Abstract
本发明提供了一种预防和治疗糖尿病的低氘水饮品的制备方法,方法为:1)制备香菇多糖;2)将香菇多糖、玉米须提取液、麦冬提取液、葛粉、山药提取液、粳米提取液、西洋参提取液、枸杞子提取液和桔梗提取液混合,搅拌均匀,灭菌,即得。与现有技术相比,本发明中控制各原料的提取工艺和用量比,香菇多糖能够通过调节糖代谢、减少肝糖元分解的途径来降低血糖;西洋参提取液中含皂苷,能够降血脂、促进血液循环;枸杞子能增强胰岛素的敏感性,降低血糖的同时,还能防止餐后血糖升高;玉米须降血糖,质量糖尿病;桔梗不仅具有降糖效果,还能能缓解糖尿病患者的烦热、口渴症状;多种成分,协同起到预防和治疗糖尿病的作用。

技术领域
本发明属于饮品领域,具体涉及一种预防和治疗糖尿病的低氘水饮品的制备方法。
背景技术
自然界里存在的水一般由2个氢原子和1个氧原子组成,但氢原子有质量不同的3个同位素,原子量量分别为1,2,3的氢(H)、氘(D,重氢)、氚(T,超重氢)。国内外研究表明,重氢对生命体的生存发展和繁衍有害。
而,自然界中水中氘氢比为150ppm。一般,水中氘的含量低于自然界常用水含量150ppm的水即为低氘水。
国内外研究表明,低氘水对人体健康有诸多好处,更有益于生命体的生存和繁衍,对于人类的健康具有重要意义。氘对生命体的生存和繁衍有害,而含氘量偏低的低氘水,具有特殊的分子结构,分子因直径比普通水小一半,进入生命体后具有吸收快、渗透力高、溶解力强的特点,能有效代谢生命体的物质,吸纳有益营养,排除有害物质。这些情况对动物如此,对植物如此,对人类也是如此。因此,含氘量偏高的重水被称做“死亡之水”,而含氘量较低的低氘水则被称做“生命之水”。
低氘水主要存在于地球两级和高山的冰雪中,采用特定方法降低自然水中的氘也可获得低氘水。
比如公开号101481088A,申请号为200910046130.8,申请名称为“一种超轻水的制备装置及超轻水的制备方法”公开的方法制备得氘丰度2-105ppm的低氘水。
糖尿病是一组以高血糖为特征的代谢性疾病。高血糖则是由于胰岛素分泌缺陷或其生物作用受损,或两者兼有引起。糖尿病时长期存在的高血糖,导致各种组织,特别是眼、肾、心脏、血管、神经的慢性损害、功能障碍。
发明内容
本发明的目的在于提供一种预防和治疗糖尿病的低氘水饮品的制备方法,以低氘水为溶剂,提取多种中药有效成分,配合香菇多糖,具有促进胰岛素分泌,降血糖的作用,达到预防和治疗糖尿病的目的。
本发明具体技术方方案如下:
一种预防和治疗糖尿病的低氘水饮品的制备方法,包括以下步骤:
1)制备香菇多糖;
2)将香菇多糖、玉米须提取液、麦冬提取液、葛粉、山药提取液、粳米提取液、西洋参提取液、枸杞子提取液和桔梗提取液混合,搅拌均匀,灭菌,即得预防和治疗糖尿病的低氘水饮品。
步骤1)所述制备香菇多糖的方法为:
1-1)将香菇粉碎过150-200筛后,置于5-8倍质量的低氘水中浸泡2-3h,然后加热至85-95℃,微波提取;
1-2)将步骤1-1)提取体系离心,收集提取液,反复冻融,离心后,收集上层清液;
1-3)向步骤1-2)得到的上层清液中加入碳酸氢钠,调节pH至6-7,然后,加入胰蛋白酶,水浴加热至35-37℃,反应0.5-1h;然后加热至90-95℃保温30-50min,使酶失活,离心,过滤,收集滤液,得到香菇多糖粗提取液;
