Abstract

taking a saussurea involucrate culture, adding 60-95 wt% of ethanol, wherein the volume ratio of the saussurea involucrate culture to the ethanol is 1 (10-20), carrying out microwave negative pressure extraction at the extraction pressure of 0.15-0.25 MPa and the temperature of 60-80 ℃, extracting for 1-10 min under 100-500W, continuously extracting for 2-3 times, concentrating the obtained liquid under reduced pressure to obtain a concentrate,
the concentrate is dried by two stages of tempering: and in the first section, adopting a mid-infrared-hot air combined drying technology until the moisture content is lower than 30wt%, and after moisture in a transition region diffuses outwards, continuously drying in the second section by adopting a microwave intermittent drying technology until the moisture content is lower than 10wt% to obtain the saussurea involucrata culture extract.
Further, in the preferred embodiment of the present invention, the wavelength range of the mid-infrared-hot air combination drying is 3-40 μm, the temperature is 60-90 ℃, the microwave output power of the microwave intermittent drying is 700-800W, and the intermittent time is 10% -30%.

The saussurea involucrate culture used in the following examples is obtained by large-scale subculture of isolated tissues of saussurea involucrate and purchased from Dali-proekang biotechnology Limited. A tablet containing saussurea involucrate culture extract is prepared from the following components in parts by weight: 10 parts of saussurea involucrate culture extract, 150 parts of cordyceps militaris powder, 150 parts of pseudo-ginseng pollen, 250 parts of sorbitol, 170 parts of microcrystalline cellulose and 10 parts of magnesium stearate.
A preparation method of a tablet containing saussurea involucrate culture extract is characterized by comprising the following steps: the method comprises the following steps:
1) weighing: weighing the raw materials and auxiliary materials according to the weight parts, and carefully rechecking.

Example 4
Preparing a saussurea involucrate culture extract: taking saussurea involucrate culture, adding 70 wt% ethanol, wherein the volume ratio of the saussurea involucrate culture to the ethanol is 1:20, performing microwave negative pressure extraction at the extraction pressure of 0.15MPa and the temperature of 60 ℃, extracting for 10min under 500W, continuously extracting for 2 times, and concentrating the obtained liquid under reduced pressure to obtain a concentrate.

A tablet containing saussurea involucrate culture extract is prepared from the following active components in parts by weight: 100 parts of saussurea involucrate culture extract, 150 parts of cordyceps militaris powder and 50 parts of pseudo-ginseng pollen.
The preparation method comprises the following steps: mixing the extract of herba Saussureae Involueratae culture, Cordyceps militaris powder, and Notoginseng radix pollen, adding medicinal adjuvants, and making into granule.
Example 5
Preparing a saussurea involucrate culture extract: taking saussurea involucrate culture, adding 70 wt% ethanol, wherein the volume ratio of the saussurea involucrate culture to the ethanol is 1:20, performing microwave negative pressure extraction at the extraction pressure of 0.15MPa and the temperature of 80 ℃ for 1min, continuously extracting for 3 times, and concentrating the obtained liquid under reduced pressure to obtain concentrate.

The preparation method comprises the following steps: mixing herba Saussureae Involueratae culture extract, Cordyceps militaris powder, and Notoginseng radix pollen, adding medicinal adjuvants, and making into oral liquid.
Example 6
Saussurea involucrate culture extract was prepared according to the preparation process in example 1.
A tablet containing saussurea involucrate culture extract is prepared from the following active components in parts by weight: 40 parts of saussurea involucrate culture extract, 150 parts of cordyceps militaris powder and 150 parts of pseudo-ginseng pollen.
The preparation method comprises the following steps: mixing herba Saussureae Involueratae culture extract, Cordyceps militaris powder, and Notoginseng radix pollen, adding medicinal adjuvants, and making into capsule.
EXAMPLE 7 clinical trials of the compositions of the invention
1. General data
800 patients with hypertension, age 30-60 years, were randomly and averagely divided into eight groups, including control 1, control 2, control 3, control 4, control 5, control 6, example 1 and example 2, and the gender, age and course of disease were statistically treated, and the differences were not significant (P >0.05) and were comparable.
All the research objects have no other cardiovascular diseases, serious liver and kidney function damage, diabetes, hepatitis, tuberculosis and other infectious diseases. Clinical symptoms: headache, dizziness, inattention, hypomnesis, numbness of limbs, nocturia, palpitation, chest distress, asthenia, emesis, vertigo, etc. Detecting a blood pressure index: the systolic pressure is more than or equal to 140mmHg, and the diastolic pressure is more than or equal to 90 mmHg.

