本发明涉及禽畜养殖技术领域,尤其涉及一种用于禽畜的的清热解毒和抗病毒中草药及其循环粉碎生物酵制球磨破壁制备工艺。
背景技术
目前,在禽畜养殖行业,中草药的使用已经日益广泛。相对于抗生素和化学合成药以及激素类药物,中药具有其特定的优势。但中草药也有不少缺陷,尤其在剂型方面表现比较突出,如不溶于水、吸收利用度差等。
荆防败毒散目前应用于禽畜病毒感染的预防和治疗,其原料药包括荆芥、防风、羌活、独活、柴胡、前胡、枳壳、茯苓、桔梗、川芍、甘草、薄荷,一般经过粗粉碎后制成散剂,其安全性和有效性已经得到验证,施用到禽畜养殖行业具有较高的可行性;但是存在剂型粗、有效成分溶出和释放效率低下等不足,严重影响了荆防败毒散的施用效果和施治范围。

本发明要解决的技术问题是提供一种禽畜用荆防败毒散破壁超微粉剂、干混悬制剂及其制备工艺,通过高效的固体分散技术开拓出禽畜用荆防败毒散新剂型,提高荆防败毒散在禽畜养殖行业的适用范围和施用效果。
为解决上述技术问题,本发明所采取的技术方案如下。
畜禽用荆防败毒散破壁超微粉,该超微粉包含细胞壁经充分破壁处理的荆芥、防风、羌活、独活、柴胡、前胡、枳壳、茯苓、桔梗、川芍、甘草、薄荷原料药;各原料药首先经循环粉碎和发酵处理后进行细胞壁处理,得到畜禽用荆防败毒散破壁超微粉。
作为本发明的一种优选技术方案,按重量份数计所述超微粉中包含:荆芥20份、防风10份、羌活10份、独活10份、柴胡12份、前胡8份、枳壳10份、茯苓18份、桔梗12份、川芍10份、甘草6份、薄荷5份。
畜禽用荆防败毒散破壁干混悬颗粒剂,将上述超微粉剂添加泊洛沙姆、微晶纤维素、葡萄糖、掩味剂,经充分混匀,添加溶出调节剂,再经流化床制粒,得到干混悬颗粒制剂。
畜禽用荆防败毒散的循环粉碎生物酵制球磨破壁制备工艺,该制备工艺包括如下步骤:
A、循环粉碎工艺:中药材备料干燥后送入粉碎机,在粉碎机的进料口设置送风机,通过气流将中药材吹送到粉碎机的粉碎部,在粉碎部的出料口端设置100-500目的网筛,在网筛外侧设置回风机,此回风机间歇开启,其鼓风压力较所述送风机的鼓风压力高出10-50%;将中药材原料送入粉碎机后通过回风机的作用循环粉碎至过100-500目网筛,得中药材粉料;
B、发酵工艺:采用由秸秆发酵剂、枯草芽孢杆菌组成的菌群并添加纤维素酶和红糖,控制发酵温度30-40℃,pH值5.0-7.0,水分含量35-60% w/w,对步骤A所得中药材粉料进行常压发酵,持续12-48小时,调节pH值至中性,烘干,得中药材酵料;
C、球磨破壁工艺:选用小型的球磨机,并采用0.5-2.5mm微珠对步骤B所得中药材酵料进行连续送料粉碎,所得产物即为经充分破壁的中药材破壁超微粉料。
作为本发明的一种优选技术方案,步骤A中,所述回风机每间隔0.5-1.5min间歇开启,每次启动后持续5-15s再次关闭;反复循环;步骤A中,在粉碎部的出料口端设置200目的网筛。
作为本发明的一种优选技术方案,步骤A中,所述鼓风压力较所述送风机的鼓风压力高出30%;回风机每次启动后持续15s再次关闭;或者所述鼓风压力较所述送风机的鼓风压力高出50%;回风机每次启动后持续5s再次关闭。
作为本发明的一种优选技术方案,步骤B中,秸秆发酵剂、枯草芽孢杆菌在使用前首先在35℃的红糖水中充分活化,使得活菌数目达到100亿/CFU以上;经活化后的菌液、中药材粉料、红糖、纤维素酶的重量份数比为(35-70):(20-40):(10-20):(0.1-1) w/w。
作为本发明的一种优选技术方案,步骤B中,烘干温度不高于65℃;边烘干边送风以加快烘干速度。
作为本发明的一种优选技术方案,步骤B中,采用乙酸、碳酸钠、碳酸氢钠第体系进行pH调节;步骤B中,常压发酵20-40小时。
作为本发明的一种优选技术方案,步骤C中,微珠直径为0.6-1.6mm;产物过400目筛。
采用上述技术方案所产生的有益效果在于:本发明研发的循环粉碎工艺,由于在网筛外侧设置回风机,此回风机间歇开启,其鼓风压力较所述送风机的鼓风压力高出10-50%,能够实现中药材原料的循环粉碎,直至通过网筛获得目标细度的粉料;本发明在循环粉碎工艺的基础上,进一步创新采用了球磨破壁工艺,选用小型的球磨机,并采用0.5-2.5mm微珠对中草药材进行连续送料粉碎,能够实现中草药细胞的充分破壁,提高其有效成分的释放率;为了进一步提高破壁效果和中草药有效成分的释放效率,本发明在球磨破壁处理之前,还对中草药进行了一定程度的发酵处理,即采用秸秆发酵剂、枯草芽孢杆菌组成的菌群并添加纤维素酶和红糖,控制温度、pH值、水分对中药材粉料进行常压发酵,这是一种轻度发酵处理,对于提升中草药的破壁效果具有积极作用。
