Abstract
一种治疗肝肿瘤的中药组合物,其特征在于,包括下列重量份配比的原料药:蔓菁15‑40份、五灵脂5‑30份、石菖蒲5‑30份、西红花1‑20份、诃子10‑40份、长松萝1‑25份、花椒0.5‑5份、白芥子5‑32份、棘豆1‑30份,以及选自冬虫夏草1‑15份、榼藤子1‑20份、人工麝香0.1‑5份,红景天1‑30份、大黄1‑20份、天胡荽1‑10份中的至少一种组成。该中药组合物可以有效的抑制肝癌病人肿瘤的生长改善患者的生存质量。

技术领域:
本发明涉及一种治疗肝肿瘤的中药制剂,属于中药技术领域。
背景技术
目前,肝肿瘤是危害人体健康的一种高发性疾病,尤以亚洲地区发病率高,在我国占各种恶性肿瘤的第三位或第四位,男性发病率高于女性;肝肿瘤属于中医学“症瘕积聚”、“肝积”、“臌胀”等范畴;本病病因与气滞血瘀,脾虚聚湿,热毒内蕴有关;现代医学认为,本病与脏腑功能障碍,代谢紊乱,免疫作用低下,以及遗传因素有关;患者由于患病后腹肿肝大,胁痛腹胀,胸闷易怒,不思饮食,食少乏力,导致身体消瘦,身体免疫力下降;现有治疗方法为手术切除,化疗,或口服“斑蝥胶囊”等药物,治疗效果不尽理想。
在当前我国诊疗水平和条件下,大多数恶性肿瘤患者确诊已属中、晚期病例,且伴随肿瘤的转移,目前的主要治疗手段为化疗、放疗、手术,但不是所有的患者病情都能适合上述三种手段,同时上述手段尤其化放疗毒副作用明显;这种现状促使人们积极寻找来自天然的植物药物,寻求药效明确但毒副作用相对少的新途径或药物,致使患者在不损伤机体正气即免疫功能状态下,尽量消除局部的癌灶,起到治疗肿瘤的作用,延长患者的生存时间。
本发明是采用中药材为原料,按照传统医药理论形成的中药组方,结合现代的制剂工艺制成中药含片。
藏医学认为,人体是由“隆、赤巴、培根”三大基础组成。一切疾病的根源都在于“隆、赤巴、培根”的失衡和紊乱。正常情况下,隆居于骨骼;赤巴居于血液、汗液;培根居于除骨骼、血液、汗液之外的其他部位,即精华、肌肉、脂肪、骨髓、精液、大便、尿等处。藏医认为正常状态下,所摄食物供养“初液”、初液营养血液、血液营养软组织肉、肉营养脂肪组织、脂肪营养骨组织、骨营养骨髓。肿瘤的形成是由于三者之间的紊乱致使所摄食物未能化为“初液”,而化作“恶液”并经血管进入全身各组织,并在某一免疫力功能衰弱的部位,被隆的结构素晃、硬、细、冷、轻、粗的逆作用收缩聚积为坚硬难消的肿块。这种似青扛果核般的肿块藏医称之为“滞癌”,从字面上可翻译为“核”,此即现代医学称之为肿瘤的疾病。
患者脉象弦细、力小而微,一般左右上下之脉除了微现外,不明显为芤脉,痞瘤在各个腑脏哪一处的具体脉象不明显而是成一片的跳动;尿液中出现状如鱼目的痞瘤之点滴;此前在上下中出现不同的颜色,有痞瘤的部位白色时为培根,黄色时为赤巴,红色时为血等;身体的哪部分患有痞瘤,该处的上面稍积污垢。
发明内容
本发明提供了一种由传统草药组成的复方口含片。该制剂可以有效的抑制肝癌病人肿瘤的生长改善患者的生存质量。
一种中药组合物,包括下列重量份配比的原料药蔓菁15-40份、五灵脂5-30份、石菖蒲5-30份、西红花1-20份、诃子10-40份、长松萝1-25份、花椒0.5-5份、白芥子5-32份、棘豆1-30份,以及选自冬虫夏草1-15份、榼藤子1-20份、人工麝香0.1-5份,红景天1-30份、大黄1-20份、天胡荽1-10份中的至少一种组成。

