Abstract
本发明涉及一种羊肚和灵芝复合制剂,包括以下重量份的各组分:羊肚菌丝体300‑600份和灵芝菌丝体300‑600份。本发明还提供一种羊肚和灵芝复合制剂的制备方法,包括以下步骤:羊肚菌丝体培养和灵芝菌丝体培养两个方面的步骤。本发明采用灵芝菌丝体,羊肚菌丝体按一定比例配制而成,为纯中药制剂,无任何副作用,安全性好,可以长期服用,是养胃抗癌、提高免疫力的高级保健品。

技术领域
本发明属于中药制剂领域,具体涉及一种羊肚和灵芝复合制剂及其制备方法。
背景技术
随着我国居民物质生活水平的不断改善,人们饮食摄入的热量不断增高,而运动量又普遍偏少,使得三高人群逐年增多,目前已接近有4亿人。再者,随着环境污染的加剧及不健康的饮食及紧张的工作节奏,也使人们的消化系统和免疫系统承受了极大的压力,因此越来越多的人患上了慢性萎缩性胃炎等消化系统疾病以及高血压、高血脂等富贵病、白血病和癌症等。目前,治疗这些疾病的药物主要以西药为主,这些药物在治疗疾病的同时,不可避免地给人们带来了不同程度的副作用,而且治疗的效果也不显著。
发明内容
鉴于此,本发明的目的在于,提供一种羊肚和灵芝复合制剂及其制备方法,旨在解决现有药物治疗效果不显著,且带有副作用的缺陷。
为了达到上述发明目的,采取的技术方案如下:
一种羊肚和灵芝复合制剂,包括以下重量份的各组分:
羊肚菌丝体300-600份 灵芝菌丝体300-600份。
本发明还提供了一种羊肚和灵芝复合制剂的制备方法,包括以下步骤:
A.羊肚菌丝体培养
(A-1)羊肚菌固体菌种培养
将琼脂25g,土豆200g,葡萄糖20g,KH2PO4 3g,MgSO4·7H2O 1.5g,维生素B1 1片,加入至饮用水1000ml中配制成固体培养基,并将该固体培养基分装到扁平瓶中,置于121℃的条件下灭菌30分钟后,将羊肚菌菌种接种到固体培养基中,22-25℃恒温培养7天;
(A-2)羊肚菌液体菌种培养
将小米50g,蔗糖20g,玉米面25g,豆粉17.5g,蛋白胨2g,葡萄糖1g,KH2PO4 3g,MgSO4·7H2O 1.5g,加入至饮用水1000ml中配制成液体培养基,并将该液体培养基分装到三角瓶中,置于121℃的条件下灭菌30分钟后,将步骤(A-1)培养得到的固体菌种接种到液体培养基中后,将三角瓶置于摇瓶机上并在22-25℃恒温条件下以转速180rpm培养96小时,然后在静置培养48h;
(A-3)羊肚菌丝体栽培种培养
将豆粉700g,谷糠200g,玉米面47g,硫酸锌3g,麦芽糖20g,蔗糖20g,石膏10g,亚硒酸氢钠0.3g,加入至饮用水200g中配制成栽培种培养基,并将该栽培种培养基分装于栽培种的袋中,置于121℃的条件下灭菌2小时后,将步骤(A-2)培养得到的液体菌种接种到栽培种培养基中,22-25℃恒温培养15天;待羊肚菌丝体长好后,在70℃的条件下沸腾干燥5分钟,将干燥后的羊肚菌丝体过80目筛待用;
B.灵芝菌丝体培养
(B-1)灵芝菌固体菌种培养
将琼脂25g,土豆200g,葡萄糖20g,KH2PO4 3g,MgSO4·7H2O 1.5g,维生素B1 1片,加入至饮用水1000ml中配制成固体培养基,并将该固体培养基分装到扁平瓶中,置于121℃的条件下灭菌30分钟后,将灵芝菌菌种接种到固体培养基中,22-25℃恒温培养7天;
(B-2)灵芝菌液体菌种培养
