Abstract
本发明提供一种马里兰烟叶发酵提取物的制备方法及其应用,其是将马里兰烟叶粉碎后,采用马里兰烟杆固定化吸附微生物菌株,对马里兰烟粉进行温育水解发酵,再结合连续逆流超声提取,制备马里兰烟叶提取物,该提取物具有天然烟草浓郁本香,刺激性降低,纯化分离步骤简单,工艺条件温和,提取率和生产效率提高;在传统卷烟、新型加热不燃烧卷烟中加料加香应用,具有天然烟草本香,劲头足,透发感强,带有果香气、奶甜香气,与卷烟烟气协调且风格一致。

技术领域
本发明涉及烟用添加剂领域,具体涉及一种马里兰烟叶发酵提取物的制备方法及其应用。
背景技术
烟用添加剂为改善烟草理化性能的助剂,包括香料香精、保润剂、助燃剂、防霉剂和吸附剂。低焦油卷烟的生产更需要加香加料以弥补烟香的不足。烟制品加香加料的目的在于掩盖天然烟草的刺激性和杂气等,并提高或改善其香味。从烟草和烟气中可鉴定出很多香味物质。合成和应用这些具有烟草本身香味的香料,正日益受到重视。天然香料因其天然性、安全性和自然复合的香气,更迎合消费者对健康的需求,其中烟草提取物更是因其天然烟草本香,能更符合卷烟风格特征,在低焦油、低危害卷烟中加香增香,没有外加香气掩盖烟香、引入烟草以外化学物质等缺陷。
马里兰烟属红花烟草,是一种淡色晾烟,口感适中,具有“低烟碱、低焦油”的特点,是生产安全性卷烟的优质原料之一,是斗烟、嚼烟、鼻烟和卷烟的原料,国内常用作混合型卷烟、雪茄烟配方中的调味原料。因此,马里兰烟提取物可以增加卷烟香气风味,是一种具有烟草本香物质的天然香料。常规、普通的烟草提取物制备方法,通常使提取香味物质的同时,带有对卷烟吸味不利的蛋白质、鞣质、淀粉、果胶、纤维素、木质素及其他大分子物质,增香的同时也具有增加刺激性、杂气和残留感的缺陷。
生物产香技术是利用微生物工程、酶工程、细胞培养、基因改造等生物技术,通过微生物代谢出特定香气成分,从而提升产品质量的一门新技术。在生物产香技术制备天然香料的过程中,多采用悬浮微生物发酵,这种方式存在天然浸膏和提取物类香料产品得率低下、产品质量不稳定、纯化步骤复杂、成本高等缺陷,严重阻碍了生物产香天然香料的应用推广。
目前有专利“一种马里兰烟提取物的制备方法及其在卷烟中的应用”(申请号201610853881.0),将马里兰烟原料采用液态二氧化碳加压膨化,采用乙醇或石油醚溶剂萃取,浓缩后分子蒸馏纯化制备;专利“马里兰烟叶多糖、提取纯化方法及其作为抗氧化剂的应用”(申请号201410070873.X),粉碎、加水超声、离心取上清液,旋蒸后醇析,离心取沉淀,Sevage法脱除蛋白,柱层析纯化制备烟叶多糖;专利“一种以马里兰烟为原料制造低烟碱再造烟叶的方法”(申请号201610946212.8),将马里兰烟烟叶用复合酶制剂(中性蛋白酶、纤维素酶、果胶酶、糖化酶、内肽酶、氨基外肽酶)发酵提取,烟梗与水65~75℃进行水提,提取液合并后,加入丙二醇、还原糖、氨基酸和碳酸钠在温度为70℃~90℃条件下减压浓缩制备涂布液,固体残余物抄造为基片,将涂布液涂布到基片上干燥制成低烟碱再造烟叶。而采用微生物菌株吸附到马里兰烟杆,与马里兰烟叶粉末在水溶液中温育水解发酵,结合溶剂连续逆流超声提取制备马里兰烟叶发酵提取物,并在传统卷烟、新型加热不燃烧卷烟中应用,尚未见报道。
发明内容