本发明还进一步提供一种中药组合物,该中药组合物包括A药物和B药物,优选由A药物和B药物组成,其中A药物由下列重量份配比的中药原料制成:蔓菁15-40份、五灵脂5-30份、石菖蒲5-30份、西红花1-20份、诃子10-40份、长松萝1-25份、花椒0.5-5份、白芥子5-32份、棘豆1-30份、榼藤子1-20份、人工麝香0.1-5份。进一步优选蔓菁15-30份、五灵脂5-20份、石菖蒲5-25份、西红花1-15份、诃子15-38份、长松萝1-18份、花椒0.5-3份、白芥子5-25份、棘豆1-20份、榼藤子1-15份、人工麝香0.1-3份。
其中B药物选自陈皮、清半夏、半枝莲、白术、党参、甘草、沙参、五味子、百合中的一种或多种。
所述肿瘤为肝癌。
方选西红花、诃子、甘草、蔓菁、螃蟹甲、红景天、人工麝香为大臣药。红花为众药之本,治一切热症之药。诃子有十七效可治二十属性病。红花、诃子合用,可增进患者体力,同时协助国王药和后妃药加强治疗作用。方选螃蟹甲治肿核疮,并治肺病喉咙干,性平。方选甘草性平,化味凉,能清一切肺热。方选红景天血清肺、止咳退烧、止痛,用于治疗肺炎。方选芳香开窍、通经络、消肿止痛等作用。方中螃蟹甲、甘草、红景天、人工麝香皆有辅助国王药治疗肺病的功效,同时兼具消肿解毒,调节培根、赤巴与隆的平衡之功效。方选蔓菁味辛,性温,治培根病、隆病,生赤巴,治疗隆病调节赤巴,平衡赤巴与隆。
方选大黄、藏菖蒲、花椒、冬虫夏草、灵芝为佐使之药。大黄有泻毒热腑热。藏菖蒲功效化食,祛隆,提升胃阳,并且治未消化症。四位药起引方中诸药以达病灶并调和诸药作用。
诸药配伍,以主治赤巴、赤巴隆合并症为主,具有清肺热之功效,兼并调节赤巴、隆与培根之间的平衡,使初液化为营养进入各脏腑、组织及血液、恶液不生,固本培元,消肿解毒。
为强化本中药组合物其中一个方面的功效,可选择陈皮、清半夏、半枝莲加入上述处方中。陈皮用于中焦寒湿脾胃气滞者,脘腹胀痛、恶心呕吐、泄泻等症状。半夏性温,味辛,归脾经、胃经、肺经,燥湿化痰、降逆止呕、消痞散结。属清热药下属分类的清热燥湿药。半枝莲味辛、苦,性寒。归肺经、肝经、肾经,清热解毒、活血化瘀、利尿。属清热药下属中的清热解毒药。三味药改善患者的痰多咳重,胸闷气短、纳呆便溏、神疲乏力等症状。根据患者的情况可独自添加或组合加入。
为强化本药方对脾、肺方面的功效,处方中还可增加包括白术、党参、甘草。三味药物可针对改善久咳痰稀、胸闷气短、纳呆食少、腹胀便溏患者的症状进行有效的改善。根据患者的情况可独自添加或组合加入。
同样的为强化本药方在清肺热方面的功效,处方中还可以增加包括沙参、五味子、百合。三味药物针对干咳无痰或痰少而粘、气短胸疼、潮热盗汗等患者症状进行有效改善根据患者的情况可独自添加或组合加入。
另一方面,运用本发明提供的中药组合物,添加合适的药学上可接受的辅料,还可以制备能够治疗白血病的中药制剂,其方法可以是任意常规的中药制剂生产方法,生产任何常规的剂型。考虑到患者的顺应性和药效,所述制剂优选为口服剂型,更优选为口含片、散剂、胶囊剂或口服液,更优选为口含片、散剂、口服液,更优选为口含片、散剂。
