技术领域
本公开内容主要涉及营养组合物领域,例如针对儿童的基于强化乳的营养组合物,所述组合物包含脂肪或脂质源、碳水化合物源和蛋白质源。
发明背景
当前存在多种膳食组合物,其力图支持儿童的正常成长和发育,并促进他们的终生健康。通常这样的膳食组合物包含大量的碳水化合物(天然存在的和精制的两种),以为成长中的儿童提供营养。
尽管针对年轻人的膳食组合物中碳水化合物普遍存在,但那些碳水化合物的来源和组成的重要性常常被忽略。因此,本公开内容的一个目标是提供具有针对性的碳水化合物组成的营养组合物。
本发明公开内容
简要地说,在一个实施方案中,本公开内容涉及包含乳糖和任选果糖的营养组合物。在一个实施方案中,营养组合物为针对儿童的基于强化乳的营养组合物,并包含:
a. 至多约5.5 g/100 kcal的脂肪或脂质源,更优选约1 g/100 kcal至约4.5 g/100kcal的脂肪或脂质源;
b. 至多约6 g/100 kcal的蛋白质源,更优选约2 g/100 kcal至约5 g/100 kcal的蛋白质源;和
c. 碳水化合物源,使得营养组合物包含至多约18 g/100 kcal的总碳水化合物,其中按重量计至少60%的总碳水化合物包括乳糖、果糖或其组合。在一些实施方案中,所述营养组合物包括针对儿童的成长乳。
优选地,营养组合物包含约11.5 g /100 kcal至约16 g/100 kcal的总碳水化合物,更优选约12 g/100 kcal至约15.5 g/100 kcal的总碳水化合物。
在某些实施方案中,碳水化合物源包含小于约0.5 g/100 kcal添加的蔗糖。在其它实施方案中,碳水化合物源包含小于约2.0 g/100 g添加的蔗糖。此外,碳水化合物源优选包含小于约1 g/100 kcal的麦芽糊精和/或小于约6 g/100 kcal的玉米糖浆固体。在其它实施方案中,碳水化合物源不包含添加的蔗糖、不包含添加的玉米糖浆固体和/或不包含添加的麦芽糊精。在某些实施方案中,营养组合物的血糖指数小于约60。
实施本发明的最佳方式
正如所提到的,本公开内容涉及营养组合物,例如针对儿童的基于强化乳的营养组合物,并包含:
a. 至多约5.5 g/100 kcal的脂肪或脂质源,优选约1 g/100 kcal至约5.5 g/100kcal的脂肪或脂质源,更优选约1 g/100 kcal至约4.5 g/100 kcal的脂肪或脂质源;
b. 至多约6 g/100 kcal的蛋白质源,优选约2 g/100 kcal至约6 g/100 kcal的蛋白质源,更优选约2 g/100 kcal至约5 g/100 kcal的蛋白质源;和
c. 碳水化合物源,其中所述营养组合物包含至多约18 g/100 kcal的总碳水化合物,并且此外当表述为重量百分数时,其中乳糖和果糖构成总碳水化合物的至少约60%。在一些实施方案中,所述营养组合物包含针对儿童的成长乳。
如本文所使用的,“总碳水化合物”指通过包含在组合物的其它组分或成分中而存在于营养组合物中的碳水化合物(称为内源性碳水化合物),以及自身添加的碳水化合物(称为添加的碳水化合物)。例如,当使用牛乳蛋白质源时,其包含碳水化合物,所述碳水化合物被归类为内源性碳水化合物。并且,如本文所用的“儿童”或“儿童们”定义为年龄在12个月至约12岁之间的人类。

在一些实施方案中,总碳水化合物包含按重量计至少约60%的乳糖。更优选,总碳水化合物包含按重量计至少约70%或至少约80%的乳糖。例如,总碳水化合物可包含按重量计约60%至约100%的乳糖。乳糖可作为所公开的营养组合物中存在的其它组分的成分存在(称为内源性乳糖),或其可单独添加。最通常地,乳糖为内源性乳糖和添加的乳糖二者的组合。