将小米50g,蔗糖20g,玉米面25g,豆粉17.5g,蛋白胨2g,葡萄糖1g,KH2PO4 3g,MgSO4·7H2O 1.5g,加入至饮用水1000ml中配制成液体培养基,并将该液体培养基分装到三角瓶中,置于121℃的条件下灭菌30分钟后,将步骤(B-1)培养得到的固体菌种接种到液体培养基中后,将三角瓶置于摇瓶机上并在22-25℃恒温条件下以转速180rpm培养96小时,然后在静置培养48h;
(B-3)灵芝菌丝体栽培种培养
将豆粉700g,谷糠200g,玉米面47g,硫酸锌3g,麦芽糖20g,蔗糖20g,石膏10g,亚硒酸氢钠0.3g,加入至饮用水200g中配制成栽培种培养基,并将该栽培种培养基分装于栽培种的袋中,置于121℃的条件下灭菌2小时后,将步骤(B-2)培养得到的液体菌种接种到栽培种培养基中,22-25℃恒温培养60天;待灵芝菌丝体长好后,在70℃的条件下沸腾干燥5分钟,将干燥后的灵芝菌丝体过80目筛待用;
C.将步骤A、B分别培养出的羊肚菌丝体和灵芝菌丝体按照羊肚菌丝体300-600份和灵芝菌丝体300-600份的比例混匀后即可制得羊肚和灵芝复合制剂。
羊肚菌(Morchella sp.),又名美味羊肚菌、俗称羊雀菌、包谷菌等,是一食药两用真菌中珍品之一。羊肚菌的菌丝体营养成分十分丰富,含有多种多糖、酶类、氨基酸、脂肪、吡喃酮抗生素、无机元素等成份。其中酶类包括谷氨酰转肽酶、羧甲基纤维素酶、漆酶、过氧化氢酶等酶类。其中氨基酸总氨基酸含量大大高于一般食用菌,特别是人体必需氨基酸含量较高,占氨基酸总量的49%,且每种必需氨基酸基本接近FAO/WHO推荐模式,是优质的蛋白源。脂肪类化合物有甘油酯、亚油酸、油酸酯、棕榈酸、麦角甾醇等。羊肚菌含有20种以上的无机元素,其中微量元素有Zn、Mn、Cu、Co、Cr、Fe、Ni、B、Sr、V等10种,并含有人体必需的必需的常量元素Ca、Mg、P等,尤其是Zn的含量最高。除了以上营养物质外,Bouillant等人还从羊肚菌中分离到几种抗菌、抗病毒的活性成分及血小板集落抑制因子、黑色素形成抑制剂酪氨酸酶抑制剂。羊肚菌的营养成分决定了其具有增强人体免疫力、抑制肿瘤、防癌、调节人体免疫力、预防动脉硬化等功能。
灵芝菌(Ganoderma lucidum sp.),俗称神仙草,是担子菌纲多孔菌科灵芝属真菌,是一种名贵食药两用真菌。灵芝菌丝体营养成分十分丰富,含有具有免疫调节、抗肿瘤、抗氧化、抗病毒、抗衰老、降血糖、降血脂等多方面的生物活性的多糖;灵芝含三萜类化合物,能够抑制各种癌细胞的生长并能诱导细胞发生凋亡有具有抗炎、抗肿瘤、抗氧化、抗病毒、保肝、抑制胆固醇合成等药理活性的三萜类化合物。灵芝多糖一般由D-葡萄糖、D-半乳糖、D-甘露糖、D-木糖、L-岩藻糖、L-鼠李糖、L-阿拉伯糖等单糖组成的一种大分子化合物,是灵芝的主要生物成分之一;其三萜类化合物是从灵芝中分离得到的一类具有生物活性的主要有效成分,是灵芝中的主要苦味成分,大约有130种以上,多数为高度氧化的羊毛甾烷衍生物,它具有较高的脂溶性。灵芝的醇提物具有明显的抗细胞增殖的活性,其具有抗乳腺癌,肝癌,骨髓性白血病肿瘤的作用。此外,灵芝孢子粉对拮抗NCAM-L1在癫痫发作时对脑组织的各种改变和损伤有重要的修复作用。
本发明的有益效果:本发明采用灵芝菌丝体,羊肚菌丝体按一定比例配制而成,为纯中药制剂,无任何副作用,安全性好,可以长期服用,是养胃抗癌、提高免疫力的高级保健品。

实施例1
一种羊肚和灵芝复合制剂,包括以下重量份的各组分:羊肚菌丝体300份,灵芝菌丝体600份。
实施例2
一种羊肚和灵芝复合制剂,包括以下重量份的各组分:羊肚菌丝体500份,灵芝菌丝体500份。
实施例3
