本发明涉及一种复方纳米蜂胶的制备方法及其产品,其制备方法如下:将蜂胶溶于丙酮至饱和制成饱和蜂胶溶液,将水溶性灵芝多糖、水溶性黄芪多糖、水溶性珍珠粉和水溶性马鞭草提取物水中至饱和制成复合饱和水溶液,然后将饱和蜂胶溶液和复合饱和水溶液混匀,‑40℃~‑80℃冷冻过夜,再真空冷冻干燥得复方纳米蜂胶,本发明采用了纳米生物技术工艺,最大限度的提高利用药物理性能,同时优化配伍,有医疗保健、药膳食疗双重功能。

蜂胶是蜜蜂从植物的树芽、树皮等部位采集的树脂、挥发性物质并混入其上颚腺、舌腺、蜡腺等腺体分泌物和少量花粉经蜜蜂加工转化而成的一种具有芳香粘性物质。每群蜂(5-6万个)每年平均仅能生产100克左右蜂胶,蜜蜂利用蜂胶保护蜂群,填补蜂箱的裂缝孔洞,涂刷巢房内壁,保存蜂粮及抵抗病虫害。养蜂者可以从蜂箱副盖、覆布、巢框上梁和蜂箱体内刮取蜂胶,这是一种很费时的方法,产量也很低。
国内外大量的研究已经证实,蜂胶有众多的医疗保健功能。蜂胶的主要有效成份是黄酮类、萜烯类,醛酮类、酯类和有机酸类化合物。蜂胶是一种天然的抗生素,具有特殊的生物学效应。其主要功能是具有广谱的抗菌消炎作用、增强抗体的免疫能力、珍贵的天然杭氧化剂,血管的“清道夫”和微循环的“保护神”,因此,蜂胶和蜂王浆一样,都是非常珍贵的蜜蜂产品,在过去的几十年年里,美国、德国、日本、加拿大、巴西、俄罗斯等几十个国家里都不断掀起蜂胶应用热潮,1972年在捷克斯洛伐克召开了首届国际蜂胶大会,1989年日本厚生省批准了蜂胶为食品的抗氧化剂。
现代药理研究显示蜂胶具有抗细菌感染、抗病毒、抗肿瘤、抗氧化;降血脂、降血糖、降血压;增强免疫、促进组织再生等广泛的药理作用,被誉为“紫黄金”。但由于蜂胶独特的物理性质,如不溶于水,吸湿性强,低温变脆,高温变粘等给蜂胶精加工带来了难度,使传统的蜂胶产品溶解性差,吸收利用度低,进入消化道后不易被人体吸收和利用,不能充分发挥其药效作用,给临床应用和销售带来了一定的困难。
纳米科学技术是20世纪80年代末刚刚诞生并正在崛起的新科技,作为现代科技领域的热门技术,已广泛应用于多个领域。它的基本涵义是纳米尺寸(1-1000nm)范围内认识和改造自然。纳米新科技诞生时间不长,但在几个重要领域有了重大的进展,其中纳米材料出现了许多传统不具备的特征。已引起世界各国科学家的极大兴趣,这些特性的产生主要基于下列效应:小尺寸效应、表面效应、量子尺寸效应、和宏观量子隧道效应。近年来研究发现,采用纳米技术将相应材料制成纳米颗粒,其理化性质和作用会发生惊人变化,可以有效地改变纳米材料的表面物化性质,提高其与水等溶剂的亲和性。现有技术也有将蜂胶制成纳米蜂胶的报道,但是未见纳米蜂胶与其他组分制成复方制剂的报道。
发明内容
有鉴于此,本发明的目的之一在于提供一种复方纳米蜂胶的制备方法,其采用纳米生物技术工艺,最大限度的提高利用药物理性能,同时优化配伍,有医疗保健、药膳食疗双重功能;本发明的目的之二在于提供由上述方法制得的产品。
