预防细胞同种异体移植排斥是当前细胞治疗策略中的主要挑战。这种排斥是由同种异体移植物反应性效应CD4和CD8 T细胞介导。用免疫抑制药物进行终身治疗减弱同种异体反应和移植排斥,但这与感染和恶性肿瘤的风险增加相关。近年来,已经开发了几种用于预防同种异体细胞移植的免疫排斥的基于生物材料的策略,依赖于细胞包裹在可透过营养物和废产物的膜囊内;然而,所述囊还防止细胞进入和血管形成,因此限制所述装置的尺寸至200μm的氧扩散距离。
理想的细胞移植装置应该能够使其整个体积的血管形成以维持所移植细胞的活力,并且应该促进宿主内的移植物整合。然而,由于经由血液循环的白细胞的大量浸润,装置血管形成通常与同种异体移植排斥的风险增加有关。
转化生长因子-β(transforming growth factor-β,TGF-β)是免疫调节细胞因子,其能增强免疫耐受微环境的产生,因为它表现出抑制炎症过程,例如自身免疫性疾病和癌症中明显的炎症过程。在TGF-β富集的环境中,摄取抗原的树突状细胞(DC)变成致耐受性并介导CD4+T细胞分化成抗炎调节性T细胞,其不仅抑制效应CD4+T细胞的产生,而且抑制细胞毒性CD8+T细胞的活化和增殖。
因此,存在用于产生局部免疫耐受微环境同时避免不期望的全身性影响的改进的组合物和方法的需求。
发明简述

本发明进一步涉及在对象中抑制或治疗自身免疫疾病或病症,同时避免全身性免疫抑制副作用的方法。因此,本发明提供生物活性多肽的局部释放,所述生物活性多肽包括但不限于TGFβ-l,其干扰自身免疫信号传导来导致其局部抑制。本发明考虑的自身免疫性疾病和病症,包含但不限于多发性硬化、I型糖尿病、和银屑病。
另一方面,本发明涉及自身免疫疾病的抑制,其中所述自身免疫疾病选自由多发性硬化、银屑病、和I型糖尿病组成的组。
因此,本发明在一方面提供在多发性硬化(MS)自身抗原特异性CD4+T细胞中诱导免疫耐受的方法,所述自身抗原包括但不限于髓磷脂碱性蛋白(MBP)、蛋白脂质蛋白(PLP)和髓磷脂少突胶质细胞糖蛋白(MOG)。在一个实施方案中,本发明提供通过施用包括硫酸酯化多糖、TGFβ1和MOG或其自身免疫原性片段的生物缀合物,在对MOG或其自身免疫原性片段特异的CD4+T细胞中诱导免疫致耐受性作用的方法。本领域已知若干自身免疫原性MOG片段,包含对应于小鼠MOG氨基酸1-22、35-55和64-96的肽(参见如,美国专利公开2009/0053249,通过引用以其整体并入本文)。在一个实施方案中,MOG自身免疫原性片段是肽MEVGWYRSPFSRV-VHLYRNGK(小鼠MOG35-55;SEQ ID NO:1)。
在其他方面,本发明提供通过在胰腺β细胞特异性的CD4+T细胞中诱导免疫耐受,预防胰腺β细胞(在一个实施方案中,其为I型糖尿病的基础)自身免疫破坏的方法。在一个实施方案中,本发明提供通过施用包含硫酸酯化多糖、TGFβ1和异基因(allogeneic)或同基因(syngeneic)的β细胞的生物缀合物,在胰腺β细胞特异性的CD4+T细胞中诱导免疫致耐受性作用的方法。
在又一个方面,本发明提供通过在皮肤(dermal)CD4+T细胞中诱导免疫耐受来抑制与异常角质形成细胞生长(其为银屑病的基础)相关的自身免疫炎症信号传导的方法。在一个实施方案中,本发明提供了经皮内施用包含硫酸多糖、TGFβ1和细胞制备物或代表组织的分子制备物(例如从皮肤活组织检查获得的材料)的生物缀合物,在皮肤CD4+T细胞中诱导免疫致耐受性作用的方法。
本文所用术语“生物活性多肽”是指在体内表现出多种药理活性的多肽,并且包括但不限于生长因子、细胞因子、趋化因子、血管生成因子、免疫调节剂、激素等。在本申请中,术语“多肽”和“蛋白质”可互换使用。所述至少一种生物活性多肽可以是带正电荷的多肽和/或肝素结合多肽。
在一个实施方案中,所述生物活性多肽是TGFβ。本发明包括所有已知的TGF同工型(isofom)(TGFβ1至TGFβ5;哺乳动物的TGFβ1-3,在鸡中发现的TGFβ4;和青蛙中发现的TGFβ5),以及它们的片段、突变体、同源物、类似物和等位基因变体。在一个实施方案中,TGFβ是哺乳动物TGFβ。在一个实施方案中,TGFβ是TGFβ1。在另一实施方案中,TGFβ是TGFβ2。在另一实施方案中,TGFβ是TGFβ3。在另一实施方案中,TGFβ是人类TGFβ1(Genbank登录号X02812)或小鼠TGFβ1(Genbank登录号AJ00986)。

因此,在一个实施方案中,本发明组合物和方法的任何生物活性多肽,无论是所述第一生物活性多肽还是额外的生物活性多肽,还是第二或第三生物活性多肽,为AT III、TPO、SLPl、C1INH、VCP、FGF、FGF受体、VEGF、HGF、IGF、PDGF、BMP、EGF、CXCL4、SDF-1、IL-6、IL-8、RANTES、MCP-1、MIP-1、淋巴细胞趋化因子、fractalkine、膜联蛋白、ApoE、HIV-1外壳蛋白gpl20、CypA、Tat蛋白、单纯疱疹病毒HSV的病毒外壳糖蛋白gC、gB或gD、登革病毒的包膜蛋白、恶性疟原虫的CS蛋白、细菌表面粘附蛋白OpaA、1-选择蛋白、P-选择蛋白、HB-GAM、TSR、MOG或AP,或其组合。
在另一个实施方案中,所述生物活性多肽是PDGF-BB、PDGF-AA、bFGF、aFGF、VEGF、IL-6、TPO、SDF-1、HGF、EGF、MOG、BMP-2、BMP-4、BMP-7、IGF或其组合。另一个实施方案中,用于本发明方法中的组合物包括TGF-β、VEGF和PDGF-ββ作为生物活性多肽。