一种羊肚和灵芝复合制剂,包括以下重量份的各组分:羊肚菌丝体600份,灵芝菌丝体300份。
本发明的羊肚和灵芝复合制剂优先选用羊肚菌丝体与灵芝菌丝体按照1:1的比例配制而成。
本发明实施例1-3中涉及的羊肚菌与灵芝菌购于中国科学院微生物保藏中心的菌种。
上述实施例1-3的一种羊肚和灵芝复合制剂的制备方法,包括以下步骤:
A.羊肚菌丝体培养
(A-1)羊肚菌固体菌种培养
将琼脂25g,土豆200g,葡萄糖20g,KH2PO4 3g,MgSO4·7H2O 1.5g,维生素B1 1片,加入至饮用水1000ml中配制成固体培养基,并将该固体培养基分装到扁平瓶中,置于121℃的条件下灭菌30分钟后,将羊肚菌菌种接种到固体培养基中,22-25℃恒温培养7天;
(A-2)羊肚菌液体菌种培养
将小米50g,蔗糖20g,玉米面25g,豆粉17.5g,蛋白胨2g,葡萄糖1g,KH2PO4 3g,MgSO4·7H2O 1.5g,加入至饮用水1000ml中配制成液体培养基,并将该液体培养基分装到三角瓶中,每500ml的三角瓶分装200ml的液体培养基,置于121℃的条件下灭菌30分钟后,将步骤(A-1)培养得到的固体菌种接种到液体培养基中后,将三角瓶置于摇瓶机上并在22-25℃恒温条件下以转速180rpm培养96小时,然后在静置培养48h,镜检无杂菌的待用;
(A-3)羊肚菌丝体栽培种培养
将豆粉700g,谷糠200g,玉米面47g,硫酸锌3g,麦芽糖20g,蔗糖20g,石膏10g,亚硒酸氢钠0.3g,加入至饮用水200g中配制成栽培种培养基,并将该500g栽培种培养基分装于栽培种的袋中,置于121℃的条件下灭菌2小时后,将步骤(A-2)培养得到的液体菌种接种到栽培种培养基中,22-25℃恒温培养15天至长满菌包;待羊肚菌丝体长好后,在70℃的条件下沸腾干燥5分钟,将干燥后的羊肚菌丝体过80目筛待用;
B.灵芝菌丝体培养
(B-1)灵芝菌固体菌种培养
将琼脂25g,土豆200g,葡萄糖20g,KH2PO4 3g,MgSO4·7H2O 1.5g,维生素B1 1片,加入至饮用水1000ml中配制成固体培养基,并将该固体培养基分装到扁平瓶中,置于121℃的条件下灭菌30分钟后,将灵芝菌菌种接种到固体培养基中,22-25℃恒温培养7天;
(B-2)灵芝菌液体菌种培养
将小米50g,蔗糖20g,玉米面25g,豆粉17.5g,蛋白胨2g,葡萄糖1g,KH2PO4 3g,MgSO4·7H2O 1.5g,加入至饮用水1000ml中配制成液体培养基,并将该液体培养基分装到三角瓶中,每500ml的三角瓶分装200ml的液体培养基,置于121℃的条件下灭菌30分钟后,将步骤(B-1)培养得到的固体菌种接种到液体培养基中后,将三角瓶置于摇瓶机上并在22-25℃恒温条件下以转速180rpm培养96小时,然后在静置培养48h,镜检无杂菌的待用;
(B-3)灵芝菌丝体栽培种培养
将豆粉700g,谷糠200g,玉米面47g,硫酸锌3g,麦芽糖20g,蔗糖20g,石膏10g,亚硒酸氢钠0.3g,加入至饮用水200g中配制成栽培种培养基,并将该500g栽培种培养基分装于栽培种的袋中,置于121℃的条件下灭菌2小时后,将步骤(B-2)培养得到的液体菌种接种到栽培种培养基中,22-25℃恒温培养60天至长满菌包;待灵芝菌丝体长好后,在70℃的条件下沸腾干燥5分钟,将干燥后的灵芝菌丝体过80目筛待用;
C.将步骤A、B分别培养出的羊肚菌丝体和灵芝菌丝体按照实施例1-3中羊肚菌丝体和灵芝菌丝体的比例混匀后即可制得羊肚和灵芝复合制剂。
