背景技术
皮肤是身体中最大的组织,其功能是保护身体免受阳光、物理和化学刺激。其对于维持人类生命是绝对必要的,并且不断地再生以维持体内平衡。皮肤由外侧依次由表皮、真皮和皮下脂肪组成。表皮是皮肤最薄的组织,起着保湿和保护皮肤的重要作用,并且有助于防止水分流失、损伤、细菌入侵等。皮肤是一种围绕整个身体的组织,具有各种各样的功能。因为皮肤在身体的内部和外部之间具有屏障功能,其具有胰岛的免疫系统和表皮的湿度控制功能。此外,皮肤在适当的阳光下进行维生素D的合成,并且还可以排泄到脂质储库和汗腺中。由健康皮肤中的正常分化过程产生的角质层具有维持皮肤保湿和保护皮肤免受外部环境刺激的功能。这种功能被称为皮肤屏障功能,它可能是表皮最重要的作用。

根据本发明的优选实施例方式,可以通过将氨基基团连接至肽的C端来诱导修饰。
上述氨基酸修饰用于显着改善本发明肽的稳定性。如本文所用,术语“稳定性”不仅指体内稳定性,还指储存稳定性(例如室温下的储存稳定性)。上述保护基团用于保护本发明的肽免受体内蛋白质裂解酶的攻击。
根据本发明的优选实施例方式,本发明的肽对人源细胞没有表现出毒性,因此作为皮肤美白肽非常有用。根据本发明,当用浓度为100ng/mL至1,000μg/mL的凝血酸-肽处理HaCat细胞和NIH3T3细胞时,没有测量到显着的细胞毒性,并且即使通过肉眼检查这些细胞的形状,也未观察到显着变化(图6)。
根据本发明,即使在50℃的温度下,凝血酸-肽也显示出优异的热稳定性,并且即使在日光条件下也证实其是稳定的(图3)。肽可以有利地应用于需要长期储存的产品,例如药物、医药部外品和化妆品。
根据本发明的优选实施例方式,本发明的肽具有抑制黑色素生成的能力。根据本发明,当黑素细胞用-MSH处理以诱导黑色素生成,用本发明的凝血酸-肽处理,用本发明的凝血酸-肽处理,培养,并比较黑色素生成量,与对照组相比,黑色素生成最多被抑制到约50%的水平(表1)。
根据本发明的优选实施例方式,本发明的肽依据浓度增加黑色素生成。根据本发明,当细胞用用各种浓度(1μ、10μ、100μ和250μ)的凝血酸-肽(NA-PS)和未结合凝血酸的肽处理细胞,培养3天,并通过肉眼检查确认这些细胞中的黑色素生成量时,确认黑色素生成量依据浓度降低(图4)。另外,当细胞用各种浓度(1μ、10μ、100μ和250μ)的4种凝血酸-肽、未结合凝血酸的肽、熊果苷和AA2G处理,培养3天,并在490nm处测量培养物的吸光度并进行分析。结果证实,与用凝血酸-肽处理的细胞相比,用未结合凝血酸的肽处理的细胞中黑色素生成量显着降低(图1)。
根据本发明的优选实施例方式,本发明的肽抑制酪氨酸酶的活性。根据本发明,当L-酪氨酸、肽或凝血酸-肽与酪氨酸酶反应,并通过测量475nm处的吸光度确认对酪氨酸酶的抑制率,证实与用未结合凝血酸的肽和熊果苷的处理相比,用凝血酸-肽的处理显示出对酪氨酸酶活性的显着抑制作用(图5)。