在本发明的一些实施方案中,所述至少一种肝素结合多肽选自PDGF-β、PDGF-α、bFGF、aFGF、VEGF、TGFβ1、IL-6、TPO、SDF-I、HGF、EGF、BMP、或IGF。在本发明的其它实施方案中,所述至少一种生物活性多肽是血管生成生长因子或呈现血管生成活性的生长因子,例如TGF-β1、VEGF、bFGF、aFGF、PDGF-β、IGF及其组合。

根据本发明,形成所述生物缀合物的所述硫酸酯化多糖可以包含不同的重复单糖单元,可以是不同的长度,并且可以具有连接所述单元的不同类型的键。所述硫酸酯化多糖是线性的例如硫酸酯化纤维素、分支的例如硫酸酯化糖原、并且长度可以变化;例如,其可以小到硫酸酯化四糖或三糖。合适的硫酸酯化多糖是同多糖,包含但不限于淀粉、糖原、纤维素、壳聚糖或壳多糖,或者是杂多糖,包含但不限于藻酸盐(alginic acid(alginate)salts)和透明质酸。
根据本发明和实施方式,所述硫酸酯化多糖包括糖醛酸残基,例如D-葡萄糖醛酸、D-半乳糖醛酸、D-甘露糖醛酸、L-艾杜糖醛酸、和L-古罗糖醛酸。包括糖醛酸残基的多糖的实例包含但不限于藻酸盐(在一个实施方案中,藻酸钠)、果胶、树胶和来自植物源的粘质;和来自动物源的粘多糖(GAG),包括透明质酸(hyaluronic acid或hyaluronan)。所述包含糖醛酸的硫酸酯化多糖可以是化学硫酸酯化的或者可以是天然硫酸酯化的多糖。
在一个实施方案中,所述硫酸酯化多糖是藻酸硫酸酯。在另一实施方案中,所述硫酸酯化多糖是透明质酸硫酸酯。
藻酸是一种获自褐藻和海藻的线性多糖,并且由β-l,4-连接的葡萄糖醛酸和甘露糖醛酸单元组成。如本文所用,术语“藻酸”是指来源于海藻(如海带(Laminariahyperborea)、L.掌状海带(L.digitata)、兴红藻(Eclonia maxima)、巨藻(Macrocystispyrifera)、淡黑巨海藻(Lessonia nigrescens)、泡叶藻(Ascophyllum codosum)、(L.japonica)、海茸(Durvillaea antarctica)、和海洋巨藻(D.potatorum))的聚阴离子多糖共聚物,并且其包括不同比例的β-D-甘露糖醛酸(M)和α-L-古罗糖醛酸(G)残基。
合适用于本发明的藻酸,在一个实施方案中,具有范围在1:1到3:1之间的α-L-古罗糖醛酸与β-D-甘露糖醛酸之间的比例,在一个实施方案中,在1.5:1和2.5:1之间,在一个实施方案中,约为2,且具有范围在1到300kDa之间的分子量,在一个实施方案中,在5到200kDa之间,在一个实施方案中,在10到100kDa之间,在一个实施方案中,在20到50kDa之间。
藻酸在二价阳离子例如Ca2+和Ba2+的存在下发生凝胶化。在制药/医药领域,其被成功地用作封装材料,主要用于细胞(细菌、植物和哺乳动物细胞)。对于分子来说,其效果差得多,并且甚至250kDa大小的大分子快速从藻酸水凝胶系统中释放出来。特别是,感兴趣的生物分子,例如细胞因子、生长因子,其大小在5到100kDa之间,被迅速释放。
透明质酸包含葡萄糖醛酸和N-乙酰氨基葡萄糖的重复二聚体单元,并形成细胞外基质中发现的核心复合蛋白多糖聚集体。
先前已经表明,硫酸酯化多糖,赋予它们能够结合和控制释放重要信号蛋白如各种细胞因子和生长因子的特性。当形成生物缀合物时,藻酸硫酸酯和透明质酸硫酸酯均被发现模拟硫酸乙酰肝素和肝素的生物学特性(参见例如WO2007/043050,其全部内容通过引用并入本文)。
本领域技术人员所理解的,所述生物活性多肽和所述硫酸酯化多糖之间的所述结合选自可逆的非共价相连,包括离子键、静电相互作用、疏水相互作用、氢键或范德华力。
应该理解,通过具有正电荷,所述生物活性多肽可以可逆地和非共价地结合到所述硫酸酯化多糖上,所述硫酸酯化多糖由于其硫基而带有负电荷。
因此,本发明还提供用于至少一种生物活性多肽的持续局部释放的方法。
根据本发明的方法,生物缀合物可以注射至或植入人体的任何部位,并用作所述生物活性多肽的递送系统。在一个实施方案中,本发明的所述生物缀合物可以是可流动凝胶的形式。在另一个实施方案中,生物缀合物可以预先配为刚性可植入支架。在又一实施方案中,所述可植入支架可以进一步含有支持基质。
所述基质用作生物缀合物的支持物或载体,并可以由颗粒或多孔材料制成。所述基质材料可以是柔性的并且可以适当地固定防止其迁移到非预期的位置。所述聚合物基质材料可以是天然的或合成的,并且包含但不限于合成聚合物,例如聚乙二醇(聚氧化乙烯)、聚乙烯醇、聚乳酸、聚乙醇酸和聚羟基丁酸酯,或者天然聚合物,如胶原蛋白、纤维蛋白和明胶,或者多糖,例如壳聚糖和藻酸。
所述基质可以是适于递送模式的任何形式,例如水凝胶、珠、微球(微珠)、水凝胶微囊、海绵、支架、泡沫、胶体分散体、纳米颗粒、悬浮液等。因此,基于硫酸酯化多糖和生物活性肽的生物缀合物的缓释剂型可以被做成液体、网状物、海绵、纤维和水凝胶的形式。
本文使用的术语“水凝胶”是指能够含水的天然或合成亲水聚合物链的网络。能够形成这种网络的化合物的例子是藻酸、部分钙交联的藻酸溶液、壳聚糖和粘性透明质酸。
本文使用的术语“生物缀合物”是指共价或非共价结合到生物活性多肽上的硫酸酯化多糖。非共价结合的例子是涉及离子键、静电相互作用、疏水相互作用、氢键或范德华力的结合。