制剂的方法可以是:
1)各原料经挑杂后,称取,分组放好备用;
2)各组原料切段,分别研磨成粉,过2号标准筛;
3)将粉末混合,即得所述药物活性成分组合物;
4)将3)得到的活性成分组合物,依据所制剂型的不同添加药用辅料,按标准制剂工序进行加工制成制剂。
例如,将提供的中药组合物制备成散剂的方法为:使用提供的中药组合物,将各原料药切段、研磨成粉,混匀即得散剂。使用时,将所述散剂按用量标准取药,置于舌下含服,约5-10分钟后吞咽,在吞咽后30分钟方可饮水。
再例如,将提供的中药组合物制备成口服液的方法:将提供的组合物经水煎煮,之后浓缩水煎液,再添加药学上可接受的矫味剂、掩味剂、稀释剂,如蔗糖、炼蜜等与之搭配,制成口服液,经灭菌后灌装成瓶。
再例如,将提供的中药组合物制备成口含片的方法:将提供的中药组合物粉末与适合的片剂辅料,如甘露醇、硬脂酸镁、糊精等,混合制粒,整粒,压片即得。
为了使组合物中各原料药更好的发挥药效,优选的对各原料药进行水提取或醇提取,如若选用了板蓝根,则对板蓝根进行水提取,取其水提液浓缩喷雾干燥得板蓝根水提物进行制剂;若选择肉豆蔻,则对肉豆蔻进行醇提取,取其醇提液经浓缩进行制剂。但这些不能用于限制本发明的保护范围。
在没有中药提取条件的情况下,优选将提供的组合物制成口含片或散剂。
优选的,15-40份、五灵脂5-30份、石菖蒲5-30份、西红花1-20份、诃子10-40份、长松萝1-25份、花椒0.5-5份、白芥子5-32份、棘豆1-30份、榼藤子1-20份、人工麝香0.1-5份。
更优选的,包括蔓菁15-30份、五灵脂5-20份、石菖蒲5-25份、西红花1-15份、诃子15-38份、长松萝1-18份、花椒0.5-3份、白芥子5-25份、棘豆1-20份、榼藤子1-15份、人工麝香0.1-3份。

西红花:西红花为鸢尾科番红花属植物藏红花(Crocus sativus L.)的干燥柱头,含有的西红花酸(Crocitin)、西红花醛(safranal)、西红花素(Crocin)和西红花苦素(Protocrocin),具有活血化瘀、凉血解毒、解郁安神。另外,现代技术研究表明,西红花具有抑制癌症、调节免疫力、抗氧化的功能。
诃子:为使君子科诃子属植物诃子(Terminalia chebula Retz.)的成熟果实,又称诃黎勒,微毛诃子。诃子具敛肺止咳、涩肠止泻、降火利咽的功效。在藏药系统中,诃子为最常用的藏药材,被奉为“藏药之王”。诃子的果实含鞣质23.60%-37.36%,内含:诃子酸,诃黎勒酸,鞣料云实精,诃子鞣质,2-3-O-连二没食子酰石榴皮鞣质,榄仁黄素,原诃子酸,葡萄糖没食子鞣甙,1,3,6-三没食子酰葡萄糖,没食子酸,并没食子酸等。又含三萜类成分:榄仁萜酸,诃王醇。还含莽草酸,去氢莽草酸,奎宁酸,三十碳酸,棕榈酸,没食子酸已酯,诃子次酸三已酯,胡萝卜甙,β-谷甾醇,阿拉伯糖,果糖,葡萄糖,蔗糖,鼠李糖和氨基酸等。另含番泻甙,诃子素以及鞣酸酶,多酚氧化酶,过氧化物酶,抗坏血酸氧化酶等。现代研究表明,诃子还具有一定的抗癌功效。石菖蒲:气味芳香,味甘、苦。功效滋补,生肌。