在其中存在果糖的本公开内容的营养组合物的实施方案中,总碳水化合物中存在的乳糖和果糖的组合量优选大于或等于总碳水化合物重量的约60%。更优选地,所述乳糖和果糖的组合量大于或等于总碳水化合物重量的约70%。优选地,在这样的实施方案中,总碳水化合物包含按重量计至少约5%、更优选至少约10%的果糖;更特别地,在一些实施方案中,总碳水化合物包含按重量计约10%至约25%的果糖。

本公开营养组合物的一些实施方案中存在的总乳糖(内源性和添加的)为约6.0g/100 kcal至约11.7 g/100 kcal,更优选约6.5 g/100 kcal至约11.2 g/100 kcal。添加的乳糖在约0.8 g/100 kcal至5.4 g/100 kcal之间,更优选约2.5 g/100 kcal至约5.2 g/100 kcal。
在一些实施方案中,总乳糖在约29 g/100 g至约48 g/100 g营养组合物之间,更优选在约30.5 g/100g至约46 g/100 g之间。添加的乳糖有利地在约3.5 g/100 g至约18.5g/100 g营养组合物之间,更优选在约9.5 g/100 g至约17 g/100 g之间。
营养组合物的某些实施方案中存在的果糖至多约3.6 g/100 kcal,更优选约1.4g/100 kcal至约3.2 g/100 kcal。在其它实施方案中,果糖至多约12.5 g/100 g,更优选约7 g/100g至约12 g/100 g。
果糖和乳糖的血糖指数(GI)低于营养组合物中通常使用的某些膨胀成分和甜味剂,例如玉米糖浆固体、蔗糖和麦芽糊精。因此,在某些实施方案中,营养组合物提供具有低血糖指数的优势。例如,在某些实施方案中,营养组合物的血糖指数低于约60,例如在约15-约60之间(更优选约15-约55,并且在一些实施方案中,低于约41、低于约36或低于约25)。
如本领域所已知的,血糖指数(GI)区分在消化过程中具有快速分解并迅速将葡萄糖释放入血流中倾向的碳水化合物(所谓的高血糖指数碳水化合物)和分解得更缓慢并更为逐渐地将葡萄糖释放入血流的碳水化合物。属于后一个范畴的碳水化合物(所谓的低血糖指数碳水化合物)在控制血糖中常常被视为潜在有利的。
通过向10个健康人一次施用50克葡萄糖(葡萄糖参照组合物)和另一次施用营养组合物来确定所述营养组合物的血糖指数。所施用的营养组合物的量为使得其包含50克可用的碳水化合物。然后在施用营养组合物和葡萄糖参照组合物后的两小时期间获取血液样品,并用于构建葡萄糖反应曲线。接着计算曲线下面积(AUC),并反映施用营养组合物或葡萄糖参照组合物后血糖(葡萄糖)水平的总上升。对于每个受试者,营养组合物的AUC除以葡萄糖参照组合物的AUC再乘以100得出每个受试者的血糖指数。将对受试者所观察到的血糖指数值平均,确定营养组合物的血糖指数值。
较低的血糖反应通常导致葡萄糖更持续地释放到血液中。因此,在本公开内容提供具有低血糖指数的营养组合物的实施方案中,这样的组合物可以支持持续的脑力、改善的认知发育和/或改善的认知功能以及健康成长的方法施用给儿童。
除了乳糖和任选果糖外,营养组合物可包含另外的较低血糖指数的碳水化合物膨胀成分,包括但不限于抗性淀粉、纤维和/或益生元,和/或另外的较低血糖指数的甜味剂,包括但不限于异麦芽酮糖(palitinose)、塔格糖、糖醇和/或非营养性甜味剂。