1-4)将步骤1-3)制备的多糖粗提取液减压浓缩后,加入无水乙醇沉降,离心,过滤,收集沉淀;
1-5)沉淀加入5-10重量倍的低氘水溶解后,加入三氟乙酸,加热水解,40-60℃真空回收三氟乙酸,得到小分子香菇多糖提取液;
1-6)将小分子香菇多糖提取液浓缩,真空干燥,得香菇多糖。
本发明中低氘水为市售,或者利用公开号101481088A,申请号为200910046130.8,申请名称为“一种超轻水的制备装置及超轻水的制备方法”公开的方法制备得2-60ppm的低氘水。
步骤1-1)中所述微波提取,是指微波功率400-500W条件下,微波提取1-2h。
步骤1-2)中所述反复冻融是指:收集提取液后,待提取液冷却至室温后,置于-18~-20℃条件下冷冻4-8h后,置于室温下溶解,然后离心,收集上层清液,再置于-18~-20℃条件下冷冻4-8h后,置于室温下溶解,然后离心,收集上层清液,重复3-5次。
步骤1-3)中胰蛋白酶的加入量为上清液质量的3-8%。
步骤1-4)中所述减压浓缩是指浓缩至原来体积的1/4-1/3。
步骤1-4)中所述加入无水乙醇沉降具体为:加入浓缩液体积2-5倍的乙醇,静置4-8h,离心,收集沉淀。
步骤1-5)中三氟乙酸的用量为处理的溶液的质量的1-2%。
步骤1-5)中所述加热水解是指加热至80-95℃条件下,水解15-25min。
步骤2)中所述玉米须提取液的制备方法为:
将玉米须剪碎,置于10-15倍质量的低氘水加热至90-98℃煎煮提取1-2h,过滤,滤渣再置于6-10倍的低氘水加热至90-98℃煎煮提取1-1.5h,过滤,合并两次滤液,滤液加热浓缩至原来体积的1/6-1/4,即可。
步骤2)中所述麦冬提取液的制备方法为:
将麦冬置于5-10倍低氘水中,20-40kHz条件超声提取20-30min,然后,加热浓缩至原来体积的1/5-1/3,即可。
步骤2)中所述葛粉为市售。
步骤2)中所述山药提取液的制备方法为:
将山药切碎后,置于13-15倍质量的低氘水加热至90-95℃煎煮提取0.5-1.5h,过滤,滤渣再置于8-10倍的低氘水加热至90-95℃煎煮提取1-1.5h,过滤,合并两次滤液,滤液加热浓缩至原来体积的1/5-1/4,即可。
步骤2)中所述粳米提取液的制备方法为:
将粳米洗净后,置于12-14倍质量的低氘水浸泡,然后加热至85-95℃煎煮提取1.5-2.5h,过滤,滤渣再置于5-9倍的低氘水加热至85-95℃煎煮提取1-2h,过滤,合并两次滤液,滤液加热浓缩至原来体积的1/5-1/4,即可。
步骤2)中所述西洋参提取液的制备方法为:
将西洋参切碎,过60-80目筛,然后置于13-15倍质量的低氘水浸泡1-2h,然后加热至85-90℃煎煮提取2-3h,过滤,滤渣再置于10-12倍的低氘水加热至85-90℃煎煮提取0.5-1.5h,过滤,合并两次滤液,滤液加热浓缩至原来体积的1/5-1/3,即可。
步骤2)中所述枸杞子提取液的制备方法为
将枸杞子洗净后,置于15-18倍质量的低氘水中浸泡1-2h,然后,加热至90-95℃,煎煮提取1-2h,过滤,滤渣再加入13-15倍质量的低氘水中,加热至90-95℃条件下煎煮提取1-1.5h,过滤,合并两次滤液,滤液加热浓缩至原来体积的1/8-1/6,即可。
