Abstract
本发明涉及一种载药黑磷壳聚糖复合纳米球及其制备方法和应用,所述载药黑磷壳聚糖复合纳米球为表面修饰有聚乙二醇并包载有黑磷量子点和药物的壳聚糖纳米球。本发明创造性地将黑磷量子点、壳聚糖和聚乙二醇相结合,发挥其协同增效优势,能够高效突破呼吸道黏液层和病菌生物膜屏障;使纳米球具有良好的载药递送能力,适用范围广;能装载多种治疗药物并且能提升药物的治疗效果,降低不良毒副作用;同时具有良好的生物相容性。本发明所述载药黑磷壳聚糖复合纳米球为呼吸类疾病的治疗提供了新的策略。
Description
技术领域
本发明属于生物医用材料技术领域,具体涉及一种载药黑磷壳聚糖复合纳米球及其制备方法和应用。
背景技术
近年来随着空气污染的加重与吸烟人群的增多,呼吸类疾病的发病情况也愈加严重。其中,慢阻肺疾病是一种发病率高且不易治愈的慢性呼吸道疾病,为我国四大慢性病之一。研究结果表明,环境污染、吸烟与呼吸道感染是慢阻肺发病的主要诱因。由于慢阻肺早期症状不明显,大多数患者容易误认为是普通的呼吸道感染疾病而未加以重视,直至出现气促等症状后才进行就医,此时患者肺功能已经下降了50%,治疗的难度增大,效果也不理想,因此常被称为沉默的杀手。
缺乏有效的治疗药物是当前呼吸类疾病特别是慢性呼吸类疾病难以治愈的主要原因。慢阻肺病情的加重通常是由呼吸道感染所致,而无论是慢阻肺稳定期或是急性加重期,流感嗜血杆菌、肺炎链球菌、铜绿假单胞菌普遍被认为是主要的致病菌与定植菌。虽然临床治疗慢阻肺的药物品类众多,如支气管扩张剂(抗胆碱能制剂、β2受体激动剂、茶碱),糖皮质激素(氟替卡松),祛痰药物(盐酸氨溴索、厄多司坦),抗氧化剂(乙酰半胱氨酸),抗生素(阿米卡星、红霉素)等,虽有一定的治疗效果,但都非特效药物,难以完全治愈慢阻肺疾病,而长期反复用药则会对患者造成较大的毒副作用并产生抗药性。
此外,慢性呼吸类疾病不易治愈的另一个重要原因是常规治疗药物难以有效递送,药物的有效给药效率低。由于气道粘液的高分泌为慢阻肺疾病的重要病理特征之一,患者的气管、支气管、和肺泡常伴随着炎症,使得呼吸道黏膜充血水肿,粘液分泌增多,毛细血管通透性增高,浆液大量渗出并与其它混合物形成粘稠的痰液。并且积累的大量粘稠痰液会在呼吸道形成粘液层“屏障”,阻碍药物作用病患部位发挥药效,降低了药物的治疗效果。同时,粘液层中大量的病菌由于没有被及时清除,病菌不断通过定植、繁殖、分化、聚集、脱落与再定植的过程可逐渐形成一层致密的病菌生物被膜保护病菌不被药物杀灭,从而导致治疗不彻底,使病菌产生耐药性。由于细菌生物膜凭借胞外基质致密的屏障作用能对药物治疗产生极强的抵抗能力,临床治疗时不得不提高药物的给药剂量并进行多次治疗,极易对患者造成较大的毒副作用并导致耐药病菌的产生与蔓延。
纳米药物相比普通药物具有更大的比表面积和更高的反应活性,能明显提升药物的生物利用度并保持较长的血药循环时间。纳米给药系统还能解决众多药物稳定性差、水溶性低、不易吸收等问题。此外,纳米药物也更为“智能”,可实现对药物的靶向递送与控释,在吸入肺部后,可渗入肺组织深部直接接触肺泡,增大肺泡组织的药物浓度快速发挥药效,在提升药效的同时显著降低药物的毒副作用。然而,目前全球慢性呼吸疾病的最有型治疗方式通常被认为是吸入疗法,当纳米药物吸入肺部后,肺部的粘液层与病菌生物膜屏障的存在明显阻碍了纳米药物的有效递送。呼吸道中大量粘稠的粘液层使得众多纳米药物无法被有效吸收,降低了药物的给药效率。另一方面,致密的病菌生物膜则使得药物难以完全杀灭病菌,导致治疗不彻底,使病菌产生耐药性。
