Abstract

The graphene oxide is a derivative of graphene after oxidation treatment, has a single atomic layer structure, is a two-dimensional plane structure, has a large specific surface area, and is randomly distributed with groups such as hydroxyl, epoxy, carboxyl and the like. The activated carbon material is amorphous carbon obtained by processing treatment, has huge specific surface area and complex void structure, and is a good hydrophobic adsorbent. Zeolite molecular sieves are a class of inorganic crystalline materials having uniform microporous, regular pore structure. The graphene oxide, the active carbon or the zeolite molecular sieve can effectively improve the solidification rate and the thermal stability of the volatile oil, and the like, thereby achieving the effect of slowly releasing the volatile oil.
A preparation method of the traditional Chinese medicine volatile oil slow-release sachet comprises the following steps: mixing the raw materials uniformly, and bagging to obtain the traditional Chinese medicine slow-release sachet. The method has simple preparation process, and can be used for large-scale mass production, and the prepared sachet can effectively improve sleep, and has long release time and uniform product quality.
In a preferred embodiment of the invention, the traditional Chinese medicine composition is mixed with a slow release carrier to obtain solidified powder, and the solidified powder is subpackaged into bags to obtain the traditional Chinese medicine slow release sachet.
In a preferred embodiment of the present invention, the slow release carrier comprises one or more of graphene oxide, activated carbon or zeolite molecular sieve, and the slow release carrier is further preferably graphene oxide. In a preferred embodiment of the present invention, the mass ratio of the traditional Chinese medicine composition to the sustained-release carrier is 1:0.5-1.2, preferably 1:0.7-1.2, and more preferably 1: 0.8-1.1. The mass ratio of the traditional Chinese medicine composition to the sustained-release carrier is typically but not limited to 1:0.5, 1:0.7, 1:0.9, 1:1 or 1: 1.2.
In a preferred embodiment of the present invention, the mixing conditions include: mixing for 3-8min under the conditions of 150-300 r/min. The mixing speed is typically, but not limited to, 150r/min, 170r/min, 190r/min, 200r/min, 220r/min, 240r/min, 260r/min, 280r/min or 300 r/min; the mixing time is typically, but not limited to, 3min, 4min, 5min, 6min, 7min or 8 min.
In a preferred embodiment of the present invention, the preparation method of the volatile oil of each raw material in the Chinese medicinal composition independently comprises steam distillation or vacuum extraction.
In a preferred embodiment of the present invention, the raw materials are pretreated, and the pretreatment comprises washing the Chinese herbal medicines and naturally drying the Chinese herbal medicines.
In a preferred embodiment of the present invention, the vacuum extraction conditions include: extracting under the conditions of pressure of 30-50kPa and temperature of 65-85 deg.C for 2-8 h. The pressure is typically, but not limited to, 30kPa, 35kPa, 40kPa, 45kPa, or 50 kPa; temperatures are typically, but not limited to, 65 ℃, 70 ℃, 75 ℃, 80 ℃ or 85 ℃; the extraction time is typically, but not limited to, 2h, 3h, 4h, 5h, 6h, 7h or 8 h.

Test example 1 Inclusion Effect
The sachet components of examples 8-17 were prepared according to the preparation method of example 20, and the inclusion rate and yield were counted, and the results are as follows:
as can be seen from the results in the above table, the inclusion rate and yield of the sachet traditional Chinese medicine volatile oil compositions in examples 8-17 are both high. Specifically, the traditional Chinese medicine volatile oil composition of the sachet in examples 8-14 uses graphene oxide as an inclusion carrier, and the inclusion rate and the yield are both significantly better than the activated carbon in example 15, the zeolite molecular sieve in example 16 and the beta-cyclodextrin in example 17. Graphene oxide, activated carbon, zeolite molecular sieve and beta-cyclodextrin are preferably used as the inclusion carrier in sequence.
The sachet components of example 14 were prepared according to the preparation method of examples 18-21, and the inclusion rate and yield were counted, and the results are as follows:

From the above results, it can be seen that the inclusion rate and the yield of the preparation methods of examples 18-20 are higher than those of the preparation method of example 21, which indicates that the product prepared by the preparation method of the sachet provided by the invention has good performance.
Test example 2 stability and sustained Release
Taking out the same mass of clathrate from the sachet in examples 8-17, storing at constant temperature of 60 deg.C for seven days with the same mass of Chinese medicinal volatile oil composition in example 7 as comparative example, extracting volatile oil in 1, 3, 5, 7 days respectively, measuring content, and comparing stability and sustained release of the volatile oil composition. The statistical results are as follows:
as can be seen from the results in the table, the stability of the composition in the sachets in examples 8-17 is higher, and compared with the traditional Chinese medicine volatile oil composition, the stability after volatile oil inclusion is remarkably improved, which indicates that the slow release carrier in the invention can remarkably improve the slow release time of the traditional Chinese medicine volatile oil composition and prolong the service life of the sachets. Specifically, the graphene oxide is used as an inclusion carrier in the inclusion compounds in examples 8 to 14, and the stability of the inclusion compounds is remarkably superior to that of the activated carbon in example 15, the zeolite molecular sieve in example 16 and the beta-cyclodextrin in example 17. Graphene oxide, activated carbon, zeolite molecular sieve and beta-cyclodextrin are preferably used as the inclusion carrier in sequence.
Test example 3 Effect
According to the traditional Chinese medicine volatile oil compositions in the examples 1-7 and the comparative examples 1-2, the sachet is prepared by taking graphene oxide as a slow release carrier according to the method for preparing the sachet in the example 20, a diazepam injection is taken as a positive control group, meanwhile, a blank control group is set, and statistics is carried out on the autonomic activity of mice. Each group had 12 mice, 6 of which were male and female.
Wherein, the specific operation of the autonomous activity condition is as follows: in the experimental groups of examples 1-7 and comparative examples 1-2, sachets of the same mass were used, the slow release carrier in the sachets containing 0.6g of the Chinese medicinal volatile oil composition, and the number of activities of the incense within 20min was recorded by a recorder; the positive control group is administered with 0.1mL/10g by intragastric administration, and the activity times within 20min of administration are recorded by a recorder; the blank control group was placed in a cotton ball soaked with 0.6g of physiological saline and the number of activities within 20min of administration was recorded by a recorder. The statistical results are given in the following table, wherein: comparing with blank group, P is less than 0.05, P is less than 0.01:

It can be seen from the data in the above table that the number of autonomic activity of the mice in examples 1 to 7, comparative examples 1 to 2 and the positive control group was significantly reduced. Specifically, the mice of examples 1 to 7 had a lower number of autonomous activities than those of comparative examples 1 to 2, and a little higher number than that of the positive control group. The traditional Chinese medicine volatile oil composition provided by the invention has reasonable components and proportion, can effectively play a role in tranquilizing and allaying excitement and is beneficial to improving sleep.
Test example 4 Effect
According to the traditional Chinese medicine volatile oil compositions in the examples 1-7 and the comparative examples 1-2, the sachet is prepared by taking graphene oxide as a slow release carrier according to the method for preparing the sachet in the example 20, the heart-benefiting mind-tranquilizing tablet is taken as a positive control group, a blank control group is arranged at the same time, and the sleep latency and the sleep duration of the mouse are counted. Each group had 12 mice, 6 of which were male and female.
The method comprises the following specific operations: in examples 1 to 7 and comparative examples 1 to 2, the test group was injected with pentobarbital sodium 50mg/kg in the abdominal cavity of each mouse 24 hours later using a sachet of the same mass of 1.2g, and the sleep latency (disappearance time of righting reflex) and the sleep duration (time from disappearance of righting reflex to recovery) were observed and recorded; the positive control group is administrated with 17mg/10g of Yixinningshen tablets by stomach irrigation, the administration is carried out once every 12h, 50mg/kg of pentobarbital sodium is injected into the abdominal cavity of each mouse after 24h, and the sleep onset latency (disappearance time of righting reflex) and the sleep duration (disappearance time of righting reflex to recovery) are observed and recorded; the blank control group was placed with a cotton ball soaked with 1.2g of physiological saline, the cotton ball was changed 2 times a day, and 24 hours later, pentobarbital sodium 50mg/kg was injected into the abdominal cavity of each mouse, and the sleep latency (disappearance of righting reflex) and the sleep duration (time from disappearance of righting reflex to recovery) were observed and recorded. The statistical results are given in the following table, wherein: comparing with blank group, P is less than 0.05, P is less than 0.01:

It can be seen from the data in the above table that the mice in examples 1 to 7, comparative examples 1 to 2 and the positive control group had significantly reduced sleep latency and prolonged sleep duration. Specifically, the mice of examples 1-7 had sleep onset latencies less than those of comparative examples 1-2, while being substantially the same as or less than that of the positive control group; the duration of sleep was significantly longer in the mice of examples 1-7 than in comparative examples 1-2, while being slightly less in the positive control group. The traditional Chinese medicine volatile oil composition provided by the invention has reasonable components and proportion, can effectively play a role in tranquilizing and allaying excitement and is beneficial to improving sleep.
While particular embodiments of the present invention have been illustrated and described, it would be obvious that various other changes and modifications can be made without departing from the spirit and scope of the invention. It is therefore intended to cover in the appended claims all such changes and modifications that are within the scope of this invention.