本发明的目的是提供一种马里兰烟叶发酵提取物的制备方法及其应用,将马里兰烟叶粉碎后,采用马里兰烟杆固定化吸附微生物菌株,对马里兰烟粉进行温育水解发酵,再结合连续逆流超声提取,制备马里兰烟叶提取物,该提取物具有天然烟草浓郁本香,刺激性降低,纯化分离步骤简单,工艺条件温和,提取率和生产效率提高;在传统卷烟、新型加热不燃烧卷烟中加料加香应用,具有天然烟草本香,劲头足,透发感强,带有果香气、奶甜香气,与卷烟烟气协调且风格一致。
本发明通过以下技术方案实现:
一种马里兰烟叶固定化发酵提取物的制备方法,包括如下步骤:
(1)原料前处理:马里兰烟叶经粉碎机粉碎至40目~100目粉末;
(2)微生物菌株的活化:将扣囊复膜孢酵母Saccharomycopsis fibuligera和桑肠杆菌Enterobacter mori VL4-3的活性固体冻干菌粉,加入到10~20倍质量的灭菌处理的质量浓度为5~20%葡萄糖溶液中,置于38~50℃水浴锅活化30~60min,得到微生物菌株活化液,其中所述的桑肠杆菌Enterobacter mori VL4-3保藏号为CCTCC NO:M2012020,于2012年2月16日保藏于武汉大学中国典型培养物保藏中心;
(3)固定化载体制备:将马里兰烟的烟杆洗净烘干后处理成碎块,高压灭菌后烘干;
(4)微生物菌株的固定化处理:称取上述步骤(3)灭菌烘干后的马里兰烟杆碎块装入容器,按照碎块与活化液质量比为1:5~30加入已培养活化好的微生物菌株活化液,放置于4+0.5℃冷藏柜固定12~30h,过滤得到固定化微生物菌株的烟杆碎块;
(5)马里兰烟叶的固定化发酵:将步骤(1)所得马里兰烟叶粉末与15~30倍质量的水混合均匀,加入步骤(4)所述固定化微生物菌株的烟杆碎块,固定化微生物菌株的烟杆碎块与马里兰烟叶粉末质量比为1:0.5~2,在温度为40~55℃,匀速搅拌,添加乙二酸调节pH值为4.5~6.5,发酵1~5d,筛网过滤去除烟杆碎块,微孔过滤除去马里兰烟粉残渣,保留马里兰烟叶发酵液A;
(6)连续逆流超声提取:将步骤(5)所得马里兰烟粉残渣按速率0.1~0.8kg/10min投料至浸出舱,开启主机和超声波发生器,调节温度60~100℃,料液比为质量比1:2~6,从浸出舱低端运行0.2~1h后调节出液阀门均匀放液,控制溶剂加入流量,保持浸出仓液位平衡,连续逆流超声提取达30~60min时,投料加入占马里兰烟粉质量0.1~1%的中性醋酸铝,当连续逆流超声提取共达50~150min时,停止生产工艺,排渣,得到浸出液B;
(7)将步骤(5)的发酵液A和步骤(6)的浸出液B合并,得到马里兰烟发酵提取液。
进一步的,还包括步骤(8):将所述马里兰烟发酵提取液经真空冷冻干燥得到马里兰烟发酵提取物。
进一步的,所述步骤(2)中的活性固体冻干菌粉为食品级,活性为10~20u/mg。
进一步的,所述步骤(3)中的马里兰烟的烟杆用剪刀剪成约1cm长的小块。
进一步的,所述步骤(5)中匀速搅拌的转速为20~100rpm。
进一步的,所述步骤(6)中使用的中性醋酸铝为分析纯。
本发明还提供一种上述方法制备的马里兰烟发酵提取液在传统卷烟配方烟丝加料中的应用,按0.01~0.1%烟丝的质量比的马里兰烟叶发酵提取液和糖分、水混匀,喷洒到制丝加料罐中,在40~60℃与传统卷烟配方烟叶不停搅拌混匀,按正常卷烟工艺加工成配方烟丝,具有协调卷烟香气,劲头增加、透发感增强,烟香醇和、带有果香气、奶甜香气和雪茄香气。
