The moisturizing effect of the whitening and moisturizing cream prepared in the embodiment 1 of the invention and the moisturizing creams prepared in the comparative examples 1-2 are subjected to a parallel comparison test:
60 young white-collar workers are selected as volunteers of the test, and the skin is dry and has the problem of dry skin. The moisture content of the inner forearm skin of each volunteer was measured using a skin moisture tester (CM820 Courage Khazaka electronic GmbH, germany) under an indoor environment of constant temperature and constant humidity (24 ± 1 ℃ and 40% ± 1 humidity), and the average value was taken by repeating the test three times, and the moisture content test value of the inner forearm skin was less than 35.
The 60 volunteers were randomly and equally divided into 3 test groups of 20 individuals each, with no statistical difference between groups. The whole test was carried out at constant temperature and constant humidity (24. + -. 1 ℃ and 40%± 1 humidity) in an indoor environment. Before formal test, the patient should sit still for at least 30min, with the forearm exposed, and be placed in the test state to maintain relaxation. Before using the moisturizing cream, the moisture content of the skin on the inner side of the forearm of each group of volunteers was measured and averaged, wherein each volunteer was averaged by repeating the measurement three times. The moisturizing cream prepared in example 1 and the moisturizing creams prepared in comparative examples 1 to 2 were used by volunteers in test groups 1 to 3, respectively. The specific use method comprises the following steps: marking the same part on the inner forearm at 4X 4cm2Test area, at 2.0. + -. 0.1mg/cm2The amount of the moisturizing cream is smeared, and the moisturizing cream is evenly massaged in a test area by using a latex finger stall so as to be fully absorbed. Skin moisture content was measured immediately after use and 2 hours, 4 hours after use, and the test was repeated three times per volunteer for averaging, and then the test values of each group of volunteers were averaged.
The test results are shown in table 2:
TABLE 2
Figure DEST_PATH_IMAGE004
In the test process, the phenomena of irritability, skin discomfort and the like do not appear in all subjects.
From the test results, it can be seen that the moisturizing cream of example 1 of the present invention has excellent moisturizing effects. The experimental test data of example 1 and comparative examples 1-2 can be used to conclude that: according to the invention, the moisturizing active ingredient obtained by compounding the cacao extract and the lycium barbarum polysaccharide in a specific ratio has an obvious synergistic effect in the aspects of moisturizing, and obtains unexpected technical effects.
The test for inhibiting tyrosinase activity was performed on the whitening active ingredients used in the present invention:
firstly, preparing a sample solution:
(1) taking a proper amount of the Brazilian cocoa extract prepared in the example 1, and then preparing a Brazilian cocoa extract sample solution with the mass concentration of 5mg/ml, 2.5mg/ml, 1.25mg/ml, 0.625mg/ml and 0.3125mg/ml by using a phosphate buffer solution with the pH of 6.8;
(2) taking a proper amount of the lycium barbarum polysaccharide prepared in the example 1, and then preparing a lycium barbarum polysaccharide sample solution with the mass concentration of 5mg/ml, 2.5mg/ml, 1.25mg/ml, 0.625mg/ml and 0.3125mg/ml by using a phosphate buffer solution with the pH value of 6.8;
(3) taking the mass ratio of 3: 2, and then preparing sample solutions with mass concentrations of 5mg/ml, 2.5mg/ml, 1.25mg/ml, 0.625mg/ml and 0.3125mg/ml by using a phosphate buffer solution with the pH of 6.8.
Secondly, a test method comprises the following steps: for sample solution with each concentration, taking four test tubes and respectively marking the test tubes with No. 1, No. 2, No. 3 and No. 4; referring to the reagent amount and concentration in Table 3, the corresponding reaction solution listed in each column was added to four tubes, mixed uniformly with shaking, preheated at 37 ℃ for 10min, then 1ml of 5 mmol/L-dopa reaction solution was added to each tube, reacted at 37 ℃ for 5min accurately, transferred to a cuvette rapidly, and the absorbance value (A) was measured at 475 nm. The sample solutions at each concentration were tested in triplicate and averaged.
Thirdly, calculating the inhibition rate of tyrosinase according to the following formula:
inhibition rate = [1- (A3-A4)/(A1-A2) ] × 100%
In the formula: a1, A2, A3 and A4 represent absorbance values at 475nm after reaction in four test tubes No. 1, No. 2, No. 3 and No. 4 in Table 3, respectively.
The inhibition data for the different concentrations of the extract solutions were plotted using GraphPad Prism software to generate standard curves and calculate the half-inhibitory concentration IC50 for each extract, the results of which are shown in table 4. The efficacy of the inhibitor was characterized by the concentration of the inhibitor (IC 50) that resulted in a 50% decrease in tyrosinase activity, with smaller values of IC50 indicating a greater ability of the extract to inhibit tyrosinase activity.

20 female white-collar workers are selected as subjects, the age range is 25-39 years, the average age is 31 years, the health and the regular work and rest are realized, no skin disease or allergic disease history exists, and cosmetics can be used according to the specification. The facial skin of the above subjects had problems of dull skin, pigmentation, and dullness.
The test method comprises the following steps: the test environment temperature is 22 deg.C + -1 deg.C, humidity is 50% + -5%, and the skin melanin content of the same part of the face is measured by MX18 skin pigment detector of CK company in Germany 1 day before the application of cosmetics and 7 days after the continuous application of skin care products, and the average value is obtained by repeating the measurement three times.
The use method of the cosmetic comprises the following steps: after the face is washed with warm boiled water respectively in the morning and before bedtime, a proper amount of the moisturizing cream of the embodiment 1 of the invention is uniformly smeared on the face, and then the face is gently massaged to uniformly absorb the cosmetics. All subjects did not use any other cosmetic or skin care product during the test.
Melanin reduction rate = (melanin content 1 day before use of cosmetics-black pigment content on the second day 7 days after continuous use of cosmetics)/melanin content 1 day before use of cosmetics × 100%. The higher the melanin reduction rate, the more obvious the whitening effect.
Effect determination criteria: the method has the following advantages: the skin color is brightened, the pigmentation fading effect is very obvious, and the melanin reduction rate is more than 30 percent; secondly, the method is effective: the skin color is improved, the pigmentation fading effect is obvious, and the melanin reduction rate is 5-30%; ③ invalid: the skin color and pigmentation are not obviously changed compared with the skin color and the pigmentation before the test, and the melanin reduction rate is lower than 5 percent.
And (3) test results: 18 cases of significant effect, 2 cases of effect and 0 case of no effect, the significant efficiency is 90 percent, and the total effective rate is 100 percent.
The above-mentioned embodiments are merely preferred embodiments of the present invention, and the scope of the present invention should not be limited thereby, and those skilled in the art should not make any insubstantial changes and substitutions based on the present invention.
