Abstract
本发明公开一种含美洲大蠊提取物的胃溃疡组合物及其制备方法,包括下列质量份的原料备料:美洲大蠊水溶浸膏0.1~20份、木蝴蝶3~15份、紫苏梗10~50份、海螵蛸10~30份、白芍10~60份、延胡索10~60份、陈皮10~30份、茯苓10~50份、葛根10~50份、三七5~25份、甘草3~15份、硬脂酸镁0.15份~1份、淀粉20~50份、糊精30份~40份。本发明制备简单,对急慢性体表溃疡(糖尿病溃疡)、褥疮、瘘窦、快速修复因手术后插入导管对消化道黏膜的损伤,食管溃疡、放射性食管炎、溃疡性结肠炎、放射性直肠炎、伴黏膜损害的腹泻、便血等疾病的有效治愈率效果显著。

技术领域
本发明涉及含美洲大蠊提取物的胃溃疡的组合物及其制备方法,特别涉及一种含美洲大蠊提取物的胃溃疡组合物及其制备方法。
背景技术
胃溃疡是一种多发病、常见病,具有病程长、病情复杂、极易复发的特点;主要是胃粘膜被胃酸消化造成超过黏膜肌层组织的损伤。主要症状表现为上腹部疼痛,常见隐痛、钝痛、胀痛、烧灼样痛,餐后发作性腹痛、烧灼性疼痛、发恶心、胃镜下表现出广泛黏膜红斑、口臭、胃痉挛、嗳气、腹部隐痛、阴虚胃痛等症状。并发症:上消化道伴有出血、溃疡穿孔、幽门梗阻、癌变等。本病活动时会出现上腹部压痛,缓解无明显体征。
胃溃疡发病与中医学的脾胃有关,脾胃升降失调则会引起胃溃疡、胃炎等胃病的发生。现代医学认为:幽门螺杆菌感染是胃溃疡的最主要原因,其次跟环境因素、自身的生活饮食习惯、长期使用的药物、遗传因素密切相关。胃酸分泌过多、幽门螺旋杆菌感染和胃黏膜保护作用降低等因素是引起胃溃疡的主要因素。
现在大多采取西药治疗,但是西药治疗副作用较大,而中药效果又不明显。
发明内容
为提供对胃溃疡有效的中药组合物,本发明提供一种含美洲大蠊提取物的胃溃疡组合物及其制备方法,可有效治疗胃溃疡,并减少西药带来的副作用及耐药性。
本发明通过下列技术方案实现:一种含美洲大蠊提取物的胃溃疡组合物,包含下列质量份的原料:
美洲大蠊水溶浸膏0.1~20份、木蝴蝶3~15份、紫苏梗10~50份、海螵蛸10~30份、白芍10~60份、延胡索10~60份、陈皮10~30份、茯苓10~50份、葛根10~50份、三七5~25份、甘草3~15份、硬脂酸镁0.15份~1份、淀粉20~50份、糊精30份~40份。
本发明的另一目的还在于提供一种含美洲大蠊提取物的胃溃疡组合物的制备方法,经过下列各步骤:
(1)按下列质量份的原料备料:
美洲大蠊水溶浸膏0.1~20份、木蝴蝶3~15份、紫苏梗10~50份、海螵蛸10~30份、白芍10~60份、延胡索10~60份、陈皮10~30份、茯苓10~50份、葛根10~50份、三七5~25份、甘草3~15份、硬脂酸镁0.15份~1份、淀粉20~50份、糊精30份~40份;
(2)将紫苏梗、海螵蛸和三七粉碎成混合细粉,过120目筛;
(3)将木蝴蝶、陈皮、甘草、白芍、葛根、茯苓和延胡索粉碎成混合粗粉,过10目筛,加入4~10体积倍的蒸馏水煎煮提取3次,第一二次各3小时,第三次2小时,合并提取液,过滤,滤液静置12~24小时,取上清液,经减压浓缩成相对密度为1.34~1.39(40~65℃)的浸膏;