CN108721219A公开了一种生物粘附性复方肺吸纳米复合微粒及其制备,所述的纳米复合微粒是由一种或两种水溶性的抗哮喘和慢阻肺药物和另外一种难溶性的抗哮喘和慢阻肺药物的纳米晶体同时镶嵌到具有生物粘附性的骨架材料中所形成的。该复合微粒采用喷雾干燥技术,提高了难溶性药物的溶出度,实现几种不同溶解性能药物的同步释放,更好地发挥其协同作用;保证了剂量差异较大的几种药物的均匀混合,大大简化制剂的工艺过程,提高药品质量,降低成本,减少能耗;延长了肺部滞留时间,提高病灶部位的药物浓度,延缓药物的吸收,达到提高疗效、减少全身毒副作用的发生。
CN105012238A公开了一种具有肺主动靶向性的甲强龙免疫纳米脂质体及其制备方法,该发明的免疫脂质体由肺表面活性蛋白A纳米抗体、治疗药物和纳米脂质体组成,以糖皮质激素为药物活性成分,以纳米脂质体为载体,以SP-A纳米抗体为特异性肺组织靶向剂。其具有明确肺靶向性,可以高效、稳定的实现药物活性成分对肺部的靶向输送,且在治疗大鼠肺损伤模型时可以有效减少肺泡灌洗液及肺组织内炎症因子的表达,减轻肺组织病理损伤,提高大鼠存活率,治疗安全,无肝、肾损伤及毒性,具有新的临床实用价值。
但现有技术中公开的递送系统渗透人体生理屏障的能力不足,药物难以有效渗透,因此,开发出一种能够高效突破呼吸道黏液层和病菌生物膜屏障的治疗方式是非常有意义的。
发明内容
针对现有技术的不足,本发明的目的在于提供一种载药黑磷壳聚糖复合纳米球及其制备方法和应用。该载药黑磷壳聚糖复合纳米球具有良好的载药能力,适用范围广,能装载多种治疗药物并且能提升药物的治疗效果,降低不良毒副作用;该载药黑磷壳聚糖复合纳米球具有良好的生物相容性,并且拥有很好的穿透黏液层的能力和抗菌能力;该载药黑磷壳聚糖复合纳米球的制备工艺简单、重现性好,能快速实现大规模制备。
为达到此发明目的,本发明采用以下技术方案:
一方面,本发明提供一种载药黑磷壳聚糖复合纳米球,所述载药黑磷壳聚糖复合纳米球为表面修饰有聚乙二醇并包载有黑磷量子点和药物的壳聚糖纳米球。
天然线性多糖结构的壳聚糖存在大量的游离氨基,其可以被溶液中的氢离子质子化,虽有广谱的抗菌功效,但其水溶性较差、抗菌能力较弱,且需在酸性条件下溶解以发挥抗菌作用,难以满足临床使用时对治疗药物的要求。本发明创造性地将黑磷量子点、壳聚糖和聚乙二醇相结合,发挥其协同增效优势,制备了一种新型的载药复合纳米球以提升药物的靶向能力与穿透人体生理屏障阻碍的能力。其中,黑磷量子点可通过氧化降解反应产生磷酸根离子,在发挥抗菌功效的同时形成酸性微环境裂解纳米球,释放治疗药物,同时形成的酸性微环境也提升了壳聚糖的水溶性以及游离氨基的数量,使得壳聚糖的抗菌功效明显增强;纳米球表面修饰的聚乙二醇能够提升纳米药物穿透黏液层的能力,协同壳聚糖粘附粘膜的特点,长效发挥抗菌功效。因此,本发明所述的纳米球各组成成分相互配合,协同增效,能够高效突破呼吸道黏液层和病菌生物膜屏障;且纳米球具有良好的载药能力,适用范围广,能装载多种治疗药物并且能提升药物的治疗效果,降低不良毒副作用;同时具有良好的生物相容性。
优选地,所述纳米球粒径为150-250nm,例如150nm、170nm、180nm、200nm、220nm、240nm或250nm等。
所述纳米球粒径选择在150-250nm范围内,是因为粒径超过此范围会导致其纳米性能降低,穿透生物屏障的能力会下降,小于此范围会导致所制备纳米药物的渗透能力不足迅速被机体排除,导致药效不佳。
优选地,所述黑磷量子点的横向尺寸为1-10nm,例如1nm、2nm、3nm、4nm、5nm、6nm、7nm、8nm、9nm或10nm等。
优选地,所述黑磷量子点的纵向尺寸为2-8nm,例如2nm、3nm、4nm、5nm、6nm、7nm或8nm等。
所述黑磷量子点的横向尺寸和纵向尺寸分别选择在1-10nm和2-8nm的原因是黑磷二维纳米材料的尺寸需在此区间才具有量子点效应。