长松萝:长松萝是独特的民族医药药用资源,由真菌和与其共生的藻类形成的稳定的共生复合体,统称为地衣(Lichens),其主要生物学特征由共生菌体现出来,因为共生关系中真菌占统治地位,因此在分类学上把地衣划分为地衣型真菌(lichen-formingfungi)。地衣的种类多,耐寒耐旱,对环境的适应性极强,在地球表面具有广阔的生长领域,其他生物难以生存的地方(包括南极和北极)地衣都能生长繁殖。微苦、涩,化后味苦;凉。功效清热解毒。用于肺热,肝热,脉热,毒热等病症。据文献长松萝次生代谢产物可归类为取代盼类、缩酷类、二苯并 喃类、绍体类、三紹类、多糖及其他类成分,这些物质具有各种各样的生物活性,包括抗生素、抗细菌、抗病毒、抗炎、镇痛、解热、抗肿瘤、细胞毒性、抗福射、植物生长抑制和昆虫拒食等。包括如木栓酮,β-香树脂醇,β-谷甾醇(4),2,4-二羟基-3,6-二甲基苯甲酸甲酯,坝巴酸,泽屋萜,苔色酸乙酯,β-羟基-粘霉-5-烯,齐墩果酸(10),(+)-松萝酸,苔色酸甲酯和4-甲基-2,6-二羟基苯甲醛。
蔓菁:别名蔓菁、诸葛菜、圆菜头、圆根、盘菜,东北人称卜留克、苤蓝,新疆人称恰玛古,拉丁文名Brassica rapa,芸薹属芸薹种芜菁亚种,能形成肉质根的二年生草本植物。肥大肉质根供食用,肉质根柔嫩、致密,供炒食、煮食。欧洲、亚洲和美洲均有栽培。味辛,性温,治培根病、隆病,生赤巴,可防病毒。
研究表明,芜菁含有如下化学成分: 皂苷、黄酮类、糖类及其苷、生物碱类、挥发油、酚类、鞣质、氨基酸、蛋白质、谷甾醇、正十六烷、β-胡萝卜苷、槲皮素、正三十二烷醇、油酸、亚油酸、芥酸、二十碳烯酸、棕榈酸和硬脂酸等脂肪酸、异硫氰酸丁酯、异硫氰酸烯丙酯、硫代葡糖糖苷等。蔓菁具有的功效与上述有效成分有关。
花椒:芸香科植物花椒Zanthoxylumbungeanum Maxim.的成熟果皮。味辛,性糙,治隆病入心,为舒脉络之上品药。含挥发油,油中含有柠檬烯、枯醇、牻牛儿醇等。药理实验表明,果皮注射液有止痛、麻醉作用;牻牛儿醇小量可引起离体肠管蠕动,大量则使之抑制;水煎剂对链球菌、葡萄球菌、肺炎球菌、炭疽杆菌、枯草杆菌、霍乱弧菌、副伤寒杆菌和绿脓杆菌均有抑制作用。
人工麝香:含麝香酮,并含麝吡啶、11种雄甾烷衍生物,尚含胆甾醇类、脂肪、蛋白质、蜡及无机盐。实验表明,本品有兴奋中枢神经和苏醒作用,可兴奋呼吸、加速心搏、升高血压;能增加肾上腺素对beta受体的作用,并有对beta-儿茶酚胺的增加作用性凉,解毒治虫病,芳香开窍、通经络、消肿止痛等作用。
榼藤子:为豆科植物榼藤的茎藤,茎的块片呈不规则形,大小不等,斜而扭曲,厚1-2cm。外皮棕褐色或灰棕色,粗糙,有地衣斑,具明显纵纹或沟纹,可见侧枝痕和点状皮也,常有1条棱脊状突起。切面皮部深棕色,有红棕色或棕黑色树脂状物,木部棕色或浅棕色,有多数小孔,可见红棕色树脂状物环绕髓部呈偏心环纹,髓部常呈小空洞状,偏于有棱脊一侧。质坚硬,不易折断。 味微苦、涩,性平。治肝毒症,并且治疗脉劳损。根据表明榼藤子乙醇提取物中分离得到25个化合物,包括4个榼藤酰胺类化合物,3个倍半萜类化合物,8个芳香苷类化合物,9个三萜皂苷类化合物及一个蔗糖。根据相关的理化性质和波谱数据对其进行结构解析,鉴定了20个化合物,其中9个未见报道的新化合物,6个为从该属植物中首次分离得到。榼藤子具有的功效与上述有效成分有关。
