Abstract

本发明属中药制药领域,涉及中药降香有效活性部位及其制备方法,本发明采用醇提取,浓缩,反向层析柱,分离方法,从传统中药降香中提取有效的活性部位DE。本发明的提取方法中,在提高提取物中DE纯度的同时,兼顾提取物的转移率和得率,制得转移率和得率稳定,且纯度较高的降香有效活性成分的提取物。进一步,可制备以降香提取物为活性成分的药物组合制剂。

Description

中药降香提取物的有效活性部位及其制备方法
技术领域
本发明属中药制药领域,涉及中药降香提取物及其制备方法,具体涉及中药降香提取物的有效活性部位—DE及其制备方法。
背景技术
现有技术公开了中药降香是传统名贵中药豆科植物降香檀Dalbergia odorifera T.Chen树干和根的干燥心材。据资料记载,降香旧时常用于治疗外伤疾病,并与其他中药配伍治疗心血管方面的疾病。《本草经疏》记载“降真香,香中之清烈者也,故能辟一切恶气。……瘀血停积胸膈骨,按之痛或并胁肋痛”。
现代中药研究表明,降香中的主要成分为挥发油以及黄酮类化合物;现代药理学表明黄酮类化合物具有很强的药理活性,例如:抗氧化作用,抗炎、镇痛作用,舒张血管,调节血脂,抑制血栓形成,促血管新生,改善心肌梗死,以及预防基因突变等作用。
目前中药降香仅作为民间应用,由于传统中药其成分复杂,难以明了其所含成分,使其制剂存在一定的可控性、稳定性的困扰,这是中医药国际化的难题之一,迄今为止,尚未见有关现代提取分离其中有效活性部位的报道,此也正是现代医学目前仍缺乏理想的含中药降香有效活性成分的治疗药物的原因之一。
基于现有技术的现状,本申请的发明人拟提供中药降香提取物及其制备方法,尤其涉及中药降香提取物的有效活性部位—DE及其制备方法。
发明内容
本发明的目的是克服现有技术的缺陷,提供一种质量可靠、稳定性良好的中药降香提取物及其制备方法,尤其涉及中药降香提取物的有效活性部位—DE及其制备方法。
本发明的另一目的是提供所述的中药降香提取物的制备方法,本发明中采用现代中药提取分离纯化和富集的方法制得中药降香提取物的有效活性部位—DE。
具体的,本发明提供了中药降香提取物的有效活性部位,其通过下述方法制备:
采用醇提取,浓缩,反向层析柱,分离的方法,按下述步骤制得所述降香有效活性部位,
降香饮片粉碎,用10倍75%的乙醇,在索氏提取器中提取回流1h,过滤,取其药渣;
‚ 将药渣加水,煮沸1h,使其浓缩至100ml,加入95%乙醇900ml,取其上清液部分;
ƒ取上述上清液经C18反向层析柱30cm×4.5cm,20-45μm,Fuji Chromatorex,Japan分离,流动相为甲醇-水;
④甲醇浓度在80-95%时分离,得到降香有效活性部位DE;
上述活性部位用HPLC测定,使用C18反向层析柱,流动相乙腈(A)-0.3%磷酸水(B),流速:1.0ml/min;柱压:10.0MPa;DAD检测器,波长为254-365nm进行分离和检测。
检测结果表明,本发明制得一种转移率和得率稳定,且纯度较高的降香有效活性成分的提取物。
本发明的优点还在于,所述的制备方法中,在提高提取物中DE纯度的同时,兼顾提取物的转移率和得率。
本发明还提供了一种药物组合,由治疗有效量的制得的降香提取物为活性成分,与药学上可接受的辅料或辅助性成分制成药物组合制剂;所述的药物组合制剂包括丸剂、散剂、胶囊、片剂、颗粒剂,注射剂以及复方制剂。
根据本发明的上述内容,按照本领域的普通技术知识和惯用手段,在不脱离本发明精神的限定内可适当变更。
具体实施方式
以下列举实施例对本发明的上述内容做更具体地和详细的说明。但不应将此理解为本发明上述主题的范围仅限于以下实例。
实施例1 制备中药降香提取物的有效活性部位—DE
取降香饮片50g(产地:中国四川,购自上海信德中药公司饮片厂),粉碎后加500ml 75%酒精,在索氏提取器中提取回流1h后,取其药渣沸水煎煮1h并浓缩至100ml,900ml 95%乙醇加入后,上清液经C18反向层析柱(30cm×4.5cm,20-45μm,Fuji Chromatorex,Japan)分离,流动相为甲醇-水;经成分测定,甲醇浓度在80-95%时分离得到中药降香提取物的有效活性部位DE。
实施例2
取实施例1制得的治疗有效量的中药降香提取物的有效活性部位DE,添加适量微晶纤维素、淀粉或药学上可接受的其他赋形剂,按常规方法分别制备胶囊剂、片剂、散剂、颗粒剂或丸剂等内服药物剂型。

Claims (7)

1.一种中药降香提取物的有效活性部位,其特征在于,所述的有效活性部位为降香有效活性部位DE,其通过醇提取,浓缩,反向层析柱,分离的方法制得。
2.权利要求1所述的中药降香提取物的有效活性部位的制备方法,其特征在于,其包括步骤:
①降香饮片粉碎,用75%的乙醇,在索氏提取器中提取回流,过滤,取其药渣;
②将药渣加水,煮沸,使其浓缩,加入95%乙醇,取其上清液部分;
③取上述上清液经C18反向层析柱30cm×4.5cm,20-45μm,Fuji Chromatorex,Japan分离,流动相为甲醇-水;
④甲醇浓度在80-95%时分离,得到降香有效活性部位DE。
3.权利要求2所述的中药降香提取物的有效活性部位的制备方法,其特征在于,所述步骤中:用10倍75%的乙醇,在索氏提取器中提取回流1h;所述步骤‚中,煮沸1h,使其浓缩至100ml,加入95%乙醇900ml。
4.权利要求2所述的中药降香提取物的有效活性部位的制备方法,其特征在于,所述步骤④中制得的降香有效活性部位DE,用HPLC测定,使用C18反向层析柱,流动相乙腈(A)-0.3%磷酸水(B),流速:1.0ml/min;柱压:10.0MPa;DAD检测器,波长为254-365nm进行分离和检测。
5.权利要求2所述的中药降香提取物的有效活性部位的制备方法,其特征在于,所述制得的降香有效活性部位DE成分转移率和得率稳定,纯度较高。
6.一种药物组合,其特征在于,由治疗有效量的所述的降香提取物为活性成分,与药学上可接受的辅料或辅助性成分制成药物组合制剂。
7.按权利要求6所述的药物组合,其特征在于,所述的药物组合制剂制成丸剂、散剂、胶囊、片剂、颗粒剂,注射剂以及复方制剂。