根据本发明的优选实施例方式,本发明的肽抑制小眼相关转录因子(MITF)的表达。黑色素通过一系列酶促反应生成(Olivares等,色素细胞和黑色素瘤研究(PigmentCell Melanoma Res)2222(6):750-760(2009)),黑色素生成的主要机制是细胞增殖和酪氨酸酶活性的增加。黑色素生成中最重要的酶是酪氨酸酶,酪氨酸酶是催化黑色素生物合成早期的速率控制因子,酪氨酸通过酪氨酸酶转化为3,4-二羟基苯丙氨酸(DOPA)和多巴醌,从而合成了基于红色的黑色素(褐黑素)和基于棕色的真黑素(Lopezet等,生物化学期刊(JBiol Chem)267:381-390(1992))。在真黑素形成中另外两种重要的酶是TRP-1和TRP-2。另外,小眼相关转录因子(MITF)是负责酪氨酸酶和相关酶(TRP)转录的重要元素,并且一旦MITF的表达被抑制,黑色素的生成就会被抑制。
根据本发明,黑素细胞用各种浓度(1μ,10μ和50μ)的-MSH和肽(PS,VS),或-MSH和凝血酸-肽(Tranexamyl-AP、Tranexamyl-HK、Tranexamyl-VS、Tranexamyl-PS)处理,培养4天,从细胞中获得mRNA并进行RT-PCR。结果证实,用凝血酸-肽的处理抑制了与黑色素生成相关的基因(MITF)的表达(图7)。这些结果表明,本发明的肽抑制与黑色素生成有关的基因的表达,从而具有显着优异的皮肤美白效果。
另外,就含有本发明肽的组合物而言,它可以非常有效地用于使用各种制剂的皮肤美白。
为了证实凝血酸-肽的皮肤美白效果,制备含有凝血酸-肽的乳霜并进行临床试验。结果证实,在临床条件下与纯凝血酸或对照组相比,凝血酸-肽具有优异的美白效果(图9)。另外,在通过将凝血酸-肽的含量降低一半进行的实验中观察到相同的结果,这表明即使当凝血酸-肽的含量降低时也可以保持美白效果(图10)。
另外,本发明的第二方面提供一种皮肤美白组合物,其含有选自由以下项组成的组中的凝血酸-肽:Tranexamil-AS、Tranexamil-AT、Tranexamil-AP、Tranexamil-GP、Tranexamil-ES、Tranexamil-KK、Tranexamil-HK、Tranexamil-MY、Tranexamil-GH、Tranexamil-MA、Tranexamil-AH、Tranexamil-CC、Tranexamil-SA、Tranexamil-WA、Tranexamil-WE、Tranexamil-KD、Tranexamil-NA、Tranexamil-TS、Tranexamil-SS、Tranexamil-EC、Tranexamil-TA、Tranexamil-PF、Tranexamil-VS、Tranexamil-VV、Tranexamil-VP、Tranexamil-AA、Tranexamil-PS、Tranexamil-HA、Tranexamil-GK、Tranexamil-KV、Tranexamil-AR、Tranexamil-RP、Tranexamil-PQ、Tranexamil-QG、Tranexamil-PP、Tranexamil-VR、Tranexamil-SV、Tranexamil-ET、Tranexamil-CG和Tranexamil-NT。
本发明的组合物可以制备为化妆品组合物。
在本发明的组合物中用作活性成分的凝血酸-肽由2至6个氨基酸残基组成,具有非常小的分子量,因此具有优异的皮肤渗透性。因此,当将本发明的组合物局部施用于皮肤时,由于其高皮肤渗透率可以实现皮肤美白效果。本发明的组合物抑制黑色素的产生,因此它使肤色变亮,保持肤色恒定,并且有效地消除皮肤色素和消除老年斑。本发明的凝血酸-肽通过几种方法抑制角质形成细胞中黑色素的产生,并通过防止或减弱角质形成细胞中产生的黑色素的释放,显示出增亮角质层颜色的效果。
