和每种抗菌元件的结果显示在下表2中。

Claims (28)
1.一种抗微生物膜,其包含:
支撑物;和
包含氧化石墨烯化合物和交联剂的复合材料,
其中所述复合材料涂布所述支撑物;
其中所述膜阻止微生物生长,这是通过具有2.0或更高的抗菌效力确定的;并且
其中所述交联剂包括聚乙烯醇、生物聚合物、丙烯酰胺共聚物中的至少一种。
2.权利要求1所述的膜,其中所述生物聚合物包含木质素磺酸盐,并且所述丙烯酰胺共聚物包含聚(N-异丙基丙烯酰胺)。
3.权利要求1或2所述的膜,其中所述丙烯酰胺共聚物包括聚(N-异丙基丙烯酰胺-共-N,N'-亚甲基-双-丙烯酰胺)。
4.权利要求1、2或3所述的膜,其中所述复合材料还包含四硼酸钾、3,5-二氨基苯甲酸和2,5-二羟基对苯二甲酸中的至少一种。
5.权利要求1、2、3或4所述的膜,其中所述复合材料还包含间隔材料。
6.权利要求5所述的膜,其中所述间隔材料包括二氧化硅、Fe3O4、TiO2、ZrO2或Al2O3的纳米颗粒。
7.权利要求5或6所述的膜,其中所述间隔材料的粒度范围为约5nm至约300nm。
8.权利要求5、6或7所述的膜,其中所述间隔材料相对于所述复合材料涂层的重量百分比为约3%至约8%。
9.权利要求5、6或7所述的膜,其中所述间隔材料相对于所述复合材料涂层的重量百分比为约5%至约7%。
10.权利要求1、2、3、4、5、6、7、8或9所述的膜,其中所述复合材料中所述氧化石墨烯与所述交联剂的质量比为1:1000至20:1范围内的值。
11.权利要求1、2、3、4、5、6、7、8或9所述的膜,其中所述复合材料中所述氧化石墨烯与所述交联剂的质量比为10:1至1:90范围内的值。
12.权利要求1、2、3、4、5、6、7、8或9所述的膜,其中所述复合材料中所述氧化石墨烯与所述交联剂的质量比为10:1至8:1范围内的值。
13.权利要求1、2、3、4、5、6、7、8、9、10、11或12所述的膜,其中所述支撑物上的复合材料涂层具有约0.01μm至约4.5μm的厚度。
14.权利要求1、2、3、4、5、6、7、8、9、10、11或12所述的膜,其中所述支撑物上的复合材料涂层具有约0.05μm至约3μm的厚度。
15.权利要求1、2、3、4、5、6、7、8、9、10、11或12所述的膜,其中所述支撑物上的复合材料涂层具有约0.05μm至约0.6μm的厚度。
16.权利要求1、2、3、4、5、6、7、8、9、10、11、12、13、14或15所述的膜,其中通过下述方法来制备所述膜:将所述复合材料施加到所述支撑物上,并将所得膜暴露于约60℃至约200℃的温度约2分钟至约150分钟的时间。
17.权利要求1、2、3、4、5、6、7、8、9、10、11、12、13、14或15所述的膜,其中通过下述方法来制备所述膜:将所述复合材料施加到所述支撑物上,并将所得膜暴露于约70℃至约150℃的温度约3分钟至约120分钟的时间。
18.权利要求1、2、3、4、5、6、7、8、9、10、11、12、13、14、15、16或17所述的膜,其中所述支撑物是要保护免于微生物生长的物品。
19.权利要求1、2、3、4、5、6、7、8、9、10、11、12、13、14、15、16、17或18所述的膜,其中所述交联剂是聚乙烯醇且所述复合材料的厚度为约50nm至约3μm。
20.权利要求19所述的膜,其中所述复合材料还包含四硼酸钾,并且所述复合材料具有约50nm至约150nm的厚度。
21.权利要求20所述的膜,其中所述复合材料还包含2,5-二羟基对苯二甲酸,氧化石墨烯相对于总复合材料的含量为约60重量%至约80重量%,并且所述复合材料具有约50nm至约150nm的厚度。
22.权利要求19所述的膜,其中所述复合材料还包含3,5-二氨基苯甲酸,并且所述复合材料具有约700nm至约2μm的厚度。
23.权利要求1、2、3、4、5、6、7、8、9、10、11、12、13、14、15、16、17或18所述的膜,其中所述交联剂包含生物聚合物并且所述复合材料的厚度为约100nm至约2μm。
24.权利要求1、2、3、4、5、6、7、8、9、10、11、12、13、14、15、16、17或18所述的膜,其中所述交联剂包含聚(N-异丙基丙烯酰胺)并且所述复合材料的厚度为约700nm至约2μm。
25.权利要求1、2、3、4、5、6、7、8、9、10、11、12、13、14、15、16、17、18、19、20、21、22、23或24所述的膜,其中所述复合材料还包含二氧化硅纳米颗粒。
26.权利要求25所述的膜,其中所述二氧化硅纳米颗粒的尺寸为约2nm至约20nm,约50nm至约100nm,或约150nm至约300nm。
27.权利要求25所述的膜,其中所述二氧化硅纳米颗粒的尺寸为约5nm至约10nm,约70nm至约9nm,或约180nm至约220nm。
28.一种防止微生物生长的方法,所述方法包括:
提供权利要求1、2、3、4、5、6、7、8、9、10、11、12、13、14、15、16、17、18、19、20、21、22、23、24、25、26、27或28所述的膜;并
将所述膜暴露于含有微生物的工作流体;
其中作为暴露于所述工作流体的结果,所述膜阻止微生物生长,这是通过具有2.0或更高的抗菌效力确定的。
