Background
Donkey-hide gelatin (Asini Corii Colla) is a gelatin block prepared by decocting and concentrating dried or fresh skin of donkey of Equidae, is a traditional and rare Chinese medicine in China, is called 'three treasures of traditional Chinese medicine' together with ginseng and pilose antler, and is called 'Sheng medicine for enriching blood'. Donkey-hide gelatin is sweet in taste and neutral in nature, enters lung, liver and kidney meridians, and has the effects of nourishing yin, supplementing blood, moistening dryness, stopping bleeding and preventing miscarriage. According to the record of this Jing, it is said that "the heart is dominated, the abdomen is collapsed, the body is strained and sprinkled like malaria, the pain of waist and abdomen, the pain of limbs, the bleeding of women, the miscarriage prevention, the body is lightened and the blood is benefited after long-term taking". Li Shizhen is also recorded in Ben Cao gang mu in great detail: "treat hematemesis, epistaxis, bloody stranguria and urine, intestinal wind and dysentery, female blood pain and blood depletion, irregular menstruation, metrorrhagia, leukorrhagia, diseases before and after fetus, all wind diseases of male and female, bone joint pain, edema of water and vapor, consumptive disease and cough, dyspnea, lung withering and saliva and pus blood, carbuncle and toxic swelling, blood nourishing, wind dispelling and dryness moistening, phlegm reducing, lung heat clearing and urination promoting, intestine regulating, and holy medicine is also included". Modern researches have extensive pharmacological actions of donkey-hide gelatin, such as increasing the intake of calcium in vivo, promoting hematopoiesis, enhancing immunity, resisting anoxia, resisting radiation, resisting fatigue, promoting bone healing, resisting shock, resisting cold, enhancing memory, resisting asthma, resisting tumor, etc.
The administration method of colla Corii Asini comprises melting method, conventional method and dietetic method. Wherein the melting method comprises crushing commercially available colla Corii Asini blocks (wrapping with letter paper or cloth, placing on a relatively hard table, and crushing into powder with hammer), placing 3-9 g (about one third of each block) into a cup, adding small amount of crystal sugar, dissolving with boiling water or decoction, stirring, and cooling. The traditional method is that 250g of donkey-hide gelatin is taken, smashed and placed in a container with a cover, 250g (or ml) of rock sugar, yellow wine and water are added respectively, the mixture is soaked for 24-48 h, after the gelatin is softened and does not have hard blocks, a proper amount of black sesame, walnut kernels, Chinese dates, longan pulp and the like are added according to needs and stirred uniformly, the mixture is placed in a pot and steamed for 0.5-1h in water in a separating way, the mixture is taken out and cooled, the pot is covered and placed in a shady place or a refrigerator, 1-2 times a day with 1-2 tablespoons each time, and the donkey-hide gelatin is taken with boiled water. The dietotherapy comprises preparing foods such as colla Corii Asini porridge, colla Corii Asini egg soup, colla Corii Asini shallot white honey, colla Corii Asini wine, and colla Corii Asini-cooked fructus Jujubae. The donkey-hide gelatin products can be taken only after post-treatment, and the use is troublesome, and if the donkey-hide gelatin products are not treated properly, the taking effect of the donkey-hide gelatin can be influenced. The existing commercially available donkey-hide gelatin slurry, such as the Dong donkey-hide gelatin compound donkey-hide gelatin slurry with the Chinese medicine standard Z20083345, has single component and unknown using effect. In the prior art, no non-medicinal donkey-hide gelatin nutrient solution is prepared by taking donkey-hide gelatin, allium macrostemon, clove, medlar, fructus amomi, mulberry leaf, dried orange peel, honeysuckle, mint, Chinese date, rose and radix codonopsitis as raw materials according to a scientific proportion.
Disclosure of Invention
The first purpose of the present invention is to provide a non-medicinal donkey-hide gelatin nutrient solution aiming at the defects in the prior art.
The second purpose of the present invention is to provide the use of the non-medicinal donkey-hide gelatin nutrient solution as mentioned above aiming at the defects in the prior art.

