Abstract
本发明提供了一种增强仔猪免疫力的微生态制剂,该制剂该微生态制剂包含菌粉,所述的菌粉是将约氏乳杆菌和胶红酵母接种于新鲜荆芥的浆液中发酵后干燥而成。约氏乳杆菌和胶红酵母在荆芥浆液中生长并产生多种蛋白酶等代谢物,同时荆芥能促使微生物产生促生长素、非特异性免疫调节因子、维生素等营养物质,这些代谢物成为抑制其它病源菌的部分物质基础,从而发挥增强仔猪免疫力的作用。
Description
技术领域
本发明属于微生态制剂技术领域,具体而言,涉及一种增强仔猪免疫力的微生态制剂及其制备方法。
背景技术
众所周知,长期滥用抗生素会破坏动物体内的菌群平衡,导致菌群紊乱免疫功能下降,而未被杀死的有害细菌会变成耐药性菌株,动物更容易致病,不易治疗。另外,抗生素的应用容易导致仔猪自身免疫力下降,当遇到传染病流行时,仔猪便大批死亡,且仔猪饲养中所使用的饲料添加剂对人体危害严重,人们甚至因此拒食猪肉。以上因素导致了仔猪市场混乱,仔猪养殖效益降低。同时,再加上药物残留问题不仅对环境造成了巨大的污染,还会影响畜禽产品质量对人类的健康造成严重的危害。因此,欧盟从1999年开始禁止在饲料中添加杆菌肽、螺旋霉素、维吉尼亚霉素、泰乐菌素4种抗生素;并在2006年开始全面禁止使用抗生素生长促进剂。
目前,寻求和开发应用一种既能替代抗生素,又能调节动物机体微生态平衡、预防疾病、提高饲料转化率、促进动物健康和保护生态环境等方面的新型添加剂,已是当前养殖业的一个热门话题。而运用动物微生态学理论,把微生态制剂运用到猪的日粮配制中,对改善猪肠道菌群的平衡,解决消化道疾病更具有重要意义。现有技术中,CN106072657A公开了一种益生菌组合物及其制备方法,其组成如下:嗜酸乳杆菌菌粉1%-3%、干酪乳杆菌菌粉0.5%-1.5%、乳双歧杆菌菌粉1%-3%、甘露低聚糖5%-8%、圆苞车前子粉3%-5%、低聚果糖40%-60%、玉米低聚肽粉2%-5%、菊粉3%-5%、蓝莓粉5%-8%和麦芽糊精10%-25%。CN107410736A公开了一种提高仔猪免疫力加速生长的益生菌组合物,其原料按重量份包括以下组分:枯草芽孢杆菌10.0-30.0份、山楂15.0-40.0份、党参15.0-40.0份、麦芽糖15.0-30.0份、尿囊素5.0-10.0份、纳豆激酶8.0-16.0份、酵母菌6.0-12.0份、乳酸菌4.0-12.0份、双歧杆菌3.0-9.0份。
然而,上述现有技术提供的益生菌组合物组分繁杂,难以制定符合规定的质量标准,同时生产成本高,因此不利于工业化大规模生产和质量控制。
发明内容
鉴于现有技术的不足,本发明的目的在于提供一种可增强仔猪免疫力,且生产成本低、易于质量控制的微生态制剂及其制备方法。
为了实现技术目的,本发明人提供了如下技术方案:一种增强仔猪免疫力的微生态制剂,该微生态制剂包含菌粉,所述的菌粉是将约氏乳杆菌(Lactobacillus johnsonii)CICC NO.10861和胶红酵母(Rhodotorula mucilaginosa)CICC No.31030接种于新鲜荆芥的浆液中发酵后干燥而成。
进一步优选地,如上所述增强仔猪免疫力的微生态制剂,其是按1:(5-10)的活菌数比将约氏乳杆菌(Lactobacillus johnsonii)CICC NO.10861和胶红酵母(Rhodotorulamucilaginosa)CICC No.31030接种于新鲜荆芥的浆液中发酵,发酵液干燥后得到微生态制剂。
再进一步优选地,如上所述增强仔猪免疫力的微生态制剂,其是按1:(6-7)的活菌数比将约氏乳杆菌(Lactobacillus johnsonii)CICC NO.10861和胶红酵母(Rhodotorulamucilaginosa)CICC No.31030接种于新鲜荆芥的浆液中发酵,发酵液干燥后得到微生态制剂。
需要说明的是,本发明所述增强仔猪免疫力的微生态制剂,其中采用的新鲜荆芥为盛花时期的荆芥地上部分,包括茎叶和花穗。
