Abstract
本发明涉及一种抑菌中药组合物、抑菌中药制剂及其制备方法、应用,属于动物饲料添加剂技术领域。本发明的抑菌中药组合物,由以下重量份数的组分组成:苦参100~400份,黄芪100~400份,十大功劳100~400份。本发明的抑菌中药组合物,将三种单味中药组合制成中药复方,取长补短,不但将三种单味中药的优点结合在一起,具有清热解毒,补中益气,燥湿利尿,调节机体免疫力,增加胃肠运动,对金黄色葡萄球菌、沙门氏菌都具有较强的抑菌作用;并且在临床使用过程中也会降低成本,在以后的市场上具有良好的应用前景。

技术领域
本发明涉及一种抑菌中药组合物、抑菌中药制剂及其制备方法、应用,属于动物饲料添加剂技术领域。
背景技术
金黄色葡萄球菌是目前兽医临床常见的致病菌之一,可引起畜禽皮肤软组织感染、败血症及相关性感染等多种疾病,是目前危及临床患病畜禽的重要致病菌。金黄色葡萄球菌多发生于哺乳仔猪,感染后会出现体质衰弱,病变部位开始是黑色痂块,有的开裂;从眼鼻面部开始向颈部后躯蔓延。到了后期全身呈现麦麸样皮肤屑,粗糙,发出异味,剖检发现皮下出血,肝、脾、肾、心脏等内脏表面有脓性坏死灶,淋巴结肿胀。
猪沙门氏菌病,又名仔猪副伤寒,是由沙门氏菌属细菌引起的仔猪的一种传染病,主要表现为败血症和坏死性肠炎,有时发生脑炎、脑膜炎、卡他性或干酪性肺炎。沙门氏菌是一种条件致病菌,对家养畜禽健康、人类公共安全、食品安全都具有严重的影响。仔猪副伤寒临床上可分为急性型、亚急性与慢性型。急性以急性败血症为特征,多见于断乳前后的仔猪突然死亡,呼吸困难、耳根、胸前和腹下皮腹出现紫色斑;亚急性与慢性仔猪副伤寒较为常见,多以坏死性肠炎、顽固性下痢、卡他性或干酪性肺炎为特征。
目前在临床上发生金黄色葡萄球菌病、猪沙门氏菌病采用抗生素进行治疗,由于抗生素的不合理运用比如滥用等,致使细菌耐药性的问题也逐步趋于严重。为防止这几类细菌病的产生,国内外研究出多种疫苗和多价灭活苗,由于这几类细菌病的血清型较多,免疫效果不甚理想。目前也有关于中药对金黄色葡萄球菌、沙门氏菌的抑制作用的研究,但效果不太理想。
发明内容
本发明的目的在于提供一种抑菌中药组合物,该中药组合物对金黄色葡萄球菌、沙门氏菌具有较好的抑制作用。
本发明的第二个目的在于提供一种抑菌中药制剂,该中药制剂对金黄色葡萄球菌、沙门氏菌具有较好的抑制作用且不易产生耐药性。
本发明的第三个目的在于提供一种上述抑菌中药制剂的制备方法。
本发明的第四个目的在于提供一种上述抑菌中药制剂在饲料添加剂方面的应用。
为实现上述目的,本发明的技术方案是:
一种抑菌中药组合物,由以下重量份数的组分组成:苦参100~400份,黄芪100~400份,十大功劳100~400份。
上述抑菌中药组合物,将各组分粉碎、混匀即得。
所述抑菌中药组合物为抑制金黄色葡萄球菌、沙门氏菌的中药组合物。
一种抑菌中药制剂,主要由以下重量份数的原料制成:苦参100~400份,黄芪100~400份,十大功劳100~400份。
上述抑菌中药制剂为抑制金黄色葡萄球菌、沙门氏菌的中药制剂。