术语“支架”是指包含空隙的任何合成或有机结构。这种支架的非限制性例子是模具(mold)、铸模(cast)和哺乳动物的受损组织中的空隙。
本发明的组合物可以通过任何合适的方法施用,例如但不限于,肝内、皮内、透皮(如在缓释制剂中)、肌肉内、腹膜内、静脉内、冠状动脉内、皮下、口服、硬膜外、外用、和鼻内途径。施用还包括手术施用、植入、插入、或注射植入物(或其部分)至对象中。所述植入物(或部分)位于皮下、肌肉内,或者位于允许植入物实现其预期功能的另一身体位置。通常,植入物(或部分)在包含但不限于对象上臂、背部或腹部的部位通过皮下植入施用。医学专业人员容易地确定其它合适的施用部位。施用多个植入物或部分以实现用于治疗的预期的剂量。也能使用其它治疗有效途径施用,施用还包括对对象全身施用或局部施用本文所述的组合物。
术语“对象”包含但不限于人类。在另一个实施方案中,所述对象是鼠,在一个实施方案中是小鼠,在另一个实施方案中是大鼠。在另一个实施方案中,所述对象是狗、猫、牛、绵羊或猪。在另一个实施方案中,所述对象是哺乳动物。
制备硫酸酯化多糖生物缀合物的方法是众所周知的(参见如美国专利公开号2015/0051148,其全部内容通过引用并入本文),并且在下文中进一步详述。
本发明考虑硫酸酯化和未硫酸酯化多糖的混合物,例如藻酸和藻酸硫酸酯。根据本发明,硫酸酯化多糖的比例范围可以是总多糖以重量计的约1%至约40%,在一个实施方案中,为总多糖以重量计的约3%至约30%,在一个实施方案中,为总多糖以重量计的约4%至约20%,在一个实施方案中,为总多糖以重量计的约5%至约10%。可选地,上述比例代表质量百分比。在一个可选的实施方案中,这些生物缀合物的结合和释放通过多糖硫酸酯化程度和并入递送系统的硫酸酯化多糖硫酸酯的范围来控制。

免疫耐受反应的例子包括但不限于,同种异体移植成功、同种异体移植排斥的缺乏、自身免疫病症的抑制、对同种异体细胞移植的免疫反应的抑制、同种异体细胞凋亡的抑制、同种异体细胞存活的增加、同种异体细胞移植物的血管形成的刺激、所述生物活性多肽的延长呈递、炎症信号传导的抑制、树突状细胞成熟的抑制、CD8+T细胞细胞毒性反应的抑制、和调节性T细胞分化的刺激。

原料和方法
化学品

动物和细胞系
C57/B16小鼠和CD1lcdnR转基因小鼠购自Jackson Laboratory(Maine,美国)。Ben-Gurion大学(Negev,以色列)的Eli Lewis博士慷慨捐赠OVA特异性、MHC类Il-限制性αβT细胞受体(OTII)转基因小鼠。所有的小鼠都被安置和饲养在Ben-Gurion大学医学中心(Negev,以色列)的动物设施中,并将其维持被在具有中有高压灭菌的寝具、食物和水的高压灭菌笼中。

藻酸基质的制备
如上所述,合成具有共价结合的粘附肽G4RGDY和肝素结合肽G4SPPRRARVTY(RGD/HBP)的藻酸,并根据Freeman等人2009(Biomaterials.2009;30(2122-31))和Freeman等人2008(Biomaterials.2008;29(3260-8))合成藻酸-硫酸酯,。通过冷冻干燥技术装配大孔支架。简而言之,1.2%(w/v)藻酸钠溶液与1.32%(w/v)D-葡萄糖酸/半钙盐通过均质化溶液而交联,以获得均匀的钙离子分布。藻酸和交联剂在交联溶液中的最终组分浓度分别为1.0%和0.22%(w/v)。96-孔板的每个孔中倒入50微升交联的藻酸溶液,冷却至4℃,在(-20)℃冷冻24小时,并在0.08bar和(-57)℃下冷冻干燥48小时。支架的灭菌是通过在生物罩中暴露于紫外线(UV)下1小时实现。
TGF-β生物活性的体外释放研究与评价
在两组支架上试验afTGF-β的释放研究:一种是原始藻酸支架,一种是藻酸/藻酸-硫酸酯(Alg/AlgS)支架(也就是,90%纯藻酸和10%藻酸硫酸酯)。载有TGF-β的支架在500μL培养基中(高糖DMEM补充100U/mL青霉素、0.1mg/mL链霉素、0.1mg/mL新霉素、和0.1%牛血清白蛋白(BSA)(w/v))在无菌条件下,在旋转振荡器上的48孔板中,在37℃下7天。根据制造商说明书,在支架负载TGF-β后的0、1、3和7天收集培养基样品,并用抗人重组TGF-βELISA试剂盒分析释放的TGF-β。为了定量在上述时间点残留的afTGF-β,取支架样品,并在乙二胺四乙酸(EDTA)中溶解支架。使溶液pH=3以将afTGF-β从藻酸链上解离,并离心以实现相分离。在ELISA之前,使溶液的pH值恢复到pH=7。在每个时间点计算afTGF-β的百分比为(TGF-β7/初始输入TGF-β量的量)x100。
总脾细胞的分离和培养
处死动物,并取出其脾脏,匀浆,经70μm筛网过滤,以获得单细胞悬浮液。用300μlACK缓冲液裂解红细胞,并计数剩余细胞。细胞在克隆培养基(补充有10%胎牛血清、10mMHEPES、1mM丙酮酸钠、10mM非必需氨基酸、1%青霉素/链霉素和50μΜβ-巯基乙醇的DMEM)中培养。
CD4+和CD8+T细胞的磁性分离
根据制造商说明书,接受全脾细胞的单细胞悬浮液后,使用EasySepTMMagnet(STEMCELL Technologies,Vancouver,加拿大),通过磁珠阴性选择试剂盒分离CD4+T细胞或CD8+T细胞。纯化的T细胞被针对CD4或CD8染色,用于如下描述的流式细胞术,并通过流式细胞术来分析以评估纯化。