一种上述增强仔猪免疫力的微生态制剂的制备方法,该制备方法包括菌种活化、液体扩大培养和荆芥发酵培养步骤,所述的荆芥发酵培养步骤为:对液体扩大培养后的约氏乳杆菌(Lactobacillus johnsonii)CICC NO.10861和胶红酵母(Rhodotorulamucilaginosa)CICC No.31030,按1:(5-10)的活菌数比混合接种于新鲜荆芥的浆液中发酵,发酵温度32-37℃,发酵时间12-18小时,所得发酵液干燥后得到微生态制剂。
进一步优选地,如上所述增强仔猪免疫力的微生态制剂的制备方法,其中的新鲜荆芥为盛花时期的荆芥地上部分,包括茎叶和花穗。
再进一步优选地,如上所述增强仔猪免疫力的微生态制剂的制备方法,其中新鲜荆芥的浆液是按料液比1:(1.5-6.0)加水打浆后灭菌,调pH=6.8-7.2而得。
再进一步优选地,如上所述增强仔猪免疫力的微生态制剂的制备方法,其中新鲜荆芥的浆液是按料液比1:(1.5-2.0)加水打浆后灭菌,调pH=6.8-7.0而得。
再进一步优选地,如上所述增强仔猪免疫力的微生态制剂的制备方法,其中的菌种活化步骤为:用pH为6.8-7.5、高压灭菌的培养基分别斜面培养约氏乳杆菌(Lactobacillus johnsonii)CICC NO.10861和胶红酵母(Rhodotorula mucilaginosa)CICC No.31030,在30-38℃培养条件下培养8-16小时。
再进一步优选地,如上所述增强仔猪免疫力的微生态制剂的制备方法,其中的液体扩大培养步骤为:分别对约氏乳杆菌(Lactobacillus johnsonii)CICC NO.10861和胶红酵母(Rhodotorula mucilaginosa)CICC No.31030进行液体扩大培养,在30-38℃培养条件下培养25-30小时。
大量实验研究表明,本发明的复合微生态制剂具有动态调节仔猪胃肠道菌群生态系统的作用,约氏乳杆菌和胶红酵母可协同增强仔猪免疫力,提高抗病菌和抗病毒的能力,提高饲料转化率,促进动物生长发育。与现有技术相比,本发明涉及的微生态制剂具有如下优点和显著进步:
(1)使用本发明的微生态制剂,一方面可通过生长繁殖消耗肠道内大量氧气,夺取其他氧型菌的生长条件;另一方面可通过自身产生一些物质以抑制其他有害菌生长,从而使肠道内有益菌重新占据优势地位,建立起良好的微生物群系。
(2)约氏乳杆菌和胶红酵母在荆芥浆液中生长并产生多种蛋白酶等代谢物,同时荆芥能促使微生物产生促生长素、非特异性免疫调节因子、维生素等营养物质,这些代谢物成为抑制其它病源菌的部分物质基础,从而发挥控制病害发生并达到预防和治疗的作用。
(3)使用本发明实施例1的微生态制剂能预防仔猪大肠杆菌病、增强免疫力和提高仔猪成活率都有显著作用。在降低仔猪发病率和死亡率的试验中显示,应用本发明的微生态制剂饲喂仔猪,可有效增强仔猪对大肠杆菌病的抵抗力,减少仔猪患病率和死亡率,提高仔猪日增重等生产性能。另外,饲喂本发明微生态制剂的仔猪粪样中的大肠杆菌数量显著降低,同时乳酸菌和双歧杆菌数量都有升高的趋势。可见,微生态制剂可改善生长猪肠道健康,降低大肠杆菌病发病率,提高饲料效率。
(4)本发明的微生态制剂具有无残留、无污染、不产生耐药性、药物残留和耐药菌株,可提高仔猪的生产性能和疾病抵抗力,适合添加在各种仔猪全价配合饲料、精料补充料、浓缩饲料和预混料中。
具体实施方式
下面通过具体实施方式对本发明作进一步详细说明。但本领域技术人员将会理解,下列实施例仅用于说明本发明,而不应视为限定本发明的范围。另外,实施例中未注明具体技术操作步骤或条件者,均按照本领域内的文献所描述的常规技术或条件或者按照产品说明书进行。另外,本发明实施例所采用的菌种均为现有技术已经公开并保藏的菌株。
实施例1:微生态制剂的制备
