Abstract

灯盏花素冻干粉及其肠溶片或肠溶胶囊的制备方法,取灯盏花素,加入2.5~3.5倍量的纯化水和0.95~1.2倍量的磷酸氢二钠加热溶解3~5分钟,并控制溶液PH值在7.2‑7.5范围内,溶液冷却后成灯盏花素混悬液;将灯盏花素混悬液冷冻干燥得到灯盏花素冻干粉,再加入淀粉、预胶化淀粉、白糖、微晶纤维素混合,加入乙醇制成软材后制粒;然后在60~75℃下干燥,经整粒后加入硬脂酸镁得到混合颗粒,将混合颗粒压片后包衣后即得灯盏花素冻干肠溶片;或将混合颗粒进行胶囊填充即得灯盏花素冻干肠溶胶囊。本发明制备的灯盏花素冻干粉及其肠溶片或肠溶胶囊安全、人体易于吸收、药效好。

Description

灯盏花素冻干肠溶片或肠溶胶囊的制备方法
技术领域
本发明属于植物药制备技术领域,具体涉及灯盏花素冻干肠溶片、灯盏花素冻干肠溶胶囊的制备方法。
背景技术
灯盏花又名灯盏细辛,属菊科短葶飞蓬属植物,学名Erigeronbreviscapus(Vant.)Hand.Mazz.,是云南特有的野生天然药物资源。目前灯盏花素的制剂主要有灯盏花素片、注射用灯盏花素、灯盏花素咀嚼片、灯盏花素分散片、灯盏花素氯化钠注射液、灯盏花素葡萄糖注射液、灯盏花素注射液、灯盏花素滴丸。以上制剂中除注射液在人体吸收较好外,口服固体制剂吸收较差,中药注射剂存在安全性和不良反应问题。如何提供一种安全、人体吸收好、药效明显的灯盏花素口服固体制剂成为行业急需解决的问题。
发明内容
本发明的目的在于针对现有灯盏花素制剂的缺陷,提供一种安全、人体易于吸收、药效好的灯盏花素冻干肠溶片或肠溶胶囊的制备方法,满足人们的用药需求。
本发明的目的通过以下技术方案予以实现。
灯盏花素冻干粉的制备方法,包括以下步骤:
(1)取灯盏花素,按质量份数加入2.5~3.5倍量的纯化水和0.95~1.2倍量的磷酸氢二钠加热溶解3~5分钟,并控制溶液PH值在7.2-7.5范围内,溶液冷却后成灯盏花素混悬液;
(2)将步骤(1)制备得到的灯盏花素混悬液冷冻干燥得到灯盏花素冻干粉。
采用本发明制备的灯盏花素冻干粉制备灯盏花素肠溶片或肠溶胶囊的方法,包括以下步骤:
(1)取制备得到的灯盏花素冻干粉,加入淀粉、预胶化淀粉、白糖、微晶纤维素混合均匀,每生产灯盏花素冻干肠溶片10000片或灯盏花素冻干肠溶胶囊10000粒,需要灯盏花素冻干粉折纯灯盏花素0.2kg、淀粉0.75~0.9kg、预胶化淀粉0.35~0.45kg、白糖0.75~0.9kg、微晶纤维素0.18~0.25kg,然后加入质量浓度63%-65%的乙醇0.55~0.65kg制成软材,过18~26目筛网制成粒;
(2)将步骤(1)制备得到的颗粒在60~75℃下干燥,将可生产灯盏花素冻干肠溶片10000片或灯盏花素冻干肠溶胶囊10000粒的干燥品经2号筛整粒后加入0.018~0.025kg硬脂酸镁混合,制得混合颗粒;
(3)将步骤(2)制备得到的混合颗粒压片得到灯盏花素片芯后包肠溶衣,包衣后即得灯盏花素冻干肠溶片;或将混合颗粒进行胶囊填充即得灯盏花素冻干肠溶胶囊。
本发明利用灯盏花素碱溶酸沉的理化特点,制备得到的灯盏花素冻干粉,最大限度地保留了灯盏花素的药效。进一步开发的灯盏花素口服固体制剂,服用安全。本发明方法制备过程中药效损失小,制备得到的灯盏花素冻干肠溶片、灯盏花素冻干肠溶胶囊可避免在胃液中溶解,而是进入肠道溶解并进行吸收,有效克服了2015版《中国药典》收录的灯盏花素片口服固体制剂成药在实际使用过程中由于人体胃液成强酸性、灯盏花素在酸性条件下无法溶解而导致的人体吸收差、生物利用度低的问题,有效提高了药品生物的利用度。本发明制备工艺简单,制作成本低,可满足人们的用药需求。
具体实施方式
实施例1
取符合2015版《中国药典》口服标准的灯盏花素500g(其野黄芩苷含量为92.25%),加入纯化水1500ml和磷酸氢二钠500g,加热溶解4分钟,PH值为7.3,溶液冷却后成灯盏花素混悬液。将灯盏花素混悬液冷冻干燥得到灯盏花素冻干粉980g,收率为98%。灯盏花素冻干粉通过高效液相检测,野黄芩苷含量为46.09%。取灯盏花素冻干粉434g,相当于纯灯盏花素200g,加入淀粉810g、预胶化淀粉400g、白糖810g、微晶纤维素208g混合均匀,然后加入质量浓度65%的乙醇600g制成软材,过20目不锈钢筛网制成颗粒,颗粒在65℃下干燥,干燥颗粒经2号筛整粒,加入硬脂酸镁20g混合得到混合颗粒。将混合颗粒压片得到灯盏花素片芯,再加入HPMCP肠溶包衣剂进行包衣,即得灯盏花素冻干肠溶片,随后包装即可。
实施例2
取符合2015版《中国药典》口服标准的灯盏花素500g(其野黄芩苷含量为92.25%),加入纯化水1250ml和磷酸氢二钠475g,加热溶解3分钟,PH值为7.4,溶液冷却后成混悬液。将灯盏花素混悬液冷冻干燥得到灯盏花素冻干粉954.53g,收率为97.9%。冻干粉通过高效液相检测,野黄芩苷含量为47.31%,取冻干粉423g,相当于灯盏花素200g,加入淀粉900g、预胶化淀粉350g、白糖900g、微晶纤维素250g混合均匀后加入质量浓度63%的乙醇650g制成软材,过26目不锈钢筛网制成颗粒,颗粒在60℃下干燥,干燥颗粒2号筛整粒,加入硬脂酸镁25g混合。将混合后的颗粒用肠溶胶囊填充,即得灯盏花素冻干肠溶胶囊。将得到的胶囊用铝塑铝包装即可。
实施例3
取符合2015版《中国药典》口服标准的灯盏花素500g(其野黄芩苷含量为92.25%),加入纯化水1750ml和磷酸氢二钠600g,加热溶解5分钟,PH值为7.5,溶液冷却后成灯盏花素混悬液。将灯盏花素混悬液冷冻干燥得到灯盏花素冻干粉1079g,收率为98.1%。灯盏花素冻干粉通过高效液相检测,野黄芩苷含量为41.91%。取灯盏花素冻干粉477g,相当于灯盏花素200g,加入淀粉750g、预胶化淀粉450g、白糖750g、微晶纤维素180g混合均匀,然后加入质量浓度64%的乙醇550g制成软材,过18目不锈钢筛网制成颗粒,颗粒在75℃下干燥,干燥颗粒经2号筛整粒,加入硬脂酸镁18g混合得到混合颗粒。将混合颗粒压片得到灯盏花素片芯,再加入HPMCP肠溶包衣剂进行包衣,即得灯盏花素冻干肠溶片,随后包装即可。
除非另有说明,本发明所采用的百分数和倍量均为质量百分数和质量倍量。

