Abstract

Description
技术领域
本发明涉及一种源自多能干细胞的血管内皮细胞的制造方法。
背景技术
哺乳类的血管由外膜、中膜、内膜的三层结构组成,内膜由一层血管内皮细胞覆盖。该血管内皮细胞除了释放各种血管活性物质,对血管的收缩/扩张进行调节以外,还抑制血小板的粘附、凝聚,对血管进行保护。
作为与血管内皮细胞相关的疾病,有缘起于位于血管内膜的内皮细胞的功能降低的动脉硬化等。内皮功能障碍、即内皮功能的消失会引起血管病,例如动脉粥样硬化症。血管内皮细胞以及血管内皮祖细胞(EPC)的治疗应用的有用性已被提出,报告有以下例子:对包括冠状动脉疾病、下肢缺血疾病(血栓闭塞性脉管炎(伯格氏病)、闭塞性动脉硬化症等)的重症缺血性疾病患者实施移植自体EPC的血管再生疗法,取得了良好的成绩(非专利文献1以及2)。然而,已确认该缺血改善效果依赖于EPC施用数,重要的是确保/施用更多的EPC,但是,对于由患者的自体血或骨髓液得到的EPC而言,往往其数量减少或者细胞功能降低,在这种情况下,无法得到充分的治疗效果。
另一方面,在此之前报告有:通过向胚胎干细胞(ES细胞)、体细胞导入未分化细胞特异性基因而得到的人工多能干细胞(iPS细胞)等具有多能性的细胞(专利文献1以及2)。这些细胞能够无限增殖,通过赋予分化刺激可能会得到用于移植治疗的血管内皮细胞,因此作为有用材料受到关注。
在此之前,研究了由人胚胎干细胞向内皮细胞分化诱导的方法(专利文献3)、由人多能干细胞向中胚层细胞的分化诱导方法(专利文献4、非专利文献3、非专利文献4)。但是,在此之前,未报告过由多能干细胞高效地诱导功能性优异的血管内皮细胞的方法。

发明内容
发明所要解决的问题
本发明的目的在于,由多能干细胞高效地制造血管内皮细胞。因此,本发明的问题在于提供一种将人多能干细胞、特别是人人工多能干细胞向血管内皮细胞高效地分化诱导的方法。
用于解决问题的方案
本发明人等为了解决上述问题,发现了:在由多能干细胞向中胚层祖细胞、进而向内皮细胞诱导的工序中,通过适当变更涂覆培养容器的基质,能够高纯度地分化诱导血管内皮细胞。进而,通过使用该方法,成功制造出不受每株多能干细胞向内皮细胞的分化偏好性的影响,功能性也优异的血管内皮细胞,从而完成了本发明。
即,本发明如下。
[1]一种方法,是包括以下(i)至(iii)的工序的由多能干细胞制造血管内皮细胞的方法,其包括:
(i)在由第一基质涂覆的培养器上,将多能干细胞在含有BMP(BoneMorphogenetic Protein:骨形态发生蛋白)的培养液中进行培养,制造中胚层祖细胞的工序;
(ii)使通过(i)得到的中胚层祖细胞解离为单细胞的工序;以及
(iii)在由选自由层粘连蛋白-411(LM411)或其片段、层粘连蛋白-511(LM511)或其片段、Matrigel(注册商标)、IV型胶原蛋白以及纤连蛋白组成的组中的第二基质涂覆的培养器上,将通过(ii)得到的细胞在含有VEGF(Vascular Endothelial Growth Factor:血管内皮生长因子)的培养液中进行培养的工序。

发明效果
根据本发明,能够高效地得到用于针对包括冠状动脉疾病、下肢缺血疾病(血栓闭塞性脉管炎、闭塞性动脉硬化症等)的缺血性疾病患者的移植治疗等的血管内皮细胞。

具体实施方式
以下详细地说明本发明。
本发明提供一种包括以下工序的由多能干细胞制造血管内皮细胞的方法:
(i)在由第一基质涂覆的培养器上,将多能干细胞在含有BMP的培养液中进行培养,制造中胚层祖细胞的工序;
(ii)使通过(i)得到的中胚层祖细胞解离为单细胞的工序;以及
(iii)在由选自由层粘连蛋白-411或其片段、层粘连蛋白-511或其片段、Matrigel、IV型胶原蛋白以及纤连蛋白组成的组中的第二基质涂覆的培养器上,将通过(ii)得到的细胞在含有VEGF的培养液中进行培养的工序。
由于上述工序(i)为由多能干细胞制造中胚层祖细胞的工序,因此,本发明也提供一种由多能干细胞制造中胚层祖细胞的方法。同样地,由于上述工序(iii)为由中胚层祖细胞制造血管内皮细胞的工序,因此,本发明也提供一种由中胚层祖细胞制造血管内皮细胞的方法。

可用于本发明的多能干细胞具有能够分化成存在于生物体的所有细胞的多能性,并且是还兼具增殖能力的干细胞,对其没有特别限定,例如包括:胚胎干(ES)细胞、源自通过核移植得到的克隆胚的胚胎干(ntES)细胞、精子干细胞(“GS细胞”)、胚胎生殖细胞(“EG细胞”)、人工多能干(iPS)细胞、源自培养成纤维细胞或骨髓干细胞的多能细胞(Muse细胞)等。优选的多能干细胞为ES细胞、ntES细胞以及iPS细胞。
(A)胚胎干细胞
ES细胞是由人、小鼠等哺乳动物的早期胚胎(例如胚泡)的内细胞团建立的、具有多能性和通过自我复制形成的增殖能力的干细胞。
ES细胞是源自来源于受精卵的8细胞期、桑椹胚后的胚(即胚泡)的内细胞团的胚的干细胞,具有分化为构成成体的所有细胞的能力、即分化多能性和通过自我复制形成的增殖能力。ES细胞在1981年发现于小鼠(M.J.Evans and M.H.Kaufman(1981),Nature 292:154-156),之后,在人、猴等灵长类中也建立了ES细胞株(J.A.Thomson et al.(1998),Science 282:1145-1147;J.A.Thomson et al.(1995),Proc.Natl.Acad.Sci.USA,92:7844-7848;J.A.Thomson et al.(1996),Biol.Reprod.,55:254-259;J.A.Thomson andV.S.Marshall(1998),Curr.Top.Dev.Biol.,38:133-165)。
ES细胞可以通过从对象动物的受精卵的胚泡取出内细胞团,并将内细胞团在成纤维细胞的饲养层上进行培养来建立。此外,通过传代培养实现的细胞的维持可以使用添加了白血病抑制因子(leukemia inhibitory factor(LIF))、碱性成纤维细胞生长因子(basic fibroblast growth factor(bFGF))等物质的培养液来进行。关于人以及猴的ES细胞的建立和维持的方法,例如记载于USP5,843,780;Thomson JA,et al.(1995),ProcNatl.Acad.Sci.U S A.92:7844-7848;Thomson JA,et al.(1998),Science.282:1145-1147;H.Suemori et al.(2006),Biochem.Biophys.Res.Commun.,345:926-932;M.Ueno etal.(2006),Proc.Natl.Acad.Sci.USA,103:9554-9559;H.Suemori et al.(2001),Dev.Dyn.,222:273-279;H.Kawasaki et al.(2002),Proc.Natl.Acad.Sci.USA,99:1580-1585;Klimanskaya I,et al.(2006),Nature.444:481-485等。
可以使用例如补充有0.1mM 2-巯基乙醇、0.1mM非必需氨基酸、2mM L-谷氨酸、20%KSR以及4ng/ml bFGF的DMEM/F-12培养液来作为用于制作ES细胞的培养液,在37℃、2%CO2/98%空气的湿润气氛下维持人ES细胞(O.Fumitaka et al.(2008),Nat.Biotechnol.,26:215-224)。此外,ES细胞需要每隔3~4天进行传代,此时,传代可以使用例如含有1mM CaCl2以及20%KSR的PBS中的0.25%胰蛋白酶以及0.1mg/ml胶原酶IV来进行。