优选地,所述药物包括支气管扩张剂、糖皮质激素、祛痰药物、抗氧化剂类药物或抗生素类药物中的任意一种或至少两种的组合,所述至少两种的组合例如支气管扩张剂和糖皮质激素的组合、祛痰药物和抗氧化剂类药物的组合、抗氧化剂类药物和抗生素类药物的组合等。
优选地,所述壳聚糖的粘度为200-400mPa.s,例如200mPa.s、220mPa.s、250mPa.s、280mPa.s、300mPa.s、320mPa.s、350mPa.s或400mPa.s等。
所述壳聚糖的粘度会影响壳聚糖纳米粒子的制备,选择在200-400mPa.s范围内的原因是超过此粘度值会导致纳米粒子出现成团现象,在溶液中的分散性不佳,小于此粘度值会不利于壳聚糖形成纳米粒子。
优选地,所述聚乙二醇为具有侧链基团修饰的聚乙二醇。
优选地,所述侧链基团包括氨基酸残基链、巯基或羧基中的任意一种或至少两种的组合,所述至少两种的组合例如氨基酸残基链和巯基的组合、巯基和羧基的组合等。
由于本发明所述的纳米球在水溶液中极易发生团聚,因此此处使用聚乙二醇进行表面修饰以提升纳米球的分散性并赋予其穿透肺部粘液屏障的能力,聚乙二醇修饰可使用具有不同侧链基团的聚乙二醇修饰剂对纳米球的疏水性和静电作用进行调整,以使纳米球具有高亲水性且能快速穿透粘液层。
优选地,所述聚乙二醇为带有荧光基团的聚乙二醇。
使用带有荧光基团的聚乙二醇进行表面修饰可实现对纳米球的定位示踪观察。
优选地,所述聚乙二醇的数均分子量为800-4000KD,例如800KD、1000KD、1200KD、1500KD、1800KD、2000KD、2200KD、2500KD、3000KD、3200KD、3500KD、3800KD或4000KD等,优选2000KD。
所述聚乙二醇的数均分子量选择在800-4000KD是因为上述分子量范围的聚乙二醇更有利于所制备的载药黑磷壳聚糖复合纳米球穿透粘液屏障。
另一方面,本发明提供一种如上所述的载药黑磷壳聚糖复合纳米球的制备方法,其特征在于,所述制备方法包括如下步骤:
(1)制备黑磷量子点;
(2)制备载药复合纳米球;
(3)对步骤(2)制得的载药复合纳米球进行表面修饰,得到所述载药黑磷壳聚糖复合纳米球。
该载药黑磷壳聚糖复合纳米球的制备工艺简单、重现性好,能快速实现大规模制备。
在本发明中,步骤(1)所述制备黑磷量子点的方法为:在真空环境下将块状黑磷研磨后分散于第一有机溶剂中,将分散液与第二有机溶剂和氢氧化钠进行混合,再将混合液加热、冷却、离心,收集上清液,得到黑磷量子点分散液。
优选地,所述块状黑磷的粒径为50-100μm,例如50μm、55μm、60μm、65μm、70μm、75μm、80μm、90μm或100μm等。
优选地,所述第一有机溶剂包括N-甲基吡咯烷酮、二甲基甲酰胺、二甲基亚砜、四氢呋喃、甲醇、异丙醇、三氯甲烷或二氯甲烷中的任意一种或至少两种的组合,所述至少两种的组合例如N-甲基吡咯烷酮和二甲基甲酰胺的组合、二甲基亚砜和四氢呋喃的组合、甲醇和异丙醇的组合等。

优选地,步骤(2)所述制备载药复合纳米球的方法为:配制壳聚糖醋酸溶液,调节pH,过滤,将其与黑磷量子点分散液、药物以及三聚磷酸钠混合,在搅拌下进行反应,离心、收集下层沉淀、洗涤,加入超纯水中进行超声分散,最后冷冻干燥,得到所述载药复合纳米球。
所述三聚磷酸钠是以溶液的方式逐滴缓慢地加入壳聚糖醋酸溶液,在搅拌下反应直至溶液出现丁达尔效应后停止加入三聚磷酸钠溶液。
优选地,所述壳聚糖醋酸溶液中壳聚糖的浓度为1-3mg/mL,例如1mg/mL、1.2mg/mL、1.5mg/mL、1.8mg/mL、2mg/mL、2.5mg/mL或3mg/mL等。
优选地,所述壳聚糖醋酸溶液中醋酸的质量浓度为1-2%,例如1%、1.2%、1.5%、1.8%或2%等。