含有本发明的用作活性成分的凝血酸-肽的组合物可以应用于凝胶型、皮肤型、乳霜型、软膏型等药物组合物,以及化妆品组合物,但是适用的类型不限于此。上述组合物可以通过已知方法,通过添加常规的软化剂、乳化剂、增稠剂或本领域已知的其他物质来适当地制备。
凝胶型组合物可以通过添加软化剂(例如,三羟甲基丙烷、聚乙二醇或甘油等),溶剂(例如,丙二醇、乙醇、异鲸蜡醇等),纯净水等来制备。
皮肤型组合物可以通过添加脂肪醇(例如,硬脂醇、肉豆蔻醇、山嵛醇、花生醇、异硬脂醇、异鲸蜡醇等)、丁二醇、甘油、尿囊素、对羟基苯甲酸甲酯、依地酸二钠、黄原胶、聚二甲基硅氧烷,聚乙二醇-60氢化蓖麻油、聚山梨醇酯60、蒸馏水等来制备。
乳霜型组合物可以通过添加脂肪醇(例如,硬脂醇、肉豆蔻醇、山嵛醇、花生醇、异硬脂醇、异鲸蜡醇等)、脂质(例如,卵磷脂、磷脂酰胆碱、磷脂酰乙醇胺、磷脂酰丝氨酸、磷脂酰肌醇等)、其衍生物、乳化剂(例如,硬脂酸甘油酯、去水山梨醇棕榈酸酯、去水山梨醇硬脂酸酯等)、天然脂肪或油(例如,鳄梨油、杏仁油、巴巴苏仁油、琉璃苣油、山茶油等)、溶剂(例如,丙二醇等)、蒸馏水等来制备。
软膏型组合物可以通过添加软化剂、乳化剂和蜡(例如,微晶蜡、石蜡、地蜡、蜂蜡、鲸蜡、凡士林等)来制备。
此外,本发明还可以作为含有一种或多种药学上可接受的载体或赋形剂的制剂和用作活性成分的组合物提供。
因此,根据需要,制剂可包括药学上可接受的载体、稀释剂、赋形剂或其组合。这些试剂有助于将活性成分给予生物体。
在下文中,将参考以下实例进一步详细描述本发明。然而,以下实例旨在说明本发明,并且本发明的范围不限于此。
实施例
实施例1:肽库的合成
为了合成肽库,将连接有nsc-氨基酸(nsc-Ala,nsc-Arg(pbf),nsc-Asp(OtBu),nsc-Asn(trt),nsc-Gly,nsc-Glu(OtBu),nsc-Gln(trt),nsc-His(trt),nsc-Ser(tBu),nsc-Thr(tBu),nsc-Tyr(tBu),nsc-Trp(Boc),nsc-Leu,nsc-Ile,nsc-Val,nsc-Phe,nsc-Met,nsc-Lys(Boc),nsc-Pro)的氯化物树脂的氯三苯甲基(CTL树脂,Novabiochem目录号01-64-0021)加入到每个系列的19行96孔Teflon反应器的每一个中,向其中加入1mL二氯甲烷(MC),搅拌3分钟。除去溶液后,加入1mL二甲基甲酰胺(DM),搅拌3分钟后,再次除去溶剂。将制备的凝血酸-肽基树脂用DMF、MC和甲醇洗涤3次,通过减慢流动的氮气进行干燥,然后在P205下减压真空干燥。将30mL混合溶液[包括三氟乙酸(TFA)81.5%、蒸馏水5%、硫代苯甲醚5%、苯酚5%、EDT(1,2-乙二硫醇)2.5%和三异丙基硅烷(TIS)1%]加入到制备的树脂中,并将反应物在冰浴中保持1小时,同时在室温下间歇振荡。过滤树脂,用少量TFA溶液洗涤,并与母液合并。然后,获得凝血酸-肽。
作为合成和纯化的结果,获得了40种与相互不同序列的肽结合的凝血酸-肽(表1)。由于肽序列的差异,合成的凝血酸-肽的产率因物理性质的差异而不同,并且平均产率为约10%。在柱分离期间,尽管洗脱时间不同,但在8分钟的时间区域发现峰。