Claims (2)

1.灯盏花素冻干粉的制备方法,其特征在于,包括以下步骤:
(1)取灯盏花素,按质量份数加入2.5~3.5倍量的纯化水和0.95~1.2倍量的磷酸氢二钠加热溶解3~5分钟,并控制溶液PH值在7.2-7.5范围内,溶液冷却后成灯盏花素混悬液;
(2)将步骤(1)制备得到的灯盏花素混悬液冷冻干燥得到灯盏花素冻干粉。
2.采用权利要求1制备的灯盏花素冻干粉制备灯盏花素肠溶片或肠溶胶囊的方法,其特征在于,包括以下步骤:
(1)取制备得到的灯盏花素冻干粉,加入淀粉、预胶化淀粉、白糖、微晶纤维素混合均匀,每生产灯盏花素冻干肠溶片10000片或灯盏花素冻干肠溶胶囊10000粒,需要灯盏花素冻干粉折纯灯盏花素0.2kg、淀粉0.75~0.9kg、预胶化淀粉0.35~0.45kg、白糖0.75~0.9kg、微晶纤维素0.18~0.25kg,然后加入质量浓度63%-65%的乙醇0.55~0.65kg制成软材,过18~26目筛网制成粒;
(2)将步骤(1)制备得到的颗粒在60~75℃下干燥,将可生产灯盏花素冻干肠溶片10000片或灯盏花素冻干肠溶胶囊10000粒的干燥品经2号筛整粒后加入0.018~0.025kg硬脂酸镁混合,制得混合颗粒;
(3)将步骤(2)制备得到的混合颗粒压片得到灯盏花素片芯后包肠溶衣,包衣后即得灯盏花素冻干肠溶片;或将混合颗粒进行胶囊填充即得灯盏花素冻干肠溶胶囊。