优选地,所述调节pH是指调节pH值至4-5,例如4.1、4.5、4.6、4.8或5等。

同时,本发明所述纳米球可被荧光分子如异硫氰酸荧光素(FITC)、罗丹明等进行标记,可实现对纳米球的定位示踪观察。其操作方法为将所制备的载药黑磷壳聚糖复合
纳米球与荧光分子充分混匀后加入透析袋中进行透析。
作为本发明的优选技术方案,所述制备方法包括如下步骤:
(1)在真空环境下将粒径为50-100μm的块状黑磷研磨后分散于N-甲基吡咯烷酮、二甲基甲酰胺、二甲基亚砜、四氢呋喃、甲醇、异丙醇、三氯甲烷或二氯甲烷中的任意一种或至少两种的组合中,将浓度为1-2mg/mL的分散液与N-甲基吡咯烷酮和氢氧化钠进行混合,再将混合液在120-160℃下加热6-18h、冷却至20-30℃、离心,收集上清液,得到黑磷量子点分散液;
(2)配制壳聚糖浓度为1-3mg/mL的1-2%醋酸溶液,调节pH至4-5,使用0.45μm和0.22μm的滤膜依次过滤,将其与黑磷量子点分散液、药物以及三聚磷酸钠混合,在转速为700-900r/min的搅拌下进行反应,以转速为6000-8000r/min离心2-5min、收集下层沉淀、洗涤,加入超纯水中以频率为10-30KHz进行超声分散1-5min,最后冷冻干燥,得到载药复合纳米球;其中,黑磷量子点与壳聚糖的质量比为1:(10-20),壳聚糖与药物的质量比为(12-18):1,壳聚糖与三聚磷酸钠的质量比为(4-8):1;
(3)将步骤(2)制得的载药复合纳米球与聚乙二醇混合,加入透析袋进行透析,得到所述载药黑磷壳聚糖复合纳米球;其中,载药复合纳米球与聚乙二醇的质量比为(5-8):1。
再一方面,本发明提供一种如上所述的载药黑磷壳聚糖复合纳米球在制备治疗呼吸类疾病的药物中的应用。
优选地,所述呼吸类疾病包括哮喘、慢性阻塞性肺病、慢性支气管炎、肺气肿、肺癌、囊性纤维化或支气管扩张。
相对于现有技术,本发明具有以下有益效果:
本发明创造性地将黑磷量子点、壳聚糖和聚乙二醇相结合,发挥其协同增效优势,制备了一种新型的纳米载药复合球以提升药物的靶向能力与穿透人体生理屏障阻碍的能力。其中,黑磷量子点可通过氧化降解反应产生磷酸根离子,在发挥抗菌功效的同时形成酸性微环境裂解纳米球,释放治疗药物,同时形成的酸性微环境也提升了壳聚糖的水溶性以及游离氨基的数量,使得壳聚糖的抗菌功效明显增强;纳米球表面修饰的聚乙二醇能够提升纳米药物穿透黏液层的能力,协同壳聚糖粘附粘膜的特点,长效发挥抗菌功效。因此,本发明所述的纳米球各组成成分相互配合,协同增效,能够高效突破呼吸道黏液层和病菌生物膜屏障;使纳米球具有良好的载药能力,适用范围广,能装载多种治疗药物并且能提升药物的治疗效果,降低不良毒副作用;同时具有良好的生物相容性。

下面通过具体实施方式来进一步说明本发明的技术方案。本领域技术人员应该明了,所述实施例仅仅是帮助理解本发明,不应视为对本发明的具体限制。
实施例1
本实施例提供一种载药黑磷壳聚糖复合纳米球,所述载药黑磷壳聚糖复合纳米球为表面修饰有聚乙二醇的并包载有黑磷量子点和药物的壳聚糖纳米球。其制备方法为:
(1)在真空环境下将粒径为70μm的块状黑磷研磨后分散于N-甲基吡咯烷酮和二甲基甲酰胺的混合溶剂中,将浓度为1.5mg/mL的分散液与N-甲基吡咯烷酮和氢氧化钠进行混合,再将混合液在140℃下加热12h、冷却至25℃、离心,收集上清液,得到黑磷量子点分散液;
(2)配制壳聚糖浓度为2mg/mL的1%醋酸溶液,调节pH至4.5,使用0.45μm和0.22μm的滤膜依次过滤,将其与黑磷量子点分散液、药物以及三聚磷酸钠混合,在转速为800r/min的搅拌下进行反应,以转速为8000r/min离心4min、收集下层沉淀、洗涤,加入超纯水中以功率为20KHz进行超声分散3min,最后冷冻干燥,得到载药复合纳米球;其中,黑磷量子点与壳聚糖的质量比为1:10,壳聚糖与药物的质量比为15:1,壳聚糖与三聚磷酸钠的质量比为6:1;
