Effects of the embodiment
1. And (3) bioavailability test: evaluation of the penetration of the formulations of example 1 and comparative examples 1 to 4 into the skin
Experimental materials: in vitro wistar rat skin made into 4cm2The large and small skin pieces are washed clean with normal saline and stored in a refrigerator at-20 ℃, and the rat skin is detected before the experiment, and no damage can be seen by naked eyes.
The experimental method comprises the following steps: the diffusion area was 2.45cm using a modified Franz diffusion chamber2Stirring at a constant speed of 150 r/min. The treated rat skin was fixed between a supply tank and a receiving tank of a diffusion cell with the stratum corneum surface facing the supply tank, 3ml of the example or comparative sample was added to the supply tank, and distilled water was added to the receiving tank. The water bath temperature is 37 +/-1 ℃, 10ml of the receiving solution is precisely absorbed into a beaker at 10min, 30min and 100min respectively, and 10ml of normal saline is immediately supplemented at the same time. The receiving solution was filtered through a microporous membrane, and the amount of the filtrate in example 1 and comparative example was quantitatively analyzed by HPLC-UVExamples 1-3HPLC-UV observations Total oat kernel extract and CalmYang, comparative example 4HPLC-UV observations diphenhydramine hydrochloride (note: good permeability by itself) was analyzed.
And (3) test results: as can be seen from fig. 1, the bioavailability of example 1 is relatively close to that of comparative example 4, and comparative example 4 replaces the pentylene glycol oat kernel extract and calmsang with the chemically synthesized antipruritic diphenhydramine hydrochloride, thus illustrating that the permeability of oat kernel extract and calmsang in example 1 is relatively close to that of diphenhydramine hydrochloride in comparative example 4.
Example 1 in comparison with comparative example 1, comparative example 1 does not contain 1, 2-pentanediol and the bioavailability is significantly different by more than several times, thus illustrating that 1, 2-pentanediol has the effect of promoting the permeability of oat kernel extract and calmsang.
Example 1 in comparison to comparative example 2, comparative example 2 used 1, 2-hexanediol instead of 1, 2-pentanediol, but the bioavailability of comparative example 2 was relatively low, indicating that other polyols did not have significant penetration enhancing effects on oat kernel extract and calmsang.
Example 1 in comparison with comparative example 3, comparative example 3 was a skin penetrating agent, water soluble azone, instead of 1, 2-pentanediol, but the bioavailability of comparative example 3 was still relatively low, indicating that the skin penetrating agent did not significantly enhance penetration of oat kernel extract and calmsang.
In summary, the following steps: the permeability of oat kernel extract and CalmYang in the presence of 1, 2-pentanediol was similar to that of the chemically synthesized antipruritic diphenhydramine hydrochloride. 1, 2-pentanediol is uniquely effective in enhancing the permeability of oat kernel extract and CalmYang.
2. Safety test
The test method comprises the following steps: according to a skin irritation/corrosivity test method of ' cosmetic hygiene code ' 2007 edition of the ministry of health of the people's republic of China, the general-grade New Zealand rabbits (two male and female, provided by the three-water base of the Guangdong province medical laboratory animal center) qualified for quarantine are directly used for the skin irritation test. Before the test, the hair on both sides of the spine of the experimental animal is cut off, and the hair removing range is about 3cm multiplied by 3cm respectively. About 0.5g of the infant soothing and cooling liquid prepared in the examples or comparative examples is smeared on the skin of the left side of an animal, the smearing area is about 2.5cm multiplied by 2.5cm, the liquid is smeared once a day for 14 days continuously, and the other side of the liquid is used as a blank control. Shearing hair before smearing every day from the next day, removing residual infant soothing and cooling liquid with purified water, treating the control area in the same way, and observing the result 1h after smearing.

TABLE 2 New Zealand rabbits multiple skin irritation test results
Sample (I) Example 1 Example 2 Comparative example 3 Comparative example 4
Mean of daily score for each animal 0 0.12 0.93 0.45
The above data (tables 1 and 2) are provided by the Guangdong provincial underwriting test center test.
The test results show that the soothing cooling liquid prepared in example 1, example 2 and comparative example 4 has no irritation according to the skin irritation intensity grading standard, the data of example 1 and example 2 are superior to the data of comparative example 4 using a chemically synthesized antipruritic agent, and the safety of the plant soothing cooling liquid using 1, 2-pentanediol together with oat kernel extract and CalmYang is slightly better than that of the soothing cooling liquid using the chemical antipruritic agent diphenhydramine hydrochloride.
Comparative example 3 the soothing and cooling liquid prepared in example 1 and example 2 has light irritation, and 1, 2-pentanediol is used as a penetration enhancer, so that the safety of the 1, 2-pentanediol is obviously better than that of water-soluble azone used as a skin penetrating agent.
3. Efficacy test
100 volunteers (mother and infant) were selected and the example samples were changed every two months during the period from 2 months to 10 months, and the example efficacy evaluation statistics were collected every two months.
TABLE 3 evaluation results of efficacy of soothing, itching-relieving and refreshing liquid
Figure BDA0002067874300000081
Efficacy test results: as can be seen from table 3, the evaluation of the relieving itching effect and the relieving itching time of example 1 and example 2 and the comparative example 4 have no obvious difference, which indicates that the relieving and cooling liquid prepared by using pentanediol together with the oat kernel extract and the CalmYang plant extract is very close to the relieving and cooling liquid prepared by using diphenhydramine hydrochloride in the relieving and itching effect. In the test of whether there was irritation during use, examples 1 and 2, which were prepared using the plant soothing essence, were slightly superior to comparative example 4, which was prepared using a chemically synthesized anti-itching agent.
Comparing example 1 with comparative example 5 and comparative example 6, it can be seen that comparative example 5 (lacking the CalmYang) and comparative example 6 (lacking the oat kernel extract) have far lower skin soothing and itching relieving effects than example 1 due to the lack of one component in the raw material formulation, thereby illustrating that the oat kernel extract and the CalmYang plant extract have good synergistic effect under the condition of 5% of 1,2 pentanediol for promoting penetration.
While the preferred embodiments and examples of the present invention have been described in detail, the present invention is not limited to the embodiments and examples described above, and various changes can be made without departing from the spirit of the present invention within the knowledge of those skilled in the art.