本发明还提供一种上述方法制备的马里兰烟发酵提取液在传统烟草配方烟叶中的应用,在制丝车间加料工艺中,将马里兰烟发酵提取液(0.01~0.1%烟丝质量比)和复合氨基酸(苯丙氨酸、天冬氨酸、半胱氨酸,食品级,与马里兰烟发酵提取液的质量比为0.1~1.5%,3种氨基酸之间的质量比为1:0.5~2:0.5~2)与水、糖料混合均匀,喷洒到加料罐的烟叶上,经后续制丝工序加工成成品配方烟丝,卷烟香气醇和、劲头足、带有奶甜香和浓烈的烘烤香气。
本发明还提供一种上述方法制备的马里兰烟发酵提取物在新型加热不燃烧卷烟烟丝中加香的应用,溶解到70%乙醇溶液中,稀释1000倍,按与烟丝0.01~0.1%的质量比加入烟丝中,卷烟香气浓郁醇厚、劲头足、带有奶甜清香气。
本发明提供的马里兰烟发酵提取物,采用马里兰烟杆作为吸附固定化载体,天然且为烟草本源物质,不引入外界杂源化学物质或香气,且固定化发酵方式使微生物菌株容易与发酵液直接过滤分离,后期纯化简单;马里兰烟叶粉末溶解在水中,与吸附在马里兰烟杆上的微生物菌株的酶体系进行弱酸性条件下的温育水解发酵,扣囊复膜孢酵母(Saccharomycopsis fibuligera)具有产淀粉酶、β-葡萄糖苷酶、酸性蛋白酶等特性,能够高效降解纤维素、淀粉、纤维二糖、蔗糖等细胞壁大分子物质转化为葡萄糖并进一步转化为酯香为主的香味物质、将非挥发性糖苷转化成挥发性的香气物质,产香产酯除杂味,破坏细胞壁更多地释放细胞中的活性化学物质,提高产率和增加香气自然复杂性;桑肠杆菌(Enterobacter mori)具有产β-半乳糖苷酶活性转化非挥发性糖苷为挥发性香气物质及转化细胞壁中阿魏酸生成天然香兰素的能力,能分解利用葡萄糖、蔗糖等糖类物质产醇、酯等香气成分,使马里兰烟发酵液中带有香兰素及其香味前体物质等奶香气成分,并破坏细胞壁释放更多有效成分,提高提取率,两种活性菌株发酵构成的复杂酶活力体系,协调强效联合作用,更好地转化非挥发性糖苷为挥发性香气成分,更有效地分解细胞壁中的纤维素、纤维二糖、阿魏酸释放胞内活性成分,降解多糖类物质为小分子成分,所有这些成分都释放到发酵液中,经简单过滤即可与固定化载体-微生物菌株分离;再结合连续逆流超声溶剂提取马里兰烟叶发酵物,工艺条件温和、缩短生产时间、提高生产效率、降低成本,生产过程中添加中性醋酸铝进一步吸附提取液中残留的杂质如鞣质成分,总的来说,使得杂质大分子成分包括蛋白质、多糖、淀粉等分解为香味物质,杂质杂味减少,刺激性降低,香气种类自然复杂性增加,杂味减少,香气品质提高,赋予马里兰固定化发酵提取液/物具有天然烟草本香和雪茄香气的同时,还带有自然复合香气包括果香气、奶甜香气,生产工艺条件温和、步骤简单、易操作、成效高、成本低,杂质少,香料品质提升,粉末状易运输、贮存,液状可直接加料应用减少稀释、混匀步骤;在传统卷烟配方烟叶加料和新型加热不燃烧烟草中加香的应用,与烟气协调、风格一致,增强烟草本香、劲头足、透发感好、香气醇和,带有奶香气和雪茄香气,口感改善,工艺条件温和易实现,操作步骤简单。
具体实施方式
以下具体实施例对本发明进行详细地说明。
实施例1
一种马里兰烟叶固定化发酵提取物的制备方法,包括如下步骤:
(1)原料前处理:马里兰烟叶经粉碎机粉碎至40目粗粉;
(2)微生物菌株的活化:将扣囊复膜孢酵母(Saccharomycopsis fibuligera)和桑肠杆菌Enterobacter moriVL4-3的活性固体冻干菌粉(食品级,活性为10u/mg),加入到10倍质量的灭菌处理的5%葡萄糖溶液中,置于38℃水浴锅活化30min,得到微生物菌株活化液;
(3)固定化载体制备:将固定化载体(马里兰烟的烟杆)洗净烘干后用剪刀剪成约1cm长的小块,高压灭菌后烘干;