步骤2)中所述桔梗提取液的制备方法为:
将桔梗粉碎过60-80目筛,置于5-8倍质量的低氘水中浸泡1-2h,然后20-40kHz条件超声提取20-30min,然后,加热浓缩至原来体积的1/5-1/4,即可。
步骤2)中所述香菇多糖、玉米须提取液、麦冬提取液、葛粉、山药提取液、粳米提取液、西洋参提取液、枸杞子提取液和桔梗提取液的质量比为0.1:1-2:1-1.5:0.2-0.4:3-4:2-5:1-2:2-3:2-4。
步骤2)中所述灭菌是指121℃条件下灭菌15-25min。
本发明中,以低氘水为主要溶剂,制备香菇多糖和多种中药提取液,低氘水可显著降低空腹血糖水平,提升空腹胰岛素水平,改善胰岛细胞形态、体积和分布。香菇多糖能够调节糖代谢,减轻糖尿病症状。香菇多糖通过促进胰岛素分泌、改善胰岛素抵抗,调节相关酶的活性,改善糖、蛋白质及脂肪代谢紊乱,促进肝糖元合成、抑制糖异生,增进免疫系统功能和抗氧化作用等多种途径发挥降血糖功能。
在制备香菇多糖的方法中,利用冰川水低氘的特性,作为提取溶剂,或者,利用冰川水制备氘含量更低的低氘水,由于冰川水富含微量元素,有利于香菇多糖的溶解和提取。提取过程中,加热低氘水,配合微波提取,有利于提高多糖的提取效率,多糖的提取率更高。在提取后,先反复冻融,除去蛋白质和其他杂质,再利用胰蛋白酶进一步除去蛋白质,提高多糖的获得率,而且,最后醇沉降一次,即可高效率的除去蛋白质。最后,加入三氟乙酸,将香菇多糖切割为小分子多糖,有利于人体吸收,提高其保健价值。具有抑制癌细胞,降血脂、降血糖等作用。
与现有技术相比,本发明中控制各原料的提取工艺和用量比,香菇多糖能够通过调节糖代谢、减少肝糖元分解的途径来降低血糖,无需通过胰岛素的作用,所以特别适合空腹血糖高和餐前血糖高者食用。西洋参提取液中含皂苷,能够降血脂、促进血液循环。枸杞子提取液中含有效成分枸杞多糖,能增强胰岛素的敏感性,降低血糖的同时,还能防止餐后血糖升高。玉米须提取液也能降血糖,治疗糖尿病。桔梗不仅具有降糖效果,还能能缓解糖尿病患者的烦热、口渴症状。配合麦冬提取液、葛粉、山药提取液、粳米提取液等多种成分,协同起到预防和治疗糖尿病的作用。
具体实施方式
本发明实施例1和2所用低氘水为市售。
实施例1
一种预防和治疗糖尿病的低氘水饮品的制备方法,包括以下步骤:
1)制备香菇多糖:
1-1)将香菇粉碎过200筛后,置于6倍质量的低氘水中浸泡2h,然后加热至95℃,微波功率500W条件下,微波提取1.5h;
1-2)将步骤1-1)提取体系离心,收集提取液,待提取液冷却至室温后,置于-18~-20℃条件下冷冻6h后,置于室温下溶解,然后离心,收集上层清液,再置于-18~-20℃条件下冷冻6h后,置于室温下溶解,然后离心,收集上层清液,再置于-18~-20℃条件下冷冻6h后,置于室温下溶解,离心后,收集上层清液;
1-3)向步骤1-2)得到的上层清液中加入碳酸氢钠,调节pH至6-7,然后,加入上清液质量的3.5%的胰蛋白酶,水浴加热至35℃,反应1h;然后加热至95℃保温30min,使酶失活,离心,过滤,收集滤液,得到香菇多糖粗提取液;
1-4)将步骤1-3)制备的多糖粗提取液减压浓缩至原来体积的1/3,加入浓缩液体积4倍的无水乙醇沉降,静置7h后,离心,过滤,收集沉淀;
