However, China is a country with a long history of wine culture, and there are thousands of years of traditional history of drinking, and nearly five hundred million wine citizens. Relatives and friends get together, business, work, etc. all drink alcohol, commonly called as no-alcohol mat. It is inevitable that the wine is not drunk excessively. CN104288652 discloses a traditional Chinese medicine composition with anti-alcoholism health efficacy, which comprises 5-15 parts of hovenia dulcis thunb, 5-15 parts of kudzu root, 0.5-7.5 parts of pueraria lobata, 0.5-7.5 parts of dried orange peel, 1-10 parts of medicated leaven, 1-10 parts of malt, 1-10 parts of scorched hawthorn fruit, 5-15 parts of oak root, 0.4-7 parts of fructus amomi, 0.8-9 parts of radish seed and 1-10 parts of poria cocos. The traditional Chinese medicine composition is prepared from pure traditional Chinese medicines according to the monarch, minister, assistant and guide traditional Chinese medicine theory, and has the effects of relieving alcoholism, quenching thirst, relieving restlessness, strengthening spleen and stomach, soothing liver, promoting qi circulation, inducing diuresis, excreting dampness and calming heart. However, the traditional Chinese medicine composition has long hangover alleviating time and uncomfortable taste.
The invention discloses an anti-alcoholism tea and a preparation method thereof, and CN 104286287. The tea for relieving alcoholism is prepared from 13 raw materials of kudzu root, lotus leaf, wrinkled gianthyssop herb, stevia rebaudiana, grosvenor momordica fruit, galangal, hovenia dulcis thunb, honeysuckle, liquorice, rhizoma polygonati, hawthorn, malt, dried orange peel and the like. But the anti-alcoholism tea still has poor curative effect and slow sobering speed.
It can be seen that for moderate and deep alcoholism, the traditional Chinese medicine preparation mainly takes chemical medicines and traditional Chinese medicine preparations, the chemical medicines can cause metabolic burden to liver and kidney, and have large side effect, while the traditional Chinese medicine preparation has small side effect, but has slow effect and insignificant curative effect.
Disclosure of Invention
The invention aims to provide a wheat seedling traditional Chinese medicine composition for nourishing liver, relieving alcoholism, dispelling effects of alcohol and preventing halitosis. Preferably, the wheat seedling traditional Chinese medicine composition for nourishing liver, removing halitosis and preventing hangover mainly comprises the following traditional Chinese medicines in parts by weight: 20 parts of wheat seedlings, 20 parts of gynostemma pentaphylla, 15 parts of hawthorn, 15 parts of kudzu root, 15 parts of hovenia dulcis thunb, 5 parts of fructus amomi, 5 parts of clove, 5 parts of polydextrose, 5 parts of vine tea, 3 parts of stevia rebaudiana, 10 parts of cinnamon and 15 parts of poria cocos.
In a specific embodiment, the wheat seedling traditional Chinese medicine composition for nourishing liver, relieving alcoholism, dispelling effects of alcohol and preventing halitosis mainly comprises the following traditional Chinese medicines in parts by weight: 20g of wheat sprout, 20g of gynostemma pentaphylla, 15g of hawthorn, 15g of radix puerariae, 15g of semen hoveniae, 5g of fructus amomi, 5g of clove, 5g of polydextrose, 5g of vine tea, 3g of stevia rebaudiana, 10g of cinnamon and 15g of poria cocos.
In the wheat seedling traditional Chinese medicine composition, the wheat seedlings are wheat seedlings or barley seedlings.

3) Putting the refined liquid obtained in the step 2) into a reaction kettle, heating to 40-60 ℃, adding 5g of polydextrose, 1-5% of sorbitol, 0.1-0.15% of mint powder essence, 0.05-0.1% of citric acid and 0.3-0.5% of magnesium stearate (the weight percentages are calculated according to the refined liquid obtained in the step 2), stirring for 5-10 minutes, continuously heating to 80-100 ℃, and preserving heat for 5-10 minutes to obtain a first mixed liquid;
4) adding the fine powder obtained in the step 1) into the first mixed solution obtained in the step 3), and stirring for 5-10 minutes to obtain a second mixed solution;
5) freeze drying the second mixture at-40 deg.C to-30 deg.C for 15-24 hr, wet granulating, and tabletting to obtain chewable tablet or oral tablet, or capsule or granule.
6) And (3) sterilizing the chewable tablet finished product obtained in the step 5) by ultraviolet rays with the wavelength of 250-300 nm, packaging, and preserving at normal temperature.