ES细胞的选择通常可以以碱性磷酸酶、Oct-3/4、Nanog等基因标记的表达为指标,通过实时定量荧光PCR法(Real-Time PCR法)来进行。特别是,在人ES细胞的选择中,可以以OCT-3/4、NANOG、ECAD等基因标记的表达为指标(E.Kroon et al.(2008),Nat.Biotechnol.,26:443-452)。
对于人ES细胞株而言,例如WA01(H1)以及WA09(H9)能够从WiCell ReserchInstitute获取,KhES-1、KhES-2以及KhES-3能够从京都大学再生医科学研究所(京都、日本)获取。
(B)精子干细胞
精子干细胞是源自精巢的多能干细胞,是作为用于形成精子的起源的细胞。该细胞与ES细胞同样地,能够分化诱导成各种系列的细胞,具有例如当移植于小鼠胚泡时能制作出嵌合体小鼠等性质(M.Kanatsu-Shinohara et al.(2003)Biol.Reprod.,69:612-616;K.Shinohara et al.(2004),Cell,119:1001-1012)。既可以在含有源自神经胶质细胞系的神经营养因子(glial cell line-derived neurotrophic factor(GDNF))的培养液中进行自我复制,此外,也可以通过在与ES细胞同样的培养条件下反复进行传代来得到精子干细胞(竹林正则等(2008),实验医学,26卷,5号(增刊),41~46页,羊土社(东京、日本))。
(C)胚胎生殖细胞
胚胎生殖细胞是由胎生期的原始生殖细胞建立的、具有与ES细胞同样的多能性的细胞,可以通过在LIF、bFGF、干细胞因子(stem cell factor)等物质的存在下培养原始生殖细胞来建立(Y.Matsui et al.(1992),Cell,70:841-847;J.L.Resnick et al.(1992),Nature,359:550-551)。
(D)人工多能干细胞
人工多能干(iPS)细胞是可以通过将某一特定的重编程因子以DNA或蛋白质的形态导入体细胞来制作的、具有与ES细胞大致同等的特性,例如分化多能性和通过自我复制形成的增殖能力的源自体细胞的人工的干细胞(K.Takahashi and S.Yamanaka(2006)Cell,126:663-676;K.Takahashi et al.(2007),Cell,131:861-872;J.Yu et al.(2007),Science,318:1917-1920;Nakagawa,M.,Nat.Biotechnol.26:101-106(2008);国际公开WO2007/069666)。重编程因子是在ES细胞中特异性表达的基因或者对于维持ES细胞的未分化起到重要的作用的基因或其基因产物即可,没有特别限定,例如是OCT3/4、SOX2以及KLF4;OCT3/4、KLF4以及C-MYC;OCT3/4、SOX2、KLF4以及C-MYC;OCT3/4以及SOX2;OCT3/4、SOX2以及NANOG;OCT3/4、SOX2以及LIN28;OCT3/4以及KLF4等的组合。
这些因子既可以以蛋白质的形态通过例如脂质体转染、与细胞膜透过性肽的结合、显微注射等方法导入体细胞内,或者也可以以DNA的形态通过例如病毒、质粒、人工染色体等载体、脂质体转染、脂质体、显微注射等方法导入体细胞内。作为病毒载体,可举例示出:逆转录病毒载体、慢病毒载体(以上,Cell,126,pp.663-676,2006;Cell,131,pp.861-872,2007;Science,318,pp.1917-1920,2007)、腺病毒载体(Science,322,945-949,2008)、腺相关病毒载体、仙台病毒载体等。此外,作为人工染色体载体,例如包括人人工染色体(HAC)、酵母人工染色体(YAC)、细菌人工染色体(BAC、PAC)等。作为质粒,可以使用哺乳动物细胞用质粒(Science,322:949-953,2008)。在载体中,既可以含有启动子、增强子、核糖体结合序列、终止子、多聚腺苷酸化位点等调控序列,以便能够表达核初始化物质,进而,根据需要也可以含有药物抗性基因(例如卡那霉素抗性基因、氨苄青霉素抗性基因、嘌呤霉素抗性基因等)、胸苷激酶基因、白喉毒素基因等选择标记序列、绿色荧光蛋白质(GFP)、β葡萄糖醛酸苷酶(GUS)、FLAG等报告基因序列等。此外,对于上述载体,为了在导入体细胞之后,切除对重编程因子进行编码的基因或者同时切除启动子和对与其结合的重编程因子进行编码的基因,也可以在其前后具有LoxP序列。
在重编程时,为了提高诱导效率,除了上述因子以外,例如可以使用:组蛋白去乙酰化酶(HDAC)抑制剂[例如,丙戊酸(VPA)(Nat.Biotechnol.,26(7):795-797(2008))、曲古抑菌素A、丁酸钠、MC 1293、M344等低分子抑制剂、针对HDAC的siRNA以及shRNA(例如,HDAC1siRNA Smartpool (注册商标:Millipore)、HuSH 29mer shRNA Constructs againstHDAC1(OriGene)等)等核酸性表达抑制剂等]、DNA甲基转移酶抑制剂(例如5’-氮杂胞苷(5’-azacytidine))(Nat.Biotechnol.,26(7):795-797(2008))、G9a组蛋白甲基转移酶抑制剂[例如,BIX-01294(Cell Stem Cell,2:525-528(2008))等低分子抑制剂、针对G9a的siRNA以及shRNA(例如,G9a siRNA(human)(Santa Cruz Biotechnology)等)等核酸性表达抑制剂等]、L-channel calcium agonist(例如Bayk8644)(Cell Stem Cell,3,568-574(2008))、p53抑制剂(例如针对p53的siRNA以及shRNA(Cell Stem Cell,3,475-479(2008))、UTF1(Cell Stem Cell,3,475-479(2008))、Wnt Signaling(例如soluble Wnt3a)(Cell Stem Cell,3,132-135(2008))、2i/LIF(2i为mitogen-activated protein kinasesignalling(丝裂原活化蛋白激酶信号传导)以及glycogen synthase kinase-3(糖原合成酶激酶-3)的抑制剂、PloS Biology,6(10),2237-2247(2008))、miR-291-3p、miR-294、miR-295等miRNA(R.L.Judson et al.,Nat.Biotech.,27:459-461)(2009)等。
作为用于iPS细胞诱导的培养基,例如包括:(1)含有10~15%FBS的DMEM、DMEM/F12或DME培养基(这些培养基中可以进一步适当含有LIF、青霉素/链霉素(penicillin/streptomycin)、嘌呤霉素(puromycin)、L-谷氨酰胺、非必需氨基酸类、β-巯基乙醇等。);(2)含有bFGF或SCF(Stem Cell Factor:干细胞因子)的ES细胞培养用培养基,例如小鼠ES细胞培养用培养基(例如TX-WES培养基、トロンボX公司)或者灵长类ES细胞培养用培养基(例如灵长类(人&猴)ES细胞用培养基、ReproCELL、京都、日本)等。