为达到上述目的,本发明提供如下技术方案:
1、一种复方纳米蜂胶的制备方法,包括如下步骤:将蜂胶溶于丙酮中至饱和制成饱和蜂胶溶液,将水溶性灵芝多糖、水溶性黄芪多糖、水溶性珍珠粉和水溶性马鞭草提取物溶于水中至饱和制成复合饱和水溶液,然后将饱和蜂胶溶液和复合饱和水溶液按体积比为5:5混匀,-40℃~-80℃冷冻过夜,再真空冷冻干燥得复方纳米蜂胶。
本发明中,通过制备将蜂胶加工为纳米蜂胶,与常规的粘稠蜂胶相比,纳米蜂胶有更强的亲水性和更活泼的化学性质,进人人体的消化系统也极易被吸收利用;同时,纳米级的蜂胶颗粒改变了其表面结构,产生具有纳米粒子特有的表面界面效应,从而提高了蜂胶的抗肿瘤、抗氧化、抗病原微生物、抗菌消炎、增强免疫、促进组织再生等广泛的生物学作用。综上所述,采用纳米蜂胶颗粒,能增加蜂胶的溶解度和生物利用度,可以更好地发挥蜂胶的特殊生物学效应。
本发明中,所述蜂胶、水溶性灵芝多糖、水溶性黄芪多糖、水溶性珍珠粉和水溶性马鞭草提取物的重量比为5-10:1-5:1-5:1-10:1-5。
优选的,所述蜂胶、水溶性灵芝多糖、水溶性黄芪多糖、水溶性珍珠粉和水溶性马鞭草提取物的重量比为5:1:1:1:1。
优选的,所述蜂胶、水溶性灵芝多糖、水溶性黄芪多糖、水溶性珍珠粉和水溶性马鞭草提取物的重量比为6:2:2:2:2。
优选的,所述蜂胶、水溶性灵芝多糖、水溶性黄芪多糖、水溶性珍珠粉和水溶性马鞭草提取物的重量比为7:4:4:6:4。
优选的,所述蜂胶、水溶性灵芝多糖、水溶性黄芪多糖、水溶性珍珠粉和水溶性马鞭草提取物的重量比为8:5:5:10:5。
2、由所述制备方法制得的复方纳米蜂胶制剂。
优选的,所述制剂为胶囊制剂、丸剂、栓剂、片剂、颗粒剂或酒剂。
本发明的有益效果在于:本发明公开了一种复方纳米蜂胶的制备方法,本发明主要组方中药品为采用纳米技术超微结构工艺制品,组方中君、臣、佐、使药物明确,处方中,蜂胶是君,灵芝是臣,黄芪多糖、珍珠是佐,马鞭草是使,剂量准确;并且通过各种药物配伍性能调理。除去药物拮杭性,利于药理协调作用及特需药味配伍组方,辨证施治、理法方药。注重生物活性高效利用,并优化选择药物的特定部位以求最佳药效,突出主要特性,合理定型,定量。
此外,本发明根据我国中医药里创新设计新组方,在药理药效方面兼顾科学性、创新性、可行性及效益性,采用现代纳米生物新技术、新工艺、新物源、新设备组成全新的现代高科技产品。根据病有缓急、症有表里、人有老幼的情况、实绩了不同的剂量、药性、药效、包装工艺上采用独特的缓控释纳米成膜材料,从而适应各种患者所需。
具体实施方式
下面将对本发明的优选实施例进行详细的描述。
实施例1
复方纳米蜂胶,按重量份计各组分如下:蜂胶5份、水溶性灵芝多糖1份、水溶性黄芪多糖1份、水溶性珍珠粉1份和水溶性马鞭草提取物1份。
