Abstract
本发明公开了一种鲍鱼养殖方法,采用模拟海水的环境对鲍鱼进行网箱养殖,可及时补充养殖池营养,避免了自然海水对鲍鱼养殖的影响,配合加硒饲料的喂养,可获得高产、高营养的富硒鲍鱼产品。所述加硒饲料由鲍鱼壳粉、羊栖菜提取物、马尾藻粉、龙须菜提取物、乌贼内脏粉、蚕蛹粉、柠檬皮粉、蛋氨酸硒、红薯叶粉、甜菜粉、鸡肉粉、香菇粉、牛肉粉组成。

本发明涉及水产动物养殖技术领域,具体涉及一种鲍鱼养殖方法。
背景技术
鲍鱼是名贵的“海珍品”之一,味道鲜美,营养丰富,被誉为海洋“软黄金”。鲍鱼是名贵的海洋食用贝类,被誉为“餐桌黄金,海珍之冠”,其肉质细嫩、营养丰富。
鲍鱼营养丰富,干品中富含蛋白质,脂肪,糖类,同时还富含多种生理活性物质如EPA、DHA、牛磺酸、超氧化物歧化酶等,对维持机体酸碱平衡、神经肌肉兴奋方面具有重要作用的金属元素(Ca2+、Mg2+等)的含量也较丰富。研究发现,鲍鱼肌肉酶解物可以显著提高小鼠机体运动耐力、应激能力和免疫功能,同时对学习和记忆有明显增强作用。鲍鱼肌肉的水提取上清液有很明显的抗凝血作用,对增强家兔纤维蛋白溶解活性有非常显著的作用,这为鲍鱼活血作用提供了佐证。还有人体所需的钙、磷、铁等矿物质和多种维生素。
冬季鲍鱼中胶原蛋白占总蛋白含量高达30%~50%,远高于一般鱼贝类。研究发现表明,胶原蛋白含有多种生物活性肽,具有很好的生理功能,如抗氧化、将血压、预防关节炎、保护胃粘膜和抗溃疡、促进皮肤胶原代谢等。此外,肌肉高含量的胶原蛋白对鲍鱼的质地也有较大影响。
硒是人和动物所必需的微量元素,对人体生命活动起着重要的作用,是多种酶,如:谷胱甘肽过氧化物酶(GDH-PX)等、免疫球蛋白的组成成份,人们通过补硒可以达到提高机体免疫功能,抗病能力,延缓衰老的目的。
由于天然产量很少,因此价格昂贵。而人工养殖也存在成本高、产量低等问题。近年来,海参鲍鱼人工养殖迅速发展,其主要养殖形式有工厂化养殖、筏式养殖、海底底播养殖和潮间带网箱养殖,由于海水的稀释效应,微生物制剂和营养添加剂稳定性差,被海参鲍鱼摄入口中的效率很低,多数溶解散失于海水中或沉入底泥,不能有效利用,反而造成环境富营养化,污染水质,进而引发水产疾病。
发明内容
为了解决上述问题,本发明提供了一种鲍鱼养殖方法,采用模拟海水的环境对鲍鱼进行网箱养殖,可及时补充养殖池营养,避免了自然海水对鲍鱼养殖的影响,配合加硒饲料的喂养,可获得高产、高营养的富硒鲍鱼产品。
本发明可以通过以下技术方案来实现:
一种鲍鱼养殖方法,包括以下步骤:
(1)控制水温23-28℃,水中溶氧量4%以上,海水盐度25-35%,pH为7-8,加入免疫调节剂使水体免疫调节剂浓度为5-7%,每3d换水一次;
(2)放养密度为800-1000个/箱,每5-7d投放1次天然饵料,投喂量为鲍鱼体重30%,每天傍晚投喂一次加硒饲料,饲料和鲍鱼重量比为1:50-100。
上步骤(1)免疫调节剂:凝结芽孢杆菌制剂2.4-2.6份、芦荟提取物0.6-0.8份、新琼寡糖0.02-0.04份、莪术16-18份、姜叶19-23份、木香20-22份、当归6-8份、扁豆花18-22份、枫荷梨8-10份、黄芩0.5-0.7份、无花果3.5-3.7份、土茯苓2.2-2.4份、山楂4-5份;所述免疫调节剂的制备是:将中药成分熬制5-7h,冷却后加入凝结芽孢杆菌制剂和芦荟提取物,充分混合。
所述凝结芽孢杆菌制剂:以20L发酵罐为平台,对凝结芽孢杆菌进行发酵,发酵结束后,按1:1的比例与麦麸进行吸附,充分混匀后,在40℃恒温中与等量金霉素和2(3,5-二叔丁基-2-羟苯基)-5氯苯并三唑,进行联合干燥至含水量为8-10%。