作为培养法的例子,例如:可以在37℃、5%CO2存在下,在含有10%FBS的DMEM或DMEM/F12培养基上,使体细胞与重编程因子(DNA或蛋白质)接触,培养约4~7天,之后,将细胞重新接种在饲养层细胞(例如,丝裂霉素C处理STO细胞、SNL细胞等)上,从体细胞与重编程因子接触开始约10天后,在含有bFGF的灵长类ES细胞培养用培养基中进行培养,从该接触开始约30~约45天或更长时间之后,产生iPS样集落。
或者,作为其替代培养法,可以在37℃、5%CO2存在下,在饲养层细胞(例如,丝裂霉素C处理STO细胞、SNL细胞等)上,在含有10%FBS的DMEM培养基(其中可以进一步适当含有LIF、青霉素/链霉素、嘌呤霉素、L-谷氨酰胺、非必需氨基酸类、β-巯基乙醇等。)中进行培养,在约25~约30天或更长时间之后,产生ES样集落。理想的是,可举例示出:使用初始化的体细胞本身(Takahashi K,et al.(2009),PLoS One.4:e8067或WO2010/137746)或者使用细胞外基质(例如,Laminin-5(WO2009/123349)以及Matrigel(BD Biosciences))来替代饲养层细胞的方法。
除此以外,还可举例示出使用不含有血清的培养基进行培养的方法(Sun N,etal.(2009),Proc Natl Acad Sci U S A.106:15720-15725)。进而,为了提高建立效率,也可以利用低氧条件(0.1%以上且15%以下的氧浓度)来建立iPS细胞(Yoshida Y,et al.(2009),Cell Stem Cell.5:237-241或者WO2010/013845)。
在上述培养期间,从培养开始第2天以后,每天进行1次新鲜的培养液与培养液的更换。此外,用于核初始化的体细胞的细胞数没有限定,在每100cm2培养皿约5×103~约5×106个细胞的范围。
iPS细胞可以根据所形成的集落的形状来选择。另一方面,在将与体细胞初始化时表达的基因(例如,Oct3/4、Nanog)连动表达的药物抗性基因作为标记基因导入的情况下,可以选择通过在含有对应的药物的培养液(选择培养液)中进行培养而建立的iPS细胞。此外,在标记基因为荧光蛋白质基因的情况下,可以通过利用荧光显微镜进行观察来选择iPS细胞,在标记基因为发光酶基因的情况下,可以通过加入发光基质来选择iPS细胞,此外,在标记基因为显色酶基因的情况下,可以通过加入显色基质来选择iPS细胞。
在本说明书中使用的术语“体细胞”是指:除了卵子、卵母细胞、ES细胞等生殖系列细胞或者分化全能细胞以外的所有动物细胞(优选为包含人的哺乳动物细胞)。体细胞没有限定,既包含胎儿(幼崽)的体细胞、新生儿(幼崽)的体细胞以及成熟的健全或疾病性的体细胞的任一种,此外,还包含原代培养细胞、传代细胞以及株化细胞的任一种。具体而言,体细胞例如可举例示出:(1)神经干细胞、造血干细胞、间充质干细胞、牙髓干细胞等组织干细胞(成体干细胞);(2)组织祖细胞;(3)淋巴细胞、上皮细胞、内皮细胞、肌肉细胞、成纤维细胞(皮肤细胞等)、毛细胞、肝细胞、胃粘膜细胞、肠细胞、脾细胞、胰腺细胞(胰腺外分泌细胞等)、脑细胞、肺细胞、肾细胞以及脂肪细胞等分化了的细胞等。
此外,在将iPS细胞用作移植用细胞的材料的情况下,从不发生排斥反应的观点考虑,理想的是,使用移植目标的个体的HLA基因型相同或者实质上相同的体细胞。在此,“实质上相同”是指,HLA基因型一致到对于所移植的细胞能够通过免疫抑制剂来抑制免疫反应的程度,例如为具有HLA-A、HLA-B以及HLA-DR的3个基因座或者加入了HLA-C的4个基因座一致的HLA型的体细胞。
(E)源自通过核移植得到的克隆胚的ES细胞
nt ES细胞是源自通过核移植技术制作出的克隆胚的ES细胞,具有与源自受精卵的ES细胞大致相同的特性(T.Wakayama et al.(2001),Science,292:740-743;S.Wakayamaet al.(2005),Biol.Reprod.,72:932-936;J.Byrne et al.(2007),Nature,450:497-502)。即,由源自通过将未受精卵的核与体细胞的核进行置换而得到的克隆胚的胚泡的内细胞团建立的ES细胞为nt ES(nuclear transfer ES:核移植胚胎干细胞)细胞。为了制作nt ES细胞,利用核移植技术(J.B.Cibelli et al.(1998),Nature Biotechnol.,16:642-646)与ES细胞制作技术(上述)的组合(若山清香等(2008),实验医学,26卷,5号(增刊),47~52页)。在核移植中,通过向哺乳动物的去核后的未受精卵注入体细胞的核并培养几小时,能够进行初始化。
<中胚层祖细胞>
在本发明中,中胚层是指,包含由具有在孳生的过程中创建体腔以及支撑其的中皮、肌肉、骨骼、皮肤真皮、结缔组织、心脏/血管(也包含血管内皮)、血液(也包含血液细胞)、淋巴管或脾脏、肾脏以及输尿管、性腺(精巣、子宮、性腺上皮)的能力的细胞构成的胚层。在本发明中,中胚层祖细胞与中胚层细胞没有区别,例如为表达选自由T(与Brachyury同义)、KDR、FOXF1、FLK1、BMP4、MOX1以及SDF1构成的标记基因中的至少一个标记基因的细胞。优选为表达T以及KDR的细胞。在本发明中制造的中胚层祖细胞可以制造为含有其他细胞类型的细胞群,例如为含有50%以上、60%以上、70%以上、80%以上或者90%以上的中胚层祖细胞的细胞群。中胚层祖细胞例如可以通过所述工序(i)的方法、专利文献4的方法、非专利文献3的方法以及非专利文献4的方法进行制作,但制造方法没有特别限定。
<血管内皮细胞>
在本发明中,血管内皮细胞是指,构成血管的内表面的扁平且薄的细胞,但在本发明中与血管内皮祖细胞没有区别。本发明中的血管内皮细胞可以为通过继续培养能够形成管结构的细胞,进一步优选是指具有摄取乙酰化低密度脂蛋白(Ac-LDL)的能力的细胞。血管内皮细胞例如没有特别限定,但可通过表达KDR、CD34以及VE钙粘蛋白之类的标记来赋予特征。本发明中制造的血管内皮细胞可以制造为含有其他细胞类型的细胞群,例如为含有20%以上、30%以上、40%以上、50%以上、60%以上、70%以上、80%以上或者90%以上的血管内皮细胞的细胞群。诱导了分化的血管内皮细胞可以提供有用的培养模型。即,通过本发明的方法生成的血管内皮细胞作为起因于基因变异的血管疾病的模型是有用的。进而,可以将通过本方法生成的血管内皮细胞用于移植。
<工序(i):在由第一基质涂覆的培养器上,将多能干细胞在含有BMP的培养液中进行培养,制造中胚层祖细胞的工序>
本工序(i)为对多能干细胞进行粘附培养的工序,在本发明中,粘附培养是指,可以通过如下方式进行:使用进行了适于细胞粘附的表面加工的培养容器或者涂覆处理有细胞外基质的培养容器来进行培养。涂覆处理可以通过如下方式来进行:在将含有基质的溶液放入培养容器之后,适当地除去该溶液。
在本发明中,基质是指优选为细胞外基质(细胞外matrix),其是存在于细胞之外的超分子结构体,可以源自天然,也可以为人工物质(重组体)。例如可列举出:胶原蛋白、蛋白聚糖、纤连蛋白、透明质酸、生腱蛋白、巢蛋白(Entactin)、弹性蛋白、肌原纤维蛋白(Fibrillin)以及层粘连蛋白这样的物质或者它们的片段。这些细胞外基质可以组合使用,例如,可以为源自EHS小鼠肉瘤细胞的细胞外基质(Matrigel)等的源自细胞的制备物。
在本发明中,层粘连蛋白是指基底膜的主要细胞粘附分子,是由α链、β链以及γ链的3条亚基链构成的异三聚体,且是分子量80万Da的巨大的糖蛋白。3条亚基链在C末端侧缔合而创建卷曲螺旋结构,形成通过二硫键而稳定的异三聚体分子。没有特别限定,例如,α链为α1、α2、α3、α4或者α5,β链为β1、β2或者β3,以及γ链为γ1、γ2或者γ3。进而,层粘连蛋白可以为片段或者变异体,只要是具有整联蛋白结合活性的片段或者变异体,就没有特别限定。例如,层粘连蛋白片段可以为利用弹性蛋白酶消化得到的片段,即E8片段。层粘连蛋白优选源自人。