(3)将步骤(2)制得的载药复合纳米球与聚乙二醇混合,加入透析袋进行透析,得到所述载药黑磷壳聚糖复合纳米球;其中,载药复合纳米球与聚乙二醇的质量比为5:1。
实施例2
本实施例提供一种载药黑磷壳聚糖复合纳米球,所述载药黑磷壳聚糖复合纳米球为表面修饰有聚乙二醇的并包载有黑磷量子点和药物的壳聚糖纳米球。其制备方法为:
(1)在真空环境下将粒径为50μm的块状黑磷研磨后分散于N-甲基吡咯烷酮中,将浓度为1mg/mL的分散液与N-甲基吡咯烷酮和氢氧化钠进行混合,再将混合液在120℃下加热18h、冷却至20℃、离心,收集上清液,得到黑磷量子点分散液;
(2)配制壳聚糖浓度为1mg/mL的1%醋酸溶液,调节pH至4,使用0.45μm和0.22μm的滤膜依次过滤,将其与黑磷量子点分散液、药物以及三聚磷酸钠混合,在转速为700r/min的搅拌下进行反应,以转速为6000r/min离心5min、收集下层沉淀、洗涤,加入超纯水中以功率为10Hz进行超声分散5min,最后冷冻干燥,得到载药复合纳米球;其中,黑磷量子点与壳聚糖的质量比为1:20,壳聚糖与药物的质量比为12:1,壳聚糖与三聚磷酸钠的质量比为4:1;
(3)将步骤(2)制得的载药复合纳米球与聚乙二醇混合,加入透析袋进行透析,得到所述载药黑磷壳聚糖复合纳米球;其中,载药复合纳米球与聚乙二醇的质量比为5:1。
实施例3
本实施例提供一种载药黑磷壳聚糖复合纳米球,所述载药黑磷壳聚糖复合纳米球为表面修饰有聚乙二醇的并包载有黑磷量子点和药物的壳聚糖纳米球。其制备方法为:
(1)在真空环境下将粒径为100μm的块状黑磷研磨后分散于N-甲基吡咯烷酮、二甲基甲酰胺和四氢呋喃的混合溶剂中,将浓度为2mg/mL的分散液与N-甲基吡咯烷酮和氢氧化钠进行混合,再将混合液在160℃下加热6h、冷却至30℃、离心,收集上清液,得到黑磷量子点分散液;
(2)配制壳聚糖浓度为3mg/mL的2%醋酸溶液,调节pH至5,使用0.45μm和0.22μm的滤膜依次过滤,将其与黑磷量子点分散液、药物以及三聚磷酸钠混合,在转速为900r/min的搅拌下进行反应,以转速为8000r/min离心3min、收集下层沉淀、洗涤,加入超纯水中以功率为20Hz进行超声分散3min,最后冷冻干燥,得到载药复合纳米球;其中,黑磷量子点与壳聚糖的质量比为1:20,壳聚糖与药物的质量比为18:1,壳聚糖与三聚磷酸钠的质量比为8:1;
(3)将步骤(2)制得的载药复合纳米球与聚乙二醇混合,加入透析袋进行透析,得到所述载药黑磷壳聚糖复合纳米球;其中,载药复合纳米球与聚乙二醇的质量比为8:1。
实施例4
本实施例采用扫描电镜观察本发明制得的载药黑磷壳聚糖复合纳米球及其裂解时的形态特征,同时观察载药与非载药给纳米球的粒径带来的影响。首先制备一种非载药的黑磷壳聚糖纳米球,其制备方法与实施例1的区别仅在于第(2)步骤中,将“壳聚糖醋酸溶液与黑磷量子点分散液、药物以及三聚磷酸钠混合”更改为“壳聚糖醋酸溶液与黑磷量子点分散液以及三聚磷酸钠混合”,其他保持不变。对其与实施例1制得的载药黑磷壳聚糖复合纳米球进行扫描电镜观察,如图1和图2所示(图1和图2分别为非载药和载药黑磷壳聚糖复合纳米球);并将实施例1制得的载药黑磷壳聚糖复合纳米球溶于磷酸盐缓冲液中,4h后对其形态进行观察,结果如图3所示。