(4)微生物菌株的固定化处理:称取上述步骤3)灭菌烘干后的马里兰烟杆碎块装到20L桶中,加入一定量(碎块与活化液质量比为1:5)步骤2)已培养活化好的微生物菌株活化液,放置于4+0.5℃冷藏柜固定12h,过滤得到固定化微生物菌株的烟杆碎块;
(5)马里兰烟叶的固定化发酵:将步骤1)所述的马里兰烟叶粉末与15倍质量的水混合均匀,加入步骤4)所述固定化微生物菌株的烟杆碎块(与马里兰烟叶粉末质量比为1:0.5),在温度为40℃,匀速搅拌(20rpm),添加乙二酸调节pH值为4.5,发酵1d,筛网过滤去除烟杆碎块,微孔过滤除去马里兰烟粉残渣,保留马里兰烟叶发酵液A;
(6)连续逆流超声提取:将步骤5)马里兰烟粉残渣按速率0.1kg/10min投料,开启主机和超声波发生器,调节温度60℃,料液比为质量比1:2,从浸出舱低端运行0.2h后调节出液阀门均匀放液,控制溶剂加入流量,保持浸出仓液位平衡,连续逆流超声提取达30min时,投料加入中性醋酸铝(分析纯,占马里兰烟粉质量的0.1%),当连续逆流超声提取共达50min时,停止生产工艺,排渣,得到浸出液B;
(7)将步骤5)的发酵液A和步骤6)的浸出液B合并,得到马里兰烟发酵提取液,经真空冷冻干燥得到马里兰烟发酵提取物。
将上述制备的马里兰烟发酵提取液按0.01%烟丝的质量比,与糖分、水混匀,喷洒到制丝加料罐中,在40℃与传统卷烟配方烟叶不停搅拌混匀,按正常卷烟工艺加工成配方烟丝,具有协调卷烟香气,劲头增加、透发感增强,烟香醇和、带有果香气、奶甜香气和雪茄香气。
实施例2
一种马里兰烟叶固定化发酵提取物的制备方法,包括如下步骤:
(1)原料前处理:马里兰烟叶经粉碎机粉碎至60目粗粉;
(2)微生物菌株的活化:将扣囊复膜孢酵母(Saccharomycopsis fibuligera)和桑肠杆菌Enterobacter moriVL4-3的活性固体冻干菌粉(食品级,活性为15u/mg),加入到15倍质量的灭菌处理的12%葡萄糖溶液中,置于45℃水浴锅活化45min,得到微生物菌株活化液;
(3)固定化载体制备:将固定化载体(马里兰烟的烟杆)洗净烘干后用剪刀剪成约1cm长的小块,高压灭菌后烘干;
(4)微生物菌株的固定化处理:称取上述步骤3)灭菌烘干后的马里兰烟杆碎块装到20L桶中,加入一定量(碎块与活化液质量比为1:20)步骤2)已培养活化好的微生物菌株活化液,放置于4+0.5℃冷藏柜固定24h,过滤得到固定化微生物菌株的烟杆碎块;
(5)马里兰烟叶的固定化发酵:将步骤1)所述的马里兰烟叶粉末与25倍质量的水混合均匀,加入步骤4)所述固定化微生物菌株的烟杆碎块(与马里兰烟叶粉末质量比为1:1.2),在温度为48℃,匀速搅拌(40rpm),添加乙二酸调节pH值为5.0,发酵3d,筛网过滤去除烟杆碎块,微孔过滤除去马里兰烟粉残渣,保留马里兰烟叶发酵液A;
(6)连续逆流超声提取:将步骤5)马里兰烟粉残渣按速率0.4kg/10min投料,开启主机和超声波发生器,调节温度80℃,料液比为质量比1:4,从浸出舱低端运行0.6h后调节出液阀门均匀放液,控制溶剂加入流量,保持浸出仓液位平衡,连续逆流超声提取达40min时,投料加入中性醋酸铝(分析纯,占马里兰烟粉质量的0.5%),当连续逆流超声提取共达80min时,停止生产工艺,排渣,得到浸出液B
(7)将步骤5)的发酵液A和步骤6)的浸出液B合并,得到马里兰烟发酵提取液,经真空冷冻干燥得到马里兰烟发酵提取物;