骨髓衍生的树突状细胞的纯化和培养
树突状细胞(DC)是根据从以前的研究中经过几处修改的方法,通过用粒细胞-巨噬细胞集落刺激因子(GM-CSF)培养骨髓衍生的前体而产生的。第0天,4-12周龄C57/B16小鼠的股骨和胫骨用纱布垫从周围组织中取出并纯化。此后,骨头放入冷PBS中,用剪刀去除它们的尖端,并通过用27G针头以冷PBS冲洗骨髓。所得悬浮液通过70μm筛网过滤,并用100μl ACK缓冲液裂解红细胞1分钟。以HBSS洗涤所述细胞悬浮液两次,在6℃以150g离心6分钟,并再悬浮于DC培养基中[补充有10%FBS、2mM L-谷氨酰胺、青霉素(100U/ml)、链霉素(1μg/ml)、制霉菌素(2.5U/ml)、50μΜβ-巯基乙醇、和20%来自J554细胞的GM-CSF上清液的IMDM(如下详述)]。细胞以5x106细胞/皿于10ml培养基中接种于细菌90mm培养皿上,并培养8天。以5%CO2在37℃的加湿培养箱内培养细胞。DC培养基和GM-CSF在第3天和第5天被替换,并准备在第8天用于进一步的实验。
含有GM-CSF的上清液的制备
Angel Porgador教授(Ben-Gurion大学(Negev,以色列))捐赠用鼠类GM-CSF构建体转染的J558细胞。通过在1mg/ml G-418存在下培养两周来选择转染的细胞,并在组织培养瓶中以~2x105细胞/ml的密度接种。然后细胞在培养基中每2-3天稀释1:5,持续3周。上清液用0.2μm过滤器过滤并储存在(-80℃)下。
基质内细胞的接种和活化:体外培养
通过将14μL细胞悬浮液(有或没有TGF-β)滴到干燥的藻酸/藻酸-硫酸酯基质上来接种细胞(0.5x106总细胞/基质)。然后细胞构建体以500rpm离心30s,并在37℃和5%CO2下培养10分钟,以在基质中使得细胞分布。然后,逐渐加入额外的200μL培养基,并孵育细胞装置2小时。
为了细胞活化,细胞构建体转移到48孔板中,并补充1ml培养基(加入或不加入OVA多肽(最终肽浓度:20mg/rnl))。在37℃含5%CO2的加湿培养箱中进行细胞培养。
IL-10抑制
纯化的抗小鼠IL-10(Biolegend;目录号505012)作为IL-10抑制剂以最终浓度为1ng/ml(比培养物中IL-10的最大浓度高一个数量级)加入。夹心ELISA显示培养物中没有IL-10的痕迹。
NIH 3T3成纤维细胞的逆转录病毒感染
PLAT-E细胞维持在含有10%FBS和0.5%青霉素、链霉素、杀稻瘟菌素、和嘌呤霉素的DMEM中。PLAT-E包装细胞以每100mm培养皿8×106个细胞铺板并孵育过夜。第二天,以PolyJet体外转染试剂(SignaGen,MD,美国)以pMX-IRES-GFP载体转染细胞。转染后24小时,培养基以新培养基替换并转染后24小时和48小时收集。
在转导前1天接种3T3/NIH成纤维细胞,每12孔板接种105个细胞。含病毒的上清液(24小时后收集)补充2μg/ml硫酸鱼精蛋白,转移到成纤维细胞培养皿中,并培养过夜。转染后24小时,以48小时上清液替换含病毒培养基。成纤维细胞经历三个转染周期。
在基质中接种NIH/3T3成纤维细胞:体内
感染表达GFP的NIH/3T3成纤维细胞(如上所述)通过30μL细胞悬浮液(也就是,补充或不补充200ng TGF-β的DMEM中的细胞)滴到干燥的共价结合的RGD/HBP藻酸/藻酸-硫酸酯基质上接种(0.5x106总细胞/基质)。我们发现30μL的细胞悬浮液是润湿整个支架而不留下进入液的合适体积,因此确保所有生长因子最大限度地结合到支架上。在移植构建体之前,基质在37℃和5%CO2下孵育5分钟以使得细胞在支架内分布。
移植装置
将细胞接种到基质后,立即将细胞移植装置移植到C57/B16野生型小鼠的左肾囊下,用异氟烷麻醉机麻醉。在移植后不同的天数(10、15或30天),细胞移植装置随着周围的纤维化一起被手术切除。免疫组织化学分析了它们的血管形成,流式细胞术分析了透过的淋巴细胞的数量和成纤维细胞的存活情况。在移植后的第15天和第30天,脾脏也被切除用于全淋巴细胞培养和CD8+细胞毒性测定。
成纤维细胞裂解物
NIH/3T3成纤维细胞(107个细胞)经历五个循环的冻融、离心,并用Bio-Rad蛋白检测法(Bio-Rad,以色列)测定其总蛋白浓度。
用成纤维细胞裂解物对总脾细胞活化
移植后15天和30天,总脾细胞在96孔U平板中培养(每孔2x104个细胞),并以成纤维细胞裂解物活化(最终蛋白质浓度:10mg/ml)。
CD8+细胞毒性检测
如上所述,CD8+T细胞在移植后15天和30天从整个脾细胞中磁性分离。CD8+T细胞和NIH3T3成纤维细胞以4:1的比例共同培养。共同培养2小时和12小时后,以FACS分析CD8+T细胞的细胞毒性和成纤维细胞的活力。
蛋白质印迹