1-5)沉淀加入7重量倍的低氘水溶解后,加入1%的氟乙酸,加热质85℃水解20min,然后,60℃真空回收三氟乙酸,得到小分子香菇多糖提取液;
1-6)将小分子香菇多糖提取液浓缩,真空干燥,得香菇多糖。
2)将香菇多糖、玉米须提取液、麦冬提取液、葛粉、山药提取液、粳米提取液、西洋参提取液、枸杞子提取液和桔梗提取液按照质量比0.1:1.3:1.2:0.3:3.5:3:1.4:2.3:3混合,搅拌均匀,121℃条件下灭菌15min,即得预防和治疗糖尿病的低氘水饮品。
步骤2)中所述玉米须提取液的制备方法为:
将玉米须剪碎,置于12倍质量的低氘水加热至95℃煎煮提取1h,过滤,滤渣再置于8倍的低氘水加热至95℃煎煮提取1.5h,过滤,合并两次滤液,滤液加热浓缩至原来体积的1/4,即可。
步骤2)中所述麦冬提取液的制备方法为:
将麦冬置于10倍低氘水中,30kHz条件超声提取20min,然后,加热浓缩至原来体积的1/3,即可。
步骤2)中所述葛粉为市售。
步骤2)中所述山药提取液的制备方法为:
将山药切碎后,置于14倍质量的低氘水加热至90℃煎煮提取1.5h,过滤,滤渣再置于9倍的低氘水加热至90℃煎煮提取1.5h,过滤,合并两次滤液,滤液加热浓缩至原来体积的1/4,即可。
步骤2)中所述粳米提取液的制备方法为:
将粳米洗净后,置于14倍质量的低氘水浸泡,然后加热至95℃煎煮提取2h,过滤,滤渣再置于8倍的低氘水加热至95℃煎煮提取2h,过滤,合并两次滤液,滤液加热浓缩至原来体积的1/4,即可。
步骤2)中所述西洋参提取液的制备方法为:
将西洋参切碎,过80目筛,然后置于13倍质量的低氘水浸泡1.5h,然后加热至90℃煎煮提取2.5h,过滤,滤渣再置于12倍的低氘水加热至90℃煎煮提取1h,过滤,合并两次滤液,滤液加热浓缩至原来体积的1/4,即可。
步骤2)中所述枸杞子提取液的制备方法为
将枸杞子洗净后,置于16倍质量的低氘水中浸泡1.5h,然后,加热至90℃,煎煮提取1.5h,过滤,滤渣再加入14倍质量的低氘水中,加热至90℃条件下煎煮提取1.5h,过滤,合并两次滤液,滤液加热浓缩至原来体积的1/6,即可。
步骤2)中所述桔梗提取液的制备方法为:
将桔梗粉碎过80目筛,置于7倍质量的低氘水中浸泡2h,然后30kHz条件超声提取20min,然后,加热浓缩至原来体积的1/4,即可。
实施例2
一种预防和治疗糖尿病的低氘水饮品的制备方法,包括以下步骤:
1)制备香菇多糖:
1-1)将香菇粉碎过180筛后,置于8倍质量的低氘水中浸泡3h,然后加热至95℃,微波功率400W条件下,微波提取2h;
1-2)将步骤1-1)提取体系离心,收集提取液,收集提取液后,待提取液冷却至室温后,置于-18~-20℃条件下冷冻8h后,置于室温下溶解,然后离心,收集上层清液,再置于-18~-20℃条件下冷冻8h后,置于室温下溶解,然后离心,收集上层清液,再置于-18~-20℃条件下冷冻8h后,置于室温下溶解,然后离心,收集上层清液,再置于-18~-20℃条件下冷冻8h后,置于室温下溶解,然后离心,收集上层清液,再置于-18~-20℃条件下冷冻8h后,置于室温下溶解,然后离心,收集上层清液;
1-3)向步骤1-2)得到的上层清液中加入碳酸氢钠,调节pH至6-7,然后,加入上清液质量的5%的胰蛋白酶,水浴加热至37℃,反应0.5h;然后加热至90℃保温40min,使酶失活,离心,过滤,收集滤液,得到香菇多糖粗提取液;