由图1和图2可知:制备得到的纳米球粒径为200nm左右,且在装载治疗药物后,其粒径与形态均未发生明显变化。由图3可知:在磷酸盐缓冲液中,黑磷量子点可迅速氧化降解,其形成的酸性环境能逐渐裂解纳米球,释放所装载的治疗药物(图中标尺尺寸为200nm)。对实施例2-3制备得到的载药黑磷壳聚糖复合纳米球进行同样的实验可以得到同样的结果。
实施例5
本实施例在体外模拟条件下研究本发明制备得到的纳米球渗透粘液屏障的能力,即通过在体外模拟人体粘液层组分从而检测纳米治疗药物在粘液层中的稳定情况和穿透速率。具体操作为:首先制备一种表面未被聚乙二醇修饰的载药黑磷壳聚糖复合纳米球,其制备方法与与实施例1的区别仅在于无第(3)步骤,其他保持不变;再制备一种非载药的黑磷壳聚糖纳米球,其制备方法与实施例4中一致;最后制备一种非载药且未被聚乙二醇修饰的载药黑磷壳聚糖复合纳米球,其制备方法与实施例1的区别仅在于将“壳聚糖醋酸溶液与黑磷量子点分散液、药物以及三聚磷酸钠混合”更改为“壳聚糖醋酸溶液与黑磷量子点分散液以及三聚磷酸钠混合”,且无第(3)步骤,其他保持不变。
将四组样品用考马斯亮蓝染料染色后加入人工模拟粘液层观察,如图4所示(a为非载药且未被聚乙二醇修饰的黑磷壳聚糖复合纳米球;b为非载药的黑磷壳聚糖纳米球;c为未被聚乙二醇修饰的载药黑磷壳聚糖复合纳米球;d为实施例1制得的载药黑磷壳聚糖复合纳米球):具有聚乙二醇修饰的载药黑磷壳聚糖复合纳米球和未载药黑磷壳聚糖复合纳米球都具有更好的“粘液惰性”,能迅速穿透粘液层,说明聚乙二醇的修饰能明显提升纳米球穿透粘液层的能力。对实施例2-3制备得到的载药黑磷壳聚糖复合纳米球进行同样的实验可以得到同样的结果。
实施例6
本实施例对本发明制备的载药黑磷壳聚糖复合纳米球进行抗菌功效评价。具体操作为:
首先制备单纯的壳聚糖纳米粒子、非载药且未被聚乙二醇修饰的黑磷壳聚糖纳米球(制备方法如实施例5所示)。
使用大肠杆菌和金黄色葡萄球菌作为细菌模型,挑取少量细菌保菌液加到含有LB液体的细菌培养基中,置于摇菌管内于37℃,220rmp细菌恒温培养箱中振荡过夜培养,随后用冷冻离心机(4000rmp,1min)对细菌进行收集,并使用无菌生理盐水洗涤细菌后,用生理盐水调节细菌浓度,用多功能酶标仪进行检测,使细菌悬液的浓度达到OD600为0.02(即(0.8-1)×106cfu/mL)。
将100μL上述各样品(浓度均为2mg/mL的单纯壳聚糖纳米粒子、非载药且未被聚乙二醇修饰的黑磷壳聚糖复合纳米球、载药黑磷壳聚糖复合纳米球)与100μL活化的菌悬液共混后均匀涂布于培养皿表面,菌液均匀涂布于培养皿后,将培养皿置于细菌培养箱中37℃恒温恒湿培养24h,观察菌落个数。每个样品平行重复5次。空白对照组将样品溶液换为等体积的生理盐水。
各组对大肠杆菌的抗菌结果如图5所示,各组对金黄色葡萄球菌的抗菌结果如图6所示,由图5和图6可知:参比空白对照组,单纯的壳聚糖纳米粒子对大肠杆菌和金黄色葡萄球菌的抑菌能力较弱,当通过离子交联法将壳聚糖和黑磷量子点共同制备成纳米粒子后,黑磷量子点氧化降解形成的酸性环境提升了壳聚糖的水溶性与游离氨基的数量,使得抗菌功效得以提升。在此基础上进一步装载治疗药物后,其抗菌功效获得明显增强。对实施例2-3制备得到的载药黑磷壳聚糖复合纳米球进行同样的实验可以得到同样的结果。
实施例7
本实施例对本发明制备的载药黑磷壳聚糖复合纳米球进行生物相容性评价。具体操作为:
首先制备单纯的壳聚糖纳米粒子、非载药且未被聚乙二醇修饰的黑磷壳聚糖纳米球,其制备方法如实施例5中所示。
通过活/死细胞染色试剂盒(Calcein AM/PI)来检测纳米球的生物相容性,利用Calcein AM为活细胞优良的荧光染色剂,其能轻易穿透活细胞的特点,当其到达细胞内会被酯酶水解为钙黄绿素留在胞内,呈现强绿色荧光;试剂盒中的碘化丙啶(PI)不能穿过活细胞膜,但能穿过死细胞膜的无序区域到达细胞核,在嵌入细胞DNA螺旋后产生红色荧光(激发波长为535nm,发射波长为617nm)从而使死细胞呈现红色荧光。