白芥子:一年或二年生植物白芥Sinapis alba L的干燥成熟种子。十字花科,呈球形,表面灰白色至淡黄色,种皮薄而脆。味辛、甘,化后味苦;润、重;功效解毒、壮阳、消肿,用于食物中毒,肾炎,瘟疫及恶病。研究表明,白芥子中含有如下化学成份:4-羟基苯乙酸-2’-醛基-5’-呋喃甲酯[(5-formylfuran-2-yl)methyl 2-(4-hydroxyphenyl)acetate]、对羟苯基乙腈[2-(4-hydroxyphenyl)acetonitrile]、对羟基苯甲醛(4-hydroxybenzaldehyde)、胡萝卜苷(daucosterol)、软脂酸-1-单甘油酯(2,3-dihydroxypropyl palmitate)、β-谷甾醇(β-sitosterol)、芥子酸(sinapic acid)、对羟基苯甲酸(4-hydroxybenzoic acid)、对羟基苯乙酸(2-(4-hydroxyphenyl)acetic acid)。白芥子具有的功效与上述有效成分有关。
棘豆:豆科,约300种,分布于北温带,中国约30种,产西南、西北至东北部。多年生草本、半灌木或矮灌木,稀垫状小半灌木。根通常发达。茎发达、缩短或成根颈状。植物体被毛、腺毛或腺点,稀被不等臂的丁字毛。功效愈疮伤、炎症、清热、消肿,干黄水,并且杀瘟治毒症。研究表明,棘豆中含有如下成份:槲皮素、芹菜素-7-O-β-D-葡萄糖苷、山奈酚、芹菜素、槲皮素-3-O-α-L-鼠李糖苷、槲皮素-3-O-β-D-葡萄糖苷、山奈酚-7-O-α-L-鼠李糖-3-O-β-D-葡萄糖苷、异鼠李素-3-O-β-D-葡萄糖苷、山奈酚-3-O-β-D-葡萄糖-7-O-β-D-葡萄糖苷、山奈酚-3-O-β-D-葡萄糖苷、槲皮素-3-O-芸香糖苷、芹菜素-7-O-新橙皮糖苷;2个酚酸类:邻羟基苯甲酸、咖啡酸;1个生物碱类:腺苷;1个内酰胺类:已内酰胺;3个其它类:棕榈酸、β-谷甾醇、胡萝卜苷。其中芹菜素-7-O-β-D-葡萄糖苷、芹菜素、异鼠李素-3-O-β-D-葡萄糖苷、槲皮素-3-O-芸香糖苷、芹菜素-7-O-新橙皮糖苷5个化合物为首次从该植物中分离得到;邻羟基苯甲酸、槲皮素-3-O-α-L-鼠李糖苷、山奈酚-7-O-α-L-鼠李糖-3-O-β-D-葡萄糖苷、咖啡酸、腺苷。棘豆具有的功效与上述有效成分有关。
五灵脂:鼯鼠科动物复齿鼯鼠Trogopterusxanthipes Milne-Edwards的粪便。味甘而微苦,化后味甘;性凉、平,用于清热,解毒,补身。含多量的树脂、尿素、尿酸及维生素A类物质。动物实验表明,本品有抑制血小板聚集作用;增加冠脉血流量作用和改善微循环作用;并有抗应激性损伤、增强免疫功能、抑制胃酸分泌和抗氧化作用。
大黄:蓼科植物掌叶大黄Rheum palmatum, L.、唐古特大黄Rheum tanguticumMaxim. ex Balf.或药用大黄Rheumoj-flcinaleBaill.的干燥根和根茎。类圆柱形、圆锥形、卵圆形或不规则块状,长3~17厘米,直径3~10厘米。除净外皮者表面黄棕色至红棕色,有的可见类白色网状纹理,残留的外皮棕褐色,多具绳孔及粗皱纹。质坚实,有的中心稍松软,断面淡红棕色或黄棕色,显颗粒性;根茎髓部宽广.有星点环列或散在;根木部发达,具放射状纹理,形成层明显,无星点。气清香,味苦而微涩,嚼之粘牙,有沙粒感。大黄泻毒热腑热,并泻除培根病。其叶、茎功效治培根病,根的功效治下半身病。本品主要含芦荟大黄素、大黄酚、大黄素甲醚、大黄酸、大黄素等成分。大黄具有的功效与上述有效成分有关。
