Abstract
本发明提供了一种用于减缓非感染性前列腺增生症状的组合物,由天然番茄果实脂溶性萃取物:油菜籽饼粕蛋白=(1‑9):(9‑1)。该组合物的制备方法包括:(1)将番茄果肉或皮干燥脱水,使其含水量低于16%;(2)萃取干燥后的番茄果肉或皮获得天然番茄果实脂溶性萃取物;(3)制备油菜籽饼粕蛋白;(4)将萃取液脱水干燥至含水量小于13%,干燥物粉碎至80‑120目;(5)灌胃液制备。该组合物以普通食品原料作为制备物,食用安全,适于除孕妇、乳母和婴幼儿外的所有人群;该组合物可缓解大鼠模型前列腺增生症状,其效果强于单独使用番茄红素;该组合物的原料丰富、经济且加工方法简并易行,适用做芯型软胶囊。

背景技术
前列腺增生症(BPH),旧称前列腺肥大,是老年男子常见疾病之一,为前列腺的一种良性病变。其发病原因与人体内雄激素与雌激素的平衡失调有关。病变起源于后尿道黏膜下的中叶或侧叶的腺组织、结缔组织及平滑肌组织,形成混合性圆球状结节。以两侧叶和中叶增生为明显,突入膀胱或尿道内,压迫膀胱颈部或尿道,引起下尿路梗阻。前列腺增生引起梗阻时,膀胱逼尿肌增厚,黏膜出现小梁、小室和憩室。长期的排尿困难使膀胱高度扩张,膀胱壁变薄,膀胱内压增高,输尿管末端丧失其活瓣作用,产生膀胱输尿管反流。
前列腺增生的病因分为两种:一种是感染性另外一种是非感染性。感染性前列腺增生由泌尿系统感染病原菌造成,非感染性前列腺增生由体内代谢失调造成。二者均可能造成严重疾病,如:前列腺癌。
目前,治疗前列腺非感染性增生的药物中包括的主要原料有松花粉和雌性激素类物质等。保健食品中的主要原料包括:番茄红素等。近期的文献报道检索植物种子蛋白及多肽也具有降低非感染性前列腺增生风险的作用。
番茄果实是人们日常生活经常食用的一种食品。番茄果实中含有丰富的番茄红素(0.1-0.3%(W/W),干重)。由于其分子结构中存在多个双键,番茄红素分子具有全反式和顺式异构体两种存在形式。番茄中的番茄红素以全反式异构体为主要存在形式,如式1所示。
从番茄果实中萃取番茄红素的方法包括:有机溶媒(如乙酸乙酯和超临界二氧化碳流体萃取法)。萃取物中番茄红素的含量在2-6%(W/W),经浓缩后可达到75%以上。已有的科学证据表明:番茄红素具有抗氧化、增强免疫、治疗前列腺增生等健康功能。
虽然已知番茄红素具有治疗前列腺增生的作用,但是如何能更好地提高番茄红素在治疗前列腺增生中的效果,仍然是一个亟待解决的问题。
发明内容
为了解决上述问题,本发明的目的在于提供一种用于减缓非感染性前列腺增生症状的组合物,该组合物通过将天然番茄果实脂溶性萃取物与油菜籽饼粕蛋白,原料简单,还可有效利用榨油废物。
本发明的另一目的在于提供上述用于减缓非感染性前列腺增生症状的组合物的制备方法。
为了实现上述目的,本发明提供一种用于减缓前列腺非感染性增生症状的组合物,该组合物按重量比由天然番茄果实脂溶性萃取物:油菜籽饼粕蛋白=(1-9):(9-1)。
其中,所述天然番茄果实脂溶性萃取物中总番茄红素含量按重量比计为6.0-80.0%,其中包括:E-异构体占88%。
本发明还提供上述述用于减缓前列腺非感染性增生症状的组合物的制备方法,包括如下步骤:
(1)将番茄果肉或皮干燥除水,使其含水量以重量计低于16%;
(2)使用超临界二氧化碳流体萃取法或有机溶酶萃取法萃取干燥后的番茄果肉或皮获得天然番茄果实脂溶性萃取物;
(3)油菜籽饼粕蛋白的制备:
将白菜型、橄榄型或芥菜型油菜籽榨油后的残渣用4-20倍按V/W物料重量计的磷酸缓冲液萃取,萃取时间=0.5-4h;萃取温度=室温-55℃,的萃取液,其中所述磷酸缓冲液0.1M,pH=5-8;
(4)将萃取液脱水干燥,至含水量以重量计小于13%,干燥物粉碎至80-120目;
(5)按重量比将天然番茄果实脂溶性萃取物与油菜籽饼粕蛋白混合制成组合物。
其中,步骤(1)中所述采用超临界二氧化碳流体萃取法制取天然番茄果实脂溶性萃取物的条件为:萃取压力=25-65MPa,萃取温度45-70℃,萃取溶媒体积400倍,按V/W物料重量计;萃取时间=0.5-6h,夹带剂:乙醇、乙酸乙酯或正己烷。
其中,步骤(1)中所述采用有机溶媒法制取天然番茄果实脂溶性萃取物的条件为:溶媒种类:正己烷、乙酸乙酯、四氢呋喃、丙酮、卤代烷烃或低级醇;溶媒用量=4-20倍,按V/W物料重量计;萃取时间=0.5-48h;萃取温度=4℃-溶剂沸点。
其中,在步骤(5)制成的组合物中以W/V计进一步加入10-40%的食用油制成组合物灌胃液。
优选地,所述食用油为花生油、豆油、葵花籽油、橄榄油、玉米油、沙棘油、紫苏籽油、鱼油、番茄籽油、茶油或棉籽油。
油菜属十字花科芸薹属的植物。在我国有白菜型、橄榄型、芥菜型等栽培品系。油菜籽中含有丰富的脂肪,是我国重要的食用油生产原料。经过榨油后,油菜籽残渣中含有丰富的蛋白质可达到30-40%(W/W)。经蛋白酶水解可形成多肽。已有的科学证据表明:油菜籽饼粕蛋白具有抗氧化等健康功能。本发明将油菜籽饼粕蛋白与天然番茄果实脂溶性萃取物组合使用,可以进一步提升天然番茄果实脂溶性萃取物中番茄红素的治疗前列腺增生效果。
本发明的优点在于:
(1)番茄提取物与油菜籽饼粕蛋白组合物为普通食品原料制备物,具有很高的食品安全性,适于除孕妇、乳母和婴幼儿外的所有人群;
(2)组合物缓解大鼠模型非感染性前列腺增生症状效果明显。其效果强于两种原料单独使用的效果,达到了协同增效的作用;
(3)组合物的原料丰富、经济且加工方法简并易行,可制成芯型软胶囊。