(4)将美洲大蠊水溶浸膏、步骤(2)的混合细粉、步骤(3)的浸膏、硬脂酸镁、淀粉和糊精混匀,制成颗粒,整粒,再进行干燥,即得含美洲大蠊提取物的胃溃疡组合物。
所得含美洲大蠊提取物的胃溃疡组合物按常规制成医药上可接受的片剂、颗粒 剂、胶囊剂。
所述美洲大蠊水溶浸膏通过下列步骤制得:
A、浸泡:将美洲大蠊干燥的成虫,加入4~10体积倍的浓度为45~95v/v%的乙醇浸泡10~720天;
B、粉碎:将浸泡的美洲大蠊连同溶剂倒入高速破壁机内粉碎;
C、过滤:将粉碎后的美洲大蠊连同溶剂进行过滤得到浸出液;
D、减压浓缩:将浸出液在真空度﹣0.07~﹣0.08MPa、温度40~45℃的条件下减压浓缩至比重为1 .05~1 .15的乙醇浓缩稠浸膏;
E、纯化分离:将乙醇浓缩稠浸膏的上层物、下层物分开收集,将上层物进行3次离心分离,合并上层,得到总上层物,再加入石油醚静置分层,分层后的上层物经浓缩成浸膏,即得到美洲大蠊水溶浸膏。该浸膏呈深红棕色,味苦、微腥,易溶于水。
美洲大蠊是属于昆虫纲蜚蠊目蜚蠊科,在地球上有三亿多年的生存历史,有“活化石”之称,是至今繁衍最成功的昆虫类群之一。美洲大蠊作为药用最开始见于《神农本草经》,被列为中品,谓“味咸、平”。用于治血痰症、寒热、破积聚、喉咽痹、内寒无子;具有活血化瘀、解毒消疳、利水消肿等功效,含有蛋白质、氨基酸、肽类、糖类、多元醇等多种有效成分,现代研究表明,美洲大蠊提取物在抗肿瘤、提高免疫力、治疗心血管疾病、组织修复、抗菌、消炎等方面具有显著的作用,减少大鼠胃粘膜损伤,修复损伤的胃粘膜,美洲大蠊提取物还富集纯化其多肽类抗肿瘤活性成分。
美洲大蠊:咸、平。入心、肝、脾、肾经。功能主治:外用消肿生肌;用于水火烫伤。内服健脾消疳、活血通脉、利水消肿;用于肋痛、症瘕、疳疾、心悸、气喘、水肿等。
木蝴蝶:为紫葳科植物木蝴蝶的干燥成熟种子。性味归经:苦、甘,凉。归肺、肝、胃经。功能与主治:清肺利咽、疏肝和胃。用于肺热咳嗽,喉痹,音哑,肝胃气痛。
紫苏梗:为唇形科植物紫苏的干燥茎。性味归经:辛、温。归肺、脾经。功能与主治:理气宽中,止痛,安胎。用于胸膈痞闷,胃脘疼痛,嗳气呕吐,胎动不安。
白芍:为毛茛科植物芍药的干燥根。性味归经:苦、酸,微寒。归肝、脾经。功能与主治:养血调经,敛阴止汗,柔肝止痛,平抑肝阳。用于血虚萎黄,月经不调,自汗,盗汗,肋痛,腹痛,四肢挛痛,头痛眩晕。
延胡索:为罂粟科植物延胡索的干燥块茎。性味归经:辛、苦,温。归肝、脾经。功能与主治:活血,行气,止痛。用于胸肋、脘腹疼痛,胸痹心痛,经闭痛经,产后淤阻,跌扑肿痛。
茯苓:为多孔菌科真菌茯苓的干燥菌核。性味归经:甘、淡,平。归心、肺、脾、肾经。功能与主治:利水渗湿,健脾,宁心。用于水肿尿少,痰饮眩悸,脾虚食少,便溏泄泻,心神不宁,惊悸失眠。