所述芦荟提取物:采集芦荟新鲜叶子,去皮,将叶肉均质5min,然后过滤得滤液,在4℃下经8000r/min离心10min,取上清液加入2倍冷丙酮,静置30min后收集沉淀,溶于2倍体积的蒸馏水中,用等量饱和硫酸铵盐析,置于4℃下12h,经8000r/min离心10min后收集沉淀,冻成干粉。
上步骤(2)加硒饲料:鲍鱼壳粉9-11份、羊栖菜提取物15-17份、马尾藻粉63-65份、龙须菜提取物5-7份、乌贼内脏粉2-4份、蚕蛹粉6-7份、柠檬皮粉2-2.4份、蛋氨酸硒0.06-0.08份、红薯叶粉12-18份、甜菜粉11-13份、鸡肉粉2-5份、香菇粉11-13份、牛肉粉4-6份。
所述羊栖菜提取物:
a、将羊栖菜干燥粉碎,过60目筛,加入料液比为1:20的90%乙醇,在60℃下回流浸提4h,真空抽滤后,取滤液于60℃,5-7kPa下减压浓缩,然后干燥;
b、收集真空抽滤的滤渣,在85℃下用2倍体积水浸提4h后过滤,向滤液中加入3倍滤液体积的95%乙醇,静置30min,经4000r/min离心5min,取沉淀加水搅拌5min,再经4000r/min离心后取上清液真空冻干;
c、将滤液和滤渣提取所得物混合均匀。
所述龙须菜提取物:将龙须菜干燥粉碎,加入料液比为1:20的12%NaOH,在60℃下处理60min,过滤,取滤渣在58℃下干燥;所述12%NaOH是采用盐度为15-20%的氯化钠溶液配制而成的。
所述加硒饲料的制备是,将各原料分别粗粉碎混合均匀,放入超微粉碎机中粉碎至80目,用水产饲料硬颗粒制粒机制成颗粒,熟化、烘干后再次粉碎300-500目。

本发明涉及一种鲍鱼养殖方法,免疫调节剂中添加凝结芽孢杆菌制剂,能够提高鲍鱼机体免疫力,增强消化系统酶活性,防病治病和促进生长;可以促进养殖水提饵料生物大量繁殖、生长,并且可分解残饵、粪便及水肿有机质,改良水体环境,抑制有害菌繁殖生长,提高鲍鱼机体抗菌防御活性;能有效调整水产鲍鱼肠道的菌群平衡,增强抗感染能力,提高鲍鱼的存活率;联合干燥凝结芽孢杆菌制剂能够更稳定持久地发挥效用;
本发明利用芦荟提取物作为鲍鱼外源免疫分子,特异性地和菌体表面的糖链及糖结合蛋白相结合,可特异地干扰病原体和宿主的结合,可增强其免疫系统的功能,避免病虫害的发生;
本发明添加的新琼寡糖具有较强的抗氧化、抗炎、及抗淀粉老化等活性,可提高鲍鱼血清的溶酶菌活性、补体活性以及鲍鱼血液白细胞总数和白细胞吞噬率,显著增强鲍鱼的免疫能力;
本发明饲料中羊栖菜提取物的加入可促进鲍鱼生长,在鲍鱼体内产生各种消化酶和维生素,提高饲料的营养含量,其中含有的生物活性物质被吸收参与能量和维生素代谢,可提高饲料转化率,加强鲍鱼体内的营养代谢;还具有促进钙、磷及微量元素的吸收利用;
本发明龙须菜提取物的加入能够延长加硒饲料在鲍鱼消化系统中停留的时间,增加了消化酶与营养物质作用的时间,能够更加高效地利用加硒饲料中的营养物质;
本发明使用的加硒饲料养殖鲍鱼,提高了鲍鱼自身的免疫力的同时有效提高鲍鱼体内的硒含量,能够达到食用者补硒的作用,同时对鲍鱼的经济化养殖具有现实意义。

实施例1
一种鲍鱼养殖方法,主要包括以下步骤:
(1)控制水温25.5℃,水中溶氧量4%以上,海水盐度30%,pH为7.5,加入免疫调节剂使水体免疫调节剂浓度为6%,每3d换水一次;
(2)放养密度为900个/箱,每6d投放1次天然饵料,投喂量为鲍鱼体重30%,每天傍晚投喂一次加硒饲料,饲料和鲍鱼重量比为1:75。
本实施例一种鲍鱼养殖方法,所述步骤(1)免疫调节剂:凝结芽孢杆菌制剂2.5份、芦荟提取物0.7份、新琼寡糖0.03份、莪术17份、姜叶21份、木香21份、当归7份、扁豆花20份、枫荷梨9份、黄芩0.6份、无花果3.6份、土茯苓2.3份、山楂4.5份;所述免疫调节剂的制备是:将中药成分熬制6h,冷却后加入凝结芽孢杆菌制剂和芦荟提取物,充分混合。
