Abstract

本发明公开了一种娑罗子皂苷提取工艺,包括粗皂苷的制备、树脂的处理及其与娑罗子提取物七叶皂苷的吸附和解吸附、七叶皂苷粗纯化、高效液相色谱、七叶皂苷精纯化,本发明涉及一种娑罗子皂苷提取工艺,本工艺采用大孔吸附树脂在对总皂苷的粗分离和除去水溶性色素、多糖和无机盐等杂质显示出其独特的效果,在实验中先用水作洗脱剂,除去一些水溶性杂质,然后再用不同浓度的乙醇进行洗脱,得到各皂苷部位,本发明将娑罗子提取物上NKA‑9吸附树脂进一步纯化,纯度为63.323%,获得率为3.67%,这为有效开发利用中药娑罗子资源提供有实用价值的分离方法,同时,为此类中药研究提供借鉴。

一种娑罗子皂苷提取工艺
技术领域
本发明属于皂苷提取技术领域,具体为一种娑罗子皂苷提取工艺。
背景技术
皂苷是苷元为三萜或螺旋甾烷类化合物的一类糖苷,主要分布于陆地高等植物中,也少量存在于海星和海参等海洋生物中。许多中草药如人参、远志、桔梗、甘草、知母和柴胡等的主要有效成分都含有皂苷。有些皂苷还具有抗菌的活性或解热、镇静、抗癌等有价值的生物活性。
皂苷类物质的提取纯化一般采用D101树脂,但实验证明,NKA-9树脂对七叶皂苷的纯化效果比D101好,而目前的娑罗子提取物上NKA-9吸附树脂的纯化程度不够,七叶皂苷的提取都是先用有机溶剂提取,过滤后或用活性炭去除杂质,或用沉淀法沉淀或经过大孔吸附树脂,用活性炭吸附和沉淀法是经典的分离方法,对分离度差别较大的化合物能起到较好的分离效果。而对分离度差异小的物质,则难以分离。
发明内容:
本发明的目的就在于为了解决上述背景技术中所提到的问题,而提供一种娑罗子皂苷提取工艺,解决了现有技术存在的问题。
为了解决上述问题,本发明提供了一种技术方案:一种娑罗子皂苷提取工艺,包括粗皂苷的制备、树脂的处理及其与娑罗子提取物七叶皂苷的吸附和解吸附、七叶皂苷粗纯化、高效液相色谱、七叶皂苷精纯化。
优选的,所述粗皂苷的制备包括娑罗子准备、粗皂苷溶液制备,具体如下:
A、娑罗子准备:将本年季的娑罗子清洗晾干后放置备用;
B、粗皂苷溶液制备:将娑罗子100g磨碎,再用60%的酒精各300mL分3次提取,抽滤,得抽提液800mL,将抽提液在60℃的恒温水浴锅上加热浓缩48h,得到78mL浓缩液,再用乙酸乙酯萃取3次,得上清液1200mL,沉淀粘稠物26.7mL,上清液水浴浓缩得沉淀12g,放置备用。
优选的,所述树脂的处理及其与娑罗子提取物七叶皂苷的吸附和解吸附将处理后的树脂与皂苷溶液充分混合30min,水洗10min,随之用三组不同浓度酒精,酒精浓度分别为60%、80%和100%,依次分别洗脱20min,收集的洗脱液经含量测定,绘制曲线图。
优选的,所述七叶皂苷粗纯化将将C18层析柱先用100mL的30%乙醇浸泡,用水洗至无醇味,将经过吸附柱NKA-9树脂纯化后的纯品溶于水,流速控制在30mL/h,水洗30mL,80%酒精60mL,用收集器收集,得到七叶皂苷的精制品.将经吸附柱NKA-9树脂纯化后的七叶皂苷分为未上过和上过反相柱的七叶皂苷分别加甲醇溶解后,制成2.5mg/ml溶液经高效液相色谱进行检测。
优选的,所述高效液相色谱用十八烷基硅烷键合硅胶为填充剂,取85%磷酸5.5mL,用水稀释至1000mL为流动相,混合比例为40∶60,检测波长为220~240nm,流速1000mL/min,取本品标准品适量,加甲醇溶解并制成3mg/ml溶液,取标准品液5μL和测试样品5μL注入高效液相色谱柱中,记录色谱图。

