本发明公开了一种含二十八烷醇表面活性剂的制备方法及应用,在水中加入γ‑氨基丁酸和吐温80,充分搅拌,形成γ‑氨基丁酸溶液,然后依次加入二十八烷醇和脂酶,搅拌反应得到γ‑氨基丁酸二十八烷醇酯溶液,最后经喷雾干燥得到γ‑氨基丁酸二十八烷醇酯粉末。本发明利用二十八烷醇和γ‑氨基丁酸合成制备了表面活性剂γ‑氨基丁酸二十八烷醇酯,解决了高级脂肪醇水溶性差的问题,其在食品、化妆品和饮料中添加不会产生任何风味及感观的问题,均一性较好;同时,γ‑氨基丁酸与二十八烷醇对机体调节具有互补,协同的作用,能够更好的控制血压,阻止血栓形成,有效预防心脑血管疾病。

背景技术
二十八烷醇是一种生物活性物质,其主要生理作用包括:1)增进耐力、精力和体力;2)缩短肌神经反应时间,提高反应敏锐性;3)促进激素功能,减轻肌肉疼痛;4)改善心肌功能,降低收缩期血压;5)提高机体基础代谢率。二十八烷醇可用于治疗血钙过多的骨质疏松,治疗高胆固醇和高脂蛋白血疾病,刺激动物及人类的性行为,含有二十八烷醇的化妆品能促进皮肤血液的循环和活化细胞,有消炎、防治皮肤病等功效。而且二十八烷醇安全性极高,用量很少。
γ-氨基丁酸是中枢神经系统中很重要的抑制性神经递质,它是一种天然存在的非蛋白组成氨基酸,具有极其重要的生理功能,它能促进脑的活化性,健脑益智,抗癫痫,促进睡眠,美容润肤,延缓脑衰老机能,能补充人体抑制性神经递质,具有良好的降血压功效。促进肾机能改善和保护作用。抑制脂肪肝及肥胖症,活化肝功能。每日补充微量的γ-氨基丁酸有利于心脑血压的缓解,又能促进人体内氨基酸代谢的平衡,调节免疫功能。而且二十八烷醇和γ-氨基丁酸联合作用于机体,不会产生任何毒性和致突变作用,能共同发挥其保健功效。
由于高级脂肪醇的水溶性较差,因此在饮料、化妆品、食品中添加会影响其风味和感观。因此,需要做适当修饰,增加其溶解性能。

本发明所要解决的技术问题是提供一种含二十八烷醇表面活性剂的制备方法,以增强二十八烷醇的溶解性,并且和γ-氨基丁酸联用达到保健功效。
本发明还要解决的技术问题是提供上述含二十八烷醇表面活性剂的应用。
为了解决上述技术问题,本发明采取的技术方案如下:
一种含二十八烷醇表面活性剂的制备方法,包括如下步骤:
步骤一:在水中加入γ-氨基丁酸和吐温80,充分搅拌,形成γ-氨基丁酸溶液;
步骤二:将二十八烷醇加入到步骤一得到的γ-氨基丁酸溶液中,加入脂酶,搅拌反应,得到γ-氨基丁酸二十八烷醇酯溶液;
步骤三:将γ-氨基丁酸二十八烷醇酯溶液喷雾干燥得到γ-氨基丁酸二十八烷醇酯粉末。
进一步地,步骤一中,所述γ-氨基丁酸按照浓度0.1~0.2g/L加入到水中(优选0.1g/L),所述吐温80按照浓度浓度0.2~0.5g/L加入到水中(优选0.2g/L),在8000~10000r/min(优选9000r/min)高速搅拌下制得γ-氨基丁酸溶液。
步骤二中,所述二十八烷醇按照浓度0.1~0.5g/L(优选0.2g/L)加入到γ-氨基丁酸溶液中,所述的脂酶按照浓度1000~5000U/ml(优选5000U/ml)加入到γ-氨基丁酸溶液中;反应温度控制在30~40℃之间(优选37℃),反应时间为4~8h(优选8h)。
步骤三中,所述喷雾干燥进气温度为200~250℃(优选250℃),空气流量为100~120kg干空气/h(优选120kg干空气/h),溶液流量为2~3kg/h(优选2kg/h)。
上述制备方法制备得到的含二十八烷醇表面活性剂粉末也在本发明的保护范围中,其中以结合形式存在的二十八烷醇质量比例为20~40%(优选40%)。
本发明还要求保护上述含二十八烷醇表面活性剂粉末作为添加剂加入食品、饮料或化妆品中的应用,将该粉末配制成100~500mg/L(优选200mg/L)的溶液加入食品、化妆品或饮料中。
有益效果:
本发明利用二十八烷醇和γ-氨基丁酸合成制备了含二十八烷醇表面活性剂,解决了高级脂肪醇水溶性差、稳定性差的问题,其在食品、化妆品和饮料中添加不会产生任何风味及感观的问题,均一性较好;同时,γ-氨基丁酸能作用于脊髓的血管运动中枢,有效促进血管扩张,达到降低血压的目的。而二十八烷醇能够有效降低低密度脂蛋白和升高高密度脂蛋白水平,是对患有高血压或血栓形成的高风险的患者的补充。两者协同作用,能够更好的控制血压,阻止血栓形成,有效预防心脑血管疾病。