The composition tablets of control 1 differed from the tablets prepared in example 1 only in not containing cordyceps militaris powder and notoginseng pollen; the composition tablets of control 2 group differed from the tablets prepared in example 1 only in not containing saussurea involucrate culture extract and notoginseng pollen; the composition tablets of the control 3 group were different from the tablets prepared in example 1 only in that they did not contain saussurea involucrate culture extract and cordyceps militaris powder; composition of control 4 tablets the tablets prepared in example 1 differ only in the absence of notoginseng pollen; the composition tablets of the control 5 group were different from the tablets prepared in example 1 only in that they did not contain cordyceps militaris powder; the composition tablets of the control 6 group were different from the tablets prepared in example 1 only in not containing the saussurea involucrate culture extract.

3. Diagnostic criteria

As can be seen from the results in table 1 above, compared with the control group, the total effective rate of the extracts obtained in examples 1 and 2 of the present invention for treating hypertension is significantly improved, and the extracts are proved to be capable of effectively treating hypertension with significant effect.
In addition, during the treatment process, no side effect occurs in each treatment group of the traditional Chinese medicine composition.
Claims (8)
1. A composition containing saussurea involucrate culture extract for treating hypertension is characterized by comprising the following components in parts by weight: 10-200 parts of saussurea involucrate culture extract, 10-150 parts of cordyceps militaris powder and 10-150 parts of pseudo-ginseng pollen; the saussurea involucrate culture extract is prepared by the following method:
taking a saussurea involucrate culture, adding 60-95 wt% of ethanol, wherein the volume ratio of the saussurea involucrate culture to the ethanol is 1 (10-20), performing microwave negative pressure extraction at the extraction pressure of 0.15-0.25 MPa and the temperature of 60-80 ℃, extracting for 1-10 min at 100-500W, continuously extracting for 2-3 times, concentrating the obtained liquid under reduced pressure to obtain a concentrate, and drying the concentrate by two stages of tempering: and in the first section, adopting a mid-infrared-hot air combined drying technology until the moisture content is lower than 30wt%, and after moisture in a transition region diffuses outwards, continuously drying in the second section by adopting a microwave intermittent drying technology until the moisture content is lower than 10wt% to obtain the saussurea involucrata culture extract.
2. The composition containing saussurea involucrate culture extract for treating hypertension according to claim 1, which is characterized by comprising the following components in parts by weight: 10-100 parts of saussurea involucrate culture extract, 50-150 parts of cordyceps militaris powder and 50-150 parts of pseudo-ginseng pollen.

The preparation of the composition containing saussurea involucrate culture extract for treating hypertension as claimed in any one of claims 1 to 2, which is prepared from the composition containing saussurea involucrate culture extract as claimed in claim 1 or 2 and pharmaceutically acceptable adjuvants.
4. The formulation of claim 3, wherein the formulation is a tablet, a granule, a capsule, or an oral liquid.

The formulation of claim 4, wherein the tablet is prepared from, by weight: 10-200 parts of saussurea involucrate culture extract, 10-150 parts of cordyceps militaris powder, 10-150 parts of pseudo-ginseng pollen, 100-400 parts of sweeteners, 0-250 parts of microcrystalline cellulose and 1-15 parts of magnesium stearate.
6. The preparation method of the preparation as claimed in claim 5, wherein the preparation method of the tablet comprises weighing the saussurea involucrate culture extract, cordyceps militaris powder, notoginseng pollen, sweeteners, microcrystalline cellulose and magnesium stearate in parts by weight, mixing and tabletting.
7. A method for preparing the formulation of claim 6, comprising the steps of:
(1) weighing: weighing the saussurea involucrate culture extract, cordyceps militaris powder, pseudo-ginseng pollen, sweeteners, microcrystalline cellulose and magnesium stearate in parts by weight;
(2) mixing: mixing the extract of herba Saussureae Involueratae culture with sweetener, mixing with Cordyceps militaris powder, Notoginseng radix pollen, and microcrystalline cellulose, adding magnesium stearate, and mixing to obtain total mixed powder;
(3) tabletting: loading the tablet machine into a die for tabletting to obtain the tablet.
