Abstract
本发明公开了一种稳定的辣根过氧化物酶底物的配方,包括底物A液和底物B液。底物A液的配方,由以下按体积百分比数计算的组分组成:0.1%‑1%硼酸,0.05%‑0.5%鲁米诺钠盐,0.01%‑0.1%吩噻嗪,0.005%‑0.05%对碘苯酚,0.05%‑0.5%吐温20,其余组分为有机溶剂和纯化水。底物B液的配方,由以下按体积百分比数计算的组分组成:0.05%‑0.5%柠檬酸三钠,0.01%‑0.1%柠檬酸,0.01%‑0.1%氯化钠,0.005%‑0.05%过氧化脲,其余组分为纯化水。本发明底物稳定性更好,且性能更优。
Description
技术领域:本发明涉及免疫检测领域,具体地讲是一种稳定的辣根过氧化物酶底物的配方。
背景技术:辣根过氧化物酶是免疫检测领域中的一种工具酶,器来源方便,价格较低,并且检测体系领命被广泛的应用于免疫检测中,但其底物种类多,在配制时需要进行准确的称量,但称量过程比较繁琐,且易出现称量的偏差,从而影响到检测的准确性,而这种底物必须现用现配,且对光敏感易变质,且底物配制时易出现不易溶解的情况,该底物稳定性较差,易出现配制失败的情况,从而严重的影响到检测的结果,满足不了人们免疫检测中的需求。
发明内容:
本发明的目的是提供一种稳定的辣根过氧化物酶底物的配方,以解决现有设备的不足。
本发明的一种稳定的辣根过氧化物酶底物的配方,包括底物A液和底物B液。
上述1的底物A液的配方,由以下按体积百分比数计算的组分组成:
0.1%-1%硼酸,0.05%-0.5%鲁米诺钠盐,0.01%-0.1%吩噻嗪,0.005%-0.05%对碘苯酚,0.05%-0.5%吐温20,其余组分为纯化水及有机溶剂。
最优选地,所述硼酸的质量分数最优选为0.62%,鲁米诺钠盐的质量分数最优选为0.1%,吩噻嗪为3-(-10-吩噻嗪基)丙烷-1-磺酸钠,且质量分数最优选为0.02%,对碘苯酚质量分数最优选为0.016%,吐温20质量分数最优选为0.2%。
最优选地,所述底物A液配置好后应在37度环境放置一周后使用。
最优选地,所述吩噻嗪和对碘苯酚需要使用有机溶剂溶解,吩噻嗪用DMF溶解为20mm/L,再按1mm/L量加入完成配制,对碘苯酚用DMF溶解为20mm/L,再按1mm/L量加入完成配制,且PH值调节到9.2-9.3为止。
上述1的底物B液的配方,由以下按体积百分比数计算的组分组成:
0.05%-0.5%柠檬酸三钠,0.01%-0.1%柠檬酸,0.01%-0.1%氯化钠,0.005%-0.05%过氧化脲,其余组分为纯化水。
最优选地,所述柠檬酸三钠的质量分数最优选为0.2%,柠檬酸的质量分数最优选为0.08%,氯化钠的质量分数最优选为0.06%,过氧化脲的质量分数最优选为0.015%。
最优选地,所述底物B液配置好后应在37度环境放置一周后使用。
最优选地,所述B液配制后后PH值为5.5。
本发明的有益效果是:本发明底物稳定性更好,且性能更优。

用于促甲状腺激素的检测:
本底物非配方用于促甲状腺素的检测值:
对比底物用于促甲状腺激素的检测值:
由表可以看出,20孔S0是做的最低检出限,显然底物的灵敏度相差较大,本底物配方S5点在900多万,另一底物信号S5点信号低一半左右,信号差别较大。
底物的稳定性,底物A/B液使用前在37度放置一周已经相对稳定性,之后2-8℃可放置1年没有什么跌幅。

由表可以看出,20孔S0是做的最低检出限,显然底物的灵敏度相差较大,本底物配方S5点在800多万,另一底物信号S5点信号低一半左右,信号差别较大。
底物的稳定性,底物A/B液使用前在37度放置一周已经相对稳定性,之后2-8℃可放置1年没有什么跌幅。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明技术原理的前提下,实现辣根过氧化物酶的稳定,还可以做出若干改进和替换,这些改进和替换也应视为本发明的保护范围。
Claims (9)
1.一种稳定的辣根过氧化物酶底物的配方,其特征在于:包括底物A液和底物B液。
2.根据权利要求1所述底物A液的配方,其特征在于:由以下按体积百分比数计算的组分组成:
0.1%-1%硼酸,0.05%-0.5%鲁米诺钠盐,0.01%-0.1%吩噻嗪,0.005%-0.05%对碘苯酚,0.05%-0.5%吐温20,其余组分为纯化水及有机溶剂。
3.根据权利要求2所述底物A液的的配方,其中所述硼酸的质量分数最优选为0.61843%,鲁米诺钠盐的质量分数最优选为0.08%,吩噻嗪为3-(-10-吩噻嗪基)丙烷-1-磺酸钠,且质量分数最优选为0.02%,对碘苯酚质量分数最优选为0.016%,吐温20质量分数最优选为0.2%。
4.根据权利要求2所述底物A液的配方,其中所述底物A液配置好后应在37度环境放置一周后使用。
5.根据权利要求2所述底物A液的配方,其中所述吩噻嗪和对碘苯酚需要使用有机溶剂溶解,吩噻嗪用DMF溶解为20mm/L,再按1.5mm/L量加入完成配制,对碘苯酚用DMF溶解为20mm/L,再按0.2mm/L量加入完成配制,且PH值调节到9.2为止。
6.根据权利要求1所述底物B液的配方,其特征在于:由以下按体积百分比数计算的组分组成:
0.05%-0.5%柠檬酸三钠,0.01%-0.1%柠檬酸,0.01%-0.1%氯化钠,0.005%-0.05%过氧化脲,其余组分为纯化水。
7.根据权利要求6所述底物B液的配方,其中所述柠檬酸三钠的质量分数最优选为0.2%,柠檬酸的质量分数最优选为0.08%,氯化钠的质量分数最优选为0.06%,过氧化脲的质量分数最优选为0.015%。
8.根据权利要求6所述底物B液的配方,其中所述底物B液配置好后应在37度环境放置一周后使用。
9.根据权利要求6所述底物B液的配方,其中所述B液配制后后PH值为5.5。