分别取对数生长期的肺细胞(HULEC-5a)与成纤维细胞(L929)以1×105/mL,每孔100μL加入96孔板内,于细胞培养箱中培养12h后轻轻吸去每孔中的细胞培养基,将含有不同待测组分(浓度均为50μg/mL的单纯的壳聚糖纳米粒子、非载药且未被聚乙二醇修饰的黑磷壳聚糖复合纳米球、载药黑磷壳聚糖复合纳米球)的细胞培养基200μL加入孔中继续培养24h,以无血清培养基作为空白对照。取出96孔细胞培养板,吸除孔内培养基,用PBS溶液小心清洗孔板3次后向每孔内加入钙黄绿素和碘化丙啶溶液,在培养箱中孵育20分钟后,吸除染料并用PBS溶液小心清洗3次后置于荧光显微镜下进行观察。
实验结果如图7所示,与空白对照组相比,所制备的各样品观察视野内的细胞均呈现强绿色荧光,表明各样品具有良好的生物相容性,在50μg/mL的浓度下对肺细胞(HULEC-5a)孵育24h后仍未见检测到明显的细胞毒性,如图8所示,在50μg/mL的浓度下对成纤维细胞(L929)孵育24h后也未见检测到明显的细胞毒性。对实施例2-3制备得到的载药黑磷壳聚糖复合纳米球进行同样的实验可以得到同样的结果。
申请人声明,本发明通过上述实施例来说明本发明的载药黑磷壳聚糖复合纳米球及其制备方法和应用,但本发明并不局限于上述实施例,即不意味着本发明必须依赖上述实施例才能实施。所属技术领域的技术人员应该明了,对本发明的任何改进,对本发明产品各原料的等效替换及辅助成分的添加、具体方式的选择等,均落在本发明的保护范围和公开范围之内。
以上详细描述了本发明的优选实施方式,但是,本发明并不限于上述实施方式中的具体细节,在本发明的技术构思范围内,可以对本发明的技术方案进行多种简单变型,这些简单变型均属于本发明的保护范围。
另外需要说明的是,在上述具体实施方式中所描述的各个具体技术特征,在不矛盾的情况下,可以通过任何合适的方式进行组合,为了避免不必要的重复,本发明对各种可能的组合方式不再另行说明。
Claims (10)
1.一种载药黑磷壳聚糖复合纳米球,其特征在于,所述载药黑磷壳聚糖复合纳米球为表面修饰有聚乙二醇并包载有黑磷量子点和药物的壳聚糖纳米球。
2.如权利要求1所述的载药黑磷壳聚糖复合纳米球,其特征在于,所述纳米球粒径为150-250nm;
优选地,所述黑磷量子点的横向尺寸为1-10nm;
优选地,所述黑磷量子点的纵向尺寸为2-8nm;
优选地,所述药物包括支气管扩张剂、糖皮质激素、祛痰药物、抗氧化剂类药物或抗生素类药物中的任意一种或至少两种的组合;
优选地,所述壳聚糖的粘度为200-400mPa.s。
3.如权利要求1或2所述的载药黑磷壳聚糖复合纳米球,其特征在于,所述聚乙二醇为具有侧链基团修饰的聚乙二醇;
优选地,所述侧链基团包括氨基酸残基链、巯基或羧基中的任意一种或至少两种的组合;
优选地,所述聚乙二醇为带有荧光基团的聚乙二醇;
优选地,所述聚乙二醇的数均分子量为800-4000kD,优选2000kD。
4.如权利要求1-3中任一项所述的载药黑磷壳聚糖复合纳米球的制备方法,其特征在于,所述制备方法包括如下步骤:
(1)制备黑磷量子点;
(2)制备载药复合纳米球;
(3)对步骤(2)制得的载药复合纳米球进行表面修饰,得到所述载药黑磷壳聚糖复合纳米球。
5.如权利要求4所述的载药黑磷壳聚糖复合纳米球的制备方法,其特征在于,步骤(1)所述制备黑磷量子点的方法为:在真空环境下将块状黑磷研磨后分散于第一有机溶剂中,将分散液与第二有机溶剂和氢氧化钠进行混合,再将混合液加热、冷却、离心,收集上清液,得到黑磷量子点分散液;
优选地,所述块状黑磷的粒径为50-100μm;