a non-medicinal donkey-hide gelatin nutrient solution is prepared from the following medicinal and edible raw materials: 20-80 parts of donkey-hide gelatin, 0.5-2.5 parts of allium macrostemon, 0.1-2.0 parts of clove, 2-8 parts of medlar, 0.1-2.0 parts of fructus amomi, 3-10 parts of mulberry leaf, 0.5-3 parts of dried orange peel, 1-6 parts of honeysuckle, 2-10 parts of mint, 5-15 parts of Chinese date, 1-8 parts of rose and 1-8 parts of radix codonopsitis.
In the above non-medicinal donkey-hide gelatin nutrient solution, as a preferable scheme, the nutrient solution is prepared from the following medicinal and edible raw materials: 40-60 parts of donkey-hide gelatin, 1-2 parts of allium macrostemon, 0.4-1 part of clove, 4-6 parts of medlar, 0.5-1 part of fructus amomi, 5-8 parts of mulberry leaf, 1-2 parts of dried orange peel, 3-4 parts of honeysuckle, 5-7 parts of mint, 8-10 parts of Chinese date, 4-6 parts of rose and 4-6 parts of codonopsis pilosula.
In the above non-medicinal donkey-hide gelatin nutrient solution, as a preferable scheme, the nutrient solution is prepared from the following medicinal and edible raw materials: 50 parts of donkey-hide gelatin, 1.5 parts of allium macrostemon, 0.6 part of clove, 5 parts of medlar, 0.6 part of amomum fruit, 6 parts of mulberry leaf, 1.5 parts of dried orange peel, 3 parts of honeysuckle, 6 parts of mint, 9 parts of Chinese date, 5 parts of rose and 5 parts of codonopsis pilosula.
In the above non-medicinal donkey-hide gelatin nutritional liquid, as a preferred embodiment, the nutritional liquid further comprises common auxiliary raw materials, wherein the auxiliary raw materials are one or more of xylitol, maltitol, polydextrose, sorbitol, magnesium stearate, maltose, soybean oligosaccharide, lactose, lactitol, galactitol and fruitol.
In the above non-medicinal donkey-hide gelatin nutrient solution, as a preferred scheme, the preparation method of the donkey-hide gelatin nutrient solution is as follows: (1) selecting a ceramic frying pan, adding allium macrostemon, clove, medlar, fructus amomi, mulberry leaf, dried orange peel, honeysuckle, mint, Chinese date, rose and radix codonopsitis, and adding water to decoct to obtain a decoction; (2) grinding colla Corii Asini into powder, adding 3-5 times of water, dissolving, adding animal protein hydrolase 4-6% of colla Corii Asini, hydrolyzing at 55-60 deg.C and pH6.0-7.0 for 3-5 hr, and inactivating enzyme to obtain colla Corii Asini solution; (3) mixing the decoction obtained in step 1) and the donkey-hide gelatin liquid obtained in step 2) to obtain the donkey-hide gelatin oral liquid.
In the above non-medicinal donkey-hide gelatin nutrient solution, as a preferred scheme, the decoction method in the step 1) is that the decoction is carried out for 2 times, 8 times of water is added for the first time, and the decoction is carried out for 1.0 to 1.5 hours; adding 6 times of water for the second time, decocting for 0.5-1.0 hr, and mixing the decoctions.
In order to achieve the second object, the invention adopts the technical scheme that:
the donkey-hide gelatin nutrient solution is applied to preparing anti-fatigue food or health care products.
The invention has the advantages that:
1. the invention solves the problem that the donkey-hide gelatin needs to be processed by oneself when being taken, and the nutrient solution is convenient to carry, can be taken at any time and is easy to store.
2. The non-medicinal donkey-hide gelatin nutrient solution is prepared from medicinal and edible raw materials, is safe, non-toxic and free of side effects, and is beneficial to improving the nutrient level of organisms and improving the physical quality after being taken for a long time.
3. The preparation method of the non-medicinal donkey-hide gelatin nutrient solution is simple, the mixture ratio of the raw materials is scientific and reasonable, and the effect of the nutrient solution is ensured.
4. The experimental result shows that the non-medicinal donkey-hide gelatin nutrient solution can obviously improve the exhaustion swimming time ratio, the liver glycogen storage and the lactate dehydrogenase activity of the mice with qi-deficiency constitution, and the non-medicinal donkey-hide gelatin nutrient solution has obvious anti-fatigue effect.