其制备方法如下:将蜂胶饱和溶于丙酮中制成饱和蜂胶溶液,将水溶性灵芝多糖、水溶性黄芪多糖、水溶性珍珠粉和水溶性马鞭草提取物水饱和溶于水中至制成复合饱和水溶液,然后将饱和蜂胶溶液和复合饱和水溶液混匀,-40℃~-80℃冷冻过夜,再真空冷冻干燥得复方纳米蜂胶。
实施例2
复方纳米蜂胶,按重量份计各组分如下:取蜂胶6份、水溶性灵芝多糖2份、水溶性黄芪多糖2份、水溶性珍珠粉2份、水溶性马鞭草提取物2份。
其制备方法如下:将蜂胶饱和溶于丙酮中制成饱和蜂胶溶液,将水溶性灵芝多糖、水溶性黄芪多糖、水溶性珍珠粉和水溶性马鞭草提取物水饱和溶于水中至制成复合饱和水溶液,然后将饱和蜂胶溶液和复合饱和水溶液混匀,-40℃~-80℃冷冻过夜,再真空冷冻干燥得复方纳米蜂胶。
实施例3
复方纳米蜂胶,按重量份计各组分如下:蜂胶6份、水溶性灵芝多糖3份、水溶性黄芪多糖4份、水溶性珍珠粉4份、水溶性马鞭草提取物3份。
其制备方法如下:将蜂胶饱和溶于丙酮中制成饱和蜂胶溶液,将水溶性灵芝多糖、水溶性黄芪多糖、水溶性珍珠粉和水溶性马鞭草提取物水饱和溶于水中至制成复合饱和水溶液,然后将饱和蜂胶溶液和复合饱和水溶液混匀,-40℃~-80℃冷冻过夜,再真空冷冻干燥得复方纳米蜂胶。
实施例4
复方纳米蜂胶,按重量份计各组分如下:蜂胶7份、水溶性灵芝多糖4份、水溶性黄芪多糖4份、水溶性珍珠粉6份、水溶性马鞭草提取物4份。
其制备方法如下:将蜂胶饱和溶于丙酮中制成饱和蜂胶溶液,将水溶性灵芝多糖、水溶性黄芪多糖、水溶性珍珠粉和水溶性马鞭草提取物水饱和溶于水中至制成复合饱和水溶液,然后将饱和蜂胶溶液和复合饱和水溶液混匀,-40℃~-80℃冷冻过夜,再真空冷冻干燥得复方纳米蜂胶。
实施例5
复方纳米蜂胶,按重量份计各组分如下:蜂胶8份、水溶性灵芝多糖5份、水溶性黄芪多糖5份、水溶性珍珠粉10份、水溶性马鞭草提取物5份。
其制备方法如下:将蜂胶饱和溶于丙酮中制成饱和蜂胶溶液,将水溶性灵芝多糖、水溶性黄芪多糖、水溶性珍珠粉和水溶性马鞭草提取物水饱和溶于水中至制成复合饱和水溶液,然后将饱和蜂胶溶液和复合饱和水溶液混匀,-40℃~-80℃冷冻过夜,再真空冷冻干燥得复方纳米蜂胶。
将实施例1~5制得的复合纳米蜂胶与药剂学上常规辅料制成胶囊制剂、丸剂、栓剂、片剂、颗粒剂、酒剂等,还可以制成保健饮品或药物食疗制品。
应用实施例
实施例1~5制得复合纳米蜂胶在溶解度和生物利用度,以及在抗肿瘤细胞生长率、抗氧化(清除氧自由基能力)、抗病原微生物(抑菌圈直径mm)、增强免疫(动物脾脏指数增加%)、促进组织再生中的数据,同时以8份普通蜂胶冻干粉作为对照,结果如表1所示。

上述检测方法具体如下:
其中溶解度百分比测定方法如下:在室温及常压条件下,100ml的水溶解复方纳米蜂胶达饱和时,所含复方纳米蜂胶的质量(单位为克)与100ml的水溶解常规蜂胶冻干粉达饱和时所含常规蜂胶冻干粉的质量(单位为克)的比率。
生物利用度测定方法如下:是以小鼠肌肉注射途径给药复方纳米蜂胶和常规蜂胶冻干粉饱和水溶液12小时后的药物活性成分吸收进入体循环的量与给药量的比值。