如所提到的,本公开内容的营养组合物包含脂肪或脂质源。用于实施本公开内容的合适的脂肪或脂质源可为本领域已知或使用的任何脂肪或脂质源,包括但不限于动物来源,例如乳脂、奶油、奶脂、蛋黄脂质;海产来源,例如鱼油、海产油、单细胞油;蔬菜和植物油,例如玉米油、芥花籽油、葵花油、大豆油、棕榈油、椰子油、高油葵花油、月见草油、菜籽油、橄榄油、亚麻仁(亚麻籽)油、棉籽油、高油红花油、棕榈硬脂、棕榈仁油、麦胚芽油;中链甘油三酸酯油和乳液及脂肪酸酯;以及其任何组合。
本公开内容的营养组合物进一步包含蛋白质源。可用于实施本公开内容的蛋白质源包括但不限于牛乳蛋白源,例如乳蛋白粉、乳蛋白浓缩物、乳蛋白分离物、脱脂乳固体、脱脂乳、脱脂奶粉、乳清蛋白、乳清蛋白分离物、乳清蛋白浓缩物、甜乳清、酸乳清、酪蛋白、酸性酪蛋白、皱胃酪蛋白、酪乳粉、共沉淀乳粉、酪蛋白酸钾、酪蛋白酸盐(例如酪蛋白酸钠、酪蛋白酸钙钠、酪蛋白酸钙)及其任何组合。在一个实施方案中,蛋白质源包含约10%至约40%的乳清蛋白。在另一个实施方案中,蛋白质源包含约50%至约80%的酪蛋白。
在一个实施方案中,蛋白质作为完整蛋白质提供。在其它实施方案中,蛋白质作为完整蛋白质和部分水解蛋白的组合(水解程度在约4%至约10%之间)提供。在又一个实施方案中,蛋白质源可添加有包含谷氨酰胺的肽。
本公开内容的基于强化乳的营养组合物可为任何形式。优选地,基于强化乳的营养组合物为液体形式或粉末形式。优选地,如果以粉末形式提供,营养组合物在施用于人之前在液体例如水中复原。优选地,营养组合物作为多样化饮食的一部分施用给儿童。更优选地,将营养组合物施用于年龄在约1岁至约12岁之间的儿童。还优选的是,营养组合物具有可接受的感觉和感官特征,例如可接受的味道、质地和/或气味。
在本公开内容的优选实施方案中,基于强化乳的营养组合物为成长乳或在液体(例如水)中复原时产生成长乳的产品(例如粉)。如普通技术人员所已知的,术语“成长乳”涉及基于强化乳的饮料范畴,其意在用作多样化饮食的一部分,以支持儿童的正常成长和发育。成长乳在本领域中为人所熟知,并且包括但不限于Mead Johnson Nutrition出售的Enfagrow®和Enfakid®。

本公开内容营养组合物还可包含一种或多种益生元,其通常被认为是总碳水化合物的一部分。如本文所使用的术语“益生元”意指不易消化的食物成分,其可通过选择性刺激结肠中可改善宿主健康的一种或有限种细菌的生长和/或活性而对宿主产生有益影响。“益生元组合物”为包含一种或多种益生元的组合物。这样的益生元可为天然存在的、合成的或通过对生物和/或植物的基因操作开发,无论这样的新来源是目前已知还是日后开发的。
用于本公开内容的益生元可包括寡糖类、多糖类和其它包含果糖、木糖、大豆、半乳糖、葡萄糖和甘露糖的益生元。更特别地,用于本公开内容的益生元可包括乳果糖、乳蔗糖、棉子糖、葡萄糖-寡糖、菊粉、聚葡萄糖、聚葡萄糖粉、半乳糖-寡糖、果糖-寡糖、异麦芽糖-寡糖、大豆-寡糖、乳蔗糖、木糖-寡糖、壳糖-寡糖、甘露糖-寡糖、阿拉伯糖-寡糖、唾液酸-寡糖、岩藻糖-寡糖和龙胆糖-寡糖。优选地,营养组合物包含聚葡萄糖、半乳糖-寡糖、果糖-寡糖和/或菊粉。例如,营养组合物可包含聚葡萄糖与半乳糖-寡糖的组合或者果糖-寡糖与菊粉的组合;或者,益生元组合可包含聚葡萄糖与果糖-寡糖,或者菊粉与半乳糖-寡糖。在某些实施方案中,本公开内容的组合物中包含的益生元包括美国专利号7,572,474中教导的那些,其公开内容通过引用结合到本文中。
如果在营养组合物中包含益生元,则所述营养组合物中存在的益生元的总量可为约0.1 g/100 kcal至约2 g/100 kcal。更优选地,营养组合物中存在的益生元的总量为约0.3 g/100 kcal至约1 g/100 kcal。