本具体实施方式采用以下技术方案:一种娑罗子皂苷提取工艺,包括粗皂苷的制备、树脂的处理及其与娑罗子提取物七叶皂苷的吸附和解吸附、七叶皂苷粗纯化、高效液相色谱、七叶皂苷精纯化。
其中,所述粗皂苷的制备包括娑罗子准备、粗皂苷溶液制备,具体如下:
A、娑罗子准备:将本年季的娑罗子清洗晾干后放置备用;
B、粗皂苷溶液制备:将娑罗子100g磨碎,再用60%的酒精各300mL分3次提取,抽滤,得抽提液800mL,将抽提液在60℃的恒温水浴锅上加热浓缩48h,得到78mL浓缩液,再用乙酸乙酯萃取3次,得上清液1200mL,沉淀粘稠物26.7mL,上清液水浴浓缩得沉淀12g,放置备用。
其中,所述树脂的处理及其与娑罗子提取物七叶皂苷的吸附和解吸附将处理后的树脂与皂苷溶液充分混合30min,水洗10min,随之用三组不同浓度酒精,酒精浓度分别为60%、80%和100%,依次分别洗脱20min,收集的洗脱液经含量测定,绘制曲线图。
其中,所述七叶皂苷粗纯化将将C18层析柱先用100mL的30%乙醇浸泡,用水洗至无醇味,将经过吸附柱NKA-9树脂纯化后的纯品溶于水,流速控制在30mL/h,水洗30mL,80%酒精60mL,用收集器收集,得到七叶皂苷的精制品.将经吸附柱NKA-9树脂纯化后的七叶皂苷分为未上过和上过反相柱的七叶皂苷分别加甲醇溶解后,制成2.5mg/ml溶液经高效液相色谱进行检测。

1.一种娑罗子皂苷提取工艺,其特征在于,包括粗皂苷的制备、树脂的处理及其与娑罗子提取物七叶皂苷的吸附和解吸附、七叶皂苷粗纯化、高效液相色谱、七叶皂苷精纯化。
2.根据权利要求1所述的一种娑罗子皂苷提取工艺,其特征在于:所述粗皂苷的制备包括娑罗子准备、粗皂苷溶液制备,具体如下:
A、娑罗子准备:将本年季的娑罗子清洗晾干后放置备用;
B、粗皂苷溶液制备:将娑罗子100g磨碎,再用60%的酒精各300mL分3次提取,抽滤,得抽提液800mL,将抽提液在60℃的恒温水浴锅上加热浓缩48h,得到78mL浓缩液,再用乙酸乙酯萃取3次,得上清液1200mL,沉淀粘稠物26.7mL,上清液水浴浓缩得沉淀12g,放置备用。
3.根据权利要求1所述的一种娑罗子皂苷提取工艺,其特征在于:所述树脂的处理及其与娑罗子提取物七叶皂苷的吸附和解吸附将处理后的树脂与皂苷溶液充分混合30min,水洗10min,随之用三组不同浓度酒精,酒精浓度分别为60%、80%和100%,依次分别洗脱20min,收集的洗脱液经含量测定,绘制曲线图。
4.根据权利要求1所述的一种娑罗子皂苷提取工艺,其特征在于:所述七叶皂苷粗纯化将将C18层析柱先用100mL的30%乙醇浸泡,用水洗至无醇味,将经过吸附柱NKA-9树脂纯化后的纯品溶于水,流速控制在30mL/h,水洗30mL,80%酒精60mL,用收集器收集,得到七叶皂苷的精制品.将经吸附柱NKA-9树脂纯化后的七叶皂苷分为未上过和上过反相柱的七叶皂苷分别加甲醇溶解后,制成2.5mg/ml溶液经高效液相色谱进行检测。