以5ng/ml TGF-β处理NIH/3T3成纤维细胞40分钟,并通过使用补充有蛋白酶抑制剂混合物和磷酸酶抑制剂的放射免疫沉淀分析(RIP A)缓冲液[150mM NaCl、1mM EGTA、50mM Tris-HCl(pH=7.5)、1%NP-40]获得细胞裂解物。细胞以RIPA在冰上孵育30分钟,并然后以13,000rpm离心。收集上清液,以Bio-Rad蛋白测定法测定蛋白质浓度,并将30μg/孔蛋白用于SDS-PAGE,并然后转移到硝酸纤维素膜上。在室温下,以补充有0.5%聚山梨醇酯20(TWEEN20)(TBST)的Tris缓冲盐水中的5%BSA(MP Biochemicals,Santa Ana,Ca)或5%牛奶封闭膜1小时,并然后在4℃与一抗在室温孵育过夜。通过使用以下一抗检测蛋白质:兔抗-磷酸-SMAD2,兔抗-SMAD2/3(均来自Cell Signaling,Danvers,MA)、和小鼠抗-肌动蛋白(MP Biochemicals,Santa Ana,Ca)。辣根过氧化物酶(HRP)缀合二抗是山羊抗小鼠IgG(Jackson labs,Sacramento,CA)和驴抗兔IgG(Amersham,GE healthcare,英国)。使用Supersignal West Pico Chemiluminescent Substrate试剂盒(Thermo Scientific,Waltham,MA)诱导化学发光,然后对其以ImageQuant LAS4000软件(GE Healthcare LifeSciences,英国)检测。以ImageJ软件量化谱带强度。
流式细胞术分析
对于体外研究后的基质分析,首先溶解基质,并然后通过离心获得细胞,用HBSS洗涤,再次离心,并悬浮在FACS缓冲液(PBS中2%的FBS)中。为了分析体内移植后的细胞移植装置,均质化基质和周围纤维化并经70μm筛网过滤以获得单细胞悬浮液,并离心细胞并重悬于FACS缓冲液(PBS中2%的FBS)。
获得单细胞悬浮液后,加入Fc-阻断剂5分钟以防止非特异性抗原结合。此后,根据制造商说明,加入荧光抗体,并在冰上孵育平板15分钟。然后以FACS缓冲液洗涤样品,并在6℃下在420g下离心两次3分钟。
对于细胞内FACS染色(Foxp3),首先以荧光抗体染色细胞,然后在固定缓冲液中孵育20分钟,洗涤两次,并在透化缓冲液中孵育15分钟。此后,用FACS缓冲液洗涤细胞,并以细胞内抗体孵育30分钟,离心,并悬浮在FACS缓冲液中。使用利用FlowJo软件的GalliosTMFlowCytometer(BeckmanCoulter,Inc.,Brea,CA)用于流式细胞术分析。
本研究中使用的所有抗体都是发光团缀合的抗体,并包括APC-Cy7缀合的抗CD4(Biolegend;目录号100414)、PerCP-缀合的抗-CD8(Biolegend;目录号100732)、PE-缀合的抗CD25(Biolegend;目录号101904)、PE-Cy7-缀合的抗-CD69(Biolegend;目录号104512)、APC-缀合的抗-CD107a(Biolegend;目录号121614)、FITC-缀合的抗-FoxP3(Biolegend;目录号137214),PE/Dazzle-缀合的抗-CDllc(Biolegend;目录号117348)、抗-CD86BrilliantViolet(Biolegend;目录号305431)、APC-缀合的Annexin V(Biolegend;目录号640920)、和PI(Biolegend;目录号421301).
用ELISA分析细胞因子分泌
通过使用OVA多肽或成纤维细胞裂解物活化后,从体外和离体研究中获得无细胞上清液。对于IL-2测量,在细胞活化后24小时收集上清液。对于IFN-γ和对于IL-10和IL-17A测量,细胞活化后分别48小时和72小时收集上清液。根据制造商说明,执行夹心ELISA用于测量上清液中细胞因子的浓度。
免疫组织化学和共聚焦成像分析
对于体内血管生成分析,从动物体内取出细胞移植装置和周围的纤维化,并在4%多聚甲醛(PFA)中固定24小时,并然后在蔗糖溶液(30%)中固定48小时。然后,在最佳切割温度化合物(OCT)中包埋这些装置,并储存在(-80)℃。用低温恒温器(Lecia CM3050S)切割水平横截面(20μm厚)并在使用前保持在(-80)℃下。在PBS/TWEEN(0.05%v/v)中洗涤横截面。染色前,使用第一抗体稀释缓冲液阻断非特异性结合。然后切片与第一单克隆抗小鼠CD31抗体一起孵育(Bio legend;目录号910003),并以Alexa Flour 633-缀合的抗大鼠IgG(Biolegend;目录号405416)染色。
以NikonC1si激光扫描共聚焦显微镜进行切片的共聚焦成像。血管密度计算为血小板内皮细胞粘附分子(PECAM)的百分比、总图像面积中CD31占据面积的百分比,并通过使用Imaris软件(Bitplane scientific solutions,South Windsor,CT)从PECAM免疫染色的横截面切片中随机选择的15个不同视野中确定。
对于CD8+细胞毒性成像,在与NIH/3T3成纤维细胞共培养之前,按照制造商说明,用SNARF1(Invitrogen,CA,美国)染色CD8+T细胞。共培养12小时后,用4%PFA固定孔中细胞。粒酶B染色前,用原抗体稀释缓冲液阻断非特异性结合,并然后细胞经历渗透。然后,以荧光缀合第一单克隆抗小鼠粒酶B(美国Biolegend;目录号515405)孵育细胞过夜。以NikonC1si激光扫描共聚焦显微镜完成对进行CD8+细胞毒性试验的切片和孔的共焦成像。