本工序(i)中使用的第一基质为Matrigel、IV型胶原蛋白、纤连蛋白、层粘连蛋白-411(由α4链、β1链、γ1链构成的层粘连蛋白)或其片段、层粘连蛋白-511(由α5链、β1链、γ1链构成的层粘连蛋白)或其片段,更优选为Matrigel或者层粘连蛋白-511或其片段,进一步优选为层粘连蛋白-511的E8片段(层粘连蛋白-511E8(LM511E8);Ido etal.J.Biol.Chem.282,11144-11154,2007)。
本工序(i)中使用的BMP是指适于向中胚层祖细胞诱导的BMP,例如可列举出:BMP2、BMP4以及BMP7。更优选为BMP4。BMP优选源自人。
本工序(i)的培养液中的BMP的浓度只要是能诱导出中胚层祖细胞的浓度,就没有特别限定,可举例示出:5ng/ml至200ng/ml、10ng/ml至100ng/ml或者20ng/ml至80ng/ml。优选为80ng/ml。
对于本工序(i)中使用的培养液而言,除了BMP以外,优选进一步含有GSK3β抑制剂以及VEGF。
在本发明中,GSK3β抑制剂定义为对GSK-3β蛋白的激酶活性(例如,针对β连环蛋白的磷酸化能力)进行抑制的物质,已知有许多物质,例如可列举出:作为靛玉红衍生物的BIO(别名:GSK-3β抑制剂IX;6-溴靛玉红3’-肟)、作为马来酰亚胺衍生物的SB216763(3-(2,4-二氯苯基)-4-(1-甲基-1H-吲哚-3-基)-1H-吡咯-2,5-二酮)、作为苯基α溴甲基酮化合物的GSK-3β抑制剂VII(4-二溴苯乙酮)、作为细胞膜透过型的磷酸化肽的L803-mts(别名:GSK-3β肽抑制剂;Myr-N-GKEAPPAPPQpSP-NH2(序列号1))以及具有高选择性的CHIR99021(6-[2-[4-(2,4-Dichlorophenyl)-5-(4-methyl-1H-imidazol-2-yl)pyrimidin-2-ylamino]ethylamino]pyridine-3-carbonitrile:6-[2-[4-(2,4-二氯苯基)-5-(4-甲基-1H-咪唑-2-基)嘧啶-2-基氨基]乙基氨基]吡啶-3-甲腈)。这些化合物例如从Calbiochem公司、Biomol公司等有售,可以容易地利用,也可以从其他获取方获取,或者也可以自行制作。本发明中使用的GSK-3β抑制剂可以优选为CHIR99021。
本工序(i)的培养液中的CHIR99021的浓度例如为1nM、10nM、50nM、100nM、500nM、750nM、1μM、2μM、3μM、4μM、5μM、6μM、7μM、8μM、9μM、10μM、15μM、20μM、25μM、30μM、40μM、50μM或者它们之间的浓度,但并不限定于此。优选为4μM。
本工序(i)的培养液中的VEGF的浓度只要是能诱导出中胚层祖细胞的浓度,就没有特别限定,可举例示出:5ng/ml至200ng/ml、10ng/ml至100ng/ml或者20ng/ml至80ng/ml。优选为80ng/ml。VEGF优选源自人。
本工序(i)中使用的培养液可以通过将用于培养动物细胞的培养基作为基础培养基并适当地添加BMP来制备。作为基础培养基,例如包含:Glasgow's Minimal EssentialMedium(GMEM:格拉斯哥极限必需培养基)培养基、IMDM培养基、Medium 199培养基、Eagle'sMinimum Essential Medium(EMEM:伊格尔极限必需培养基)培养基、αMEM培养基、Dulbecco's modified Eagle's Medium(DMEM:杜尔贝科改良伊格尔培养基)培养基、Ham'sF12培养基、RPMI 1640培养基、Fischer's培养基、Neurobasal Medium(LifeTechnologies)、mTesR1培养基(Life Technologies)、Essential 8(Life Technologies)、Stempro34SFM培养基(Life Technologies)以及它们的混合培养基等。培养基中可以含有血清,或者也可以没有血清。根据需要,培养基例如既可以含有:白蛋白、铁传递蛋白、Knockout Serum Replacement(KSR)(ES细胞培养时的FBS的血清替代物)、N2补充物(Invitrogen)、B27补充物(Invitrogen)、脂肪酸、胰岛素、胶原蛋白前体、微量元素、2-巯基乙醇、3’-硫醇丙三醇等1种以上的血清替代物,也可以含有:脂质、氨基酸、L-谷氨酰胺、Glutamax(Invitrogen)、非必需氨基酸、维生素、生长因子、低分子化合物、抗生素、抗氧化剂、丙酮酸、缓冲剂、无机盐类等1种以上的物质。优选的基础培养基为mTesR1培养基或者Essential 8。
关于本工序(i)的培养条件,培养温度没有特别限定,为约30~40℃,优选为约37℃,在含CO2的空气的气氛下进行培养,CO2浓度优选为约2~5%。培养时间为2天以上,优选为2天~3天。
<工序(ii):使细胞解离为单细胞的工序>
本工序(ii)为使通过工序(i)得到的细胞群实质上解离为单细胞的工序,作为使细胞解离的方法,例如可列举出:力学解离的方法、使用具有蛋白酶活性和胶原酶活性的解离溶液(例如,Accutase(商标)以及Accumax(商标)等)或者仅具有蛋白酶活性或胶原酶活性的解离溶液的解离方法。优选采用使用作为胰蛋白酶替代物的TrypLE Express(LifeTechnologies)进行解离的方法。
<工序(iii)或者由中胚层祖细胞制造血管内皮细胞的工序(以下,称为工序(iii)等):在由选自由层粘连蛋白-411或其片段、层粘连蛋白-511或其片段、Matrigel、IV型胶原蛋白以及纤连蛋白组成的组中的第二基质涂覆的培养器上,在含有VEGF的培养液中进行培养的工序>
本工序(iii)等为对中胚层祖细胞进行粘附培养的工序。本工序(iii)等中使用的中胚层祖细胞的制造方法没有特别限定,可以为通过所述工序(i)的方法制造出的中胚层祖细胞,该中胚层祖细胞也可以为经由所述工序(ii)被解离为单细胞的细胞。粘附培养可以使用所述工序(i)记载的基质来进行,本工序(iii)等中使用的优选的基质为Matrigel、IV型胶原蛋白、纤连蛋白、层粘连蛋白-411或其片段、层粘连蛋白-511或其片段,优选为层粘连蛋白-411的片段。层粘连蛋白-411的片段是指,对整联蛋白α6β1具有结合活性的片段,更优选为层粘连蛋白-411的E8片段(层粘连蛋白-411E8:LM411E8)。需要说明的是,层粘连蛋白可以包括其变异体,但不包括将从层粘连蛋白-411E8的γ链的C末端起第三位的谷氨酸置换为谷氨酰胺而使对整联蛋白α6β1的结合活性消失的变异体(层粘连蛋白-411E8(EQ))。