所述抑菌中药制剂为散剂、颗粒剂或口服液。
苦参:苦参为豆科植物苦参的干燥根。根中含有多种生物碱(苦参碱等)、黄酮类化合物、醌类化合物等成分。苦参有清热燥湿、杀虫、利尿的功效,可以治疗湿热、黄疽、痢疾、肠炎、皮肤瘙痒等症状。苦参对结核杆菌和皮肤真菌有抑制作用。体外试验有抗滴虫功效。对多种病毒、细菌有显著的抑制作用,尤其对金黄色葡萄球菌具有较强的抑菌作用。
黄芪:又名黄耆,为植物和中药材的统称。中药材黄芪为豆科草本植物蒙古黄芪、膜荚黄芪的根,具有补气固表、利水退肿、托毒排脓、生肌等功效。黄芪含皂甙、蔗糖、多糖、多种氨基酸、叶酸及硒、锌、铜等多种微量元素。黄芪用于提高肺与呼吸系统免疫能力,用于因免疫力低下导致的虚汗病症,也可以用于补气和增进脾脏功能和代谢能力低下。黄芪有益气固表、利水消肿、脱毒、生肌的功效,适用于自汗、盗汗、浮肿、痈疽不溃或溃久不敛等症。蜜炙黄芪有补气、养血、益中功效,适用于内伤劳倦、脾虚泄泻、气虚、血虚、气衰等症。
十大功劳:属暖温带植物,具有较强的抗寒能力,不耐暑热。喜温暖湿润的气候,性强健、耐荫、忌烈日曝晒,有一定的耐寒性,也比较抗干旱。十大功劳花性凉,味甘。根、茎性寒,味苦。含小檗碱、药根碱、木兰花碱等。有清热解毒、止咳化痰之功效。对金黄色葡萄球菌、痢疾杆菌、大肠菌有抑制作用。主治细菌性痢疾、胃肠炎、传染性肝炎、支气管炎、咽喉肿痛、结膜炎、烧伤、烫伤等症。秋末采果实、挖根,全年采叶,晒干或烘干备用。

本发明的抑菌中药制剂,以黄芪、苦参、十大功劳复配,其中苦参中的苦参碱、十大功劳中的小劈碱,再加上黄芪中的皂甙、多种氨基酸等成分的协同作用,可以干扰细菌细胞壁的合成,损伤胞浆膜,影响细胞蛋白合成;影响核酸合成,干扰遗传密码复制等进而对金黄色葡萄球菌、沙门氏菌具有良好的抑制作用。
本发明的抑菌中药制剂,原料简单,给药方便,治疗时间短,治愈率高,原料质量可控,毒副作用低,不易产生耐药性。
上述抑菌中药制剂的制备方法,包括以下步骤:
将原料药苦参、黄芪、十大功劳分别进行提取浓缩处理,得苦参浓缩液、黄芪浓缩液、十大功劳浓缩液;将苦参浓缩液、黄芪浓缩液、十大功劳浓缩液合并,即得。
上述原料苦参、黄芪、十大功劳在加水浸泡前洗净、晾干。将晾干后的原料切至长度为1cm左右。
所述提取浓缩处理包括如下步骤:
1)将原料药分别在水中浸泡20~40min后弃水,然后用乙醇回流提取并过滤2~4次,合并滤液及滤渣,将滤液浓缩至密度为1.00~1.10g/mL,得第一浓缩液;所述回流提取的时间为2~3h;
2)将滤渣加水真空提取3~5h后过滤,将滤液浓缩至密度为0.90~1.10g/mL,得第二浓缩液;将第一浓缩液和第二浓缩液合并。
步骤1)中将滤液浓缩前蒸发回收乙醇。
步骤1)中的浓缩温度为45~50℃。
步骤2)中真空提取的温度≤70℃。
上述浓缩液合并后加入赋形剂。所述赋形剂为葡萄糖、糊精中的一种或两种。
上述加入赋形剂后于40~50℃干燥。
上述抑菌中药制剂在饲料添加剂方面的应用。
本发明的抑菌中药组合物,将黄芪、苦参、十大功劳三味药复配,具有较好的抑制金黄色葡萄球菌、沙门氏菌的作用。