在制丝车间加料工艺中,将上述制备的马里兰烟发酵提取液(0.05%烟丝质量比)和复合氨基酸(苯丙氨酸、天冬氨酸、半胱氨酸,食品级,与马里兰烟发酵提取液的质量比为0.1%,3种氨基酸之间的质量比为1:0.6:1.2)与水、糖料混合均匀,经后续制丝工序加工为成品配方烟丝,具有协调烟气,香气醇和、劲头足、带有果香、奶香和浓烈烘烤香气;
实施例3
一种马里兰烟叶固定化发酵提取物的制备方法,包括如下步骤:
(1)原料前处理:马里兰烟叶经粉碎机粉碎至100目粉末;
(2)微生物菌株的活化:将扣囊复膜孢酵母(Saccharomycopsis fibuligera)和桑肠杆菌Enterobacter moriVL4-3的活性固体冻干菌粉(食品级,活性为20u/mg),加入到20倍质量的灭菌处理的20%葡萄糖溶液中,置于50℃水浴锅活化60min,得到微生物菌株活化液;
(3)固定化载体制备:将固定化载体(马里兰烟的烟杆)洗净烘干后用剪刀剪成约1cm长的小块,高压灭菌后烘干;
(4)微生物菌株的固定化处理:称取上述步骤3)灭菌烘干后的马里兰烟杆碎块装到20L桶中,加入一定量(碎块与活化液质量比为1:30)步骤2)已培养活化好的微生物菌株活化液,放置于4+0.5℃冷藏柜固定30h,过滤得到固定化微生物菌株的烟杆碎块;
(5)马里兰烟叶的固定化发酵:将步骤1)所述的马里兰烟叶粉末与30倍质量的水混合均匀,加入步骤4)所述固定化微生物菌株的烟杆碎块(与马里兰烟叶粉末质量比为1:2),在温度为55℃,匀速搅拌(100rpm),添加乙二酸调节pH值为6.5,发酵5d,筛网过滤去除烟杆碎块,微孔过滤除去马里兰烟粉残渣,保留马里兰烟叶发酵液A;
(6)连续逆流超声提取:将步骤5)马里兰烟粉残渣按速率0.8kg/10min投料,开启主机和超声波发生器,调节温度100℃,料液比为质量比1:6,从浸出舱低端运行1h后调节出液阀门均匀放液,控制溶剂加入流量,保持浸出仓液位平衡,连续逆流超声提取达60min时,投料加入中性醋酸铝(分析纯,占马里兰烟粉质量的1%),当连续逆流超声提取共达150min时,停止生产工艺,排渣,得到浸出液B;
(7)将步骤5)的发酵液A和步骤6)的浸出液B合并,得到马里兰烟发酵提取液,经真空冷冻干燥得到马里兰烟发酵提取物。
本发明还提供一种上述方法制备的马里兰烟发酵提取物粉末在新型加热不燃烧卷烟烟丝中加香的应用,溶解到70%乙醇溶液中,稀释1000倍,按0.01%烟丝的质量比加香应用,与传统卷烟烟气接近,烟香协调,具有香气浓郁醇厚、劲头足,并带有果香气和奶甜清香气。
上述实施例中,所述的桑肠杆菌Enterobacter moriVL4-3保藏号为CCTCC NO:M2012020,于2012年2月16日保藏于武汉大学中国典型培养物保藏中心。
试验例1:应用的感官效果对比评价
将实施例1-3制备的马里兰烟叶发酵提取物与空白对照,在传统烟丝和新型加热不燃烧卷烟烟丝中进行感官评吸对比实验。
结果表明(如表1所示):本发明提供的马里兰烟叶发酵提取物在传统卷烟配方烟叶加料和新型加热不燃烧烟草中加香的应用,与烟气协调、风格一致,能增强烟草本香、劲头足、透发感好、香气醇和,带有奶香气和雪茄香气,口感改善,刺激性降低、杂气减少。

Claims (9)
1.一种马里兰烟叶发酵提取物的制备方法,其特征在于包括如下步骤:
(1)原料前处理:马里兰烟叶经粉碎机粉碎至40目~100目粉末;