如果在益生元组合物中使用聚葡萄糖,则在一个实施方案中,所述营养组合物中聚葡萄糖的量可在约0.1 g/100 kcal至约1.0 g/100 kcal的范围内。在另一个实施方案中,聚葡萄糖的量在约0.2 g/100 kcal至约0.5 g/100 kcal的范围内。
如果在益生元组合物中使用半乳糖-寡糖,则在一个实施方案中,所述营养组合物中寡聚半乳糖的量可为约0.1 g/100 kcal至约0.5 g/100 kcal。在另一个实施方案中,所述营养组合物中半乳糖-寡糖的量可为约0.2 g/100 kcal至约0.5 g/100 kcal。在某些实施方案中,所述益生元组合物中聚葡萄糖与半乳糖-寡糖的比率在约9:1至约1:9之间。
在本公开内容的一个实施方案中,营养组合物可包含一种或多种益生菌。“益生菌”为对宿主健康发挥有益作用的低或无致病性微生物。本领域已知的任何益生菌在本实施方案中均可接受,只要其取得预期的结果。在一个具体实施方案中,益生菌可选自乳杆菌(Lactobacillus)种、鼠李糖乳杆菌(Lactobacillus rhamnosus) GG、双歧杆菌(Bifidobacterium)种、长双歧杆菌(Bifidobacterium longum)、短双歧杆菌(Bifidobacterium brevis)和动物双歧杆菌乳亚种(Bifidobacterium animalis subsp. lactis) BB-12。
如果在组合物中包含益生菌,则所述益生菌的量可在约104至约1010菌落形成单位(cfu)/kg体重/天之间变化。在另一个实施方案中,所述益生菌的量可在约106至约109 cfu/kg体重/天之间变化。在又一个实施方案中,所述益生菌的量可至少为约106 cfu/kg体重/天。在其它实施方案中,本公开内容的营养组合物中存在的益生菌的量在1 x 107至约1 x109 cfu/g产品之间。
在一个实施方案中,所述益生菌的一种或多种是有生活力的。在另一个实施方案中,所述益生菌的一种或多种是无生活力的。如本文所使用的术语“有生活力的”指有生命的微生物。术语“无生活力的”或“无活力的益生菌”意指无生命的益生菌微生物、其细胞组分或其代谢产物。所述无生活力的益生菌可已被加热灭活或其它方式灭活,但其保留有利影响宿主健康的能力。用于本发明的益生菌可为天然存在的、合成的或通过对生物的基因操作而开发的,无论这些新的来源是目前已知还是日后开发的。
在一些实施方案中,本公开内容的营养配方可进一步包含长链多不饱和脂肪酸(LCPUFA)源。优选地,LCPUFA源包括二十二碳六烯酸(DHA)。其它合适的LCPUFA包括但不限于α-亚油酸、γ-亚油酸、亚油酸、α-亚麻酸、二十碳五烯酸(EPA)和花生四烯酸(ARA)。
在一些实施方案中,营养组合物中包含的LCPUFA为DHA,以约5 mg/100 kcal至约20 mg/100 kcal的水平存在,更优选以约6.5 mg/100 kcal至约16 mg/100 kcal的水平存在。
在另一个实施方案中,营养组合物补充有DHA和ARA二者。在该实施方案中,ARA:DHA的重量比率可为约1:3至约9:1。在本公开内容的一个实施方案中,ARA:DHA的重量比率为约1:2至约4:1。
可使用本领域已知的标准技术,向营养组合物中添加包含DHA(ARA同样如此)的油类。例如,可通过替代等量的组合物中通常存在的油将DHA添加至组合物中,所述油例如高油葵花油。如另一个实例,可通过替代等量的无DHA组合物中通常存在的其余全部脂肪共混物将包含DHA的油类添加至组合物中。