冬虫夏草:麦角菌科真菌冬虫夏草Cordycepssinensis (Berk.) Sacc.寄生在蝙蝠蛾科昆虫幼虫上的子座及幼虫尸体的复合体。性温,味淡。气特殊,味微苦涩。含粗蛋白、氨基酸、脂肪、D-甘露醇、虫草菌素、虫草酸、麦角甾醇、虫草多糖、生物碱、尿嘧啶、腺嘌呤、腺嘌呤核苷、维生素B12和多种微量元素。虫草酸和虫草菌素是虫草的主要活性物质。具有抑菌作用;能提高细胞吞噬功能,增强心血管和机体免疫机能,还具有抗癌和抗心律失常等功能。
红景天:为景天科植物大花红景天Rhodiolacrenulataook .f.et.Thoms.)H.Ohba干燥根及根茎。秋季采挖,除去粗皮,晒干。甘、苦、涩;寒。活血,清肺,止咳解热止痛。用于腊度(高山反应),恶心,呕吐,嘴唇和手心等发紫,全身无力,胸闷,难于透气,身体虚弱等症。研究表明红景天的有效成分为红景天苷、红景天苷元络醇、酪萨维、二苯甲基六氢吡啶、红景天素以及21种微量元素和18种氨基酸,氨基酸包含7种人体必需氨基酸。红景天具有的功效与上述有效成分有关。
灵芝:多孔菌科植物真菌紫芝GanodermasinenseZhao,Xu et Zhang或多孔菌科真菌赤芝Gan。derma lucidum(Ley-ss.ex Fr.)Karst.的子实体。味甘,性和。补气安神,止咳平喘。用于心神不宁,失眠心悸,肺虚咳喘,虚劳短气,不思饮食。含麦角、甾醇、真菌溶酶菌、酸性蛋白质、水溶性蛋白质、氨基酸、多肽、生物碱及多糖类、三萜化合物等。对神经系统有镇静作用;对心血管有增强其功能的作用;对呼吸系统具有镇咳、祛痰作用;还能提高代谢,增强免疫功能;对肺炎球菌、甲型链球菌、白色葡萄球菌及流感杆菌有抑制作用。灵芝具有的功效与上述有效成分有关。
天胡荽:伞形科植物天胡荽Hydrocotylesibthorpioides Lam.的全草。清热,利尿,消肿,解毒。研究表明,天胡荽含有chlorogenic acid methylester、正丁基-O-β-吡喃果糖苷、当归棱子芹醇葡萄糖苷、槲皮素3-O-β-D半乳糖苷、槲皮素、3’-O-methylquercetin、芹菜素、山奈酚、5-hydroxymaltol、齐墩果酸、B-谷甾醇、豆甾醇。天胡荽具有的功效与上述有效成分有关。
清半夏:又称清水半夏、清夏、清夏片。为生半夏用白矾加工炮制后入药者。清半夏毒性及辛燥之性降低,化痰作用增强,宜用于体弱多痰、寒湿较轻者。含挥发油,内含3-乙酰氨基-5-甲基异唑等60多种成分及16种氨基酸和多种无机元素。其中,所含的草酸钙针晶为半夏的刺激性成分之一,经炮制后,晶形发生变化,含量急剧下降,刺激性明显减弱。另含琥珀酸、丁基乙烯基醚、苯甲醛、半夏蛋白、姜辣醇、胰蛋白酶抑制剂等。清半夏具有的功效与上述有效成分有关。