本发明的抑菌中药制剂,配伍合理,中药复方是中药的精髓,任何一味药其作用不是单一的,根据临床的不同需要进行适当配伍,可充分发挥一药多能的效果。本发明的抑菌中药制剂,将黄芪、苦参、十大功劳按照配方量进行复配,不仅对金黄色葡萄球菌、沙门氏菌具有较强的抑制作用,还能增强基体免疫力,清热解毒,补中益气,燥湿利尿,治愈率高,毒副作用低,不易产生耐药性。
本发明的抑菌中药制剂的制备方法,简单可行,可根据需要制成不同形态,给药方便,更适合全群给药并且还容易被机体吸收。

1)将苦参洗净、晒干,并切至长度为1cm左右,然后加入适量蒸馏水,蒸馏水不超过药面,浸泡20min,弃水,将弃水后的苦参用乙醇回流提取3h,过滤;重复上述乙醇回流提取、过滤3次,合并滤液及滤渣,蒸发回收乙醇,收集浓缩液,将浓缩液密度浓缩至1.00g/mL,得苦参第一浓缩液;将滤渣加水于≤70℃的温度下真空提取4h,之后过滤,将各滤液浓缩至密度为1.00g/mL,得苦参第二浓缩液;合并苦参第一浓缩液和苦参第二浓缩液,得苦参浓缩液;
2)将黄芪洗净、晒干,并切至长度为1cm左右,然后加入适量蒸馏水,蒸馏水不超过药面,浸泡20min,弃水,将弃水后的黄芪用乙醇回流提取3h,过滤;重复上述乙醇回流提取、过滤3次,合并滤液及滤渣,回收乙醇,收集浓缩液,将浓缩液密度浓缩至1.00g/mL,得黄芪第一浓缩液;将滤渣加水于≤70℃的温度下真空提取4h,之后过滤,将各滤液浓缩至密度为1.00g/mL,得黄芪第二浓缩液;合并黄芪第一浓缩液和黄芪第二浓缩液,得黄芪浓缩液;
3)将十大功劳洗净、晒干,并切至长度为1cm左右,然后加入适量蒸馏水,蒸馏水不超过药面,浸泡20min,弃水,将弃水后的十大功劳用乙醇回流提取3h,过滤;重复上述乙醇回流提取、过滤3次,合并滤液及滤渣,回收乙醇,收集浓缩液,将浓缩液密度浓缩至1.00g/mL,得十大功劳第一浓缩液;将滤渣加水于≤70℃的温度下真空提取4h,之后过滤,将各滤液浓缩至密度为1.00g/mL,得十大功劳第二浓缩液;合并十大功劳第一浓缩液和十大功劳第二浓缩液,得十大功劳浓缩液;
4)合并步骤1)~3)所得的苦参浓缩液、黄芪浓缩液、十大功劳浓缩液,然后加入40份葡萄糖、40份糊精制成颗粒,于40℃干燥即得。
上述抑菌中药制剂可作为饲料添加剂。
实施例2
本实施例的抑菌中药组合物,由以下重量份数的组分组成:苦参400份,黄芪400份,十大功劳400份。

2)将黄芪洗净、晒干,并切至长度为1cm左右,然后加入适量蒸馏水,蒸馏水不超过药面,浸泡40min,弃水,将弃水后的黄芪用乙醇回流提取2h,过滤;重复上述乙醇回流提取、过滤4次,合并滤液及滤渣,回收乙醇,收集浓缩液,将浓缩液密度浓缩至1.10g/mL,得黄芪第一浓缩液;将滤渣加水于≤70℃的温度下真空提取3h,之后过滤,将各滤液浓缩至密度为1.05g/mL,得黄芪第二浓缩液;合并黄芪第一浓缩液和黄芪第二浓缩液,得黄芪浓缩液;