The animal protein hydrolase may be commercially available product, such as animal protein hydrolysis complex enzyme from Nanning Pont bioengineering, Inc., which is high-efficiency proteolytic complex enzyme mainly composed of endoprotease, exonuclease and flavor enzyme.
Example 11 anti-fatigue effect experiment of donkey-hide gelatin nutrient solution on qi-deficiency constitution mice
1 laboratory animal
SPF grade healthy 4 week old male ICR mice, 60, weight 18-22g, were provided by Shanghai Jiesei laboratory animals, Inc. Mice were acclimatized for one week before the experiment, with a temperature stabilized at about 22 degrees, a relative humidity of 70%, and an illumination time of 12 hours from 8 am to 8 am. All mice are raised in boxes, 6 mice are fed in each box, water is freely drunk, standard feed is freely taken, and the mice are subjected to adaptive swimming exercise for 3 days and 10min/d, the water depth is 50cm, and the water temperature is 25 +/-1 ℃ before the experiment is started.
2 method of experiment
2.1 Molding method
The mice are subjected to negative 6% body weight exhaustive swimming (the tail root of the mice is loaded with 6% body weight and swims to be exhausted in water with the water depth of 50cm and the temperature of 25 ℃, the exhaustion standard is that the mice can not return to the water surface after sinking for 10s at last in swimming), the initial exhaustion swimming time is recorded, the mice with large difference of exhaustion swimming time are removed, and the mice are fasted for 24 h. Weighing the mice with larger weight difference on the day of the beginning of the experiment, and screening 45 male mice meeting the experiment requirements. The molding method comprises the following steps: the mice are placed in a 60 x 80cm swimming box at 9:00 a.m., a cubic platform with the side length of 5cm is placed in the box (the platform can only accommodate one mouse to stand on each platform, the distance between the box wall and each platform is not less than 4cm), water is added into the box to submerge the platform and keep the platform 0.8cm away from the water surface, the mice can selectively swim or stand on the platform, the mice cannot sleep normally due to the fact that the mice are out of water, and partial sleep deprivation is carried out. Taking the swimming pool out of the swimming box on the day 21:00 and then feeding the swimming pool regularly. After continuous molding for 6 days, the molding time is reduced from 12h/d to 8h/d, and the total training time is 9 d.
2.2 methods of administration
The male mice meeting the experimental requirements are randomly divided into five groups, namely a normal control group, a model group, an experimental group and an experimental group. Grinding colla Corii Asini into powder, adding 5 times of warm water, dissolving, adding animal protein hydrolase 5% of colla Corii Asini, hydrolyzing at 55-60 deg.C and pH6.0-7.0 for 3-5 hr, and inactivating enzyme to obtain colla Corii Asini solution; mixing colla Corii Asini solution and the decoction to obtain colla Corii Asini nutritional liquid. Grinding colla Corii Asini into powder, adding 5 times of warm water, dissolving, adding animal protein hydrolase 5% of colla Corii Asini, hydrolyzing at 55-60 deg.C and pH6.0-7.0 for 3-5 hr, and inactivating enzyme to obtain colla Corii Asini solution; mixing colla Corii Asini solution and the decoction to obtain colla Corii Asini nutritional liquid. The dosage is 1mg per 100g body mass (raw material), the intragastric volume is 1mL, and the continuous 12 days.
2.3 index determination
2.3.1 determination of anti-fatigue index
The exhaustive swimming time of the mice is measured the next day after the end of the intervention, and the measuring method comprises the following steps: the tail root of the mouse is loaded with 6 percent of body weight, and the mouse swims in water with the water depth of 50cm and the temperature of 25 ℃ until exhaustion, wherein the exhaustion standard is that the mouse can not return to the water surface after finally sinking for 10s in swimming. Recording the exhaustive swimming time of the mouse, and judging whether the mouse is in a fatigue state according to the exhaustive swimming time ratio P. The exhaustion swimming time ratio P is the exhaustion swimming time of the experimental mouse/the exhaustion swimming time of the normal control group mouse.
2.3.2 determination of liver glycogen
Muscle of mouse liver was used for determination of hepatic glycogen, and mouse hepatic glycogen detection kit of Yaji Biotech Ltd, Shanghai was used for detection.
2.3.3 determination of the lactate dehydrogenase Activity in mice
The activity of the serum lactate dehydrogenase of each group of mice is detected by adopting a lactate dehydrogenase test box of Nanjing institute of bioengineering.
3 results
3.1 Effect of donkey-hide gelatin nutrient solution on exhaustion swimming time ratio of qi-deficiency constitution mice
The results are shown in table 1, and the first exhaustive swimming time ratio P1 of each group has no significant difference; the ratio P2 of the last exhausted swimming time of the model control group is obviously reduced compared with the normal control group (P is less than 0.01), which indicates that the model building is successful, and the mouse is in a fatigue state after the model building; the ratio of the last swimming time spent in the experimental group P2 is obviously increased compared with the normal control group and the model control group (P < 0.01); the ratio of the final swimming time spent in the two experimental groups and the three experimental groups P2 is obviously increased compared with the model control group (P <0.05), and the ratio of the final swimming time spent in the two experimental groups and the three experimental groups P2 has no obvious difference compared with the normal control group (P > 0.05). The experimental results show that each experimental group has certain anti-fatigue activity, wherein the anti-fatigue effect of the experimental group is most obvious.

3.2 Effect of donkey-hide gelatin nutrient solution on liver glycogen of mice with qi-deficiency constitution
The results are shown in table 2, the liver glycogen content of the model group is obviously reduced compared with that of the normal control group (P is less than 0.01), which indicates that the model building is successful, and the mice are in a fatigue state after the model building; the liver glycogen content of an experimental group is obviously increased compared with that of a normal control group and a model control group (P < 0.01); the liver glycogen content of the two experimental groups and the three experimental groups is obviously increased compared with that of a model control group (P <0.05), and the liver glycogen content of the two experimental groups and the three experimental groups is not obviously different compared with that of a normal control group (P > 0.05). The experimental results show that all experimental groups can effectively improve the liver glycogen storage level of the mice with qi-deficiency constitution, wherein the two experimental groups and the three experimental groups can obviously improve the liver glycogen storage to a normal level, and the one experimental group can obviously improve the normal value of the liver glycogen storage of the mice with qi-deficiency constitution.

3.3 Effect of donkey-hide gelatin nutrient solution on LDH activity of serum of mice with qi-deficiency constitution

The experimental results show that all experimental groups can effectively improve the LDH activity of the serum of the mice with qi-deficiency constitution, wherein two experimental groups and three experimental groups can obviously improve the LDH activity of the serum of the mice to a normal level, and one experimental group can obviously improve the normal value of the LDH activity of the serum of the mice with qi-deficiency constitution.