(2)微生物菌株的活化:将扣囊复膜孢酵母Saccharomycopsis fibuligera和桑肠杆菌Enterobacter mori VL4-3的活性固体冻干菌粉,加入到10~20倍质量的灭菌处理的质量浓度为5~20%葡萄糖溶液中,置于38~50℃水浴锅活化30~60min,得到微生物菌株活化液,其中所述的桑肠杆菌Enterobacter moriVL4-3保藏号为CCTCC NO:M2012020,于2012年2月16日保藏于武汉大学中国典型培养物保藏中心;
(3)固定化载体制备:将马里兰烟的烟杆洗净烘干后处理成碎块,高压灭菌后烘干;
(4)微生物菌株的固定化处理:称取上述步骤(3)灭菌烘干后的马里兰烟杆碎块装入容器,按照碎块与活化液质量比为1:5~30加入已培养活化好的微生物菌株活化液,放置于4±0.5℃冷藏柜固定12~30h,过滤得到固定化微生物菌株的烟杆碎块;
(5)马里兰烟叶的固定化发酵:将步骤(1)所得马里兰烟叶粉末与15~30倍质量的水混合均匀,加入步骤(4)所述固定化微生物菌株的烟杆碎块,固定化微生物菌株的烟杆碎块与马里兰烟叶粉末质量比为1:0.5~2,在温度为40~55℃,匀速搅拌,添加乙二酸调节pH值为4.5~6.5,发酵1~5d,筛网过滤去除烟杆碎块,微孔过滤除去马里兰烟粉残渣,保留马里兰烟叶发酵液A;
(6)连续逆流超声提取:将步骤(5)所得马里兰烟粉残渣按速率0.1~0.8kg/10min投料至浸出舱,开启主机和超声波发生器,调节温度60~100℃,料液比为质量比1:2~6,从浸出舱低端运行0.2~1h后调节出液阀门均匀放液,控制溶剂加入流量,保持浸出仓液位平衡,连续逆流超声提取达30~60min时,投料加入占马里兰烟粉质量0.1~1%的中性醋酸铝,当连续逆流超声提取共达50~150min时,停止生产工艺,排渣,得到浸出液B;
(7)将步骤(5)的发酵液A和步骤(6)的浸出液B合并,得到马里兰烟发酵提取液。
2.如权利要求1所述的马里兰烟叶发酵提取物的制备方法,其特征在于:还包括步骤(8):将所述马里兰烟发酵提取液经真空冷冻干燥得到马里兰烟发酵提取物。

4.如权利要求1所述的马里兰烟叶发酵提取物的制备方法,其特征在于:所述步骤(3)中的马里兰烟的烟杆用剪刀剪成1cm长的小块。
5.如权利要求1所述的马里兰烟叶发酵提取物的制备方法,其特征在于:所述步骤(5)中匀速搅拌的转速为20~100rpm。
6.如权利要求1所述的马里兰烟叶发酵提取物的制备方法,其特征在于:所述步骤(6)中使用的中性醋酸铝为分析纯。
7.一种如权利要求1所述方法制备的马里兰烟叶发酵提取液在传统卷烟配方烟丝加料中的应用,将马里兰烟叶发酵提取液和糖分、水混匀喷洒到制丝加料罐中,在40~60℃与传统卷烟配方烟叶不停搅拌混匀,按正常卷烟工艺加工成配方烟丝,其中马里兰烟叶发酵提取液与烟丝质量比为0.01%或0.05%。
8.一种如权利要求1所述方法制备的马里兰烟叶发酵提取液在传统烟草配方烟叶中的应用,在制丝车间加料工艺中,将马里兰烟发酵提取液和复合氨基酸与水、糖料混合均匀,喷洒到加料罐的烟叶上,经后续制丝工序加工成成品配方烟丝,其中马里兰烟发酵提取液与烟丝质量比为0.01%或0.05%,复合氨基酸与马里兰烟发酵提取液的质量比为0.1%,复合氨基酸由苯丙氨酸、天冬氨酸、半胱氨酸按照1:0.6:1.2质量比混合而成。
9.一种如权利要求2所述方法制备的马里兰烟叶发酵提取物在新型加热不燃烧卷烟烟丝中加香的应用,将所述马里兰烟叶发酵提取物溶解于70%乙醇溶液中,稀释1000倍,按与烟丝0.01%的质量比加入烟丝中。