3)将十大功劳洗净、晒干,并切至长度为1cm左右,然后加入适量蒸馏水,蒸馏水不超过药面,浸泡40min,弃水,将弃水后的十大功劳用乙醇回流提取2h,过滤;重复上述乙醇回流提取、过滤4次,合并滤液及滤渣,回收乙醇,收集浓缩液,将浓缩液密度浓缩至1.10g/mL,得十大功劳第一浓缩液;将滤渣加水于≤70℃的温度下真空提取3h,之后过滤,将各滤液浓缩至密度为1.05g/mL,得十大功劳第二浓缩液;合并十大功劳第一浓缩液和十大功劳第二浓缩液,得十大功劳浓缩液;
4)合并步骤1)~3)所得的苦参浓缩液、黄芪浓缩液、十大功劳浓缩液,然后加入40份葡萄糖、40份糊精制成颗粒,于50℃干燥即得。
上述中药制剂可作为饲料添加剂。
实施例3
本实施例的抑菌组合物,由以下重量份数的组分组成:苦参200份,黄芪200份,十大功劳200份。
本实施例的抑菌中药制剂,由以下重量份数的原料制成:苦参200份,黄芪200份,十大功劳200份。
上述抑菌中药制剂的制备方法,包括以下步骤:
1)将苦参洗净、晒干,并切至长度为1cm左右,然后加入适量蒸馏水,蒸馏水不超过药面,浸泡30min,弃水,将弃水后的苦参用乙醇回流提取2.5h,过滤;重复上述乙醇回流提取、过滤2次,合并滤液及滤渣,回收乙醇,收集浓缩液,将浓缩液密度浓缩至1.10g/mL,得苦参第一浓缩液;将滤渣加水于≤70℃的温度下真空提取5h,之后过滤,将各滤液浓缩至密度为1.00g/mL,得苦参第二浓缩液;合并苦参第一浓缩液和苦参第二浓缩液,得苦参浓缩液;
2)将黄芪洗净、晒干,并切至长度为1cm左右,然后加入适量蒸馏水,蒸馏水不超过药面,浸泡30min,弃水,将弃水后的黄芪用乙醇回流提取2.5h,过滤;重复上述乙醇回流提取、过滤2次,合并滤液及滤渣,回收乙醇,收集浓缩液,将浓缩液密度浓缩至1.10g/mL,得黄芪第一浓缩液;将滤渣加水于≤70℃的温度下真空提取5h,之后过滤,将各滤液浓缩至密度为1.00g/mL,得黄芪第二浓缩液;合并黄芪第一浓缩液和黄芪第二浓缩液,得黄芪浓缩液;
3)将十大功劳洗净、晒干,并切至长度为1cm左右,然后加入适量蒸馏水,蒸馏水不超过药面,浸泡30min,弃水,将弃水后的十大功劳用乙醇回流提取2.5h,过滤;重复上述乙醇回流提取、过滤2次,合并滤液及滤渣,回收乙醇,收集浓缩液,将浓缩液密度浓缩至1.10g/mL,得十大功劳第一浓缩液;将滤渣加水于≤70℃的温度下真空提取5h,之后过滤,将各滤液浓缩至密度为1.00g/mL,得十大功劳第二浓缩液;合并十大功劳第一浓缩液和十大功劳第二浓缩液,得十大功劳浓缩液;
4)合并步骤1)~3)所得的苦参浓缩液、黄芪浓缩液、十大功劳浓缩液即得。
试验例1
对实施例1中的抑菌中药制剂进行金黄色葡萄球菌及猪沙门氏菌的抑制试验。
一、金黄色葡萄球菌:
(1)断奶仔猪试验组:
饲喂前的临床症状:病初精神沉郁,体温升高,触摸患猪皮肤温度增高,被毛粗乱,渗出物直接粘连,蹄球部的角质脱落。发病猪食欲不振和脱水,严重者体重迅速减轻。
试验组:每50kg饲料中加入100-150g中药制剂,每日饲喂2次,连用6-8天。