葛根:为豆科植物野葛的干燥根。 性味归经:甘、辛,凉。归肺、脾、胃经。功能与主治:解肌退热,生津止渴,透疹,升阳止泻,通经活络,解酒毒。用于外感发热头痛,项背强痛,口渴,消渴,麻疹不透,热痢,泄泻,眩晕头痛,中风偏瘫,胸痹心痛,酒毒伤中。
陈皮:为芸香科植物橘及其栽培的变种的干燥成熟果实。性味归经:苦、辛、温。归肺、脾经。功能与主治:理气健脾,燥湿化痰。用于腕腹胀满,食少吐泻,咳嗽痰多。
海螵蛸:为乌贼科动物无针乌贼或金乌贼的干燥内壳。性味归经:咸、涩,温。归脾、肾经。功能与主治:收敛止血,涩精止带,制酸止痛,收湿敛疮。用于吐血衄血,崩漏便血,遗精滑精,赤白带下,胃痛吞酸;外治损伤出血,湿疹湿疮,溃疡不敛。
三七:为五加科植物三七的干燥根和根茎。 性味归经:甘、淡,平。归肝、胃经。功能与主治:散瘀止血,消肿定痛。用于咯血,吐血,衄血,便血,崩漏,外伤出血,胸腹刺痛,跌扑肿痛。
甘草:为豆科植物甘草、胀果甘草或光果甘草的干燥根和根茎。性味归经:甘,平。归心、肺、脾、胃经。功能与主治:补脾益气,清热解毒,祛痰止咳,缓急止痛,调和诸药。用于脾胃虚弱,倦怠乏力,心悸气短,咳嗽痰多,脘腹、四肢挛急疼痛,痈肿疮毒,缓解药物毒性,烈性。
本发明具备的优点及效果:目前临床上根据发病机制及药物作用特点分成:抗酸制剂、受体阻断剂、黏膜保护剂、抗幽门螺杆菌类药等;为了减少西药带来的副作用及耐药性,容易复发,治疗周期长等缺点。依据中医中药的传统理论,选出了十一种中药,本配方能够标本兼治,治疗各种胃溃疡临床症状作用迅速、效果明显,没有毒副作用,方便服用,服用后无不适反应。本发明所得含美洲大蠊提取物的胃溃疡组合物呈浅黄色、无腥臭味、易溶。通过本发明方法制备的组合物中游离氨基酸、低聚糖、维生素等活性成分含量较多,制备方法简单。本发明的组合物用于治疗急慢性胃溃疡(包括浅表性胃炎、出血性胃炎、胃烧痛),对急慢性体表溃疡(糖尿病溃疡)、褥疮、瘘窦、快速修复因手术后插入导管对消化道黏膜的损伤,食管溃疡、放射性食管炎、溃疡性结肠炎、放射性直肠炎、伴黏膜损害的腹泻、便血等疾病的有效治愈率效果显著。
具体实施方式
下面结合实施例对本发明做进一步说明。
实施例1
(1)按下列质量份的原料备料:
美洲大蠊水溶浸膏15份、木蝴蝶5份、紫苏梗30份、海螵蛸30份、白芍60份、延胡索30份、陈皮15份、茯苓30份、葛根25份、三七15份、甘草12份、硬脂酸镁0.8份、淀粉30份、糊精35份;
所述美洲大蠊水溶浸膏通过下列步骤制得:
A、浸泡:将美洲大蠊干燥的成虫,加入8体积倍的浓度为75v/v%的乙醇浸泡360天;
B、粉碎:将浸泡的美洲大蠊连同溶剂倒入高速破壁机内粉碎;
C、过滤:将粉碎后的美洲大蠊连同溶剂进行过滤得到浸出液;
D、减压浓缩:将浸出液在真空度﹣0.07~﹣0.08MPa、温度40~45℃的条件下减压浓缩至比重为1.05~1.15的乙醇浓缩稠浸膏;
E、纯化分离:将乙醇浓缩稠浸膏的上层物、下层物分开收集,将上层物进行3次离心分离,合并上层,得到总上层物,再加入石油醚静置分层,分层后的上层物经浓缩成浸膏,即得到美洲大蠊水溶浸膏。该浸膏呈深红棕色,味苦、微腥,易溶于水。