具体实施方式
下面结合附图1所示的各实施方式对本发明进行详细说明,但应当说明的是,这些实施方式并非对本发明的限制,本领域普通技术人员根据这些实施方式所作的功能、方法、或者结构上的等效变换或替代,均属于本发明的保护范围之内。
实施例1建立雄性大鼠前列腺增生模型
(1)实验动物及分组
选健康SD雄性大鼠(150±5g体重)48只(包括:建模空白对照组、给药空白对照组、番茄红素阳性对照组、番茄红素和油菜籽饼粕蛋白组合物低剂量给药组、番茄红素和油菜籽饼粕蛋白组合物中剂量给药组、番茄红素和油菜籽饼粕蛋白组合物高剂量给药组),随机分为6组,每组8只。大鼠购自北京大学医药部实验动物中心,实验动物合格证号,SCXK京2008-0008。在SPS级动物房做适用性饲养6天。所有饲料由北京奥科协力饲料有限公司提供。饲料中确认不含番茄红素和油菜种子蛋白。
(2)建立雄性大鼠前列腺增生模型
皮下注射丙酸睾注射液,用量4-6mg/Kg BW,1次/每日,连续注射30天。建模对照组注射等量豆油,测定大鼠前列腺湿重、体重,按式(I)计算前列腺指数(PI):
前列腺指数[%]=(前列腺鲜重/体重)*100% (I)
雄性大鼠经皮下注射丙酸睾可诱发前列腺增生。这一动物模型可用于缓解前列腺增生病症的药物筛选。
实施例2制备组合物灌胃液
1.将100kg番茄果肉干燥脱水至含水量低于16%。
2.应用超临界二氧化碳流体萃取法,制备番茄果实脂溶性萃取物,具体条件为:萃取压力=45MPa,萃取温度60℃,萃取溶媒体积400倍,按V/W物料重量计;萃取时间=4h,夹带剂:乙醇。制得天然番茄果实脂溶性萃取物
其中,天然番茄果实脂溶性萃取物中总番茄红素含量6.0%(W/W),其中包括:E-异构体占88%(W/W)。
3.用磷酸缓冲液(0.1M,pH=5)萃取白菜型油菜(江苏镇江当地品种)籽榨油残渣中的蛋白质,缓冲液用量为榨油残渣的14倍(V/W,物料重量),萃取时间=2.5h,萃取温度室温。
4.萃取液在45℃、-0.06MPa条件下脱水干燥至含水量为12.9%(W/W),干燥物粉碎至100目。
5.按天然番茄果实脂溶性萃取物与白菜型油菜籽饼粕蛋白的比例为1:1(W/W)制成灌胃液中的固形物,用豆油按固形物含量20%(W/V)配制灌胃液。制成灌胃液中含有天然番茄脂溶性萃取物100mg/mL(其中总番茄红素含量6mg/mL),饼粕蛋白粉含量100mg/mL。
实施例3
对实施例1建模成功的大鼠,用实施例2制备的灌胃液灌胃,每次1mL,每天两次(间隔12小时),连续灌胃3周。给药对照组给予等量的食用油。阳性对照组给予含有天然番茄脂溶性萃取物100mg/mL(其中总番茄红素含量≥6mg/mL)的灌胃液。
按上述方法测定受试动物前列腺指数,使用SPS软件对所得收据进行统计分析。
结果与分析
实验结果见表1:

表1结果表明:按天然番茄果实脂溶性萃取物与白菜型油菜籽饼粕蛋白1:1配制的组合物所得结果与单独使用天然番茄果实脂溶性萃取物所得结果差异显著。即:组合物比单独使用天然番茄果实脂溶性萃取物对雄性大鼠的非感染性前列腺增生具有更好的治疗作用。
实施例4
1.将100kg番茄果肉干燥脱水至含水量低于16%。
2.用400L乙酸乙酯萃取干粉,萃取温度50℃,萃取时间30min,搅拌速度30rpm。重复萃取3次。合并收集萃取液。在45℃、-0.06MPa条件下蒸发出去萃取溶媒,至萃取物恒重,制成番茄果实脂溶性萃取物。
其中,总番茄红素含量6%(W/W),其中包括:E-异构体占88%(W/W)。
3.用磷酸缓冲液(0.1M,pH=5)萃取橄榄型油菜(甘肃当地品种)籽榨油残渣中的蛋白质。缓冲液用量为榨油残渣的14倍(V/W,物料重量)。萃取时间=3h。,萃取温度室温。
4.萃取液在45℃、-0.06MPa条件下脱水干燥至含水量为12.2%(W/W)。干燥物粉碎至100目。
5.按天然番茄果实脂溶性萃取物与橄榄型油菜籽饼粕蛋白的比例为1:2(W/W)制成灌胃液中的固形物。用食用油按固形物含量30%(W/V)配制灌胃液。制成灌胃液中含有天然番茄脂溶性萃取物100mg/mL(其中总番茄红素含量6mg/mL),饼粕蛋白粉含量200mg/mL。
实施例5
对实施例1建模成功的大鼠,用实施例4制备的灌胃液灌胃,每次1mL,每天两次(间隔12小时),连续灌胃3周。给药对照组给予等量的食用油。阳性对照组给予含有天然番茄脂溶性萃取物100mg/mL(其中总番茄红素含量6mg/mL)的灌胃液。
(5)按上述方法测定受试动物前列腺指数,使用SPS软件对所得收据进行统计分析。

从上述实施例可以看出,本发明提供的组合物对雄性大鼠的非感染性前列腺增生比单独使用番茄红素具有更好的治疗作用,因此,天然番茄果实脂溶性萃取物与白菜型油菜籽饼粕蛋白的组合具有协同增效的作用。

Claims (5)
1.一种用于减缓非感染性前列腺增生症状的组合物,其特征在于,按重量比由天然番茄果实脂溶性萃取物:油菜籽饼粕蛋白为1:1或1:2;其中,所述天然番茄果实脂溶性萃取物中总番茄红素含量按重量比计为6.0-80.0%,其中包括:E-异构体占总番茄红的88%。
2.一种如权利要求1所述用于减缓非感染性前列腺增生症状的组合物的制备方法,其特征在于,包括如下步骤:
(1)将番茄果肉或皮干燥脱水,使其含水量以重量计低于16%;
(2)使用超临界二氧化碳流体萃取法或有机溶媒萃取法萃取干燥后的番茄果肉或皮获得天然番茄果实脂溶性萃取物;
(3)制备油菜籽饼粕蛋白:
将白菜型、橄榄型或芥菜型油菜籽榨油后的残渣用4-20倍按V/W物料重量计的磷酸缓冲液萃取,萃取时间=0.5-4h;萃取温度=室温-55℃,的萃取液,其中所述磷酸缓冲液0.1M,pH=5-8;
(4)将萃取液脱水干燥至含水量以重量计小于13%,干燥物粉碎至80-120目;
(5)按重量比将天然番茄果实脂溶性萃取物与油菜籽饼粕蛋白混合制成组合物;
其中:步骤(2)中所述采用超临界二氧化碳流体萃取法制取天然番茄果实脂溶性萃取物的条件为:萃取压力=25-65 MPa,萃取温度45-70℃,萃取溶媒体积400倍,按V/W物料重量计;萃取时间=0.5-6h,夹带剂:乙醇、乙酸乙酯或正己烷;
步骤(2)中所述采用有机溶媒萃取法制取天然番茄果实脂溶性萃取物的条件为:溶媒种类:正己烷、乙酸乙酯、四氢呋喃、丙酮、卤代烷烃或低级醇;溶媒用量=4-20倍,按V/W物料重量计;萃取时间=0.5-48h;萃取温度=4℃-溶剂沸点。
3.如权利要求2所述用于减缓非感染性前列腺增生症状的组合物的制备方法,其特征在于,在步骤(5)制成的组合物中以W/V计进一步加入10-40%的食用油制成组合物灌胃液。
4.如权利要求3所述用于减缓非感染性前列腺增生症状的组合物的制备方法,其特征在于,所述食用油为花生油、豆油、葵花籽油、橄榄油、玉米油、沙棘油、紫苏籽油、鱼油、番茄籽油、茶油或棉籽油。
5.如权利要求1所述的减缓非感染性前列腺增生症状的组合物或权利要求2-4任一项所述用于减缓非感染性前列腺增生症状的组合物的制备方法,其特征在于,所述非感染性前列腺增生症状为雄性大鼠的非感染性前列腺增生症状。