本工序(iii)等中使用的培养液可以通过将用于培养动物细胞的培养基作为基础培养基并适当地添加VEGF来制备。作为基础培养基,例如包含:Glasgow's MinimalEssential Medium(GMEM:格拉斯哥极限必需培养基)培养基、IMDM培养基、Medium 199培养基、Eagle's Minimum Essential Medium(EMEM:伊格尔极限必需培养基)培养基、αMEM培养基、Dulbecco's modified Eagle's Medium(DMEM:杜尔贝科改良伊格尔培养基)培养基、Ham's F12培养基、RPMI 1640培养基、Fischer's培养基、Neurobasal Medium(LifeTechnologies)、mTesR1培养基(Life Technologies)、Essential 8(Life Technologies)、Stempro34SFM培养基(Life Technologies)、Endothelial Serum Free Medium(LifeTechnologies)以及它们的混合培养基等。培养基中可以含有血清,或者也可以没有血清。根据需要,培养基例如既可以含有:白蛋白、铁传递蛋白、Knockout Serum Replacement(KSR)(ES细胞培养时的FBS的血清替代物)、N2补充物(Invitrogen)、B27补充物(Invitrogen)、脂肪酸、胰岛素、胶原蛋白前体、微量元素、2-巯基乙醇、3'-硫醇丙三醇等1种以上的血清替代物,也可以含有:脂质、氨基酸、L-谷氨酰胺、Glutamax(Invitrogen)、非必需氨基酸、维生素、生长因子、低分子化合物、抗生素、抗氧化剂、丙酮酸、缓冲剂、无机盐类等1种以上的物质。优选的基础培养基为Stempro34SFM培养基或者Endothelial SerumFree Medium(Life Technologies)。
本工序(iii)等的培养液中的VEGF的浓度只要是能诱导出血管内皮细胞的浓度,就没有特别限定,可举例示出:5ng/ml至200ng/ml、10ng/ml至100ng/ml或者20ng/ml至80ng/ml。优选为80ng/ml。
关于本工序(iii)等的培养条件,培养温度没有特别限定,为约30~40℃,优选为约37℃,在含CO2的空气的气氛下进行培养,CO2浓度优选为约2~5%。培养时间为2天以上,优选为4天~7天,特别优选为4天。
<血管再生剂>
本发明中得到的血管内皮细胞可以施用于包括冠状动脉疾病、下肢缺血疾病(血栓闭塞性脉管炎、闭塞性动脉硬化症等)的重症缺血性疾病患者的治疗。即,通过将所得到的血管内皮细胞移植于缺血部位,能够用作血管再生疗法(Takayuki Asahara,YAKUGAKUZASSHI 127(5)841-845,2007)。因此,在本发明中,提供一种血管再生剂,其含有通过上述方法由多能干细胞得到的血管内皮细胞。
<筛选方法>
在本发明中,提供一种包括以下工序的对包括冠状动脉疾病、下肢缺血疾病(血栓闭塞性脉管炎、闭塞性动脉硬化症等)的重症缺血性疾病的治疗药进行筛选的方法:
(i)使候选药物与通过上述方法得到的血管内皮细胞接触的工序;
(ii)对所述血管内皮细胞的功能障碍进行测定的工序;以及
(iii)选择与未使候选药物接触的情况相比使所述血管内皮细胞的功能障碍减少了的候选药物作为重症缺血性疾病的治疗药的工序。
在本发明中,血管内皮细胞的功能障碍是指,可举例示出:NO合成酶的表达或者NO的产生等氧化应激的增大、内皮细胞粘附分子的表达增强、血管紧张素II、内皮素-1和纤溶酶原激活剂/抑制剂-1的产生增加、以及Ac-LDL等脂质摄取量的增加等。
在本发明中,候选药物例如可举例示出:细胞提取物、细胞培养上清液、微生物发酵产物、源自海洋生物的提取物、植物提取物、纯化蛋白质或粗蛋白质、肽、非肽化合物、合成低分子化合物以及天然化合物。
在本发明中,候选药物还可以使用包括(1)生物学库、(2)使用解卷积(Deconvolution)的合成库法、(3)“一珠一化合物(one-bead one-compound)”库法、以及(4)使用亲和层析分选的合成库法的本技术领域公知的组合库法中众多途径的任一种来得到。使用亲和层析分选的生物学库法限定于肽库,其他四种途径可以适用于肽、非肽寡聚物或者化合物的低分子化合物库(Lam(1997)Anticancer Drug Des.12:145-67)。分子库的合成方法的例子可以在本技术领域中找到(DeWitt et al.(1993)Proc.Natl.Acad.Sci.USA90:6909-13;Erb et al.(1994)Proc.Natl.Acad.Sci.USA 91:11422-6;Zuckermann etal.(1994)J.Med.Chem.37:2678-85;Cho et al.(1993)Science 261:1303-5;Carell etal.(1994)Angew.Chem.Int.Ed.Engl.33:2059;Carell et al.(1994)Angew.Chem.Int.Ed.Engl.33:2061;Gallop et al.(1994)J.Med.Chem.37:1233-51)。化合物库可以制作为:溶液(参照Houghten(1992)Bio/Techniques 13:412-21)或珠(Lam(1991)Nature 354:82-4)、芯片(Fodor(1993)Nature 364:555-6)、细菌(美国专利第5,223,409号)、孢子(美国专利第5,571,698号、美国专利第5,403,484号以及美国专利第5,223,409号)、质粒(Cull et al.(1992)Proc.Natl.Acad.Sci.USA 89:1865-9)或者噬菌体(Scottand Smith(1990)Science 249:386-90;Devlin(1990)Science249:404-6;Cwirla et al.(1990)Proc.Natl.Acad.Sci.USA 87:6378-82;Felici(1991)J.Mol.Biol.222:301-10;美国专利申请第2002103360号)。
<试剂盒>
在本发明的其他实施方案中,包括由多能干细胞制作血管内皮细胞的试剂盒。该试剂盒包含:在上述制作血管内皮细胞的各工序中使用的培养液、添加剂或者培养容器等。例如,可列举出:包含选自由基质(优选层粘连蛋白-411E8)、BMP4、VEGF以及GSK3β抑制剂组成的组中的1种以上的试剂的试剂盒。在本试剂盒中,可以进一步包含记载有制造工序的流程的文书、说明书。
以下,列举实施例来更具体地说明本发明,但本发明当然不限定于这些实施例。
[实施例]
细胞以及培养
人ES细胞(KhES1)取自京都大学再生医科学研究所,通过现有的方法进行了培养(Suemori H,et al.Biochem Biophys Res Commun.345:926-32,2006)。人iPS细胞(253G4、409B2以及223Q5)取自京都大学的山中教授。
在SNL饲养层细胞上,使用添加了5mg/mL的bFGF(Wako)的ES培养基(ReproCELL)进行了人ES细胞以及人iPS细胞的维持培养。SNL饲养层细胞可以从DS Pharma BiomedicalCo.Ltd.等获取。此外,对于传代而言,利用CTK溶液(0.25%胰蛋白酶(LifeTechnologies)、0.1%胶原酶IV(Life Technologies)、20%KSR以及1mM CaCl2)在室温下处理约30秒来使细胞解离,通过现有的方法(Suemori,H.et al.Biochemical andBiophysical Research Communications 345,926932(2006)),去除了SNL细胞。
LM411E8片段的制作
按照Ido等(Ido H,et al.,J.Biol.Chem.,282,11144-11154,2007)记载的方法,将α4链E8片段、β1E8片段、γ1链E8片段的表达载体导入源自人肾脏的293F细胞(Invitrogen)来使LM411E8表达。