陈皮:芸香科植物橘和其栽培变种的成熟果皮。采摘成熟果实,剥取果皮,晒干。常纵剖成整齐3瓣,基部相连。理气开胃,燥湿化痰,治脾胃病。属理气药,还具有抗胃溃疡、保肝利胆、升压、强心、扩张冠脉及抗肿瘤作用。
党参:条党、单枝党、板桥党、防党参、上党参、狮头参、中灵草。桔梗科植物党参Codonopsispilosula(Franch.)Nannf.、素花党参CodonopsispilosulaNannf.var.modesta(Nannf.)L.T.Shen或川党参CodonopsistangshenOliv.的干燥根。研究表明,党参含有挥发油、黄芩素葡萄糖苷、微量生物碱、多糖及皂苷。水煎液灌胃,使小鼠对钴射线有一定保护作用。多糖对实验动物应激性溃疡等多种溃疡均有明显的对抗作用。水、醇提物有中枢镇静作用,能改善小鼠的记忆过程和加强造血功能,并有抗血栓和调血脂的作用。
本发明具有以下有益效果:本发明公开的一种复方中药口含片,可以有效的抑制肿瘤,改善患者的生存质量。

实施例1
将生长旺盛的MCF-7细胞用含10%胎牛血清的DMEM培养基调整浓度为1×107/mL的细胞液,按0.2ml/只接种于裸鼠右侧腋窝皮下,观察瘤体的生长情况。待瘤体体积>100mm3时,测量瘤体的长(a,mm)和宽(b,mm),按计算瘤体体积,选择肿瘤生长旺盛且无破溃的荷瘤裸鼠,处死动物,用75%酒精消毒动物瘤体表面及周围皮肤,在无菌条件下剥离瘤体,用0.9%氯化钠注射液以1:1(V:V)比例制成活细胞计数为107/mL的瘤体组织匀浆液,接种于正常裸鼠右侧腋窝下,每只动物接种0.2mL,观察瘤体的生长情况,待瘤体体积>100mm3时,同法处理操作进行传代接种,用传至2代及以上的瘤体组织匀浆液按同法操作接种于正式实验裸鼠右侧腋下。
取瘤体体积为100mm3以上的裸鼠按瘤体体积随机分组,每组10只,实验设模型对照组、空白对照组、本发明药物给药组,按20mL/kg灌胃给药,1次/日,连续28日。试验期间每天观察动物的饮食、饮水、行为活动和瘤体的状况,每5天测量并记录瘤体的长、宽和动物体重,实验结束时处死动物,剥离瘤体,称重并拍照。
肿瘤体积及相对肿瘤增殖率:采用游标卡尺测量肿瘤的长、宽,计算肿瘤体积T;抗
肿瘤活性评价指标为相对肿瘤增殖率:。疗效评价标准:T/C % >40 %
为无效;T/C % ≤40%为有效。
从实验结果可以得出,通过与设立空白对照组和阳性药物对照组与本发明的药物设立的实验组进行对比实验。本发明的给药组肿瘤抑制率大于40%的动物数明显多于阳性对照组和空白给药组,证明药物对肿瘤细胞的增殖有明显的抑制作用。
实施例2
治疗方法:含服,一日一次,一次4片,一个月为一疗程。
疗效判断标准:
治愈:症状全部消失,两年及以上未复发;
显效:病灶处疼痛消失或明显减轻,CT 显示病灶处肿瘤明显缩小;
有效:病灶处疼痛缓解,CT 显示病灶处肿瘤未继续增长,其他部位未见新肿瘤;
无效:病灶处疼痛未缓解,肿瘤继续增长或转移。
处方6使用患者:患者陈某,55岁,检查患有肝癌。2015 年8 月入我院,入院时为肝癌晚期,慢性乙肝,腹胀腹痛食欲不振,乏力,尤其肝区疼痛难忍,CT 及彩超检查肝内肿瘤巨大,且有多发子灶,肿瘤组织占肝脏体积超过70%,肝硬化明显,少量腹水,门脉右支癌栓形成,肝癌转移。使用本发明的药物后症状逐步好转,一周后疼痛明显减轻,无需再注射止痛药,半月后,食欲得到改善,腹胀减轻,精神较治疗前好,继续服用病情稳定,无需再用止痛药,每餐吃两小碗饭,生活能自理。