实施例2:凝血酸-肽的筛选
在加入了10%胎牛血清(FBS,Sigma)的杜氏改良(Dulbecco's modified)Eagle培养基(DMEM,Sigma)中37℃的CO2条件下培养B16F1细胞(小鼠黑素细胞)。将细胞在24孔板中以1×105个细胞/孔的浓度培养,确认细胞附着。然后,对照组未用任何物质处理,只向其中加入溶剂,而将α-MSH(20μg/mL)加入阳性对照组。其余培养皿用α-MSH(20μg/mL)、40种凝血酸-肽处理而对照组用熊果苷和AA2G处理,直到浓度分别为1μg/mL、10μg/mL、100μg/mL和250μg/mL。将试验物质加入每个培养皿中并培养3天。3天后,通过离心除去培养物,使细胞裂解并收集细胞内黑色素,并在490nm处观察以测量细胞内黑色素的生成量。
根据浓度处理40种凝血酸-肽,并测量它们对黑色素生成的抑制作用。与其中凝血酸未与肽结合的对照组相比,证实在用结合有凝血酸的肽处理的组中黑色素的量迅速减少。此外,就4种对α-MSH(%)具有优异抑制作用的合成的凝血酸-肽(Tranexamil-AP、-HK、-VS和-PS)而言,这些凝血酸-肽显示为优于具有美白功能的现有物质熊果苷和AA2G(图2)。从这些结果可以认为,当应用诱导皮肤中黑色素生成的因子时,合成的凝血酸-二肽可以抑制它们的活性,从而使肤色变亮。
通过筛选显示出良好滴度的候选肽被分类为单独的一组并且大规模制备并进行实验。表1显示了凝血酸-肽的筛选结果。

实施例3:证实凝血酸-肽稳定性的实验
为了证实制备的肽的热稳定性和光稳定性,将HPLC纯化的至少98%纯度的高纯度的Tranexamyl-AP、Tranexamyl-HK、Tranexamyl-VS和Tranexamyl-PS溶解于50mM Tris-HCl(pH 8.0)缓冲液至浓度为100μg/mL,等分到玻璃瓶中,并在50℃下保存7、14、21、28、35、42和60天,测量热量导致的肽损失,并使用相同的方法在日光条件下测量稳定性(图3)。热量导致的肽损失和阳光条件下稳定性的测量结果为,仅显示出少于10%的肽损失,因此证实这些凝血酸-肽具有高抗热稳定性和储存期稳定性。另外,比较稳定性实验的结果显示,与未结合凝血酸的肽相比,凝血酸-二肽的含量平均高出约20%。因此,与未结合凝血酸的肽相比,凝血酸-肽在热和阳光下更稳定。
实施例4:凝血酸-肽的美白效果的证实
根据浓度处理凝血酸-肽,并测量它们对黑色素生成的抑制作用。在37℃的CO2条件下,在含有10%胎牛血清(BSA,Sigma)的杜氏改良Eagle培养基(DMEM,Sigma)中培养小鼠黑素细胞。
将细胞在24孔板中以1×105个细胞/孔的浓度培养,确认细胞附着。然后,对照组未用任何物质处理,只向其中加入溶剂,而将α-MSH(20μ)加入到阳性对照组中。其余培养皿用α-MSH(20μg/mL)、4种凝血酸-肽处理而对照组用熊果苷和AA2G处理,直到浓度分别为1μg/mL、10μg/mL、100μg/mL和250μ。将试验物质加入每个培养皿中并培养3天。3天后,通过离心除去培养物,通过肉眼检查确认细胞内黑色素的生成量。