对于激活成像后72小时支架内的体外脾细胞培养,收集支架,并在DMEM缓冲液中用4%PFA固定20分钟,之后在蔗糖溶液中孵育24h。固定后,在O.C.T中包埋装置,并储存在(-80)℃。以低温恒温器(Lecia CM3050S)切割水平横截面(30μm厚),并在使用前保持在(-80)℃。在DMEM缓冲液中洗涤横截面。染色前,用DMEM缓冲液中的2%BSA阻断非特异性结合。然后以第一单克隆抗小鼠CDllc抗体(Biolegend,目录号117302))和以第一单克隆抗小鼠CD4抗体(Biolegend,目录号100402)孵育切片,并且以AlexaFlour 488缀合的抗鼠IgG抗体(Biolegend,目录号405418))和以Alexa Flour 546抗亚美尼亚仓鼠IgG抗体(Biolegend,目录号405423)染色。
统计学
使用GraphPad Prism version 5.02 for Windows(GraphPad Software,SanDiego,CA)完成所有的统计分析。所有变量表示为均值±SEM。体外研究通过双侧不配对t检验(p<0.05=比较。所有来自体外的实验通过单因素ANOVA用Tukey事后检验进行比较。

实施例1
细胞移植装置的构建及免疫调节特性
细胞移植装置是由于其大孔结构,在移植后已显示有效的血管形成的藻酸支架(平均孔径为80μm)。为加强血管形成,所述藻酸支架补充亲和结合血管内皮生长因子(VEGF)和血小板源生长因子(PDGF-ββ)。所述生长因子与藻酸基质的亲和结合是通过它们与构成所述支架干重的10%的藻酸-硫酸酯的相互作用介导的。我们之前表明,肝素结合蛋白家族的生长因子,例如VEGF、PDGF-ββ和TGF-β,以类似于它们与肝素/硫酸乙酰肝素的天然相互作用的方式与藻酸-硫酸酯相互作用(图1A)。因此,我们期望所述生长因子与所述藻酸支架基质的所述亲和结合将改善其生物学功能。
在此期望基础上,我们通过亲和结合TGF-β到藻酸硫酸酯或藻酸支架上产生免疫耐受微环境(图1A,详见材料和方法)。与当TGF-β非特异性吸附于原始藻酸支架相比,该过程延长TGF-β的呈递。TGF-βELISA揭示,在含藻酸硫酸酯支架中7天后,30.2%±0.8%的初始负载afTGF-β仍呈递在基质中,然而仅5.2%±0.3%保留在原始支架中(图IB)。重要的是,在培养基中发现的释放的TGF-β和与藻酸或硫酸酯化藻酸基质(afTGF-β)亲和结合的TGF-β相似地激活成纤维细胞单层中的SMAD2磷酸化(图1C,D)。值得注意的是,afTGF-β在其仍与基质亲和结合时和与支架解离后显示出相似的活性,指示细胞与TGF-β在其亲和结合形态和其释放时都相互作用。
接下来,我们检查藻酸-硫酸酯或藻酸支架(afTGF-β构建体)中afTGF-β的空间呈递是否会产生免疫调节微环境。我们进行初步研究以测试总脾细胞白细胞能否在支架内培养、维持存活、和诱出抗原特异性T细胞反应。为此,在作为T细胞激活肽的OVA存在或不存在时,从OTII OVA-TCR Tg小鼠(其淋巴细胞是OVA特异性的)中分离出剔除红细胞的脾白细胞并在支架中接种(图2A)。图13A显示典型的构建体,在细胞接种后3天,具有彻底分布支架的白细胞并具有紧密邻近的DC和CD4T细胞,允许细胞-细胞相互作用。验证OVA诱导的T细胞增殖,揭示32%和6%的T细胞分别在OVA激活和未激活的情况下存活(图13B)。在OVA激活的构建体中大约有20%的存活淋巴细胞是CD4+CD25+(图14A,B)。
afTGF-β的免疫调节作用表现为细胞因子谱和脾细胞中CD4+CD25+Foxp3+调节性T细胞(Treg)的出现(图2B-E),以及在在afTGF-β构建体内接种的纯化DC中(图15A-E)显现。如图15B-E所示,afTGF-β显著抑制促炎细胞因子IL-17A的分泌,显著增加调节性细胞因子IL-10,并且不影响分泌的IL-2或IFN-γ的水平。重要的是,所述装置中afTGF-β的存在导致CD4+CD25+Foxp3+T细胞在CD4+T-细胞群体中的频率增加2.88倍(含有或不含有OVA的总T细胞群体的分别30.6%±3.4%及11.8%±3.1%;p=0.008,图3,图4A-C)。总之,这一发现表明,afTGF-β促进所述细胞移植装置内的抗炎环境。值得注意的是,在抗IL-10存在时(图5A-F)或在CD1lc启动子下使用来自CD1lcdnrTg小鼠脾细胞表达人类TGF-β受体Ⅱ基因的显性阴性形式(图5G-H,图6A-6E)时,未观察到在afTGF-β构建体中观察到的降低的分泌的IL-17水平和增加的Treg频率。
总的来说,我们的数据指出借助DC中的afTGF-β信号转导诱导IL-10上调的机制,其然后促成Treg分化,并减弱CD4T细胞的效应子功能。这种机制在移植装置的三维(3D)微环境中似乎比补充可溶性TGF-β的2D细胞单层时明显更为突出(图16A-E,图17A-B)。
实施例2
装置的体内血管形成
移植后细胞移植装置的血管形成对支持移植的同种异体细胞的存活能力和功能至关重要,特别是在高密度接种的细胞装置中。为增强血管形成,我们的装置补充以VEGF和PDGF-ββ。为了了解afTGF-β构建体中细胞行为和存活能力的差异(与TGF-β缺乏构建体相比)是由于血管形成程度的不同的可能性,我们比较afTGF-β构建体和TGF-β缺乏构建体的血管形成程度;两种构建体以被感染以表达GFP瑞士鼠源同基因的NIH或3T3成纤维细胞接种(5xl05细胞或支架)。
图7A显示,在肾包膜下移植后10天内,两种细胞接种的装置上均产生PECAM+(CD31)内皮细胞的群体,形成散布在GFP成纤维细胞中的毛细血管。第15天,在移植的构建体中观察到清晰的毛细管(图7B,C),占据含有和不含TGF-β的细胞装置的3-4%(图8)。