上述凝结芽孢杆菌制剂的制取:以20L发酵罐为平台,对凝结芽孢杆菌进行发酵,发酵结束后,按1:1的比例与麦麸进行吸附,充分混匀后,在40℃恒温中与等量金霉素和2(3,5-二叔丁基-2-羟苯基)-5氯苯并三唑,进行联合干燥至含水量为9%。
上述芦荟提取物的制取:采集芦荟新鲜叶子,去皮,将叶肉均质5min,然后过滤得滤液,在4℃下经8000r/min离心10min,取上清液加入2倍冷丙酮,静置30min后收集沉淀,溶于2倍体积的蒸馏水中,用等量饱和硫酸铵盐析,置于4℃下12h,经8000r/min离心10min后收集沉淀,冻成干粉。
本实施例一种鲍鱼养殖方法,所述步骤(2)加硒饲料为:鲍鱼壳粉10份、羊栖菜提取物16份、马尾藻粉64份、龙须菜提取物6份、乌贼内脏粉3份、蚕蛹粉6.5份、柠檬皮粉2.2份、蛋氨酸硒0.07份、红薯叶粉15份、甜菜粉12份、鸡肉粉3.5份、香菇粉12份、牛肉粉5份。
上述羊栖菜提取物的制取步骤如下:
(1)将羊栖菜干燥粉碎,过60目筛,加入料液比为1:20的90%乙醇,在60℃下回流浸提4h,真空抽滤后,取滤液于60℃,6kPa下减压浓缩,然后干燥;
(2)收集真空抽滤的滤渣,在85℃下用2倍体积水浸提4h后过滤,向滤液中加入3倍滤液体积的95%乙醇,静置30min,经4000r/min离心5min,取沉淀加水搅拌5min,再经4000r/min离心后取上清液真空冻干;
(3)将滤液和滤渣提取所得物混合均匀。
上述龙须菜提取物的制取:将龙须菜干燥粉碎,加入料液比为1:20的12%NaOH,在60℃下处理60min,过滤,取滤渣在58℃下干燥;所述12%NaOH是采用盐度为17.5%的氯化钠溶液配制而成的。
上述加硒饲料的制备是,将各原料分别粗粉碎混合均匀,放入超微粉碎机中粉碎至80目,用水产饲料硬颗粒制粒机制成颗粒,熟化、烘干后再次粉碎400目。
实施例2
一种鲍鱼养殖方法,主要包括以下步骤:

效果验证选择在潮间带一个条件适宜的地方,围堤建池、池内放置石块,水泥板,瓦片等作鲍的附着隐蔽物,向海一侧的池壁上部留有若干个通水口,作为张落潮时池内与外海水的交换通道,落潮后应使池内海水保持1米左右的水深,养殖器材为2米×2米×0.8米钢骨架大型网箱,下沉于池底进行养殖,按照上述实施例1~3和对比例1~3的方法分别进行鲍鱼的养殖,养殖完一个周期分别对上述方法得出的效果进行评价,结果如下:
由上表分析可得,实施例1~3之间的数据相差不大,在鲍鱼的抗病、抗逆、特定生长率、鲍肉的含量指标均比对比例的要好,说明要达到实施例那样的效果,本发明的步骤成分缺一不可;
实施例1和对比例1相比较,在加硒饲料中添加羊栖菜提取物和龙须菜提取物,产生各种消化酶和维生素,参与能量和维生素代谢,促进钙、磷及微量元素的吸收利用,延长加硒饲料在鲍鱼消化系统中停留的时间,使吸收更充分,所以在特定生长率和鲍鱼品质方面比对比例1明显要好;
对比例2、对比例3比较于实施例1,各项目有所差距,尤其在抗病能力增长率、抗逆能力增长率上,所以它们两者的情况的致病、致死相对较多,其中对比例2缺乏的凝结芽孢杆菌群会分泌蛋白酶、淀粉酶、脂肪酶以及多种氨基酸,起到调整水中菌群、抑制病菌害菌等作用,提高鲍鱼机体免疫力,增强消化系统酶活性,防病治病和促进生长;同时促进养殖水提饵料生物大量繁殖、生长,并且可分解残饵、粪便及水肿有机质,改良水体环境,抑制水提有害菌繁殖生长,调节菌藻平衡,能够激活宿主细胞并提高细胞吞噬活性,提高鲍鱼机体抗逆活性;对比例3缺乏的新琼寡糖有抗氧化、抗炎、及抗淀粉老化等活性,可以提高鲍鱼血清的溶酶菌活性、补体活性以及鲍鱼血液白细胞总数和白细胞吞噬率,从而提高鲍鱼的抗病能力增长率。