(2)将紫苏梗、海螵蛸和三七粉碎成混合细粉,过120目筛;
(3)将木蝴蝶、陈皮、甘草、白芍、葛根、茯苓和延胡索粉碎成混合粗粉,过10目筛,加入8体积倍的蒸馏水煎煮提取3次,第一二次各3小时,第三次2小时,合并提取液,过滤,滤液静置20小时,取上清液,经减压浓缩成相对密度为1.34~1.39(40~65℃)的浸膏;
(4)将美洲大蠊水溶浸膏、步骤(2)的混合细粉、步骤(3)的浸膏、硬脂酸镁、淀粉和糊精混匀,制成颗粒,整粒,再进行干燥,即得含美洲大蠊提取物的胃溃疡组合物。
实施例2
(1)按下列质量份的原料备料:
美洲大蠊水溶浸膏0.1份、木蝴蝶3份、紫苏梗10份、海螵蛸10份、白芍10份、延胡索10份、陈皮10份、茯苓10份、葛根10份、三七5份、甘草3份、硬脂酸镁0.15份、淀粉20份、糊精30份;
所述美洲大蠊水溶浸膏通过下列步骤制得:
A、浸泡:将美洲大蠊干燥的成虫,加入4体积倍的浓度为45v/v%的乙醇浸泡10天;
B、粉碎:将浸泡的美洲大蠊连同溶剂倒入高速破壁机内粉碎;
C、过滤:将粉碎后的美洲大蠊连同溶剂进行过滤得到浸出液;
D、减压浓缩:将浸出液在真空度﹣0.07~﹣0.08MPa、温度40℃的条件下减压浓缩至比重为1 .05~1 .15的乙醇浓缩稠浸膏;

(3)将木蝴蝶、陈皮、甘草、白芍、葛根、茯苓和延胡索粉碎成混合粗粉,过10目筛,加入4体积倍的蒸馏水煎煮提取3次,第一二次各3小时,第三次2小时,合并提取液,过滤,滤液静置12小时,取上清液,经减压浓缩成相对密度为1.34~1.39(40~65℃)的浸膏;
(4)将美洲大蠊水溶浸膏、步骤(2)的混合细粉、步骤(3)的浸膏、硬脂酸镁、淀粉和糊精混匀,制成颗粒,整粒,再进行干燥,即得含美洲大蠊提取物的胃溃疡组合物。
实施例3
(1)按下列质量份的原料备料:
美洲大蠊水溶浸膏20份、木蝴蝶15份、紫苏梗50份、海螵蛸20份、白芍50份、延胡索60份、陈皮30份、茯苓50份、葛根50份、三七25份、甘草15份、硬脂酸镁1份、淀粉50份、糊精40份;
所述美洲大蠊水溶浸膏通过下列步骤制得:
A、浸泡:将美洲大蠊干燥的成虫,加入10体积倍的浓度为95v/v%的乙醇浸泡720天;
B、粉碎:将浸泡的美洲大蠊连同溶剂倒入高速破壁机内粉碎;
C、过滤:将粉碎后的美洲大蠊连同溶剂进行过滤得到浸出液;
D、减压浓缩:将浸出液在真空度﹣0.07~﹣0.08MPa、温度45℃的条件下减压浓缩至比重为1 .05~1 .15的乙醇浓缩稠浸膏;
E、纯化分离:将乙醇浓缩稠浸膏的上层物、下层物分开收集,将上层物进行3次离心分离,合并上层,得到总上层物,再加入石油醚静置分层,分层后的上层物经浓缩成浸膏,即得到美洲大蠊水溶浸膏;该浸膏呈深红棕色,味苦、微腥,易溶于水;
(2)将紫苏梗、海螵蛸和三七粉碎成混合细粉,过120目筛;
(3)将木蝴蝶、陈皮、甘草、白芍、葛根、茯苓和延胡索粉碎成混合粗粉,过10目筛,加入10体积倍的蒸馏水煎煮提取3次,第一二次各3小时,第三次2小时,合并提取液,过滤,滤液静置24小时,取上清液,经减压浓缩成相对密度为1.34~1.39(40~65℃)的浸膏;