1) weighing the traditional Chinese medicinal materials of wheat sprout, gynostemma pentaphylla, hawthorn, vine tea, kudzu root, hovenia dulcis thunb, fructus amomi, clove, polydextrose and stevia rebaudiana in parts by weight, adding cinnamon and poria cocos, and adding 8 times of water to extract for three times, wherein each time lasts for 1.5 hours;
2) mixing decoctions, filtering, and concentrating to relative density of 1.10; adding adjuvants such as sorbitol, herba Menthae powder essence, citric acid, etc., stirring for dissolving, and filtering to obtain filtrate;
3) adding water into the filtrate, adjusting the relative density of the solution to 1.06, bottling, sterilizing, labeling, and packaging.

The formula is as follows:
20g of wheat seedlings, 20g of gynostemma pentaphylla, 15g of hawthorn, 15g of radix puerariae, 15g of hovenia dulcis thunb, 5g of fructus amomi, 5g of clove, 5g of polydextrose, 5g of vine tea, 3g of stevia rebaudiana, 10g of cinnamon and 15g of poria cocos.
The preparation process of the chewable tablet of the composition preparation comprises the following steps:
1) weighing wheat seedlings according to weight, boiling the wheat seedlings (wheat seedlings or barley seedlings) for 5-10 minutes, and drying at low temperature for 1-2 hours, so as to reduce nutrition loss and keep the original flavor and taste of the wheat seedlings; grinding the wheat seedlings to reach the fineness of 800-;
2) adding gynostemma pentaphyllum, hawthorn, vine tea, kudzuvine root, hovenia dulcis thumb, stevia rebaudiana, poria cocos and cinnamon into 6 times of water by weight of the medicine, decocting for 30-45 minutes, adding fructus amomi and clove, decocting for 3-10 minutes, preferably 5 minutes, and keeping volatile active ingredients of the fructus amomi and clove from volatilizing to obtain a decoction; concentrating the Chinese medicinal water extract to 1: 1-1: 2 (ml: g), cooling the medicinal liquid, slowly adding ethanol under stirring to reach specified ethanol content of 70-80%, sealing, refrigerating for 24-48 hr, filtering, and recovering ethanol from the filtrate to obtain refined solution.
3) Putting the refined liquid obtained in the step 2) into a reaction kettle, heating to 40-60 ℃, adding 5g of polydextrose, 3% of sorbitol, 0.12% of mint powder essence, 0.08% of citric acid and 0.4% of magnesium stearate (calculated by the weight percentage of the refined liquid obtained in the step 2), stirring for 5-10 minutes, continuously heating to 80-100 ℃, and preserving heat for 5-10 minutes to obtain a first mixed liquid;
4) adding the fine powder obtained in the step 1) into the first mixed solution obtained in the step 3), and stirring for 5-10 minutes to obtain a second mixed solution;
5) freeze drying the second mixture at-40 deg.C to-30 deg.C for 15-24 hr, granulating by wet method, and tabletting to obtain chewable tablet (0.5 g/tablet or 0.3 g/tablet), 1-3 tablets for each administration.
6) And (3) sterilizing the chewable tablet finished product obtained in the step 5) by ultraviolet rays with the wavelength of 250-300 nm, packaging, and preserving at normal temperature.
The preparation process of the oral beverage of the composition preparation comprises the following steps:

2) mixing decoctions, filtering, and concentrating to relative density of 1.10; adding appropriate amount of flavoring agent (or called additive) such as sorbitol, herba Menthae powder essence, citric acid, etc., stirring for dissolving, and filtering to obtain filtrate;
3) adding water into the filtrate, adjusting the relative density of the solution to 1.06, bottling, sterilizing, labeling, and packaging to obtain beverage or solution. Specification: 125 ml/bottle or 330 ml/bottle or 500 ml/bottle.
Examples the recipe of the composition of the Maiqingmiao traditional Chinese medicine, each example and comparative example is shown in Table 1.

In conclusion, the traditional Chinese medicine composition with the anti-alcoholism and health-care effects can obviously prolong the drunk latency period and obviously reduce the blood ethanol content within two hours after drinking; the enlightenment can effectively improve the total activity of the alcohol dehydrogenase and the acetaldehyde dehydrogenase, and has obvious antialcoholic and health-care effects.
Hangover alleviation test for healthy volunteers

Healthy volunteers were recruited 60, male, 20-30 years old, with no history of disease and no allergy to alcohol. Volunteers were divided into 6 groups: positive control group a, medication test groups B1 and B2, control test group C, control test group D, control test group E, 10 people per group. Wherein, the positive control group takes the Riben Anhuixian pill for relieving alcoholism, and the medication test groups B1 and B2 respectively take the chewable tablets and the oral liquid of example 1, and the comparison test group C, the comparison test group D and the comparison test group E respectively take the chewable tablets of comparison examples 1, 2 and 3. The volunteers drunk wine at the same time, and drunk the Jiangjin white spirit with the alcohol content of 52 degrees within a specified time. The time each volunteer was awake was measured.