如果利用DHA源,则所述DHA源可为本领域已知的任何来源,例如海产油、鱼油、单细胞油、蛋黄脂质和脑脂质。在一些实施方案中,DHA来源于单细胞Martek油DHASCO®或其变体。DHA可为天然形式,只要LCPUFA源的其余部分对婴儿不造成任何实质性的有害作用。或者,DHA可以以精制的形式使用。
在本公开内容一个实施方案中,如美国专利号5,374,567、5,550,156和5,397,591所教导的,DHA源为单细胞油,所述专利的公开内容通过引用以其整体结合到本文中。然而,本公开内容不仅限于这些油类。
在一些实施方案中,本公开内容营养组合物还包含乳铁蛋白。乳铁蛋白为约80 kD的单链多肽,包含1-4个聚糖,取决于物种。不同物种的乳铁蛋白的3-D结构非常相似,但不相同。每个乳铁蛋白包含两个同源叶(lobe),称为N-和C-叶,分别指该分子的N-末端和C-末端部分。每个叶进一步由两个亚叶或结构域组成,此两个亚叶或结构域形成裂缝,高铁离子(Fe3+)与碳酸根(碳酸氢根)阴离子在此处紧密协同结合。这些结构域分别被称为N1、N2、C1和C2。乳铁蛋白的N-末端具有强阳离子肽区域,其决定着多种重要的结合特性。乳铁蛋白具有非常高的等电点(~pI 9),并且其阳离子性质在其抵抗细菌、病毒和真菌病原体的能力中发挥重要作用。乳铁蛋白的N-末端区域中存在数个阳离子氨基酸残基簇,介导乳铁蛋白对广泛的微生物的生物学活性。例如,人乳铁蛋白的N-末端残基1-47 (牛乳铁蛋白的1-48)对于乳铁蛋白的铁-非依赖性生物学活性是关键性的。在人乳铁蛋白中,残基2-5 (RRRR)和28-31 (RKVR)为N-末端富含精氨酸的阳离子结构域,其对乳铁蛋白的抗微生物活性尤为关键。牛乳铁蛋白中存在相似的N-末端区域(残基17-42;FKCRRWQWRMKKLGAPSITCVRRAFA)。
如刊物Biochemistry and Cell Biology, pp 275-281 (2006)所发表的“Perspectives on Interactions Between Lactoferrin and Bacteria (对乳铁蛋白与 细菌之间相互作用的看法)”中所述,来自不同宿主物种的乳铁蛋白在氨基酸序列上可能不同,尽管通常具有相对高的等电点以及在内叶的末端区域具有带正电荷的氨基酸。用于本公开内容的合适的乳铁蛋白包括与HLf (349-364)片段上的氨基酸序列AVGEQELRKCNQWSGL具有至少48%同源性的那些乳铁蛋白。例如,合适的乳铁蛋白包括但不限于人乳铁蛋白、牛乳铁蛋白、猪乳铁蛋白、马乳铁蛋白、水牛乳铁蛋白、山羊乳铁蛋白、鼠乳铁蛋白和骆驼乳铁蛋白。
在优选的实施方案中,乳铁蛋白为获自非人类来源的乳铁蛋白。如本文所使用,“获自非人类来源的乳铁蛋白”意指来自除人母乳外的来源的乳铁蛋白。例如,在某些实施方案中,乳铁蛋白为通过基因修饰的生物产生的人乳铁蛋白和/或非人乳铁蛋白。如本文所用的术语“非人乳铁蛋白”指具有与人乳铁蛋白的氨基酸序列不同的氨基酸序列的乳铁蛋白。
在一个实施方案中,乳铁蛋白以约5 mg/100 kcal至约16 mg/100 kcal的量存在于营养组合物中;在另一个实施方案中,乳铁蛋白以约9 mg/100 kcal至约14 mg/100 kcal的量存在。