(4)将美洲大蠊水溶浸膏、步骤(2)的混合细粉、步骤(3)的浸膏、硬脂酸镁、淀粉和糊精混匀,制成颗粒,整粒,再进行干燥,即得含美洲大蠊提取物的胃溃疡组合物。
毒理学试验:
1.1急性毒性试验
1.1.1取昆明小鼠16只,17~22g/只,雌雄各半,随机分为4组,每组4只,4组按照组距灌胃实施例1制备的片剂、按组距1.5倍设置剂量组:15000mg/kg 、7500mg/kg、3750mg/kg、1875mg/kg。
配制浓度:75mg/ml、37.5mg/ml、18.75mg/ml、9.375mg/ml
每只小鼠灌胃体积是:0.2ml/10g。
注:M—小鼠体重。
1.1.2配制药物:
称量药物组合物4.5g,溶于6ml蒸馏水中,得到浓度为750mg/ml的混溶液:
3ml(750mg/ml)3ml溶剂:375mg/ml
3ml(750mg/ml)+3ml溶剂:187.5mg/ml
3ml(187.5mg/ml)+3ml溶剂:93.75mg/ml
1.1.3结果:
活动较活跃、精神状态好、食欲正常、大小便的颜色正常、呼吸正常,眼部、鼻、口腔、生殖器无异常分泌物,动物未出现不良反应及死亡现象,解剖各脏器没有异常。
1.2长期毒性试验:
试验中应用昆明小鼠16只,17~22g/只,雌雄各半,将实验例1制备的片剂在灭菌的氯化钠注射液中溶解,配制成溶液灌胃,根据设置的剂量组连续灌胃3个月,观察本发明的组合物对小鼠的各项指标影响。
试验结果表明:连续灌胃3个月,观察的外观体征、活动、精神状态、食欲、大小便的颜色、背部毛、肤色、呼吸,等均无明显影响。还有观察眼部、鼻、口腔、生殖器无异常分泌物。
根据本发明实施例所得含美洲大蠊提取物的胃溃疡组合物治疗各种胃溃疡的临床资料统计:
一、治疗对象
胃溃疡患者180例;其中女患者120例,男患者60例,年龄15岁-81岁。随机分为治疗组和对照组各90例。
二、服用方法
治疗组分为三个小组,分别服用实施例1-3所得组合物,每日3次,每次3-5g,连服20日。
对照组按常规胃溃疡治疗,连服奥美拉唑胶囊20日。
三、结果
本发明提供的组合物治疗效果:20-30天就有明显效果,对胃溃疡患者的有效率达到98%,而对照组对胃溃疡患者的有效率只达68%。
四、结论
本发明提供的组合物为各组分之间相互协同增效,使所得组合物能够标本兼治,治疗各种胃溃疡临床症状作用迅速、效果明显,没有毒副作用,方便服用,服用后无不适反应。本发明的组合物用于治疗急慢性胃溃疡(包括浅表性胃炎、出血性胃炎、胃烧痛),对急慢性体表溃疡(糖尿病溃疡)、褥疮、瘘窦、快速修复因手术后插入导管对消化道黏膜的损伤,食管溃疡、放射性食管炎、溃疡性结肠炎、放射性直肠炎、伴黏膜损害的腹泻、便血等疾病的有效治愈率效果显著。其作用优于对照组。
案例1