人层粘连蛋白α4链E8片段表达载体的制作
为了获得从5’侧依次对小鼠Ig-κ链V-J2-C信号肽、6×His标签和α4链E8片段进行编码的cDNA片段,分别取得对小鼠Ig-κ链V-J2-C信号肽和6×His标签进行编码的cDNA片段、以及对α4链E8进行编码的cDNA片段,通过延伸PCR对这两种片段进行连结/扩增。
首先,将人层粘连蛋白α5链E8表达载体(Ido et al.,J.Biol.Chem.,282,11144-11154,2007)作为模板,使用以下的引物组(i)进行PCR,对相当于小鼠Ig-κ链V-J2-C信号肽/6×His标签的区域进行扩增。需要说明的是,在反向(reverse)引物的5’侧附加用于延伸PCR的序列。
(i)信号肽序列/6×His标签序列扩增用引物
5’-GAGGTCTATATAAGCAGAGCTCTCTGGCTAACTA-3’(正向(forward)、序列号2)
5’-CATTGGCTTCATCATGATGATGATGATGATGAAGC-3’(反向(reverse)、序列号3)。
接着,将含有人层粘连蛋白α4链的全长cDNA序列的质粒(Hayashi et al.,Biochem Biophys Res Commun.,299,498-504,2002)作为模板来进行PCR,对相当于α4链(注册号:NP_002281的Glu629-His1449)的区域进行扩增。需要说明的是,分别在正向(forward)引物的5’侧附加用于延伸PCR的序列,在反向(reverse)引物的5’侧附加EcoRI识别序列。
(ii)层粘连蛋白α4链E8序列扩增用引物
5’-CATCATGATGAAGCCAATGAAACAGCAGAATTTGC-3’(正向(forward)、序列号4)
5’-GCAGAATTCTCAATGAGAGTTTCTTGGAGTATTCC-3’(反向(reverse)、序列号5)。
通过延伸PCR使所得到的两种cDNA片段连结/扩增,得到了对小鼠Ig-κ链V-J2-C信号肽/6×His标签/α4链E8进行编码的cDNA片段。利用限制酶HindIII和EcoRI对扩增后的cDNA进行消化,插入哺乳细胞用表达载体pSecTag2B(Invitrogen)的该部位,制作出人α4链E8片段(在N末端侧含有6×His标签)的表达载体。
人β1链E8片段(在N末端侧含有HA标签)、人γ1链E8片段(在N末端侧含有FLAG标签)的表达载体通过Ido等(Ido H,et al.,J.Biol.Chem.,282,11144-11154,2007)的方法来制作。
将所制作的各链的表达载体导入源自人肾脏的293F细胞而制作出LM411E8。使用转染试剂293fectin(商标:Life Technologies)以及Opti-MEM I(注册商标:Invitrogen),向1000ml的293F细胞(1.0×106个/ml)同时转染各链表达载体各400μg,进行72小时培养之后,回收培养液。培养液以1000×g离心10分钟,进一步将其上清液以15000×g离心30分钟,去除细胞、不溶物。向培养上清液中加入PMSF(最终浓度:1mM)和叠氮化钠(最终浓度:0.02%),使其充分混合。之后,向培养上清液中添加15ml的Ni-NTA agarose(Qiagen),培育一晚,使其吸附目标蛋白质。回收Ni-NTA agarose,利用TBS(-)(不含Ca、Mg的Tris缓冲生理盐水)进行洗涤之后,利用200mM咪唑/TBS(-)进行洗脱。利用分光光度计来确认洗脱馏分的A280值,向A280值高的馏分中添加5ml的ANTI-FLAG M2affinity Gel(Sigma),在4℃下使其旋转一晚。将凝胶转移至Econo柱,利用TBS(-)进行洗涤之后,利用含有100μg/ml FLAGpeptide(注册商标:Sigma)的TBS(-)进行洗脱。通过CBB染色来确认洗脱馏分,汇总LM411E8的洗脱出的馏分,对PBS(-)(不含Ca、Mg的磷酸缓冲生理盐水)进行透析。
涂覆
Laminin-411(LM411)(Biolamina)以及Laminin-511(LM511)(Biolamina)利用PBS(-)进行稀释,制备为20μg/mL的浓度之后,以达到2μg/cm2的浓度的方式分注至培养皿。GFR-Matirgel(BD Biosciences)利用PBS(-)进行稀释,在冰上制备为200μg/mL的浓度之后,以达到20μg/cm2的浓度的方式分注至培养皿。Type IV collagen(BD Biosciences)利用0.05M盐酸进行稀释,制备为100μg/mL的浓度之后,以达到10μg/cm2的浓度的方式分注至培养皿。Fibronectin(Millipore)利用PBS(-)进行稀释,制备为20μL/mL的浓度之后,以达到2μg/cm2的浓度的方式分注至培养皿。LM511E8fragment(Nippi)以及LM411E8利用PBS(-)进行稀释,制备为4μL/mL的浓度之后,以达到0.4μg/cm2的浓度的方式分注至培养皿。
任一种涂覆剂均通过在分注后以37℃培育2小时来进行涂覆。
实施例1
由多能干细胞向中胚层祖细胞的分化诱导
由多能干细胞向中胚层祖细胞的分化诱导按照Niwa等的记载来进行(Niwa A,etal,PLoS One.6:e22261 2011,Yanagimachi MD,et al,PLoS One.8:e59243,2013)。详细而言,将多能干细胞的集落以2colonies/cm2的密度接种于由GFR-Matrigel涂覆的板,在mTeSR1培养基(STEMCELL TECHNOLOGIES)中进行培养。使其增殖至集落的直径达到约750μm之后,更换为含有20ng/mL的BMP4(R&Dsystems)的mTeSR1培养基,培养3天(图1中第3天(Day3))。
由中胚层祖细胞向内皮细胞的分化诱导
将含有如上所述得到的中胚层祖细胞的培养物的培养基更换为含有40ng/mLVEGF(R&D systems)的Stempro34SFM(Life Technologies),进一步培养4天(图1中第7天(Day7))。
血管内皮细胞的提取/功能确认
利用流式细胞仪对分化诱导第7天的标记的表达进行了分析。详细而言,将所得到的细胞利用TrypLE Express在37℃下处理20分钟,在Stempro34培养基中进行了抗体反应。抗人KDR抗体(Biolegend)、抗人CD34抗体(Beckman coulter)以及抗人VE-cadherin抗体(eBioscience)均以1:100稀释来使用。其结果是,在约10%的细胞中观察到KDR/CD34/VE-钙粘蛋白(VE-cadherin)的表达(图2)。
接着,在继续培养的分化诱导第10天提取CD34+/VE-钙粘蛋白(VE-cadherin)+馏分,进行了Ac-LDL摄取试验以及免疫染色。详细而言,将DiI-Ac-LDL(Life Technologies)以1:100的稀释率加入Endothelial Serum Free Medium(Life Technologies),使其与在37℃、5%CO2的条件下得到的细胞反应5小时之后,利用PBS(-)洗涤两次,使用Cytofix(BDBiosciences)来固定细胞。固定在室温下进行5分钟。之后,使用Perm/Wash(BDBiosciences)在室温下进行30分钟封闭,使抗人CD31抗体(R&D systems)(1:10)在4℃下反应一晚。利用Perm/Wash洗涤两次之后,在室温下进行二次抗体反应(FITC标记抗羊IgG抗体,Jackson immunoresearch,1:100)1小时。接着,利用Perm/Wash洗涤两次,通过荧光显微镜(Keyence)进行拍摄。其结果是,观察到作为血管内皮细胞标记的CD31的表达、以及作为血管内皮细胞的功能之一的Ac-LDL的摄取(图3)。根据以上内容,给出了如下启示:诱导出的CD34+/VE-钙粘蛋白(VE-cadherin)+细胞为血管内皮细胞。