处方3使用患者:王某,男,49岁。患者半年前检查出患有肝癌,用本本发明的中药制剂,治疗1个疗程后,患者腹痛、腹胀症状消失,继续治疗6个疗程,患者临床症状及体征全部消失,CT检查肿块没有明显增大,没有出现肿瘤转移的情况,半年后随访,患者身体状况良好,生活能自理。
治疗结果:52例恶性肝瘤患者服药一个疗程后症状不同程度缓解,三个疗程后明显改善,显效25例,有效15例,无效12例。
Claims (10)
1.一种治疗肝肿瘤的中药组合物,其特征在于,包括下列重量份配比的原料药:蔓菁15-40份、五灵脂5-30份、石菖蒲5-30份、西红花1-20份、诃子10-40份、长松萝1-25份、花椒0.5-5份、白芥子5-32份、棘豆1-30份,以及选自冬虫夏草1-15份、榼藤子1-20份、人工麝香0.1-5份,红景天1-30份、大黄1-20份、天胡荽1-10份中的至少一种组成。
2.根据权利要求1所述的中药组合物,其特征在于,包括下列重量份配比的原料药:蔓菁15-40份、五灵脂5-30份、石菖蒲5-30份、西红花1-20份、诃子10-40份、长松萝1-25份、花椒0.5-5份、白芥子5-32份、棘豆1-30份,以及选自冬虫夏草1-15份、榼藤子1-20份、人工麝香0.1-5份,红景天1-30份、大黄1-20份、天胡荽1-10份中的至少二种组成。
3.根据权利要求1所述的中药组合物,其特征在于,包括下列重量份配比的原料药::蔓菁15-40份、五灵脂5-30份、石菖蒲5-30份、西红花1-20份、诃子10-40份、长松萝1-25份、花椒0.5-5份、白芥子5-32份、棘豆1-30份、榼藤子1-20份、人工麝香0.1-5份。
4.根据权利要求1-3任意之一所述的中药组合物,其特征在于,原料药还包括至少一种选自陈皮、清半夏、半枝莲。
5.根据权利要求1-3任意之一所述的中药组合物,其特征在于,原料药还包括至少一种选自白术、党参、甘草。
6.根据权利要求1-3任意之一所述的中药组合物,其特征在于,原料药还包括至少一种选自沙参、五味子、百合。
7.根据权利要求1-3任意之一所述的中药组合物,其特征在于,包括下列重量份配比的原料药::蔓菁15-40份、五灵脂5-30份、石菖蒲5-30份、西红花1-20份、诃子10-40份、长松萝1-25份、花椒0.5-5份、白芥子5-32份、棘豆1-30份、榼藤子1-20份、人工麝香0.1-5份。
8.根据权利要求1-3任意之一所述的中药组合物,其特征在于,包括下列重量份配比的原料药::蔓菁15-30份、五灵脂5-20份、石菖蒲5-25份、西红花1-15份、诃子15-38份、长松萝1-18份、花椒0.5-3份、白芥子5-25份、棘豆1-20份、榼藤子1-15份、人工麝香0.1-3份。
9.一种中药制剂,其特征在于,由权利要求1-3任意一项提供的组合物,以及药学上可以接受的辅料制成。
10.一种保健食品,其特征在于将权利要求1-3任意一项提供的组合物,作为添加剂或成分之一制成。
