The Agaricus blazei fine powder is prepared by pulverizing fruiting body of Agaricus blazei, and sieving with 100 mesh sieve; then adding water which is 5 times of the weight of the agaricus blazei powder into the agaricus blazei powder, adding a mixture of cellulase and papain, wherein the mass ratio of the cellulase to the papain in the mixture is 1:1, the mixture is 3% of the weight of the agaricus blazei powder, adjusting the pH value of the solution to 6.0-6.5, and performing enzymolysis for 30 min; then extracting the obtained enzymolysis liquid by ultrasonic wave for 20-30min, and ultrafiltering; concentrating the obtained filtrate to dryness, adding water for dissolving, and evaporating to dryness again; and repeating the steps of dissolving and evaporating for 3-5 times until the dried substance has no acid taste, adding water with the weight of 4-6 times of that of the dried substance into the dried substance for dissolving, adding absolute ethyl alcohol with the water content of 60-70%, uniformly mixing, standing overnight, centrifuging, taking the precipitate, and freeze-drying to obtain the agaricus blazei fine powder.
The obtained health composition has good anti-fatigue effect, and can be used for preparing anti-fatigue health food or medicine.
The invention has the following remarkable advantages:
(1) the invention combines the traditional tonic medicines of the curculigo orchioides and the astragalus and improves the nutritive value of the product and simultaneously improves the efficacies of tonifying the kidney, strengthening the body, tonifying qi and strengthening the body resistance by adding the agaricus blazei murill, the willow tea and the south Africa leaves.
(2) The south Africa leaves contain chemical components of coumarin and flavonoid, and have antioxidant effect, gastric acid secretion promoting, and digestion and absorption promoting effects.
(3) The invention adopts a special method to extract the astragalus, the agaricus blazei and the willow tea, and can obviously improve the content of effective components in the raw materials.
(4) The composition obtained by the invention has good anti-fatigue effect and obvious improvement effect on symptoms such as headache, weakness, inappetence and the like caused by fatigue.
Detailed Description
In order to make the present invention more comprehensible, the technical solutions of the present invention are further described below with reference to specific embodiments, but the present invention is not limited thereto.
Preparing the rhizoma curculiginis powder: pulverizing rhizoma Curculiginis, sieving with 100 mesh sieve, adding 5 times of water, decocting for 1 hr, filtering, and collecting filtrate and residue respectively; concentrating the obtained filtrate to 1/2 of the original volume, adding anhydrous ethanol with the volume of 60% of the concentrated solution, mixing uniformly, standing overnight, centrifuging, taking the precipitate, and freeze-drying to obtain powder A; adding an ethanol solution with the weight 3 times that of the obtained filter residue and the volume of 60% into the obtained filter residue, carrying out ultrasonic extraction for 30min, then filtering, recovering ethanol from the obtained filtrate, and carrying out freeze drying to obtain powder B; mixing the obtained powders A, B.
Preparing the astragalus fine powder: crushing astragalus membranaceus, sieving the crushed astragalus membranaceus by a 100-mesh sieve, adding an ethanol solution with the weight 2 times and 60 vol% into the obtained powder, carrying out ultrasonic treatment for 30min to enable the powder to be fully soaked, quickly adding hot water with the weight 8 times and the temperature of 90-100 ℃ into the obtained feed liquid, carrying out heat preservation extraction for 10min, filtering, concentrating the obtained filtrate under reduced pressure to 1/2 of the original volume, adding anhydrous ethanol with the volume 60% of that of the concentrated solution, uniformly mixing, standing overnight, centrifuging, taking the precipitate, and carrying out freeze drying to obtain the astragalus membranaceus powder. The obtained radix astragali fine powder contains 81% of polysaccharide.
Preparing the agaricus blazei fine powder: pulverizing Agaricus blazei Murill fruiting body, and sieving with 100 mesh sieve; then adding water which is 5 times of the weight of the agaricus blazei powder into the agaricus blazei powder, adding a mixture of cellulase and papain, wherein the mass ratio of the cellulase to the papain in the mixture is 1:1, the mixture is 3% of the weight of the agaricus blazei powder, adjusting the pH value of the solution to 6.0-6.5, and performing enzymolysis for 30 min; then extracting the obtained enzymolysis liquid by ultrasonic wave for 20-30min, and ultrafiltering; concentrating the obtained filtrate to dryness, adding water for dissolving, and evaporating to dryness again; and repeating the steps of dissolving and evaporating for 3-5 times until the dried substance has no acid taste, adding water with the weight of 4-6 times of that of the dried substance into the dried substance for dissolving, adding absolute ethyl alcohol with the water content of 60-70%, uniformly mixing, standing overnight, centrifuging, taking the precipitate, and freeze-drying to obtain the agaricus blazei fine powder. The polysaccharide content of the obtained Agaricus blazei fine powder is 87%.
Preparing the fine willow tea powder: pulverizing the willow tea, sieving with a 100-mesh sieve, adding an ethanol solution with the weight 2 times and 60 vol% into the obtained powder, carrying out ultrasonic treatment for 20min to fully infiltrate the willow tea, quickly adding hot water with the weight 8 times and the temperature of 90-100 ℃ into the obtained feed liquid, carrying out heat preservation extraction for 10min, filtering, concentrating the obtained filtrate under reduced pressure to 1/2 of the original volume, adding anhydrous ethanol with the volume 60% of the concentrated solution, uniformly mixing, standing overnight, centrifuging, taking the precipitate, and freeze-drying to obtain the fine willow tea powder. The content of polysaccharide in the obtained fine powder of folium Salicis Babylonicae is 78%.