抗肿瘤细胞生长率(%)测定方法如下:肿瘤细胞在添加了相同比例的复方纳米蜂胶和常规蜂胶冻干粉饱和水溶液的培养基上生长72小时后肿瘤细胞死亡的数量与原数量的比值。
抗氧化(清除氧自由基能力)测定方法如下:参见参考文献《生姜提取物对邻苯三酚自氧化生成超氧自由基的清除》2014年39卷11期《中国调味品》杂志35-39页。
抗病原微生物(抑菌圈直径mm)测定方法如下:采用牛津杯法进行测试。
增强免疫(动物脾脏指数增加%)测定方法如下:小鼠分别注射相同剂量的复方纳米蜂胶和常规蜂胶冻干粉饱和水溶液及对照组注射生理盐水10天后处死小鼠,小鼠脾脏湿重与其体重的比值。
促进细胞再生率(%)测定方法如下:成纤维细胞在添加了相同比例的复方纳米蜂胶和常规蜂胶冻干粉饱和水溶液的培养基上生长24小时后细胞的数量与原数量的比值。
从上述结果可以看出,使用本发明制得的复合纳米蜂胶在溶解度、生物利用度、在抗肿瘤、抗氧化、抗病原微生物、增强免疫、促进组织再生的效果均显著提升。
最后说明的是,以上优选实施例仅用以说明本发明的技术方案而非限制,尽管通过上述优选实施例已经对本发明进行了详细的描述,但本领域技术人员应当理解,可以在形式上和细节上对其作出各种各样的改变,而不偏离本发明权利要求书所限定的范围。
Claims (8)
1.一种复方纳米蜂胶的制备方法,其特征在于,包括如下步骤:将蜂胶溶于丙酮中至饱和制成饱和蜂胶溶液,将水溶性灵芝多糖、水溶性黄芪多糖、水溶性珍珠粉和水溶性马鞭草提取物溶于水中至饱和制成复合饱和水溶液,然后将饱和蜂胶溶液和复合饱和水溶液按体积比为5:5混匀,-40℃~-80℃冷冻过夜,再真空冷冻干燥得复方纳米蜂胶。
2.根据权利要求1所述复方纳米蜂胶的制备方法,其特征在于:所述蜂胶、水溶性灵芝多糖、水溶性黄芪多糖、水溶性珍珠粉和水溶性马鞭草提取物的重量比为5-10:1-5:1-5:1-10:1-5。
3.根据权利要求2所述复方纳米蜂胶的制备方法,其特征在于:所述蜂胶、水溶性灵芝多糖、水溶性黄芪多糖、水溶性珍珠粉和水溶性马鞭草提取物的重量比为5:1:1:1:1。
4.根据权利要求2所述复方纳米蜂胶的制备方法,其特征在于:所述蜂胶、水溶性灵芝多糖、水溶性黄芪多糖、水溶性珍珠粉和水溶性马鞭草提取物的重量比为6:2:2:2:2。
5.根据权利要求2所述复方纳米蜂胶的制备方法,其特征在于:所述蜂胶、水溶性灵芝多糖、水溶性黄芪多糖、水溶性珍珠粉和水溶性马鞭草提取物的重量比为7:4:4:6:4。
6.根据权利要求2所述复方纳米蜂胶的制备方法,其特征在于:所述蜂胶、水溶性灵芝多糖、水溶性黄芪多糖、水溶性珍珠粉和水溶性马鞭草提取物的重量比为8:5:5:10:5。
7.由权利要求1~6任一项所述制备方法制得的复方纳米蜂胶制剂。
8.根据权利要求7所述的复方纳米蜂胶制剂,其特征在于:所述制剂为胶囊制剂、丸剂、栓剂、片剂、颗粒剂或酒剂。