以上所述实施例仅表达了本发明的几种实施方式,其描述较为具体和详细,但并不能因此而理解为对本发明专利范围的限制。应当指出的是,对于本领域的普通技术人员来说,在不脱离本发明构思的前提下,还可以做出若干变形和改进,这些都属于本发明的保护范围。
Claims (8)
1.一种鲍鱼养殖方法,其特征在于,包括以下步骤:
(1)控制水温23-28℃,水中溶氧量4%以上,海水盐度25-35%,pH为7-8,加入免疫调节剂使水体免疫调节剂浓度为5-7%,每3d换水一次;
(2)放养密度为800-1000个/箱,每5-7d投放1次天然饵料,投喂量为鲍鱼体重30%,每天傍晚投喂一次加硒饲料,饲料和鲍鱼重量比为1:50-100。
2.根据权利要求1所述的一种鲍鱼养殖方法,其特征在于,所述步骤(1)免疫调节剂:凝结芽孢杆菌制剂2.4-2.6份、芦荟提取物0.6-0.8份、新琼寡糖0.02-0.04份、莪术16-18份、姜叶19-23份、木香20-22份、当归6-8份、扁豆花18-22份、枫荷梨8-10份、黄芩0.5-0.7份、无花果3.5-3.7份、土茯苓2.2-2.4份、山楂4-5份;所述免疫调节剂的制备是:将中药成分熬制5-7h,冷却后加入凝结芽孢杆菌制剂和芦荟提取物,充分混合。
3.根据权利要求2所述的一种鲍鱼养殖方法,其特征在于,所述凝结芽孢杆菌制剂:以20L发酵罐为平台,对凝结芽孢杆菌进行发酵,发酵结束后,按1:1的比例与麦麸进行吸附,充分混匀后,在40℃恒温中与等量金霉素和2(3,5-二叔丁基-2-羟苯基)-5氯苯并三唑,进行联合干燥至含水量为8-10%。
4.根据权利要求2所述的一种鲍鱼养殖方法,其特征在于,所述芦荟提取物:采集芦荟新鲜叶子,去皮,将叶肉均质5min,然后过滤得滤液,在4℃下经8000r/min离心10min,取上清液加入2倍冷丙酮,静置30min后收集沉淀,溶于2倍体积的蒸馏水中,用等量饱和硫酸铵盐析,置于4℃下12h,经8000r/min离心10min后收集沉淀,冻成干粉。
5.根据权利要求1所述的一种鲍鱼养殖方法,其特征在于,所述步骤(2)加硒饲料:鲍鱼壳粉9-11份、羊栖菜提取物15-17份、马尾藻粉63-65份、龙须菜提取物5-7份、乌贼内脏粉2-4份、蚕蛹粉6-7份、柠檬皮粉2-2.4份、蛋氨酸硒0.06-0.08份、红薯叶粉12-18份、甜菜粉11-13份、鸡肉粉2-5份、香菇粉11-13份、牛肉粉4-6份。
6.根据权利要求5所述的一种鲍鱼养殖方法,其特征在于,所述羊栖菜提取物:
(1)将羊栖菜干燥粉碎,过60目筛,加入料液比为1:20的90%乙醇,在60℃下回流浸提4h,真空抽滤后,取滤液于60℃,5-7kPa下减压浓缩,然后干燥;
(2)收集真空抽滤的滤渣,在85℃下用2倍体积水浸提4h后过滤,向滤液中加入3倍滤液体积的95%乙醇,静置30min,经4000r/min离心5min,取沉淀加水搅拌5min,再经4000r/min离心后取上清液真空冻干;
(3)将滤液和滤渣提取所得物混合均匀。
7.根据权利要求5所述的一种鲍鱼养殖方法,其特征在于,所述龙须菜提取物:将龙须菜干燥粉碎,加入料液比为1:20的12%NaOH,在60℃下处理60min,过滤,取滤渣在58℃下干燥;所述12%NaOH是采用盐度为15-20%的氯化钠溶液配制而成的。
8.根据权利要求5所述的一种鲍鱼养殖方法,其特征在于,所述加硒饲料的制备是,将各原料分别粗粉碎混合均匀,放入超微粉碎机中粉碎至80目,用水产饲料硬颗粒制粒机制成颗粒,熟化、烘干后再次粉碎300-500目。