The results in table 3 show that the anti-hangover Chinese medicinal composition of the present invention has significant advantages over the Chinese medicinal compositions of the comparative examples in the time of sobering up. The results of animal experiments and human body experiments show that the wheat seedling Chinese medicinal composition has very good anti-inebriation effect. The composition of the invention is also proved to have obviously different antialcoholic effects due to the deletion of components, particularly wheat seedlings or the replacement change of other components, and the antialcoholic efficacy of the composition is proved to be extremely superior.
The anti-alcoholism liver-protecting effect of the anti-alcoholism traditional Chinese medicine composition on acute alcoholism of rats is researched, and related action mechanisms are discussed.
The method comprises the step of randomly dividing 45 SD rats into a normal group (15 rats), a model group (15 rats) and an experimental group (15 rats) (the anti-alcoholism traditional Chinese medicine composition beverage of the invention is taken by filling).
Preparation of the test: the sobering-up traditional Chinese medicine composition beverage of the embodiment 1 of the invention is used for intragastric administration in a dosage of 1.5ml/100gwt, twice a day, 7h interval every time, and the normal group and the model group are administrated with the same dosage of drinking water for intragastric administration for 3 d. After the 2 nd administration on the third day, after fasting and water deprivation for 14 hours, the experimental group is subjected to 1.5ml/100gwt intragastric administration on the hangover-alleviating traditional Chinese medicine composition beverage, and the normal group and the model group are subjected to intragastric administration on the same amount of drinking water. After 1h, the model and experimental groups replicated the rat alcoholism model by gavage with 55% ethanol at 1.5ml/100 gpt.
Observation indexes are as follows: after the stomach is perfused with ethanol, observing the disappearance time of turn-over reflex and the lethargy time of the rats after the alcohol is perfused, carrying out intraperitoneal injection on 3 percent sodium pentobarbital for anesthetizing the rats 24 hours later, and reserving serum and liver tissue samples for detecting related indexes, namely AST and ALT activities of the serum; liver tissue HE staining; ③ measuring the alcohol metabolizing enzyme index, namely the Acetaldehyde Dehydrogenase (ADH) activity of serum and liver tissues; and fourthly, lipid peroxidation indexes, namely the content of reduced Glutathione (GSH) and serum iron in the liver, the activity of glutathione-peroxidase (GSH-Px), Nitric Oxide Synthase (NOS) and Inducible Nitric Oxide Synthase (iNOS).
And (3) test results:
compared with a model group, the disappearance time of the turning-over reflex of the rats in the experimental group after the rats are filled with alcohol is obviously prolonged (P0.01), and the lethargy time is shortened (P0.05);
compared with a model group, the serum AST activity of an experimental group is remarkably reduced (P0.01), the ALT activity has a descending trend, but no statistical difference (P0.05) exists;
the HE staining of the liver tissue shows that the fatty degeneration of the liver cells of the experimental group is reduced compared with the fatty degeneration of the liver cells of the model group, and the formation of fat vacuoles in the liver cells is reduced compared with the fatty vacuoles of the model group;
compared with a model group, the ADH activity of the liver tissue in the alcohol metabolizing enzyme indexes of the experimental group is obviously improved (P0.01).
Compared with the model group, the GSH activity of the liver tissue of the experimental group is obviously improved (P0.01), and the TNOS and iNOS activities of the liver tissue are obviously reduced (P0.05).
The conclusion is that the anti-alcoholism traditional Chinese medicine composition has the effects of protecting liver and relieving alcoholism on rats with acute alcoholism for 24 hours. The anti-alcohol action mechanism is as follows: slow down the absorption of ethanol in vivo, promote the ADH activity of the liver and accelerate the metabolism of ethanol. The liver protection mechanism of the medicine is to improve GSH level, inhibit TNOS and iNOS expression, protect mitochondria and reduce lipid peroxidation.
Meanwhile, animal and human experiments using the compositions formulated in examples 2 to 4 found that they had the same anti-hangover effect and liver-protecting effect as the composition of example 1.
Simple variations of the composition ingredients within the spirit of the present invention, including combinations of extracts of each of the medicinal materials or single actives thereof in the composition, are also within the scope of the present invention.