现有技术中,荆防败毒散是各原料成分经过简单的粗粉碎后混匀制备,本发明研发的工艺与现有技术相比效果十分显著,大幅提高了药物的溶出和吸收效率,提高了中草药的生物利用度,有效的拓展了荆防败毒散在禽畜养殖行业的适用范围和施用效果。
具体实施方式
以下实施例详细说明了本发明。本发明所使用的各种原料及各项设备均为常规市售产品,均能够通过市场购买直接获得。
实施例1、荆防败毒散破壁固体超微粉。
本实施例的荆防败毒散破壁固体超微粉,该超微粉包含细胞壁经充分破壁处理的荆芥、防风、羌活、独活、柴胡、前胡、枳壳、茯苓、桔梗、川芍、甘草、薄荷原料药;具体的,按重量份数计算包含:荆芥20份、防风10份、羌活10份、独活10份、柴胡12份、前胡8份、枳壳10份、茯苓18份、桔梗12份、川芍10份、甘草6份、薄荷5份;主要用于禽畜病毒感染的防治。
实施例2、荆防败毒散破壁干混悬制剂。
将实施例1中的超微粉剂添加泊洛沙姆、微晶纤维素、葡萄糖、掩味剂,经充分混匀,添加溶出调节剂,再经流化床制粒,得到干混悬颗粒制剂;主要用于禽畜病毒感染的防治。
实施例3、荆防败毒散的循环粉碎工艺。
循环粉碎工艺:十二组中药材备料干燥后送入粉碎机,在粉碎机的进料口设置送风机,通过气流将中药材吹送到粉碎机的粉碎部,在粉碎部的出料口端设置网筛,在网筛外侧设置回风机,回风机的鼓风压力较送风机的鼓风压力高出40%,回风机每间隔1min间歇开启,每次启动后持续8s再次关闭,反复循环;将中药材原料送入粉碎机后通过回风机的作用循环粉碎至过300目网筛,得中药材粉料。
实施例4、破壁前处理——轻度发酵工艺。
轻度发酵工艺:采用由秸秆发酵剂、枯草芽孢杆菌组成的菌群并添加纤维素酶和红糖,控制发酵温度35℃,pH值5.0-7.0,水分含量40-52% w/w,对所得中药材粉料进行常压发酵,持续30小时;其中,秸秆发酵剂、枯草芽孢杆菌在使用前首先在35℃的红糖水中充分活化,使得活菌数目达到100亿/CFU以上;经活化后的菌液、中药材粉料、红糖、纤维素酶的重量份数比为50:25:10:0.5 w/w;发酵完成后,调节pH值至中性,65℃以下边烘干边送风完成快速烘干,得中药材酵料。
实施例5、球磨破壁工艺。
选用小型的球磨机,并采用1.2mm微珠对所得中药材酵料进行连续送料粉碎,产物过400目筛,得到经充分破壁的中药材破壁料。
本发明创新开发了针对中草药的独特的球磨工艺,并配合循环粉碎+轻度酵制的前处理工艺,能够实现中草药的高效率破壁处理,大幅提高了药物的溶出和吸收效率,提高了中草药的生物利用度,有效的拓展了荆防败毒散在禽畜养殖行业的适用范围和施用效果。
上述描述仅作为本发明可实施的技术方案提出,不作为对其技术方案本身的单一限制条件。
Claims (10)
1.畜禽用荆防败毒散破壁超微粉,其特征在于:该超微粉包含细胞壁经充分破壁处理的荆芥、防风、羌活、独活、柴胡、前胡、枳壳、茯苓、桔梗、川芍、甘草、薄荷原料药;各原料药首先经循环粉碎和发酵处理后进行细胞壁处理,得到畜禽用荆防败毒散破壁超微粉。
2.根据权利要求1所述的畜禽用荆防败毒散破壁超微粉,其特征在于:按重量份数计所述超微粉中包含:荆芥20份、防风10份、羌活10份、独活10份、柴胡12份、前胡8份、枳壳10份、茯苓18份、桔梗12份、川芍10份、甘草6份、薄荷5份。
3.畜禽用荆防败毒散破壁干混悬颗粒剂,其特征在于:将权利要求1中的超微粉剂添加泊洛沙姆、微晶纤维素、葡萄糖、掩味剂,经充分混匀,添加溶出调节剂,再经流化床制粒,得到干混悬颗粒制剂。
4.畜禽用荆防败毒散的循环粉碎生物酵制球磨破壁制备工艺,其特征在于:该制备工艺包括如下步骤:
A、循环粉碎工艺:中药材备料干燥后送入粉碎机,在粉碎机的进料口设置送风机,通过气流将中药材吹送到粉碎机的粉碎部,在粉碎部的出料口端设置100-500目的网筛,在网筛外侧设置回风机,此回风机间歇开启,其鼓风压力较所述送风机的鼓风压力高出10-50%;将中药材原料送入粉碎机后通过回风机的作用循环粉碎至过100-500目网筛,得中药材粉料;

7.根据权利要求4所述的畜禽用荆防败毒散的循环粉碎生物酵制球磨破壁制备工艺,其特征在于:步骤B中,秸秆发酵剂、枯草芽孢杆菌在使用前首先在35℃的红糖水中充分活化,使得活菌数目达到100亿/CFU以上;经活化后的菌液、中药材粉料、红糖、纤维素酶的重量份数比为(35-70):(20-40):(10-20):(0.1-1) w/w。