实施例3
体内局部免疫调节与同种异体成纤维细胞移植存活
接下来的一系列实验证实,在体内,在afTGF-β支架中局部免疫调节微环境的建立。此外,鉴于所述装置广泛的血管形成,检查这种潜在的移植装置维持移植的同种异体成纤维细胞的存活能力和功能的功效。
移植后10天和15天取回的构建体的FACS分析揭示,相比TGF-β缺乏构建体,在afTGF-β构建体中,表达共刺激分子CD86的成熟CDllc+DC的比例显著更低(图9A-C),且CD4+CD25+Foxp3+Treg的比例显著更高(图9D-F,p<0.05)。Treg的增加仅在移植后第10天才明显。因此,在移植后第10天,与缺乏TGF-β的构建体相比,afTGF-β构建体中CD8+T细胞的百分比显著更低(分别是2.1%±0.4%对比5.5%±0.9%,p=0.019)(图9G-I)。此外,在移植后10天(分别是13.4%±3.5%对比22.9%±9.3%,p=0.015)和15天(分别是4%±0.8%对比12.0%±2.5%,p=0.0001),与缺乏TGF-β的构建体相比,afTGF-β构建体中观察到表达活化标记物CD69的CD8 T细胞比例显著减少(图9J-L)。
为了确定免疫耐受环境是否影响同种异体成纤维细胞的存活,我们确定存活的同种异体成纤维细胞的数量和经历凋亡的细胞的频率。移植后3天从装置中取回的GFP+细胞计算为来自装置中存在的总GFP+细胞的GFP+细胞百分比(图17C、D)。选择这一基准以消除与T细胞介导的细胞毒性无关的细胞死亡。移植后15天取回的同种异体成纤维细胞百分比在afTGF-β和TGF-β缺乏构建体中分别为50.4%±2%和44.6%±0.7%(p<0.05,图10A、B),伴随在移植后30天观察到从afTGF-β装置回收的成纤维细胞的更高比例的趋势。通过以AnnexinV和PI标记的GFP细胞的FACS分析,进一步验证存活的同种异体成纤维细胞的存活能力(图10C)。在移植后15天,afTGF-β构建体中存活的同种异体成纤维细胞的比例(GFP+Annexin-PI-)显著高于TGF-β缺乏的构建体(分别为38.5%±3.5%及23.2%±5.0%,p=0.038,图10D)。因此,在移植后15天和30天,在afTGF-β构建体中来自GFP+同种异体成纤维细胞的总数的GFP+AnnexinV+PI-凋亡细胞和GFP+AnnexinV+PI+细胞的百分比明显低于在TGF-β缺乏的构建体中(图10E、F)。这些数据指示,构建体中afTGF-β的局部呈递在DC未成熟状态下维持其浸润,从而增加Treg的频率并抑制细胞毒性CD8 T细胞的活性。
实施例4
亲和结合的TGF-β抑制同种异体细胞特异性外周T细胞的活化
细胞或组织移植中一个主要障碍在于免疫抑制性药物不是抗原特异性的,并因此损害宿主的防御机制。此外,停药通常导致免疫反应的反弹,最终导致移植物排斥。通过诱导同种异体移植物特异性T细胞耐受,实现在不损害宿主免疫下移植物的长期存活。为了确定血管形成的afTGF-β构建体能否诱导同种异体成纤维细胞特异性的外周耐受,在移植后第15天和第30天从小鼠分离脾细胞,并检查同种异体成纤维细胞特异性T-辅助因子和T-细胞毒性反应。与TGF-β缺乏构建体的移植相比,在第15天,切自afTGF-β构建体移植的小鼠的脾细胞具有显著降低的同种异体成纤维细胞特异性Thl7效应子功能(图11A;分泌的IL-17为与切自以TGF-β缺乏构建体移植的小鼠的脾细胞相比的55.5%,p<0.05)。值得注意的是,这些脾细胞在移植后第30天也显示减弱的Thl和/或CD8 T细胞效应子功能(图11B-C;分泌的IL-2和IFN-γ分别为与切自以TGF-β缺乏构建体移植的小鼠的脾细胞相比的45.2%和20.9%,p<0.05)。各组间IL-10分泌水平无显著性差异(图11D)。
接下来,我们确定以afTGF-β构建体移植的小鼠中同种异体特异性CD8 T细胞细胞毒性反应是否也被抑制。因此从移植的小鼠和从野生型小鼠中分离脾细胞,并然后分离CD8T细胞(如“方法”和图17E所述),分别以1:4的细胞比例与同种异体成纤维细胞共培养。共聚焦成像显示,从以TGF-β缺乏构建体移植的小鼠分离的CD8+T细胞与同种异体成纤维细胞形成免疫突触并发挥细胞毒性(图HE和图18)。
CD8 T细胞与同种异体成纤维细胞共培养2小时和12小时后表达CD107的细胞毒性CD8 T细胞的频率的FACS评估指示,在两个时间点,细胞毒性CD8+CD107+T细胞的百分比在来自移植afTGF-β构建体的小鼠的脾细胞中显著低于在来自移植TGF-β缺乏构建体的小鼠的脾细胞中。
此外,与以TGF-β缺乏构建体移植的小鼠相比,与从以afTGF-β构建体移植的小鼠分离的脾衍生CD8 T细胞共培养时的经历凋亡的同种异体成纤维显著减少(分别为35.5%±5.1%对比7.2%±0.9%,第15天;图12B,C)。值得注意的是,获自以afTGF-β移植的小鼠的活化CD8 T细胞的频率和细胞毒性与野生小鼠的相似,指示在TGF-β构建体的条件下的同种异体成纤维细胞的移植引起很轻微的细胞毒性反应。
我们检验在细胞移植装置中与基质亲和结合的TGF-β的局部呈递促进免疫调节微环境,其保护细胞移植物不被排斥的假设。我们显示,afTGF-β通过维持DC在不成熟的表型以及通过增加Treg的频率和以IL-10依赖的方式降低CD4和CD8细胞毒性T细胞的效应子功能来发挥其调节功能。重要的是,我们显示afTGF-β的局部免疫调节作用投射到脾脏,其显示同种异体成纤维细胞特异性CD4和CD8 T细胞的显著降低的效应子功能。