实施例1
1)将γ-氨基丁酸以浓度0.1g/L,吐温80以浓度0.2g/L加入水中,在9000r/min高速搅拌下制得γ-氨基丁酸溶液;
2)向步骤1)制得的γ-氨基丁酸溶液中以浓度0.2g/L加入二十八烷醇,以浓度5000U/ml加入脂酶,在37℃下搅拌反应8h,得到γ-氨基丁酸二十八烷醇酯溶液;
3)将γ-氨基丁酸二十八烷醇酯溶液进行喷雾干燥,喷雾干燥参数:进气温度为250℃,空气流量为120kg干空气/h,溶液流量为2kg/h,得到γ-氨基丁酸二十八烷醇酯粉末。
实施例2
1)将γ-氨基丁酸以浓度0.1g/L,吐温80以浓度0.2g/L加入水中,在8000r/min高速搅拌下制得γ-氨基丁酸溶液;
2)向步骤1)制得的γ-氨基丁酸溶液中以浓度0.1g/L加入二十八烷醇,以浓度1000U/ml加入脂酶,在30℃下搅拌反应4h,得到γ-氨基丁酸二十八烷醇酯溶液;

1)将γ-氨基丁酸以浓度0.2g/L,吐温80以浓度0.5g/L加入水中,在10000r/min高速搅拌下制得γ-氨基丁酸溶液;
2)向步骤1)制得的γ-氨基丁酸溶液中以浓度0.5g/L加入二十八烷醇,以浓度5000U/ml加入脂酶,在40℃下搅拌反应8h,得到γ-氨基丁酸二十八烷醇酯溶液;
3)将γ-氨基丁酸二十八烷醇酯溶液进行喷雾干燥,喷雾干燥参数:进气温度为250℃,空气流量为120kg干空气/h,溶液流量为3kg/h,得到γ-氨基丁酸二十八烷醇酯粉末。
对比例1
1)将吐温80以浓度0.2g/L加入水中,在9000r/min高速搅拌下制得吐温80溶液;
2)再向步骤1)制备的吐温80溶液中以浓度0.2g/L加入二十八烷醇,以浓度5000U/ml加入脂酶,在37℃下搅拌反应8h,得到二十八烷醇酯悬浊液;
3)将二十八烷醇悬浊液进行喷雾干燥,喷雾干燥参数:进气温度为250℃,空气流量为120kg干空气/h,溶液流量为2kg/h,得到二十八烷醇酯粉末。
对比例2
1)将γ-氨基丁酸以浓度0.1g/L加入水中,在9000r/min高速搅拌下制得γ-氨基丁酸溶液;
2)向步骤1)制备的γ-氨基丁酸溶液中以浓度0.2g/L加入二十八烷醇,以浓度5000U/ml加入脂酶,在37℃下搅拌反应8h,得到γ-氨基丁酸二十八烷醇酯溶液;
3)将γ-氨基丁酸二十八烷醇酯溶液进行喷雾干燥,喷雾干燥参数:进气温度为250℃,空气流量为120kg干空气/h,溶液流量为2kg/h,得到γ-氨基丁酸二十八烷醇酯粉末。
对比例3
1)将γ-氨基丁酸以浓度0.1g/L,吐温80以浓度0.2g/L加入水中,在9000r/min高速搅拌下制得γ-氨基丁酸溶液;
2)向步骤1)制得的γ-氨基丁酸溶液中以浓度0.2g/L加入二十八烷醇,在37℃下搅拌8h,得到γ-氨基丁酸二十八烷醇悬浊液;
3)将γ-氨基丁酸二十八烷醇悬浊液进行喷雾干燥,喷雾干燥参数:进气温度为250℃,空气流量为120kg干空气/h,溶液流量为2kg/h,得到γ-氨基丁酸二十八烷醇粉末。
将实施例1~3、对比例1~3制备的粉末分别配制成200mg/L水溶液,然后进行300W超声分散10min,再用10000×g离心10min,检查溶液均一性及是否有不溶物,离心上清液经酸水解后采用GB/T 32947-2016《蜂蜡中二十八烷醇、三十烷醇的测定气相色谱法》检测二十八烷醇质量比例。
酸水解方法:加入10mmol/L硫酸,煮沸30min,冷却到室温,用5mol/L氢氧化钠溶液调pH至7.0。
结果见表1。