VEGF刺激前的传代培养的效果
在分化诱导第3天,KDR阳性中胚层祖细胞出现80%以上(图4),但是,之后即使利用VEGF进行刺激,成为血管内皮细胞的也少。因此,以消除在第3天之前形成的细胞间相互作用为目的,在利用BMP/Matrigel进行培养之后,即将利用VEGF进行刺激之前,通过利用TrypLE Express(Life Technologies)在37℃下处理20分钟而使细胞解离,再次通过平面培养(VEGF/Matrigel)使单细胞(single cell)分化。其结果是,在解离至单细胞(singlecell)的组(passage组:传代组)中,观察到CD34+/VE-钙粘蛋白(VE-cadherin)+细胞的纯度上升(图5)。
使用了单一基质板的分化诱导
如上所述,通过在第3天对细胞进行解离,观察到CD34+/VE-钙粘蛋白(VE-cadherin)+细胞的纯度上升,但是,其效果根据细胞株而不同(图5)。因此,进行使用了由各种单一基质蛋白质涂覆的第二基质板的分化诱导(第3天(Day3)至第7天(Day7)),探索到能高纯度且株距稳定地诱导血管内皮细胞的基质。将分化诱导方法示于图6。其结果是,示出了:对于非涂覆(non-coating)和LM411而言,无论哪种细胞株均能够高纯度地诱导血管内皮细胞(图7)。另一方面,在被认为对于维持多能干细胞有用的LM511中,存在纯度降低的细胞株。此外,对血管内皮细胞的产量进行比较的结果是,LM411为高产量。
进而,为了对使用LM411得到的血管内皮细胞的功能进行评价,进行了Ac-LDL摄取试验以及管形成试验。管形成试验通过以下方法来进行。通过将Matrigel(BDBiosciences)以50μL/well分注至96well plate并在37℃下放置30分钟来使其固形化。接着,将细胞悬浮于Endothelial Serum Free Medium,以达到80ng/mL的方式加入VEGF。将该细胞悬浮液以细胞密度为4×104/well的方式分注至先前固形化后的Matrigel上,在37℃、5%CO2的条件下培养一晚(Kurian L,et al.,Nat.Methods.10:77-83,2013)。其结果是,确认到:使用LM411诱导出的血管内皮细胞具有管形成能力以及Ac-LDL摄取能力(图8)。根据以上内容,确认到:通过使中胚层祖细胞解离并在由LM411涂覆的板上进行培养,能够高效地诱导血管内皮细胞。
由层粘连蛋白-411 E8片段(Laminin-411 E8 fragment)实现的产量的改善
接着,关于使用了作为具有与LM411的整联蛋白(integrin)结合的部分的片段的LM411E8的情况下的、对于针对中胚层祖细胞的粘附活性以及分化的影响进行了研究。详细而言,将多能干细胞的集落以2colonies/cm2的密度接种于由GFR-Matrigel涂覆的板,在mTeSR1培养基(STEMCELL TECHNOLOGIES)中进行培养。使其增殖至集落的直径达到约750μm之后,更换为含有80ng/mL的BMP4(R&D systems)的mTeSR1培养基。在培养3天之后,通过利用TrypLEExpress(Life Technologies)在37℃下处理20分钟而使细胞解离,悬浮于含有80ng/mL VEGF(R&D systems)的Stempro34SFM(Life Technologies)培养基,以4×104/cm2的密度接种于由LM411E8涂覆的板,进一步培养4天。
其结果是,在使用了LM411E8的情况下,以与LM411大致相同的纯度得到了血管内皮细胞,产量比使用了LM411的情况多(图9左)。此外,依赖于LM411E8的浓度,血管内皮细胞的数量有所增加(图9右)。
此外,观察到使用LM411E8分化出的血管内皮细胞具有管形成能力以及Ac-LDL摄取能力(图10)。
进而,对使用非涂覆(non-coating)、LM411E8或者LM411分化诱导出的血管内皮细胞进行比较的结果是,在非涂覆(non-coating)条件下未形成管,LM411E8的管的长度、分支点的数量均比LM411高(图11以及图12)。根据以上内容,示出了:通过使用LM411E8来进行诱导,可得到功能性更多的血管内皮细胞。
对在上述三个条件下分化出的内皮细胞的基因表达谱进行了由RNA-seq实现的单细胞微阵列分析的结果是,示出了:接近于人脐带静脉血管内皮细胞(HUVEC)的基因表达。进一步进行了聚类分析的结果是,LM411E8显示出与非涂覆(non-coating)、LM411不同的基因表达谱。特别是,观察到:对于VEGF刺激的响应、涉及血管形成的基因表达在LM411E8中增强。
源自多能干细胞的中胚层祖细胞与LM411E8的相互作用
作为通过使用LM411E8提高了内皮细胞的产量的理由,可以认为是与细胞的粘附活性。因此,对分化诱导第3天的中胚层祖细胞的对于非涂覆(non-coating)、LM411或者LM411E8的粘附活性进行比较的结果是,LM411E8显著高于其他两种。
已知LM411与integrinα6β1、α7x1β1进行结合。因此,针对分化诱导第3天的中胚层祖细胞使用integrinα6β1的中性抗体进行了细胞粘附试验(celladhesion assay)的结果是,粘附细胞数降低至与非涂覆(non-coating)相同的程度。LM411E8(EQ)是通过将从LM411E8的γ链的C末端起第三位的谷氨酸置换为谷氨酰胺而对integrinα6β1的结合活性消失的变异体。对针对该LM411E8(EQ)进行的分化诱导第3天的中胚层祖细胞的结合活性进行了评价的结果是,显示出与非涂覆(non-coating)大致相同的值。根据以上内容,可以认为:分化诱导第3天的中胚层祖细胞的对于LM411E8的初始粘附(initial adhesion)依赖于integrinα6β1。
LM411-E8的功能
为了研究LM411是分选了向血管内皮细胞分化的细胞,还是促进了所粘附的细胞的血管内皮分化,进行了如下双转换试验(double-switching assay),即,在使利用BMP4/Matrigel进行的分化诱导第3天得到的中胚层祖细胞解离之后,接种于LM411,将所粘附的细胞再次重新传代于Matrigel(MG)或者LM411。其结果是,与LM411-LM411相比,LM411-MG纯度显著降低(图13)。根据以上内容,可以认为:LM411不仅受体特异性地分选了细胞,对所粘附的细胞之后的血管内皮分化也带来了影响。此外,为了考查LM411E8是否具有单独诱导血管内皮分化的功能,在LM411E8之上以没有VEGF的条件对分化诱导第3天的中胚层祖细胞进行了培养的结果是,示出了:未诱导出血管内皮细胞,即LM411E8本身不具有诱导血管内皮分化的功能。
根据以上内容,给出了如下启示:在中胚层祖细胞的VEGF依赖性的血管内皮分化中,LM411E8对于祖细胞的分选和之后的分化的方向性确定发挥了重要的作用。
单细胞RNA测序(Single-cell RNA-sequencing)
