Abstract
本发明涉及药用植物资源提取技术领域,特别涉及一种从麦冬须根中提取纯化总黄酮和总皂苷的方法,以体积浓度86%乙醇,料液比1:15热回流提取,提取3次,每次95min;减压浓缩得到麦冬须根提取液;麦冬须根提取液先加入等量无水乙醇涡旋后静置5min,醇沉除杂,再经过D101大孔树脂纯化得到洗脱液:大量水冲洗,至洗脱液不浑浊,洗脱液弃去;以体积浓度10%乙醇洗脱,洗脱液弃去,再以50%乙醇洗脱,收集洗脱液,得到纯化的提取物,其中总黄酮和总皂苷含量大于30%。本发明同时从麦冬须根中提取总黄酮和总皂苷,并对其进行纯化,精简工艺,节省时间,并提高了总黄酮和皂苷的纯度。
Description
技术领域
本发明涉及药用植物资源提取技术领域,特别涉及一种从麦冬须根中提取纯化总黄酮和总皂苷的方法。
背景技术
麦冬为百合科植物麦冬Ophiopogonjaponicus(L.f)Ker-Gawl的干燥块根。具有养阴生津,润肺清心的作用,用于肺燥干咳,阴虚痨嗽,喉痹咽痛,津伤口渴,内热消渴,心烦失眠和肠燥便秘。
麦冬中主要成分为甾体皂苷类、高异黄酮类和多糖类等。具有降血糖、保护心血管系统、增强免疫力、抗皮肤衰老、抗炎、抗肿瘤等药理作用。除此之外还具有镇静、催眠、降血糖等作用,尤其对保持老年人健康具有多方面功效。
现2015版《中国药典》要求以麦冬块根入药,而弃去须根,造成了大量的须根浪费。通过对麦冬块根和麦冬须根中的成分和含量进行对比研究发现:麦冬须根的化学成分在种类和含量上皆与块根相似,有些成分甚至高于块根。而且发明人初步对麦冬须根的药理作用进行研究还发现,麦冬须根提取物对烟气所致小鼠气道损伤具有明显的保护作用。所以,须根完全可以作为块根的替代品应用于保健品及药物研发等领域。
发明内容
本发明针对现有技术的缺陷,提供了一种从麦冬须根中高效提取和纯化总黄酮和总皂苷的方法,使废弃资源再利用,具有较大经济价值。
为了实现以上发明目的,本发明采取的技术方案如下:
从麦冬须根中提取纯化总黄酮和总皂苷的方法,包括以下步骤:
(1)以体积浓度86%乙醇水溶液,料液比1:15热回流提取,提取3次,每次95min;减压浓缩得到麦冬须根提取液(密度1.25~1.30g/mL);
(2)麦冬须根提取液加入等量无水乙醇涡旋后静置5min后10000rpm离心,醇沉除杂,上清液40℃减压旋干得浸膏;
(3)浸膏用纯水按固液比1:20溶解配制为上样液,再按照大孔树脂比上样液1:5(固液比,g:mL)反复3次上样;
(4)大量水冲洗,至洗脱液不浑浊,洗脱液弃去;
(5)以体积浓度10%乙醇洗脱4个柱床体积(4BV),洗脱液弃去,再以50%乙醇洗脱4BV,收集洗脱液,旋干得纯化物,其中总黄酮和总皂苷含量大于30%。
其中,步骤(2)中先将麦冬须根提取液加入等量无水乙醇涡旋后静置5min,醇沉除杂质;(5)采用D101大孔树脂纯化,收集10%~50%乙醇洗脱液。
与现有技术相比本发明的优点在于:
1通过响应面实验优化提取工艺,同时从麦冬须根中提取总黄酮和总皂苷,并通过大孔吸附树脂对其进行纯化,精简工艺,节省时间,提高了总黄酮和总皂苷的纯度。
2麦冬在中药中以块根入药,此次从麦冬须根中提取出总黄酮和总皂苷,并进行纯化,使得麦冬须根最大限度地被利用,变废为宝,节省成本。

为使本发明的目的、技术方案及优点更加清楚明白,以下结合实施例,对本发明做进一步详细说明。
如图1所示,从麦冬须根中提取纯化总黄酮和总皂苷的方法,包括以下步骤:
(1)以体积浓度86%乙醇,料液比1:15热回流提取,提取3次,每次95min;减压浓缩得到麦冬须根提取液(密度1.25~1.30g/mL);;
(2)麦冬须根提取液先加入等量无水乙醇涡旋后静置5min,醇沉除杂,再经过大孔树脂纯化得到洗脱液;采用D101大孔树脂上样,反复上样3次;
(3)大量水冲洗,至洗脱液不浑浊,洗脱液弃去;
(4)以体积浓度10%乙醇洗脱,洗脱液弃去,再以50%乙醇洗脱,收集洗脱液,得到纯化的提取物。

Claims (2)
1.从麦冬须根中提取纯化总黄酮和总皂苷的方法,其特征在于,包括以下步骤:
(1)以体积浓度86%乙醇,料液比1:15热回流提取,提取3次,每次95min;减压浓缩得到麦冬须根提取液,密度1.25~1.30g/mL;
(2)麦冬须根提取液加入等量无水乙醇涡旋后静置5min后10000rpm离心,醇沉除杂,上清液40℃减压旋干得浸膏;
(3)浸膏用纯水按固液比1:20溶解配制为上样液,再按照D101大孔树脂比上样液1:5,固液比,g:mL,反复3次上样;
(4)大量水冲洗,至洗脱液不浑浊,洗脱液弃去;
(5)以体积浓度10%乙醇洗脱4个柱床体积4BV,洗脱液弃去,再以50%乙醇洗脱4BV,收集洗脱液,旋干得纯化物,其中总黄酮和总皂苷含量大于30%。
2.根据权利要求1所述的从麦冬须根中提取纯化总黄酮和总皂苷的方法,其特征在于,步骤(2)中先将麦冬须根提取液加入等量无水乙醇涡旋后静置5min后离心,醇沉除杂质;步骤(5)采用D101大孔树脂纯化,收集10%~50%乙醇洗脱液。