The traditional Chinese medicine composition for tranquilizing and allaying excitement is characterized by comprising the following components in parts by weight: 15-25 parts of wild jujube seed volatile oil, 3-10 parts of gardenia volatile oil, 1-6 parts of sandalwood volatile oil, 6-15 parts of horse-tree lotus volatile oil, 3-10 parts of melissa volatile oil, 10-20 parts of lavender volatile oil, 3-10 parts of chamomile volatile oil, 5-15 parts of orange flower volatile oil, 3-10 parts of sweet orange volatile oil and 1-8 parts of grapefruit volatile oil.

The traditional Chinese medicine composition according to claim 1, which is characterized by comprising the following components in parts by weight: 17-24 parts of wild jujube seed volatile oil, 3-8 parts of gardenia volatile oil, 1-5 parts of sandalwood volatile oil, 6-13 parts of horse-tree lotus volatile oil, 3-8 parts of melissa volatile oil, 10-17 parts of lavender volatile oil, 3-8 parts of chamomile volatile oil, 7-13 parts of orange flower volatile oil, 5-10 parts of sweet orange volatile oil and 3-8 parts of grapefruit volatile oil.

The traditional Chinese medicine composition according to claim 1, which is characterized by comprising the following components in parts by weight: 18-22 parts of wild jujube seed volatile oil, 3-6 parts of gardenia volatile oil, 2-5 parts of sandalwood volatile oil, 6-10 parts of horse-tree lotus volatile oil, 3-6 parts of melissa volatile oil, 10-15 parts of lavender volatile oil, 3-6 parts of chamomile volatile oil, 8-12 parts of orange flower volatile oil, 6-9 parts of sweet orange volatile oil and 3-7 parts of grapefruit volatile oil.
4. The Chinese medicinal composition according to any one of claims 1 to 3, wherein the preparation method of each volatile oil in the Chinese medicinal composition independently comprises steam distillation or vacuum extraction.
5. The traditional Chinese medicine composition of claim 4, wherein the vacuum extraction conditions comprise: extracting under the conditions of pressure of 30-50kPa and temperature of 65-85 deg.C for 2-8 h.
6. A slow release sachet comprising the Chinese medicinal composition of any one of claims 1 to 5.
7. The traditional Chinese medicine slow-release sachet of claim 6, further comprising a slow-release carrier.
8. The traditional Chinese medicine slow-release sachet of claim 7, wherein the slow-release carrier comprises one or more of graphene oxide, activated carbon or zeolite molecular sieve.
9. The traditional Chinese medicine slow-release sachet of claim 7, wherein the slow-release carrier is graphene oxide.
10. The preparation method of the traditional Chinese medicine slow-release sachet of claim 6, which comprises the following steps: mixing the raw materials uniformly, and bagging to obtain the traditional Chinese medicine slow-release sachet.
11. The preparation method according to claim 10, wherein the Chinese medicinal composition is mixed with a sustained-release carrier to obtain a solidified powder, and the solidified powder is separately packaged into a bag to obtain the Chinese medicinal sustained-release sachet.
12. The preparation method of claim 11, wherein the slow release carrier comprises one or more of graphene oxide, activated carbon or zeolite molecular sieves.

16. The preparation method of claim 15, wherein the mass ratio of the traditional Chinese medicine composition to the sustained-release carrier is 1: 0.8-1.1.
17. The method of claim 11, wherein the mixing conditions comprise: mixing for 3-8min under the conditions of 150-300 r/min.
18. The method of any one of claims 10-17, wherein the volatile oils are extracted from the composition by steam distillation or vacuum extraction.
19. The preparation method according to claim 18, wherein the conditions of the vacuum extraction method comprise: extracting under the conditions of pressure of 30-50kPa and temperature of 65-85 deg.C for 2-8 h.