图4显示了生成的黑素细胞的图像,其证实了根据凝血酸-肽和VS肽的每种浓度的黑色素生成的减少速率。从图像中可以看出,与用α-MSH处理的细胞相比,细胞数没有相对减少,而在相似浓度处理熊果苷等的情况下,细胞数迅速减少。从与细胞量相比黑色素生成量减少的结果发现,凝血酸-肽具有抑制黑色素生成的作用。
实施例5:通过凝血酸-肽降低的酪氨酸酶活性
酪氨酸酶是一种已知参与黑色素生物合成的酶,在从L-酪氨酸开始的黑色素生物合成过程中起催化剂的作用,并在从酪氨酸到3,4-二羟基-L-苯丙氨酸(DOPA)和从DOPA到多巴醌的氧化过程中起催化剂的作用。由于在这两个氧化反应之后的后续反应自发发生,所以涉及酪氨酸酶的反应决定了整体反应速率。因此,酪氨酸酶抑制活性是评价美白活性的一个非常重要的因素。在该实验中,比较了合成的凝血酸-二肽的体外酪氨酸酶抑制活性(图5)。
将作为底物的L-酪氨酸溶解在磷酸盐缓冲液(0.05M,pH 6.8)中至1.5mM的浓度,将所得溶液(0.01mL)加入0.3mL分光光度计比色杯中,并向其中加入0.01mL浓度为0.06mM的L-DOPA(辅因子)。向其中加入本发明的肽,并加入磷酸盐缓冲溶液至0.1mL。通过向其中加入0.1mL酶溶液进行反应,酶溶液是酪氨酸酶以60U/mL溶解在磷酸盐缓冲液中。另外,以100ppm的量加入熊果苷、凝血酸-肽和未与凝血酸结合的肽中的每一种,以进行对比实验。特别地,作为对照组(空白),仅添加0.1mL缓冲液代替酪氨酸酶。反应在37℃下进行10分钟,用分光光度计(Beckman DU-7500)在475nm处测量吸光度,得到酪氨酸酶抑制率(图5)。作为实验结果,与AA2G或熊果苷相比,4种凝血酸-肽显示出相对较高的酪氨酸酶抑制作用,而未结合凝血酸的肽没有良好的抑制作用。
实施例6:细胞毒性实验
为了确认本发明的肽是否在角质形成细胞中具有细胞毒性,使用MTT测定法评估细胞毒性。将HACAT细胞和3T3细胞以4×104个细胞/24孔的浓度接种到24孔板的每个孔中,并在37℃的CO2条件下恒温培养。将培养物用PBS洗涤两次,培养,用各种浓度(1mg/mL至1μg/mL)的合成的凝血酸-二肽处理,并在恒温下培养24小时。培养后,将DPBS中0.5%的MTT与培养基以1:9(v/v)的比例混合并加入其中,并在CO2培养箱中培养2小时。将产生的甲臜溶解在DMSO中,并通过ELISA在570nm处测量其吸光度。从图6中可以看出,对于HACAT细胞(一种角质形成细胞)和3T3细胞(一种成纤维细胞),未观察到以低浓度至高浓度施用的肽的细胞数减少,即使在显微镜下观察这些细胞也未观察到显着变化。
从以上结果可以确认,凝血酸-肽对皮肤相关细胞没有细胞毒性,因此可以预期本发明的这些肽可以安全地施用于皮肤而不会引起任何严重的影响。
实施例7:黑色素生成标记物的抑制
为了更清楚地验证凝血酸-肽在小鼠黑素瘤细胞中通过-MSH处理诱导黑色素生成的美白活性,通过实时逆转录聚合酶链反应(qRT-PCR)证实了小眼相关转录因子(MITF)的RNA生成。
首先,将实施例2中培养的B16F10细胞分别用浓度为0.1%和0.05%的两种凝血酸-肽(TXA-VS和TXA-PS)处理。然后,对于实时PCR测量,提取RNA样品如下。
使用intron RNA提取试剂盒(Intron公司,韩国)进行提取。具体实验方法如下。
(1)在1.5mL试管中加入1×106个细胞
(2)通过13,000rpm离心10秒钟除去培养基,并且
(3)加入1mL裂解缓冲液。