制得的羊肚和灵芝复合制剂可以做成片剂、胶囊剂和颗粒剂。将羊肚和灵芝复合制剂用乙醇制成颗粒装入复合膜即为颗粒剂;将羊肚和灵芝复合制剂装入硬胶囊即为胶囊剂;在羊肚和灵芝复合制剂中加入适量的糊精、淀粉等辅料压制成片即成片剂。
本发明涉及的羊肚菌丝体含有多种多糖、酶类、氨基酸、脂肪、吡喃酮抗生素、无机元素等成份,具有增强人体免疫力、抑制肿瘤、防癌、调节人体免疫力、预防动脉硬化等功能。
灵芝菌丝体含有多方面的生物活性的多糖和三萜类化合物,营养成分十分丰富,具有免疫调节、抗肿瘤、抗氧化、抗病毒、抗衰老、降血糖、降血脂等多方面的功能。
本发明采用灵芝菌丝体,羊肚菌丝体按一定比例配制而成的羊肚和灵芝复合制剂是一种可以提高免疫活性、抗疲劳、抗肿瘤、降血脂、防癌的一种多糖制剂,适应于亚健康,易于疲倦的人;头晕、失眠多梦的人;脾胃虚弱、消化不良、饮食不振者;高血压、高血脂、高血糖及动脉硬化者;冠心病患者;理气化痰、痰多咳嗽、气短、气喘者等。
将本发明的羊肚和灵芝复合制剂应用于临床上进行对此实验:选取符合病例对象200人,年龄在40-67岁,男女比例1:1,服用量每晚10-12g,每天一次,7天见效,痊愈150例,好转50例,无效0人。此药疗效高于当初实验设想。

1.一种羊肚和灵芝复合制剂,其特征在于,包括以下重量份的各组分:
羊肚菌丝体300-600份 灵芝菌丝体300-600份。
2.一种羊肚和灵芝复合制剂的制备方法,其特征在于,包括以下步骤:
A.羊肚菌丝体培养
(A-1)羊肚菌固体菌种培养
将琼脂25g,土豆200g,葡萄糖20g,KH2PO4 3g,MgSO4·7H2O 1.5g,维生素B1 1片,加入至饮用水1000ml中配制成固体培养基,并将该固体培养基分装到扁平瓶中,置于121℃的条件下灭菌30分钟后,将羊肚菌菌种接种到固体培养基中,22-25℃恒温培养7天;
(A-2)羊肚菌液体菌种培养
将小米50g,蔗糖20g,玉米面25g,豆粉17.5g,蛋白胨2g,葡萄糖1g,KH2PO4 3g,MgSO4·7H2O 1.5g,加入至饮用水1000ml中配制成液体培养基,并将该液体培养基分装到三角瓶中,置于121℃的条件下灭菌30分钟后,将步骤(A-1)培养得到的固体菌种接种到液体培养基中后,将三角瓶置于摇瓶机上并在22-25℃恒温条件下以转速180rpm培养96小时,然后在静置培养48h;
(A-3)羊肚菌丝体栽培种培养
将豆粉700g,谷糠200g,玉米面47g,硫酸锌3g,麦芽糖20g,蔗糖20g,石膏10g,亚硒酸氢钠0.3g,加入至饮用水200g中配制成栽培种培养基,并将该栽培种培养基分装于栽培种的袋中,置于121℃的条件下灭菌2小时后,将步骤(A-2)培养得到的液体菌种接种到栽培种培养基中,22-25℃恒温培养15天;待羊肚菌丝体长好后,在70℃的条件下沸腾干燥5分钟,将干燥后的羊肚菌丝体过80目筛待用;

(B-2)灵芝菌液体菌种培养

将豆粉700g,谷糠200g,玉米面47g,硫酸锌3g,麦芽糖20g,蔗糖20g,石膏10g,亚硒酸氢钠0.3g,加入至饮用水200g中配制成栽培种培养基,并将该栽培种培养基分装于栽培种的袋中,置于121℃的条件下灭菌2小时后,将步骤(B-2)培养得到的液体菌种接种到栽培种培养基中,22-25℃恒温培养60天;待灵芝菌丝体长好后,在70℃的条件下沸腾干燥5分钟,将干燥后的灵芝菌丝体过80目筛待用;
C.将步骤A、B分别培养出的羊肚菌丝体和灵芝菌丝体按照羊肚菌丝体300-600份和灵芝菌丝体300-600份的比例混匀后即可制得羊肚和灵芝复合制剂。