廖某,女,21岁,云南人,半年前胃镜检查为原发消化性溃疡,临床症状主要表现为胃疼,近几日疼痛加剧,成放射性疼痛,伴有便血,舌苔白腻,服用过奥美拉唑肠溶片、三九胃泰颗粒,多酶片、654-2等药物当时症状缓解,但停药时,过一会又复发。患者随后服用本发明实施例1的组合物,7天胃疼缓解很多,大便正常,气色较好,饭量较好。连续服用2个疗程后复查,溃疡愈合,继续服药半个月巩固治疗,至今没在出现胃痛。
案例2
吴某,男,80岁,云南人,15年前胃镜检查为出血性胃炎,临产症状表现为上腹部隐痛,偶尔剧痛,伴有恶心、口臭,便血,服用过中药汤剂、三九胃泰颗粒、藿香正气丸、奥美拉唑肠溶片、雷尼替丁胶囊等药物,服用多年,停药随即复发。患者随后服用本发明实施例2的组合物,11天胃疼缓解很多,精神较好。连续服用3个疗程后复查,大便正常,溃疡消失,继续服药1个疗程巩固治疗,半年回访后,至今没有出现胃痛。
以上所述,只是本发明选出的较佳实施例,并非本发明做任何的限制,凡是依据本发明的技术实质上是对实施例简单的调整修改,类似的改变,均是本发明的保护范围。
Claims (3)
1.一种含美洲大蠊提取物的胃溃疡组合物,其特征在于包含下列质量份的原料:
美洲大蠊水溶浸膏0.1~20份、木蝴蝶3~15份、紫苏梗10~50份、海螵蛸10~30份、白芍10~60份、延胡索10~60份、陈皮10~30份、茯苓10~50份、葛根10~50份、三七5~25份、甘草3~15份、硬脂酸镁0.15份~1份、淀粉20~50份、糊精30份~40份。
2.一种含美洲大蠊提取物的胃溃疡组合物的制备方法,其特征在于经过下列各步骤:
(1)按下列质量份的原料备料:
美洲大蠊水溶浸膏0.1~20份、木蝴蝶3~15份、紫苏梗10~50份、海螵蛸10~30份、白芍10~60份、延胡索10~60份、陈皮10~30份、茯苓10~50份、葛根10~50份、三七5~25份、甘草3~15份、硬脂酸镁0.15份~1份、淀粉20~50份、糊精30份~40份;
(2)将紫苏梗、海螵蛸和三七粉碎成混合细粉,过120目筛;
(3)将木蝴蝶、陈皮、甘草、白芍、葛根、茯苓和延胡索粉碎成混合粗粉,过10目筛,加入4~10体积倍的蒸馏水煎煮提取3次,第一二次各3小时,第三次2小时,合并提取液,过滤,滤液静置12~24小时,取上清液,经减压浓缩成浸膏;
(4)将美洲大蠊水溶浸膏、步骤(2)的混合细粉、步骤(3)的浸膏、硬脂酸镁、淀粉和糊精混匀,制成颗粒,整粒,再进行干燥,即得含美洲大蠊提取物的胃溃疡组合物。
3.根据权利要求2所述的制备方法,其特征在于:所述美洲大蠊水溶浸膏通过下列步骤制得:
A、浸泡:将美洲大蠊干燥的成虫,加入4~10体积倍的浓度为45~95v/v%的乙醇浸泡10~720天;
B、粉碎:将浸泡的美洲大蠊连同溶剂粉碎;
C、过滤:将粉碎后的美洲大蠊连同溶剂进行过滤得到浸出液;
D、减压浓缩:将浸出液在真空度﹣0.07~﹣0.08MPa、温度40~45℃的条件下减压浓缩成乙醇浓缩稠浸膏;
E、纯化分离:将乙醇浓缩稠浸膏的上层物、下层物分开收集,将上层物进行3次离心分离,合并上层,得到总上层物,再加入石油醚静置分层,分层后的上层物经浓缩成浸膏,即得到美洲大蠊水溶浸膏。