1-4)将步骤1-3)制备的多糖粗提取液减压浓缩至原来体积的1/3后,加入4倍体积的无水乙醇沉降静置8h,离心,过滤,收集沉淀;
1-5)沉淀加入10重量倍的低氘水溶解后,加入1.4%的三氟乙酸,加热至90℃水解20min,50℃真空回收三氟乙酸,得到小分子香菇多糖提取液;
1-6)将小分子香菇多糖提取液浓缩,真空干燥,得香菇多糖。
2)将香菇多糖、玉米须提取液、麦冬提取液、葛粉、山药提取液、粳米提取液、西洋参提取液、枸杞子提取液和桔梗提取液按照质量比0.1:1.8:1.5:0.36:3.5:4.2:1.2:2.5:3.8混合,搅拌均匀,121℃条件下灭菌20min,即得预防和治疗糖尿病的低氘水饮品。
步骤2)中所述玉米须提取液的制备方法为:
将玉米须剪碎,置于15倍质量的低氘水加热至98℃煎煮提取1h,过滤,滤渣再置于8倍的低氘水加热至898℃煎煮提取18h,过滤,合并两次滤液,滤液加热浓缩至原来体积的1/4,即可。
步骤2)中所述麦冬提取液的制备方法为:
将麦冬置于10倍低氘水中,40kHz条件超声提取20min,然后,加热浓缩至原来体积的1/5,即可。
步骤2)中所述葛粉为市售。
步骤2)中所述山药提取液的制备方法为:
将山药切碎后,置于15倍质量的低氘水加热至95℃煎煮提取1h,过滤,滤渣再置于10倍的低氘水加热至95℃煎煮提取1h,过滤,合并两次滤液,滤液加热浓缩至原来体积的1/5,即可。
步骤2)中所述粳米提取液的制备方法为:

步骤2)中所述枸杞子提取液的制备方法为
将枸杞子洗净后,置于18倍质量的低氘水中浸泡1h,然后,加热至90℃,煎煮提取1h,过滤,滤渣再加入15倍质量的低氘水中,加热至90℃条件下煎煮提取1h,过滤,合并两次滤液,滤液加热浓缩至原来体积的1/6,即可。
步骤2)中所述桔梗提取液的制备方法为:
将桔梗粉碎过80目筛,置于7倍质量的低氘水中浸泡1.5h,然后30kHz条件超声提取30min,然后,加热浓缩至原来体积的1/5,即可。
实施例1制备的饮品,有131位不同性别不同年龄的糖尿病患者,按照自愿原则饮用,每次250ml,每天2次,连续饮用1个月,有5人糖尿病未好转,有102人病情不同程度的好转,有24人治愈。
实施例2制备的饮品,有97位不同性别不同年龄的糖尿病患者,按照自愿原则饮用,每次250ml,每天2次,连续饮用1个月,有3位糖尿病人未好转,无效,有81人病情人不同程度的好转,有13人治愈。
Claims (10)
1.一种预防和治疗糖尿病的低氘水饮品的制备方法,其特征在于,所述制备方法包括以下步骤:
1)制备香菇多糖;
2)将香菇多糖、玉米须提取液、麦冬提取液、葛粉、山药提取液、粳米提取液、西洋参提取液、枸杞子提取液和桔梗提取液混合,搅拌均匀,灭菌,即得预防和治疗糖尿病的低氘水饮品。
2.根据权利要求1所述的制备方法,其特征在于,步骤1)所述制备香菇多糖的方法为:
1-1)将香菇粉碎过150-200筛后,置于5-8倍质量的低氘水中浸泡2-3h,然后加热至85-95℃,微波提取;
1-2)将步骤1-1)提取体系离心,收集提取液,反复冻融,离心后,收集上层清液;