(1)斜面菌种活化:将pH为7.0、120℃高压灭菌30分钟的LB培养基制备成试管斜面,分别接种约氏乳杆菌CICC NO.10861和胶红酵母CICC No.31030,约氏乳杆菌在35℃条件下培养12小时,胶红酵母在30℃条件下培养16小时;
(2)液体扩大培养:对约氏乳杆菌CICC NO.10861在培养基中进行液体扩大培养,37℃条件下培养25小时,培养基组成为:酪蛋白胨10.0g,牛肉膏10.0g,酵母粉5.0g,葡萄糖5.0g,乙酸钠5.0g,柠檬酸二铵2.0g,吐温80 1.0g,K2HPO4 2.0g,MgSO4.7H2O0.2g,MnSO4.H2O0.05g,CaCO320.0g,琼脂15.0g,蒸馏水1.0L,pH6.8。对胶红酵母CICC NO.31030在培养基中进行液体扩大培养,在30℃条件下培养30小时,培养基为5°Bé麦芽汁1.0L,自然pH;
(3)荆芥发酵培养:对液体扩大培养后的约氏乳杆菌CICC NO.10861和胶红酵母CICC NO.31030按1:6的活菌数比混合接种于新鲜荆芥的浆液中发酵,发酵温度32℃,发酵时间15小时,其中的新鲜荆芥浆液是按料液比1:2(1g:2mL)在新鲜荆芥茎叶和花穗中加水打浆后灭菌,调pH=6.8-7.0而得;发酵结束后,发酵液经冻干得到微生态制剂。
实施例2:微生态制剂的制备
(1)斜面菌种活化:将pH为7.0、120℃高压灭菌30分钟的LB培养基制备成试管斜面,分别接种约氏乳杆菌CICC NO.10861和胶红酵母CICC No.31030,约氏乳杆菌在35℃条件下培养16小时,胶红酵母在30℃条件下培养16小时;
(2)液体扩大培养:对约氏乳杆菌CICC NO.10861在培养基中进行液体扩大培养,37℃条件下培养25小时,培养基组成为:酪蛋白胨10.0g,牛肉膏10.0g,酵母粉5.0g,葡萄糖5.0g,乙酸钠5.0g,柠檬酸二铵2.0g,吐温80 1.0g,K2HPO4 2.0g,MgSO4.7H2O0.2g,MnSO4.H2O0.05g,CaCO320.0g,琼脂15.0g,蒸馏水1.0L,pH6.8。对胶红酵母CICC NO.31030在培养基中进行液体扩大培养,在30℃条件下培养30小时,培养基为5°Bé麦芽汁1.0L,自然pH;
(3)荆芥发酵培养:对液体扩大培养后的约氏乳杆菌CICC NO.10861和胶红酵母CICC NO.31030按1:10的活菌数比混合接种于新鲜荆芥的浆液中发酵,发酵温度32℃,发酵时间15小时,其中的新鲜荆芥浆液是按料液比1:2(1g:2mL)在新鲜荆芥茎叶和花穗中加水打浆后灭菌,调pH=6.8-7.0而得;发酵结束后,发酵液经冻干得到微生态制剂。
实施例3:微生态制剂的制备
(1)斜面菌种活化:将pH为7.0、120℃高压灭菌30分钟的LB培养基制备成试管斜面,分别接种约氏乳杆菌CICC NO.10861和胶红酵母CICC No.31030,约氏乳杆菌在35℃条件下培养12小时,胶红酵母在30℃条件下培养16小时;
(2)一级液体扩大培养:对约氏乳杆菌CICC NO.10861在培养基中进行液体扩大培养,37℃条件下培养25小时,培养基组成为:酪蛋白胨10.0g,牛肉膏10.0g,酵母粉5.0g,葡萄糖5.0g,乙酸钠5.0g,柠檬酸二铵2.0g,吐温80 1.0g,K2HPO42.0g,MgSO4.7H2O 0.2g,MnSO4.H2O 0.05g,CaCO320.0g,琼脂15.0g,蒸馏水1.0L,pH6.8。对胶红酵母CICC NO.31030在培养基中进行液体扩大培养,在30℃条件下培养30小时,培养基为5°Bé麦芽汁1.0L,自然pH;
(3)荆芥发酵培养:二级液体扩大培养:对一级液体扩大培养后的约氏乳杆菌CICCNO.10861和胶红酵母CICC NO.31030分别进行二级液体扩大培养,发酵结束后,发酵液经冻干得菌粉;按重量比为10:10:1的比例将约氏乳杆菌菌粉、胶红酵母菌粉和荆芥粉混合,得到微生态制剂。
实施例4:微生态制剂对仔猪大肠杆菌病的影响