Preparing south Africa leaf powder: pulverizing south Africa leaves, sieving with 100 mesh sieve, adding 6 times of water, decocting for 1 hr, filtering, and collecting filtrate and residue; concentrating the filtrate to 1/2 of the original volume, adding 70% anhydrous ethanol of the volume of the concentrated solution, mixing well, standing overnight, centrifuging, taking the precipitate, and freeze-drying to obtain powder A; adding an ethanol solution with the weight 5 times that of the obtained filter residue and the volume percent of 60% into the obtained filter residue, carrying out ultrasonic extraction for 40min, then filtering, recovering ethanol from the obtained filtrate, and carrying out freeze drying to obtain powder B; mixing the obtained powders A, B.
Example 1
Mixing rhizoma Curculiginis powder 120g, radix astragali fine powder 100g, Agaricus Blazei Murr fine powder 100g, folium Salicis Babylonicae fine powder 80g, and folium Cucurbitae powder 30g to obtain health composition.
Example 2
Mixing rhizoma Curculiginis powder 100g, radix astragali fine powder 120g, Agaricus Blazei Murr fine powder 90g, folium Salicis Babylonicae fine powder 70g, and folium Cucurbitae powder 50g to obtain health composition.
Example 3
Mixing rhizoma Curculiginis powder 150g, radix astragali fine powder 80g, Agaricus Blazei Murr fine powder 80g, folium Salicis Babylonicae fine powder 60g, and folium Nanfanae powder 60g to obtain health composition.
Efficacy test
After 70 adult male rats are subjected to adaptive culture for 5 days, the adult male rats are randomly divided into a test group, a curculigo orchioides control group, an astragalus membranaceus control group, an agaricus blazei murill control group, an willow tea control group, a south African leaf control group and a blank control group according to weight, 10 adult male rats are respectively subjected to intragastric administration of 2mL of the composition aqueous solution (0.5 g/mL), a curculigo orchioides powder aqueous solution (0.14 g/mL), an astragalus membranaceus fine powder aqueous solution (0.14 g/mL), an agaricus blazei murill fine powder aqueous solution (0.12 g/mL), a willow tea fine powder aqueous solution (0.1 g/mL) and a south African leaf powder aqueous solution (0.05 g/mL) of the composition of example 1, the blank control group is subjected to intragastric administration of equal amount of clear water, and the adult male rats are placed into water for weight-bearing (.
As can be seen from Table 1, the swimming time of the mice in the test group is significantly prolonged compared to the control group, and it is proved that the anti-fatigue effect of the mice in the test group can be improved by using the combination of Curculigo orchioides, Astragalus membranaceus, Agaricus blazei, willow tea and leaves of south Africa.

2) concentrating the filtrate to 1/2, adding 60-70% anhydrous ethanol, mixing, standing overnight, centrifuging, collecting precipitate, and freeze drying to obtain powder A;
3) adding 3-5 times of 60 vol% ethanol solution into the obtained residue, ultrasonic extracting for 30-50min, filtering, recovering ethanol from the filtrate, and freeze drying to obtain powder B;
4) mixing the obtained powders A, B to obtain rhizoma Curculiginis powder or south Africa leaf powder;
the preparation method of the astragalus fine powder and the willow tea fine powder comprises the steps of crushing astragalus or willow tea, sieving the crushed astragalus or willow tea by a 100-mesh sieve, adding an ethanol solution with the weight being 2 times and 60 vol% of the weight of the powder into the obtained powder, carrying out ultrasonic treatment for 20-30min to enable the powder to be fully soaked, then quickly adding hot water with the weight being 8 times and the temperature being 90-100 ℃ into the obtained feed liquid, carrying out heat preservation extraction for 10min, filtering, carrying out reduced pressure concentration on the obtained filtrate to 1/2 of the original volume, adding absolute ethanol with the volume being 60-70% of the volume of the concentrated solution, uniformly mixing, standing overnight, centrifuging, taking a precipitate, and freeze-drying to obtain;
the Agaricus blazei fine powder is prepared by pulverizing fruiting body of Agaricus blazei, and sieving with 100 mesh sieve; then adding water which is 5 times of the weight of the agaricus blazei powder into the agaricus blazei powder, adding a mixture of cellulase and papain which is 3% of the agaricus blazei powder by weight, adjusting the pH value of the solution to 6.0-6.5, and carrying out enzymolysis treatment for 30 min; then extracting the obtained enzymolysis liquid by ultrasonic wave for 20-30min, and ultrafiltering; concentrating the obtained filtrate to dryness, adding water for dissolving, and evaporating to dryness again; dissolving repeatedly, evaporating for 3-5 times until the dried product has no acid smell, adding water 4-6 times the weight of the dried product to dissolve, adding 60-70% anhydrous alcohol, mixing, standing overnight, centrifuging, collecting precipitate, and freeze drying to obtain Agaricus blazei Murill refined powder;
the mass ratio of the cellulase to the papain in the mixture is 1: 1;
the health-care composition is applied to preparing anti-fatigue health-care food or medicine.