(4)剧烈进行涡旋。
(5)加入200μL氯仿并进行涡旋。
(6)在4℃下以13,000rpm离心10分钟。将400μL转移到空试管中。
(7)加入400μL结合缓冲液以及混合物,平稳移液2或3次
(8)放在柱子上。
(9)以13,000rpm离心30秒。
(10)用700μL缓冲液A洗涤
(11)盖上试管盖,以13,000rpm离心30秒。
(12)用缓冲液B重复10到11次。
(13)以13,000rpm干燥1至2分钟。
(14)向柱中加入50μL洗脱缓冲液并在室温下培养1分钟。
(15)以13,000rpm离心1分钟。
使用提取的RNA进行实时PCR以确认MITF的表达量,实验中使用的试剂和条件如下。

实验实施例:临床试验
从实施例2中对黑色素合成显示出高抑制作用的4种凝血酸-二肽中,选择了两种(VS和PS),从而制备了含有1%凝血酸-二肽的两种乳霜,使其具有与下面实施例10中相同的组成。为了证实这些肽的皮肤美白效果,收集了54名皮肤色素沉着但没有器质性疾病的女性。特别地,将含有凝血酸的乳霜作为阳性对照组,标准基础乳霜作为阴性对照组。
将每个受试者的面部垂直分成两部分,并将上面制备的两种乳霜施用于每个面部。要求受试者在早晨和晚上洗脸后使用乳霜,每天2次,连续8周。
为了客观评价使用乳霜前后皮肤色素沉着的变化,在使用乳霜前和使用乳霜2周、4周、6周和8周后,使用Mexameter(CK,德国)测定色素沉着的变化。测量结果如图9所示。
如图9所示,证实含有两种凝血酸-二肽的乳霜与仅含有凝血酸的阴性对照组和阳性对照组相比都具有更大的美白效果。此外,在回顾了黑色素指数的每周变化率后,证实在8周后,使用凝血酸-二肽(VS)的第1组显示减少22.9%,使用凝血酸-二肽(PS)的第2组显示减少183.2%,凝血酸组显示减少11.4%,使用基础乳霜的组显示减少6.6%(图9)。
上述实验的结果是,在两种凝血酸-二肽之间选择具有更大作用的凝血酸-二肽,并且为了降低乳霜中的含量,制备了含有0.5%凝血酸-二肽的乳霜。收集10名女性受试者,并以与上述相同的方式进行8周的临床实验。
如图10所示,证实含有0.5%凝血酸-二肽的乳霜与仅含有凝血酸的乳霜和基础乳霜相比具有更大的美白效果。此外,在回顾了黑色素指数的每周变化率后,证实在8周后,使用含有0.5%凝血酸-二肽(VS)的第1组显示减少23.9%,使用凝血酸的组显示减少10.2%,使用基础乳霜的组显示减少4.7%。

Claims (8)
1.一种具有皮肤美白活性的凝血酸-肽,其中所述凝血酸-肽是由凝血酸和肽通过N端相连来制备的,所述肽选自由以下项组成的组:AP、HK、VS和PS。
2.根据权利要求1所述的凝血酸-肽,其中所述凝血酸-肽抑制黑色素合成。
3.根据权利要求1所述的凝血酸-肽,其中所述凝血酸-肽抑制酪氨酸酶的活性。
4.根据权利要求1所述的凝血酸-肽,其中所述凝血酸-肽抑制小眼相关转录因子(MITF)的表达。
5.根据权利要求1所述的凝血酸-肽,其中所述凝血酸-肽在0.01%至5.0%的浓度下具有皮肤美白效果。
6.根据权利要求1所述的凝血酸-肽,其中所述凝血酸-肽通过其N端的凝血酸增加稳定性。
7.一种用于皮肤美白的组合物,包含根据权利要求1所述的凝血酸-肽。
8.根据权利要求7所述的组合物,其中所述组合物是化妆品组合物。