为了详细地研究在从中胚层向血管内皮细胞的分化过程中,基质分选对各个细胞带来了何种影响,进行了单细胞RNA测序(single-cell RNA-sequencing)。在此,对利用BMP4/Matrigel使多能干细胞分化而得到的中胚层祖细胞(Day3:第3天)进行解离之后,使其在Matrigel或者LM411E8上分化,以单细胞等级对分化诱导第5天(Day5)以及第7天(Day7)的基因表达进行了比较(图14)。进行了主要成分分析的结果是,在分化诱导第0天以及第3天比较散乱的基因表达被重新接种在LM411E8上,结果变成均匀的群。另一方面,在Matrigel上,细胞的表达的不均匀性(heterogeneity)未改善。根据以上内容,从基因表达谱的观点考虑,也示出了:在细胞的命运的方向性确定中,LM411E8发挥了对基因表达谱进行限定的导向的作用。进而,针对分化诱导第5天的使用LM411E8以及Matrigel得到的血管内皮细胞进行微阵列,进行了Gene Set Enrichment Analysis(GSEA:基因集富集分析)的结果是,给出了如下启示:Rho家族GTP酶途径(Rho family GTPase pathway)在LM411E8中得以激活。
实施例2
由Wnt信号的增强实现的中胚层祖细胞的分化效率的改善
已知通过激活Wnt/β-catenin信号来提高中胚层分化诱导效率(Sumi,T.,et al.,Development.135:2969-2979,2008),作为强大的GSK3β抑制剂的CHIR99021用于血细胞、内皮的分化诱导(Sturgeon,C,et al.,Nat Biotechnol.32:554-561,2014)。根据单细胞RNA-seq(Single cell RNA-seq)的结果,示出了:分化诱导第3天的、作为Wnt信号的已知的报告基因的AXIN2的增加在单细胞等级上是不均匀的,由此,明确了:Wnt/β-catenin路径的内源性激活(endogenous activation)在初始分化中并不充分。根据单细胞RNA测序(Singlecell RNA seq)分析,可以认为在现有方法中初始分化的内源性wnt信号的激活是不均匀的,因此,可以认为可能是:通过利用CHIR99021同样地激活Wnt/β-catenin信号,能够增加粘附于LM411E8且分化为血管内皮细胞的祖细胞的数量。此外,可以认为也可能是:通过从分化的初期添加VEGF,能够逐步促进从表达了KDR的中胚层祖细胞向血管内皮细胞的分化。
因此,将多能干细胞以5colonies/cm2的密度接种于由LM511E8 fragment涂覆的板,在mTeSR1中进行培养。使其增殖至集落的直径达到750μm之后,更换为含有4μMCHIR99021(Wako)、80ng/mL BMP4、80ng/mL VEGF的Essential8(Life Technologies)培养基,由此开始分化诱导(图15)。在分化开始后两天,通过利用TrypLE Express在37℃下处理20分钟而使细胞解离,悬浮于含有80ng/mL VEGF的Stempro34培养基,以4×104/cm2的密度接种于由LM411E8涂覆的板,培养4天。
如上所述,示出了:在改良后的初始分化的条件下诱导出的中胚层祖细胞也适合于使用LM411E8作为第二基质的培养体系,能够高纯度地诱导血管内皮细胞(图16左图),能够由一个多能干细胞诱导10个以上的血管内皮细胞(图16右图)。此外,在这些内皮细胞还确认到具有绳状结构形成(管形成)以及Ac-LDL的摄取的功能(图17)。
在体内(in vivo)的血管形成能力
将上述得到的诱导血管内皮细胞以1×107/mL的密度悬浮于以达到300ng/mL的方式加入bFGF(Wako)的Matrigel。将该悬浮液100μL皮下注射于NOG小鼠(6周龄前后)的背部(Nakahara M,et al,cloning and stem cells,11:509-522,2009)。在移植后第21天回收Matrigel,通过免疫荧光染色进行分析。详细而言,将Matrigel利用4%多聚甲醛(Wako)在4℃下固定一晚。之后,利用20%蔗糖溶液在4℃下置换一晚,使用O.C.T.compound进行了冻结包埋。将其制成厚度6μm的切片,使其吸附于载玻片。使切片干燥之后,使用Cytofix在室温下处理5分钟,由此进行固定。接着,通过使用Perm/Wash在室温下培育30分钟来增强透过性,进行抗体反应。对于一次抗体而言,将羊抗CD31抗体(BD Biosciences,1:10)、小鼠抗人核抗体(Millipore,1:100)、大鼠Alexa fluor647标记抗小鼠TER-119抗体(BDBiosciences,1:10)溶解于Perm/Wash,在室温下使其反应2小时。对于二次抗体反应而言,将抗羊IgG抗体(Jackson immunoresearch,1:100)、抗小鼠IgG抗体(Jacksonimmunoresearch,1:100)溶解于Perm/Wash,在室温下使其反应1小时。使用荧光显微镜(OLYMPUS,fluoview)进行拍摄。
其结果是,观察到:通过本方法诱导出的血管内皮细胞在生物体内形成管腔结构,在内腔中存在小鼠红细胞(图18)。即,示出了:通过使用了LM411E8的基质转换法,即使在体内(in vivo)也能够诱导功能性血管内皮细胞。
工业上的可利用性
通过本发明,能够由ES细胞、iPS细胞等多能干细胞制作血管内皮细胞。血管内皮细胞可以使用于以包括冠状动脉疾病、下肢缺血疾病(血栓闭塞性脉管炎、闭塞性动脉硬化症等)的缺血性疾病的治疗为目的的血管再生医疗的领域。
Claims (14)
1.一种方法,是包括以下(i)至(iii)的工序的由多能干细胞制造血管内皮细胞的方法,其包括:
(i)在由第一基质涂覆的培养器上,将多能干细胞在含有BMP的培养液中进行培养,制造中胚层祖细胞的工序;
(ii)使通过(i)得到的中胚层祖细胞解离为单细胞的工序;以及
(iii)在由是层粘连蛋白-411或其具有整联蛋白结合活性的片段的第二基质涂覆的培养器上,将通过(ii)得到的细胞在含有VEGF的培养液中进行培养的工序。
2.根据权利要求1所述的方法,其中,所述(iii)的第二基质为层粘连蛋白-411的具有整联蛋白结合活性的片段。
3.根据权利要求1所述的方法,其中,所述层粘连蛋白-411的具有整联蛋白结合活性的片段为层粘连蛋白-411E8。
4.根据权利要求1所述的方法,其中,所述(i)的第一基质为Matrigel或者层粘连蛋白-511或其具有整联蛋白结合活性的片段。
5.根据权利要求4所述的方法,其中,所述层粘连蛋白-511的片段为层粘连蛋白-511E8。
6.根据权利要求1~5中任一项所述的方法,其中,所述BMP为BMP4。
7.根据权利要求1~5中任一项所述的方法,其中,所述(i)的培养液进一步含有GSK3β抑制剂以及VEGF。
8.根据权利要求7所述的方法,其中,所述GSK3β抑制剂为CHIR99021。
9.根据权利要求1~5中任一项所述的方法,其中,所述工序(i)进行2天或者3天。
10.根据权利要求1~5中任一项所述的方法,其中,所述工序(iii)进行4天。
11.一种方法,是包括以下工序的、由中胚层祖细胞制造血管内皮细胞的方法,其包括:
在由是层粘连蛋白-411或其具有整联蛋白结合活性的片段的基质涂覆的培养器上,将中胚层祖细胞在含有VEGF的培养液中进行培养的工序。
12.根据权利要求11所述的方法,其中,所述基质为层粘连蛋白-411的具有整联蛋白结合活性的片段。
13.根据权利要求12所述的方法,其中,所述层粘连蛋白-411的具有整联蛋白结合活性的片段为层粘连蛋白-411E8。
14.根据权利要求11~13中任一项所述的方法,其中,所述工序进行4天。