对照组:用温水洗全身,擦干后用红霉素软膏涂抹全身,每日1次,用青霉素和复合维生素B结合治疗,连用6-8天。
(2)保育猪试验组:
饲喂前的临床症状:发病猪精神不振,轻度脱水,体温升高,在眼睛周围、耳廓、面颊及鼻背部皮肤,以及肛门周围和下腹部等无被毛处皮肤出现红斑等症状。
试验组:每50kg饲料中加入150-200g中药制剂,每天饲喂2次,连用6-8天。
对照组:用复方蒽诺沙星和氧氟沙星按说明书肌肉注射,2次/天,用青霉素和复合维生素B结合治疗连用6-8天。
(3)育肥猪试验组:
饲喂前临床症状:发病猪精神不振,体温略有升高,食欲减退,全身病变较轻,发病猪多为全身症状感染。
试验组:每50kg饲料中加入250-300g中药制剂,每天饲喂2次,连用6-8天。
对照组:用抗生素青霉素、20%磺胺嘧啶钠注射液加地塞米松治疗,连用6-8天。
二、猪沙门氏菌:
(1)断奶仔猪试验组:
饲喂前临床症状:发病猪常突然死亡。病程稍长者,表现体温升高(41~42℃),腹痛,下痢,呼吸困难,耳根、胸前和腹下皮肤有紫斑,多以死亡告终。
试验组:每50kg饲料中加入100-150g中药制剂,每日饲喂2次,连用6-8天。
对照组:用磺胺甲基异恶唑或磺胺嘧啶每千克体重20-40毫克,加甲氧苄氨嘧啶每千克体重4-8毫克,混合后分两次内服,呋喃唑酮(痢特灵),每日每千克体重20-40毫克,分两次口服,连用6-8天。
(2)保育猪试验组:
饲喂前临床症状:发病猪表现体温升高,眼结膜发炎,有脓性分泌物。初便秘后腹泻,排灰白色或黄绿色恶臭粪便。病猪消瘦,皮肤有痂状湿疹。
试验组:每50kg饲料中加入150-200g中药制剂,每日饲喂2次,连用6-8天。
对照组:用磺胺甲基异恶唑或磺胺嘧啶每千克体重20-40毫克,加甲氧苄氨嘧啶每千克体重4-8毫克,混合后分两次内服,呋喃唑酮(痢特灵),每日每千克体重20-40毫克,分两次口服,连用6-8天。
(3)育肥猪试验组:
饲喂前临床症状:发病猪精神异常,出现轻度腹泻,粪便呈黄绿色或暗绿色。症状时好时坏,反复发作,持续数周,伴以消瘦、脱水而死。
试验组:每50kg饲料中加入250-300g中药制剂,每日饲喂2次,连用6-8天。
对照组:用磺胺甲基异恶唑或磺胺嘧啶每千克体重20-40毫克,加甲氧苄氨嘧啶每千克体重4-8毫克,混合后分两次内服,呋喃唑酮(痢特灵),每日每千克体重20-40毫克,分两次口服,连用6-8天。

注:表1-2中的治愈是指体温正常,精神正常,活泼好动,皮毛光滑,食欲增加,呼吸正常,临床症状消失;有效是指病情好转,精神较好,食欲增加,皮毛不是很好,活动量较少,呼吸有点困难,临床症状逐渐消失;无效是指病情没有好转,甚至有的临床症状逐渐严重。
由表1-2可知,在饲料中添加本发明实施例1的抑菌中药制剂,试验动物的临床症状和生命体征都得到了较好的改善,所发生的细菌病也得到了有效的控制。本发明的中药制剂对金黄色葡萄球菌、猪沙门氏菌等病猪治疗后,病猪的有效率均在88%以上,治疗效果都可达100%,而对照组的较低。说明本发明将三种不同的单味中药相结合,在治疗过程中不但把不同单味中药的优点同时体现了出来,具有清热解毒,补中益气,燥湿利尿等功效,而且在治疗过程中,中药制剂对金黄色葡萄球菌、沙门氏菌具有明显的治疗效果。