从表1数据可以看出,采用二十八烷醇和γ-氨基丁酸合成γ-氨基丁酸二十八烷醇酯溶液,然后喷雾干燥得到的含二十八烷醇表面活性剂粉末能够有效解决高级脂肪醇水溶性差的问题,具有显著的进步。
本发明提供了一种含二十八烷醇表面活性剂的制备方法及应用的思路及方法,具体实现该技术方案的方法和途径很多,以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。本实施例中未明确的各组成部分均可用现有技术加以实现。
Claims (10)
1.一种含二十八烷醇表面活性剂的制备方法,其特征在于,包括如下步骤:
步骤一:在水中加入γ-氨基丁酸和吐温80,充分搅拌,形成γ-氨基丁酸溶液;
步骤二:将二十八烷醇加入到步骤一得到的γ-氨基丁酸溶液中,再加入脂酶,搅拌反应,得到γ-氨基丁酸二十八烷醇酯溶液;
步骤三:将γ-氨基丁酸二十八烷醇酯溶液喷雾干燥得到γ-氨基丁酸二十八烷醇酯粉末。
2.根据权利要求1所述的一种含二十八烷醇表面活性剂的制备方法,其特征在于,步骤一中,所述γ-氨基丁酸按照浓度0.1~0.2g/L加入到水中。
3.根据权利要求1所述的一种含二十八烷醇表面活性剂的制备方法,其特征在于,步骤一中,所述吐温80按照浓度浓度0.2~0.5g/L加入到水中。
4.根据权利要求1所述的一种含二十八烷醇表面活性剂的制备方法,其特征在于,步骤一中,在8000~10000r/min高速搅拌下制得γ-氨基丁酸溶液。
5.根据权利要求1所述的一种含二十八烷醇表面活性剂的制备方法,其特征在于,步骤二中,所述二十八烷醇按照浓度0.1~0.5g/L加入到γ-氨基丁酸溶液中。
6.根据权利要求1所述的一种含二十八烷醇表面活性剂的制备方法,其特征在于,步骤二中,所述的脂酶按照浓度1000~5000U/ml加入到γ-氨基丁酸溶液中。
7.根据权利要求1所述的一种含二十八烷醇表面活性剂的制备方法,其特征在于,步骤二中,反应温度为30~40℃,反应时间为4~8h。
8.根据权利要求1所述的一种含二十八烷醇表面活性剂的制备方法,其特征在于,步骤三中,所述喷雾干燥进气温度为200~250℃,空气流量为100~120kg干空气/h,溶液流量为2~3kg/h。
9.权利要求1~8中任意一项制备方法制备得到的含二十八烷醇表面活性剂,其特征在于,以结合形式存在的二十八烷醇质量比例为20~40%。
10.权利要求6所述含二十八烷醇表面活性剂作为添加剂加入食品、饮料或化妆品中的应用。