1-3)向步骤1-2)得到的上层清液中加入碳酸氢钠,调节pH至6-7,然后,加入胰蛋白酶,水浴加热至35-37℃,反应0.5-1h;然后加热至90-95℃保温30-50min,使酶失活,离心,过滤,收集滤液,得到香菇多糖粗提取液;
1-4)将步骤1-3)制备的多糖粗提取液减压浓缩后,加入无水乙醇沉降,离心,过滤,收集沉淀;
1-5)沉淀加入5-10重量倍的低氘水溶解后,加入三氟乙酸,加热水解,40-60℃真空回收三氟乙酸,得到小分子香菇多糖提取液;
1-6)将小分子香菇多糖提取液浓缩,真空干燥,得香菇多糖。
3.根据权利要求2所述的制备方法,其特征在于,步骤1-1)中所述微波提取,是指微波功率400-500W条件下,微波提取1-2h。
4.根据权利要求2所述的制备方法,其特征在于,步骤1-3)中胰蛋白酶的加入量为上清液质量的3-8%。
5.根据权利要求2所述的制备方法,其特征在于,步骤1-5)中三氟乙酸的用量为处理的溶液的质量的1-2%。
6.根据权利要求1或2所述的制备方法,其特征在于,步骤2)中所述玉米须提取液的制备方法为:
将玉米须剪碎,置于10-15倍质量的低氘水加热至90-98℃煎煮提取1-2h,过滤,滤渣再置于6-10倍的低氘水加热至90-98℃煎煮提取1-1.5h,过滤,合并两次滤液,滤液加热浓缩至原来体积的1/6-1/4,即可;
步骤2)中所述麦冬提取液的制备方法为:
将麦冬置于5-10倍低氘水中,20-40kHz条件超声提取20-30min,然后,加热浓缩至原来体积的1/5-1/3,即可。
7.根据权利要求1或2所述的制备方法,其特征在于,步骤2)中所述山药提取液的制备方法为:
将山药切碎后,置于13-15倍质量的低氘水加热至90-95℃煎煮提取0.5-1.5h,过滤,滤渣再置于8-10倍的低氘水加热至90-95℃煎煮提取1-1.5h,过滤,合并两次滤液,滤液加热浓缩至原来体积的1/5-1/4,即可;
步骤2)中所述粳米提取液的制备方法为:
将粳米洗净后,置于12-14倍质量的低氘水浸泡,然后加热至85-95℃煎煮提取1.5-2.5h,过滤,滤渣再置于5-9倍的低氘水加热至85-95℃煎煮提取1-2h,过滤,合并两次滤液,滤液加热浓缩至原来体积的1/5-1/4,即可。
8.根据权利要求1或2所述的制备方法,其特征在于,步骤2)中所述西洋参提取液的制备方法为:
将西洋参切碎,过60-80目筛,然后置于13-15倍质量的低氘水浸泡1-2h,然后加热至85-90℃煎煮提取2-3h,过滤,滤渣再置于10-12倍的低氘水加热至85-90℃煎煮提取0.5-1.5h,过滤,合并两次滤液,滤液加热浓缩至原来体积的1/5-1/3,即可;
步骤2)中所述枸杞子提取液的制备方法为

9.根据权利要求1或2所述的制备方法,其特征在于,步骤2)中所述桔梗提取液的制备方法为:
将桔梗粉碎过60-80目筛,置于5-8倍质量的低氘水中浸泡1-2h,然后20-40kHz条件超声提取20-30min,然后,加热浓缩至原来体积的1/5-1/4,即可。
10.根据权利要求1或2所述的制备方法,其特征在于,步骤2)中所述香菇多糖、玉米须提取液、麦冬提取液、葛粉、山药提取液、粳米提取液、西洋参提取液、枸杞子提取液和桔梗提取液的质量比为0.1:1-2:1-1.5:0.2-0.4:3-4:2-5:1-2:2-3:2-4。