优选地,所述第一有机溶剂包括N-甲基吡咯烷酮、二甲基甲酰胺、二甲基亚砜、四氢呋喃、甲醇、异丙醇、三氯甲烷或二氯甲烷中的任意一种或至少两种的组合;
优选地,所述分散液的浓度为1-2mg/mL;
优选地,所述第二有机溶剂为N-甲基吡咯烷酮;
优选地,所述加热的温度为120-160℃;
优选地,所述加热的时间为6-18h;
优选地,所述冷却是指冷却至20-30℃。
6.如权利要求4或5所述的载药黑磷壳聚糖复合纳米球的制备方法,其特征在于,步骤(2)所述制备载药复合纳米球采用的是多聚磷酸钠静电作用的离子交联法;
优选地,步骤(2)所述制备载药复合纳米球的方法为:配制壳聚糖醋酸溶液,调节pH,过滤,将其与黑磷量子点分散液、药物以及三聚磷酸钠混合,在搅拌下进行反应,离心、收集下层沉淀、洗涤,加入超纯水中进行超声分散,最后冷冻干燥,得到所述载药复合纳米球。
7.如权利要求6所述的载药黑磷壳聚糖复合纳米球的制备方法,其特征在于,所述壳聚糖醋酸溶液中壳聚糖的浓度为1-3mg/mL;
优选地,所述壳聚糖醋酸溶液中醋酸的质量浓度为1-2%;
优选地,所述调节pH是指调节pH值至4-5;
优选地,所述过滤是使用0.45μm和0.22μm的滤膜依次过滤;
优选地,所述混合后黑磷量子点与壳聚糖的质量比为1:(10-20);
优选地,所述混合后壳聚糖与药物的质量比为(12-18):1;
优选地,所述混合后壳聚糖与三聚磷酸钠的质量比为(4-8):1;
优选地,所述搅拌的转速为700-900r/min;
优选地,所述离心的转速为6000-8000r/min;
优选地,所述离心的时间为2-5min;
优选地,所述超声的频率为10-30KHz;
优选地,所述超声的时间为1-5min。
8.如权利要求4-7中任一项所述的载药黑磷壳聚糖复合纳米球的制备方法,其特征在于,步骤(3)所述对载药复合纳米球进行表面修饰的方法为:将步骤(2)制得的载药复合纳米球与聚乙二醇混合,加入透析袋进行透析,得到所述载药黑磷壳聚糖复合纳米球;
优选地,所述载药复合纳米球与聚乙二醇的质量比为(5-8):1。
9.如权利要求4-8中任一项所述的载药黑磷壳聚糖复合纳米球的制备方法,其特征在于,所述制备方法包括如下步骤:
(1)在真空环境下将粒径为50-100μm的块状黑磷研磨后分散于N-甲基吡咯烷酮、二甲基甲酰胺、二甲基亚砜、四氢呋喃、甲醇、异丙醇、三氯甲烷或二氯甲烷中的任意一种或至少两种的组合中,将浓度为1-2mg/mL的分散液与N-甲基吡咯烷酮和氢氧化钠进行混合,再将混合液在120-160℃下加热6-18h、冷却至20-30℃、离心,收集上清液,得到黑磷量子点分散液;
(2)配制壳聚糖浓度为1-3mg/mL的1-2%醋酸溶液,调节pH至4-5,使用0.45μm和0.22μm的滤膜依次过滤,将其与黑磷量子点分散液、药物以及三聚磷酸钠混合,在转速为700-900r/min的搅拌下进行反应,以转速为6000-8000r/min离心2-5min、收集下层沉淀、洗涤,加入超纯水中以频率为10-30KHz进行超声分散1-5min,最后冷冻干燥,得到载药复合纳米球;其中,黑磷量子点与壳聚糖的质量比为1:(10-20),壳聚糖与药物的质量比为(12-18):1,壳聚糖与三聚磷酸钠的质量比为(4-8):1;
(3)将步骤(2)制得的载药复合纳米球与聚乙二醇混合,加入透析袋进行透析,得到所述载药黑磷壳聚糖复合纳米球;其中,载药复合纳米球与聚乙二醇的质量比为(5-8):1。
10.如权利要求1-3中任一项所述的载药黑磷壳聚糖复合纳米球在制备治疗呼吸类疾病的药物中的应用;
优选地,所述呼吸类疾病包括哮喘、慢性阻塞性肺病、慢性支气管炎、肺气肿、肺癌、囊性纤维化或支气管扩张。