TGF-β是一种重要的免疫调节细胞因子,其依赖于相邻的细胞因子环境驱动抗炎和促炎反应两者。当与藻酸支架亲和结合时,呈递的TGF-β能否调节活化的效应T细胞尚不清楚。我们的体外结果指示,afTGF-β当DC或当总脾细胞用作抗原呈递细胞(APC)时(后者至更大程度地),抑制CD4T细胞的活化,并且其增加分泌的IL-10的水平和Treg的频率。以抗IL-10或以CDllc-DNR脾细胞体外完成阻断实验,暗示afTGF-β在两种途径中具有调节功能:(1)增强对DC的IL-10信号转导和维持其不成熟表型。因此,不成熟的DC降低T细胞的效应子功能并增加CD4+CD25+Foxp3+Treg的频率,如在以前的研究中也有观察;及(2)它直接向效应T细胞信号传导,并减弱其效应子功能。
值得注意的是,在2D培养中,afTGF-β比可溶性TGF-β更有效。这种现象可能缘于亲和结合在基质上的TGF-β对细胞的局部呈递,由此增强信号转导。此外,与其吸附在原始支架时的快速释放相比,TGF-β的亲和结合导致延长的活性细胞因子的持续释放。这两种机制都有助于免疫调节环境的成功产生。TGF-β-控释系统曾用于体外和体内诱导和扩展Treg的群体。然而,这些系统都没有开发细胞因子的亲和力结合或试图探索该策略,以创造一个将促使其作为同种异体细胞移植装置使用的3D免疫调节环境。
作为移植装置,藻酸支架补充有血管生成因子VEGF和PDGF-β,其广泛地增强支架血管形成。这种细胞移植装置的一个潜在分支是,虽然促进血管形成以使能够去往和来自移植的细胞的有效物质输送,但增强的白细胞浸润会促进同种异体移植排斥。我们的数据显示在体外和体内观察到,afTGF-β发挥相似的调节作用。与TGF-β缺乏构建体相比,移植后10天和15天取回的带有afTGF-β的构建体显示显著降低的成熟DC的频率、增加的Treg的频率、和降低的细胞毒性T细胞的频率。此外,以afTGF-β移植的小鼠也会引起外周耐受,因为脾细胞衍生的同种异体成纤维细胞反应性CD4和CD8 T细胞呈现出显著降低的促炎细胞因子水平,和几乎完全消失的细胞毒性活性。
因此,只要TGF-β在支架中局部呈递,移植装置的血管形成不仅提高移植物的存活,而且减弱响应移植细胞引起的外周免疫反应。
我们提出一个两阶段模型来解释afTGF-β构建体相对于TGF-β缺乏构建体的独特性质。第一阶段,afTGF-β在其吞噬移植细胞的同时,向浸润APC信号传导并维持其在不成熟状态。毛细管在移植后几天内形成,APC通过其迁移到引流淋巴结,并然后促进幼稚的同种异体成纤维细胞反应性CD4和CD8 T细胞的失能或轻度激活。第二阶段,afTGF-β信号转导到支架浸润的T细胞,和未成熟DC的存在一起,进一步减弱DC的激活,并促进Treg的分化和/或扩展。在移植afTGF-β构建体后15天和30天观察,这些过程总体上导致了显著降低的同种异体成纤维细胞的细胞毒性。
随着藻酸支架在宿主中侵蚀,TGF-β富含的环境扩展局部抗原特异性免疫调节至全身性和抗原特异性免疫耐受是十分重要的。例如,在1型糖尿病的情况下,响应移植的β细胞引起的这种外周耐受允许阻断胰腺内的免疫攻击,并可能预防胰岛再生。类似的策略能用于治疗其他组织特异性自身免疫疾病,同时避免损害宿主对病原体的防御,例如免疫抑制药物的情况。
实施例5
亲和结合至藻酸或硫酸酯化藻酸基质的TGF-β(afTGF-β)用于治疗多发性硬化(MS)
实验用自身免疫性脑脊髓炎(Experimental Autoimmune Encephalomyelitis,EAE)用作MS模型。以髓磷脂少突胶质细胞糖蛋白(MOG)残基35-55免疫C57BL6小鼠。7~10天后,由进行临床评分,小鼠出现尾、肢麻痹。该疾病通过白细胞浸润到脊髓中形成典型的MS病变特征显现。所述疾病经常以复发缓解方式出现。在免疫前后的不同时间点(如发病前第5天,临床评分出峰的第15天或缓解后的第20天)植入携带或缺乏TGF-β和MOG35-55肽的藻酸或硫酸酯化藻酸支架,并检查EAE临床评分(正如如Miller等“Experimental AutoimmuneEncephalomyelitis in the Mouse.”Curr Protoc Immunol.2007年5月:CHAPTER:Unit-15.1所述,通过参考文献在此引入),连同MOG特异性CD4T细胞的表型(细胞因子谱及活化标记物)及脊髓组织病理学检查,即浸润白细胞与脱髓鞘。
结果显示MOG特异性T细胞是耐受的,并从而预防或缓解急性的和慢性的疾病过程,包含在接受携带TGF-β和MOG35-55支架的动物体内EAE临床评分的降低、淋巴细胞浸润和减少的MS损伤数量。
在临床试验中,TGFβ支架携带若干依赖于患有MS的对象中的特定HLA等位基因的免疫显性表位。
实施例6
afTGF-β用于治疗I型糖尿病
在其中白细胞破坏胰腺中β细胞的Ⅰ型糖尿病小鼠模型(例如NOD小鼠)中,在发病前后(大约10周)移植表达TGFP的同种异体的或同系的胰腺β细胞,并检测小鼠中胰岛素和葡萄糖水平,连同支架中和胰腺中的免疫浸润、病理生理和β细胞存活。
结果显示,β细胞特异性淋巴细胞是耐受的,并从而预防或缓解急性的和慢性的疾病过程,包含接受表达ΤGFβ的胰岛β细胞的动物中的β细胞死亡的减少、胰岛素水平的增加和血糖水平的减少。