试验例2
试验组:用蒸馏水设不同的梯度将实施例2中的中药制剂进行稀释,混匀,备用;用灭菌棉拭子蘸取菌液涂布于平板表面,不断旋转平板,使菌液均匀,最后在平板的周围涂抹菌液。用制备好的药敏纸片蘸取稀释后的中药制剂药液,紧贴于平板上,标记,放入37℃恒温箱中培养18-24h,观察结果,测量抑菌圈直径,结果如表3所示。
对照组:分别以苦参、黄芪、十大功劳的药液为对比例,用灭菌棉拭子蘸取菌液涂布于平板表面,不断旋转平板,使菌液均匀,最后在平板的周围涂抹菌液。用制备好的药敏纸片蘸取药液后,紧贴于平板上,标记,放入37℃恒温箱中培养18-24h,观察结果,测量抑菌圈直径,结果如表4所示。

针对金黄色葡萄球菌、沙门氏菌做体外抑菌试验,由表3内容可知,上述所制备的中药制剂对金黄色葡萄球菌、沙门氏菌均有不同程度的抑菌作用,延缓细菌耐药性的产生。通过对比表3及表4内容可知,本发明中药制剂,将黄芪、苦参、十大功劳复配,在总药量相同的情况下,本发明中药制剂的抑菌效果明显优于任何单味药抑菌效果;当本发明中药制剂含量为0.2g/mL时,其抑菌效果也明显优于0.4g/mL的苦参、黄芪或十大功劳单味药的抑菌效果。
Claims (7)
1.一种抑菌中药制剂在制备饲料添加剂方面的应用,其特征在于,所述饲料添加剂用于抑制金黄色葡萄球菌、沙门氏菌;所述抑菌中药制剂主要由以下重量份数的原料制成:苦参100~400份,黄芪100~400份,十大功劳100~400份;所述抑菌中药制剂的制备方法包括以下步骤:将原料药苦参、黄芪、十大功劳分别进行提取浓缩处理,得苦参浓缩液、黄芪浓缩液、十大功劳浓缩液;将苦参浓缩液、黄芪浓缩液、十大功劳浓缩液合并,即得。
2.根据权利要求1所述的抑菌中药制剂在制备饲料添加剂方面的应用,其特征在于,所述中药制剂为散剂、颗粒剂或口服液。
3.根据权利要求1所述的抑菌中药制剂在制备饲料添加剂方面的应用,其特征在于,所述制备方法中提取浓缩处理包括如下步骤:
1)将原料药分别在水中浸泡20~40min后弃水,然后用乙醇回流提取并过滤2~4次,合并滤液及滤渣,将滤液浓缩至密度为1.00~1.10g/mL,得第一浓缩液;所述回流提取的时间为2~3h;
2)将滤渣加水真空提取3~5h后过滤,将滤液浓缩至密度为0.90~1.10g/mL,得第二浓缩液;将第一浓缩液和第二浓缩液合并。
4.根据权利要求3所述的抑菌中药制剂在制备饲料添加剂方面的应用,其特征在于,步骤1)中的浓缩温度为45~50℃。
5.根据权利要求3所述的抑菌中药制剂在制备饲料添加剂方面的应用,其特征在于,步骤2)中真空提取的温度≤70℃。
6.根据权利要求3所述的抑菌中药制剂在制备饲料添加剂方面的应用,其特征在于,浓缩液合并后加入赋形剂。
7.根据权利要求6所述的抑菌中药制剂在制备饲料添加剂方面的应用,其特征在于,加入赋形剂后于40~50℃干燥。