在某些实施方案中,本公开内容的营养组合物还包含贝塔-葡聚糖。贝塔-葡聚糖(β-葡聚糖)为葡萄糖聚合物的不同子集,其由葡萄糖单体链组成,所述葡萄糖单体通过β型糖苷键连接在一起以形成复合的碳水化合物。源自于面包酵母菌酿酒酵母(Saccharomyces cerevisiae)的β-葡聚糖由在1和3位连接的D-葡萄糖分子链组成,其在1和6位具有葡萄糖侧链连接。酵母菌来源的β-葡聚糖为不溶性的纤维样复合糖,其具有以下通用结构:具有β-1,3主链的直链葡萄糖单元,其交替有β-1,6侧链,所述侧链长度通常为6-8个葡萄糖单元。更特别地,源自于面包酵母菌的β-葡聚糖为聚-(1,6)-β-D-吡喃葡萄糖基-(1,3)-β-D-吡喃葡萄糖。
β-1,3;1,6葡聚糖为在小儿受试者中引发免疫监视的多糖级分,当其作为营养组合物的一部分施用时,可通过刺激免疫功能减少儿童或婴儿中的微生物相关疾病。此外,β-葡聚糖在小儿受试者中耐受良好且不产生或导致过剩气体、腹胀、胃气胀或腹泻。当口服施用时,β-1,3-葡聚糖不是通过消化系统的代谢过程直接吸收。事实上,酵母菌β-葡聚糖摄入后未发生明显的全身性暴露;然而,少量的不溶性β-葡聚糖颗粒通过小肠的淋巴集结(Peyer's patch)摄取,这些颗粒随后随着其通过巨噬细胞转运而进入体循环。β-葡聚糖可以以约3.5 mg/100 kcal至约14 mg/100 kcal之间的量包含在小儿营养组合物中;或者,β-葡聚糖可以以约0.010至约0.050 g/100 g组合物之间的水平存在。
在某些实施方案中,本公开内容的营养组合物可包含天然和/或人工调味剂和食用香料,以使所述组合物对于儿童更适口。例如,所述组合物可包含香草、巧克力、蜂蜜和其它所需的调味剂,以增加营养组合物对儿童的适口性。

Claims (15)
1.基于乳的营养组合物,包含:
a) 至多约5.5 g/100 kcal的脂肪或脂质源;
b) 至多约6 g/100 kcal的蛋白质源;和
c) 碳水化合物源,使得所述营养组合物包含至多约18 g/100 kcal的总碳水化合物,其中所述总碳水化合物的至少60%包括果糖、乳糖或其组合。
2.权利要求1的营养组合物,其中所述总碳水化合物包含按重量计至少约10%的果糖。
3.权利要求1的营养组合物,其中所述总碳水化合物包含按重量计至少约60%的乳糖。
4.权利要求1的营养组合物,其中在所述总碳水化合物中存在的果糖和乳糖的组合量为按重量计至少约70%。
5. 权利要求1的营养组合物,其中在所述营养组合物中的总乳糖为约6.0 g/100 kcal至约11.7 g/100 kcal。
6. 权利要求5的营养组合物,其中添加的乳糖为约0.8 g/100 kcal至约5.4 g/100kcal。
7. 权利要求5的营养组合物,其中在所述营养组合物中存在的果糖为至多约3.6 g/100 kcal。
8. 权利要求1的营养组合物,包含约11.5 g/100 kcal至约16 g/100 kcal的总碳水化合物,并且此外其中乳糖以约6.5 g/100 kcal至约11.2 g/100 kcal的水平存在于所述营养组合物中,果糖以约1.4 g/100 kcal至约3.2 g/100 kcal的水平存在于所述营养组合物中。
9.权利要求1的营养组合物,其中所述营养组合物不包含添加的蔗糖。
10.权利要求1的营养组合物,其中所述营养组合物为成长乳。
11.权利要求1的营养组合物,其中所述营养组合物进一步包含长链多不饱和脂肪酸源。
12.权利要求11的营养组合物,其中所述长链多不饱和脂肪酸源包括二十二碳六烯酸。
13. 权利要求1的营养组合物,其进一步包含至少一种约0.1 g/100 kcal至约2 g/100kcal水平的益生元。
14.权利要求13的营养组合物,其中所述益生元包括半乳糖-寡糖、聚葡萄糖、菊粉、果糖-聚糖或其组合。
15.权利要求1的营养组合物,其中所述营养组合物具有小于约60的血糖指数。