选取健康的仔猪76头,体质量在20kg±2kg范围内,没有使用任何抗菌药物。将所有仔猪随机分为如下四组:正常对照组、模型对照组、试验A组和试验B组。除正常对照组10头外,其余各组仔猪数为22头。所有试验猪圈养,正常对照组和模型对照组饲喂不含任何添加剂的全价饲料,试验A组饲喂在全价饲料的基础上添加0.5%实施例1微生态制剂的混合饲料,试验B组饲喂在全价饲料的基础上添加0.5%实施例3微生态制剂的混合饲料。饲喂一周后,除正常对照组外,其它各组每头猪肌注5mL猪大肠杆菌O78菌液(将猪大肠杆菌O78接种新鲜肉汤培养基,置于37℃恒温箱培养16-18h,琼脂平板涂抹法计算其含菌数,猪大肠杆菌O78含菌数为1.17×1010/mL),接种后接种后观察各组试验仔猪的精神、采食、饮水、粪便,并按原方式继续饲喂一周。
试验结束后,统计各组仔猪的患病率,死亡率、平均增重和相对增重。患病标准:仔猪的精神状态、饮食和粪便中有2项以上不正常时均判定为患病;平均增重:根据给予微生态制剂前及试验结束时每头猪的体重,计算每头猪的增重量,据此计算每组平均增重。

与模型对照组比较,*P<0.01;与试验Ⅱ组比较,#P<0.01。
根据表1的试验结果可以看出,应用本发明的微生态制剂饲喂仔猪,可有效增强仔猪对大肠杆菌病的抵抗力,减少仔猪患病率和死亡率,提高仔猪日增重等生产性能。另外,饲喂本发明微生态制剂的仔猪粪样中的大肠杆菌数量显著降低,同时乳酸菌和双歧杆菌数量都有升高的趋势。
Claims (10)
1.一种增强仔猪免疫力的微生态制剂,该微生态制剂包含菌粉,所述的菌粉是将约氏乳杆菌CICC NO.10861和胶红酵母CICC No.31030接种于新鲜荆芥的浆液中发酵后干燥而成。
2.根据权利要求1所述增强仔猪免疫力的微生态制剂,其特征在于,按1:(5-10)的活菌数比将约氏乳杆菌CICC NO.10861和胶红酵母CICC No.31030接种于新鲜荆芥的浆液中发酵,发酵液干燥后得到微生态制剂。
3.根据权利要求2所述增强仔猪免疫力的微生态制剂,其特征在于,按1:(6-7)的活菌数比将约氏乳杆菌CICC NO.10861和胶红酵母CICC No.31030接种于新鲜荆芥的浆液中发酵,发酵液干燥后得到微生态制剂。
4.根据权利要求1所述增强仔猪免疫力的微生态制剂,其特征在于,所述的新鲜荆芥为盛花时期的荆芥地上部分。
5.一种根据权利要求1所述增强仔猪免疫力的微生态制剂的制备方法,其特征在于,该制备方法包括菌种活化、液体扩大培养和荆芥发酵培养步骤,所述的荆芥发酵培养步骤为:对液体扩大培养后的约氏乳杆菌CICC NO.10861和胶红酵母CICC No.31030,按1:(5-10)的活菌数比混合接种于新鲜荆芥的浆液中发酵,发酵温度32-37℃,发酵时间12-18小时,所得发酵液干燥后得到微生态制剂。
6.根据权利要求5所述增强仔猪免疫力的微生态制剂的制备方法,其特征在于,所述的新鲜荆芥为盛花时期的荆芥地上部分。
7.根据权利要求6所述增强仔猪免疫力的微生态制剂的制备方法,其特征在于,所述新鲜荆芥的浆液是按料液比1:(1.5-6.0)加水打浆后灭菌,调pH=6.8-7.2而得。
8.根据权利要求7所述增强仔猪免疫力的微生态制剂的制备方法,其特征在于,所述新鲜荆芥的浆液是按料液比1:(1.5-2.0)加水打浆后灭菌,调pH=6.8-7.0而得。
9.根据权利要求5所述增强仔猪免疫力的微生态制剂的制备方法,其特征在于,所述的菌种活化步骤为:用pH为6.8-7.5、高压灭菌的培养基分别斜面培养约氏乳杆菌CICCNO.10861和胶红酵母CICC No.31030,在30-38℃培养条件下培养8-16小时。
10.根据权利要求5所述增强仔猪免疫力的微生态制剂的制备方法,其特征在于,所述的液体扩大培养步骤为:分别对约氏乳杆菌CICC NO.10861和胶红酵母CICC No.31030进行液体扩大培养,在30-38℃培养条件下培养25-30小时。
